In this study freeze-drying and oven-drying (at 50 and 65 °C) were applied for the preparation of dried pumpkin waste. The effect of drying methods on physical properties (moisture content, water activity, hygroscopicity, water solubility, bulk and tapped density, flowability and colour), content of bioactive compounds (carotenoids and phenolics) and antioxidant activity were investigated. Also, the influence of one-year storage at –20 °C on the bioactive compounds content and the antioxidant activity of dried pumpkin waste was examined. Results indicated that drying method showed a significant impact on the investigated characteristics. Bioactive compounds content and antioxidant activity by DPPH test of freeze-dried were higher than of oven-dried pumpkin waste, while higher antioxidant activity in reducing power assay was determined by oven-dried pumpkin waste. An artificial neural network model was developed, for the anticipation of antioxidant activity according to bioactive compounds content (phenolics and carotenoids), in oven-dried (at 50 and 65 °C) and in freeze-dried pumpkin waste after one-year storage. These models showed good prediction properties (the r2 value during training cycle for output variables was 0.999). It was demonstrated that pumpkin waste is potentially an important source of bioactive compounds, which can be used after extraction in suitable forms in the development of functional food products.

Modern trends in phytochemical extraction from alternative crops support the use of alternative technologies, such as ultrasound- and microwave-assisted extractions. Considering the reduction of toxic solvents, rapid and effective extraction process, the abovementioned technologies have become the foundation of green chemistry approaches in a wide range of applications. These extractions have not been studied on Satureja kitaibelii Wierzb. ex Heuff, which is a highly potent plant when considering its aromatic and medicinal properties. This preliminary study presents an in vitro evaluation of biological activities of ultrasound- and microwave-assisted extracts of S. kitaibelii, for the first time. Furthermore, it offers a totally green, modern, fast and reproducible method for extraction of phytochemicals from S. kitaibelii herba (Rtanj Mountain, Serbia). This short communication suggests that the applied microwave-assisted extraction, using only water as the solvent, can be a promising approach for obtaining green products with commercial potential. As a secondary industrial product, peach waste (PW) presents an ecological problem, but is potentially a rich source of natural antioxidants. A potential and novel way to improve the phytochemical stability of waste rich in phytochemicals is encapsulation in yeast cells that possess good structural characteristics. In the present study, PW extract was encapsulated in non-plasmolyzed, plasmolyzed and living Saccharomyces cerevisiae cells using the freeze-drying method. HPLC analysis revealed that β-carotene is the most abundant carotenoid, while epicatechin and catechin are the most abundant phenolics in PW. The highest encapsulation efficiency of carotenoids (86.59 %), as well as phenolics (66.98 %), was obtained with freeze-dried non-plasmolyzed yeast cells used as carriers. Although plasmolysis can cause some changes in the structure and properties of yeast cells, it did not enhance the encapsulation efficiency of present phytochemicals. Successful encapsulation of PW extract in yeast cells was confirmed by FTIR spectroscopy and SEM imaging. The obtained results present the encapsulation of sensitive compounds in yeast cells by freeze-drying as an excellent method for preserving valuable compounds and their potential use in the food and pharmaceutical industries.

Background/Aim. Plant polyphenols are well known to show antimutagenic, anticarcinogenic, antiviral and antioxidative activity. The aim of this study was to investigate bio-active potential of Armoracia rusticana root juice and extracts: their polyphenol content, as well as in vitro antitumor activity and cell-death mechanism. Methods. Liquid-liquid extraction of polar and non-polar compounds was used and poly-phenolic compounds were identified and quantified by high performance liquid chomatography (HPLC) analysis. Anti-proliferative activity was examined in vitro on human cervix carcinoma (HeLa), breast adenocarcinoma (MCF7, MDA-MB-231), colon adenocarcinoma (HT-29), lung adenocarcinoma (A549), prostate adenocarcinoma (PC-3), melanocyte carcinoma (Hs 294T), hepatocyte carcinoma (Hep G2), as well as rat hepatocyte carcinoma (H-4-II-E), and normal human fetal lung (MRC-5) cell line using sulforhodamine B assay. The mechanism of cell-death in cell line was determined using Cell Death Detection ELISAPLUS kit. Results. Dichloromethane extracts had the highest content of cate-chin, p-hydroxybenzoic, syringic and gallic acid (pulp, E1), and epicatechin (juice, E3). The results showed strong and non-selective antiproliferative activity of chloroform and di-chloromethane extracts and root juice – highest being to-wards liver, breast and lung tissue cells. IC50 values of ex-tracts and juice had low range of concentrations (IC50 = 3.49–26.5 μg/mL) and high range of dilutions (IC50 = 418–1,590). High and unfavorable potential of horseradish juice and chloroform juice extract (E4) to induce necrotic cell death was detected. Conclusion. Strong and non-selective in vitro antiproliferative activity of chloroform and dichloromethane extracts and root juice of horseradish was detected, with necrosis as a main mechanism of induced cell death. In order to utilize horseradish root bioactive potential further investigations that will pinpoint active components with more favourable apoptosis/necrosis inducing properties are needed.

The application of bioactive antioxidant compounds is limited because of their instability. To overcome this drawback, different protection systems including encapsulation have been developed. In this study, for the stabilisation of bioactive compounds (carotenoids and phenolics) extracted from tomato waste, encapsulation with carbohydrate (inulin and gum arabica) and protein (soy protein and pea protein) wall materials by freeze drying method was applied. Content of bioactive compounds and antioxidant properties (determined by DPPH, reducing power and β-carotene bleaching assay) of encapsulates before and after in vitro digestion was investigated. Also, in obtained encapsulates carotenoid stability during storage at ambient (25 ± 5 °C) light and dark conditions for four weeks was assessed. The results indicated that release behavior of bioactive compounds and antioxidant activity after digestion of encapsulates varied according to the type of wall material. Protein carriers showed better ability to preserve phenolic compounds through in vitro gastrointestinal digestion in comparison to carbohydrate wall materials. Pea protein showed best carrier properties for delivery of carotenoids, while differently to other used carriers, gum arabica showed minor ability to release carotenoids after in vitro gastrointestinal digestion. During storage higher content of carotenoids was preserved in encapsulates prepared with carbohydrate carriers. All encapsulates retained higher amount of carotenoids under dark conditions. The results of this study showed that assessment of the behavior of encapsulates during digestion and storage is necessary in order of selection appropriate delivery system of bioactive compounds in powder form which can be used as ingredient in functional food products.

Utilization of carotenoids as a nutraceutical ingredients or natural colorants within foods is currently limited due to their instability. Encapsulation is one of the prominent means used to improve carotenoid stability. In this study the content of bioactive compounds (lycopene, β-carotene and phenolics) and antioxidant activity (DPPH, ABTS, superoxide anion and reducing power test) were investigated in the tomato waste extract. Based on the obtained results, tomato waste showed high contents of lycopene (13.72 mg/100 g DW), β-carotene (12.36 mg/100 g DW) and phenolic compounds (203.33 mg/100 g DW). Also, high values of antioxidant activity were determined in all applied tests. To protect extracted sensitive carotenoid compounds (lycopene and β-carotene) tomato waste extract was encapsulated with various encapsulating materials (soy protein, pea protein, inulin and gum arabica) by freeze drying method. The influence of applying different encapsulating materials on the encapsulation efficiency of carotenoids (lycopene and β-carotene) was studied. Also, obtained encapsulates were characterized in terms of water activity, moisture content, hygroscopicity, flowability, density ratios and particles size. Encapsulate on the basis of gum arabic showed highest encapsulation efficiency of β-carotene (53.47%), while encapsulate prepared with soy protein as encapsulating material showed highest encapsulation efficiency of lycopene (51.44%). The results of this study support the need for preparation of encapsulates with different encapsulating material and evaluation of their characteristics with the aim of development powder forms suitable for supplementation and food fortification.

The aim of this study was to compare the chemical compositions and biological potentials (antioxidant potentials, α-glucosidase test and antimicrobial activity) of wild and cultivated blackberry varieties from two different locations. The chemical compositions were evaluated in fresh samples, while the biological potentials were measured in dry blackberry extracts. The highest dry matter (15.73 g/100 g fw) was obtained for wild blackberry from Verići (Bw2). The significantly higher content of sugars (6.07 g/100 g fw) and ascorbic acid (21.36 g/100 g dw) was found in Chester Thornless blackberry (Bc1) comparing to other samples. The wild blackberry from Javorani (Bw1) showed the highest amount of polyphenols, as well as antioxidant potentials on DPPH (184.26 mmol Trolox/kg dw), ABTS (340.26 mmol Trolox/kg dw) and OH (944.03 mmol BHT/kg dw) radicals. Carrot is considered to be rich in bioactive antioxidants, both lipophilic (carotenoids) and hydrophilic (phenolic compounds). In the present study, the conditions for accelerated solvent extraction (ASE) of bioactive compounds from carrots (Daucus carota L.) were optimized using response surface methodology (RSM). Box–Behnken design was employed for the experimental design to obtain the optimized combination of extraction temperature, time, and number of extraction cycles. Total carotenoid content (TCar), total polyphenol content (TPh), free radical scavenging activity (SA) and reducing power (RP) of the obtained extracts were used as responses for the optimization. Considering the four quality indicators, the ideal extraction conditions were found to be: 120 °C, 60 min and three extraction cycles. Under these conditions, predicted values of 28.84 mg β-carotene/100 g for TCar; 530.81 mg GAE/100 g for TPh; 2572.29 μmol TE/100 g for SA and 1336.26 μmol TE/100 g for RP were obtained with high desirability (0.975) and no significant difference (p < 0.05) with the experimental values. The processing of fruits results in high amounts of waste materials without having any significant commercial values. Fruit wastes represent an important source of phytochemicals with human health benefits, so isolation and further utilization of these compounds is an important issue in agro-industrial waste management. New aspects concerning the use of these wastes for the production of food additives or supplements with high nutritional value have gained increasing interest because these are economically attractive and high-value products. This study discusses the potential of the quince waste, which remains after alcoholic distillation, as a source of bioactive phytochemicals (phenolics, flavonoids and carotenoids). Antioxidant activities were measured by scavenging effect on 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,20-azinobis(3-ethyl-benzothiazoline- 6-sulfonic acid) (ABTS) radicals, β-carotene bleaching assay and reducing power. The anti-inflammatory activity was evaluated by protein denaturation assay. Quince waste extract expressed good concentration-dependent antioxidant and anti-inflammatory activities. Research background. The aim of this study is to determine and compare the antioxidant and antiproliferative activities of juices and extracts of the peel, aril and membrane of the cultivated and wild pomegranate fruits. Experimental approach. The content of total phenols, total flavonoids, total flavonols, total flavan-3-ols and total anthocyanins was determined spectrophotometrically. The individual phenolics were quantified by HPLC. Antioxidant activity was determined by DPPH and ABTS tests and neutralisation of hydroxyl radical, while the antiproliferative activity was measured in vitro by sulforhodamine B (SRB) assay. Results and conclusions. Total phenolics were statistically highest in wild pomegranate peel extract, expressed in gallic acid equivalents, 340.92 mg/g (p<0.05), while total flavonoid content was the highest in cultivated pomegranate peel extract, expressed in quercetin equivalents, 31.84 mg/g (p<0.05). The sample of wild pomegranate peel extract showed the highest antioxidant activity with respect to free DPPH and ABTS radicals. The samples of cultivated pomegranate peel and membrane extracts had almost identical and the strongest effect on the inhibition of hydroxyl radicals (41.24 and 41.23 μg/mL, respectively). The sample of wild pomegranate peel extract showed the strongest effect on the growth inhibition of all tested tumour cell lines. Novelty and scientific contribution. In this study, the bioactivity of different parts of cultivated and wild pomegranates was determined and compared. In the available literature, the individual antioxidant and antiproliferative activity of only some parts of the pomegranate fruit was investigated. All parts of the pomegranate fruit were investigated, including the membrane, which was barely analysed in other works. The wild pomegranate has also been less analysed in previous studies. Carrot is an important root vegetable rich in bioactive compounds like carotenoids and polyphenols with appreciable levels of several other functional components having significant health-promoting properties. Utilization of carotenoids is limited due to their instability. Encapsulation is one of the alternatives used to improve carotenoid stability. The objectives of this study were to optimize the extraction and encapsulation of carotenoids from carrot. Freeze-dried carrots were extracted by conventional solvent extraction (CSE) using four solvents, i.e. ethanol, acetone, ethyl acetate, and hexane. Although CSE using hexane and ethyl acetate resulted in highest carotenoid contents (18.27 and 15.73 mg β-carotene/100 g), acetone and ethanol at a solid to solvent ratio 1:10 w/v with slightly lower carotenoid contents (14.52 and 11.45 mg β-carotene/100 g) were chosen for further studies due to their higher food compatibility and higher polyphenol content (88.86 and 66.21 mg GAE/100 g) than with lower solid to solvent ratio (1:5 w/v). Ethanol and acetone carrot extracts were encapsulated using different carriers by a freeze-drying method in order to obtain the optimum encapsulate with the highest carotenoid encapsulation efficiency (EE). Encapsulation using the maltodextrin, whey and soy protein as a wall material yielded an EE of carotenoids ranging from 41.95% to 100%. The encapsulated β-carotene content was evaluated during two months of storage at ambient temperature under light and dark conditions. Generally, the retention of carotenoids was significantly higher in dark conditions, where maximum retention (65.94-87.32%) occurred in the samples encapsulated in maltodextrin and soy protein. Samples of three types of honey: polyfloral (PH), linden (LH) and acacia (AH,) without and with addition of dried cherries (40%) were analyzed before and after three months of storage. The total phenol (TPh), flavonoid (TFd) and anthocyanin (TAn) contents, antioxidant activities and sensory properties of honeys with and without the addition of dry cherries were evaluated. TPh and TFd increased with addition of dried cherries to the honey, while enriched honeys showed high TAn. The LH sample with dried cherries showed the highest anthocyanins content (41.41mgCGE/100g). The antioxidant activity increased with addition of dried cherries in honey in the DPPH• test and reducing power. The PH and enriched PH exibited the best antiradical activity compared to LH and AH. The EC50 DPPH values were: 23.81 for PH and 24.19 mg/mL for PH, while the EC50 DPPH were: 1.16 mg/mL for PH40 and 1.18 mg/mL for PH40s. RP0.5 values were: 57.00 mg/mL for PH40 and 56.00 mg/ml for PH40s, while RP0.5 were: 15.05 mg/mL for PH40 and 15.18 mg/mL for PH40s. The statistical analysis showed that TPh, TFd and TAn, and antioxidant activity of honeys and enriched honeys showed significant correlation. Sensory analysis of honey with dried cherries, before and after storage, indicated very good sensory characteristics.

Kombucha is a traditional beverage obtained by fermenting sweetened black tea with tea fungus, which represents a consortium of acetic acid bacteria and yeasts. Also, CoffeeBerry® products, which derived from the whole fruit of the coffee plant, are valuable ingredients with nutritional and health-enhancing potential. Samples of fermentation broths enriched with CoffeeBerry® extract and traditional Kombucha were analysed. The fermentation was performed in a bioreactor at 28±1°C for nine days. The results showed that the CoffeeBerry® extract has contributed to a faster fermentation of cultivation medium. Some individual polyphenolic compounds and catehins in fermentation broth samples were identified and quantified by High Performance Liquid Chromatography (HPLC). Among the bioactive compounds present in investigated samples obtained during Kombucha fermentation of the sweetened black tea enriched with CoffeeBerry® extract, chlorogenic acid (188.94-458.56 μg/mL) was the predominant. The antioxidant activity of investigated samples was tested by measuring their ability to scavenge DPPH and reactive hydroxyl radicals by electron spin resonance (ESR) spectroscopy. The scavenging activities on DPPH and hydroxyl radicals were increased with duration of fermentation. IC50 values for Kombucha fermentation broth enriched with CoffeBerry®, based on DPPH and hydroxyl radical scavenging activities, were in the range 26.33-170.13 μL/mL and 11.33-102.22 μL/mL, respectively.

The main chemical composition, i.e. the total content of bioactive compounds (phenolics 2209.86 ± 70.32 mg GAE/100g FDR, flavonoids 831.87 ± 12.61 mg R/100g FDR and anthocyanins 144.55 ± 0.39 mg CGE/100g FDR), in freeze-dried raspberry (FDR) was evaluated spectrophotometrically. Vitamin C content was determined by HPLC analysis (88.81 ± 4.38 mg vit C/100g FDR). Antioxidant activities of FDR extract were evaluated spectrophotometrically on stable 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals and by electron spin resonance spectroscopy (ESR) method on hydroxyl radicals (•OH). EC50 values were evaluated. EC50 DPPH• was 0.127 ± 0.013 mg/ml, while EC50 •OH was 1.366 ± 0.026 mg/ml. Antiproliferative activity of the FDR extract was evaluated in vitro in three human cell lines by colorimetric sulphorhodamine B (SRB) assay. The most pronounced effects were obtained in the breast adenocarcinoma cell line (MCF7). EC50 value was 395.07 ± 96.38 μg/ml. Antimicrobial activity was determined by disk diffusion method. The FDR extract produced a clear inhibition zone (without visible colonies) only toward Staphylococcus aureus. The minimal inhibitory (MIC) and minimal bactericidal (MBC) concentrations of FDR extract were evaluated. Dried apricot was extracted using two different solvents: 80% ethanol and hexane. The contents of total phenolics, flavonoids and anthocyanins were determined in ethanol apricot extract (EAE), while the carotenoid content was determined in hexane apricot extracts (HAE) by spectrophotometric method. The detected amounts of bioactive compounds in dried apricot were: phenolics 498.13 ± 12.04 mg GAE/100g DA, flavonoids 218.45 ± 14.14 mg R/100g DA and anthocyanins 3.08 ± 0.40 mg CGE/100g DA. In dried apricot the β-carotene was present in the amount of 0.56 ± 0.03 mg/100g DA. Some individual phenolic compounds in EAE were determined by HPLC analysis and the most dominant compound was gallic acid (35.02 ± 1.65 mg/100g DA). Free radical scavenging activities of EAE and HAE were tested spectrophotometrically using stable DPPH radicals and reducing power method. The EC50 value for EAE was 2.05 ± 0.06 mg/ml and for HAE was 6.28 ± 0.01 mg/ml. The RP0.5 values determined by reducing power method were 6.11 ± 0.01 mg/ml for EAE and 62.04 ± 0.03 mg/ml for HAE. The results indicate that dried apricot can be a valuable source of natural antioxidants and can be used as a functional food ingredient in the food industry.

The main chemical composition, contents of total phenolic (TPh), total flavonoid (TF), and total monomeric anthocyianin (TMA), as well as the antioxidant activity of two raspberry cultivars (Meeker and Willamette), two blackberry cultivars (Čačanska bestrna and Thornfree) and wild bilberry were studied. The raspberry cultivars had the highest total solids among fruits investigated. Bilberry fruits had the highest sugar-to-acid ratio. Blackberry fruits were richer in crude fibers (cellulose) in comparison to raspberry and bilberry fruits. The content of pectic substances was highest in the bilberry. Also, bilberry had a highest content of TPh (808.12 mg GAE/100 g FW), TF (716.31 mg RE/100 g FW) and TMA (447.83 mg CGE/100 g FW). The antioxidant activity was evaluated spectrophotometrically, using 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity assay. The DPPH free radical scavenging activity, expressed as the EC50 value (in mg of fresh weight of berry fruit per ml of the reaction mixture), of bilberry (0.3157 ± 0.0145 mg/ml) was the highest. These results also showed that the antioxidant value of 100 g FW bilberry, raspberry - Willamette, raspberry - Meeker, blackberry - Čačanska bestrna and blackberry - Thornfree is equivalent to 576.50 mg, 282.74 mg, 191.58 mg, 222.28 mg and 272.01 mg of vitamin C, respectively. There was a significant positive correlation between the antioxidant activities and content of total phenolics (RTPh 2=0.9627), flavonoids (RTF 2=0.9598) and anthocyanins (RTMA 2=0.9496) in berry fruits. The changes in total phenol and flavonoid content, as well as antioxidant activity was monitored in acacia honey supplemented with prunes in 20, 30 and 40% mass concentrations. The total phenolic content increased by 2.5 times (from 16.18 to 41.64 mg GAE/100 g) with increasing concentration of prunes in honey, while the increase in flavonoid content was even higher, approximately 11.5-fold (from 2.65 to 30.86 mg RE/100 g). The addition of prunes also improved the antioxidant activity of acacia honey. The honey samples with highest content of prunes, 40%, exhibited the best antioxidant activity measured by hydroxyl radical sacvenging assay (EC50 •OH=4.56 mg/ml), 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging assay (EC50 DPPH=16.48 mg/ml), and reducing power (EC50 RP=81.17 mg/ml). Judging from the high correlation coefficients, ranging from 0.771 to 0.947 for total phenolics, and from 0.862 to 0.993 for total flavonoids, it is obvious that these compounds were associated with the antioxidant mechanisms. On the other hand, sensorial properties of supplemented honeys were lower than that of pure acacia honey, where flavor of supplemented honey was the least affected. Our results indicate that the supplementation of honey with prunes improves antioxidant activity of honey by enriching the phenolic composition, with slight modifications in sensorial characteristics.

The macro- and micro-chemical composition, as well as antioxidant activity of two strawberry cultivars, Marmolada and Clery, were studied. Results showed a noticeable difference in the sugar, protein and pectin contents. Clery had 6.92% and Marmolada 4.93% of total sugar. Also, protein and pectin contents were higher in the Clery cultivar. No significant difference was observed in acidity, as well as in ash and cellulose content. Marmolada had a higher content of total phenolics and flavonoids (228.04 mg GAE /100 g FW and 136.01 mg RE/100 g FW, respectively ). The anthocyanins content in Marmolada (32.0 mg CGE/100 g FW) was slightly lower than in Clery (36.0 mg CGE/100 g FW). The antioxidant activity was evaluated spectrophotometrically, using 2,2-diphenyl- 1-picrylhydrazyl (DPPH) radical scavenging activity assay. The DPPH free radical scavenging activity, expressed as EC50 value, of Marmolada (0.77 mg/ml )was higher than of Clery (0.83 mg/ml). There was a significant positive correlation (R2>0.90) between the concentration of phenolics/flavonoids/anthocyanins and DPPH radical scavenging activity of both strawberry cultivars. These results also showed that the antioxidant value of 100 g FW Marmolada and Clery is equivalent to 237.91 mg and 219.01 mg of vitamin C, respectively.

Four berry (bilberry, blackberry, strawberry and raspberry) fruits pomace, a waste product coming from juice production, were extracted with 80% methanol containing 0.05% acetic acid. The highest content of antioxidant components, total polyphenols, flavonoids and anthocyanins, was found in the bilberry pomace extract (1116.24, 1047.39 and 1279.49 mg/100g fresh pomace, respectively). The antioxidant activity of the four berry pomace extracts (BPE) was determined using DPPH free radical scavenging assay and reducing power. There was a high linear correlation between the IC50 and content of anthocyanins (r2=0.98) and polyphenols (r2=0.85). Also, a good correlation was found between the IC50 and the content of flavonoids. The strawberry pomace extract showed the highest (RP0.5=0.35 mg/ml), while the blackberry pomace extract showed the lowest (RP0.5=0.57 mg/ml) reducing power. The results showed that BPE can be a valuable source of natural ingrediences of the products in the food, cosmetical and pharmaceutical industry.

Antibacterial activity of Beta vulgaris L. (beetroot) pomace extract (concentration 100 mg/ml) was tested against five Gram positive and seven Gram negative bacterial strains (reference cultures and natural isolates). Disc diffusion method with 15 µl of extract and agar-well diffusion method with 50 and 100 µl were used. Antibiotic (cefotaxime/clavulanic acid) was used as a control sample. The tested extract showed the highest activity against Staphylococcus aureus and Bacillus cereus, where clear zones (without growth) appeared. There was no any activity against other tested Gram-positive bacteria, except for Staphylococcus epidermidis, with a small zone of reduced growth. Growth of all tested Gram-negative bacteria was inhibited usually with 100 µl of extract. The most susceptible were Citrobacter freundii and Salmonella typhymurium. The tested antibiotic gave clear, usually large zones for all tested strains except for Staphylococcus cohni spp. cohni, where only a zone of reduced growth appeared.
