Introduction: Physical activity (PA) is associated with higher quality of life and probably better prognosis among colorectal cancer (CRC) patients. This study focuses on determinants of PA among CRC patients from diagnosis until 5 yr postdiagnosis., Methods: Sociodemographic and disease-related factors of participants of two large CRC cohort studies were combined. Moderate-to-vigorous PA during sport and leisure time (MVPA-SL) was measured at diagnosis (T0) and 6, 12, 24, and 60 months (T6 to T60) postdiagnosis, using the SQUASH questionnaire. Mixed-effects models were performed to identify sociodemographic and disease-related determinants of MVPA-SL, separately for stage I-III colon (CC), stage I-III rectal cancer (RC), and stage IV CRC (T0 and T6 only). Associations were defined as consistently present when significant at ≥4 timepoints for the stage I-III subsets. MVPA-SL levels were compared with an age- and sex-matched sample of the general Dutch population., Results: In total, 2905 CC, 1459 RC and 436 stage IV CRC patients were included. Patients with higher fatigue scores, and women compared with men had consistently lower MVPA-SL levels over time, regardless of tumor type and stage. At T6, having a stoma was significantly associated with lower MVPA-SL among stage I-III RC patients. Systemic therapy and radiotherapy were not significantly associated with MVPA-SL changes at T6. Compared with the general population, MVPA-SL levels of CRC patients were lower at all timepoints, most notably at T6., Conclusions: Female sex and higher fatigue scores were consistent determinants of lower MVPA-SL levels among all CRC patients, and MVPA-SL levels were lowest at 6 months postdiagnosis. The natural antimicrobial peptide Polybia-MP1 is a promising candidate for developing new treatment therapy for infection and cancer. It showed broad-spectrum antimicrobial and anticancer activity with high safety on healthy cells. However, previous sequence modification usually resulted in at least one of two consequences: a notable increase in hemolytic activity or a considerable decrease in activity against Gram-negative bacteria and cancer cells. Herein, a new approach was applied by replacing the amino acid Glutamine at position 12 with Lysine and generating the MP1-Q12K analog. Our preliminary data suggested an enhancement in antibacterial and antifungal activity, whereas the anticancer and hemolytic activity of the two peptides were comparable. Moreover, MP1-Q12K was found to be less self-assembly than Polybia-MP1, which further supports the enhancement of antimicrobial properties. Hence, this study provides new information regarding the structure-activity relationships of Polybia-MP1 and support for the development of potent, selective antimicrobial peptides., (© 2023. The associations of glucose, fructose, and sucrose intake with type 2 diabetes mellitus (T2DM) have been inconsistent. Furthermore, there is a lack of studies focusing on early markers of T2DM that provide insight into the process of T2DM progression: impaired pancreatic β-cell function (BCF) and insulin sensitivity. This study evaluated associations cross-sectionally in a population-based cohort consisting of 2818 individuals (mean ± SD age 59.7 ± 8.18, 49.5% male, n = 120 newly diagnosed T2DM). Glucose, fructose, and sucrose intake were assessed by a food frequency questionnaire. Glucose metabolism status, insulin sensitivity, and BCF were measured by a seven-points oral glucose tolerance test. Linear regression analysis revealed a positive association of glucose intake with insulin sensitivity in the fully adjusted model (standardized beta (95% CI) 0.07 (0.05, 0.14) SD for ≥23 g vs. <10 g of glucose). Fructose and sucrose intake were not associated with insulin sensitivity after full adjustments. In addition, no associations of dietary glucose, fructose, and sucrose with BCF were detected. In conclusion, higher intake of glucose, not fructose and sucrose, was associated with higher insulin sensitivity, independent of dietary fibre. No convincing evidence was found for associations of dietary glucose, fructose, and sucrose with BCF in this middle-aged population.

Salicylic acid (SA) and jasmonic acid (JA) cross-communicate in the plant immune signaling network to finely regulate induced defenses. In Arabidopsis, SA antagonizes many JA-responsive genes, partly by targeting the ETHYLENE RESPONSE FACTOR (ERF)-type transcriptional activator ORA59. Members of the ERF transcription factor family typically bind to GCC-box motifs in the promoters of JA- and ethylene-responsive genes, thereby positively or negatively regulating their expression. The GCC-box motif is sufficient for SA-mediated suppression of JA-responsive gene expression. Here, we investigated whether SA-induced ERF-type transcriptional repressors, which may compete with JA-induced ERF-type activators for binding at the GCC-box, play a role in SA/JA antagonism. We selected ERFs that are transcriptionally induced by SA and/or possess an EAR transcriptional repressor motif. Several of the 16 ERFs tested suppressed JA-dependent gene expression, as revealed by enhanced JA-induced PDF1.2 or VSP2 expression levels in the corresponding erf mutants, while others were involved in activation of these genes. However, SA could antagonize JA-induced PDF1.2 or VSP2 in all erf mutants, suggesting that the tested ERF transcriptional repressors are not required for SA/JA cross-talk. Moreover, a mutant in the co-repressor TOPLESS, that showed reduction in repression of JA signaling, still displayed SA-mediated antagonism of PDF1.2 and VSP2. Collectively, these results suggest that SA-regulated ERF transcriptional repressors are not essential for antagonism of JA-responsive gene expression by SA. We further show that de novo SA-induced protein synthesis is required for suppression of JA-induced PDF1.2, pointing to SA-stimulated production of an as yet unknown protein that suppresses JA-induced transcription., (© The Author 2016. Camellia japonica L. is an evergreen shrub whose cultivars are of great ornamental value. In autumn, after flower bud differentiation, dormancy is initiated. As in many other spring flowering woody ornamentals, winter low temperatures promote dormancy release of both flower and vegetative buds. However, warm spells during late autumn and winter can lead to unfulfilled chilling requirements leading to erratic and delayed flowering. We hypothesized that storing plants at no light and low temperature could favor dormancy breaking and lead to early and synchronized flowering in response to forcing conditions in C. japonica 'Nuccio's Pearl'. Plants with fully developed floral primordia were stored at dark, 7°C, and RH > 90% for up to 8 weeks. To monitor endodormancy release during the storage, we evaluated the content of abscisic acid (ABA) in flower buds and the expression profiles of five putative genes related to dormancy and cold acclimation metabolism in leaves and flower buds. In addition, the expression of four anthocyanin biosynthesis pathway genes was profiled in flower buds to assess the effect of the treatment on flower pigment biosynthesis. At 0, 4, 6, and 8 weeks of cold treatment, 10 plants were transferred to the greenhouse and forced to flower. Forced plant flower qualities and growth were observed. The ABA content and the expression profiles of two dormancy-related genes (CjARP and CjDEH) suggested that dormancy breaking occurred after 6-8 weeks of cold treatment. Overall, plants treated for 6-8 weeks showed earlier vegetative sprouting, enhanced, and homogeneous flowering with reduced forcing time. Prolonged cold treatments also reduced flower size and longevity, anthocyanin content, and pigment biosynthesis-related gene transcripts. In conclusion, the cold treatment had a promotive effect on dormancy breaking but caused severe drawbacks on flower quality.

Volatile fatty acids (VFA) are proposed platform molecules for the production of basic chemicals and polymers from organic waste streams. In this study we developed a granular sludge process to produce VFA at high rate, yield and purity while minimizing potential operational costs. A lab-scale anaerobic sequencing batch reactor (ASBR) was fed with 10 g l(-1) glucose as model substrate. Inclusion of a short (2 min) settling phase before effluent discharge enabled effective granulation and very high volumetric conversion rates of 150-300 gCOD l(-1)  d(-1) were observed during glucose conversion. The product spectrum remained similar at the tested pH range with acetate and butyrate as the main products, and a total VFA yield of 60-70% on chemical oxygen demand (COD) basis. The requirement for base addition for pH regulation could be reduced from 1.1 to 0.6 mol OH(-) (mol glucose)(-1) by lowering the pH from 5.5 to 4.5. Solids concentrations in the effluent were 0.6 ± 0.3 g l(-1) but could be reduced to 0.02 ± 0.01 g l(-1) by introduction of an additional settling period of 5 min. The efficient production of VFA at low pH with a virtually solid-free effluent increases the economic feasibility of waste-based chemicals and polymer production. Avian cell lines derived from germinal crescent primordial germ cells and gonadal gonocytes with long-term proliferative capacity in vitro and their subsequent rates of colonization and germline transmission are described. In general, male cultures proliferate more rapidly than female cultures although both can be developed into cell lines of >2 × 10(6) cells, at which time, they can be grown indefinitely and a cell bank can be established. All the cell lines injected into embryos transmitted through the germline with the percentage of germline transmission of both male and female cell lines varying from single digits to the high 90s. The derivation of these primordial germ cell and gonadal cell lines and the subsequent robustness of germline transmission validates these cells as suitable for establishment of lines of chickens bearing novel genetic modifications.

We report a case of a 26 year old woman with rash, lymphadenopathy, liver enzyme abnormalities and spiking fever. She was diagnosed with drug-induced hypersensitivity syndrome (DHS) to lamotrigine. Spiking fever in relation to drug-induced hypersensitivity syndrome has not earlier been described in adults. Spiking fever is an important symptom of the wide spectrum of disease presentation. The syndrome is commonly referred to as either Drug Rash with Eosinophilia and Systemic Symptoms (DRESS) or DHS. In accord with previous authors we see both syndromes as two ends of a spectrum, with a wide range of symptoms and presentations. Therefore we plea for unity in nomenclature.

Some Mallotus species are commonly used as traditional medicine (TM) ingredients in Vietnam and China, but only a few are studied for their activities. In Part I, high-performance liquid chromatography (HPLC) fingerprints of 39 Mallotus samples (17 species) were developed and, because of the complexity of and the large differences between the samples, it was chosen to analyse the unaligned fingerprints. The peaks, potentially responsible for the antioxidant activity in given Mallotus species, were indicated by the regression coefficients from an orthogonal projections to latent structures (O-PLS) model. In the present study, an in depth discussion on the need for alignment of the Mallotus fingerprints for the indication of the potentially active compounds is made, as well as an experimental analysis and identification of the previously indicated peaks by HPLC-mass spectrometry (HPLC-MS). Additionally, to thoroughly study and discuss the alignment problem, the modelling and prediction of the antioxidant activity of green tea samples based on HPLC fingerprints were also considered., (Copyright © 2012 Elsevier B.V. In fixed mouse brain magnetic resonance images, a high prevalence of fixation artifacts have been observed. Of more than 1700 images of fixed brains acquired at our laboratory, fixation artifacts were present in approximately 30%. In this study, two of these artifacts are described and their causes are identified. A hyperintense rim around the brain is observed when using perfusates reconstituted from powder and delivered at a high flow rate. It is proposed that these perfusion conditions cause blockage of the capillary beds and an increase in pressure that ruptures the vessels, resulting in a blister of liquid below the dura mater. Secondly, gray-white matter contrast inversion is observed when too short a fixation time or too low a concentration of fixative is used, resulting in inadequate fixation. The deleterious consequences of these artifacts for quantitative data analysis are discussed, and precautions for their prevention are provided., (Copyright © 2012 Elsevier Inc. The growth of iron-oxidizing bacteria, generally regarded as obligate microaerophilic at neutral pH conditions, has been reported in a wide range of environments, including engineered systems for drinking water production. This research focused on intensively aerated trickling filters treating deep anaerobic and subsurface aerated groundwater. The two systems, each comprising groundwater abstraction and trickling filtration, were monitored over a period of 9 months. Gallionella spp. were quantified by qPCR with specifically designed 16S rRNA primers and identified directly in the environmental samples using clone libraries with the same primers. In addition, enrichments in gradient tubes were evaluated after DGGE separation with general bacterial primers. No other iron-oxidizing bacteria than Gallionella spp. were found in the gradient tubes. qPCR provided an effective method to evaluate the growth of Gallionella spp. in these filter systems. The growth of Gallionella spp. was stimulated by subsurface aeration, but these bacteria hardly grew in the trickling filter. In the uninfluenced, natural anaerobic groundwater, Gallionella spp. were only present in low numbers, but they grew extensively in the trickling filter. Identification revealed that Gallionella spp., growing in the trickling filter were phylogenetically distinct from the species found growing during subsurface aeration, indicating that the different conditions in both systems selected for niche organisms, while inhibiting other groups. A 3D Biofilm model, appropriate for complex porous media support structures, is successfully modified such that non-zero permeability of biofilms structures is enabled. A systematic study is then conducted into the influence of biofilm permeability on overall biomass growth rate. This reveals a significant influence at large biofilm concentrations; even when the permeability of the biomass is 1.25% of that of the free pore space, biomass accumulation increased by a factor of ∼3 over 40 h. The effect is shown to be retained when allowing for biomass detachment or erosion as a consequence of adjacent velocity shear. We conclude that biofilm permeability should be included in biofilm models and that further experimental work is required to better describe the link between biofilm permeability and local microstructure., (Copyright © 2011 Wiley Periodicals, Inc.)

Local renal complement activation by the donor kidney plays an important role in the pathogenesis of renal injury inherent to kidney transplantation. Contradictory results were reported about the protective effects of the donor C3F allotype on renal allograft outcome. We investigated the influence of the donor C3F allotype on renal transplant outcome, taking all different donor types into account. C3 allotypes of 1265 donor-recipient pairs were determined and divided into four genotypic groups according to the C3F allotype of the donor and the recipient. The four genotypic groups were analyzed for association with primary nonfunction (PNF), delayed graft function, acute rejection, death-censored graft survival and patient survival. Considering all donor types, multivariable analysis found no association of the donor C3F allotype with renal allograft outcome. Also, for living and deceased brain-dead donors, no association with allograft outcome was found. Post hoc subgroup analysis within deceased cardiac dead (DCD) donors revealed an independent protective association of donor C3F allotype with PNF. This study shows that the donor C3F allotype is not associated with renal allograft outcome after kidney transplantation. WHO have long called for universal vaccination against hepatitis B worldwide. However, in north-western Europe low incidence of the disease has fueled debate whether targeted or universal vaccination strategies are the way to go for. Careful assessment has made it clear that the extensive targeted hepatitis B vaccination programmes in the Netherlands nevertheless fail to reach a significant part of the risk groups and have not succeeded in eliminating the disease. Modelling suggests that the public health benefits obtained through targeted programmes could be augmented considerably by universal vaccination. Therefore, the Minister of Health of the Netherlands has decided to implement universal vaccination by October 2011. We illustrate the case of the Netherlands and explore lessons, which can be learnt from the vaccination programmes against HPV and influenza A/H1N1 and how to prepare for a potential public debate that might arise when implementing universal vaccination against hepatitis B., (Copyright © 2011 Elsevier Ltd. Nanophotonic structures have attracted attention for light trapping in solar cells with the potential to manage and direct light absorption on the nanoscale. While both randomly textured and nanophotonic structures have been investigated, the relationship between photocurrent and the spatial correlations of random or designed surfaces has been unclear. Here we systematically design pseudorandom arrays of nanostructures based on their power spectral density, and correlate the spatial frequencies with measured and simulated photocurrent. The integrated cell design consists of a patterned plasmonic back reflector and a nanostructured semiconductor top interface, which gives broadband and isotropic photocurrent enhancement.

Several Mallotus species (Euphorbiaceae) are used in Vietnam as edible plants or as traditional medicines for different indications, some related to the treatment of inflammatory diseases. This study investigated the antioxidant activities of 33 samples from 17 Vietnamese Mallotus species. We also evaluated potential cytotoxic activity against human cervix carcinoma HeLa and human lung fibroblast WI-38 cells. Our aim is to develop safe dietary supplements with a protective effect against various diseases caused by tissue damage and the acceleration of the aging process linked to reactive oxygen species. These tests allowed the identification of non-cytotoxic plant species exhibiting significant antiradical properties. These antioxidant properties may be explained by their polyphenol composition. The antioxidant activity of the most active Mallotus species was further analyzed with and without tannins removal. We also identified by LC-ESI-MS some flavonoids responsible for a part of this activity.

Because of its eminent high resolution potential and minimal solvent consumption, pressurized capillary electrochromatography (pCEC) may offer an interesting alternative to HPLC for screening applications that need to resolve complex samples. In this paper, its potential was assessed in a screening of plant extracts from Mallotus species to indicate compounds with possible antioxidant activities by means of a PLS model built from their pCEC fingerprints. The main aim of this research was to find out whether pCEC can have an added value for this application. To get a complete overview of the techniques potential for this application, it was also assessed whether the technique can meet the requirements in terms of precision, sensitivity and column robustness. Encountered benefits and downsides were reported. Fingerprints with satisfactory sensitivity and precision could be obtained by concentrating the sample 5-fold and using optimized rinsing procedures, respectively. From the generated pCEC fingerprints of 39 Mallotus samples and their respective DPPH radical scavenging activity test results, a three-component PLS model was being built. The model proved good predictive abilities and easily allowed the indication of possible antioxidant compounds in the fingerprints. Despite its much higher peak capacity, the performance of pCEC to fingerprint the majority of the Mallotus extracts did not surpass that of a custom HPLC method. This was also reflected in its comparable power to indicate possible antioxidant compounds in the fingerprints after modeling. Because of its low detection sensitivity and modest column robustness, the benefit of the lower solvent consumption was partly paid-off by the current need for more system maintenance, also limiting the sample throughput. Six flavonoids including two new flavones, luteolin 7-O-(4''-O-(E)-coumaroyl)-beta-glucopyranoside), chrysoeriol-7-O-(4''-O-(E)-coumaroyl)-beta-glucopyranoside) and a mixture of two pairs of diastereoisomeric flavonolignans, (+/-)-hydnocarpin 7-O-(4''-O-(E)-coumaroyl)-beta-glucopyranoside)/(+/-)-hydnocarpin-D 7-O-(4''-O-(E)-coumaroyl)-beta-glucopyranoside) with a 2:1 ratio were isolated from the whole plant of Mallotus metcalfianus Croizat, in addition to 10 known compounds. Their structures were evaluated on the basis of different spectroscopic methods, including extensive 1D and 2D NMR spectroscopy. Some extracts have moderate antimicrobial properties and interesting antiradical (DPPH) activity, as well as some compounds isolated from this species. Tannins were also identified in some active extracts.

Phototrophic biofilms occur on surfaces exposed to light in a range of terrestrial and aquatic environments. Oxygenic phototrophs like diatoms, green algae, and cyanobacteria are the major primary producers that generate energy and reduce carbon dioxide, providing the system with organic substrates and oxygen. Photosynthesis fuels processes and conversions in the total biofilm community, including the metabolism of heterotrophic organisms. A matrix of polymeric substances secreted by phototrophs and heterotrophs enhances the attachment of the biofilm community. This review discusses the actual and potential applications of phototrophic biofilms in wastewater treatment, bioremediation, fish-feed production, biohydrogen production, and soil improvement.

The determination of diuron, atrazine, desisopropylatrazine (DIA) and desethylatrazine (DEA) were investigated using conductometric tyrosinase biosensor. Tyrosinase was immobilised on the biosensor sensitive part by allowing it to mix with bovine serum albumin (BSA) and then cross-linking in saturated glutaraldehyde (GA) vapour for 30min. The determination of pollutants in a solution was performed by comparison of the output signal (i.e percentage of the enzymatic activity) of the biosensor before and after contact with pollutants. The measurement of the enzymatic activity was performed using 4-chlorophenol, phenol and catechol substrates and response times ranging from 1 to 5min were observed. A 4-chlorophenol substrate was used to detect pesticides. A 30min contact time of the biosensor in the pollutant solution was used. Under the experimental conditions employed, detection limits for diuron and atrazine were about 1ppb and dynamic range of 2.3-2330 and 2.15-2150ppb were obtained for diuron and atrazine, respectively. A relative standard deviation (n=3) of the output signal was estimated to be 5% and a slight drift of 1.5muSh(-1) was observed. The 90% of the enzyme activity was still maintained after 23 days of storage in a buffer solution at 4 degrees C.
