Abstract

Herpes simplex virus (HSV) persists in its human host and evades the immune response by undergoing a latent infection in sensory neurons, from which it can reactivate periodically. HSV expresses >80 gene products during productive ('lytic') infection, but only the latency-associated transcript (LAT) gene is expressed at abundant levels during latent infection. The LAT gene has been shown to repress lytic-gene expression in sensory neurons. In this study, we use chromatin immunoprecipitation to show that HSV lytic-gene promoters become complexed with modified histones associated with heterochromatin during the course of establishment of latent infection. Experiments comparing LAT-negative and LAT-positive viruses show that a function encoded by the LAT gene increases the amount of dimethyl lysine 9 form of histone H3 or heterochromatin and reduces the amount of dimethyl lysine 4 form of histone H3, a part of active chromatin, on viral lytic-gene promoters. Thus, HSV, and in particular the HSV LAT gene, may manipulate the cellular histone modification machinery to repress its lytic-gene expression and contribute to the persistence of its genome in a quiescent form in sensory neurons. chromatin modification | gene regulation | herpes simplex virus

Abstract

In this study, we investigated the immune enhancing effects of different adjuvants used in a pentavalent vaccine for turbots. The pentavalent vaccine consisted of inactive bacterial cells from five common pathogenic strains (Vibrio anguillarum, Vibrio scophtalmi, Edwardsiella tarda, Vibrio harveyi and Vibrio alginolyticus) and the adjuvants were astragalus polysaccharides (APS), propolis, and the Freund's complete adjuvant (FCA). Turbots were immunized with the pentavalent vaccine alone or with one of the adjuvants, and the immune efficiency was evaluated by measuring the activities of lysozyme (LSZ) and superoxide dismutase (SOD), and serum antibody titers. Fish were also challenged with the pathogens after immunization and the relative percent survival (RPS) was assessed. Our results showed that APS, propolis, and FCA had significant immune-enhancing effects on turbots as shown by the higher titers of antibodies against the pathogens, increased LSZ and SOD activities, and enhanced RPS after challenge with pathogens. Among the three adjuvants, FCA had the most significant immune synergistic effects with the vaccine, and APS and propolis had lower and similar immune synergies.

Abstract

White spot syndrome virus (WSSV) is one of the major pathogens in cultured shrimp. However, up to now, the mechanisms of virus attachment and entry into shrimp cells are poorly understood. Recent research has indicated that WSSV-VP37 is one of the virus attachment proteins of WSSV. Therefore it is important to express the WSSV-VP37 gene in order to determine how it binds to the shrimp cells and interacts in the shrimp. In this study, Pichia pastoris was used as a host to express the WSSV-VP37 gene. The VP37 gene was amplified from the genomic DNA of a Chinese isolate of WSSV and cloned in P. pastoris expression vector pGAPZa-A. SDS-PAGE and Western blot analyses showed that VP37 migrated as a single band with a molecular weight of 37 kDa, and that mouse anti-(His)x6-antibodies bound specifically to VP37. The culture supernatant of the VP37 expression strain, SMD1168/pGAPZaA-VP37, was purified to greater than 95% homogeneity using ammonium sulfate precipitation and an Ni-NTA column. Binding assay by ELISA and fluorescence microscopy in vitro indicated that VP37 is capable of binding to shrimp cells. These results indicate that the P. pastoris expression system is useful for production of bioactive WSSV-VP37 and that WSSV-VP37 is an attachment protein in the infection of shrimp.

Abstract

Eight hundred and thirty eight base pair fragment of mitochondrial COI gene of wild and cultured populations (CP1, CP4, CP5 and CP6) of Fenneropenaeus chinensis was amplified and sequenced. The A, T, G and C contents of the sequence were 235 bp (28.0%), 307 bp (36.6%), 138 bp (16.5%) and 158 bp (18.9%), respectively. Furthermore, 556 bp fragment of the sequence was used to discuss the phylogenetic relationship among 14 Penaeidae species using Alpheus armillatus as the outgroup. From the molecular phylogenetic tree constructed by neighbor-joining method, we obtained three large shrimp groups: Farfantepenaeus, Litopenaeus and Fenneropenaeus group. The results also indicated that there were a closer genetic relationships between F. aztecus and F. paulensis, L. schmitti and L. setiferus, F. indicus and F. merguiensis, and the genus Farfantepenaeus was closer to Litopenaeus.

Abstract

We introduce the concepts of information, uncertainty information, systems and uncertainty systems, analyze the intension of these concepts, and point out the differences and connections among various concepts of systems. We put forward a mathematical method to research uncertainty systems and present a problem that can be solved with our method but cannot be solved with the interval analyzing method. Otherwise, from analyzing the purpose of introducing the concepts of information, uncertainty information, systems and uncertainty systems, we conclude that uncertainty information and uncertainty systems are among the most important subjects studied in scientific research, especially in applied research, both that being presently conducted and the abundance which is to come in the future.
