Objective: This study examined saccadic reaction time (SRT) in visually guided saccades according to age and its relationship with attention and working memory. Materials and Methods: The study participants were divided into three groups: young adulthood (18–39 years), middle adulthood (40–59 years old), and older age groups (over 60 years). A total of 85 participants, including 20 young aged, 26 middle aged, and 39 older aged, participated in the study. SRT was recorded using the EyeLink 1000 Plus eye tracker, and 32 trials were conducted. In addition, neuropsychological tests assessing attention and working memory including the Trail Making Test (TM), Digit Span (DS), and Stroop test were applied to the participants. Results: SRTs were prolonged in the middle adulthood (P = 0.026) and older age group compared with young adulthood (P = 0.002). However, SRT did not differ between the middle adulthood and older age groups (P > 0.05). In addition, SRT was moderately positively and negatively correlated with TM-A (r = 0.355, P = 0.001), TM-B (r = 0.460, P < 0.001), TM-B-A (r = 0.433, P < 0.001), and DS backward (r = −0.409, P < 0.001) that is visual attention and working memory. Conclusion: SRT may be affected by age. This appears to be part of the visual attention and working memory associated with the saccadic decision-making process.

Parkinson's disease (PD) presents a significant clinical challenge due to its profound motor and cognitive impacts. Early diagnosis is crucial for implementing effective, stage-based treatment strategies. Recently, eye-tracking technology has emerged as a promising tool for the non-invasive diagnosis and monitoring of various neurological disorders, including PD. This retrospective study analyzed eye-tracking parameters, specifically visually-guided saccades (VGS), in PD patients within a clinical setting. We reviewed eye-tracking data from 62 PD patients, focusing on eye movement performance in horizontal and vertical VGS tasks. Our findings revealed significant correlations between demographic profiles, Mini-Mental State Examination (MMSE) scores, pattern recognition, and spatial working memory tests with saccadic performance in PD patients. Despite the retrospective nature of the study, our results support the potential of eye-tracking technology as a valuable diagnostic tool in PD assessment and monitoring. Future research should prioritize longitudinal studies and more comprehensive assessments to further understand and enhance the clinical application of eye-tracking in PD. [ABSTRACT FROM AUTHOR]

Under natural behavioral conditions, visually guided eye movements are linked to direction-specific modulations of cortico-spinal system (CSS) excitability in upper-limb muscles, even in absence of a manual response. These excitability changes have been shown to be compatible with a covert motor program encoding a manual movement toward the same target of the eyes. The aim of this study is to investigate whether this implicit oculo-manual coupling is enforced following every saccade execution or it depends on the behavioral context. Twenty-two healthy young adults participated in the study. Single-pulse transcranial magnetic stimulation was applied to the motor cortex at nine different time epochs during a double-choice eye task, in which the decision to execute a prosaccade or an antisaccade was made on the color of a peripheral visual cue. By analyzing the amplitude of the motor evoked potentials (MEP) in three distal muscles of the resting upper-limb, a facilitation peak of CSS excitability was found in two of them at 120 ms before the eyes begin to move. Furthermore, a long-lasting, generalized reduced corticomotor excitability develops following the eye response. Finally, a quite large modulation of MEP amplitude, depending on the direction of the saccade, is observed only in the first dorsal interosseous muscle, in a narrow time window at about 150 ms before the eye movement, irrespective of the type of the ocular response (pro-/anti-saccade). This change in CSS excitability is not tied up to the timing of the occurrence of the visual cue but, instead, appears to be tightly time-related to the saccade onset. Observed excitability changes differ in many respects from those previously reported with different behavioral paradigms. A main finding of our study is that the implicit coupling between eye and hand motor systems is contingent upon the particular motor set determined by the cognitive aspects of the performed oculomotor task. In particular, the direction-specific modulation in CSS excitability described in this study appears to be related to perceptual and decision-making processes rather than representing an implicit upper-limb motor program, coupled to the saccade execution.

The latency of visually guided saccades executed after a cueing condition was adopted as a more ecological index to explore how spatial attention, usually assessed by manual responses, operates in the visual field. Subjects executed saccades aimed towards one of 4 possible positions, equally distributed around a central fixation cross. Before the onset of the saccadic target, a visual cue was briefly presented at the same or at a different spatial location. The visual cue was non-predictive of the target position. Two experimental sessions were carried out, differing for the onset asynchrony between cue and target. A time-dependent coupling between the task-irrelevant location of the cue and the direction of a following overt shift of attention emerged. Clinical applications of the adopted experimental setting to neurological and psychiatric patients with motor impairments are discussed.

Memory-guided saccades show effect of a perceptual illusion whereas visually guided saccades do not. J Neurophysiol 119: 62-72, 2018. First published September 27, 2017; doi:10.1152/jn.00229.2017.--The doubledrift stimulus (a drifting Gabor with orthogonal internal motion) generates a large discrepancy between its physical and perceived path. Surprisingly, saccades directed to the double-drift stimulus land along the physical, and not perceived, path (Lisi M, Cavanagh P. Curr Biol 25: 2535-2540, 2015). We asked whether memory-guided saccades exhibited the same dissociation from perception. Participants were asked to keep their gaze centered on a fixation dot while the doubledrift stimulus moved back and forth on a linear path in the periphery. The offset of the fixation was the go signal to make a saccade to the target. In the visually guided saccade condition, the Gabor kept moving on its trajectory after the go signal but was removed once the saccade began. In the memory conditions, the Gabor disappeared before or at the same time as the go-signal (0- to 1,000-ms delay) and participants made a saccade to its remembered location. The results showed that visually guided saccades again targeted the physical rather than the perceived location. However, memory saccades, even with 0-ms delay, had landing positions shifted toward the perceived location. Our result shows that memory- and visually guided saccades are based on different spatial information. NEW & NOTEWORTHY We compared the effect of a perceptual illusion on two types of saccades, visually guided vs. memory-guided saccades, and found that whereas visually guided saccades were almost unaffected by the perceptual illusion, memory-guided saccades exhibited a strong effect of the illusion. It has been recently demonstrated that visually guided saccades are linked to changes in muscle excitability in the relaxed upper limb, which are compatible with a covert motor plan encoding a hand movement toward the gaze target. In this study we investigated whether these excitability changes are time locked to the visual stimulus, as predicted by influential attention models, or are strictly dependent on saccade execution. Single-pulse transcranial magnetic stimulation was applied to the motor cortex at eight different time delays during a ’go’/’no-go’ task, which involved overt or covert orienting of attention. By analyzing the time course of excitability in three hand muscles, synchronized with the onset of either the attentional cue or the eye movement, we demonstrated that side- and muscle-specific excitability changes were strictly time locked to the saccadic response and were not correlated to the onset of the visual attentive stimulus. Furthermore, muscle excitability changes were absent following a covert shift of attention. We conclude that a sub-threshold manual motor plan is automatically activated by the saccade decision-making process, as part of a covert eye-hand coordination program. We found no evidence for a representation of spatial attention within the upper limb motor map.

Saccadic adaptation maintains the correct mapping between eye movements and their targets, yet the dynamics of saccadic gain changes in the presence of systematically varying disturbances has not been extensively studied. Here we assessed changes in the gain of saccade amplitudes induced by continuous and periodic postsaccadic visual feedback. Observers made saccades following a sequence of target steps either along the horizontal meridian (Two-way adaptation) or with unconstrained saccade directions (Global adaptation). An intrasaccadic step--following a sinusoidal variation as a function of the trial number (with 3 different frequencies tested in separate blocks)--consistently displaced the target along its vector. The oculomotor system responded to the resulting feedback error by modifying saccade amplitudes in a periodic fashion with similar frequency of variation but lagging the disturbance by a few tens of trials. This periodic response was superimposed on a drift toward stronger hypometria with similar asymptotes and decay rates across stimulus conditions. The magnitude of the periodic response decreased with increasing frequency and was smaller and more delayed for Global than Two-way adaptation. These results suggest that--in addition to the well-characterized return-to-baseline response observed in protocols using constant visual feedback--the oculomotor system attempts to minimize the feedback error by integrating its variation across trials. This process resembles a convolution with an internal response function, whose structure would be determined by coefficients of the learning model. Our protocol reveals this fast learning process in single short experimental sessions, qualifying it for the study of sensorimotor learning in health and disease. [ABSTRACT FROM AUTHOR]

Recent studies demonstrated abnormalities in several types of eye movements (smooth pursuit, optokinetic nystagmus, fixation and vergence) in ST patients. The present study investigates horizontal and vertical saccades in patients with tinnitus lateralized predominantly to the left or to the right side. Compared to left sided ST, tinnitus perceived on the right side impaired almost all the parameters of saccades (latency, amplitude, velocity, etc.) and noticeably the upward saccades. Relative to controls, saccades from both groups were more dysmetric and were characterized by increased saccade disconjugacy (i.e. poor binocular coordination). Although the precise mechanisms linking ST and saccadic control remain unexplained, these data suggest that ST can lead to detrimental auditory, visuomotor and perhaps vestibular interactions.

When the eyes fixate at a point in a visual scene, small saccades rapidly shift the image on the retina. The effect of these microsaccades on the latency of subsequent large-scale saccades may be twofold. First, microsaccades are associated with an enhancement of visual perception. Their occurrence during saccade target perception could, thus, decrease saccade latencies. Second, microsaccades are likely to indicate activity in fixation-related oculomotor neurons. These represent competitors to saccade-related cells in the interplay of gaze holding and shifting. Consequently, an increase in saccade latencies would be expected after microsaccades. Here, we present evidence for both aspects of microsaccadic impact on saccade latency. In a delayed response task, participants made saccades to visible or memorized targets. First, microsaccade occurrence up to 50 ms before target disappearance correlated with 18 ms (or 8%) faster saccades to memorized targets. Second, if microsaccades occurred shortly (i.e., <150 ms) before a saccade was required, mean saccadic reaction time in visual and memory trials was increased by about 40 ms (or 16%). Hence, microsaccades can have opposite consequences for saccade latencies, pointing at a differential role of these fixational eye movements in the preparation of saccade motor programs. The Müller-Lyer illusion produces differences in visual length perception when lines of equal length are presented as double-arrows with inward-pointing and outward-pointing arrowheads, respectively. The aim of this study was to investigate the influence of length perception on the amplitude of saccadic eye movements and on postsaccadic fixation positions. Video recordings of eye movements were obtained from 35 healthy volunteers (age: 20-30 years) while they scanned the endpoints of Müller-Lyer targets. The targets were presented in horizontal and vertical orientations with an arrow length of 15°. Fixation periods between the saccades were adjusted at 1 s and 4 s. The final saccadic amplitudes between the endpoints of Müller-Lyer figures reflect the visually perceived length differences: lines with outward-pointing arrowheads were scanned with smaller saccades than lines with inward-pointing arrowheads. This bias in final saccadic amplitude was equal in both the horizontal and vertical target orientations as well as for 1-s and 4-s fixation periods. Postsaccadic drift after initial saccadic movements reduced the effect of the Müller-Lyer illusion depending on arrowhead and target orientation: outward-pointing horizontal arrows caused hypometric pulses, while inward-pointing vertical arrows caused hypermetric pulses. The results of this study demonstrate that spatial coding of both saccadic eye movements and fixation is strongly influenced by visual perception. While the influence of the Müller-Lyer illusion on final saccadic amplitudes did not depend on target orientation or fixation period, an additional pulse-step mismatch appeared in the horizontal (hypometria) and vertical (hypermetria) direction. To determine whether the periarcuate frontal cortex spatially encodes visual and oculomotor parameters, we trained monkeys to repeatedly execute saccades of the same amplitude and direction toward visual targets and we obtained quantitative images of the distribution of metabolic activity in 2D flattened reconstructions of the arcuate sulcus (As) and prearcuate convexity. We found two topographic maps of contraversive saccades to visual targets, separated by a region representing the vertical meridian: the first region straddled the fundus of the As and occupied areas 44 and 6-ventral, whereas the second one occupied areas 8A and 45 in the anterior bank of the As and the prearcuate convexity. The representation of the vertical meridian runs along the posterior borders of areas 8A and 45 (deep in the As). In both maps, the upper part of visuo-oculomotor space is represented ventrally and laterally and the lower part dorsally and medially whereas dorsal and ventral regions are separated by the representation of the horizontal meridian., (© The Author 2014. Saccadic eye movements of a normal subject were assessed through semi-quantitative analysis algorithms based on linear and non-linear test application in order to highlight the dynamics type characterizing saccadic neural system behavior. These movements were recorded during a simple visually-guided saccade test and one with a cognitive load involving button pressing to show a decision. Following the application of specific computational tests, chaotic dynamical trend dominancy was mostly revealed with some differences between the two saccade recording conditions: auto-correlation time was increased from 170 to 240 by cognitive task superposition and the Hurst exponent was enhanced from 0.52 to 0.76, denoting more persistence in the dynamics of saccadic system during increased neural activity related to cognitive task.
