The aim of the present study was to assess dietary and sex-related effects on selected blood indicators of 40-day-old mallard ducks undergoing fattening. The ducks were divided into three dietary groups (control, L50 and L100). The control (C) group was fed a diet containing soybean meal as the main protein component of the feed. In both experimental groups L50 and L100 soybean meal was replaced with meal of whole seeds of white lupine, Zulika variety, in proportions of 50 and 100%, respectively. With respect to the assessed dietary effect, the serum of the L50 group showed a significantly higher albumin concentration as compared to the C group (P < 0.05) accompanied by a significantly lower proportion of γ-globulins in the L50 group as compared to the L100 group (P < 0.01). As for the blood plasma, the C group showed a significantly higher concentration of total cholesterol as compared to the L100 group (P < 0.05) and a highly significant increase in the triacylglyceride (TAG) level as compared to the L50 and L100 groups (P < 0.01). Concerning plasma mineral indicators, ducks of the C group showed a significantly higher calcium concentration (P < 0.05) in comparison to the L100 group and a lower phosphorus concentration (P < 0.01) in comparison to the L50 as well as L100 group. Further, the C group showed a significantly lower level of potassium (K) as compared to the L50 group (P < 0.05) as well as the L100 group (P < 0.01). The sex of the ducks significantly (P < 0.05) affected the proportion of α-globulins in serum and the magnesium level in the plasma and affected the plasma levels of TAG and K and the activity of aspartate aminotransferase (P < 0.01).

The aim of the present study was to evaluate the effects of the inclusion of whole seeds of white lupine and different binder types in complete pelleted diets on selected haematological and plasma biochemical indicators of growing dwarf rabbits. The individual litters of Dwarf Lop rabbit kits were randomised after birth into control (n = 12) and experimental (E) groups. The young rabbits of the E group (n = 24) received an experimental pre-weaning diet containing a 25% share of white lupine seeds. At the time of weaning, the live weights of eight-week-old rabbits in the control group and E group were 727 g and 743 g, respectively. The weaned rabbits of the E group were further equally divided into two post-weaning experimental dietary groups (M = 12; G = 12); these diets contained a 20% share of white lupine seeds each and their composition only differed in the type of the binder (molasses or glycerol component). The rabbits of the control group received a commercial pet rabbit diet with no white lupine seeds during the entire experimental period. At the age of eight weeks, the diet showed a significant effect on the level of albumin (P < 0.05), A/G ratio (P < 0.05) and ALP activity (P < 0.01) in plasma of dwarf rabbit kits. Significant effects of diet were found in 15-week old rabbits in relation to the plasma level of albumin (P < 0.05), Ca (P < 0.05), A/G ratio (P < 0.05) and ALT activity (P < 0.01). The dietary inclusion of white lupine seeds resulted in beneficial health effects on certain blood indicators of rabbits, and their use in the proportion of 25% in the pre-weaning diet and 20% in the post-weaning diet can be recommended as a suitable feed additive for dwarf rabbits. The use of crude glycerol as binder in the experimental diet had no adverse effect on the blood indicators of the growing rabbits and thus it can be recommended for the manufacture of feed pellets intended for dwarf rabbits.

The present study was conducted to determine the effect of dietary inclusion of the wormwood (Artemisia absinthium L.) meal on growth performance, carcass composition, physico-chemical traits and amino acid profile in meats of fattened broilers. In a completely randomised block design, a total of 288 female broilers that were 21 d old (Ross 308) were divided into four dietary groups and fed for 3 weeks: the control basal broiler diet (C), without any anticoccidial or wormwood herb (WH) supplementation, and the C diet plus 10 g (WW1 group), 50 g (WW5 group) or 100 g (WW10 group) of WH meal supplementation per kilogram of basal diet. At the end of the experiment (day 42), broilers were randomly selected for carcass composition and meat quality trait evaluation. In conclusion, the final live weight of chickens was not affected by diets with higher WH levels (P>0.05). For the entire experimental period, the feed conversion ratio raised with an increasing WH level in diets, showing the highest value in chickens of the WW10 group (P<0.01). Dietary supplementation with wormwood (WW) had no negative effects on the carcass composition or on the chemical and physical traits of meat quality assessed. By contrast, it can be assumed that WH dietary supplementation influenced, predominantly, proteosynthesis of chickens, resulting in alteration of amino acid profiles in meats, where especially increasing aspartic acid and valine contents (P<0.001) in the leg meat were found. Our findings indicate that the supplementation of 5 % WH to the diet showed favourable results for chicken performance. However, it is necessary to conduct further studies dealing with WH dietary effects on metabolism and heath control in chickens.

The aim of the present study was to evaluate the effect of breed on haematological and biochemical indicators in 3 dwarf rabbit breeds. In the experiment, 30 sexually intact dwarf rabbit females aged 6 mo were used. With the sole exception of white blood cells and haematocrit value, breed had the most significant effect on the majority of haematological indicators monitored. We investigated the role of the 5-lipoxygenase (5-LOX) pathway of arachidonic acid metabolism in tumour necrosis factor-alpha (TNF-alpha)-induced differentiation of human leukemic HL-60 cells using MK-886, an inhibitor of 5-LOX activating protein. MK-886 augmented cell cycle arrest and differentiation induced by TNF-alpha; however, both effects were probably 5-LOX-independent, because a general LOX inhibitor, NDGA, had no effect. Apoptosis was significantly elevated after combined TNF-alpha and MK-886 treatment, which could be partially associated with changes of Mcl-1 protein expression. NF-kappaB signalling or activation of JNKs were not modulated by MK-886. Thus, in addition to apoptosis, MK-886 can enhance TNF-alpha-induced differentiation.

Nitric oxide production by the hemocytes of the last instar larvae and sessile pharate prepupa of Galleria mellonella (Lepidoptera: Pyralidae) was demonstrated in vitro in response to preparations of bacterial lipopolysaccharide (LPS) from Escherichia coli using the Griess reaction. Augmented, dose dependent nitric oxide production was observed in the pharate prepupal hemocytes compared with larval hemocytes. This was partially reversed in a dose dependent manner with S-methyl thiourea (SMT), a specific inhibitor of inducible nitric oxide synthase (iNOS). A decrease in NO production was also observed when non-selective inhibitors such as N(G)-nitro-L-arginine (L-NAME) and N-omega-nitro-L-arginine (L-NNA) were used, albeit the inhibition was not to the extent of SMT. Challenge with the entomopathogenic Gram-negative bacterium Photorhabdus asymbiotica also enhanced NO production by hemocytes of both stages. SMT, alone or in combination with P. asymbiotica significantly decreased levels of NO production. However, it was observed that phenoloxidase activity (a cascade for innate immune responses) was independent of NO production stimulation. NO donors, S-nitroso-N-acetyl-penicillamine (SNAP) and diethylenetriamine NO adduct (DETA/NO) at various concentrations (100-500 microM) resulted in the lysis of hemocytes dose dependently. The nitrite production in these cases was however similar to LPS stimulation (10 microg/mL) and 1.5-3 fold lower than those observed upon P. asymbiotica (2.5 x 10(7) cfu/mL) stimulation. Survival analysis (Kaplan-Meier) following injection of P. asymbiotica alone or in combination with SMT revealed that only 12.5% (median survival 25.5 h) of co-injected larvae of G. mellonella survived in comparison to 28.6% (median survival 29 h) survivors in P. asymbiotica alone-injected groups till the end of the study. In contrast, co-injected pharate prepupa survived longer (median survival 28 h) than the P. asymbiotica alone-injected individuals (median survival 24 h); however, both co-injected and P. asymbiotica-injected groups showed 100% mortality at the end of the study. Based on the above, we propose that although NO production is involved in cellular immune responses of this insect to bacterial infection it does not appear to be a part of the signalling pathway that initiates the prophenoloxidase (PPO) cascade, and the extended NO production/overproduction by pharate prepupal hemocytes could result in cytotoxic rather than cytoprotective effects compared with larval hemocytes.

Male Wistar rats were offered food for either 24, 6, or 1 h a day at two different ambient temperatures -21 or 6 degrees C. At room temperature, rats offered food for 6 h a day matched their food intake to that of rats with 24 h of food-availability in 2-3 days, so that no main changes in total food intake, gross food efficiency, and body weight were recorded. No impairment of thermoregulatory capacity was recorded as judged by the unaltered oxidative capacity of brown adipose tissue and skeletal muscle. Nonetheless, a transient hypothermia was recorded at the beginning of the experiment in the food-restricted group. Rats that were only offered food 1 h a day were unable to sustain the food intake of the control animals, so that a continuous decline in body weight and a negative gross food efficiency ensured. In the cold, the food intake of rats offered food for only 6 or 1 h a day was similar to that showed by the corresponding groups at 21 degrees C. These results suggest a maximum rate for digestive processes, irrespective of ambient temperature. However, such levels of food intake were clearly insufficient to cope with the thermoregulatory enhanced energy demand at 6 degrees C, ad judged by the decrease in body weight.

The simultaneous effect of intermittent starvation and a high-fat diet were investigated in mice after several weeks of experimental feeding. The animals adapted to intermittent fasting fed a high-fat diet showed a lower degree of hyperphagia than animals adapted to intermittent fasting fed a standard laboratory diet. The weight of both individual portions of the stomach was elevated in adapted animals fed both a standard laboratory diet and the high-fat diet. The weight of the small intestine was increased in adapted animals fed a high-fat diet. The length of the small intestine was not changed after 8 weeks of intermittent starvation in both adapted groups (standard laboratory diet, high-fat diet). A higher amount of body fat was found in both groups of animals adapted to intermittent fasting (standard laboratory diet, high-fat diet) but adapted animals fed a high-fat diet showed less body fat than adapted animals fed a standard laboratory diet. Lower levels of serum lipids were found in adapted animals fed a high-fat diet. These results suggest that both lipogenesis and lipid oxidation are accentuated by intermittent starvation and a high-fat diet act concomitantly.

After 8 weeks of intermittent fasting, mice fed both a standard laboratory diet and a high-fat diet became hyperphagic and showed an increased amount of glycogen storage in the liver. An important effect of the adaptation to intermittent feeding with a high-fat diet seems to be an activation of the oxidation of lipids. Lipid oxidation prevails over lipogenesis so that the protein levels in the liver and skeletal muscle are preserved and maintained constant.

In golden hamster adapted to intermittent starvation (with access to food for 3 days a week only), the glycogen and lipid content of the liver, total body fat and RQ values measured in vivo were investigated. These parameters were studied in dependence on the duration of the adaptation period (6 weeks versus 20 weeks), season of the year (winter versus summer) and sex. The liver glycogen content in intermittently starved males and females increases after 6 weeks of adaptation during winter. In summer an increase was found after long-term adaptation, but only in males. Male hamsters also exhibited a higher ability to accumulate liver lipids during both seasons. In spite of their lower weight intermittently starving males have a higher percentage of body fat in comparison with females, a stimulating effect being observed in winter. Increased lipogenesis in golden hamsters adapted to intermittent starvation may be concluded from the increased RQ values when compared with relative data gained in controls fed daily ad libitum. Significant differences between RQ measurements according to season and sex were not found.

Fed animals have a higher resting metabolic rate in the thermoneutral zone than fasting ones. The metabolic increase is due to the specific dynamic action of food. With a decline of environmental temperature this increase in metabolism either declines or remains unchanged; decisive is whether the heat is used for thermoregulation or not (Mejsnar and Janský 1971). The objective of our work was to find out to what extent a single intake of a diet with a different ratio of nutrients can influence resting metabolism in the golden hamster and whether this heat can be used for thermoregulation in the cold. Female golden hamsters aged 6-8 weeks kept at a constant temperature of 22 +/-1 degrees C with twelve-hour alternation of light (6 a.m. - 6 p.m.) and darkness ( 6 p.m. - 6 a.m.) were used for the experiments. The oxygen consumption was assessed after a single intake of a standard, high-carbohydrate (76 cal.% starch), high-fat (80 cal.% margarine) and high-protein (82 cal.% casein) diet-for detailed composition see Fábry (1959). The food was given at 6.m. after previous 20 hours of fasting. Animals were then transferred into the respiration chamber and kept there for three hours, including one hour when they were left to settle down; during this period the oxygen consumption was not measured. Oxygen consumption measurement started at 9 a.m. and lasted till 11 a.m. The metabolism of the animals at rest was assessed at temperatures of 10, 20 and 30 degrees C by measuring the oxygen consumption by the interferometric method (Wollschitt et al. 1935). The results are expressed in ml of oxygen per g of body weight per hour. The relationship between the metabolism at rest and environmental temperatures in hamsters given a single dose of standard, high-proetin, high-fat or high-carbohydrate diet is apparent from Table 1. The maximum increase of oxygen consumption after administration of the experimental diets was found at a temperature of 30 degrees C. At an environmental temperature of 20 degrees C the administration of the high-protein and high-fat diet causes roughly the same increase of metabolism. The high-carbohydrate diet increase is only one third of thevalues found, in the remaining two diets at the same temperature and is non-significant. At the environmental temperature of 10 degrees C all the diets used increased the oxygen consumption insignificantly. The changes in metabolism at different environmental temperatures after administration of various diets expressed as percentage of metabolism at 30 degrees C in animals fed the standard diet indicate that the specific dynamic action of the high-protein and high-fats diets is lower at lowered temperatures. We may thus assume that the heat produced as a result of specific dynamic action of the high-protein and high-fat diets is perhaps used for thermoregulation. The role of specific dynamic action of high-carbohydrate diet for thermoregulation is not clear from our experiments. The role of specific dynamic action of food was assessed by several authors...

Golden hamsters (Mesocricetus auratus) were subjected to intermittent fasting for six weeks, for three of which their environmental temperature was reduced to 4 degrees C. They were compared with a control group (fed ad libitum and kept at 22 degrees C), a group subjected six weeks only to intermittent fasting at 22 degrees C and a group fed ad libitum for six weeks, but exposed to a temperature of 4 degrees C after the third week. The effect of these various nutritional and temperature regimens on growth, on food consumption, on the formation of liver glycogen and lipid reserves and on energy metabolism was studied. Adaptation to cold raised the hamsters' food intake above the hyperphagia level observed during intermittent fasting. Cold and intermittent fasting acted synergically in raising basal oxygen consumption values and reducting the amount of body lipids. They acted antagonistically on the liver glycogen concentration; when they were used simultaneously, the effect of intermittent fasting preponderated.

As a result of the specific dynamic action of a high-protein diet the resting metabolism of golden hamsters within the zone of thermoneutrality is increased on a average by 40%. The specific dynamic action diminishes markedly, with declining environmental temperature. It is concluded from this results that part of the heat produced by the specific dynamic action of a high-protein diet leads to a rise in the lower critical temperature in the zone of thermoneutrality by 2 degrees C.
