Today, about 74% of medications are taken orally, and many people do not find them as effective as they would like. To improve these properties, transdermal drug delivery systems have been developed. The deck system of the transition drug includes the production of pharmaceuticals on the skin to have a topical therapy effect, and the largest of the drug is transported by systematic circulation and quality. The modern administration of therapies offers a lot of advantages compared to the traditional methods of recovering oral and invasive drugs. In addition to transitional medicines, other than the drug, the first metabolism and therapeutic efficiency increases and maintains its normal condition.

The aim of the present research is to formulate Sertraline Hydochloride solid dispersion (SRT-SD) fast disintegrating tablets. The speed of onset of action of antidepressant drug is clinically important for several reasons. Fast disintegrating tablets of SRT-SD was formulated by using natural superdisintegrant adopting direct compression method to minimize time of onset of action and also become economic. The study describes formulation of solid dispersion of SRT using HPMC E5 as a carrier and further converting SRT-SD into fast disintegrating tablets utilizing Plantago ovata mucilage as natural superdisintegrant. All tablets formulations were evaluated for various pre and post compression parameters. The F1 batch showed release kinetics closest to Serta® with maximum r2 (0.9991) value was considered to be the optimized batch and showed hardness of 3.10 kg/cm2, disintegration time of 28.00 second and dissolution of 95 % in 12 min. The stability study clearly indicate that optimized batch can increase the disintegrating and dissolution time of SRT and utilized as prominent alternative for the treatment of depression.

Drug delivery via the oral mucous membrane is considered to be a promising alternative to the oral route. Sublingual route is a useful when rapid onset of action is desired with better patient compliance than oraly ingested tablets. In terms of permeability, the sublingual area of the oral cavity (i.e. the floor of the mou) is more permeable than the buccal (cheek) area, which in turn is more permeable than the palatal (roof of the mouth) area. A rapid onset of pharmacological effect is often desired for some drugs, especially those used in the treatment of acute disorders. Sublingual tablets disintegrate rapidly and the small amount of saliva present is usually sufficient for achieving disintegration of the dosage form coupled with better dissolution and increased bioavailability.

Mannich bases identified by Professor Carl Mannich have been the most extensively explored scaffolds for more than 100 years now. The versatile biological roles that they play have promoted their applications in many clinical conditions. The present review highlights the application of Mannich bases as cytotoxic agents, categorizing them into synthetic, semisynthetic, and prodrugs classes, and gives an exhaustive account of the work reported in the last two decades. The methods of synthesis of these cytotoxic agents, their anti-cancer potential in various cell lines, and promising leads for future drug development have also been discussed. Structure-activity relationships, along with the targets on which these cytotoxic Mannich bases act, have been included as well.

Aim of the present investigation was to assess and compare the in-vitro and in-vivo cancer targeting propensity of DPPE–FA–DOX Micelles and free DOX in tumor bearing BALB/c mice. The DOX was conjugated with 1, 2-Dihexadecanoyl-sn-glycero-3-phosphoethanolamin (DPPE) and folic acid using Di-cyclohexyl-carbodiimide, confirmed by Fourier transform infrared spectroscopy (FTIR) and proton NMR. DPPE–FA–DOX micelles were prepared using thin film method and evaluated for zeta potential, particle size, surface morphology, in- vitro drug release study etc. In-vitro anticancer activity and apoptosis assay was evaluated in breast cancer (MCF-7) cells using MTT assay and flow cytometer respectively. In-vivo biodistribution and toxicity assessment were evaluated in rats whereas antitumor activity in tumor bearing BALB/c mice. Prepared micelles were spherical with size and zeta potential of 295.6 + 84.4 nm and 0.8 ± 0.24 mV respectively. Apoptosis assay for DPPE–FA–DOX micelles treated cells using Annexin V/PI staining demonstrated 56.2% apoptotic cells. Remarkably, DPPE–FA–DOX micelles improved DOX bioavailability by 7 fold and diminished plasma elimination with no sign of tissue toxicity compared to free DOX. In-vivo biodistribution studies revealed that micelles facilitated higher accumulation of DOX in tumor than free DOX. DPPE–FA–DOX micelles treated mice survived for 62 days than Free DOX (40 days), revealed by Kaplan-Meier survival curve analysis. Histopathological examination of liver, kidney and heart tissues of micelles treated rat’s corroborated reduced systemic toxicity than free DOX. Conclusively, DPPE–FA–DOX micelles could potentially facilitate the targeted delivery of DOX to tumors.

The main aim of the study is to formulate and evaluate hydrogel containing azithromycin and silver nanoparticles to treat wound dressing, burn infections, acne, Pharyngitis and Tonsillitis, psoriasis, etc. The Silver nanoparticles were prepared by green synthesis approach with the help of reduction of silver nitrate by Azadirachta indica leaves extract. Azithromycin was chosen as antimicrobial drug, however it shows adverse effect when administered in high dose, hence to reduce dose of azithromycin it was decided to make formulation containing silver nanoparticles and azithromycin. The drug loading and drug delivery techniques were searched for topical as well as bone grafting purpose. Initially mixing of Silver nanoparticles, Azithromycin and Bis acrylamide in ammonium persulphate were done and added N, N dimethyl amino ethyl methacrylate (DEM) to form semisolid mass that is hydrogel. Hydrogel formulation was found fantastic results with azithromycin and silver nanoparticles. The compatibility study was done for silver nanoparticles with polymer and azithromycin and all are found compatible with each other. All the formulations were characterized for consistency, spreadability, efficacy, safety and stability. From all F1-F12 Hydrogel Formulations F4 batch was optimized by parameters like pH measurement, spreadability, Drug content, Drug release and antimicrobial assay. So F4 batch was further evaluated for SEM, XRD, FTIR, skin Irritation study and showed good results. Hydrogel formulation of azithromycin and silver nanoparticles formulated for Azithromycin as a antimicrobial drug, however it shows adverse effect when administered in high dose, hence to reduce dose of azithromycin it was decided to make formulation containing silver nanoparticles and azithromycin. It was concluded that formulation was compatible with animal as well as human being with excellent evaluation results.
