3. CRISPR-transient expression in soybean for simplified gRNA screening in planta

Abstract

Abstract The objective of this work was to develop a method to create and validate CRISPR-Cas systems and different gRNAs in soybean (Glycine max) embryos. Two model genes were used for simple mutation with one gRNA or partial gene deletion with two guides. The gRNAs were inserted into the CRISPR transformation vectors by a type IIS restriction enzyme or by subcloning and inserting the promoter + gRNA2 in the final transformation vector using the classic restriction enzyme cloning method. The vectors were successfully constructed for one and two gRNAs. Agrobacterium-mediated transient transformation in soybean was carried out to test the quality of gRNAs and of the system itself (expression cassette). Simple mutation and gene deletion were detected in the embryos transformed after DNA enrichment by enzyme digestion followed by polymerase chain reaction and sequencing, which indicates that the CRISPR-Cas system and guides were working. This protocol can be used to accelerate CRISPR-based genome editing strategies for genetic transformation in soybean.

Abstract

The new scientific decade (2023-2032) of the International Association of Hydrological Sciences (IAHS) aims at searching for sustainable solutions to undesired water conditions – whether it be too little, too much or too polluted. Many of the current issues originate from global change, while solutions to problems must embrace local understanding and context. The decade will explore the current water crises by searching for actionable knowledge within three themes: global and local interactions, sustainable solutions and innovative cross-cutting methods. We capitalise on previous IAHS Scientific Decades shaping a trilogy; from Hydrological Predictions (PUB) to Change and Interdisciplinarity (Panta Rhei) to Solutions (HELPING). The vision is to solve fundamental water-related environmental and societal problems by engaging with other disciplines and local stakeholders. The decade endorses mutual learning and co-creation to progress towards UN sustainable development goals. Hence, HELPING is a vehicle for putting science in action, driven by scientists working on local hydrology in coordination with local, regional, and global processes.

Abstract

Climate change and population size records threaten food security. Therefore, the call for a more sustainable and efficient crop production has never been more urgent. Traditional plant breeding was one of the first successful approaches to expand cultivation areas and crop yield. Later, biotechnological tools, such as genetically modified organisms containing exogenous DNA, further broadened the limits of agricultural results, yet bringing huge financial, bureaucratic, and public rejection hurdles. In the 90s, scientific advances brought the opportunity to drive mutations using engineered nucleases and since 2013, CRISPR-Cas has emerged as the most practical toolkit to edit genomes. One of the most striking possibilities is to generate edited and non-transgenic plants. In this review, we present the working mechanism of CRISPR-induced mutations, pinpoint the latest techniques developed, as well as its myriad of applications in agriculture. The enhancing scope of CRISPR ranges from introducing traits of agronomic interest – such as herbicide resistance, resistance/tolerance to biotic and abiotic stresses, and quality and durability of products – to accelerating plant breeding processes, including haploid induction, generating male-sterile lines, fixating hybrid vigor, and overcoming self-incompatibility. We also discuss regulatory issues surrounding edited plants and derived products around the world, challenges that must be overcome, and future prospects to harness all the potential of this amazing tool to guarantee the new crop production revolution.

17. Continuous rainfall-runoff modelling in ungauged basins: steps forwards, bottlenecks and possible applications in early warning system design

Abstract

The benefit of continuous modelling in hydrological studies is widely recognized, indeed it is particularly promising for estimating the design hydrological input for a variety of practical applications and there is a clear tendency to overcome the concept of design hydrograph in favor of the design runoff simulation. Recently, it was underlined the possibility and the opportunity to apply the continuous framework also in the challenging case of ungauged basins. However, while promising, this approach is still not commonly adopted in practice mainly because it needs as input a simulated rainfall time series, that still is not user-friendly task.In this contribution we summarize the recent steps forward of the continuous modelling in ungauged basins and the residual procedural bottlenecks, moreover we explore its use for designing flood early warning systems. We illustrate a framework based on hydrological-hydraulic synthetic scenarios for selecting the best-performing machine learning model for forecasting discharges. Finally, feature importance measures are introduced for discerning the most influential sub-basins where the measurement instrumentations should be installed enabling the implementation of a cost-effective flood early warning system., The 28th IUGG General Assembly (IUGG2023) (Berlin 2023)

23. Analysis of large-scale monitoring data to identify spatial and temporal trend of risk for terbuthylazine and desethyl-terbuthylazine in surface water bodies of Po plain (Italy)

Abstract

In Italy, the National Environmental Protection Agency (ISPRA) publishes annual reports containing the results of monitoring programs for pesticides in surface and groundwater. These programs were established to accomplish the EU Directives addressing the quality of water resources (drinking water, groundwater and surface water) such as the Water Framework Directive or the Directive on the Sustainable Use of Pesticides. These data can be extremely helpful for risk management actions as they can provide indications where to implement mitigation measures at the territorial scale. In this study, we extrapolated the monitoring data (period 2008–2017) of terbuthylazine and its metabolite desethyl-terbuthylazine in the surface water bodies of the Po plain (Northern Italy). Then, these data were utilized to apply a methodology based on a previous work proposed and published by our research team, which combine, in two distinct phases, historical series of monitoring data and GIS. Overall, the results allowed to identify three areas where to focus risk mitigation measures, which correspond to the low part of the Lombardy (Lodi, Cremona and Mantua area), the area between the Padua and Venice provinces in Veneto and the area of Ferrara in Emilia Romagna.

Abstract

BACKGROUND AND OBJECTIVE: Cardiovascular (CV) diseases represent a major cause of death and severe medical condition worldwide. Different therapeutic options are available to control low-density lipoprotein cholesterol (LDL-C) level in order to prevent CV events. In recent years, two new drugs were approved for patients who are unable to reduce circulating LDL-C with the current therapies: evolocumab and alirocumab (proprotein convertase subtilisin/kexin type nine [PCSK9] inhibitors). This study was aimed to characterise patients who started treatment with PCSK9 inhibitors in the Tuscany region of Italy during the first year of public healthcare service reimbursement and to describe the pattern of PCSK9 inhibitor use in the first 6 months of treatment. METHODS: Patients on PCSK9 inhibitor treatment in Tuscany (3.7 million inhabitants) from 07/2017 to 06/2018 were selected from regional healthcare administrative databases. Concomitant use of lipid-lowering therapies (LLTs), adherence and persistence during the 6 months preceding the first PCSK9 inhibitor dispensing, as well as comorbidities since 1996, were described. In the first 6 months of PCSK9 inhibitor treatment, adherence, persistence and concomitant LLTs were assessed. RESULTS: There were 269 (176 evolocumab, 93 alirocumab) new users of PCSK9 inhibitors. Patients (mean age of 59.1 years) were mainly male (71.0%) in secondary prevention (70.2%) and affected by familial hypercholesterolaemia (53.5%). Sixty-six patients (24.5%) had diabetes mellitus and 12 (4.5%) chronic renal failure. In the 6 months prior to the first PCSK9 inhibitor administration, 61.3% of patients received at least one prescription of ezetimibe or high-intensity statins and 45.7% were persistent to these drugs. During follow-up, 79.9% of patients were adherent to PCSK9 inhibitor and 73.3% were persistent. Abstract

A pregnancy related cancer is defined as a neoplasm diagnosed in a woman during gestation or within 12 months following childbirth or abortion. Breast, thyroidal, melanoma, cervical and hematological malignancies are those most commonly diagnosed during pregnancy, with a reported incidence considered lower than those estimated of 1 case on 1000 pregnancies. The clinical complexity is generated by the co-presence of mother and fetus, and the consequent conflict represented by the need to identify a therapeutic strategy that is effective for the maternal prognosis and, at the same time, safeguards the fetal well-being. Moreover, the counseling, diagnosis, staging and management of these cases are further complicated because most of the scientific literature consists in case reports or small retrospective studies and lacks in guidelines and prospective clinical trials. On that basis, the Study Group on Cancer in Pregnancy (members of S.I.G.O.-Italian Society of Gynecology and Obstetrics) presents the TIGRE Trial, an Italian multicenter observational study aimed at establishing a shared database for the collection of epidemiological and clinical data of the patients affected by pregnancy related cancer in Italy, with the final objective to improve the outcome of a mother and a child involved in a such pathology.

36. Regulation of α-expansins genes in Arabidopsis thaliana seeds during post-osmopriming germination

Abstract

Seed osmopriming is a pre-sowing treatment that involves limitation of the seed water imbibition, so that pre-germinative metabolic activities proceed without radicular protrusion. This technique is used for improving germination rate, uniformity of seedling growth and hastening the time to start germination. In Arabidopsis thaliana, seed germination has been associated with the induction of enzymes involved in cell wall modifications, such as expansins. The α-expansins (EXPAs) are involved in cell wall relaxation and extension during seed germination. We used online tools to identify AtEXPA genes with preferential expression during seed germination and RT-qPCR to study the expression of five EXPA genes at different germination stages of non-primed and osmoprimed seeds. In silico promoter analysis of these genes showed that motifs similar to cis-acting elements related to abiotic stress, light and phytohormone responses are the most overrepresented in promoters of these AtEXPA genes, showing that their expression is likely be regulated by intrinsic developmental and environmental signals during Arabidopsis seed germination. The osmopriming conditioning had a decreased time and mean to 50% germination without affecting the percentage of final seed germination. The dried PEG-treated seeds showed noticeable high mRNA levels earlier at the beginning of water imbibition (18 h), showing that transcripts of all five EXPA isoforms were significantly produced during the osmopriming process. The strong up-regulation of these AtEXPA genes, mainly AtEXPA2, were associated with the earlier germination of the osmoprimed seeds, which qualifies them to monitor osmopriming procedures and the advancement of germination.

Abstract

Regular aerobic exercise strongly influences muscle metabolism in elderly and young; however, the acute effects of aerobic exercise on protein metabolism are not fully understood. We investigated the effect of a single bout of moderate walking (45 min at ~40% of peak [O.sub.2] consumption) on postexercise (POST-EX) muscle metabolism and synthesis of plasma proteins [albumin (ALB) and fibrinogen (FIB)] in untrained older (n = 6) and younger (n = 6) men. We measured muscle phenylalanine (Phe) kinetics before (REST) and POST-EX (10, 60, and 180 min) using L-[ring-[sup.2][H.sub.5]]phenylalanine infusion, femoral arteriovenous blood samples, and muscle biopsies. All data are presented as the difference from REST (at 10, 60, and 180 min POST-EX). Mixed muscle fractional synthesis rate (FSR) increased significantly at 10 min POST-EX in both the younger (0.0363%/h) and older men (0.0830%/h), with the younger men staying elevated through 60 min POST-EX (0.0253%/ h). ALB FSR increased at 10 min POST-EX in the younger men only (2.30%/day), whereas FIB FSR was elevated in both groups through 180 min POST-EX (younger men = 4.149, older men = 4.107%/ day). Muscle protein turnover was also increased, with increases in synthesis and breakdown in younger and older men. Phe rate of disappearance (synthesis) was increased in both groups at 10 min POST-EX and remained elevated through 60 min POST-EX in the older men. Abstract

A pregnancy related cancer is defined as a neoplasm diagnosed in a woman during gestation or within 12 months following childbirth or abortion. Breast, thyroidal, melanoma, cervical and hematological malignancies are those most commonly diagnosed during pregnancy, with a reported incidence considered lower than those estimated of 1 case on 1000 pregnancies. The clinical complexity is generated by the co-presence of mother and fetus, and the consequent conflict represented by the need to identify a therapeutic strategy that is effective for the maternal prognosis and, at the same time, safeguards the fetal well-being. Moreover, the counseling, diagnosis, staging and management of these cases are further complicated because most of the scientific literature consists in case reports or small retrospective studies and lacks in guidelines and prospective clinical trials. On that basis, the Study Group on Cancer in Pregnancy (members of S.I.G.O.-Italian Society of Gynecology and Obstetrics) presents the TIGRE Trial, an Italian multicenter observational study aimed at establishing a shared database for the collection of epidemiological and clinical data of the patients affected by pregnancy related cancer in Italy, with the final objective to improve the outcome of a mother and a child involved in a such pathology.
