No. of bitstreams: 0 Previous issue date: 2020-10-01 Introduction: This study aimed to evaluate the in vitro effectiveness of N-acetylcysteine (NAC), photodynamic therapy (PDT) and NAC with supplemental PDT in optimizing the removal of bacteria from infected dentinal tubules of root canals infected with Enterococcus (E.) faecalis biofilm. Methods and Materials: Eighty human teeth were randomly divided into 5 groups (n=16) according to the intracanal medication used: saline solution (control); calcium hydroxide (CH); NAC; PDT; NAC+PDT. Ten samples from each group were prepared for microbiological culture analysis (CFU/mL) and were inoculated with E.faecalis suspension for 21 days for biofilm development; the other six samples from each group were prepared for scanning electron microscopy (SEM) and confocal laser scanning microscopy (CLSM) and submitted to a 5-days contamination protocol including eight centrifugation cycles on every other day for dentinal tubules infection. For antimicrobial activity analysis by microbiological culture (CFU/mL), the root canals were contaminated with E. faecalis biofilm, instrumented and then medicated according to the experimental groups. Three samples were collected from the root canals: after 21-days of contamination, immediately after the instrumentation and 14-days after the medication according to the experimental groups. The morphology of E. faecalis biofilm on the root canal walls and bacterial cells viability were assessed by means of SEM and CLSM, respectively. One-way ANOVA and Repeated Measures ANOVA tests were used to analyze the obtained data statistically. Results: CFU/mL analysis showed that CH, NAC and NAC+PDT promoted greater antibacterial activity with statistically significant difference compared to saline solution and PDT (P > 0.0001). However, saline solution and PDT were statistically similar (P > 0.07). Illustrative images by SEM confirmed partially the CFU/mL results. CLSM showed that all groups were effective eliminating E. faecalis except for the saline solution group. Conclusions: Based on this in vitro study NAC was bactericidal against E. faecalis biofilms regardless PDT stimulation, presenting similar antimicrobial activity to CH. This study evaluated the cleanliness of post space after different irrigation protocols, associated or not to the use of ultrasound. Thirty-six single-rooted bovine roots were instrumented with rotary system, irrigated with 1 % hypochlorite and filled with Ah Plus sealer and gutta-percha cones. After seven days in stove at 37 °C, two-thirds of canal filling was removed by drilling and the post space preparation was irrigated with 2.5 % NaOCl (group NaOCl); 0.2 % chlorhexidine (group CLX); distilled water (group Water); 2.5 % NaOCl + ultrasound (group NaOCl-US); 0.2 % chlorhexidine + US (Group CLX-US); or water + US (Group Water-US). Roots were cleaved in mesiodistal direction, prepared for scanning electron microscope (SEM) to evaluate the smear layer and debris presence and for energy dispersive spectroscopy (EDS) analyses, to evaluate chemical components of dentin after treatment. Objetivo: Além da agressão química devido à penetração dos géis clareadores na polpa durante o clareamento de dentes com vitalidade pulpar, outro possível fator agressor pode ser o calor gerado pela reação de oxidação exotérmica do gel clareador, que pode também ser agravada pela fotoativação. Este estudo avaliou a variação da temperatura na câmara pulpar de dentes humanos, utilizando-se três diferentes géis clareadores, com ou sem fotoativação. Metodologia: Trinta pré-molares humanos foram cortados longitudinalmente para obtenção de duas metades: vestibular e lingual. Os 60 espécimes foram divididos em 3 grupos e os agentes clareadores utilizados variaram como segue: peróxido de hidrogênio 35% (WHP), peróxido de carbamida 37% (W) e peróxido de hidrogênio 38% (OX). Metade dos espécimes foi submetida ao clareamento com fotoativação e, a outra metade, sem fotoativação. A fonte de luz utilizada foi o aparelho à base de diodo emissor de luz (LED, 3-Light, Clean Line) e as temperaturas foram medidas por um termômetro digital. Os dados foram analisados por análise de variância e teste de Tukey (alfa=5%). Objetivo: O objetivo deste trabalho foi avaliar o uso do ultra-som na remoção de retentores intra-radiculares metálicos fundidos (RI). Metodologia: Foram utilizados 30 dentes humanos unirradiculados tratados endodonticamente e divididos em três grupos: grupo G1 - retentores intra-radiculares do tipo convencional, sem nenhum tipo de retenção mecânica adicional (tipo liso); grupo G2 - retentores intra-radiculares com sulcos longitudinais (tipo canaleta) e grupo G3 - retentores intra-radiculares com sulcos transversais (tipo anelado). Os retentores foram cimentados nos canais radiculares com cimento fosfato de zinco e os espécimes armazenados por 24 h em temperatura ambiente. Para a remoção dos retentores utilizou-se o ultra-som e a tração manual com pinça hemostática. Foi medido o tempo necessário para remoção dos retentores; os dados foram analisados por ANOVA e teste de Tukey (5% de significância). O objetivo deste trabalho foi realizar um levantamento sobre as causas de traumatismos maxilo-faciais e dentais em pacientes atendidos no ambulatório do setor de Traumatologia Buco-Maxilo-Facial do Pronto Socorro Municipal de uma cidade do Estado de São Paulo com 600. 000 habitantes no período de janeiro de 1998 a abril de 2002. Foram selecionados 756 casos e os dados referentes ao sexo, idade, data de atendimento, causa do trauma, áreas envolvidas, envolvimento dentário e tipo de trauma dental foram anotados em uma ficha. Os resultados mostraram que: 80,29% dos traumatismos ocorreram no sexo masculino; a faixa etária mais envolvida foi 25-29 anos (51,87%); a causa mais freqüente foi acidente automobilístico/bicicleta (25,33%); a região dos ossos nasais foi a mais acometida representando 32% dos casos; os dentes permanentes anteriores os mais afetados (69,39%) e o tipo de trauma dental mais freqüente foi a avulsão (35,37%). Verificou-se pelo teste qui-quadrado de aderência a uma distribuição uniforme que houve diferença estatística em todos os 6 itens analisados (p < 0,05). Concluiu-se que a ocorrência de traumatismo maxilo-facial foi mais freqüente em adultos do sexo masculino, relacionados a acidente automobilístico/bicicleta, sendo a região dos ossos nasais a área mais acometida, os dentes permanentes anteriores os mais envolvidos e a avulsão, o trauma dental mais freqüente

No. of bitstreams: 0 Previous issue date: 2015-01-01 In this study, the push-out method was used to evaluate the bond strengths of 3 types of endodontic cements according to their composite base: methacrylate, epoxy resin, and an experimental copaiba oil resin. The study hypothesis was that the methacrylate-based and experimental cements would have bond strengths equal to or greater than that of the epoxy resin-based cement. Thirty bovine tooth roots, 18 mm long, were divided into 3 groups (n = 10) based on the chosen cement treatment. After treatment, the specimens were sectioned and submitted to a push-out test. Results showed that there was no statistically significant difference (P < 0.05) between the cements used or between the middle and apical thirds of the roots. It could be concluded that the tested cements had satisfactory and similar bond strengths to dentin. No. of bitstreams: 0 Previous issue date: 2015-01-01 This clinical study evaluated the use of 0.11% topical sodium fluoride (SF) desensitizing agent to treat tooth sensitivity during a nightguard tooth whitening procedure. Thirty-two subjects bleached their teeth with 10% carbamide peroxide (CP) gel using an at-home bleaching technique with custom trays. During bleaching treatment, subjects were divided into 2 groups (n = 16). The subjects in Group 1 received a topical gel containing 0.11% SF; the subjects in Group 2 received a placebo gel (PG). Each subject was instructed to place the gel in his/her bleaching tray for 30 min every day following bleaching treatment. Results showed the use of SF did not affect the whitening efficacy of the 10% CP gel. Subjects who received the PG had significantly higher tooth sensitivity when compared with subjects who received SF (P < 0.00). The use of daily 0.11% SF after 10% CP bleaching gel reduced tooth sensitivity during the bleaching treatment. No. of bitstreams: 0 Previous issue date: 2014-01-01 This in vitro study sought to evaluate the biomechanical preparation action on microorganisms and endotoxins by using sodium hypochlorite (NaOCl) and an intracanal medication containing Zingiber officinale, with or without calcium hydroxide. Single-rooted teeth were contaminated, and root canal instrumentation (using 2.5% NaOCl) was performed. Samples were divided into 4 groups, according to the intracanal medication employed. The root canal content was gathered 28 days after contamination (baseline), immediately after biomechanical preparation, 7 days after biomechanical preparation, 14 days after intracanal medication, and 7 days after intracanal medication was removed. The results (submitted to Kruskal-Wallis and Dunn tests) showed that the NaOCl eliminated 100% of root canal microorganisms and reduced 88.8% of endotoxins immediately after biomechanical preparation, and 83.2% at 7 days after biomechanical preparation.
