Abstract

The biomedical research community addresses reproducibility challenges in animal studies through standardized nomenclature, improved experimental design, transparent reporting, data sharing, and centralized repositories. The ARRIVE guidelines outline documentation standards for laboratory animals in experiments, but genetic information is often incomplete. To remedy this, we propose the Laboratory Animal Genetic Reporting (LAG-R) framework. LAG-R aims to document animals genetic makeup in scientific publications, providing essential details for replication and appropriate model use. While verifying complete genetic compositions may be impractical, better reporting and validation efforts enhance reliability of research. LAG-R standardization will bolster reproducibility, peer review, and overall scientific rigor.

Abstract

Azoospermia (the complete absence of spermatozoa in the semen) is a common cause of male infertility. The etiology of azoospermia is poorly understood. Whole-genome analysis of azoospermic men has identified a number of candidate genes, such as the X-linked testis-expressed 11 (TEX11) gene. Using a comparative genomic hybridization array, an exonic deletion (exons 10–12) of TEX11 had previously been identified in two non-apparent azoospermic patients. However, the putative impact of this genetic alteration on spermatogenesis and the azoospermia phenotype had not been validated functionally. We therefore used a CRISPR/Cas9 system to generate a mouse model (Tex11Ex9-11del/Y) with a partial TEX11 deletion that mimicked the human mutation. Surprisingly, the mutant male Tex11Ex9-11del/Y mice were fertile. The sperm concentration, motility, and morphology were normal. Similarly, the mutant mouse line's testis transcriptome was normal, and the expression of spermatogenesis genes was not altered. These results suggest that the mouse equivalent of the partial deletion observed in two infertile male with azoospermia has no impact on spermatogenesis or fertility in mice, at least of a FVB/N genetic background and until 10 months of age. Mimicking a human mutation does not necessarily lead to the same human phenotype in mice, highlighting significant differences species. [ABSTRACT FROM AUTHOR]

Abstract

Background Atypical/Nor98 scrapie (AS) is an idiopathic infectious prion disease affecting sheep and goats. Recent findings suggest that zoonotic prions from classical bovine spongiform encephalopathy (C-BSE) may copropagate with atypical/Nor98 prions in AS sheep brains. Investigating the risk AS poses to humans is crucial. Methods To assess the risk of sheep/goat-to-human transmission of AS, we serially inoculated brain tissue from field and laboratory isolates into transgenic mice overexpressing human prion protein (Met129 allele). We studied clinical outcomes as well as presence of prions in brains and spleens. Results No transmission occurred on the primary passage, with no clinical disease or pathological prion protein in brains and spleens. On subsequent passages, 1 isolate gradually adapted, manifesting as prions with a phenotype resembling those causing MM1-type sporadic Creutzfeldt-Jakob disease in humans. However, further characterization using in vivo and in vitro techniques confirmed both prion agents as different strains, revealing a case of phenotypic convergence. Importantly, no C-BSE prions emerged in these mice, especially in the spleen, which is more permissive than the brain for C-BSE cross-species transmission. Conclusions The results obtained suggest a low zoonotic potential for AS. Rare adaptation may allow the emergence of prions phenotypically resembling those spontaneously forming in humans. Abstract

The kinesin light chain 3 protein (KLC3) is the only member of the kinesin light chain protein family that was identified in post-meiotic mouse male germ cells. It plays a role in the formation of the sperm midpiece through its association with both spermatid mitochondria and outer dense fibers (ODF). Previous studies showed a significant correlation between its expression level and sperm motility and quantitative semen parameters in humans, while the overexpression of a KLC3-mutant protein unable to bind ODF also affected the same traits in mice. To further assess the role of KLC3 in fertility, we used CRISPR/Cas9 genome editing in mice and investigated the phenotypes induced by the invalidation of the gene or of a functional domain of the protein. Both approaches gave similar results, i.e. no detectable change in male or female fertility. Testis histology, litter size and sperm count were not altered. Apart from the line-dependent alterations of Klc3mRNA levels, testicular transcriptome analysis did not reveal any other changes in the genes tested. Western analysis supported the absence of KLC3 in the gonads of males homozygous for the inactivating mutation and a strong decrease in expression in males homozygous for the allele lacking one out of the five tetratricopeptide repeats. Overall, these observations raise questions about the supposedly critical role of this kinesin in reproduction, at least in mice where its gene mutation or inactivation did not translate into fertility impairment.Although the kinesin light chain 3 protein has been proposed to play a critical role in midpiece formation and sperm parameters in humans and mice, mutation or inactivation of its encoding gene has no adverse effect on mouse fertility.Graphical Abstract

Synthèse sur la détection des produits issus des nouvelles technologies génomiques (NGT) appliquées aux plantesLa question du statut réglementaire des NGT (New Genomic Techniques), qui ont été définies comme techniques de modification du génome « apparues » depuis 2001 , est actuellement débattue en Europe. Ces techniques n’existaient pas, ou étaient peu développées lors de l’adoption des directives et règlements européens qui encadrent actuellement les organismes génétiquement modifiés (OGM). A l’époque le législateur européen avait choisi de réglementer les techniques de transgénèse et d’exempter les méthodes de mutagénèse notamment sur la base d’une expérience d’usage n’ayant révélé aucun problème particulier. Certaines NGT pouvant être utilisées pour induire une mutagenèse, ciblée ou non , la questionde leur soumission à la réglementation européenne en vigueur fait l’objet de discussions juridiques et politiques depuis quelques années. Les données scientifiques relatives à ces technologies (définitions, méthodes et résultats) doivent permettre de guider ces discussions.

Abstract

Le Haut Conseil des biotechnologies (HCB) a été saisi le 22 juin 2021 par les autorités compétentes françaises (Ministère de l'Agriculture et de l'Alimentation) d'une demande d'avis relative au dossier EFSA-GMO-NL-2020-172 de demande d'autorisation de mise sur le marché du maïs génétiquement modifié DP915635 à des fins d'importation, transformation et alimentation humaine et animale. Ce dossier a été déposé par la société Pioneer Hi-Bred International, Inc. auprès des autorités compétentes néerlandaises sur le fondement du règlement (CE) n° 1829/2003. Dans le cadre de ce règlement, l'évaluation des dossiers de demande de mise sur le marché est confiée à l'Autorité européenne de sécurité des aliments (EFSA). Les Etats membres disposent de trois mois pour envoyer leurs commentaires à l'EFSA en contribution à l'évaluation du dossier. Dans ce contexte, le HCB est invité à proposer des commentaires à destination de l'EFSA au plus tard le 07 septembre 2021.

Abstract

Abstract We recently reported the site-independent and copy-number-related expression in mice of a goat α-lactalbumin gene with 150 kb and 10 kb of 5'- and 3'-flanking sequences, respectively. In the present study, we observed that the resection of the 5'-flanking region, leaving only 70 kb, resulted in a site-dependent expression of this milk protein-encoding transgene. This suggests that important cis-regulatory elements are located within the distal-deleted sequence. Within this region, we localised the promoter of the cyclin T1 gene, an ubiquitously expressed gene. So far, no other gene has been located between these two loci. Since these two genes are differentially expressed, our data suggest the potential location of an insulator within the deleted region that allows the two genes to be independently regulated.

Abstract

Scrapie in small ruminants belongs to transmissible spongiform encephalopathies (TSEs), or prion diseases, a family of fatal neurodegenerative disorders that affect humans and animals and can transmit within and between species by ingestion or inoculation. Conversion of the host-encoded prion protein (PrP), normal cellular PrP (Pr[P.sup.c]), into a misfolded form, abnormal PrP (Pr[P.sup.Sc]), plays a key role in TSE transmission and pathogenesis. The intensified surveillance of scrapie in the European Union, together with the improvement of Pr[P.sup.Sc] detection techniques, has led to the discovery of a growing number of so-called atypical scrapie cases. These include clinical Nor98 cases first identified in Norwegian sheep on the basis of unusual pathological and Pr[P.sup.Sc] molecular features and 'cases' that produced discordant responses in the rapid tests currently applied to the large-scale random screening of slaughtered or fallen animals. Worryingly, a substantial proportion of such cases involved sheep with PrP genotypes known until now to confer natural resistance to conventional scrapie. Here we report that both Nor98 and discordant cases, including three sheep homozygous for the resistant Pr[P.sup.ARR] allele ([A.sub.136][R.sub.154][R.sub.171]), efficiently transmitted the disease to transgenic mice expressing ovine PrP, and that they shared unique biological and biochemical features upon propagation in mice.
