The lichen Cetraria islandica (L.) Ach. has been used in traditional and modern medicines for its many biological properties such as immunological, immunomodulating, antioxidant, antimicrobial, and anti-inflammatory activities. This species is gaining popularity in the market, with interest from many industries for selling as medicines, dietary supplements, and daily herbal drinks. This study profiled the morpho-anatomical features by light, fluorescence, and scanning electron microscopy; conducted an elemental analysis using energy-dispersive X-ray spectroscopy; and phytochemical analysis was performed using high-resolution mass spectrometry combined with a liquid chromatography system (LC-DAD-QToF) of C. islandica. In total, 37 compounds were identified and characterized based on comparisons with the literature data, retention times, and their mass fragmentation mechanism/s. The identified compounds were classified under five different classes, i.e., depsidones, depsides, dibenzofurans, aliphatic acids, and others that contain simple organic acids in majority. Two major compounds (fumaroprotocetraric acid and cetraric acid) were identified in the aqueous ethanolic and ethanolic extracts of C. islandica lichen. This detailed morpho-anatomical, EDS spectroscopy, and the developed LC-DAD-QToF approach for C. islandica will be important for correct species identification and can serve as a useful tool for taxonomical validation and chemical characterization. Additionally, chemical study of the extract of C. islandica led to isolation and structural elucidation of nine compounds, namely cetraric acid (1), 9′-(O-methyl)protocetraric acid (2), usnic acid (3), ergosterol peroxide (4), oleic acid (5), palmitic acid (6), stearic acid (7), sucrose (8), and arabinitol (9).

This article describes the morpho-anatomies of the leaves and stems of Ocotea paranaensis Brotto, Baitello, CerviSantos (Lauraceae) using light and scanning electron microscopy. The main anatomical features characterizing the species are the presence of simple non-glandular trichomes in the leaves and stems, large secretory cells in the leaves, flat-convex petioles with two small lateral ribs, brachysclereids in the stem cortex and pith, and the presence of starch grains and various types of calcium oxalate crystals in the pith. Histochemical tests indicated the presence of lignin in stone cells, fibers, and xylem. Lipophilic contents were found in the secretory cells. Phenolic compounds were detected in the epidermis, hypodermis, phloem, and xylem. The present study's findings can contribute to the taxonomy and authentication of O. paranaensis. RESEARCH HIGHLIGHTS: This article is the first morpho-anatomical study of Ocotea paranaensis. Anatomy and histochemistry of the leaves and stems were studied by light microscopy, FESEM and EDS. The findings of this study would aid in the species identification and taxonomy.

This study provides a comprehensive account of the various types of calcium oxalate crystals found in the genus Baccharis and assesses the exceptional value of crystal macropatterns for the taxonomy of the genus. The morphotype, occurrence and chemical composition of the crystals found in the stems and leaves are studied. The 44 species included in this study were selected based on a broad phylogeny-based sampling covering seven subgenera and 31 sections. These species were chosen to represent all the main phylogenetic lineages of Baccharis; thus, the sampling also represents a comprehensive coverage concerning evolutionary significance for such a large and environmentally and economically important plant group. The samples were analyzed by light microscopy, scanning electron microscopy (SEM), and energy-dispersive X-ray spectroscopy (EDS). Several morphotypes of crystals, including druses, crystal sand, styloids and prisms, were present. Based on their chemical composition, the crystals were classified as pure calcium oxalate, mixtures of oxalates and sulfates, and mixtures of oxalates, sulfates and silica. The crystal macropatterns observed in this study aid in the species identification and provide novel data for the taxonomy of Baccharis.

This paper provides detailed morpho-anatomical characteristics of the leaves, stems and berries of Viscum album L. subsp. album (Santalaceae) growing on the branches of Malus domestica. Additionally, microchemical analyses of all tissues and Energy Dispersive X-Ray Spectroscopy (EDS) analyses of the calcium oxalate crystals are provided for the first time. The plant grows as a semi-parasite on the branches of host trees and shrubs; it develops as dichasium with pseudo-dichotomous branching stems and well-developed green leaves with parallel veins; the dioecious inflorescences usually consist of three flowers, with female flowers generating white fleshy berries, in which a seed is embedded in the mucilaginous mesocarp, normally containing two embryos. The analyzed leaves were isobilateral, amphistomatic, and showed straight anticlinal epidermal cell walls, thick cuticles with epicuticular wax crystalloids, and paracytic stomata. The midrib is flat on both sides and has a single vascular bundle, whereas the petiole is concave-convex in shape and contains five bundles. The stems show a primary structure with a ring of nine vascular bundles enclosing the pith. Calcium oxalate druses and cubic and quadrangular prisms were observed in different parts of the plant. The microscopic features of the tissues and structures are illustrated with light and scanning electron micrographs.

This research reports for the first time the anatomical characteristics of all species belonging to Baccharis subgenus Coridifoliae (Asteraceae). The anatomy and micro-morphology of aerial vegetative organs of ten species: B. albilanosa, B. artemisioides, B. bicolor, B. coridifolia, B. erigeroides, B. napaea, B. ochracea, B. pluricapitulata, B. scabrifolia, and B. suberectifolia are investigated by light and scanning electron microscopy. The number of secretory ducts, crystal morphology, presence or absence of conical nonglandular trichomes, leaves cross-section shape, margin morphology, anticlinal epidermal cell walls shape, and cuticle structure were identified as characters with diagnostic value for species. Similarity cluster analysis allows the formation of three groups based on a percentage of similarity between 45 and 84%. Some species showed differential characteristics as the presence of up to four secretory ducts in the midrib in B. albilanosa; smooth cuticles onboth sides of the leaf epidermis in B. erigeroides; flat midrib shape on both sides of the leaves in B. napaea; and convex–flat midrib shape in B. suberectifolia. The remaining species can be differentiated by a set of anatomical features. Anatomical and histochemical characteristics of stems and leaves provided data to support species identification.

This paper provides detailed anatomy and histochemistry of the leaves and stems ofEucalyptus tereticornisillustrated with brightfield and scanning electron microscopy. The key microscopic features that can aid in the species identification include the presence of crust-like epicuticular waxes on the leaf surfaces, platy aggregations of cluster crystals in the epidermal cells, presence of prismatic crystals in the epidermal cells, in the parenchymatous sheath in the blade and petiole, and in the cortex and pith of the stems, and leaf homogenous mesophyll consisting of palisade cells. Histochemical analyses confirmed the presence of lipophilic and phenolic compounds in the contents of secretory cavities, starch grains in the xylem parenchyma of the stem, and lignified elements in the sclerenchymatous ring adjoining the phloem and in the xylem in the leaves and stems.

This paper provides detailed morpho-anatomical and histochemical characteristics of the leaves, peduncles, stems, and roots of Eryngium yuccifolium Michx. (Apiaceae) by bright-field light and scanning electron microscopy. Noteworthy morpho-anatomical features include the monocot-like leaves with parallel venation and distant marginal bristles, amphistomatic leaves with paracytic or anisocytic stomata and heterogenous isobilateral mesophyll, the presence of epicuticular wax crystalloids on both leaf surfaces, calcium oxalate druses and secretory canals in all the studied plant parts, presence of hypodermis, stem and peduncle with longitudinal ribs containing collenchyma, presence of aerenchyma in the peduncle and root, and absence of sclerenchyma in all the plant parts analyzed. These characteristics would aid in the taxonomy, species identification as well as quality control of the botanical.

The raw materials of “licorice root” in the commerce consist of roots and/or rhizomes (stolons) of different species of Glycyrrhiza. Licorice products and raw materials are frequently mislabeled and often have mixed, misidentified, or unidentified species and parts. This paper provides a detailed comparative analysis of the morpho-anatomies of the rhizomes and roots of five species of Glycyrrhiza, namely G. glabra, G. uralensis, G. echinata, G. inflata, and G. lepidota, by bright-field light microscopy and scanning electron microscopy. The studied species showed some similarities in their basic anatomical features due to the fact that they are phylogenetically closely related and belong to the same genus. However, differences in microscopic features such as the thickness of cork and medullary rays, pore frequency, and size of the vessels were observed. The rhizomes can readily be distinguished by the presence of a distinct pith. The roots lack a well-defined pith and instead have primary xylem in the center.

The roots and rhizomes of several Glycyrrhiza species are widely used as sweetening and flavoring agents in food, as well as important ingredients in formulations of traditional medicines. Five Glycyrrhiza species, G. uralensis, G. glabra, G. inflata, G. echinata, and G. lepidota, often share the name "licorice roots" in the botanicals' marketplace. Unfortunately, misidentification/mislabeling is very common due to their similarities in morpho-anatomical features. Significant metabolite alterations among the different Glycyrrhiza species and their hybrids have been reported, suggesting that the biological activities could vary with the licorice roots or products derived from different species. Development of simple, effective methods for species identification and differentiation is of key importance. In this study, 78 licorice samples were investigated using HPTLC and NMR as analytical tools. Significant metabolite variations were observed between the five species. The species-specific fingerprint patterns for the five Glycyrrhiza species were determined with HPTLC and NMR; then applied to the sample identification and discrimination. The results obtained from these two orthogonal analytical methods agreed with each other. Furthermore, the NMR signals and the species-specific constituents that made significant contributions to the differentiation of the five Glycyrrhiza species were confirmed based on the multivariate analysis of the NMR spectral data. Using the established OPLS-DA models, the classification of hybrids was evaluated and confirmed. The developed methods, particularly the HPTLC method with its simplicity and low cost, could be used as a rapid and reliable approach for the authentication of licorice species and quality control of licorice raw material and products.

This paper studied the chemical profile, cytotoxicity, and insecticidal activity of the volatile oil, as well as provided a detailed anatomy of the leaves and stems from Eugenia involucrata DC., Myrtaceae. Its volatile oil was identified by gas chromatography coupled with mass spectrometry. The insecticidal activity was performed using the volatile oil against two strains of Cimex lectularius L., using topical, residual, and fumigant methods. Cytotoxicity was verified against B16F10, MCF7, and McCoy cell lines. The cytotoxic mechanism was studied by the morphology feature changes. The microscopic analysis was performed by usual microscopy techniques. The main volatile compounds of E. involucrata were germacrene B (22.17%), bicyclogermacrene (19.76%), and β-elemene (10.86%). Volatile oil demonstrated low toxicity against two strains of the bed bug in the topical test and did not present toxicity in fumigation and residual methods. Cytotoxicity for the B16F10 murine melanoma cells was demonstrated. Moreover, the volatile oil induces morphological characteristic changes of the cell death process of apoptosis. The microscopic data of the leaf and stem provided information to support the identification of E. involucrata.

The State of Parana has a diversity of plants that are used in the traditional medicine and among them are some of the representatives of the Piperaceae family. Piper is the one of the most important genera of this family and its species show similar morphology and different biological activities. The literature shows that approximately 54 species of Piper have been described in the State of Parana. The purpose of this study is to gather key information by means of electronic databases, about the morphoanatomy and biological activities of species of Piper found in the State of Parana. The results with regard to the morphoanatomy show that, on the whole, the species that have been described possess hypostomatic leaves, non-glandular and glandular trichomes, and calcium oxalate crystals in the leaves and stems. Several biological activities have been reported, including antimicrobial, anti-inflammatory, antifungal, digestive, diuretic, insecticidal, leishmanicidal, pesticide and trypanocide.

Phytochemical investigation of the methanolic extract of Vangueria agrestis aerial parts led to the isolation and characterization of four monoterpene rhamnosides, two ursane saponins, and six flavonoid glycosides. Of these constituents, are three undescribed monoterpene glycosides, vangagrestosides A-C. The reported constituents comprise the monoterpene glycoside [(2E,6Z)-2,6-dimethyl-8-[O-α-L-rhamnopyranosyl-(1→3)-α-L-rhamnopyranosyl)-oxy]-octadien-1-yl α-L-rhamnopyranoside], the saponins [3-O-[α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl]pomolic acid, and 3-O-[α-L-rhamnopyranosyl-(1→2)-β-D-xylopyranosyl]pomolic acid], and the flavonoids [narcissoside, rutin, hyperoside, guajavarin, biorobin, and nicotifloroside].

This article enumerates the detailed anatomy of Cantinoa althaeifolia (Lamiaceae) illustrated with light and scanning electron microscopy images. The anatomical markers include the presence of branched nonglandular trichomes and capitate, peltate, and clavate types of glandular trichomes; prismatic crystals on the leaf and stem surfaces; and oil droplets in the leaf mesophyll. Histochemical tests and EDS analyses were performed in order to detect the composition of certain cells and their contents. The key findings of the present study can contribute to the taxonomy, species identification, and quality control of Cantinoa althaeifolia.

Eucalyptus species possess anti-inflammatory, antifungal, antibacterial, and insecticidal properties. In this study, the chemical composition and biological activities of Eucalyptus cinerea essential oil (EO) and the leaf and stem anatomy were investigated. EO was extracted by Clevenger apparatus and the compounds were identified by GC/MS. The antioxidant activity was evaluated by DPPH, ABTS, and reducing phosphomolybdenum complex. Broth microdilution was used to determine antimicrobial activity. Cytotoxicity was verified against HeLa, HRT-18, and Calu-3 cells by MTT assay. The cytotoxic mechanism was studied by cell DNA content, cell cycle, and DNA fragmentation. The microscopic analyzes of the leaves and the stems were performed by light microscopy, field emission scanning electron microscopy, and energy-dispersive X-ray spectroscopy. The main constituent of the EO was 1,8-cineole (55.24%). The EO showed low antioxidant and antimicrobial activities. Calu-3 cells showed a significant reduction in viability with IC50 of 689.79 ± 29.34 μg/mL. EO at 1000 μg/mL decreased the DNA content in Jurkat cells. In general, EO increased cell percentage in sub-G0 and S phases with concomitant reduction of cell percentage in G0/G1 and G2/M phases and provided DNA fragmentation of 29.73%. Anatomical and micromorphological features of the leaves and stems can help in the species identification and its differentiation from other Eucalyptus species.

The essential oil of Lavandula dentata L. possesses sedative, antioxidant, antibacterial, antifungal, carminative, antidepressant, and cytotoxic properties. The aims of this study were to investigate the cytotoxic effects of L. dentata essential oil on Calu-3 lung cancer cells in both vapor and liquid phases, and to provide further insights into its cytotoxic mechanisms. Usual MTT, SRB, and DNA-PI assays were used for evaluating cytotoxicity. In addition, LDH, acridine orange with ethidium bromide, Hoechst 33342, propidium iodide, Rhodamine 123, and Annexin V analyses were performed to explore the cytotoxic mechanisms. Rhodamine 123 accumulation assay was used for evaluating P-glycoprotein inhibitory potential. L. dentata essential oil provided a significant reduction of cell viability for all cytotoxic experiments by reaching 84% of cytotoxicity in vapor phase and showed a time-dependent profile. Both necrosis and apoptosis were involved in Calu-3 cell death. However, necrosis appeared to be the dominant cell death pathway. L. dentata essential oil did not inhibit P-glycoprotein.

Fadogia agrestis Schweinf. ex Hiern (Vangueria agrestis (Schweinf. ex Hiern) Lantz), Rubiaceae, is an African traditional medicinal plant also used as a dietary supplement in the US. The present paper is the first report of the pharmacognostic study of the leaf, stem and root of F. agrestis by microscopy, HPTLC and total phenolic/flavonoid content analyses. Noteworthy microscopic features that can help in identification and quality control are septate and lignified non-glandular trichomes on leaf and stem epidermises, paracytic stomata on leaf abaxial epidermis, numerous cells containing yellow substances of presumably phenolic compounds in leaf and stem, calcium oxalate druses and prismatic crystals in leaf and styloids in stem, primary phloem fibers in stem, brachysclereids in stem and root, spherical starch grains in root, and vessels with vestured pits and simple perforated end walls. In addition to microscopy, a total phenolic/flavonoid content determination and an HPTLC method were also developed for rapid chemical fingerprint analyses of Fadogia samples and dietary supplements. Two new epimeric bibenzylated monoterpenes machaerifurogerol (1a) and 5-epi-machaerifurogerol (1b), and four known isoflavonoids (+)-vestitol (2), 7-O-methylvestitol (3), (+)-medicarpin (4), and 3,8-dihydroxy-9-methoxypterocarpan (5) were isolated from Machaerium Pers. This plant was previously assigned as Machaerium multiflorum Spruce, from which machaeriols A-D (6–9) and machaeridiols A-C (10–12) were reported, and all were then re-isolated, except the minor compound 9, for a comprehensive antimicrobial activity evaluation. Structures of the isolated compounds were determined by full NMR and mass spectroscopic data. Among the isolated compounds, the mixture 10 + 11 was the most active with an MIC value of 1.25 μg/mL against methicillin-resistant Staphylococcus aureus (MRSA) strains BAA 1696, −1708, −1717, −33591, and vancomycin-resistant Enterococcus faecium (VRE 700221) and E. faecalis (VRE 51299) and vancomycin-sensitive E. faecalis (VSE 29212). Compounds 6–8 and 10–12 were found to be more potent against MRSA 1708, and 6, 11, and 12 against VRE 700221, than the drug control ciprofloxacin and vancomycin. A combination study using an in vitro Checkerboard method was carried out for machaeriols (7 or 8) and machaeridiols (11 or 12), which exhibited a strong synergistic activity of 12 + 8 (MIC 0.156 and 0.625 µg/mL), with >32- and >8-fold reduction of MIC’s, compared to 12, against MRSA 1708 and −1717, respectively. In the presence of sub-inhibitory concentrations on polymyxin B nonapeptide (PMBN), compounds 10 + 11, 11, 12, and 8 showed activity in the range of 0.5–8 µg/mL for two strains of Acinetobacter baumannii, 2–16 µg/mL against Pseudomonas aeruginosa PAO1, and 2 µg/mL against Escherichia coli NCTC 12923, but were inactive (MIC > 64 µg/mL) against the two isolates of Klebsiella pneumoniae.

Ethnopharmacological relevance Cecropia pachystachya Trecul (Urticaceae) is a medicinal plant popularly known as ‘embauba’. In Brazil, the leaves of this species are used for the treatment of various kidney and cardiovascular diseases. However, there are no detailed studies on the renal and cardiovascular activities of this species. No studies on the anatomy or the quality control of this herbal drug is available thus far. Aim This study was aimed to investigate the ethnopharmacological properties of the leaves of C. pachystachya. Material and methods The leaves of C. pachystachya were analyzed by light and scanning electron microscopy for pharmacobotanical and anatomical characterization. The ethanol-soluble fraction of C. pachystachya leaf extract (ESCP) was characterized by high-performance liquid chromatograph equipped with diode array detector and mass spectrometry (HPLC-DAD-MS). The acute oral toxicity of ESCP on female Wistar rats was assessed. The acute and prolonged diuresis and antioxidant effects of ESCP (30, 100, and 300 mg/kg) were evaluated in male Wistar rats. In addition, the hypotensive effects of the ESCP as well as the vasodilatory activity in isolated and perfused mesenteric vascular beds were investigated. Results The anatomical markers obtained in this study can help in the identification of C. pachystachya, as well as to distinguish it from the other ‘embaubas’. The metabolites found in the ESCP were phenolic compounds, mainly C- and O-glycosylated flavonoids. The ESCP did not exhibit any toxic effects at a dose of 2000 mg/kg. Significant diuretic activities were observed at the doses of 30, 100, and 300 mg/kg. In addition, a significant modulating activity of the tissue redox state was observed after prolonged treatment. On the other hand, no hypotensive or vasodilator activity was observed. Conclusion The key findings of the present study can contribute to the taxonomy, species identification and quality control of C. pachystachya. Chemical studies have shown the presence of glycosylated flavonoids, phenylpropanoid derivative and proanthocyanidins. The pharmacological studies showed significant diuretic and antioxidant effects of C. pachystachya leaf extract, indicating a possible validation of its popular medicinal use.

Ocotea porosa (Nees & Mart.) Barroso, commonly known as “imbuia”, “canela-imbuia” or “imbuia-amarela” in Brazil, is a tree of the Southern Atlantic Forest. The present study investigates the anatomy of leaf and stem, volatile oil chemistry, as well as cytotoxicity and insecticidal activities of the essential oil of O. porosa. Species identification was achieved by anatomy features, mainly due to paracytic and anomocytic stomata; non-glandular trichomes; biconvex midrib and petiole with a collateral open arc vascular bundle; presence of a sclerenchymatous layer, starch grains and crystal sand in the stem; and the presence of phenolic compounds in the epidermis, phloem and xylem of the midrib, petiole and stem. The main volatile components of the essential oil were α-pinene (19.71%), β-pinene (13.86%) and bicyclogermacrene (24.62%). Cytotoxicity against human cancer cell (MCF-7), mouse cancer cell (B16F10) and mouse non-tumoral cell (McCoy) was observed as well as insecticidal activity of the essential oil against susceptible ‘Ft. Dix’ bed bugs (Cimex lectularius L.) by topical application.

This study aimed to investigate the anatomy and histochemistry of Mollinedia clavigera leaves and stems through photonic microscopy and scanning electron microscopy. Noteworthy features of leaves were: presence of paracytic stomata on both surfaces; simple as well as bifurcate non-glandular trichomes; prismatic calcium oxalate crystals; flat-convex midrib with a central and two dorsal bundles; concave-convex petiole with a single vascular bundle in open archh. Stems were cylindrical and showed prismatic and styloid crystals in the pith. Histochemical analysis detected lipophilic and phenolic compounds, starch grains and lignified elements such as brachysclereids and fibers. These features may assist in future identifications and quality control of M. clavigera, avoid misidentification between other genus members, once species and genus studies are scarce.

The present study examined the effect of pyrolysis temperature on the physicochemical properties of biochar, activation process and carbon capture. Two different categories of biochars were synthesized from herbaceous (miscanthus and switchgrass) or agro-industrial (corn stover and sugarcane bagasse) feedstock under four different pyrolysis temperatures- 500, 600, 700 and 800 oC. The synthesized biochars underwent sono-amination activation comprising low-frequency acoustic treatment followed by amine functionalization to prepare adsorbents for CO2 capture. As per the elemental analysis, the elevated pyrolysis temperature resulted in increased %C and %ash contents and reduced %N contents of biochar. The textural analysis exhibited almost 3-times enhancement of micro surface area and pore volume upon increasing the pyrolysis temperature from 500 to 700 oC, though further increase to 800 oC reduced the micro-porosity and the surface area. The intermediate temperatures of 600 and 700 oC revealed the highest interactions with ultrasound-amination, which significantly intensified CO2 adsorption. Accordingly, the CO2 capture capacity of sono-aminated biochars synthesized at 600 and 700 oC were almost 200% greater than that of raw biochars. There were 127-159% and 115-151% increases in adsorption capacity of biochars synthesized at 800 and 500 oC upon ultrasono-amine functionalization. Miscanthus biochar synthesized at 700 oC and treated sono-chemically demonstrated the highest adsorption ability of 2.89 mmol/g at 70 oC and 0.10 atm partial pressure, which is 211% higher than its pristine condition. The superior adsorption capacity of miscanthus (at 700 oC) can be attributed to its large surface area (324.35 m2/g), high carbon content (84%), and low ash content (4.9%), as well as its %N content after sono-amination that was twice that of raw char.

Background: As part of the efforts to find natural alternatives for cancer treatment and to overcome the barriers of cellular resistance to chemotherapeutic agents, polymeric nanocapsules containing curcumin and/or methotrexate were prepared by an interfacial deposition of preformed polymer method. Methods: Physicochemical properties, drug release experiments and in vitro cytotoxicity of these nanocapsules were performed against the Calu-3 lung cancer cell line. Results: The colloidal suspensions of nanocapsules showed suitable size (287 to 325 nm), negative charge (&minus, 33 to &minus, 41 mV) and high encapsulation efficiency (82.4 to 99.4%). Spherical particles at nanoscale dimensions were observed by scanning electron microscopy. X-ray diffraction analysis indicated that nanocapsules exhibited a non-crystalline pattern with a remarkable decrease of crystalline peaks of the raw materials. Fourier-transform infrared spectra demonstrated no chemical bond between the drug(s) and polymers. Drug release experiments evidenced a controlled release pattern with no burst effect for nanocapsules containing curcumin and/or methotrexate. The nanoformulation containing curcumin and methotrexate (NCUR/MTX-2) statistically decreased the cell viability of Calu-3. The fluorescence and morphological analyses presented a predominance of early apoptosis and late apoptosis as the main death mechanisms for Calu-3. Conclusions: Curcumin and methotrexate co-loaded nanocapsules can be further used as a novel therapeutic strategy for treating non-small-cell lung cancer.
