8. Bcl-2 May Contribute to Evolution of Endometrial Hyperplasia, but It Isn’t a Factor in Subsequent Carcinogenesis.

Abstract

Endometrial carcinoma is a prevalent disease in the Western world and is experiencing an upward trend in developing countries as well. Endometrial hyperplasia is regarded as a precancerous lesion. Apoptosis plays an important role in the neoplastic transformation of cells, with Bcl-2 serving as an anti-apoptotic cellular marker. It is possible that Bcl-2 may plays an important role in the development of endometrial carcinoma. The objective was to evaluate and compare the expression of the Bcl-2 markers accross the spectrum of normal endometrium endometrial hyperplasia and endometrial adenocarcinoma. A total of 50 cases were included in this study, comprising of 10 cases of normal endometrium, 10 cases of endometrial hyperplasia without atypia, 10 cases of atypical endometrial hyperplasia and 20 cases of endometrial adenocarcinomas. The cases were collected from January 2017 to June 2018. Immunohistochemical staining with Bcl-2 was performed and the results were subsequently analyzed. Bcl-2 staining demonstrated a notable increase in cases exhibiting with strong staining intensity, from 20% in normal endometrial tissue to 75% in cases of endometrial hyperplasia. However, there was a notable decline in the number of cases exhibiting with strong intensity Bcl-2 staining as the lesions progressed from endometrial hyperplasia to endometrial carcinoma (30% of the cases). The results were statistically significant (P =0.00309). However, there was no significant association was observed between staining and either atypical hyperplasia or endometrial carcinomas (p=0.429), or between the degree of carcinoma and staining (p=0.6903). Bcl-2 expression demonstrated an increase from cases of normal endometrium to endometrial hyperplasia, which supports the hypothesis that there is an increase in anti-apoptotic activity in endometrial hyperplastic lesions. The observed decrease in Bcl-2expression in endometrioid adenocarcinoma when compared to endometrial hyperplasia may indicate the involvement of alternative mechanisms of carcinogenesis, potentially beyond the failure of apoptosis. Abstract

The focus is now on sustainable development, which is inevitable without harnessing renewable energy sources. The fundamental element in wind wave generation is the interaction between air and sea which helps in momentum exchange between atmosphere and ocean. The Indian coastline is under a dynamic wave climate with the action of wind. Indian landmass has two tropical basins, the Bay of Bengal and the Arabian Sea, which have tremendous potential to tap renewable energy. The variations in wave climate due to dynamic-wind have to be assessed. Hindcast data obtained from Global Climate Models help us in the long-term analysis of wind and wave climate. In an attempt to explore the renewable energy potential along the Indian coast, a numerical wave model is developed using MIKE 21 SW module to assess the wind and wave climate. A gridded global wind speed dataset from ECMWF called ERA-Interim wind speed data of 38 years (1981 to 2018) is used as input for the numerical model. The dataset and numerical model performance were validated against in-situ measurements. The results showed amongst the locations studied off Goa, Karnataka, Kerala, Tamil Nadu, and Andhra Pradesh had good potential to extract offshore wind energy using offshore wind turbines. 13. Plant species composition and product utility pattern of Garo homegardens in Meghalaya, India

Abstract

Home garden is a traditional landuse system practiced by many rural households in the tropical region. The composition and management practices within homegardens are largely driven by cultural setup and ecological conditions. The present study characterized the plant species composition, utility patterns and management of Garo homegardens in Dadenggre block, West Garo Hill district of Meghalaya, India. Fifty households from 5 villages were randomly selected and interviewed using a semi-structured questionnaire. The homegardens size ranged between 0.07 and 1.29ha, harbouring 132 plant species, out of which 74 species were trees, 19 shrubs and 39 herbs. Among the perennials, Areca catechu (areca nut) was the most common contributor to household earnings. When species were grouped into 9 utility classes (timber, medicinal, fruit, fuelwood, fodder, vegetables, ornamental, spice, and others), highest number was for fuelwood, followed by vegetables and fruits. The average household income was Rs. 318/100m2, the highest contribution from the sale of vegetables. Various home garden management activities were conducted, engaging family members and generating employment for others. Animal rearing is common in many households and the application of animal manure and household waste has helped maintain soil fertility of homegardens’ soils. Homegardens are integral to the Garo society, contributing significantly to household needs and activities.

Abstract

Identification of genes/proteins involved in pluripotency and their inter-relationships is important for understanding the induction/loss and maintenance of pluripotency. With the availability of large volume of data on interaction/regulation of pluripotency scattered across a large number of biological databases and hundreds of scientific journals, it is required a systematic integration of data which will create a complete view of pluripotency network. Describing and interpreting such a network of interaction and regulation (i.e., stimulation and inhibition) links are essential tasks of computational biology, an important first step in systems-level understanding of the underlying mechanisms of pluripotency. To address this, we have assembled a network of 166 molecular interactions, stimulations and inhibitions, based on a collection of research data from 147 publications, involving 122 human genes/proteins, all in a standard electronic format, enabling analyses by readily available software such as Cytoscape and its Apps (formerly called "Plugins"). The network includes the core circuit of OCT4 (POU5F1), SOX2 and NANOG, its periphery (such as STAT3, KLF4, UTF1, ZIC3, and c-MYC), connections to upstream signaling pathways (such as ACTIVIN, WNT, FGF, and BMP), and epigenetic regulators (such as L1TD1, LSD1 and PRC2). We describe the general properties of the network and compare it with other literature-based networks. Gene Ontology (GO) analysis is being performed to find out the over-represented GO terms in the network. We use several expression datasets to condense the network to a set of network links that identify the key players (genes/proteins) and the pathways involved in transition from one state of pluripotency to other state (i.e., native to primed state, primed to non-pluripotent state and pluripotent to non-pluripotent state).
