Surface radiometer data from Ross Island, Antarctica, collected during the austral summer 2015–2016 by the US Department of Energy Atmospheric Radiation Measurement (ARM) program West Antarctic Radiation Experiment (AWARE), are used to evaluate how shortwave and longwave irradiance respond to changing cloud properties as governed by contrasting meteorological regimes. Shortwave atmospheric transmittance is derived from pyranometer measurements, and cloud conservative-scattering optical depth is derived from filter radiometer measurements at 870 nm. With onshore flow associated with marine air masses, clouds contain mostly liquid water. With southerly flow over the Transantarctic Mountains, orographic forcing induces substantial cloud ice water content. These ice and mixed-phase clouds attenuate more surface shortwave irradiance than the maritime-influenced clouds and also emit less longwave irradiance due to colder cloud base temperature. These detected irradiance changes are in a range that can mean onset or inhibition of surface melt over ice shelves. This study demonstrates how basic and relatively low-cost broadband and filter radiometers can be used to detect subtle climatological influences of contrasting cloud microphysical properties at very remote locations.

Abstract Clouds, crucial for understanding climate, begin with droplet formation from aerosols, but observations of this fleeting activation step are lacking in the atmosphere. Here we use a time-gated time-correlated single-photon counting lidar to observe cloud base structures at decimeter scales. Results show that the air–cloud interface is not a perfect boundary but rather a transition zone where the transformation of aerosol particles into cloud droplets occurs. The observed distributions of first-arriving photons within the transition zone reflect vertical development of a cloud, including droplet activation and condensational growth. Further, the highly resolved vertical profile of backscattered photons above the cloud base enables remote estimation of droplet concentration, an elusive but critical property to understanding aerosol–cloud interactions. Our results show the feasibility of remotely monitoring cloud properties at submeter scales, thus providing much-needed insights into the impacts of atmospheric pollution on clouds and aerosol-cloud interactions that influence climate.

Background and Aims: Data on the prevalence and characteristics of so-called rare HCV genotypes (GTs) in larger cohorts is limited. This study investigates the frequency of rare GT and resistance-associated substitutions and the efficacy of retreatment in a European cohort. Methods: A total of 129 patients with rare GT1-6 were included from the European resistance database. NS3, NS5A, and NS5B were sequenced and clinical parameters and retreatment efficacies were collected retrospectively. Results: Overall 1.5% (69/4,656) of direct-acting antiviral (DAA)-naive and 4.4% (60/1,376) of DAA-failure patients were infected with rare GT. Although rare GTs were almost equally distributed throughout GT1-6 in DAA-naive patients, we detected mainly rare GT4 (47%, 28/60 GT4; of these n = 17, subtype 4r) and GT3 (25%, 15/60 GT3, of these n = 8, subtype 3b) among DAA-failures. A total of 62% (37/60) of DAA failures had not responded to first-generation regimes and the majority was infected with rare GT4 (57%, 21/37). In contrast, among patients with failure to pangenotypic DAA regimens (38%, 23/60), infections with rare GT3 were overrepresented (57%, 13/23). Although NS5A RASs were uncommon in rare GT2, GT5a, and GT6, we observed combined RASs in rare GT1, GT3, and GT4 at positions 28, 30, 31, which can be considered as inherent. DAA failures with completed follow-up of retreatment, achieved a high SVR rate (94%, 45/48 modified intention-to-treat analysis; 92%, 45/49 intention-to-treat). Three patients with GT4f, 4r, or 3b, respectively, had virological treatment failure. Conclusions: In this European cohort, rare HCV GT were uncommon. Accumulation of specific rare GT in DAA-failure patients suggests reduced antiviral activities of DAA regimens. The limited global availability of pangenotypic regimens for first line therapy as well as multiple targeted regimens for retreatment could result in HCV elimination targets being delayed. The highlight of the meanwhile 50 years of lidar-based aerosol profiling at Garmisch-Partenkirchen has been the measurements of stratospheric aerosol since 1976. After a technical breakdown in 2016, they have been continued with a new, much more powerful system in a vertical range up to almost 50 km a.s.l. (above sea level) that allowed for observing very weak volcanic aerosol up to almost 40 km. The observations since 2017 are characterized by a number of spectacular events, such as the Raikoke volcanic plume equalling in integrated backscatter coefficient that of Mt St Helens in 1981 and severe smoke from several big fires in North America and Siberia with backscatter coefficients up to the maximum values after the Pinatubo eruption. The smoke from the violent 2017 fires in British Columbia gradually reached more than 20 km a.s.l., unprecedented in our observations. The sudden increase in frequency of such strong events is difficult to understand. Finally, the plume of the spectacular underwater eruption on the Tonga Islands in the southern Pacific in January 2022 was detected between 20 and 25 km.

Cloud droplet number concentration (Nd) is crucial for understanding aerosol–cloud interactions (ACI) and associated radiative effects. We present evaluations of four ground-based Nd retrievals based on comprehensive datasets from the Atmospheric Radiation Measurement (ARM) Aerosol and Cloud Experiments in the Eastern North Atlantic (ACE-ENA) field campaign. The Nd retrieval methods use ARM ENA observatory ground-based remote sensing observations from a micropulse lidar, Raman lidar, cloud radar, and the ARM NDROP (Droplet Number Concentration) value-added product (VAP), all of which also retrieve cloud effective radius (re). The retrievals are compared against aircraft measurements from the fast cloud droplet probe (FCDP) and the cloud and aerosol spectrometer (CAS) obtained from low-level marine boundary layer clouds on 12 flight days during summer and winter seasons. Additionally, the in situ measurements are used to validate the assumptions and characterizations used in the retrieval algorithms. Statistical comparisons of the probability distribution function (PDF) of the Nd and cloud re retrievals with aircraft measurements demonstrate that these retrievals align well with in situ measurements for overcast clouds, but they may substantially differ for broken clouds or clouds with low liquid water path (LWP). The retrievals are applied to 4 years of ground-based remote sensing measurements of overcast marine boundary layer clouds at the ARM ENA observatory to find that Nd (re) values exhibit seasonal variations, with higher (lower) values during the summer season and lower (higher) values during the winter season. The ensemble of various retrievals using different measurements and retrieval algorithms such as those in this paper can help to quantify Nd retrieval uncertainties and identify reliable Nd retrieval scenarios. Of the retrieval methods, we recommend using the micropulse lidar-based method. This method has good agreement with in situ measurements, less sensitivity to issues arising from precipitation and low cloud LWP and/or optical depth, and broad applicability by functioning for both daytime and nighttime conditions.

Between 14 March and 21 April 2022, an extensive investigation of an extraordinary Saharan dust intrusion over Europe was performed based on lidar measurements obtained by the European Aerosol Research Lidar Network (EARLINET). The dust episode was divided into two distinct periods, one in March and one in April, characterized by different dust transport paths. The dust aerosol layers were studied over 18 EARLINET stations, examining aerosol characteristics during March and April in four different regions (M-I, M-II, M-III, and M-IV and A-I, A-II, A-III, and A-IV, respectively), focusing on parameters such as aerosol layer thickness, center of mass (CoM), lidar ratio (LR), particle linear depolarization ratio (PLDR), and Ångström exponents (ÅE). In March, regions exhibited varying dust geometrical and optical properties, with mean CoM values ranging from approximately 3.5 to 4.8 km, and mean LR values typically between 36 and 54 sr. PLDR values indicated the presence of both pure and mixed dust aerosols, with values ranging from 0.20 to 0.32 at 355 nm and 0.24 to 0.31 at 532 nm. ÅE values suggested a range of particle sizes, with some regions showing a predominance of coarse particles. Aerosol Optical Depth (AOD) simulations from the NAAPS model indicated significant dust activity across Europe, with AOD values reaching up to 1.60. In April, dust aerosol layers were observed between 3.2 to 5.2 km. Mean LR values typically ranged from 35 to 51 sr at both 355 nm and 532 nm, while PLDR values confirmed the presence of dust aerosols, with mean values between 0.22 and 0.31 at 355 nm and 0.25 to 0.31 at 532 nm. The ÅE values suggested a mixture of particle sizes. The AOD values in April were generally lower, not exceeding 0.8, indicating a less intense dust presence compared to March. The findings highlight spatial and temporal variations in aerosol characteristics across the regions, during the distinctive periods. From 15 to 16 March 2022, Saharan dust significantly reduced UV-B radiation by approximately 14% over the ATZ station (Athens, GR). Backward air mass trajectories showed that the dust originated from the Western and Central Sahara when, during this specific case, the air mass trajectories passed over GRA (Granada, ES) and PAY (Payerne, CH) before reaching ATZ, maintaining high relative humidity and almost stable aerosol properties throughout its transport. Lidar data revealed elevated aerosol backscatter (baer) and PLDR values, combined with low LR and ÅE values, indicative of pure dust aerosols.

Background: Left prefrontal intermittent theta-burst stimulation (iTBS) has emerged as a safe and effective transcranial magnetic stimulation (TMS) treatment protocol in depression. Though network effects after iTBS have been widely studied, the deeper mechanistic understanding of target engagement is still at its beginning. Here, we investigate the feasibility of a novel integrated TMS-fMRI setup and accelerated echo planar imaging protocol to directly observe the immediate effects of full iTBS treatment sessions. Objective/hypothesis: In our effort to explore interleaved iTBS-fMRI feasibility, we hypothesize that TMS will induce acute BOLD signal changes in both the stimulated area and interconnected neural regions. Methods: Concurrent TMS-fMRI with full sessions of neuronavigated iTBS (i.e. 600 pulses) of the left dorsolateral prefrontal cortex (DLPFC) was investigated in 18 healthy participants. In addition, we conducted four TMS-fMRI sessions in a single patient on long-term maintenance iTBS for bipolar depression to test the transfer to clinical cases. Results: Concurrent TMS-fMRI was feasible for iTBS sequences with 600 pulses. During interleaved iTBS-fMRI, an increase of the BOLD signal was observed in a network including bilateral DLPFC regions. In the clinical case, a reduced BOLD response was found in the left DLPFC and the subgenual anterior cingulate cortex, with high variability across individual sessions. Conclusions: Full iTBS sessions as applied for the treatment of depressive disorders can be established in the interleaved iTBS-fMRI paradigm. In the future, this experimental approach could be valuable in clinical samples, for demonstrating target engagement by iTBS protocols and investigating their mechanisms of therapeutic action.

In this study ozone profiles of the differential-absorption lidar at Garmisch-Partenkirchen are compared with those of ozone sondes of the Forschungszentrum Jülich and of the Meteorological Observatory Hohenpeißenberg (German Weather Service). The lidar measurements are quality assured by the highly accurate nearby in situ ozone measurements at the Wank (1780 m a.s.l.) and Zugspitze (2962 m a.s.l.) summits and at the Global Atmosphere Watch station Schneefernerhaus (UFS, 2670 m a.s.l.), at distances of 9 km or less from the lidar. The mixing ratios of the lidar agree with those of the monitoring stations, with a standard deviation (SD) of 1.5 ppb, and feature a slight positive offset of 0.6 ± 0.6 ppb (SD) conforming to the known −1.8 % calibration bias of the in situ instruments. Side-by-side soundings of the lidar and electrochemical (ECC) sonde measurements in February 2019 by a team of the Forschungszentrum Jülich shows small positive ozone offsets for the sonde with respect to the lidar and the mountain stations (0.5 to 3.4 ppb). After applying an altitude-independent bias correction to the sonde data an agreement to within just ±2.5 ppb in the troposphere was found, which we regard as the wintertime uncertainty of the lidar. We conclude that the recently published uncertainties of the lidar in the final configuration since 2012 are realistic and rather small for low to moderate ozone concentrations. Comparisons of the lidar with the Hohenpeißenberg routine measurements with Brewer-Mast sondes are more demanding because of the distance of 38 km between the two sites implying significant ozone differences in some layers, particularly in summer. Our comparisons cover the 3 years September 2000 to August 2001, 2009, and 2018. A slight negative average offset (−3.64 ± 3.72 ppb (SD)) of the sondes with respect to the lidar was found. We conclude that most Hohenpeißenberg sonde data could be improved in the troposphere by recalibration with the Zugspitze station data (1978 to 2011 summit, afterwards UFS). This would not only remove the average offset but also greatly reduce the variability of the individual offsets. The comparison for 2009 suggests a careful partial re-evaluation of the lidar measurements between 2007 and 2011 for altitudes above 6 km, where occasionally a negative bias occurred.

The pronounced increase in ozone observed at the Alpine station Zugspitze (2962 ma.s.l.) since the 1970s has been ascribed to an increase in stratospheric air descending to the Alps. In this paper, we present a reanalysis of the data from for both ozone (1978 to 2011) and carbon monoxide (1990–2011), which has been extended until 2020 by the data from the Global Atmosphere Watch site at the Umweltforschungsstation Schneefernerhaus (UFS; 2671 ma.s.l. – above sea level), which is located just below the Zugspitze summit. For ozone between 1970 and 1977, a constant annual average of 36.25 ppb (parts per billion) was assumed to have been obtained by extrapolation. The analysis is based on data filtering, utilizing the isotope 7Be (measured between 1970 and 2006) and relative humidity (1970 to 2011; UFS from 2002 to 2020). We estimate both the influence of stratospheric intrusions directly descending to the northern rim of the Alps from the full data filtering and the aged (“indirect”) intrusions from applying a relationship between ozone and the 7Be data. The evaluated total stratospheric contribution to the annual average ozone rises roughly from 12 ppb in 1970 to 24 ppb in 2003. It turns out that the increase in the stratospheric influence is particularly strong in winter. A lowering in positive trend is seen afterwards, with a delay of roughly 1 decade after the beginning of the decrease in the solar irradiation. The air masses hitting the Zugspitze summit became drier until 2003, and we see the growing stratospheric contribution as being an important factor for this drying. Both an increase in the lower-stratospheric ozone and the growing thickness of the intruding layers departing downward from just above the tropopause must be taken into consideration. Carbon monoxide in the intrusions did not change much during the full measurement period from 1990 to 2020, with a slight increase until 2005. This is remarkable since, for air outside intrusions, a decrease by approximately 44 % was found, indicating a substantial improvement in the tropospheric air quality.

Abstract Background Adolescents and young adults (AYA) with a chronic somatic disease (CD) have a 3-fold higher risk of post-traumatic stress disorder (PTSD) than healthy controls. In addition, elevated post-traumatic stress symptoms (PTSS) have a negative impact on CD severity, treatment adherence, health problems and functional impairment. However, a more detailed understanding of this comorbidity is lacking. Methods AYA with type 1 diabetes mellitus, juvenile idiopathic arthritis or cystic fibrosis (12–21 years of age) and elevated anxiety and/or depression symptoms, as well as their reference persons (≥ 18 years of age), completed online questionnaires in self- or observer report. The most stressful event related to the CD was reported descriptively. Questionnaires were used to assess PTSS, anxious and depressive symptoms, actual overall health, coping, personal growth and social support. Qualitative content analysis, linear regression models and correlations were used for mixed methods analysis. Results According to the reports of n = 235 AYA (mean age 15.61; 73% girls) and n = 70 reference persons, four categories were identified as the most stressful events due to CD: (1) psychological burden (40% of AYA / 50% of reference persons); (2) CD self-management (32% / 43%); (3) social burden (30% / 27%); and (4) physical impairment (23% / 16%). 37% of AYA reported clinically relevant PTSS due to CD. BackgroundRandomized evidence suggested improved outcomes in fractional flow reserve (FFR) guidance of coronary revascularization compared to medical therapy in well-defined patient cohorts. However, the impact of FFR-guided revascularization on long-term outcomes of unselected patients with chronic or acute coronary syndromes (ACS) is unknown.AimsThe FLORIDA (Fractional FLOw Reserve In cardiovascular DiseAses) study sought to investigate outcomes of FFR-guided vs. angiography-guided treatment strategies in a large, real-world cohort.MethodsThis study included patients enrolled into the German InGef Research Database. Patients undergoing coronary angiography between January 2014 and December 2015 were included in the analysis. Eligible patients had at least one inpatient coronary angiogram for suspected coronary artery disease between January 2014 and December 2015. Patients were stratified into FFR arm if a coronary angiography with adjunctive FFR measurement was performed, otherwise into the angiography-only arm. Matching was applied to ensure a balanced distribution of baseline characteristics in the study cohort. Patients were followed for 3 years after index date and primary endpoint was all-cause mortality.ResultsIn the matched population, mortality at 3 years was 9.6% in the FFR-assessed group and 12.6% in the angiography-only group (p = 0.002), corresponding to a 24% relative risk reduction with use of FFR. This effect was most pronounced in patients in whom revascularization was deferred based on FFR (8.7% vs. 12.3%, p = 0.04) and in high-risk subgroups including patients aged ≥75 years (14.9% vs. 20.1%, p

The Far-Infrared Radiation Mobile Observation System (FIRMOS) is a Fourier transform spectroradiometer developed to support the Far-infrared Outgoing Radiation Understanding and Monitoring (FORUM) satellite mission by validating measurement methods and instrument design concepts, both in the laboratory and in field campaigns. FIRMOS is capable of measuring the downwelling spectral radiance emitted by the atmosphere in the spectral band from 100 to 1000 cm−1 (10–100 µm in wavelength), with a maximum spectral resolution of 0.25 cm−1. We describe the instrument design and its characterization and discuss the geophysical products obtained by inverting the atmospheric spectral radiance measured during a campaign from the high-altitude location of Mount Zugspitze in Germany, beside the Extended-range Atmospheric Emitted Radiance Interferometer (E-AERI), which is permanently installed at the site. Following the selection of clear-sky scenes, using a specific algorithm, the water vapour and temperature profiles were retrieved from the FIRMOS spectra by applying the Kyoto protocol and Informed Management of the Adaptation (KLIMA) code. The profiles were found in very good agreement with those provided by radiosondes and by the Raman lidar operating from the Zugspitze Schneefernerhaus station. In addition, the retrieval products were validated by comparing the retrieved integrated water vapour values with those obtained from the E-AERI spectra.

The ground-based Global Positioning System (GPS) has been used extensively to retrieve integrated water vapour (IWV) and has been adopted as a unique tool for the assessments of atmospheric reanalyses. In this study, we investigated the multi-temporal-scale variabilities and trends of IWV over Europe by using IWV time series from 108 GPS stations for more than 2 decades (1994–2018). We then adopted the GPS IWV as a reference to assess six commonly used atmospheric reanalyses, namely the Climate Forecast System Reanalysis (CFSR); ERA5; ERA-Interim; the Japanese 55-year Reanalysis (JRA-55); the Modern-Era Retrospective Analysis for Research and Applications, version 2 (MERRA-2); and NCEP-DOE AMIP-II Reanalysis (NCEP-2). The GPS results show that the peaks of the diurnal harmonics are within 15:00–21:00 in local solar time at 90 % of the stations. The diurnal amplitudes are 0–1.2 kg m−2 (0 %–8 % of the daily mean IWV), and they are found to be related to seasons and locations with different mechanisms, such as solar heating, land–sea breeze, and orographic circulation. However, mismatches in the diurnal cycle of ERA5 IWV between 09:00 and 10:00 UTC as well as between 21:00 and 22:00 UTC were found and evaluated for the first time, and they can be attributed to the edge effect in each ERA5 assimilation cycle. The average ERA5 IWV shifts are −0.08 and 0.19 kg m−2 at the two epochs, and they were found to be more significant in summer and in the Alps and in Eastern and central Europe in some cases. Nevertheless, ERA5 outperforms the other reanalyses in reproducing diurnal IWV anomalies at all the 1-, 3-, and 6-hourly temporal resolutions. ERA5 is also superior to the others in modelling the annual cycle and linear trend of IWV. For instance, the IWV trend differences between ERA5 and GPS are quite small, with a mean value and a standard deviation of 0.01 % per decade and 0.97 % per decade, respectively. However, due to significant discrepancies with respect to GPS, CFSR and NCEP-2 are not recommended for the analysis of IWV trends over southern Europe and the whole of Europe, respectively.

Non-naturalists about the normative face the problem of providing a metaphysical explanation for the supervenience of the normative on the natural. Recently, Gideon Rosen has argued that non-naturalists can side-step this problem by rejecting strong supervenience and the view that normative truths are metaphysically necessary. Rosen proposes to take normative truths to be normatively necessary, where normative necessity is different from and irreducible to metaphysical necessity. I argue that if Rosen is right, that creates a deeper problem for robust ethical realism (the view that there are mind-independent, non-natural moral facts). According to robust ethical realism, it is a normative fact that persons are an especially valuable kind of being. But if Rosen is right, that is a metaphysically contingent fact. The existence of persons is also contingent. According to robust ethical realism, then, there is a striking match between what the normative facts happen to be and the kinds of beings that happen to exist. Persons could have failed to exist and they could have failed to be valuable, but it just so happens to be a fact about the natural world that they exist and a normative fact that they have value. Given that this match is accidental, it amounts to a miraculous coincidence. To the extent that commitment to unexplained coincidences counts against a view, robust ethical realism faces a problem.

Die Diabetische Polyneuropathie (DPN) geht mit deutlich erhöhten Gesundheitskosten und somit einer wirtschaftlichen Belastung für die Gesellschaft und das Gesundheitssystem einher (Happich et al. 2008; Khdour 2020; Shillo et al. 2019). Eine hohe Anzahl an Krankenhausaufenthalten, Pflegebedürftigkeit, Arbeitsunfähigkeit sowie Folgekosten der DPN (bspw. Amputationen) und eine insgesamt stärkere Inanspruchnahme von Gesundheitsressourcen sind für die höheren medizinischen Kosten verantwortlich (Sadosky et al. 2015; Schroeter/Hartung 2005; Ziegler 2020) und verdeutlichen die Belastung der an DPN erkrankten Patient:innen. Tendenziell steigen die Kosten dabei mit zunehmender Krankheitslast an (Happich et al. 2008).

We conceive of ourselves as capable of acting in response to normative reasons. Given that normative reasons are facts, this self-conception entails that we are capable of acting in response to facts. Arguments from error cases might seem to force us to deflate this self-conception, for they seem to show that to act in light of a fact must simply be a way of acting in light of a belief. The starting point of this paper is the rejection of this deflationary view. In order to reject the argument from error cases, we should adopt a disjunctive view of motivating reasons. According to this view there are two distinct ways of acting in light of a consideration: acting in light of a fact and acting in light of a belief. Disjunctivism about motivating reasons, however, is the target of a skeptical challenge grounded in a cognitivist account of the mind. According to this account, cognition is to explained in terms of the manipulation of representations and there is no meaningful difference between acting in light of a fact and acting in light of a belief: in both cases, one decides in light of a representation. The goal of this paper is to defend disjunctivism from this objection. In order to do so I appeal to Gibson’s ecological psychology. I argue that the ecological account of perception allows us to hold that in the case of actions guided by perception we act in light of the facts themselves (not representations of those facts) and that the same approach can be extended to cover cases in which we act in light of sensorily unavailable facts.

Zooplankton, integral to aquatic ecosystems, face diverse environmental influences. To comprehend their dynamics, critical for ecological insights and fisheries management, traditional morphological analysis proves laborious. Recent advances include automated systems like ZooScan and DNA metabarcoding. This study examines two methods on the same samples to identify similarities and dependencies between them, potentially reducing the required workload and enhancing the quality of the results. Ten Lake Starnberg vertical tows in September 2021 provided zooplankton samples preserved in ethanol. Subsamples underwent ZooScan morphological identification and subsequent DNA metabarcoding. High concordance between ZooScan counts and DNA reads (86.8%) was observed, while biomass calculations from body length (major axis) and equivalent spherical diameter (ESD) showed slightly lower agreement (78.1% and 79.6%, respectively). Linear regression analysis revealed a correlation between counts and DNA reads (r2 = 0.59). This study underscores the complementary strengths and limitations of ZooScan and DNA metabarcoding for zooplankton analysis. ZooScan aids biomass estimation and morphological differentiation, whereas DNA metabarcoding offers superior taxonomic resolution and low-abundance taxon detection. Combining both methods on the same sample enhances understanding and facilitates future advanced analyses.

We investigate a significant model-observation difference found between cloud-base vertical velocity for continental shallow cumulus simulated using large-eddy simulations (LES) and observed by Doppler lidar measurements over the U.S. Southern Great Plains Atmospheric Radiation Measurement Facility. The LES cloud-base vertical velocity is dominated by updrafts that are consistent with a general picture for convective clouds but is inconsistent with Doppler lidar observations that also show the presence of considerable downdrafts. The underestimation of simulated downdrafts is found to be a robust feature, being insensitive to various numerical, physical, or dynamical choices. We find that simulations can more closely reproduce observations only after improving the model physics to use size-resolved microphysics and horizontal longwave radiation, both of which modify the cloud buoyancy and velocity structure near cloud side edges. The results suggest that treatments that capture these structures are needed for the proper simulation and subsequent parameterization development of shallow cumulus vertical transport.

Abstract Helicobacter (H.) pylori-induced gastritis is a risk factor for gastric cancer (GC). Deleted-in-liver-cancer-1 (DLC1/ARHGAP7) inhibits RHOA, a downstream mediator of virulence factor cytotoxin-A (CagA) signalling and driver of consensus-molecular-subtype-2 diffuse GC. DLC1 located to enterochromaffin-like and MIST1+ stem/chief cells in the stomach. DLC1+ cells were reduced in H. pylori gastritis and GC, and in mice infected with H. pylori. DLC1 positivity inversely correlated with tumour progression in patients. GC cells retained an N-terminal truncation variant DLC1v4 in contrast to full-length DLC1v1 in non-neoplastic tissues. H. pylori and CagA downregulated DLC1v1/4 promoter activities. DLC1v1/4 inhibited cell migration and counteracted CagA-driven stress phenotypes enforcing focal adhesion. CagA and DLC1 interacted via their N- and C-terminal domains, proposing that DLC1 protects against H. pylori by neutralising CagA. H. pylori-induced DLC1 loss is an early molecular event, which makes it a potential marker or target for subtype-aware cancer prevention or therapy.
