This study reports a peculiar case of systemic candidiasis infection associated with pulmonary aspergillosis in an apparently immunocompetent alpaca. A captive 7-year-old female alpaca exhibited respiratory symptoms, underwent treatment with benzylpenicillin and dexamethasone, and succumbed to the infection 40 days later. During the post-mortem examination, subcutaneous emphysema, widespread pneumonia with multiple suppurative foci, scattered necro-suppurative lesions throughout the renal and hepatic parenchyma were evident. Histopathological analysis of the collected tissues revealed multifocal mild lymphoplasmacytic chronic interstitial nephritis, necro-suppurative pneumonia with the presence of fungal hyphae, multifocal foci of mineralization, and fibrosis in the liver. Fungal cultures confirmed the growth of Aspergillus fumigatus from the lungs, and Candida albicans from the liver, kidney, and heart. The only recognizable risk factor for candidiasis and pulmonary aspergillosis in this case was prior corticosteroid and antibiotic therapy. Nevertheless, it is crucial to consider systemic candidosis and pulmonary aspergillosis as potential differential diagnoses in respiratory infections among camelids. This research aimed to assess the influence of red dragon fruit peels ratio (RDF-PR) from two species, Hylocereus hybridum (HH) and Hylocereus undatus (HU), and particle size (PS) on quality parameters of red dragon fruit peel powder (RDF-PP) and its further application in emulsified alpaca-based sausages as partial substitutes of pork-back fat. A three-level full factorial design (nine treatments) was employed to evaluate the effect of RDF-PR (HH(0%):HU(100%), HH(50%):HU(50%), and HH(100%):HU(0%)) and PS (499–297, 296–177, and <177 µm) on the dependent variables: L*, a*, b*, C, h°, water-holding capacity, oil-holding capacity, swelling capacity, pectin yield, degree of esterification (analysed through FT-IR), and crude fibre content. The data analysed through a response surface methodology showed that treatment one (T1) is the best with the optimised conditions at 100% HU RDF-PR and PS of <177 µm. The statistical validation of T1 exhibited the highest water-holding capacity (32.1 g/g peel), oil-holding capacity (2.20 g oil/g peel), and pectin yield (27.1%). A completely randomised design (four formulations) was then used to assess the effect of partial replacement of pork-back fat by T1 in emulsified alpaca-based sausages on the colourimetric, physicochemical, and texture properties (hardness, chewiness, cohesiveness, springiness, adhesiveness, and adhesive force). Likewise, a sensory hedonic scale was employed to evaluate the appearance, colour, odour, flavour, texture, and overall acceptability of sausages. The results revealed that 65.7% of pork-back fat content was successfully replaced compared with a control formulation. Additionally, F3 showed significantly (p < 0.05) better colourimetric, physicochemical, and textural characteristics, such as lower hardness (34.8 N) and chewiness (21.7 N) and higher redness (a* = 19.3) and C (22.9), compared to a control formulation. Alpacas have a high incidence of congenital reproductive tract abnormalities, including ovarian hypoplasia/dysgenesis. Diagnosis of this condition is often challenging. The present study describes the clinical, ultrasonographic, and histologic features of ovarian hypoplasia/dysgenesis syndrome in 5 female alpacas. Additionally, serum AMH levels were compared between female alpacas diagnosed with ovarian hypoplasia/dysgenesis and a group of reproductively sound females (n = 11). The syndrome was suspected based on the presence of an infantile uterus and lack of ovaries by ultrasonography and laparoscopy. All females had normal female karyotype (n = 74 XX), but one presented a minute chromosome. The ovaries from these cases showed 3 main histological classifications: hypoplasia (n = 2), dysgenesis (n = 2), and dysplasia (n = 1). Serum AMH levels in affected females were significantly lower (P < 0.05) than those of reproductively sound control females. In conclusion, Serum AMH level may be helpful in the rapid diagnosis of ovarian hypoplasia/dysgenesis syndrome in alpacas. Furthermore, this syndrome in alpacas presents a variety of histological features. Different mechanisms may be involved in the derangement of ovarian differentiation. Further studies are needed to elucidate the causes of the syndrome.

Universally, in microbiological diagnostics the detection of live bacteria is essential. Rapid identification of pathogens enables appropriate remedial measures to be taken. The identification of many bacteria simultaneously facilitates the determination of the characteristics of the accompanying microbiota and/or the microbiological complexity of a given environment.

Early pregnancy detection in alpacas, whose breeding season is limited to the rainy season and has a long gestation period, is important for reproductive management. Conventional detection methods such as ultrasonography cannot be used to detect pregnancy before 30 days after mating. In this study, we examined the feasibility of using fecal steroid hormones as an early detection method in pregnant and non‐pregnant alpacas. Fecal and blood samples were collected from pregnant and non‐pregnant alpacas after mating. Progesterone (P4) and estradiol 17‐β were extracted and quantified from blood and fecal samples. A positive correlation exists between the steroid hormones in serum and feces, indicating that serum steroid hormone concentrations can be estimated from fecal steroid hormones. Within 10 days after mating, both pregnant and non‐pregnant alpacas had fecal P4 concentrations greater than 1.0 ng/mg dry matter (DM), but by 15 days after mating, fecal P4 concentrations decreased to the pre‐mating concentration in non‐pregnant alpacas. From 15 days after mating, non‐pregnant alpacas had a low fecal P4 concentration (< 1 ng/mg DM), whereas a high fecal P4 concentration indicated the possibility of pregnancy, suggesting that this test is clinically beneficial as a supportive test for pregnancy detection. Eighty‐five sperm samples were cryopreserved and SYBR14/PI, MitoTracker Deep Red FM, FITC‐PSA/PI and chlortetracycline were used for imaging flow cytometry evaluation of sperm viability, mitochondrial membrane potential (MMP), acrosome integrity and sperm capacitation, respectively. Sperm motility was also registered. Sperm motility (46.1 ± 7.7 vs. 24.1% ± 6.5%), sperm viability (49.8 ± 11.5 vs. 32.3% ± 9.6%) and high MMP (49.8% ± 12.4% vs. 34.9% ± 9.9%) decreased significantly (p <.05) during cryopreservation process, in contrast to acrosome‐reacted in viable spermatozoa (1.0% ± 1.6% vs. 1.0% ± 1.0%) and sperm capacitation (10.0 ± 9.8 vs. 8.2% ± 12.4%) that were similar (p >.05) before and after cryopreservation. Positive correlations were found between sperm motility versus high MMP (r =.63), sperm motility versus sperm viability (r =.67) and sperm viability versus high MMP (r =.88). In conclusion, cryopreservation of alpaca spermatozoa is related to a decrease in sperm motility, sperm viability and high MMP, meanwhile acrosome integrity and sperm capacitation are not affected. Abstract Background Epididymal sperm can be collected post‐castration or post‐mortem. This method has already been described for alpaca (Vicugna pacos), but little data are available on success rate and the influence of diluent on it. Objective The aim was to investigate the effectiveness of epididymis incision as an extraction method. Method Post‐castration epididymides (n = 78) were incised and semen was collected from cut surfaces. Further sperm were flushed with two semen extenders. For the left epididymis, a diluent without animal proteins and for the right, a diluent with egg yolk was utilised. Collected sperms were immediately spermatologically examined. Results Due to incorrect measurements, the samples of seven epididymides were not analysed. An evaluation was possible in 58 samples. Average density was 108.80 ± 83.28 million/mL and motility was 53.30 ± 18.17%. On average, 76.70 ± 11.60% of the sperm were vital in eosin‐stained specimens. In the hypoosmotic swelling test, an average of 69.50 ± 10.48% of the sperm had an intact plasma membrane. Semen extender had no effect on spermatological parameters (p > 0.05). Overall success rate of sperm recovery was 83%. Extraction of epididymal sperm was possible in 68.80% of the 2‐year‐old males. The 13 epididymides from which no sperm recovery was possible were histologically examined and for 10, there was no histological evidence of sperm. The corrected success rate was 95.60%. Conclusion Testicular volume (p = 0.0453), but not age (p = 0.62), had an effect on the probability of obtaining sperm.

The breeding of alpaca (Vicugna pacos) is one of the most important economic activities in the high Andean areas of Peru. The commercialization of products derived from alpaca represents more than 80% of the income of high-Andean families. However, the infestation of parasites such as Sarcocystis lamacanis in the alpacas causes economic losses that deteriorate the already diminished quality of life of the alpaca breeder. The search for biomarkers that allow the early detection of these parasites is one of the most critical challenges in Peru, a country with the largest population of alpacas worldwide. This work aimed to analyze and quantify the microcysts formed by the parasite and relate them to the troponin cTnI level in the blood serum. Troponins are proteins secreted when there is damage to the cardiac muscle. 60 blood and cardiac tissue samples were collected from Tisco and La Raya slaughterhouses, localities of Caylloma Province in Arequipa, and Chucuito District in Puno, both regions in southern Peru. The cardiac muscle samples were processed with the routine histology technique and stained with hematoxylin and eosin. In addition, serum samples were processed with the ELISA and immunochromatography methods for troponin cTnI. Results were 100% positive for the presence of Sarcocystis lamacanis microcysts in all cardiac muscle samples. The average microcyst quantification per field of 100x were 3.5 and 5.7 for the Tisco and La Raya samples. In addition, several microscopic lesions were observed in the cardiac muscles: microcyst infiltration between muscle fibers, basophilic microcysts with a thick outer membrane and bradyzoites inside, and tissue displacement. On the other hand, all serum blood samples were negative for troponin cTnI, with both methods, ELISA and immunochromatography. For results, we infer troponin cTnI do not can be used as a biomarker for heart damage caused by Sarcocystis lamacanis parasite in alpacas.

Abstract Background Little is known about the epidemiology of Toxoplasma gondii and Neospora caninum infections in alpacas (Vicugna pacos) and llamas (Lama glama) outside South America. The study aimed to estimate the seroprevalence of T. gondii and N. caninum infections in South American camelids (SAC) in Switzerland, to optimize serological tests for SAC and to identify risk factors, which may favour infection. Methods A total of 571 sera from 132 Swiss farms (374 alpacas and 197 llamas, mean 4.3 animals/farm) were obtained. Four commercial enzyme-linked immunosorbent assays (ELISA) for detecting antibodies against T. gondii (ID Screen® Toxoplasmosis Indirect (TOXO-MS)) or N. caninum (i.e. ID Screen® Neospora caninum Indirect Multi-species (NCS-MS); ID Screen® Neospora caninum Competition (NCC) and ID Screen® Neospora caninum Indirect (NCS)) were first assessed for their use on SAC comparing their results with those in immunoblot, and optimizing cut-offs. Subsequently, two kits (TOXO-MS and NCS-MS) were selected for seroprevalence estimation. Additionally, a risk factor analysis for infection was performed on 41 farms, which agreed to participate in a web-based survey. Results Three kits (TOXO-MS, NCS-MS and NCC) showed almost perfect agreement (kappa > 0.901) with immunoblot results when the cut-offs were optimized, and one kit (NCS) proved not to be useful for detecting N. caninum seropositive SAC. By TOXO-MS ELISA, 82.3% (308/374) of the alpacas and 84.8% (167/197) of the llamas were seropositive for T. gondii, and 131/132 (99.2%) farms had seropositive animals. By NCS-MS ELISA, 3.5% (13/374) of the alpacas and 2.5% (5/197) of the llamas evidenced antibodies against N. caninum, and 9.1% (12/132) of the farms had seropositive animals. The variables “age” and “female sex” were identified as risk factors for T. gondii infection and “absence of cats in the farm during the last two years” as a protective factor. No risk or protective factors for N. caninum infection could be identified. Conclusions This nationwide cross-sectional study demonstrated for the first time the presence of antibodies against T. gondii and N. caninum in the Swiss SAC population, highlighting a high seroprevalence for T. gondii, the presence of cats as a risk factor and suggesting that SAC meat might represent an additional infection source for humans.

This study aimed to determine the pharmacokinetics of prednisolone following intravenous and oral administration in healthy adult alpacas. Healthy adult alpacas were given prednisolone (IV, n = 4), as well as orally (PO, n = 6). Prednisolone was administered IV once (1 mg/kg). Oral administration was once daily for 5 days (2 mg/kg). Each treatment was separated by a minimum 4 month washout period. Samples were collected at 0 (pre-administration), 0.083, 0.167, 0.25, 0.5, 0.75, 1, 2, 4, 8, 12, and 24 h after IV administration, and at 0 (pre-administration), 0.25, 0.5, 0.75, 1, 2, 4, 8, 12, 24 after the first and 5th PO administration. Samples were also taken for serial complete blood count and biochemistry analysis. Prednisolone concentration was determined by high pressure liquid chromatography. Non-compartmental pharmacokinetic parameters were then determined. After IV administration clearance was 347 mL/kg/hr, elimination half-life was 2.98 h, and area under the curve was 2,940 h*ng/mL. After initial and fifth oral administration elimination half-life was 5.27 and 5.39 h; maximum concentration was 74 and 68 ng/mL; time to maximum concentration was 2.67 and 2.33 h; and area under the curve was 713 and 660 hr*ng/mL. Oral bioavailability was determined to be 13.7%. Packed cell volume, hemoglobin, and red blood cell counts were significantly decreased 5 days after the first PO administration, and serum glucose was significantly elevated 5 days after the first PO administration. In conclusion, serum concentrations of prednisolone after IV and PO administration appear to be similar to other veterinary species. Future research will be needed to determine the pharmacodynamics of prednisolone in alpacas.

The domestic South American camelid Vicugna pacos L. is distributed along Peru, Chile, Bolivia, and Argentina. Here, we contribute to the bioinformatics and evolutionary systematics of the Camelidae by performing high-throughput sequencing analysis on the black Huacaya breed of V. pacos from Puno, Peru. The black Huacaya breed mitogenome is 16,664 base pairs (bp) in length and contains 37 genes (GenBank accession MT044302). The mitogenome shares a high-level of gene synteny to other Camelidae (Camelops, Camelus, Lama, and Vicugna). The mitogenome of the black Huacaya breed of V. pacos situates it in a clade with V. vicugna Molina, sister to Lama. We anticipate that further mitogenome sequencing of different breeds from Vicugna pacos will improve our understanding of the evolutionary history of this taxon.

Rift Valley fever (RVF) is a zoonotic, viral, mosquito-borne disease that causes considerable morbidity and mortality in humans and livestock in Africa and the Arabian Peninsula. In June 2018, 4 alpaca inoculated subcutaneously with live attenuated RVF virus (RVFV) Smithburn strain exhibited pyrexia, aberrant vocalization, anorexia, neurologic signs, and respiratory distress. One animal died the evening of inoculation, and 2 at ~20 d post-inoculation. Concern regarding potential vaccine strain reversion to wild-type RVFV or vaccine-induced disease prompted autopsy of the latter two. Macroscopically, both alpacas had severe pulmonary edema and congestion, myocardial hemorrhages, and cyanotic mucous membranes. Histologically, they had cerebral nonsuppurative encephalomyelitis with perivascular cuffing, multifocal neuronal necrosis, gliosis, and meningitis. Lesions were more severe in the 4-mo-old cria. RVFV antigen and RNA were present in neuronal cytoplasm, by immunohistochemistry and in situ hybridization (ISH) respectively, and cerebrum was also RVFV positive by RT-rtPCR. The virus clustered in lineage K (100% sequence identity), with close association to Smithburn sequences published previously (identity: 99.1–100%). There was neither evidence of an aberrant immune-mediated reaction nor reassortment with wild-type virus. The evidence points to a pure infection with Smithburn vaccine strain as the cause of the animals' disease. [ABSTRACT FROM AUTHOR]

Abstract Background This case report describes a focal brain lesion in an alpaca (Vicugna pacos). Although this is a restricted study based on a single animal, neuropathological features are reported that are most likely attributed to a vascular event with either ischemic or hemorrhagic pathology. Concerning translational issues, these findings extend neurovascular unit concept to the alpacas’ brain and qualify a larger panel of stroke tissue markers for further exploration of ischemic or hemorrhagic consequences beyond the usually used small animal models in stroke research. Case presentation A brain lesion indicative of a stroke was diagnosed in a 3-year-old female alpaca as an incidental finding during a post mortem examination. The rostral portion of the right frontal lobe contained a 1.0 × 1.5 × 1.7 cm lesion that extended immediately to the overlying leptomeninges. Microscopically, it was composed of liquefactive necrosis with cholesterol crystal deposition and associated granulomatous inflammation as well as vascularized fibrous connective tissue rimmed by proliferated astrocytes. Multiple fluorescence labeling of the affected brain regions revealed strong microgliosis as shown by immunostaining of the ionized calcium binding adapter molecule 1 and astrogliosis as demonstrated by enhanced immunoreactivity for glial fibrillary acidic protein. In parallel, a drastic neuronal loss was detected by considerably diminished immunolabeling of neuronal nuclei. Concomitantly, up-regulated immunoreactivities for collagen IV and neurofilament light chains were found in the affected tissues, indicating vascular and cytoskeletal reactions. Conclusions Driven by these neuropathological features, the incidental brain lesion found in this alpaca strongly suggests an ischemic or hemorrhagic etiology. However, since typical hallmarks became verifiable as previously described for other species affected by focal cerebral ischemia, the lesion is more likely related to an ischemic event. Nevertheless, as such cellular alterations might be difficult to distinguish from other brain lesions as for instance caused by inflammatory processes, adjuvant observations and species-related features need to be considered for etiological interpretations. Indeed, the lack of neurological deficits is likely attributed to the location of the lesion within the rostral aspect of the right frontal lobe of the alpacas’ brain. Further, fibroblast migration from the meninges likely caused the intralesional scar formation.

The article presents the second part of the series.The authors have focused on husbandry, breeding and feeding of alpacas. Alpacas must be well protected from rain, cold and high air temperature. When choosing this type of animals, you should consider how much profit you want to achieve from breeding. Will animals be treated as a source of income or a hobby? Alpacas live more than 20 years and their reproductive life is also long. Alpacas are not farm animals within the meaning of Polish regulations, and thus no additional permits or registration are required. However, possession of the herd should be reported to the appropriate County Veterinary Inspectorate. Alpacas are herd animals even the males, so it is strongly discouraged to isolate a single male from other alpacas. The herd gives them a sense of security that is fundamental to their welfare. Only within a herd can they live relaxed, graze peacefully, rest, sleep and build harmonious relationships. Lonely alpacas live under constant stress, which can lead to a serious illness or even death. They are pasture animals. 80-90% of alpacas food is roughage, i.e. hay, grass. It is quite difficult to create one perfect recipe for a good feeding program because a lot depends on the conditions on a given farm, on the quality of the soil, pasture and the hay quality.Here, we have also presented the breeding program and some aspects of veterinary surveillance for alpacas. This article describes the general features of the alpacas, a new breeding species in Poland. Alpaca (Vicugna pacos), belongs to the camelid family (Camelidae). It is a medium-sized animal with quite original body conformation. Its head is firmly set on a long neck and the body rests on slender limbs. The alpacas are bred mainly for their high quality natural fiber as well as for agritourism, recreation and zootherapy. Due to their easy anatomophysiological adaptation they can be bred in any climate. There are two breeds in the alpaca world population: about 85-90% are Huacaya and 10-15% are Suri. Good quality Huacaya and Suri fleece is dense, silky to the touch and has a beautiful luster. The annual fiber growth is from 6 to 12 cm. Alpaca fiber is resistant and keeps its form, so why the alpaca yarns and textiles may last for years. It is warmer than sheep's wool or bird's down, very soft in handle and being hypoallergenic it may be a great option for allergic people. Australia, USA, New Zealand and the United Kingdom, alongside Peru, Bolivia and Chile, have become in last 30 years the leading centres for alpacas breeding - both in quality of the breed and the export of animals to European countries or recently also to China. According to data from The British Alpaca Society, it is calculated that currently the largest European population of alpacas lives is in Great Britain and is about 35,000 individuals. Background: Little is known about the epidemiology of Toxoplasma gondii and Neospora caninum infections in alpacas (Vicugna pacos) and llamas (Lama glama) outside South America. The study aimed to estimate the seroprevalence of T. gondii and N. caninum infections in South American camelids (SAC) in Switzerland, to optimize serological tests for SAC and to identify risk factors, which may favour infection. Methods: A total of 571 sera from 132 Swiss farms (374 alpacas and 197 llamas, mean 4.3 animals/farm) were obtained. Four commercial enzyme-linked immunosorbent assays (ELISA) for detecting antibodies against T. gondii (ID Screen® Toxoplasmosis Indirect (TOXO-MS)) or N. caninum (i.e. ID Screen® Neospora caninum Indirect Multi-species (NCS-MS); ID Screen® Neospora caninum Competition (NCC) and ID Screen® Neospora caninum Indirect (NCS)) were first assessed for their use on SAC comparing their results with those in immunoblot, and optimizing cut-offs. Subsequently, two kits (TOXO-MS and NCS-MS) were selected for seroprevalence estimation. Additionally, a risk factor analysis for infection was performed on 41 farms, which agreed to participate in a web-based survey. Results: Three kits (TOXO-MS, NCS-MS and NCC) showed almost perfect agreement (kappa > 0.901) with immunoblot results when the cut-offs were optimized, and one kit (NCS) proved not to be useful for detecting N. caninum seropositive SAC. By TOXO-MS ELISA, 82.3% (308/374) of the alpacas and 84.8% (167/197) of the llamas were seropositive for T. gondii, and 131/132 (99.2%) farms had seropositive animals. By NCS-MS ELISA, 3.5% (13/374) of the alpacas and 2.5% (5/197) of the llamas evidenced antibodies against N. caninum, and 9.1% (12/132) of the farms had seropositive animals. The variables "age" and "female sex" were identified as risk factors for T. gondii infection and "absence of cats in the farm during the last two years" as a protective factor. No risk or protective factors for N. caninum infection could be identified. Conclusions: This nationwide cross-sectional study demonstrated for the first time the presence of antibodies against T. gondii and N. caninum in the Swiss SAC population, highlighting a high seroprevalence for T. gondii, the presence of cats as a risk factor and suggesting that SAC meat might represent an additional infection source for humans. Vγ9VΣ2 T cells are a major γΣ T cell population in the human blood expressing a characteristic Vγ9JP rearrangement paired with VΣ2. This cell subset is activated in a TCR-dependent and MHCunrestricted fashion by so-called phosphoantigens (PAgs). PAgs can be microbial [(E)-4-hydroxy-3-methyl-but-2-enyl pyrophosphate, HMBPP] or endogenous (isopentenyl pyrophosphate, IPP) and PAg sensing depends on the expression of B7-like butyrophilin (BTN3A, CD277) molecules. IPP increases in some transformed or aminobisphosphonate-treated cells, rendering those cells a target for Vγ9VΣ2 T cells in immunotherapy. Yet, functional Vγ9VΣ2 T cells have only been described in humans and higher primates. Using a genome-based study, we showed in silico translatable genes encoding Vγ9, VΣ2, and BTN3 in a few nonprimate mammalian species. Here, with the help of new monoclonal antibodies, we directly identified a T cell population in the alpaca (Vicugna pacos), which responds to PAgs in a BTN3-dependent fashion and shows typical TRGV9- and TRDV2-like rearrangements. T cell receptor (TCR) transductants and BTN3-deficient human 293T cells reconstituted with alpaca or human BTN3 or alpaca/human BTN3 chimeras showed that alpaca Vγ9VΣ2 TCRs recognize PAg in the context of human and alpaca BTN3. Furthermore, alpaca BTN3 mediates PAg recognition much better than human BTN3A1 alone and this improved functionality mapped to the transmembrane/cytoplasmic part of alpaca BTN3. In summary, we found remarkable similarities but also instructive differences of PAg-recognition by human and alpaca, which help in better understanding the molecular mechanisms controlling the activation of this prominent population of γΣ T cells. Cryptosporidiosis has been reported as an important cause of neonatal diarrhea and mortality in cattle, sheep, and other ruminants, but its impact on alpaca health has not been studied thoroughly. In this study, we have determined the prevalence and evaluated the role of cryptosporidiosis as a risk factor for diarrhea occurrence in newborn alpacas. During the calving season (January–March) of 2006, stool specimens (N = 1312) were collected from 24 herds of newborn alpacas in Puno and Cuzco, departments that account for the largest populations of alpacas in Peru. All the specimens were microscopically screened for Cryptosporidium spp. using the acid-fast technique. The association between Cryptosporidium detection and diarrhea was analyzed using χ2 test and generalized lineal model. Cryptosporidium species were determined by PCR-RFLP analysis of the small subunit rRNA gene. Cryptosporidium oocysts were detected in 159 of 1312 (12.4%) newborn alpacas. Results of the analyses demonstrated that crypstosporidiosis was significantly associated with diarrhea (PR = 3.84; CI95% 2.54–5.81; p < 0.0001). Only Cryptosporidium parvum was detected in the 153 Cryptosporidium-infected animals. Thus, there is an association of C. parvum infection with diarrhea in neonatal alpacas. [ABSTRACT FROM AUTHOR]

This article aims at the presentation of an epizootic problem of alpacas tuberculosis in our country. Tuberculosis is caused by bacteria of Mycobacterium genus, members of Mycobacterium Tuberculosis Complex group. Animal tuberculosis is usually caused by M. bovis and M. caprae - the bovine type bacilli that mainly affect cattle, bisons and pigs, however in recent years cases of tuberculosis in alpacas, an animal species belonging to the Camelidae family, have been reported with increased frequency. According to available literature, M. bovis infections prevail in this animal species. In addition, alpacas proved to be very sensitive to infection with M. microti, whose rodents are the natural reservoir. These features, along with the very frequent absence of clinical symptoms or their late occurrence, contribute to the rapid transmission of the disease among alpacas. Infection spreads by respiratory route. Usually the source of infection are asymptomatic carriers introduced to the tuberculosis-free herd. Alpaca tuberculosis is diagnosed by tuberculin skin test, supplemented with serological tests such as ELISA, EnferplexTM Camelid TB Assay, Dual path platform (DPP) assay or Multi-antigen print immunoassays (MAPIA). Currently, the epizootic situation of alpacas tuberculosis in Poland seems to be under control however, to maintain the surveillance it is necessary to amend Polish legislation accordingly. • High humidity and temperature of Taiwan diminishes alpaca productivity and welfare. • Quantifiable behavioral change may serve as early indicator of heat stress in alpaca. • Fans, mists, and especially pools are effective alpaca heat stress reducing tools. Despite their ability to adapt to a variety of environments, alpacas suffer from heat stress symptoms when raised in warm and humid regions of the world. Hyperthermia in alpacas not only raises animal welfare concerns, but also decreases productivity. To better understand the relationship between heat stress and alpaca mortality, Taiwanese alpaca facilities were surveyed. Additionally, to address the dearth of experimental research on heat stress reducing amenities in alpacas, an experiment was designed to test the effectiveness of different cooling amenities (water pool, frequent mist spraying, and fans) through temperature, behavioral, location and postural observations of alpacas at the Taipei Zoo during manipulations. Experimental result supports the use of mist sprinklers, water pools, and fans as alpaca cooling amenities, with water pools as being the most effective. Survey result shows a significant portion of alpaca deaths in Taiwan occurred during the summer and that heat stress was a leading cause of death. The domestic South American camelid Vicugna pacos L. is distributed along Peru, Chile, Bolivia, and Argentina. Here, we contribute to the bioinformatics and evolutionary systematics of the Camelidae by performing high-throughput sequencing analysis on the black Huacaya breed of V. pacos from Puno, Peru. The black Huacaya breed mitogenome is 16,664 base pairs (bp) in length and contains 37 genes (GenBank accession MT044302). The mitogenome shares a high-level of gene synteny to other Camelidae (Camelops, Camelus, Lama, and Vicugna). The mitogenome of the black Huacaya breed of V. pacos situates it in a clade with V. vicugna Molina, sister to Lama. We anticipate that further mitogenome sequencing of different breeds from Vicugna pacos will improve our understanding of the evolutionary history of this taxon. [ABSTRACT FROM AUTHOR]

Existen cuatro especies de camélidos sudamericanos, dos de ellos silvestres, guanaco (Lama guanicoe) y vicuña (Vicugna vicugna), y dos formas domésticas, alpaca (Lama pacos) y llama (Lama glama), cuyo origen ha sido objeto de debate. En el presente estudio la variación en el patrón de bandas G de los cromosomas de llamas y alpacas y la secuencia de dos genes mitocondriales han sido usados para estudiar el origen y la clasificación de llamas y alpacas. Patrones de bandas cromosómicas similares fueron observados en las cuatro especies de Lamini, incluso similares a los descritos para camello, Camelus bactrianus. Sin embargo, se encontraron finas y consistentes diferencias en los brazos cortos del cromosoma 1, permitiendo separar a camellos, guanacos y llamas, de las de vicuñas y alpacas. Este patrón fue consistente incluso en un híbrido guanaco x alpaca. Relaciones equivalentes fueron encontradas en las secuencias completas del gen para citocromo b, así como en el árbol de expansión mínima de las secuencias parciales de la región control, agrupando a guanacos con llamas y a vicuñas con alpacas. Los análisis filogenéticos mostraron a V. vicugna y a L. guanicoe como grupos recíprocamente monofHéticos. El análisis de las secuencias de ambos genes mostró dos ciados entre las vicuñas, concordantes con las subespecies reconocidas para esta especie, pero los resultados obtenidos para guanacos no reflejaron la existencia de las cuatro subespecies previamente propuestas. El análisis combinado de variaciones cromosómicas y moleculares demostraron una alta similitud genética entre alpacas y vicuñas, así como entre llamas y guanacos. Aunque se revela hibridización direccional, nuestros resultados apoyan fuertemente la hipótesis de que la llama se deriva de L. guanicoe, y la alpaca de V. vicugna, apoyando la reclasificación de la alpaca como V. pacosFour camelid species exist in South America: two wild, the guanaco (Lama guanicoe) and the vicuña (Vicugna vicugna), and two domestic, the alpaca (Lama pacos) and the llama (Lama glama). However, the origin of the domestic species has been a matter of debate. In the present study, variations in chromosome G banding patterns and in two mitochondrial gene sequences have been used to study the origin and classification of the llama and alpaca.-Similar patterns in chromosome G band structure were observed in all four Lamini species, and these in turn were similar to the bands described for camels, Camelus bactrianus. However, fine and consistent differences were found in the short arms of chromosome 1, separating camels, guanacos and llamas from vicuñas and alpacas. This pattern was consistent even in a hybrid guanaco x alpaca. Equivalent relationship showed the complete cytochrome b gene sequences, and the minimum expansion tree of the partial control region sequence, grouping guanaco with llama and vicuña with alpaca. Phylogenetic analyses showed V. vicugna and L. guanicoe as monophyletic groups. Analysis of both gene sequences revealed two clades within vicuña, concordant with the two described subspecies, but the results for guanaco did not confirm existence of the four previously proposed subspecies. The combined analysis of chromosomal and molecular variation showed close genetic similarity between alpacas and vicuñas, as well as between llamas and guanacos. Although directional hybridization was revealed, our results strongly support the hypothesis that the llama would have derived from L. guanicoe and the alpaca from V. vicugna, supporting reciassification as V. pacos

Epididymal alpaca sperm represent an alternative model for the study of alpaca semen. The objective of this study was to characterize the normal values of some functional variables in epididymal alpaca sperm using imaging flow cytometry. Alpaca testicles ( n = 150) were processed and sperm were recovered from the cauda epididymides. Only 76 samples with acceptable motility and sperm count were considered for assessment by imaging flow cytometry. Acrosome integrity and integrity/viability were assessed by FITC-PSA/PI and FITC-PNA/PI. Mitochondrial membrane potential (MMP) was assessed by MitoTracker CMXRos and MitoTracker Deep Red FM. Lipid peroxidation was evaluated using BODIPY 581/591 C11. Results show that the mean values for acrosome-intact sperm were 95.03 ± 6.39% and 93.34 ± 7.96%, using FITC-PSA and FITC-PNA, respectively. The mean values for acrosome-intact viable sperm were 60.58 ± 12.12% with FITC-PSA/PI and 58.81 ± 12.94% with FITC-PNA/PI. Greater MMP was detected in 65.03 ± 15.92% and 59.52 ± 19.19%, using MitoTracker CMXRos and MitoTracker Deep Red FM, respectively. Lipid peroxidation was 0.84 ± 0.95%. Evaluation of acrosome-intact and acrosome-intact viable sperm with FITC-PSA/PI compared with. FITC-PNA/PI or MMP with MitoTracker CMXRos compared with MitoTracker Deep Red FM were correlated ( P < 0.05). The MMP using MitoTracker CMXRos was the only variable correlated ( P < 0.05) with sperm motility ( r = 0.3979). Middle East respiratory syndrome coronavirus is a recently emerged pathogen associated with severe human disease. Zoonotic spillover from camels appears to play a major role in transmission. Because of logistic difficulties in working with dromedaries in containment, a more manageable animal model would be desirable. We report shedding and transmission of this virus in experimentally infected alpacas (n = 3) or those infected by contact (n = 3). Infectious virus was detected in all infected animals and in 2 of 3 in-contact animals. All alpacas seroconverted and were rechallenged 70 days after the original infection. Experimentally infected animals were protected against reinfection, and those infected by contact were partially protected. Necropsy specimens from immunologically naive animals (n = 3) obtained on day 5 postinfection showed virus in the upper respiratory tract. These data demonstrate efficient virus replication and animal-to-animal transmission and indicate that alpacas might be useful surrogates for camels in laboratory studies. [ABSTRACT FROM AUTHOR]

Background: Dysuria in camelids is usually associated with the presence of lower urinary tract disease such as urolithiasis. As another differential diagnosis, urine retention may be caused by neurological disturbances resulting from infections of the spinal cord, discospondylitis or trauma. Case presentation: A 2.5-year-old male Huacaya alpaca (Vicugna pacos) presented with dysuria due to damage of the lumbosacral intumescence of the spinal cord. On presentation the alpaca was recumbent. Clinical examination revealed abdominal pain, oliguria, leucopenia with neutrophilia, and slightly elevated creatinine kinase. Ultrasonography of the abdomen showed an irregularly shaped, dilated urinary bladder with hyperechoic serosa. Magnetic resonance imaging revealed discospondylitis of the fourth and fifth lumbar vertebrae and herniation of the intervertebral disc between these vertebrae and the spinal cord. Postmortem examination confirmed severe chronic purulent discospondylitis with ventral spondylosis and narrowing of the spinal canal. Urolithiasis could not be verified. Conclusion: Although rare, diseases of the spinal cord should be considered as a differential diagnosis for impaired micturition in camelids. [ABSTRACT FROM AUTHOR]
