An active and constructive process whereby individuals possess the ability to develop, implement, and flexibly maintain planned behavior in order to achieve a desired achievement goal is referred to as self-regulation. The aim of the present study is to examine the factorial structure and psychometric properties of the Self-Regulation Questionnaire, validated in the Czech educational context (SRQ-CZ). The other target is to identify any usage of self-regulation strategies which may differ as nationality, gender, age, education, and internal motivation to learn come into play. A total of 1,711 adult learners from Poland, Serbia, Slovakia, and the Czech Republic who were enrolled in a formal secondary or higher education system pursuing professions in the field of humanities, social and health care sciences participated in this study. A measurement-invariant four-factor model was obtained across all countries (min. CFI, TLI = 0.918, and 0.902, max. RMSEA = 0.059; ω between 0.625 and 0.838, and Cronbach's α between 0.622 and 0.837), including the Impulse Control, Goal Orientation, Self-Direction, and Decision Making subscales. Generally, the study confirmed a gradual increase in scores that measure positive self-regulatory qualities (Goal Orientation and Decision Making) and a decrease in unfavorable self-regulatory qualities (Impulse Control and Self-Direction) with higher age, education, and motivation, with no gender differences reported within countries. Moreover, scores on Goal Orientation and Decision Making dominated between countries. DNA-protein crosslinks (DPCs) are highly toxic DNA lesions represented by proteins covalently bound to the DNA. Persisting DPCs interfere with fundamental genetic processes such as DNA replication and transcription. Cytidine analog zebularine (ZEB) has been shown to crosslink DNA METHYLTRANSFERASE1 (MET1). Recently, we uncovered a critical role of the SMC5/6-mediated SUMOylation in the repair of DPCs. In an ongoing genetic screen, we identified two additional candidates, HYPERSENSITIVE TO ZEBULARINE 2 and 3, that were mapped to REGULATOR OF TELOMERE ELONGATION 1 (RTEL1) and polymerase TEBICHI (TEB), respectively. By monitoring the growth of hze2 and hze3 plants in response to zebularine, we show the importance of homologous recombination (HR) factor RTEL1 and microhomology-mediated end-joining (MMEJ) polymerase TEB in the repair of MET1-DPCs. Moreover, genetic interaction and sensitivity assays showed the interdependency of SMC5/6 complex, HR, and MMEJ in the homology-directed repair of MET1-DPCs in Arabidopsis. Structural maintenance of chromosome (SMC) complexes play roles in cohesion, condensation, replication, transcription, and DNA repair. Their cores are composed of SMC proteins with a unique structure consisting of an ATPase head, long arm, and hinge. SMC complexes form long rod-like structures, which can change to ring-like and elbow-bent conformations upon binding ATP, DNA, and other regulatory factors. These SMC dynamic conformational changes are involved in their loading, translocation, and DNA loop extrusion. Here, we examined the binding and role of the PpNSE5 regulatory factor of Physcomitrium patens PpSMC5/6 complex. We found that the PpNSE5 C-terminal half (aa230-505) is required for binding to its PpNSE6 partner, while the N-terminal half (aa1-230) binds PpSMC subunits. Specifically, the first 71 amino acids of PpNSE5 were required for binding to PpSMC6. Interestingly, the PpNSE5 binding required the PpSMC6 head-proximal joint region and PpSMC5 hinge-proximal arm, suggesting a long distance between binding sites on PpSMC5 and PpSMC6 arms. Therefore, we hypothesize that PpNSE5 either links two antiparallel SMC5/6 complexes or binds one SMC5/6 in elbow-bent conformation, the later model being consistent with the role of NSE5/NSE6 dimer as SMC5/6 loading factor to DNA lesions. In addition, we generated the P. patens Ppnse5KO1 mutant line with an N-terminally truncated version of PpNSE5, which exhibited DNA repair defects while keeping a normal number of rDNA repeats. The unpredictable spread of the COVID-19 pandemic, national lockdowns, and public health measures implemented in the Czech Republic had a negative effect on older adults' mental health and loneliness. The 2,631 older adults in 2020 and 2,083 older adults in 2021 used in this study formed a nationally representative sample of the Survey of Health, Ageing and Retirement in Europe (SHARE). Almost every third older adult experienced feelings of loneliness in both stages of the COVID-19 outbreak. Loneliness increased in 2021 in those individuals who reported their physical health as poor, who felt nervous, sad or depressed, and who had ever left their home since the outbreak. According to age-related drivers of loneliness, feelings of loneliness were prevalent (40% vs. 45%) among younger retirees in both waves. In both data sets, declared feelings of sadness or depression were the strongest sustainable predictor of loneliness (2020 and 2021 models, OR  = 3.69; 95% CI [2.90, 4.69] and OR  = 2.55; [1.97, 3.30]). Being a woman and feeling nervous equated with a higher likelihood of feeling lonely compared to counterparts. Background: One of the most significant human qualities is the ability to develop, implement, and flexibly maintain planned behaviour in order to achieve one's goals. Self-regulation has become a relatively well-researched area in the field of psychology and pedagogy. However, empirically valid and reliable instrument is still missing across European context. The primary goal of this research was to analyze the psychometric properties of the Czech version of the Self-Regulation Questionnaire (SRQ-CZ) among adult learners from Poland, Serbia, Slovakia, and the Czech Republic., Objective: The aim of the present study was to examine the factor structure and psychometric properties of the SRQ-CZ validated in the Czech educational context in a multi-cultural sample., Methods: A total of 1711 adult learners from European countries including Poland, Serbia, Slovakia, and the Czech Republic completed the SRQ-CZ. The first step to reviewing the validity of the SRQ-CZ included testing face validity. Furthermore, exploratory factor analysis (EFA) was performed on half the sample and confirmatory factor analysis (CFA) on the other half. Measurement invariance was conducted across gender, age, and country followed by the evaluation of the reliability of the final instrument., Results: EFA showed that a three-factor structure best fit the data. The originally proposed Impulse Control and Self-Direction are merged into one distinct factor, while Decision Making and Goal Orientation comprise the other two. Goodness-of-fit statistics yielded from CFA showed a good fit for the model, introducing a reliable and measurement invariant instrument., Conclusion: The present study used a diverse multi-cultural sample to explore the factorial structure and psychometric properties of the SRQ-CZ. A three-factor model was obtained as the result of the exploratory and confirmatory factor analyses. Further analysis aiming at measurement invariance, comparing the sample according to gender, age, and country, led to satisfactory results. The only exception was a lack of model fit in the case of Serbia. Although further psychometric evaluation of the SRQ-CZ is still needed, the presented results constitute significant findings, confirming instrument validity and utility as a measure of generalized self-regulation capacity across adult learners in European educational context., (© 2022. Combining theory-driven and data-driven approaches, this study used both self-reported and observational measures to examine: (1) the joint contributions of students' self-reported undergraduates' motivation and emotion in their self-regulated learning, their observed online learning interactions, and their academic success in blended course designs; and (2) the extent to which the self-reported and observational measures were consistent with each other. The participants in the study were 54 social sciences undergraduates in the Czech Republic. The participants' self-reported self-efficacy, intrinsic goals, and anxiety were assessed using a Czech version of three scales from the Motivated Strategies for Learning Questionnaire. Their online engagement was represented by students' observed frequency of interactions with the six online learning activities recorded in the learning management system. The results of a hierarchical regression analysis showed that the self-reported and observational measures together could explain 71% of variance in academic success, significantly improving explanatory power over using self-reported measures alone. Departing from the theory-driven approach, students were clustered as better and poorer self-regulated learners by their self-reports, and one-way ANOVAs showed that better self-regulated learners had significantly more frequent online interactions with four out of six online learning activities and better final exam results. Departing from the data-driven approach, students were clustered as higher and lower online-engaged learners by the observed frequency of their interaction with online learning activities. One-way ANOVAs showed that higher online-engaged learners also reported having higher self-efficacy and lower anxiety. Furthermore, the strong association between the students' profiles in both self-reported measures and observational measures in cross-tabulation analyses showed that the majority of better self-regulated learners by self-reporting also had higher online engagement by observation, whereas the majority of poorer self-regulated learners by self-reporting were lower online-engaged learners, demonstrating consistency between theory-driven and data-driven approaches., Competing Interests: The authors declare that the research was conducted in the absence of any commercial or financial relationships that could be construed as a potential conflict of interest., (Copyright © 2022 Han, Vaculíková and Juklová.)

This study deals with key factors affecting mental health during the COVID-19 pandemic. The results are based on the longitudinal representative 50+ population-based multi-country panel study Survey of Health, Aging and Retirement in Europe (SHARE) in which representative samples of individuals in European countries plus Israel were collected. The goal is to turn the challenges of aging into opportunities as well as provide valid and reliable data for evidence-based policies. A total of 2,631 participants were interviewed by telephone in the Czech Republic during the initial stages of the COVID-19 outbreak in 2020. After a descriptive analysis showed that participants complied well with most preventative and protective COVID-19 measures, subsequent modeling using logistic regression analysis and a decision tree algorithm identified key determinants that have contributed to an understanding of variation in declared feelings of depression. We found that nervousness, previously unexperienced trouble with sleep, and self-rated health status before the COVID-19 outbreak and gender remained significant predictors of depression, from which statistically significant different categories concerning the percentage of depression could be formed. These identified potential risk factors should be considered in planning further supportive strategies for high-risk 50+ population.
