To investigate vascular endothelium damage in rats exposed to hypoxic and cold and the effect of salidroside in protecting against this damage. A rat isolated aortic ring hypoxia/cold model was established to simulate exposure to hypoxic and cold. The levels of endothelial cell injury markers were measured by ELISA. TEM was performed to observe the ultrastructure of vascular ring endothelial cells. In vitro assays were performed to verify the effect of salidroside on endothelial cells. CCK-8 and flow cytometry were performed to analyze endothelial cell survival and apoptosis, respectively. Ca2+ concentrations were measured by Flow cytometry, and the expressions of NOS/NO pathway-related proteins were measured by WB. Endothelial cell damage, mitochondrial swelling, autophagy, and apoptosis were increased in the hypoxia group and hypoxia/hypothermia group. All of these effects were inhibited by salidroside. Moreover, exposure to cold combined with hypoxia reduced the NO levels, Ca2+ concentrations and NOS/NO pathway-related protein expression in the hypoxia group and hypoxia/hypothermia group. Salidroside treatment reversed these changes. Salidroside protected against endothelial cell injury induced by cold and hypoxia through reduction of Ca2+-CaM-CAMKII-dependent eNOS/NO activation, thereby preventing mitochondrial damage, reducing ROS levels, and inhibiting apoptosis. Turmeric extract is a β-diketone polyphenol extract isolated from theCurcuma longa L, a plant of the genus Curcuma of the Zingberaceae family. It mainly includes curcumin (CUR), demethoxycurcumin (DMC), and bisdemethoxycurcumin (BDMC). It is a commonly used food, natural pigment and cosmetic additive, which has various pharmacological activities, such as hypolipidemia, antioxidant, anti-inflammation, antiviral and anti-Alzheimer's disease. However, poor stability and low bioavailability limit the clinical application of curcuminoids. Curcumin hydrogenated metabolites also have pharmacological activity, and they are more stable and bioavailable than curcumin. The metabolites of curcuminoids (CUR, DMC and BDMC) in turmeric extract in rats were identified. Turmeric extract was suspended in 0.5% carboxymethlcellulose sodium (CMC-Na). Healthy male Sprague Dawley (SD) rats in drug group were gavaged with a single dose of 1 703 mg/kg turmeric extract (approximately 480 mg/kg curcuminoids), and the rats in control group were given equal volume of 05% CMC-Na solution. And then, plasma at different time points was collected after oral gavage. Urine, faeces and bile were collected after oral gavage in rats within 12 h. Plasma were prepared by protein precipitation method, the urine, faeces and bile were extracted by methanol. The biological samples of faeces, urine, bile and plasma were analyzed by ultra high performance liquid chromatography-quadrupole extractive orbitrap mass spectrometry (UPLC-Q-Extractive MS) in negative ion mode. The accurate molecular weight, primary and secondary mass spectrometry information of metabolites in faeces, urine, bile and plasma were identified. As a result, a total of 59 metabolites (B1-W12) were identified. B3, B7, B8, B11, B12, B13, C3, D3, D5, D8, D10, W1, W2, W4, W5, W7 and W10 were new metabolites. W3/W6, W11/W12 and W12 were firstly found in plasma, faces and urine, respectively. The results showed that the main metabolic pathways of curcumin were glucuronidation, sulfation, reductive hydrogenation, oxidation, reduction, hydroxylation, dehydroxylation and their composite reactions. This method can provide a theoretical basis for the later screening of active metabolites of curcumin compounds.

Introduction The genus Dasypyrum contains two species, namely the annual D. villosum (L.) P. Candargy (2x = 2n = 14) and the perennial D. breviaristatum (L.) Frederiksen (2x = 2n [...], The perennial species Dasypyrum breviaristatum (genome [V.sup.b]) contains many potentially valuable genes for the improvement of common wheat. Construction of a detailed karyotype of D. breviaristatum chromosomes will be useful for the detection of Dasypyrum chromatin in wheat background. We established the standard karyotype of 1[V.sup.b]-7[V.sup.b] chromosomes through nondenaturing fluorescence in situ hybridization (ND-FISH) technique using 28 oligonucleotide probes from the wheat - D. breviaristatum partial amphiploid TDH-2 (AABB[V.sup.b][V.sup.b]) and newly identified wheat - D. breviaristatum disomic translocation and addition lines D2138 (6[V.sup.b]S.2[V.sup.b]L), D2547 (4[V.sup.b]), and D2532 (3[V.sup.b]S.6[V.sup.b]L) by comparative molecular marker analysis. The ND-FISH with multiple oligo probes was conducted on the durum wheat - D. villosum amphiploid TDV-1 and large karyotype differences between D. breviaristatum and D. villosum was revealed. These ND-FISH probes will be valuable for screening the wheat - Dasypyrum derivative lines for chromosome identification, and the newly developed wheat - D. breviaristatum addition lines may broaden the gene pool of wheat breeding. The differences between D. villosum and D. breviaristatum chromosomes revealed by ND-FISH will help us understand evolutionary divergence of repetitive sequences within the genus Dasypyrum. Alzheimer's Disease (AD) is a progressive neurodegenerative disorder affecting millions of people worldwide, with no effective treatment currently available. In recent decades, various traditional Chinese medicines (TCMs) and their active ingredients have shown the potential to attenuate the pathogenesis of AD in cellular and animal models. However, the effects of TCM formulas, which are typically administered in practice, have been less studied. This study aims to investigate the therapeutic effects of several formulas consisting of 4 components herbal components: catalpol, puerarin, gastrodin, and borneol, on streptozotocin (STZ)-induced AD models in cells and rats. The new object recognition (NOR), elevated plus maze (EMP), and Morris water maze (MWM) tests were used to evaluate the cognitive functions of rats. Golgi staining, Haematoxylin and Eosin (HE) staining, and Nissl staining analyses were employed assess the physiology of hippocampal tissues. Gene expression profiles were analyzed used transcriptomics and reverse transcription quantitative polymerase chain reaction analysis, while protein expression levels were determined using immunoblotting, immunohistochemical, and immunofluorescence. The production of cytokines was evaluated with enzyme-linked immunosorbent assay. The results demonstrated that the combined administration of these 4 components (CPGB) had superior mitigating effects on AD cell model, as evidenced by the reduced pro-inflammatory cytokine production and decreased deposition of Aβ protein. Further in vivo and in vitro experiments confirmed that varying doses of CPGB formula effectively ameliorated STZ-induced cognitive deficits, as shown by NOR, MWM, and EMP tests, as well as pathological changes in hippocampal tissues and a 3-dimensional brain neurovascular unit (3D-NVU) model, including decreased deposition of Aβ protein and formation of plaques. Transcriptome sequencing and analysis identified 35 genes with significantly altered expression levels due to STZ and CPGB treatment in hippocampal tissues, which were enriched in the Tlr4/Myd88/NF-κB signaling pathway. Interference with this pathway significantly influenced the therapeutic effects of CPGB in the 3D-NVU model. Collectively, these findings suggest that the combined administration of catalpol, puerarin, gastrodin, and borneol offers superior therapeutic effects on AD by modulating the Tlr4/Myd88/NF-κB signaling pathway. As an essential part of integrated circuit, chip manufacturing has increasingly demanding requirements for quality, cost and others. Some technical limitations and the development direction of miniaturisation and high density have led to defects in the manufacturing process, which affect the performance and life of the chip. Therefore, the detection, identification and classification of defects are very important to improve the chip reliability. Based on the orderly and concise literature research, this paper focuses on a comprehensive review of the main research results in the field of non-contact non-destructive testing imaging methods for chip defects. Firstly, various defects that may occur in the chip design and packaging process are introduced. Then the contact detection methods including manual and probe method are clarified, and the principles, properties and development applications of NDT methods are introduced in detail, also the defect identification methods after imaging are briefly introduced. Moreover, the advantages and limitations of NDT technologies are summarised through comparative studies. Finally, the future development trend of non-destructive testing methods is predicted. This work hopes to provide some reference for the non-destructive testing of chips, which is of great significance for the health testing of chips in the electronics manufacturing industry.
