Aims. In this work we use the Gaia DR3 set of ultracool dwarf candidates and complement the Gaia spectrophotometry with additional photometry in order to characterise its global properties. This includes the inference of the distances, their locus in the Gaia colour-absolute magnitude diagram and the (biased through selection) luminosity function in the faint end of the Main Sequence. We study the overall changes in the Gaia RP spectra as a function of spectral type. We study the UCDs in binary systems, attempt to identify low-mass members of nearby young associations, star forming regions and clusters, and analyse their variability properties. Results. We detect 57 young, kinematically homogeneous groups some of which are identified as well known star forming regions, associations and clusters of different ages. We find that the primary members of 880 binary systems with a UCD belong mainly to the thin and thick disk components of the Milky Way. We identify 1109 variable UCDs using the variability tables in the Gaia archive, 728 of which belong to the star forming regions defined by HMAC. We define two groups of variable UCDs with extreme bright or faint outliers. Conclusions. The set of sources identified as UCDs in the Gaia archive contains a wealth of information that will require focused follow-up studies and observations. It will help to advance our understanding of the nature of the faint end of the Main Sequence and the stellar/substellar transition., Accepted by Astronomy and Astrophysics. 29 pages, 20 figures plus 3 appendices

Aims. We produce a clean and well-characterised catalogue of objects within 100 pc of the Sun from the Gaia Early Data Release 3. We characterise the catalogue through comparisons to the full data release, external catalogues, and simulations. We carry out a first analysis of the science that is possible with this sample to demonstrate its potential and best practices for its use., Methods. Theselection of objects within 100 pc from the full catalogue used selected training sets, machine-learning procedures, astrometric quantities, and solution quality indicators to determine a probability that the astrometric solution is reliable. The training set construction exploited the astrometric data, quality flags, and external photometry. For all candidates we calculated distance posterior probability densities using Bayesian procedures and mock catalogues to define priors. Any object with reliable astrometry and a non-zero probability of being within 100 pc is included in the catalogue., Results. We have produced a catalogue of 331 312 objects that we estimate contains at least 92% of stars of stellar type M9 within 100 pc of the Sun. We estimate that 9% of the stars in this catalogue probably lie outside 100 pc, but when the distance probability function is used, a correct treatment of this contamination is possible. We produced luminosity functions with a high signal-to-noise ratio for the main-sequence stars, giants, and white dwarfs. We examined in detail the Hyades cluster, the white dwarf population, and wide-binary systems and produced candidate lists for all three samples. We detected local manifestations of several streams, superclusters, and halo objects, in which we identified 12 members of Gaia Enceladus. We present the first direct parallaxes of five objects in multiple systems within 10 pc of the Sun., Conclusions. We provide the community with a large, well-characterised catalogue of objects in the solar neighbourhood. This is a primary benchmark for measuring and understanding fundamental parameters and descriptive functions in astronomy.

Machine learning approaches promise to accelerate and improve success rates in medicinal chemistry programs by more effectively leveraging available data to guide a molecular design. A key step of an automated computational design algorithm is molecule generation, where the machine is required to design high-quality, drug-like molecules within the appropriate chemical space. Many algorithms have been proposed for molecular generation; however, a challenge is how to assess the validity of the resulting molecules. Here, we report three Turing-inspired tests designed to evaluate the performance of molecular generators. Profound differences were observed between the performance of molecule generators in these tests, highlighting the importance of selection of the appropriate design algorithms for specific circumstances. One molecule generator, based on match molecular pairs, performed excellently against all tests and thus provides a valuable component for machine-driven medicinal chemistry design workflows.

The growing number of multi-epoch optical and infrared sky surveys are uncovering unprecedented numbers of new variable stars, of an increasing number of types. The short interval between observations in adjacent near infrared filters in the UKIDSS Galactic Plane Survey (UGPS) allows for the discovery of variability on the timescale of minutes. We report on the nature of one such object, through the use of optical spectroscopy, time-series photometry and targeted X-ray observations. We propose that UGPS J194310.32+183851.8 is a magnetic cataclysmic variable star of novel character, probably featuring a longer than average spin period and an orbital period likely to be shorter than the period gap (i.e. P$_{\text{orb}}$, 9 pages, 9 figures

SynopsisBy linking anatomical structure to mechanical performance we can improve our understanding of how selection shapes morphology. Here we examined the functional morphology of feeding in fishes of the subfamily Danioninae (order Cypriniformes) to determine aspects of cranial evolution connected with their trophic diversification. The Danioninae comprise three major lineages and each employs a different feeding strategy. We gathered data on skull form and function from species in each clade, then assessed their evolutionary dynamics using phylogenetic-comparative methods. Differences between clades are strongly associated with differences in jaw protrusion. The paedomorphic Danionella clade does not use jaw protrusion at all, members of the Danio clade use jaw protrusion for suction production and prey capture, and members of the sister clade to Danio (e.g., Devario and Microdevario) use jaw protrusion to retain prey after capture. The shape of the premaxillary bone is a major determinant of protrusion ability, and premaxilla morphology in each of these lineages is consistent with their protrusion strategies. Premaxilla shapes have evolved rapidly, which indicates that they have been subjected to strong selection. We compared premaxilla development in giant danio (Devario aequipinnatus) and zebrafish (Danio rerio) and discuss a developmental mechanism that could shift danionine fishes between the feeding strategies employed by these species and their respective clades. We also identified a highly integrated evolutionary module that has been an important factor in the evolution of trophic mechanics within the Danioninae.

The bromodomain and extraterminal (BET) family of bromodomain-containing proteins are important regulators of the epigenome through their ability to recognize N-acetyl lysine (KAc) post-translational modifications on histone tails. These interactions have been implicated in various disease states and, consequently, disruption of BET-KAc binding has emerged as an attractive therapeutic strategy with a number of small molecule inhibitors now under investigation in the clinic. However, until the utility of these advanced candidates is fully assessed by these trials, there remains scope for the discovery of inhibitors from new chemotypes with alternative physicochemical, pharmacokinetic, and pharmacodynamic profiles. Herein, we describe the discovery of a candidate-quality dimethylpyridone benzimidazole compound which originated from the hybridization of a dimethylphenol benzimidazole series, identified using encoded library technology, with an N-methyl pyridone series identified through fragment screening. Optimization via structure- and property-based design led to I-BET469, which possesses favorable oral pharmacokinetic properties, displays activity in vivo, and is projected to have a low human efficacious dose.

Aims. We aim to demonstrate the scientific potential of the Gaia Early Data Release 3 (EDR3) for the study of different aspects of the Milky Way structure and evolution and we provide, at the same time, a description of several practical aspects of the data and examples of their usage., Methods. We used astrometric positions, proper motions, parallaxes, and photometry from EDR3 to select different populations and components and to calculate the distances and velocities in the direction of the anticentre. In this direction, the Gaia astrometric data alone enable the calculation of the vertical and azimuthal velocities; also, the extinction is relatively low compared to other directions in the Galactic plane. We then explore the disturbances of the current disc, the spatial and kinematical distributions of early accreted versus in situ stars, the structures in the outer parts of the disc, and the orbits of open clusters Berkeley 29 and Saurer 1., Results. With the improved astrometry and photometry of EDR3, we find that: (i) the dynamics of the Galactic disc are very complex with oscillations in the median rotation and vertical velocities as a function of radius, vertical asymmetries, and new correlations, including a bimodality with disc stars with large angular momentum moving vertically upwards from below the plane, and disc stars with slightly lower angular momentum moving preferentially downwards; (ii) we resolve the kinematic substructure (diagonal ridges) in the outer parts of the disc for the first time; (iii) the red sequence that has been associated with the proto-Galactic disc that was present at the time of the merger with Gaia-Enceladus-Sausage is currently radially concentrated up to around 14 kpc, while the blue sequence that has been associated with debris of the satellite extends beyond that; (iv) there are density structures in the outer disc, both above and below the plane, most probably related to Monoceros, the Anticentre Stream, and TriAnd, for which the Gaia data allow an exhaustive selection of candidate member stars and dynamical study; and (v) the open clusters Berkeley 29 and Saurer 1, despite being located at large distances from the Galactic centre, are on nearly circular disc-like orbits., Conclusions. Even with our simple preliminary exploration of the Gaia EDR3, we demonstrate how, once again, these data from the European Space Agency are crucial for our understanding of the different pieces of our Galaxy and their connection to its global structure and history.

We present the early installment of the third Gaia data release, Gaia EDR3, consisting of astrometry and photometry for 1.8 billion sources brighter than magnitude 21, complemented with the list of radial velocities from Gaia DR2. Gaia EDR3 contains celestial positions and the apparent brightness in G for approximately 1.8 billion sources. For 1.5 billion of those sources, parallaxes, proper motions, and the (G_BP-G_RP) colour are also available. The passbands for G, G_BP, and G_RP are provided as part of the release. For ease of use, the 7 million radial velocities from Gaia DR2 are included in this release, after the removal of a small number of spurious values. New radial velocities will appear as part of Gaia DR3. Finally, Gaia EDR3 represents an updated materialisation of the celestial reference frame (CRF) in the optical, the Gaia-CRF3, which is based solely on extragalactic sources. The creation of the source list for Gaia EDR3 includes enhancements that make it more robust with respect to high proper motion stars, and the disturbing effects of spurious and partially resolved sources. The source list is largely the same as that for Gaia DR2, but it does feature new sources and there are some notable changes. The source list will not change for Gaia DR3. Gaia EDR3 represents a significant advance over Gaia DR2, with parallax precisions increased by 30 percent, proper motion precisions increased by a factor of 2, and the systematic errors in the astrometry suppressed by 30--40 percent for the parallaxes and by a factor ~2.5 for the proper motions. The photometry also features increased precision, but above all much better homogeneity across colour, magnitude, and celestial position. A single passband for G, G_BP, and G_RP is valid over the entire magnitude and colour range, with no systematics above the 1 percent level.

Spectroscopic observations of white dwarfs reveal that many of them are polluted by exoplanetary material, whose bulk composition can be uniquely probed this way. We present a spectroscopic and photometric analysis of the DA white dwarf WDJ181417.84$-$735459.83, an object originally identified to have a strong infrared excess in the 2MASS and WISE catalogues that we confirmed to be intrinsic to the white dwarf, and likely corresponding to the emission of a dusty disc around the star. The finding of Ca, Fe and Mg absorption lines in two X-SHOOTER spectra of the white dwarf, taken 8 years apart, is further evidence of accretion from a dusty disc. We do not report variability in the absorption lines between these two spectra. Fitting a blackbody model to the infrared excess gives a temperature of 910$\pm50$ K. We have estimated a total accretion flux from the spectroscopic metal lines of $|\dot{\rm M}| = 1.784 \times 10^{9}\, $g s$^{-1}$., Accepted for publication in MNRAS. 11 pages, 10 figures, 6 tables

The Gaia DR2 radial velocity sample (GDR2RVS), which provides six-dimensional phase-space information on 7.2 million stars, is of great value for inferring properties of the Milky Way. Yet a quantitative and accurate modelling of this sample is hindered without knowledge and inclusion of a well-characterized selection function. Here we derive the selection function through estimates of the internal completeness, i.e. the ratio of GDR2RVS sources compared to all Gaia DR2 sources (GDR2all). We show that this selection function or "completeness" depends on basic observables, in particular the apparent magnitude GRVS and colour G-GRP, but also on the surrounding source density and on sky position, where the completeness exhibits distinct small-scale structure. We identify a region of magnitude and colour that has high completeness, providing an approximate but simple way of implementing the selection function. Context. Gaia Early Data Release 3 (Gaia EDR3) provides accurate astrometry for about 1.6 million compact (QSO-like) extragalactic sources, 1.2 million of which have the best-quality five-parameter astrometric solutions., Aims. The proper motions of QSO-like sources are used to reveal a systematic pattern due to the acceleration of the solar systembarycentre with respect to the rest frame of the Universe. Apart from being an important scientific result by itself, the acceleration measured in this way is a good quality indicator of the Gaia astrometric solution., Methods. Theeffect of the acceleration was obtained as a part of the general expansion of the vector field of proper motions in vector spherical harmonics (VSH). Various versions of the VSH fit and various subsets of the sources were tried and compared to get the most consistent result and a realistic estimate of its uncertainty. Additional tests with the Gaia astrometric solution were used to get a better idea of the possible systematic errors in the estimate., Results. Our best estimate of the acceleration based on Gaia EDR3 is (2.32 +/- 0.16) x 10(-10) m s(-2) (or 7.33 +/- 0.51 km s(-1) Myr-1) towards alpha = 269.1 degrees +/- 5.4 degrees, delta = -31.6 degrees +/- 4.1 degrees, corresponding to a proper motion amplitude of 5.05 +/- 0.35 mu as yr(-1). This is in good agreement with the acceleration expected from current models of the Galactic gravitational potential. We expect that future Gaia data releases will provide estimates of the acceleration with uncertainties substantially below 0.1 mu as yr(-1).

Synthetic organic reactions are a fundamental enabler of small-molecule drug discovery, and the vast majority of medicinal chemists are initially trained--either at universities or within industry--as synthetic organic chemists. The sheer breadth of synthetic methodology available to the medicinal chemist represents an almost endless source of innovation. But what reactions do medicinal chemists use in drug discovery? And what criteria do they use in selecting synthetic methodology? Why are arrays (small focused libraries) so powerful in the lead-optimization process? In this Minireview, we suggest some answers to these questions and also describe how we have tried to expand the number of robust reactions available to the medicinal chemist.

Aryl aminopyrazole amides capped with N-alkylbenzamides 13-16 are selective glucocorticoid receptor agonists. 2,6-Disubstituted benzamides have prednisolone-like potency or better in vitro. Good oral exposure was demonstrated in the rat, with compounds with lower lipophilicity, for example N-hydroxyethyl benzamides (e.g., 16e).

The efficient synthesis of an α-trifluoromethyl-α-tosyloxymethyl epoxide is reported. This highly versatile building block may be reacted sequentially with two different nucleophiles to furnish α-trifluoromethyl tertiary alcohols. Furthermore, the two enantiomers of this key intermediate have been separated using chiral HPLC and the stereochemistry shown to be conserved during subsequent chemical manipulations. Finally, an enzyme-driven desymmetrisation approach has been successfully employed to confer chirality on an intermediate in the sequence.

Synthetic organic reactions are a fundamental enabler of small-molecule drug discovery, and the vast majority of medicinal chemists are initially trained--either at universities or within industry--as synthetic organic chemists. The sheer breadth of synthetic methodology available to the medicinal chemist represents an almost endless source of innovation. But what reactions do medicinal chemists use in drug discovery? And what criteria do they use in selecting synthetic methodology? Why are arrays (small focused libraries) so powerful in the lead-optimization process? In this Minireview, we suggest some answers to these questions and also describe how we have tried to expand the number of robust reactions available to the medicinal chemist.

A novel series of indazole non-steroidal glucocorticoid receptor agonist has been discovered. This series features a sulfonamide central core and meta amides which interact with the extended ligand binding domain. This series has produced some of the most potent and least lipophilic agonists of which we are aware such as 20a (NFκB pIC50 8.3 (100%), c log P 1.9). Certain analogues in this series also display evidence for modulated pharmacology.

Crystallography and computer modeling have been used to exploit a previously unexplored channel in the glucocorticoid receptor (GR). Highly potent, nonsteroidal indazole amides showing excellent complementarity to the channel were designed with the assistance of the computational technique AlleGrow. The accuracy of the design process was demonstrated through crystallographic structural determination of the GR ligand-binding domain–agonist complex of the D-prolinamide derivative 11 . The utility of the channel was further exemplified through the design of a potent phenylindazole in which structural motifs, seen to interact with the traditional GR ligand pocket, were abandoned and replaced by interactions within the new channel. Occupation of the channel was confirmed with a second GR crystal structure of this truncated D-alaninamide derivative 13 . Compound 11 displays properties compatible with development as an intranasal solution formulation, whereas oral bioavailability has been demonstrated with a related truncated exemplar 14 . Data with the pyrrolidinone amide 12 demonstrate the potential for further elaboration within the “meta” channel to deliver compounds with selectivity for the desired transrepressive activity of glucocorticoids. The discovery of these interactions with this important receptor offers significant opportunities for the design of novel GR modulators.

Neighborhood behavior describes the extent to which small structural changes defined by a molecular descriptor are likely to lead to small property changes. This study evaluates two methods for the quantification of neighborhood behavior: the optimal diagonal method of Patterson et al. and the optimality criterion method of Horvath and Jeandenans. The methods are evaluated using twelve different types of fingerprint (both 2D and 3D) with screening data derived from several lead optimization projects at GlaxoSmithKline. The principal focus of the work is the design of chemical arrays during lead optimization, and the study hence considers not only biological activity but also important drug properties such as metabolic stability, permeability, and lipophilicity. Evidence is provided to suggest that the optimality criterion method may provide a better quantitative description of neighborhood behavior than the optimal diagonal method.

Various approaches using Diels–Alder chemistry have been established for the synthesis of truncated carbocyclic analogues 4 and 6 of 4-guanidino-Neu5Ac2en. In the case of compound 4, elaboration of an initial adduct from Danishefsky's diene and the dienophile 7 allowed access to the key enone 26. Methylenation of the carbonyl group and azide-induced opening of an intermediate oxazoline established the required framework regio- and stereo-specifically. Compounds 4 and 6 were found to retain interesting levels of antiviral activity comparable to those shown by their oxygen-containing counterparts.

An evaluation of five commercially available test kit procedures which measure free available chlorine has been conducted on both synthetic water (chemically treated distilled water) and five natural waters. Synthetic water samples were used to evaluate parameters of interest, such as accuracy and precision, as influenced by temperature and pH. The ability of test kit procedures to differentiate free available chlorine from other previously reported chlorine test interfering ions, Mn(+2) and NO2(-), and the compounds mono- and dichloramine was also studied. The primary objective of the natural water studies was to evaluate the test kit procedures on a variety of natural waters that could be encountered around the world. Accuracy, precision and specificity for free available chlorine were evaluated. Procedural modifications should be attempted in order to increase the specificity of the N,N-diethyl-p-phenylene diamine procedure and to improve the accuracy of the Syringaldazine (Liquid) procedure, without adversely affecting their other qualities. The design and operation of an adiabatic low‐temperature calorimeter employing a somewhat different design of automatic shield control is described. Thermocouple difference is sensed by a dc μv amplifier, which is followed in the circuit by a recorder‐controller and a Leeds & Northrup Series 60 (current adjusting type) control unit. The output of the latter is used to control the voltage across the shield heaters through a Westinghouse ``Magamp'' saturable reactor. This design has been used successfully with a conventional low‐temperature adiabatic calorimeter (50‐ml sample size). In most cases the recorder showed no variation from zero (±0.002°) during heating and equilibrating periods, while momentary disturbance at ``on'' and ``off'' was less than 0.05°.

The Medicare Plus project of the Oregon Region Kaiser-Permanente Medical Care Program was designed as a model for prospective payment to Increase Health Maintenance Organization (HMO) participation in the Medicare program. The project demonstrated that it is possible to design a prospective payment system that costs the Medicare program less than services purchased in the community from fee-for-service providers; would provide appropriate payment to the HMO; and in addition, creates a “savings” to return to beneficiaries in the form of comprehensive benefits to motivate them to enroll in the HMO. Medicare Plus was highly successful in recruiting 5,500 new and 1,800 conversion members into the demonstration, through use of a media campaign, a recruitment brochure, and a telephone information center. Members recruited were a representative age and geographic cross section of the senior citizen population in the Portland, Oregon metropolitan area. Utilization of inpatient services by Medicare Plus members in the first full year (1981) was 1679 days per thousand members and decreased to 1607 in the second full year (1982). New members made an average of eight visits per year to ambulatory care facilities.
