A widespread pattern of the Tibetan plateau is mosaics of grasslands of Cyperaceae and grasses with forbs, interspersed with patches covered by lichen crusts induced by overgrazing. However, the fate of inorganic and organic N in non-crusted and crusted patches in Kobresia grasslands remains unknown. We reported on a field 15N-labeling experiment in two contrasting patches to compare retention of organic and inorganic N over a period of 29 days. 15N as KNO3, (NH4)2SO4 or glycine was sprayed onto soil surface. Crusted patches decreased plant and soil N stocks. More 15N from three N forms was recovered in soil than plants in both patches 29 days after the labeling. In non-crusted patches, 15N recovery by the living roots was about two times higher than in crusted ones, mainly because of higher root biomass. Microorganisms in non-crusted patches were N-limited because of more living roots and competed strongly for N with roots. Inorganic N input to non-crusted patches could alleviate N limitation to plants and microorganisms, and leads to higher total 15N recovery (plant + soil) for inorganic N forms. Compared to non-crusted patches, microorganisms in crusted patches were more C-limited because of depletion of available C caused by less root exudation. Added glycine could activate microorganisms, together with the hydrophobicity of glycine and crusts, leading to higher 15N-glycine than inorganic N. We conclude that overgrazing-induced crusts in Kobresia grasslands changed the fate of inorganic and organic N, and lead to lower total recovery from inorganic N but higher from organic N. Since the late 1950s, governmental rangeland policies have changed the grazing management on the Tibetan Plateau ( TP). Increasing grazing pressure and, since the 1980s, the privatization and fencing of pastures near villages has led to land degradation, whereas remote pastures have recovered from stronger overgrazing. To clarify the effect of moderate grazing on the carbon ( C) cycle of the TP, we investigated differences in below-ground C stocks and C allocation using in situ 13 CO2 pulse labeling of (i) a montane K obresia winter pasture of yaks, with moderate grazing regime and (ii) a 7-year-old grazing exclosure plot, both in 3440 m asl. Twenty-seven days after the labeling, 13 C incorporated into shoots did not differ between the grazed (43% of recovered 13 C) and ungrazed (38%) plots. In the grazed plots, however, less C was lost by shoot respiration (17% vs. 42%), and more was translocated below-ground (40% vs. 20%). Within the below-ground pools, <2% of 13 C was incorporated into living root tissue of both land use types. In the grazed plots about twice the amount of 13 C remained in soil (18%) and was mineralized to CO2 (20%) as compared to the ungrazed plots (soil 10%; CO2 9%). Despite the higher contribution of root-derived C to CO2 efflux, total CO2 efflux did not differ between the two land use types. C stocks in the soil layers 0-5 and 5-15 cm under grazed grassland were significantly larger than in the ungrazed grassland. However, C stocks below 15 cm were not affected after 7 years without grazing. We conclude that the larger below-ground C allocation of plants, the larger amount of recently assimilated C remaining in the soil, and less soil organic matter-derived CO2 efflux create a positive effect of moderate grazing on soil C input and C sequestration. The montane grassland soils of Europe store significant amounts of nitrogen (N), and climate change might drive their volatilization due to the stimulation of gaseous nitrous oxide (N2O) and dinitrogen (N2) losses. Hence, a thorough, mechanistic understanding of the processes responsible for N loss and retention such as denitrification and dissimilatory nitrate reduction to ammonium (DNRA) in these soils is urgently needed. Here we aimed to explore the relationships between denitrifier gene abundance and expression with N2 and N2O production and the importance of DNRA versus denitrification in nitrate consumption and N2O production for typical montane grassland soils of Southern Germany. In a laboratory incubation experiment with glucose and nitrate addition, we combined direct measurements of N2O and N2 production with a molecular analysis of the denitrifier communities involved in nitrite, nitric oxide (NO) and N2O reduction and with the quantification of DNRA. The soils originated from a space-for-time climate change experiment, where intact plant-soil mesocosms were exposed for three years either to ambient conditions at a high elevation site ("HE" control treatment) or to predicted climate change conditions (warming, reduced summer precipitation and reduced winter snow cover) by translocation to lower elevation ("LE" climate change treatment). The abundance (DNA) of cnorB genes was significantly reduced in LE soils, whereas the abundance of nosZ genes did not differ between the HE and LE soils. However, the decreased abundance of cnorB genes unexpectedly resulted in slightly increased rather than decreased potential N2O emissions. This effect could be explained by the increased levels of cnorB mRNA and, therefore, the higher physiological activity of the NO reducers in the LE soils. In contrast with the DNA levels, the dynamics of the cnorB mRNA levels followed N2O emission patterns, whereas the nosZ expression was strongly correlated with the N2 emission (R2 = 0.83). The potential rates of DNRA were approximately one-third of the rates of denitrification, and DNRA was not a source for N2O. We conclude that DNRA significantly competes with denitrification in these soils, thus contributing to N conservation. This work demonstrates that the molecular analysis of nosZ gene expression has great potential to contribute to solving the enigmatic problem of understanding N2 loss from soil. With 450,000 km 2 Kobresia (syn. Carex) pygmaea dominated pastures in the eastern Tibetan highlands are the world's largest pastoral alpine ecosystem forming a durable turf cover at 3000-6000 m a.s.l. Kobresia's resilience and competitiveness is based on dwarf habit, predominantly below-ground allocation of photo assimilates, mixture of seed production and clonal growth, and high genetic diversity. Kobresia growth is co-limited by livestock-mediated nutrient withdrawal and, in the drier parts of the plateau, low rainfall during the short and cold growing season. Overstocking has caused pasture degradation and soil deterioration over most parts of the Tibetan highlands and is the basis for this man-made ecosystem. Natural autocyclic processes of turf destruction and soil erosion are initiated through polygonal turf cover cracking, and accelerated by soil-dwelling endemic small mammals in the absence of predators. The major consequences of vegetation cover deterioration include the release of large amounts of C, earlier diurnal formation of clouds, and decreased surface temperatures. These effects decrease the recovery potential of Kobresia pastures and make them more vulnerable to anthropogenic pressure and climate change. Traditional migratory rangeland management was sustainable over millennia, and possibly still offers the best strategy to conserve and possibly increase C stocks in the Kobresia turf., (Copyright © 2018 The Authors.
