28. Sustained Exposure to Helicobacter pylori Lysate Inhibits Apoptosis and Autophagy of Gastric Epithelial Cells

Abstract

Helicobacter pylori is designated as a class I carcinogen of human gastric cancer following long-term infection. During this process, H. pylori bacteria persist in proliferation and death, and release bacterial components that come into contact with gastric epithelial cells and regulate host cell function. However, the impact of long-term exposure to H. pylori lysate on the pathological changes of gastric cells is not clear. In this study, we aimed to investigate the regulation and mechanisms involved in gastric cell dysfunction following continuous exposure to H. pylori lysate. We co-cultured gastric cell lines GES-1 and MKN-45 with H. pylori lysate for 30 generations, and we found that sustained exposure to H. pylori lysate inhibited GES-1 cell invasion, migration, autophagy, and apoptosis, while it did not inhibit MKN-45 cell invasion or migration. Furthermore, Mongolian gerbils infected with H. pylori ATCC 43504 strains for 90 weeks confirmed the in vitro results. The clinical and in vitro data indicated that sustained exposure to H. pylori lysate inhibited cell apoptosis and autophagy through the Nod1-NF-κB/MAPK-ERK/FOXO4 signaling pathway. In conclusion, sustained exposure to H. pylori lysate promoted proliferation of gastric epithelial cells and inhibited autophagy and apoptosis via Nod1-NF-κB/MAPK-ERK/FOXO4 signaling pathway. In the process of H. pylori-induced gastric lesions, H. pylori lysate plays as an “accomplice” to carcinogenesis.

Abstract

The complete mitochondrial genome of the Tamarisk jird, Meriones tamariscinus, was sequenced. The 16,389bp genome contains 37 genes, typical for rodent mitogenomes, including 22 tRNA genes, 2 rRNA genes, and 13 protein-coding genes. The total GC content of the mitochondrial genome is 36.8%, with a base composition of 34.0% A, 24.5% C, 12.3% G, and 29.2% T. The phylogenetic analysis showed that M. tamariscinus was classified in the genus Meriones, Muridae.

Abstract

Based on the geochemical data obtained from the national project about the prevention and control of soil contamination, this paper explored the properties of soil chemical elements in Huanghuaihai Plain, Shandong Province. The results showed that among the grade-one nutritive elements in soil, organic matter, nitrogen and phosphorus were relatively deficient while potassium was rich. Meanwhile, as the grade-two nutritive elements, calcium oxide and magnesium oxide were relatively short and sulfur's content was abundant. About the other beneficial and trace nutritive elements, iron oxide, manganese, molybdenum and boron were deficient, but the content of chlorine was high, hardly lack. The main barriers to improving land productivity were soil salinization and soil heavy metal contamination. The values of soil integrated fertility index that most of the soil in the study area is middle-lower fertilized. Specifically, the low fertility area and lower fertility area are 6 1604 and 1 244 km2 respectively, occupying about 97.43% and 1.97% of the total area. The moderate fertility soil has an area of 172 km2, occupying about 0.27% of the total area. The higher fertility soil covers an area of 128 km2, while the high fertility area of only 76 km2. This article proposed scientific fertilization, elimination of soil obstacle, remediation of heavy-metal-contaminated soil and other effective measures to improve land productivity according to the basic investigation results, which provides a good technological support for the planning and development of good-quality and high-benefit agriculture.
