1. Carica papaya Leaf Extract Inhibits SARS-CoV-2 Main Proteases but not Human TMPRSS2: An In-vitro and In-silico Study

Abstract

Carica papaya (CP) leaf is long known for its traditional pharmacological effects against dengue virus and malaria. Therefore, CP could also be a potential solution for the treatment of other infectious diseases, such as coronavirus. In this study, we evaluate the potential effect of the ethanolic CP leaf extract in inhibiting the enzymatic activity of three protein targets in SARS-CoV-2 life cycle, which include the 3-chymotrypsin-like protease (3CLpro), papain-like protease (PLpro) and the human transmembrane protein serine 2 (TMPRSS2). Results demonstrate that CP leaf extract inhibits 3CLpro and PLpro significantly with IC50 of 0.02 microgram/mL and 0.06 microgram/mL, respectively, but it is inactive towards TMPRSS2. Phenol, 2-methyl-5-(1,2,2-trimethylcyclopentyl)-(S)- (17a) and beta-mannofuranoside, farnesyl- (21a) were identified in the extract using GC-MS. These two compounds demonstrated a stronger binding affinity towards the main proteases than TMPRSS2 during the docking simulation, which agrees with the in-vitro study. Further pharmacophore mapping suggests that 17a has a fit score higher than 21a to the SARS-CoV-2 3CLpro pharmacophore model concluding that CP leaf extract has the potential to be developed as a herbal SARS-CoV-2 antiviral agent., Comment: 28 pages, 7 figures, 2 tables

15. Formulation and Characterisation of pH-Sensitive Eudragit L-100 Nanoparticles for Quercetin Delivery in Lung Carcinoma.

Abstract

Quercetin is a flavonoid found abundantly in many vegetables and fruits. Quercetin is known for its antioxidant, anti-inflammatory, and anti-proliferative properties. The present study aims to formulate quercetin nanoparticles (Qu-NPs) for targeting lung carcinoma cells. Qu-NPs were formulated using the pH-sensitive polymer Eudragit® L-100. The physicochemical properties of the Qu-NPs were characterized by dynamic light scattering (DLS), Fourier transform infrared (FT-IR) spectroscopy, X-ray diffraction (XRD), differential scanning calorimetry (DSC), and scanning electron microscopy (SEM). In vitro drug release of Qu-NPs was assessed using the dialysis bag method. Cytotoxic activity of Qu-NPs against non-small cell lung cancer cell line (NCI-H460), and normal human lung fibroblasts cell line (CCD-19Lu) was assessed using the 3-[4,5-dimethylthiazol-2-yl]2,5-diphenyltetrazolium bromide (MTT) assay. DLS results indicated a mean particle size of 242.86 ± 20.61 nm, zeta potential of -18.2 ± 2.97 mV and polydispersion index of 0.266 ± 0.008 for Qu-NPs. FT-IR spectra confirmed the presence of hydrogen bonds between quercetin and the polymer, which is crucial for drug entrapment and release. XRD and SEM analysis demonstrated the amorphous nature of quercetin within the nanoparticles. In vitro drug release studies revealed a sustained drug release across different pH conditions including simulated gastric fluid (pH 1.2), and intestinal fluid (pH 6.8, and pH 7.4). Cytotoxicity assay revealed a potent cytotoxic effect of Qu-NPs against NCI-H460. This study underscores the potential of Eudragit® L-100 nanoparticles (NPs) as carriers for quercetin in cancer therapy. Abstract

In the present, the study on Alzheimer disease is exceptionally important since the number of cases are increasing by almost 10 million every year. According to World Health Organization, Alzheimer disease is one of the major effects of disability and dependency among older people and is currently the seventh leading cause of death among other diseases. There are many experiments conducted by scholars focusing on Alzheimer disease but most of them give little attention to raise awareness on the early detection of Alzheimer disease. Early detection of Alzheimer symptoms can be beneficial to help prevent the development of Alzheimer or slow the progression in people who have symptoms. One of the perspectives that integrate Alzheimer disease in Artificial Intelligence is by using a logic mining model with based Reverse Analysis method that operates in Discrete Hopfield Neural Network. In this paper, a method named Permutation 2 Satisfiability based Reverse Analysis method is implemented to obtain the logical relationship among the factors that contribute towards Alzheimer Disease. The proposed method essentially produces a pattern from analyzing the data and provides an induced logic that represents the possible outcome of the dataset with an average of 95% accuracy. The dataset used in this paper was retrieved from a reputable site of the Alzheimer Disease Neuroimaging Initiative which consists of researchers in the field of dementia science. The investigation shows that several medical factors such as the severity of gross findings in cerebral cortex atrophy, lobar atrophy, and hypopigmentation are highly contributed to the neuropathology in Alzheimer disease classification. 45. Quassinoids from Eurycoma longifoliaas Potential Dihydrofolate Reductase Inhibitors: A Computational Study

Abstract

Background: Quassinoids are degraded triterpene compounds that can be obtained from various species of the Simaroubaceae plant family, including Eurycoma longifolia. Quassinoids are the major compounds in E. longifolia, and they are known to have various medicinal potentials, such as anticancer and antimalarial properties. Dihydrofolate reductase (DHFR) was reported to be one of the important targets for certain anticancer and antimalarial drugs. Twelve quassinoids from E. longifoliawere identified to have anticancer effects based on their IC50 values. This study aimed to evaluate the interactions of these twelve quassinoids with DHFR via Autodock 4.2 software and Biovia Discovery Studio Visualiser.Methods: Twelve quassinoids from E. longifoliaand their interactions with DHFR were evaluated via Autodock 4.2 software and Biovia Discovery Studio Visualiser. Their drug-likeness and pharmacokinetic properties were also assessed using the ADMETlab 2.0 program.Results: The molecular docking results showed that eleven quassinoids showed better docking scores than methotrexate, in which the binding energy (BE) of these quassinoids ranged from - 7.87 to -9.58 kcal/mol. Their inhibition constant (Ki) ranged from 0.095 to 1.71 µM. At the same time, the BE and Ki values for methotrexate were -7.80 kcal/mol and 1.64 µM, respectively.Conclusion: From the analysis, 6-dehydrolongilactone and eurycomalide B are among the twelve compounds that showed great potential as hit-to-lead compounds based on the docking score on DHFR, drug-likeness, and ADMET properties. These results suggest a great potential to pursue validation studies via in vitroand in vivomodels.
