The Permian Basin is one of the most prolific, and currently one of the most active, oil and gas basins in the USA. The Lower Permian strata in the Permian Basin have produced more than 14 billion barrels of oil (BBO), making it the largest volume of hydrocarbon in the basin. Sedimentation in the Midland Basin during late Leonardian through early Guadalupian (ca. 272e269 Ma) resulted in progradation of shelf edge and ultimately closure of the basin by Middle Permian time. We analyzed a merged seismic survey covering parts of the Permian Basin (i.e., Central Basin Platform and Midland Basin) in Andrews, Ector, and Midland Counties, Texas. The seismic survey and well logs show the presence of gently dipping (ca. 1°) clinoforms in the Upper San Andres and Grayburg Formations on the eastern edge of the Central Basin Platform and western Midland Basin. The seismic attributes, curvature, and spectral decomposition identify low sinuosity slope channels oriented north-south, but such channels do not appear beyond the slope. The shelf edge shifts from north to south during deposition of the Upper San Andres and Grayburg Formations. We identify five basinward shifts noted by the migration of the shelf edge toward the basin center and the presence of channel features along the depositional slope. The petrophysical analysis indicates that channels cut into carbonate rocks and are filled by siliciclastic sediments; this interpretation is supported by the most negative curvature anomalies along the channel axes caused by the differential compaction between the carbonate and siliciclastic rocks. There are a few channels with a northwest-southeast strike, which matches the direction of the Concho Lineament observed by satellite data. The tannery is one of the leading revenue-generating sectors in developing countries. The ever-increasing demand for leather products in the global market requires converting large amounts of rawhide/skins into resilient non-putrescible finished leather. Only 20% of the raw material is converted into a finished product; the rest 80% is discarded as solid and liquid wastes during leather processing. A heavy discharge of improperly treated solid tannery waste (STW) causes a severe impact on the surrounding environment by polluting soil, surface water, and groundwater resources, posing severe hazards to human and animal health. STW comprises proteinaceous untanned and tanned waste, which requires proper treatment for eco-friendly disposal. Several strategies have been developed over the years for the reduction and recycling of STW for producing renewable energy (biogas and biohydrogen), biofuels (biodiesel and briquettes), construction material, fertilizers, commercial products (adsorbents, animal feeds, proteins, fats, and enzymes), and biodegradable packaging and non-packaging materials. In this review, we discuss various strategies adopted for recycling, reutilization, and reduction of STW in an environment-friendly manner. Furthermore, an overview of the current perspectives toward achieving a zero-waste policy is also presented to reduce the environmental burden using green-clean technology to aid the survival of present-day tanneries. Adeno-associated virus (AAV)-based gene therapies are emerging strategies in Duchenne muscular dystrophy (DMD) treatment. Exposure to wild-type AAV can lead to development of neutralizing antibodies (NAbs) and blocking of AAV transduction, thereby limiting the delivery of AAV vector-based gene therapy. Therefore, it is imperative to check for the presence of AAV NAbs in a patient who is a candidate for gene therapy. We prospectively enrolled 101 genetically confirmed males with DMD (median age 11 years, 48% ambulatory and 59% on steroids) and performed AAV neutralization assays against AAV2, AAV8, AAV9, and AAVrh74 serotypes. The serotype analysis showed that AAV9 (36%) and AAVrh74 (32%) seroprevalence was lower compared with AAV2 (56%) and AAV8 (47%). Interestingly, age was not correlated with NAb titer for any of the capsids. NAb responses were observed at a higher frequency in African American participants and at a lower frequency in Caucasian participants for all four serotypes. Further analysis showed no significant differences in NAb titers regardless of serotype and whether participants were taking steroids or not. Finally, we observed higher AAV8, AAV9, and AAVrh74 seroprevalence and significantly higher AAV2 and AAV8 NAb titers in participants who were ambulatory compared with nonambulatory participants. Overall, these data identify AAV9 and AAVrh74 as the two serotypes with lower pre-existing NAb titers in this study's cohort of 101 males with DMD, possibly showing their utility in future gene therapy applications in treatment of this cohort of patients with DMD. Active macro seepages of methane that occur in between the north bank of the Brahmaputra river and Himalayan foothill region of Assam Arakan Basin, India, indicate the presence of hydrocarbon accumulation in the subsurface, but the hydrocarbon prospects in this region are not well studied. We carried out an extensive field sampling, which included a total of 58 sediment core collections from an active gas seepage location and nearby areas at a depth of 2–2.5 m. Our sample locations are placed at 1 km intervals laterally. We performed laboratory investigations and mapped near-surface chemical alterations associated with active macro seepages and microseepages. The analysis of geochemical composition of hydrocarbon gases in the sediment indicates both the biogenic and thermogenic origins of seeped hydrocarbons. The stable isotope analysis of methane suggests the presence of thermogenic as well as mixed biogenic-thermogenic gases. The presence of such mixing of gases is caused by the secondary alteration processes during their migration through potential faults and fractures. The trace elements of the sediments show anomalous concentrations at different parts of the study area, with a wide range of concentrations for Ba (54 to 492 ppm), Cu (1–25 ppm), Cr (61–329 ppm), Ni (1–42 ppm), Pb (2–48 ppm), Th (2–32 ppm), U (4–39 ppm), V (19–133 ppm) and U (0.87–6.5 ppm). There are higher concentrations of adsorbed gases, trace elements, and microbes along the identified lineaments. Such higher concentration can be triggered by high hydrocarbon-oxidizing bacteria count, which is greater than 104 cfu/gm of soil of bacterial growth around the lineaments. We identified potential hydrocarbon prospects based on the macro and micro seepage analysis using integrated geological, geochemical and microbial techniques in the study area. Metagenomics facilitates the isolation of novel enzymes from the unculturable microorganisms inhabiting a particular ecosystem. In the present study, solid tannery waste (STW) metagenome was explored as a potential source of microbial serine proteases. KEGG annotated functional profile of the UNK metagenome was exploited for in silico mining of candidate serine proteases. A novel serine protease was identified, namely Prt1SU, corresponding to an 1149 bp full-length gene encoding a 383 amino acid protein. The identity of the Prt1SU was evaluated by PSI-BLAST against the databases at NCBI and BLAST against the MEROPS database. Identities ranged from 30–45.7% to their respective nearest homologs of the serine protease family. Utilizing the DNA template isolated from the UNK metagenome, the candidate serine protease gene was cloned in the pET28a(+) vector and expressed in E. coli BL21 (DE3) strain. Although the Prt1SU was overexpressed at 18ºC using 0.1 mM IPTG, it remained inactive because of the formation of inclusion body. After solubilization with 8 M urea, the Prt1SU enzyme was purified to homogeneity using Ni-NTA chromatography under denaturing conditions and then renatured to retain protease activity. The purified Prt1SU with a molecular weight of ~ 40 kDa exhibited the maximum enzyme activity at pH 9.0 and temperature 37ºC. The enzyme activity of the Prt1SU was enhanced by the addition of organic solvents, and the enzyme was found stable in the presence of anionic and non-ionic surfactants and oxidizing agents, suggesting that Prt1SU is suitable for various industrial applications as well as wastewater and STW treatment. We report structural characterization of proteases mined in silico from solid tannery waste (STW) metagenome. The physico-chemical analysis revealed the molecular weight of selected query proteases in the range of 34–43 kDa except 52.46 kDa for Cp-6. Secondary structure analysis suggested the dominance of α-helices (26–51 %) followed by β-sheets (13–34 %). Conserved regions in the selected proteases were identified using multiple sequence alignment. Diversity analysis of the proteases was performed by aligning with their best hits and already characterized proteases of similar families. The 3D structures of 19 selected amino acid sequences were deduced using homology modeling and evaluated following Ramachandran plots (R-plots), stability of the 3D conformation, and overall quality factor (G factor). The R-plots of all structures had 96.4–98.9 % residues in the favoured region, whereas 0-0.8 % lied in the outlier region. Fifteen modeled protein structures passed the quality assessment criteria and were subsequently used for molecular docking. All models showed two domains except Cp-6, which had an extra domain. Ligand binding sites and active sites were identified using sequence and structural homologs of the respective protease molecules. Serine, lysine, and serine of different conserved motifs were specified as the active site residues in carboxypeptidases, whereas serine, aspartate, and histidine were specific to aminopeptidases Ap-1, Ap-2, and Ap-4. Further, Ap-8 and Ap-9 required Mn2+ for enzyme activity along with histidine and glutamate in their active sites. These 3D structures were used for molecular docking with their specific peptide ligands to obtain stable docked conformations. Successfully docked complexes indicated the catalytic activity of these enzymes in hydrolyzing the peptide ligand. The structural and functional insights gained from our study may help in the identification of novel industrially important proteases after validation through experimental studies. Background: Soft tissues of the head and neck areas are a very frequent site of occurrence of certain benign tumors of the peripheral nerve sheath, especially the neurofibromas. Hence, the present study was conducted for assessing clinical, radiographic, and treatment profile of 10 cases of neurogenic tumors and tumor-like lesions of the oral and maxillofacial region. Materials and Methods: Data records of a total of 10 patients who were diagnosed with tumors of neurogenic origin were enrolled in the present study. Data files were analyzed over a time period of 2 years, and complete clinical and radiographic details were evaluated. All the patients in which incomplete information was present in the record files were excluded from the present study. The assessment of the histopathologic reports was done, and final diagnosis was recorded separately in the master chart. Results: Neurofibroma was the diagnosis in two cases. In another set of two cases, final diagnosis of traumatic neuroma was achieved. A single case Schwannoma of mandible depicting multilocular radiolucency was present. Granular cell tumor was present in three cases. It was present clinically in the form of swelling, ulcerative nodule, and nodular growth in the three respective cases. Surgical excision was carried out in all the cases, and follow-up records did not depict any case of recurrence of complication posttreatment. Conclusion: Neurogenic tumors of oral and maxillafacial region are a rare phenomenon and mainly present in the form of benign neoplasm. Despite tremendous resources being invested in prevention and treatment, breast cancer remains a leading cause of cancer deaths in women globally. The available treatment modalities are very costly and produces severe side effects. Drug repurposing that relate to new uses for old drugs has emerged as a novel approach for drug development. Repositioning of old, clinically approved, off patent non-cancer drugs with known targets, into newer indication is like using old weapons for new battle. The advances in genomics, proteomics and information computational biology has facilitated the process of drug repurposing. Repositioning approach not only fastens the process of drug development but also offers more effective, cheaper, safer drugs with lesser/known side effects. During the last decade, drugs such as alkylating agents, anthracyclins, antimetabolite, CDK4/6 inhibitor, aromatase inhibitor, mTOR inhibitor and mitotic inhibitors has been repositioned for breast cancer treatment. The repositioned drugs have been successfully used for the treatment of most aggressive triple negative breast cancer. The literature review suggest that serendipity plays a major role in the drug development. This article describes the comprehensive overview of the current scenario of drug repurposing for the breast cancer treatment. Background: Motor unit number index (MUNIX) is a validated electrophysiological biomarker in amyotrophic lateral sclerosis. MUNIX studies in spinal muscular atrophy (SMA) are limited.Methods: Later-onset SMA children (n = 13; three SMN2 copy number) were evaluated for Hammersmith Motor Function Scale Expanded (HMFSE) and MUNIX of right abductor pollicis brevis (APB) and abductor digiti minimi (ADM) muscles. Age-matched healthy controls (n = 8) were enrolled to obtain normative APB and ADM MUNIX values.Results: Mean APB and ADM MUNIX values in SMA subjects were significantly reduced (P < .001) compared with controls. HMFSE scores strongly correlated with ADM MUNIX (r 0.63).Conclusions: APB and ADM muscle MUNIX studies are feasible in SMA type 2 children. Tanneries pose a serious threat to the environment by generating large amount of solid tannery waste (STW). Two metagenomes representing tannery waste dumpsites Jajmau (JJK) and Unnao (UNK) were sequenced using Illumina HiSeq platform. Microbial diversity analysis revealed domination of Proteobacteria , Firmicutes , Bacteroidetes , Actinobacteria , and Planctomycetes in both metagenomes. Presence of pollutant degrading microbes such as Bacillus , Clostridium , Halanaerobium and Pseudomonas strongly indicated their bioremediation ability. KEGG and SEED annotated main functional categories included carbohydrate metabolism, amino acids metabolism, and protein metabolism. KEGG displayed 5848 and 9633 proteases encoding ORFs compared to 5159 and 8044 ORFs displayed by SEED classification in JJK and UNK metagenomes, respectively. Abundantly present serine- and metallo-proteases belonging to Bacillaceae , Clostridiaceae , Xanthomonadaceae , Flavobacteriaceae and Chitinophagaceae families exhibited proteinaceous waste degrading ability of these metagenomes. Coronavirus infection is a transmissible disease. It was first described in China in December, 2019. It has been said to have a person-to-person transmission after prolonged and unprotected exposure. Patients with a potential SARS-CoV-2 exposure present with symptoms of low-grade pyrexia, dry cough, or shortness of breath. People with these symptoms should contact health-care providers before seeking medical intervention so that appropriate preventive actions may be implemented. Tangential velocity (V t) of tornadoes is the major parameter that causes building damage. In-field tornado measurements are less reliable at less than 20 m above ground level (AGL). Laboratory tornado simulators suggest that swirl ratio (S) and radius (r o) are the major tornado parameters that influence the V t. However, due to scaling problems, the laboratory simulators also report the V t at greater than 20 m AGL. Well-refined computational fluid dynamics (CFD) models can evaluate the V t at less than 10 m AGL. However, the CFD models are limited to r o = 1.0 km, and the effect of r o on V t is not investigated. The aim of this study is to investigate the maximum V t for different r o close to ground. Simulation results show that increasing r o decreases the maximum V t with respect to V ro. Moreover, by increasing ro, the corresponding elevation of occurrence of maximum Vt (zmax) will increase. However, for all tornado radii, the zmax is between 20 m and 64 m AGL. In addition, results show that for all r o , the radial V t profile has two peaks at z < 10 m AGL due to strong shear force close to the ground and at higher elevation the profile transits to Rankine Combined Vortex Model (RCVM). • When the tornado or tornado chamber radius (r o) increases the touchdown swirl ratio increases. • o The increase in radius (r) reduces the ratio of the maximum tangential velocity to radial velocity. • Close to the ground, the radial profile of the tangential velocity has two peaks. • Close to the ground the radial velocity profile is not same as the Rankine Combined Vortex Model. The long non-coding RNAs (lncRNAs) are the crucial regulators of human chronic diseases. Therefore, approaches such as antisense oligonucleotides, RNAi technology, and small molecule inhibitors have been used for the therapeutic targeting of lncRNAs. During the last decade, phytochemicals and nutraceuticals have been explored for their potential against lncRNAs. The common lncRNAs known to be modulated by phytochemicals include ROR, PVT1, HOTAIR, MALAT1, H19, MEG3, PCAT29, PANDAR, NEAT1, and GAS5. The phytochemicals such as curcumin, resveratrol, sulforaphane, berberine, EGCG, and gambogic acid have been examined against lncRNAs. In some cases, formulation of phytochemicals has also been used. The disease models where phytochemicals have been demonstrated to modulate lncRNAs expression include cancer, rheumatoid arthritis, osteoarthritis, and nonalcoholic fatty liver disease. The regulation of lncRNAs by phytochemicals can affect multi-steps of tumor development. When administered in combination with the conventional drugs, phytochemicals can also produce synergistic effects on lncRNAs leading to the sensitization of cancer cells. Phytochemicals target lncRNAs either directly or indirectly by affecting a wide variety of upstream molecules. However, the potential of phytochemicals against lncRNAs has been demonstrated mostly by preclinical studies in cancer models. In laboratories, core cannot always be cut exactly along the axis of symmetry (normal to the bedding plane), which leads to a minor core cutting rotation (CCR) error. The presence of a small CCR error can give rise to an error in computation of anisotropic parameters, which will result in erroneous P-wave and S-wave velocities (V P, V SV and V SH). In this study, we test the sensitivity of Thomson's anisotropic parameters, epsilon (ε), gamma (γ) and delta (δ), to the CCR error. In the vertical transverse isotropy (VTI) system where no rotation exists, values of ε , γ and δ will not vary with azimuth. Similarly, in VTI the values of V P, V SV and V SH, measured at orientations with respect to the axis of symmetry, will also not vary with azimuth. We modelled and analysed the error generated, in anisotropic parameters and phase velocities, due to rotation error of 5° on VTI ANNIE model data, and the Niobrara Shale core sample measurements. For these two cases, we obtained values of P-wave and S-wave velocities along with anisotropy parameters ε , γ and δ , at 0°, 45° and 90° orientations with respect to the axis of symmetry, before and after a rotation of 5°, for different azimuth directions. This study shows that, when the CCR error exists, ε , γ and δ vary with azimuth. For Niobrara Shale samples, we observed that, among all three Thomsen's parameters δ is the most sensitive parameter to the CCR error; when we varied azimuth, we observed a sign change of δ from positive to negative. For ε and γ , variation in azimuth lead to only slight changes in values without any change of sign. The CCR error affects V P and V s measurements the most at 45° orientation, and the least at 90° orientation. The maximum error occurred at 0° azimuth, whereas the minimum error occurs at 90° azimuth. This analysis suggests that, if core cutting rotation exists, phase velocities should be measured at 90° azimuth for accurate results. Transmission expansion planning (TEP) has become a complex problem in restructured electricity markets. This article presents the symbiotic organisms search (SOS) algorithm, a novel metaheuristic optimization technique for solving TEP problems in power systems. The SOS algorithm is inspired by the interactions among organisms in an ecosystem. The TEP problem is formulated here as an optimization problem to determine the cost-effective expansion planning of electrical power systems. Several constraints, such as power flow of the lines, right-of-way validity and maximum line addition, are taken into consideration. First, the SOS algorithm is tested with several benchmark functions. Then, it is applied on three standard power system networks (IEEE 24-bus system, Brazilian 46-bus system and Brazilian 87-bus system) in a TEP study to demonstrate the optimization capability of the proposed SOS algorithm. Hypoplasia of the right hepatic lobe is a rare and usually asymptomatic anomaly, which sometimes may be associated with portal hypertension but rarely with retroperitoneal fibrosis. We report one such rare case of hypoplastic right hepatic lobe associated with portal hypertension and retroperitoneal fibrosis. A 45-year-old nonalcoholic male presented with a chief complaint of hematemesis. Serum investigations were normal and inconclusive, so computed tomography was performed which showed right hepatic lobe hypoplasia, esophageal varices, and retroperitoneal fibrosis. Since all other causes of acquired liver atrophy had been ruled out, we considered this case to be of congenital origin. Background: Accurate working length determination helps to decide the extent to which the instruments are placed. The present study was conducted to evaluate the effect of canal curvature on accuracy of apex locator. Materials & Methods: The present study was conducted on 54 mandibular molar teeth. The file clip of apex locator was attached to the file and the file inserted until the "Apex" reading was reached in Root ZX. This was the electronic working length (EWL). Differences between EWL and actual working length (AWL) were calculated. Results: Group I teeth were with root curvature < 20°, group II with 20-36° and group III with >36°. Each group had 17 teeth. Maximum difference was observed in group III, followed by group II and group I. Background: Prevention of oral diseases is preferable to treatment and is the key method of achieving cost effectiveness for oral health improvement programs. The present study was conducted to determine the retention of pit and fissure sealants with different techniques. Materials & Methods: The present study was conducted on 60 patients of both genders. Patients were divided into 2 groups. In group I, mandibular permanent first molar of both sides was treated by acid etching alone and in group II, mandibular permanent second molar of both sides was pretreated by with air abrasion followed by acid etching. All patients were clinically determined after 3 and 6 months of sealant placement. Results: Out of 60 patients, males were 32 and females were 28. Completely retained sealants after 6 months was seen in 30 patients in group I and 28 in group II, partially in 15 in group I and 16 in group II and missing in 15 in group I and 16 in group II. In this paper, we propose and perform the experimentally calibrated simulation of a novel structure of a GaN/AlGaN high electron mobility transistor (HEMT). The novelty of the structure is the realization of enhancement mode operation by employing polarization engineering approach. In the proposed polarization engineered HEMT (PE-HEMT) a buried Aluminum Nitride (AlN) box is employed in the GaN layer just below the gate. The AlN box creates a two-dimensional hole gas (2DHG) at the GaN/AlN interface, which creates a conduction band barrier in the path of the already existing two-dimensional electron gas (2DEG) at GaN/AlGaN. Therefore, there is no direct path between the source and drain regions at zero gate voltage due to the barrier created by AIN and the device is initially OFF, an enhancement mode operation. A two dimensional (2D) calibrated simulation study of proposed PE-HEMT shows that the device has a threshold voltage (V th ) of 2.3 V. The PE-HEMT also reduces the electron spillover and thus improves the breakdown voltage by 108% as compared to conventional HEMT. The thermal analysis of the GaN PE-HEMT shows that a hot zone occurs on the drain side gate edge. Electrodeposition of CuAg alloy films from plating baths containing 3,5-diamino-1,2,4-triazole (DAT) as an inhibitor yields high surface area catalysts for the active and selective electroreduction of CO2 to multicarbon hydrocarbons and oxygenates. EXAFS shows the co-deposited alloy film to be homogeneously mixed. The alloy film containing 6% Ag exhibits the best CO2 electroreduction performance, with the Faradaic efficiency for C2H4 and C2H5OH production reaching nearly 60 and 25%, respectively, at a cathode potential of just -0.7 V vs RHE and a total current density of ~ - 300 mA/cm². Such high levels of selectivity at high activity and low applied potential are the highest reported to date. In situ Raman and electroanalysis studies suggest the origin of the high selectivity toward C2 products to be a combined effect of the enhanced stabilization of the Cu2O overlayer and the optimal availability of the CO intermediate due to the Ag incorporated in the alloy. Introduction: This study's objective was to evaluate quantitative electromyography (QEMG) using multiple-motor-unit (multi-MUP) analysis in Duchenne muscular dystrophy (DMD).Methods: Ambulatory DMD boys, aged 5-15 years, were evaluated with QEMG at 6-month intervals over 14 months. EMG was performed in the right biceps brachii (BB) and tibialis anterior (TA) muscles. Normative QEMG data were obtained from age-matched healthy boys. Wilcoxon signed-rank tests were performed.Results: Eighteen DMD subjects were enrolled, with a median age of 7 (interquartile range 7-10) years. Six-month evaluations were performed on 14 subjects. QEMG showed significantly abnormal mean MUP duration in BB and TA muscles, with no significant change over 6 months.Conclusions: QEMG is a sensitive electrophysiological marker of myopathy in DMD. Preliminary data do not reflect a significant change in MUP parameters over a 6-month interval; long-term follow-up QEMG studies are needed to understand its role as a biomarker for disease progression. Pretreatment and hydrolysis of lignocellulosic biomass are affected by several uncertainties, which must be systematically considered for a robust process design. In this work, stochastic simulations for expected uncertainties in feedstock composition, kinetic parameter values, and operational parameter values for these two steps were performed. The results indicated that these uncertainties significantly impacted the concentration profiles, which could also affect the optimal batch time. Global sensitivity analysis was then used to identify the critical uncertain parameters. In the feedstock components, cellulose and xylan fractions for acid pretreatment and cellulose fraction for enzymatic hydrolysis were important. Temperature was the most sensitive operating parameter for both acid pretreatment and hydrolysis. The activation energies for different reactions were ranked in terms of their impact on process output. Present study aimed to evaluate the changes in proximate composition and physical attributes in brown shrimp (Metapenaeus dobsonii) exposed to different methods of cooking followed by freezing. For this, three different grades (100/200, 200/300, and 300/500 numbers per kg) of brown shrimp were cooked at 90°C till the core temperature of the product reaches 85°C using hot water, steam, and microwave (400W) techniques. The changes in yield, cooking loss, proximate composition, textural, and colour profile were assessed for cooked shrimps. The cooking loss was higher for larger grades of shrimp, whereas shrimp cooked using hot water exhibited the highest cooking loss. Lowest cooking loss was observed for microwave-cooked shrimp. Moisture content decreased after cooking whereas protein, fat, ash, and calorie content increased. After cooking, different grades of shrimp showed an increase in their lightness (L*), redness (a*), and yellowness (b*) values. The smaller grade shrimp exhibited lower value for cohesiveness, hardness, chewiness, and gumminess. Different cooking techniques yielded cooked shrimp of varying hardness values. Standardization of metagenomic DNA extraction protocol is a pre-requisite for a successful metagenomic study aiming to screen and exploit the variety of microorganisms inhabiting a particular soil environment. Six methods reported earlier were used for isolation of metagenomic DNA in the present study. These methods suffered with regard to either poor yield or quality of DNA. Therefore, we developed an improved method for isolation of high-molecular weight and good quality metagenomic DNA from different soil samples. Our protocol combines the enzymatic (lysozyme and proteinase K) and chemical (CTAB and CaCl) strategies to ensure efficient cell lysis and use of PEG and isopropanol for precipitation of humic impurities-free DNA. Our improved method gave high yield of good quality metagenomic DNA from diverse soils collected from garden, domestic waste dumping site, cellulose waste dumping site, sewage site, and tannery waste site. Power sector reform, which circumscribes independent regulatory commission, unbundling of the State Electricity Boards (SEBs), and eventual privatization, has been the dominant theme in the Indian economic policy discourse and power sector. In India, the power sector reforms were initiated in 1991 and began with the concurrence of the private investors in generation, and then centralized on the unbundling of vertically integrated utilities. In this paper, an attempt has been made to analyze the changing importance of energy conservation and efficiency measures been initiated in India between 1970 and 2013. The study also examines how far the reform process has been effective in realizing its set objectives and ameliorates the development of the power sector and the nation unrestrained. The paper briefly discusses the issue involved in the introduction of competition in the power sector primarily through the development of a market for bulk power, and a careful comparison of the costs and benefits of the old regulated system and the new deregulated system is made. DNA double-strand breaks are critical lesions that can lead to chromosomal aberrations and genomic instability. In response to DNA damage, Chk1, a serine/threonine kinase, is responsible for cell cycle arrest to prevent damaged cells from progressing through the cell cycle. Here, we report that the disruption of wat1, a WD repeat-containing protein, leads to the phosphorylation of Chk1. The double-deletion of chk1 and wat1 had a grave effect on the survival of fission yeast cells, and the spontaneous recombination rate was also high upon double-deletion of wat1 and chk1, as compared to the single-mutant. In the absence of wat1, the cells exhibited a high level of nuclear fragmentation that resulted in the accumulation of Rad22 yellow fluorescent protein foci. Furthermore, we show that wat1 is required for the regulation of the oxidative stress response. We observed elevated levels of reactive oxygen species (ROS) generation in wat1-null mutant that led to a high degree of propidium iodide staining at nonpermissive temperature. Based on the results presented here, we hypothesize that ROS production in wat1-null mutant cells generates DNA fragmentation that could trigger a checkpoint response and that, in the absence of checkpoint kinase Chk1, the cells exhibit severe growth defects leading to a synthetic lethal phenotype. Several studies have been carried out to investigate the magnitude of forces induced by tornado-like winds on buildings from experimental as well as computational side. The number of experimental studies evaluating the wind loads due to tornado-like vortices are comparatively greater in number than studies based on computational fluid dynamics (CFD). Furthermore, the limited number of CFD studies were often not validated by comparing with experimental measurements. In this work, an attempt is made to validate a CFD model by comparing the pressures induced by tornado-like vortices on a building model with experimental measurements from the tornado simulator at Texas Tech University (TTU). Results of the comparison indicate that the pressure coefficients obtained from the CFD model agree well with TTU experimental datasets. Besides, the effect of building size as well as the effect of flow structure of vortex on pressure coefficients on the building induced by tornado-like vortex is also investigated. Considering system uncertainties prevailing in the restructured power system, congestion management (CM) is a vital issue in power system operation and control. The objective of CM is to remove congestion in the lines while satisfying all the constraints with minimum congestion cost. This study proposes a generation rescheduling‐based approach for CM in electricity market using a novel ant lion optimiser (ALO) algorithm. ALO is a recently developed the nature‐inspired algorithm based on the hunting mechanism of antlions. The effectiveness of the proposed approach is tested on modified IEEE 30‐bus, modified IEEE 57‐bus and IEEE 118‐bus test systems. The security constraints like load bus voltage and line loading impact are incorporated in this study. To prove the validity of the proposed technique, the obtained results are compared to the results offered by various recent optimisation algorithms. The results show that the proposed ALO algorithm outperforms the other comparative algorithms. The proposed approach uses less number of fitness function evaluations, not traps into local minima and offers promising convergence characteristic. Introduction: Patients with connective tissue disorders (CTD) and pulmonary arterial hypertension (PAH) have a poorer prognosis than those with other PAH etiologies. This study assessed the impact of CTD on healthcare outcomes among PAH patients with and without CTD comorbidities that were treated with oral selexipag. Methods: The study utilized Optum's de-identified Clinformatics® Data Mart Database (2007–2021) from January 1, 2014 to June 30, 2019, and identified patients with PAH without CTD and PAH with CTD treated with oral selexipag. Patients had ≥ 12-month baseline period with no requirement for a minimum follow-up period. Patients were followed until any of the following events: discontinuation of oral selexipag, or health plan disenrollment, or death, or presence of a diagnosis claim for CTEPH, or study end date, whichever occurred first. PAH-related hospitalizations, PAH disease progression, and healthcare utilizations and costs were assessed in the follow-up period. The Cox proportional hazards model was used to evaluate the time to hospitalization and generalized linear models were used to examine healthcare costs and utilization between the two cohorts. Results: In the analysis, 237 PAH without CTD, and 80 PAH patients with CTD comorbidities prescribed oral selexipag were included. The PAH without CTD comorbidities cohort was older (65 vs. 63 years old), had proportionately less females (72 vs. 83%), and higher comorbidity burden than PAH with CTD comorbidities (mean CCI index 3 vs. 2). After adjusting for potential confounders, the risk for PAH-related hospitalization (hazard ratio (HR) 1.13, p value 0.641), all-cause hospitalization (HR 1.09, p value: 0.765), and PAH disease progression (HR 1.14, p value 0.522) between the two cohorts were similar. After adjusting for baseline demographic and clinical characteristics, PAH with CTD comorbidities incurred higher total mean all-cause PAH-related medical care costs compared to PAH without CTD comorbidities. Conclusions: In this real-world study, the risk of hospitalization and PAH disease progression were similar between the two cohorts who received oral selexipag. Phenols are ubiquitous pollutants, mainly from industrial effluent, causing pollution of natural water resources. The research focused on screening efficient phenol-degrading bacteria and kinetic modelling of phenol biodegradation and growth. Membrane filtration was used for the isolation of bacteria from the wastewater sample. The screening of phenol-degrading bacteria was based on the efficiency of phenol utilization. The strain with efficient phenol degradation capacity was characterized by 16S rDNA sequencing and designated Acinetobacter baumannii W29. Biomass growth and phenol utilization rate of the strain were evaluated at different initial phenol concentrations (100-800 mgL-1). Specific growth rate data were fitted to five models, i.e. Monod, Haldane, Aiba, Teisser, and Webb model. The yield coefficient at different initial phenol concentrations was calculated from the slope of the specific growth rate (μ) versus the specific phenol utilization rate (q). The strain showed complete phenol degradation potential up to 1000 mgL-1. The maximal growth rate was achieved at 400 mgL-1, which coincided with the maximum substrate utilization rate at the same concentration. The specific growth rate showed the best fit with the Haldane model. The strain had a yield coefficient of 0.70 (mg cell mg-1 phenol). The value of µ and Ks revealed the affinity of the strain for high-concentration phenol and the its ability to withstand high phenol concentrations. The kinetic growth behaviour of the strain fitted well with the Haldane model. The findings of the study could be applied to wastewater treatment with a high phenol load. [ABSTRACT FROM AUTHOR]

Aberrant chromosome segregation defects can lead to aneuploidy, a common characteristic of human solid tumors. Aneuploidy is generated due to defects in the mitotic spindle or due to inefficient mitotic checkpoint response. We have isolated a novel mutant allele of wat1, a WD repeat containing protein that exhibits conditional synthetic lethality with chk1 knock out. We observed only a marginal decrease in the level of α tubulin protein level in wat1-17 mutants after prolong exposure at semi permissive temperature. Interestingly the protein level of α-tubulin was reduced in the chk1Δ wat1-17 double mutant at 18°C with defective microtubule structure. Consistent with loss of microtubule structure in the chk1 deletion background, the double mutant of wat1-17 chk1Δ was hypersensitive to the microtubule destabilizing agent TBZ suggesting severe defects in microtubule integrity in wat1-17 mutant in the absence of Chk1. Combination of wat1-17 with the chk1 deletion also aggravates the defects in the maintenance of genome ploidy. The mutation in wat1-17 was mapped to Cys 233 that was changed to tyrosine. Based on the molecular modeling studies, we hypothesize that the substitution of the bulky Tyr residue at Cys233 position in wat1-17 mutant results in conformational changes. Abstract: This paper examines the effect of mixer size on the density of the propellant and it also explores the role of the propellant density with regards to burning rate and burn rate pressure index. Propellant samples were prepared in the four different size (15, 70, 200 and 1000g) mixers with identical input of ingredients to examine the effect of mixer size on density and burning rate of the composite solid propellant. It was noticed that density and burning rate of the propellant changes significantly with change in the mixer size from 15g to 1000g. This is because for the smaller mixers the surface area to volume ratio is large and the actual percentage of aluminum and ammonium perchlorate (especially coarse) that goes into the propellant reduces. High burning rate with decrease in mixer size is also accompanied with increase in burn rate pressure index. The composition analysis was also carried out and it is noticed that the actual percentage of the ingredients is different from the intended percentage of the ingredients and percentage change is more when the mixer size decreases. Abstract: Evaluation of acute ataxia in a child poses a dilemma for the clinician in determining the extent and timing of initial screening tests. This article reviews the evidence concerning the diagnostic yield of commonly ordered tests in evaluating the child with acute ataxia. The literature revealed the following frequencies of laboratory screening abnormalities in children with acute ataxia: CT (∼2.5%), MRI (∼5%), lumbar puncture (43%), EEG (42%), and toxicology (49%). In most studies, abnormalities detected by these screening tests were nondiagnostic. There are insufficient data to assess yields of testing for autoimmune disorders or inborn errors of metabolism. A toxicology screen should be considered in all children presenting with acute ataxia. Neuroimaging should be considered in all children with new onset ataxia. Cerebrospinal fluid analysis has limited diagnostic specificity unless clinically indicated. Studies to examine neurophysiology testing did have sufficient evidence to support their use. There is insufficient evidence to establish a role for autoantibody testing or for routine screening for inborn error of metabolism in children presenting with acute ataxia. Finally, in a child presenting with ataxia and opsoclonus myoclonus, urine catecholamine testing for occult neuroblastoma is recommended. Abstract: Rate constants of surface diffusion processes in metals often depend on the local atomic environment. Accurate models that describe this local environment dependence for specific material systems are not easily available in the literature despite the need for such information in kinetic models. In this work, we find the prefactor and activation barrier associated with adatom hop processes for a large number of local environments and use this information to systematically train cluster expansion (CE) models for Ag, Al, Cu, Ni, Pd and Pt. We demonstrate that although the local environments used for training is a small subset of all possible local environments, the CE model can predict the rate constants for many local environments that have not been used while training the model with reasonable accuracy. It is found that up to fourth nearest neighbor positions can be relevant to the activation barrier. Furthermore, many-body effects play an important role. DNA topoisomerases are specialized nuclear enzymes that perform topological modifications on double-stranded DNA (dsDNA) and hence are essential for DNA metabolism such as replication, transcription, recombination, condensation and segregation. In a genetic screen, we identified a temperature-sensitive mutant allele of topoisomerase 2 that exhibits conditional synthetic lethality with a chk1 knockout strain. The mutant allele of topoisomerase 2 is defective in chromosome segregation at a non-permissive temperature and there was increase in chromosome segregation defects in the double mutant of top2–10 and chk1 delete at a non-permissive temperature. More importantly, topoisomearse 2 mutant cells mildly delay the mitotic progression at non-permissive temperature that is mediated by checkpoint protein kinase Chk1. Additionally, top2–10 mutant cells also activate the Chk1 at a non-permissive temperature and this activation of Chk1 takes place at the time of mitosis. Interestingly, top2–10 mutant cells retain their viability at a non-permissive temperature if the cells are not allowed to enter into mitosis. Taking together our results, we speculate that in the top2–10 mutant, the segregation of entangled chromatids during mitosis could result in delaying the mitotic progression through the activation of Chk1 kinase. It states that muscle diseases in clinical practice can be screened through creatine kinase (CK) because its levels increase in condition with active muscle fiber necrosis and injury. The differential diagnosis of the disease, the use of glucocorticoids for treatment and the long-term management of comorbid conditions are also noted.

Abstract: This paper reports the discovery of activated charcoal as an effective burn rate enhancer of aluminized composite solid propellant. Experiments were carried out using a strand burner at pressures ranging from 10 to 70bar with varying fractions of activated charcoal. The results show that with the addition of activated charcoal in an aluminized composite propellant higher burn rates (in excess of 25mm/s at 70bar) were obtained. This paper also explores the possibility of further enhancing the burn rates of aluminized composite propellant by using iron oxide along with activated charcoal. Burn rates recorded with iron oxide and activated charcoal both present in the propellant were in excess of 50mm/s at 70bar. It was noticed that these high burn rates were accompanied by a higher (0.65) pressure index of combustion. A series of experiments were designed to understand the burn rate enhancement with activated charcoal in aluminized composite propellant. TGA and DSC tests were carried out on AP pellets with and without activated charcoal to detect any condensed phase activity of activated charcoal. These tests did not show any significant difference between the AP pellets with and without activated charcoal. The AP pellets with and without activated charcoal were also burnt in a strand burner to understand if any gas phase reaction was being catalyzed by activated charcoal. AC does not enhance any gas phase activity while addition of AC in AP increased the LPDL of AP and reduced the burn rate. Lastly, sandwich propellant with activated charcoal at the interface was prepared to determine if interface reactions were being affected with the addition of activated charcoal. It has been observed that with the addition of activated charcoal at the AP–binder interface the burn rates are higher than those recorded without activated charcoal at the AP–binder interface. The restructuring of power systems and the ever-increasing demand for electricity have given rise to congestion in power networks. The use of distributed generators (DGs) may play a significant role in tackling such issues. DGs may be integrated with electrical power networks to regulate the drift of power in the transmission lines, thereby increasing the power transfer capabilities of lines and improving the overall performance of electrical networks. In this article, an effective method based on the Harris hawks optimization (HHO) algorithm is used to select the optimum capacity, number, and site of solar-based DGs to reduce real power losses and voltage deviation. The proposed HHO has been tested with a complex benchmark function then applied to the IEEE 33 and IEEE 69 bus radial distribution systems. The single and multiple solar-based DGs are optimized for the optimum size and site with a unity power factor. It is observed that the overall performance of the systems is enhanced when additional DGs are installed. Moreover, considering the stochastic and sporadic nature of solar irradiance, the practical size of DG has been suggested based on analysis that may be adopted while designing the actual photovoltaic (PV) plant for usage. The obtained simulation outcomes are compared with the latest state-of-the-art literature and suggest that the proposed HHO is capable of processing complex high dimensional benchmark functions and has capability to handle problems pertaining to electrical distribution in an effective manner. The article presents a clinical case of an 18-year-old woman who was diagnosed with exercise-induced syncope. Prior to the diagnosis, the patient had experienced fainting during a track workout. She underwent several clinical examinations, including complete blood count (CBC) and liver function test, which yield normal results. However, interpretations of clinical symptoms revealed a brief precipitous vasodepressor syncope which is triggered by exercise. According to the author, patients with exercise-induced syncope are advised to increase fluid and salt intake while exercising.

Cytoplasmic and nuclear iron-sulfur (Fe-S) enzymes that are essential for genome maintenance and replication depend on the cytoplasmic Fe-S assembly (CIA) machinery for cluster acquisition. The core of the CIA machinery consists of a complex of CIAO1, MMS19 and FAM96B. The physiological consequences of loss of function in the components of the CIA pathway have thus far remained uncharacterized. Our study revealed that patients with biallelic loss of function in CIAO1 developed proximal and axial muscle weakness, fluctuating creatine kinase elevation, and respiratory insufficiency. In addition, they presented with CNS symptoms including learning difficulties and neurobehavioral comorbidities, along with iron deposition in deep brain nuclei, mild normocytic to macrocytic anemia, and gastrointestinal symptoms. Mutational analysis revealed reduced stability of the variants compared with WT CIAO1. Functional assays demonstrated failure of the variants identified in patients to recruit Fe-S recipient proteins, resulting in compromised activities of DNA helicases, polymerases, and repair enzymes that rely on the CIA complex to acquire their Fe-S cofactors. Lentivirus-mediated restoration of CIAO1 expression reversed all patient-derived cellular abnormalities. Our study identifies CIAO1 as a human disease gene and provides insights into the broader implications of the cytosolic Fe-S assembly pathway in human health and disease. [ABSTRACT FROM AUTHOR]

The primary safety and efficacy endpoints were freedom from LBBAP lead‐related serious adverse events and the composite of LBBA capture threshold of ≤2.0 V and R‐wave amplitudes ≥5 mV (or ≥value at implant), respectively.Of 221 patients with attempted implants of the Tendril 2088 lead in the LBBA, 91.4% (202/221) had successful implants for LBBAP. Regardless of the LBBAP implant success, all patients were followed for at least 6 months (8.7 ± 7.3 months). Baseline characteristics: 44% female, 84% ≥65 years old, 34% coronary artery disease, and 86% of primary indications for pacemaker implant. Both primary safety and effectiveness endpoints were met (freedom from LBBAP lead‐related serious adverse device effects of 99.5% and electrical performance composite success rate of 93%). The capture thresholds in LBBAP at implant and 6 months were 0.8 ± 0.3 V@0.4 ± 0.1 ms and 0.8 ± 0.3 V@0.4 ± 0.1 ms. The rate of patients with capture threshold rise ≥1 V was 1.5% through 6 months. The R‐wave amplitudes in LBBAP at implant and 6 months were 9.3 ± 3.2 mV and 10.6 ± 3.0 mV.This large multicenter study demonstrates that the stylet‐driven Tendril™ STS 2088 lead is safe and effective for LBBAP with high success and low complication rates. Introduction/Aims: Objective outcome measures in children undergoing treatment for chronic inflammatory demyelinating polyradiculoneuropathy (CIDP) are lacking. The aim of the study was to record serial grip strength and motor nerve conduction studies to assess interval change. Methods: This was a retrospective review of 16 children (8 females and 8 males; median age, 9.7 years; interquartile range, 6–13 years) with CIDP followed at a tertiary children's hospital from 2013 to 2021. Subjects were treated with intravenous immunoglobulin (IVIG). Right and left grip strength measurements were obtained at each clinic visit using a handheld dynamometer. Annual right median motor nerve conduction study data were recorded during the study period. Results: Mean duration of follow‐up was 2.9 years. Grip strength (right: 0.19 kg/month, p < 0.001; left 0.23 kg/month, p < 0.001) and median F‐wave latencies (−0.23/month, p = 0.015) showed significant improvement over time. Akaike information criterion showed time + IVIG frequency <21 days as best fit for grip strength and distal compound muscle action potential amplitude. Discussion: Our study results indicate serial grip strength measurements are a feasible and objective way to assess motor strength improvement in children with CIDP receiving immunotherapy. Electrocatalytic carbon monoxide reduction has been previously reported to yield a range of carbonaceous products including alcohols, hydrocarbons and carboxylic acids. However, esters, an important family of organic compounds, have not been formed. Herein, we report the electrosynthesis of C3 to C6 acetate esters (H3C−(C=O)−O−R) from carbon monoxide using copper catalysts in a membrane electrode assembly cell. Ethyl acetate and propyl acetate could be produced with an unprecedented total Faradaic efficiency (FE) of ∼22 % and with a current density of up to −55 mA cm−2, alongside minor quantities of methyl acetate and butyl acetate. The esters are produced via the addition reaction of ethenone (H2C=C=O) and alcohols produced during CO reduction. Pyrolyzing Cu(II) precursors absorbed on carbon black gave an eletrocatalytic material for the electroreduction of CO 2. Controlled potential electrolysis and GC analysis revealed an unexpected selectivity towards methane even in alkaline media, with a methane Faradaic efficiency as high as 42%, a partial current density of 100 mA/cm2 (at −1 V vs. RHE) and a methane/ethylene ratio of 4:1. XPS, EXAFS, and EDX mapping results indicate a single-site Cu(I) center as the catalytically active site.
