17. Phosphorus(III)-assisted regioselective C-H silylation of heteroarenes.

Abstract

Heteroarenes containing carbon-silicon (C-Si) bonds are important building blocks that play an important role in the construction of natural products, pharmaceuticals, and organic materials. In this context, the C-H silylation of heteroarenes is a topic of intense interest. Indole C-H silylation can preferentially occur at the nucleophilic C3 and C2 position (pyrrole core), while accessing the C4-C7 positions (benzene core) of the indole remains highly challenging. Here, we show a general strategy for the regioselective C7-H silylation of indole derivatives. Mainly, the regioselectivity is determined by strong coordination of the palladium catalyst with phosphorus (III) directing group. Using this expedient synthetic strategy, the diverse C7-silylated indoles are synthesized effectively which exhibits the broad functional group compatibility. Moreover, this protocol also been extended to other heteroarenes such as carbazoles. The obtained silylated indoles have been employed in various transformations to enable the corresponding differently functionalized indole derivatives. Significantly, a cyclopalladated intermediate is successfully synthesized to test the hypothesis about the P(III)-directed C-H metalation event. A series of mechanistic experiments and density functional theory (M06-2X) calculations has shown the preferred pathway of this directed C-H silylation process.

21. LncRNA SNHG3 promoted cell proliferation, migration, and metastasis of esophageal squamous cell carcinoma via regulating miR-151a-3p/PFN2 axis

Abstract

Esophageal squamous cell carcinoma (ESCC) is an aggressive malignant tumor with a poor prognosis. The dysregulation of long non-coding RNAs (lncRNAs) is closely related to the tumorigenesis and progression of ESCC. However, the effects of lncRNA small nucleolar RNA host gene 3 (lncRNA SNHG3) in ESCC are still unclear. Therefore, a series of experiments methods, such as quantitative real-time polymerase chain reaction, function gain/loss experiments, western blots, and animal xenograft tumor model, were employed to explore the biological function and molecular mechanism of SNHG3 in ESCC. As results, we first reported that SNHG3 was significantly up-regulated in ESCC tissues and cells. SNHG3 knockdown obviously inhibited cell proliferation, migration, invasion, and promoted apoptosis. Mechanism analysis revealed that SNHG3 sponged miR-151a-3p to regulate PFN2. Inhibition of miR-151a-3p and overexpression of PFN2 attenuated the positive effect of SNHG3 knockdown on suppressing tumor progression. Furthermore, the anti-tumor effects of SNHG3 knockdown were also observed in vivo. In summary, our results indicated that SNHG3 knockdown suppressed tumor development via the miR-151a-3p/PFN2 axis, and targeting SNHG3 may provide a new opportunity for ESCC patients.

Abstract

Ketones, prevalent in many biologically significant molecules, require the development of novel methods to synthesize these structures, which is a critical endeavor in organic synthesis. Transition metal catalysis has proven to be an effective method for synthesizing ketones. However, the scope of these substrates remains relatively limited, particularly due to their incompatibility with sensitive functional groups. Herein, we report a Ni-catalyzed three-component 1,2-carboacylation of alkenes, which activates secondary/tertiary alkyl bromides. This method offers significant advantages: simplicity of operation, ready availability of substrates, and broad substrate applicability. Abstract

Direct and selective installation of two different functional groups into the unsaturated bonds of alkenes constitutes a long-standing goal in organic synthesis. Herein, we first report a photoelectrochemical approach for the carboacylation/silanoylation of alkenes with unactivated alkanes/hydrosilanes via the LMCT process. A number of ketones with high complexity and diversity were rapidly obtained from commercially available materials in an economical and sustainable manner. Furthermore, this protocol can be scaled up to the gram-level. This paired electrocatalysis strategy features excellent regioselectivity, and ample substrate scope, is external-oxidant-free, and features a high resource economy. A series of mechanistic experiments were performed to determine the preferred pathway for the photoelectrochemical Ni(I)/Ni(II)/Ni(III) cycles in these transformations. [ABSTRACT FROM AUTHOR]

48. Visual Landmark-Aided LiDAR–Inertial Odometry for Rail Vehicle

Abstract

The precise localization of rail vehicles is crucial for their secure operation. In this article, we present a visual landmark-assisted light detection and ranging (LiDAR) and inertial fusion localization scheme, which adeptly addressing the key issues of absent global navigation satellite system (GNSS) signal and loopback-free areas encountered in railroad. To obtain a reliable visual landmark, we design a novel easy-to-deploy trackside kilometer post. Advanced deep learning-based programs are employed for its identification and character detection. View cone and deskewed point cloud plane are combined to achieve accurate positioning of the kilometer post’s vertices. Furthermore, we construct a positioning framework based on fast direct LiDAR-inertial odometry (FAST-LIO2) that adapts to the train operating environment. This framework employs an error-state extended kalman filter (ESKF) to fuse LiDAR and inertial data as a frontend, constructs a factor graph for optimizing the multimodal information to obtain the best position estimation at the backend. It is worth noting that we leverage kilometer post plane factor and virtual vertex factor to carry out trajectory correction. Finally, our practical experiments demonstrate the excellent performance of the proposed method, achieving meter-level localization errors over long sequence of 8 km. Kilometer post identification and digit extraction excel in a variety of complex situations. Thanks to the auxiliary role of the kilometer post, it still provides stable and accurate position estimation even in severely degraded tunnel scenarios. Also, it can meet the real-time localization requirements of trains.
