BACKGROUND: The dropout rate from the treatment of patients with substance use disorders (SUD) negatively affects overall treatment outcomes. Little is known about the specific role of various psychiatric and psychosocial factors in the individual patients. Limited data is available in regards to mindset and addiction treatment, giving the opportunity of testing such interventions in the future. AIM: To identify the key psychological, psychosocial, and other treatment related factors in the patients from the inpatient substance use treatment with addiction who have dropped out of the program based on correlations with the patients' mindset. SETTING: Men's ward for treatment of SUD at the Department of Addictology, General University Hospital in Prague, the Czech Republic. SAMPLE: 120 male patients aged between 18 and 70 years sampled by convenience sampling in the Department of Addictology at the General University Hospital in Prague, Czech Republic. METHODS: The prospective study with the follow-up will be based on correlation analysis of self-reports questionnaires which are completed on the first week and the last week of the treatment, followed up by a short phone interview 6 months after the beginning of the treatment for the patients that have completed the treatment. The following is a protocol for the study that began in 2020 and will end at the end of 2023. Data will be stored according to the guidelines and treatment confidentially. DISCUSSION: Findings will be disseminated in peer reviewed scientific journals, national and international conferences, and in briefings to inform clinical decision making. It will provide information which will help to personalize treatment to meet unique needs of each patient and serve as dropout prevention. [ABSTRACT FROM AUTHOR]

- Abstract
Introduction: The importance of biomarkers for pharmaceutical drug development and clinical diagnostics is more significant than ever in the current shift toward personalized medicine. Biomarkers have taken a central position either as companion markers to support drug development and patient selection, or as indicators aiming to detect the earliest perturbations indicative of disease, minimizing therapeutic intervention or even enabling disease reversal. Protein biomarkers are of particular interest given their central role in biochemical pathways. Hence, capabilities to analyze multiple protein biomarkers in one assay are highly interesting for biomedical research. Areas covered: We here review multiple methods that are suitable for robust, high throughput, standardized, and affordable analysis of protein biomarkers in a multiplex format. We describe innovative developments in immunoassays, the vanguard of methods in clinical laboratories, and mass spectrometry, increasingly implemented for protein biomarker analysis. Moreover, emerging techniques are discussed with potentially improved protein capture, separation, and detection that will further boost multiplex analyses. Expert commentary: The development of clinically applied multiplex protein biomarker assays is essential as multi-protein signatures provide more comprehensive information about biological systems than single biomarkers, leading to improved insights in mechanisms of disease, diagnostics, and the effect of personalized medicine.

- Abstract
Excessive and regular alcohol consumption affects negatively the whole organism and the organ that is damaged most often is liver. A whole range of diseases, from the lighter forms such as steatosis to a serious, life-shortening illness – the liver cirrhosis, can be caused by alcohol consumption. Therapeutic procedures for people with alcoholic liver damages include above all change of lifestyle, abstinence from alcohol consumption and nutritional and pharmacological therapy. Liver transplant surgery is required in serious cases. The main aim of this communication is to find out how the liver transplant influences quality of patients' everyday lives improves after liver transplant. We have divided this aim in several questions: do the patients' attitudes towards use of alcohol change in relation to the subsequent outpatient treatments? Is it possible to collect objective markers related to alcohol usage in patients after liver transplant? Are there any interconnections between the patients' subjective attitude towards alcohol, quality of their lives and objective evidence of their alcohol use? Data collection method. To this end a structured questionnaire was used based on standardised instruments WHOQOL-BREF and AUDIT. The research sample. Consisted of 96 patients after liver transplant, who were treated in the Institute for Clinical and Experimental Medicine in Prague. Data analysis method. The collected data was processed by the method of descriptive statistics and the results presented in individual tables and charts. Conclusion and results. The results indicate that patients after liver transplant perceive the quality of their lives relatively without regard to their age and sex. The overall number of patients evaluate their quality of life as average, no significant differences between quality of life in men and women have been found. Answers from the AUDIT questionnaire demonstrate that absolute majority of patients experience no problems in relation to alcohol use, as proved also by their laboratory values. The patients who underwent liver transplant can return to their regular pre-transplant lifestyle without serious complications. These people can even get back to their original professions, especially if they are middle-aged and their jobs do not require manually demanding activities. [ABSTRACT FROM AUTHOR]

INTRODUCTION: The foreign studies indicate that parental substance use makes it more likely that their children will engage in risk behaviour. Risk behaviour may be positively influenced by prevention programmes. AIM: The aim of this research project was to assess whether the Unplugged prevention programme has the desired effect on drinking among children at the age of approximately 15 years. The study addresses any differences in the effects on the children that depend on whether their mothers drink more than weekly. We regarded drinking as a sign of risk behaviour engaged in by children from families where the mothers use alcohol more or less than weekly. METHODS: The data of the children who were exposed to the prevention programme was compared to that of the children who did not participate in the programme. RESULTS: The Unplugged prevention programme was found to have no statistically measurable effect on drinking among children who came from families where the mother uses alcohol more than weekly. Children from the families where the mother reported using alcohol weekly or less frequently than that and who had been exposed to the Unplugged prevention programme were found to be less likely (by 63%) to report drunkenness in the last 30 days. - Abstract
Musculus piriformis is an important muscle for human biomechanics. It is the most powerful and resistant external rotator of the hip joint as well as a strong abductor. In many locomotive activities including sports it is the subject to reflex long term hyperactivation, and we therefore encounter excessive tonus (hypertonus) and often hypertrophy and shortened muscle. In view of the intimate contact with the narvus ischiadicus the nervous filaments are irritated. It is either due to the pressure of the hypertrophic muscle or the situation when the nerve passes at least partly through the muscle and a traction neuropathy develops. It results to ischialgia which belongs to the low back pain group. Several clinical tests used in practice include the FAIR text (flexion, adduction, internal rotation), which stretches the muscle and in case of increased or provoked ischialgia it is evaluated as positive. In our study we paid attention to the correlation of the FAIR results with MRI finding. Our cohort included 25 patients with chronic ischialgia, 11 men and 14 women with normal clinical neurological findings. We excluded root lesion S1, discopathy L5/S1, rheumatological or infectious disease and a pathology of the hip joint. After a clinical examination the patients were examined on the MRI projection to the area of m. piriformis. In 11 out of the 25 patients examined by NRI we found compression of n. ischiadicus (ischiadic nerve) in foramen infrapiriforme, compression the nerve in passing the muscle in four, no sign of irritation of the nerve in other four patients and other kinds of pathology in six. The results indicated high sensitivity of the GFSAIR test in diagnostics of irritation of n. ischiadicus in the region of foramen infrapiriforme or in passage through the muscle. At the same time the results revealed that without adequate imaging it is impossible to differentiate functional irritation of ischiadic nerve from an irritation by another pathological process. Therefore, from the point of view of correct diagnostics until the recommendation to make it a part of precise diagnostics. [ABSTRACT FROM AUTHOR]

- Abstract
The European Cooperation in Science and Technology (COST) provides an ideal framework to establish multi-disciplinary research networks. COST Action BM1203 (EU-ROS) represents a consortium of researchers from different disciplines who are dedicated to providing new insights and tools for better understanding redox biology and medicine and, in the long run, to finding new therapeutic strategies to target dysregulated redox processes in various diseases. This report highlights the major achievements of EU-ROS as well as research updates and new perspectives arising from its members. The EU-ROS consortium comprised more than 140 active members who worked together for four years on the topics briefly described below. The formation of reactive oxygen and nitrogen species (RONS) is an established hallmark of our aerobic environment and metabolism but RONS also act as messengers via redox regulation of essential cellular processes. The fact that many diseases have been found to be associated with oxidative stress established the theory of oxidative stress as a trigger of diseases that can be corrected by antioxidant therapy. However, while experimental studies support this thesis, clinical studies still generate controversial results, due to complex pathophysiology of oxidative stress in humans. For future improvement of antioxidant therapy and better understanding of redox-associated disease progression detailed knowledge on the sources and targets of RONS formation and discrimination of their detrimental or beneficial roles is required. In order to advance this important area of biology and medicine, highly synergistic approaches combining a variety of diverse and contrasting disciplines are needed.

Here we describe the electrochemical behavior of a group of polyaromatics designated helicenes. These compounds are finding many applications in optoelectronics, catalysis and chiral separation. Thus it is important to investigate the electrochemical and optical behavior of helicenes with respect to improving their application potential and our knowledge of strategies for their surfaces immobilization. Electrochemical analyses were performed in acetonitrile and in aqueous media with glassy carbon and indium tin oxide electrodes. Specifically, [n]helicenes and their salts 1-buty1-3-(2-methy1[6]heliceny)-immidazolium bromide and 3-([7]helicene-9-y1)thiophene were investigated, the application of acquired electrochemical data is disussed, especially in terms of optoelectronics and the construction of semi-conductive devices.

Ferroelec. materials have promising applications in frequency control and filtering (e.g. Miniaturization in cellular phone technol.). One approach to such materials is to use a textured insulator surface with a trigonal array of channels. When mols. with rotatable elec. dipoles are partially inserted into such a matrix, with the rotors protruding outside, an array of elec. dipoles on a surface of the crystal is formed. The preferred alignment of the rotors in the array is detd. by the location of the channels in the porous material. Trigonal arrangment may lead to a ferroelec. ground states. We describe our progress in the design, fabrication, and characterization of such surface inclusion compds. using mainly solid-state NMR techniques and dielec. spectroscopy

In this contribution, we report that electrooxidation of PCD, phenolic cyclopentenedione, 2,2'-bis[4,5-bis(4-hydroxybenzyl)-2-(4-hydroxyphenyl)-cyclopent-4-ene-1,3-dione] results in the formation of stable films with permselective properties on metal and carbon surfaces. The electropolymerization process proceeds in neutral and alkaline aqueous medium at potentials higher than +0.6 V (vs. Ag/AgCl/3M KCl). The PCD polymer prepared was characterized by electrochemical methods, quartz crystal microbalance and optical spectroscopy techniques. The PCD film can be used for electrode coating and preparation of an anti-interference barrier. Thus, we performed SAR study which could open the door to developing novel functional films and electrochemical tools for bioactive compounds sensing.\n

Research on the employability of university graduates provides key feedback for universities, applicants, students, and decision makers. The topic is even more important at a time when the first international curricula (UPC/UTC) and professional platforms are being developed. The profession of an addiction specialist in the Czech Republic is a young one and it is still seeking to establish its position within the system of services for people with substance use disorders. Therefore, there is only limited data on addictologists' success in finding a job. AIMS: To identify the rate and characteristics of the employability of addictologists on the labour market, whether addictology graduates work in the field and under what conditions, and, if not, what prevents them from doing so. METHODS: The study population comprised the graduates of the academic programmes in addictology from 2005 to 2016. Data was collected using an online questionnaire survey, with a 59% response rate, and analysed using MS Excel and IBM SPSS 22. RESULTS: 58% of the respondents work in the field of addictology, mostly in low-threshold and outpatient services. 62% of them had already worked during their studies. 53% work in Prague, two-thirds of them on a full-time basis. One-third has another employment too. One half keeps the position of addictologists, more men than women. With statistical significance, men are more likely to work in the field. 69 persons do not work in the field of addictology, with 39 never having looked for such a job. The reasons for working in other fields included low salaries, the offer of a different job, and a lack of work opportunities. Those working in addictology tended to rate both the quality of the study programme and the demand for the profession in more positive terms and they are also significantly more likely to be members of the Czech Association of Addictologists. CONCLUSION: The results are of great value and will be used for adjusting the study programmes to meet the requirements of practice and for the internationalization process involving the implementation of the UPC and UTC curricula.
