Considering as basis the structure of a new model for evaluating performances in health and safety on workplace, the measures are introduced to allow the quantification of the goodness of the system, in order to verify how much the implemented system matches with the workers safeguard as requested by the in force law. Quantification consists in the inclusion of two different types of tools: check-lists and indicators. These tools are based on the tree structure of the model that is constituted by 6 key-elements, each of them formed by several themes (up to an amount of 27 themes). The quantification of check-lists and indicators allows to assess the importance of themes and key-elements and to lay the foundation of a complete score, that means the index of global performance of the examined company is obtained. In conclusion, once the quantification is obtained, or in other words, once the performance of company management system is estimated, it will be possible to identify, if it is the case, the priority of interventions for improvements, so that the management process becomes more effective and efficient.

Pituitary gonadotrope cells are essential for the endocrine regulation of reproduction in vertebrates. These cells emerge early during embryogenesis, colonize the pituitary glands and organize in tridimensional networks, which are believed to be crucial to ensure proper regulation of fertility. However, the molecular mechanisms regulating the organization of gonadotrope cell population during embryogenesis remain poorly understood. In this work, we characterized the target genes of NEUROD1 and NEUROD4 transcription factors in the immature gonadotrope αT3-1 cell model by in silico functional genomic analyses. We demonstrated that NEUROD1/4 regulate genes belonging to the focal adhesion pathway. Using CRISPR/Cas9 knock-out approaches, we established a double NEUROD1/4 knock-out αT3-1 cell model and demonstrated that NEUROD1/4 regulate cell adhesion and cell motility. We then characterized, by immuno-fluorescence, focal adhesion number and signaling in the context of NEUROD1/4 insufficiency. We demonstrated that NEUROD1/4 knock-out leads to an increase in the number of focal adhesions associated with signaling abnormalities implicating the c-Src kinase. We further showed that the neurotrophin tyrosine kinase receptor 3 NTRK3, a target of NEUROD1/4, interacts physically with c-Src. Furthermore, using motility rescue experiments and time-lapse video microscopy, we demonstrated that NTRK3 is a major regulator of gonadotrope cell motility. Finally, using a Ntrk3 knock-out mouse model, we showed that NTRK3 regulates gonadotrope cells positioning in the developing pituitary, in vivo. Altogether our study demonstrates that the Neurod1/4-Ntrk3-cSrc pathway is a major actor of gonadotrope cell mobility, and thus provides new insights in the regulation of gonadotrope cell organization within the pituitary gland., (© 2023. This article considers the intersection of multiple and, at times, seemingly conflicting temporalities in Andean childhoods. We draw on Silvia Rivera Cusicanqui's scholarship on Andean sociology and our ethnographic research with Cañari families to argue that Cañari families' and children's relations with cows and milk production are fueled by both capitalist and Andean temporalities that cannot be thought as opposites. These temporal relations do not create confusion or limiting binaries but are, we propose, itinerant. Background: Gonadotrope lineage differentiation is a stepwise process taking place during pituitary development. The early step of gonadotrope lineage specification is characterized by the expression of the Nr5a1 transcription factor, a crucial factor for gonadotrope cell fate determination. Abnormalities affecting Nr5a1 expression lead to hypogonadotropic hypogonadism and infertility. Although significant knowledge has been gained on the signaling and transcriptional events controlling gonadotrope differentiation, epigenetic mechanisms regulating Nr5a1 expression during early gonadotrope lineage specification are still poorly understood., Results: Using ATAC chromatin accessibility analyses on three cell lines recapitulating gradual stages of gonadotrope differentiation and in vivo on developing pituitaries, we demonstrate that a yet undescribed enhancer is transiently recruited during gonadotrope specification. Using CRISPR/Cas9, we show that this enhancer is mandatory for the emergence of Nr5a1 during gonadotrope specification. Furthermore, we identify a highly conserved estrogen-binding element and demonstrate that the enhancer activation is dependent upon estrogen acting through ERα. Lastly, we provide evidence that binding of ERα is crucial for chromatin remodeling of Nr5a1 enhancer and promoter, leading to RNA polymerase recruitment and transcription., Conclusion: This study identifies the earliest regulatory sequence involved in gonadotrope lineage specification and highlights the key epigenetic role played by ERα in this differentiation process.

Background: Breastfeeding is definitely one of the measures capable of improving not only children's but also mother's health. Human milk banks are institutions providing human milk to babies with limited access to breastfeeding for various reasons., Study Design: The aim of this observational retrospective study was to examine principal characteristics of breast milk donors in the province of Siena and to identify variables influencing Milk Quantity (MQ) and Length of donation period (LD)., Methods: Information was extracted from medical records of Human Milk Bank of Siena, all women that donated their breast milk during the period from January 2010 until August 2017 were included. Examined variables were: age, place of birth (Italy/outside Italy), residence (Siena city/Siena province), education, profession, type of labor (preterm/in-term) and type of delivery (vaginal/cesarean section), gestational age, number of children, previous donations (blood, milk), quantity of donated milk and length of donation., Results: A total of 304 donors were included: 75.7% of Italian nationality. The mean age was 32.4±5.2, Italian donors were older (33.5±5.0 vs. 28.7±4.2; Mann-Whitney; p<0.001). Socio-economic situation of Italian donors was better compared to non-Italian donors (chi-squared; p<0.005). Non-Italian donors had more probability to go through preterm labor (OR=3.68; p<0.001). Average length of donation was 2.7±1.8 months. Mean quantity of donated milk was 4.8±7.6l. From multiple linear regression, preterm birth (p<0.005) and length of donation (p<0.001) emerged as a variable that can predict higher quantity of donated milk., Conclusions: Preterm babies are usually recovering in a neonatology intensive therapy unit, with mothers staying close to them, which facilitates the whole donation process. Mothers of premature babies have a higher perception of the meaningfulness of donation and a need of adequate nutrition for fragile infants.

This study has tested the effects of hyperbaric oxygen in periodontal structures in agreement with the theories supported by literature research. Eight patients, from 30 to 50 years-of-age, were tested with pure oxygen inhalation, at the 2.5 ATA absolute pressure. Main approved tests of periodontal health were evaluated before and after HBOT’s cycles. The results in all patients treated with HBOT, have founded clear improvement of clinical and instrumental parameters.

The aim of this research was to evaluate the antioxidant capacity and physical-chemical characteristics of commercial white myrtle berry (Myrtus communis L. var. leucocarpa DC) liqueur (WMBL). The total phenolic (TP) content was measured spectrophotometrically, applying a modified Folin-Ciocalteu's method, and phenolic compounds were identified by high-performance liquid chromatography (HPLC) coupled with electrospray mass spectrometry, and quantified by HPLC coupled with ultraviolet/visible detection. The antioxidant capacities were evaluated by FRAP, CUPRAC, DPPH • , and ABTS •+ assays. The volatiles were assessed by gas chromatography and mass spectrometry (GC-MS/FID) after headspace solid-phase microextraction (HS-SPME) and liquid-liquid extraction (LLE). WMBL showed lower TP levels (636.3 ± 39.2 mg GAE/L) than in purple myrtle berry liqueur (PMBL). Nevertheless, WMBL exhibited better antioxidant capacities, potentially due to high concentrations of gallic acid (294.2 ± 14.2 mg/L) and its derivatives (58.3 ± 2.1 mg/L). Other phenolic compounds detected by HPLC-DAD and LC-MS/MS were flavonols like myricetin and its derivatives (myricetin-3-O-galactoside and myricetin-3-O-rhamnoside) with concentrations similar to those found in PMBL. GC-MS/FID analysis revealed 44 compounds (terpenes, higher aliphatic compounds and shikimic acid pathway derivatives). 1,8-Cineole was the most abundant terpene in the liqueur (26.5% (HS-SPME) and 9.6% (LLE)).
