Spatial transcriptomics extends single-cell RNA sequencing (scRNA-seq) by providing spatial context for cell type identification and analysis. Imaging-based spatial technologies such as multiplexed error-robust fluorescence in situ hybridization (MERFISH) can achieve single-cell resolution, directly mapping single-cell identities to spatial positions. MERFISH produces a different data type than scRNA-seq, and a technical comparison between the two modalities is necessary to ascertain how to best integrate them. We performed MERFISH on the mouse liver and kidney and compared the resulting bulk and single-cell RNA statistics with those from the Tabula Muris Senis cell atlas and from two Visium datasets. MERFISH quantitatively reproduced the bulk RNA-seq and scRNA-seq results with improvements in overall dropout rates and sensitivity. Finally, we found that MERFISH independently resolved distinct cell types and spatial structure in both the liver and kidney. Computational integration with the Tabula Muris Senis atlas did not enhance these results. We conclude that MERFISH provides a quantitatively comparable method for single-cell gene expression and can identify cell types without the need for computational integration with scRNA-seq atlases.

Green hydrogen is essential to achieving carbon neutrality, and solid oxide electrolysis cells can produce hydrogen using renewable power and waste heat. Insufficient long-term durability of solid oxide electrolysis cells has impeded their commercialization. Here, coatings in the porous stainless steel support of metal-supported solid oxide electrolysis cells (MS-SOECs) are used to dramatically improve their performance and durability. The long-term degradation rate of uncoated MS-SOECs is highly dependent on the current density, with the fastest degradation occurring at the highest current density tested, 0.5 A cm-2. At this current density, coatings are quite effective. Three protective coatings, Co3O4deposited by atomic layer deposition (ALD), Co3O4deposited by electroplating deposition (ED), and CuMn1.8O4(CMO) deposited by electrophoretic deposition (EPD), are explored to enhance the performance of MS-SOECs with La0.6Sr0.4Co0.2Fe0.8O3-Sm0.2Ce0.8O3(LSCF-SDC) as the oxygen catalyst and SDC-Ni as the fuel catalyst. The initial average current density at 1.4 V is increased with coatings. It is 0.83 mA cm-2for the ALD cells, 1.05 mA cm-2for the ED cells, and 1.13 mA cm-2for the EPD cells, compared to 0.65 mA cm-2for the bare cells at 700 °C with 50% H2-50% H2O. The degradation rate over 1000 h of continuous operation is reduced from 36% to 26%, 27%, and 19% kh-1with the three coatings, respectively. These improvements are ascribed to reduced Cr poisoning on the oxygen catalyst, which is one of the primary degradation modes for this type of MS-SOEC.

Hepatitis A virus (HAV) is a positive-sense RNA virus causing acute inflammation of the liver. Here, using a genome-scale CRISPR screen, we provide a comprehensive picture of the cellular factors that are exploited by HAV. We identify genes involved in sialic acid/ganglioside biosynthesis and members of the eukaryotic translation initiation factor complex, corroborating their putative roles for HAV. Additionally, we uncover all components of the cellular machinery for UFMylation, a ubiquitin-like protein modification. We show that HAV translation specifically depends on UFM1 conjugation of the ribosomal protein RPL26. Furthermore, we find that components related to the yeast Trf4/5-Air1/2-Mtr4 polyadenylation (TRAMP) complex are required for viral translation independent of controlling viral poly(A) tails or RNA stability. Finally, we demonstrate that pharmacological inhibition of the TRAMP-like complex decreases HAV replication in hepatocyte cells and human liver organoids, thus providing a strategy for host-directed therapy of HAV infection.

Metal-supported solid oxide fuel cells with symmetric architecture, having metal supports on both sides of the cell, are scaled up from button cell size to large 50 cm2 active area cell size. The cells remain flat after sintering assisted by the symmetric structure. Equivalent performance is achieved for button cells and large cells, and thermal cycling and redox cycling tolerance are demonstrated for the large cells. The catalyst infiltration process is improved to enable high-throughput manufacturing. The cumbersome lab-scale molten nitrate infiltration process is replaced with a room-temperature process in which a shelf-stable aqueous solution of nitrate salts is applied to the cell by spraying, painting, or other scalable techniques. A fast-ramp thermal conversion of the nitrate salts to the final oxide catalyst composition is implemented, allowing many infiltration cycles to be accomplished in a single work shift. Increasing the number of infiltration cycles from 5 to 10 led to an increase in peak power density from approximately 0.3 to 0.52 W cm−2.

The Coronaviridae are a family of viruses that cause disease in humans ranging from mild respiratory infection to potentially lethal acute respiratory distress syndrome. Finding host factors common to multiple coronaviruses could facilitate the development of therapies to combat current and future coronavirus pandemics. Here, we conducted genome-wide CRISPR screens in cells infected by SARS-CoV-2 as well as two seasonally circulating common cold coronaviruses, OC43 and 229E. This approach correctly identified the distinct viral entry factors ACE2 (for SARS-CoV-2), aminopeptidase N (for 229E), and glycosaminoglycans (for OC43). Additionally, we identified phosphatidylinositol phosphate biosynthesis and cholesterol homeostasis as critical host pathways supporting infection by all three coronaviruses. By contrast, the lysosomal protein TMEM106B appeared unique to SARS-CoV-2 infection. Pharmacological inhibition of phosphatidylinositol kinases and cholesterol homeostasis reduced replication of all three coronaviruses. These findings offer important insights for the understanding of the coronavirus life cycle and the development of host-directed therapies.

Hydrogen is a renewable energy carrier, and electrolysis to split water is the most environmentally friendly method to produce hydrogen. This work reports long-term durability and degradation mode analysis for metal-supported solid oxide electrolysis cells (MS-SOECs). Catalyst screening showed that MS-SOECs with composite electrode catalysts (samarium-doped ceria-nickel [SDC-Ni] serving as a fuel electrode catalyst, and praseodymium oxide [PrOx]-SDC or La0.6Sr0.4Co0.2Fe0.8O3 [LSCF]-SDC serving as an air electrode catalyst) exhibit the highest electrochemical performance at 700 °C. The degradation rate of cells with LSCF-SDC as the air electrode catalyst was as low as 1.3%/100 h in long term durability tests at a current density of 0.33 A cm−2, in contrast to rapid degradation observed for a cell with a PrOx-SDC air electrode. Post-mortem analysis reveals the degradation is dependent on the primary modes of fuel electrode catalyst coarsening and Cr poisoning on the air electrode catalyst, as well as secondary modes of oxidation of the metal support and local elemental accumulation of Ni. Other degradation modes reported in conventional anode-supported SOECs, such as Ni migration, foreign element contamination, delamination of the cell, and nano-voids on the electrolyte, are not observed in the present MS-SOECs.

Protonic ceramic fuel or electrolysis cells (PCFC/PCEC) have shown promising performance at intermediate temperatures. However, these technologies have not yet been demonstrated in a stack, hence the oxidation behavior of the metallic interconnect under relevant operating environments is unknown. In this work, ferritic stainless steels 430 SS, 441 SS, and Crofer 22 APU were investigated for their use as interconnect materials in the PCFC/PCEC stack. The bare metal sheets were exposed to a humidified air environment in the temperature range from 450 °C to 650 °C, to simulate their application in a PCFC cathode or PCEC anode. Breakaway oxidation with rapid weight gain and Fe outward diffusion/oxidation was observed on all the selected stainless steel materials. A protective coating is deemed necessary to prevent the metallic interconnect from oxidizing. To mitigate the observed breakaway oxidation, state-of-the-art protective coatings, Y2O3, Ce0.02Mn1.49Co1.49O4, CuMn1.8O4 and Ce/Co, were applied to the stainless steel sheets and their oxidation resistance was investigated. Dual atmosphere testing further validated the effectiveness of the protective coatings in realistic PCFC/PCEC environments, with a hydrogen gradient across the interconnect. Several combinations of metal and coating material were found to be viable for use as the interconnect for PCFC/PCEC stacks.

High power density and longevity are required to commercialize solid oxide fuel cells for vehicular applications. In this work, electrochemical durability of high-performance metal-supported solid oxide fuel cells (MS-SOFCs) with infiltrated catalysts is investigated. Durability screening of various cathode and anode compositions is conducted. The Pr6O11 cathode and SDCN40 (40 vol% Ni-60 vol% SDC) anode are selected due to preferential tradeoff between performance and durability. X-ray diffraction indicates the catalyst phases are stable after prolonged thermal annealing. Electrochemical impedance and scanning electron microscopy analyses show that the evolution of SDCN40 is minimal, and coarsening and Cr poisoning of the Pr6O11 catalyst are the dominant degradation mechanisms. These degradation mechanisms are addressed by implementing three additional fabrication steps: (1) preoxidation of the metal support, (2) deposition of thin and protective atomic layer deposition (ALD) coatings throughout the cathode-side electrode and support, and (3) in situ pre-coarsening of the catalysts. A favorable tradeoff is achieved: the combination of the three fabrication steps reduces degradation rate at 0.7 V and 700 °C by two orders of magnitude to 2.3% kh−1, while sacrificing 35% of initial power density. The catalyst pre-coarsening step is primarily responsible for the loss of performance.

Proton conducting oxide electrolyte materials could potentially lower the operating temperature of metal-supported solid oxide cells (MS-SOCs) to the intermediate range 400 to 600 °C. The porous metal substrate provides the advantages of MS-SOCs such as high thermal and redox cycling tolerance, low-cost of structural materials, and mechanical ruggedness. In this work, the viability of co-sintering fabrication of metal-supported proton conducting solid oxide cells using BaZr1-x-yCexYyO3-δ (BZCY) is investigated. BZCY ceramics are sintered at 1450 °C in reducing environment alone and supported on Fe–Cr alloy metal support, and key characteristics such as Ba loss, sintering behavior, and chemical compatibility with metal support are determined. Critical challenges are identified for this fabrication approach, including: Contamination of the electrolyte with Si and Cr from the metal support, incomplete electrolyte sintering, and evaporation of electrolyte constituents. Various approaches to overcome these limitations are proposed, and preliminary assessment indicates that the use of barrier layers, low-Si-content stainless steel, and sintering aids warrant further development.

High-temperature electrolysis (HTE) using solid oxide electrolysis cells (SOECs) is a promising hydrogen production technology and has attracted substantial research attention over the last decade. While most studies are conducted on hydrogen electrode-supported type cells, SOEC operation using metal-supported cells has received minimal attention. The development of metal-supported SOECs with performance similar to the best conventional SOECs is reported. These cells have stainless steel supports on both sides, 10Sc1CeSZ electrolyte and electrode backbones, and nano-structured catalysts infiltrated on both hydrogen and oxygen electrode sides. Samarium-doped ceria (SDC) mixed with Ni is infiltrated as a hydrogen electrode catalyst, and the effect of ceria:Ni ratio is studied. On the oxygen electrode side, catalysts including lanthanum strontium manganite (LSM), lanthanum strontium cobalt ferrite (LSCF), praseodymium oxide (Pr6O11), and their composite catalysts with SDC (i.e., LSM-SDC, LSCF-SDC, and Pr6O11-SDC) are compared. Using the materials with highest catalytic activity (Pr6O11-SDC and SDC40-Ni60) and optimizing the catalyst infiltration processes, excellent electrolysis performance of metal-supported cells is achieved. Current densities of −5.31, −4.09, −2.64, and −1.62 A cm−2 are achieved at 1.3 V and 50 vol% steam content at 800, 750, 700, and 650 °C, respectively.

Candidate proton conducting oxides including perovskite oxides BaZr 0.7 Ce 0.2 Y 0.1 O 3−δ , SrZr 0.5 Ce 0.4 Y 0.1 O 3−δ , and Ba 3 Ca 1.18 Nb 1.82 O 9−δ , pyrochlore oxides La 1.95 Ca 0.05 Zr 2 O 7−δ and La 2 Ce 2 O 7 , and acceptor doped rare-earth ortho-niobate La 0.99 Ca 0.01 NbO 4 are synthesized via solid state reactive or sol-gel methods. These ceramics are sintered at 1450 °C in reducing environment alone and supported on Fe-Cr alloy metal support, and their key characteristics such as phase formation, sintering property, and chemical compatibility with metal support are determined. Most electrolyte candidates suffer from one or more challenges identified for this fabrication approach, including: phase decomposition in reducing atmosphere, evaporation of electrolyte constituents, contamination of the electrolyte with Si and Cr from the metal support, and incomplete electrolyte sintering. In contrast, La 0.99 Ca 0.01 NbO 4 is found to be highly compatible with the metal support and co-sintering processing in reducing atmosphere. A metal-supported cell is fabricated with La 0.99 Ca 0.01 NbO 4 electrolyte, ferritic stainless steel support, Pt air electrode and nanoparticulate ceria-Ni hydrogen electrocatalyst. The total resistance is 50 Ω·cm 2 at 600 °C. This work clearly demonstrates the challenges, opportunities, and breakthrough of metal-supported proton-conducting solid oxide cells by co-sintering fabrication.

High power density is required to commercialize solid oxide fuel cells for vehicular applications. In this work, high performance of metal supported solid oxide fuel cells (MS-SOFCs) is achieved via catalyst composition, electrode structure, and processing optimization. The full cell configuration consists of a dense ceramic electrolyte and porous ceramic backbones (electrodes) sandwiched between porous stainless steel metal supports. The conventional YSZ electrolyte and backbones are replaced with more conductive and thinner 10Sc1CeSZ ceramics. MS-SOFCs are co-sintered in a single step and subsequently infiltrated with nanocatalysts. Five categories of cathode catalysts are screened in full cells, including: perovskites, nickelates, praseodymium oxide, binary layered composites, and ternary layered composites. Various anode compositions are also tested. The conventional LSM cathode catalyst is replaced with more active Pr6O11 and the Ni content of the SDC-Ni anode is increased. The resulting cells achieve a peak power of 1.56, 2.0, and 2.85 W cm−2 at 700, 750, and 800 °C, respectively, with 3%H2O/H2 as fuel and cathode exposed to air. Multiple cells show reproducible performance (Pmax = 1.50 ± 0.06 W cm−2) and OCV (1.10 ± 0.02 V). The performance is further increased with cathode exposed to pure oxygen (2.0 W cm−2 at 700 °C).

Objective: To provide evidence for choosing surgical or nonsurgical treatment for epilepsy in patients with unilateral multilobar and hemispheric polymicrogyria (PMG). Methods: We searched published studies until September 2022 related to unilateral multilobar and hemispheric PMG and included patients who were followed up at the Pediatric Epilepsy Centre of Peking University First Hospital in the past 10 years. We summarized the clinical characteristics and compared the long‐term outcomes after surgical or nonsurgical (anti‐seizure medications, ASMs) treatment. Results: A total of 70 patients (49 surgical, 21 non‐surgical) with unilateral multilobar and hemispheric PMG were included. The median age at epilepsy onset was 2.5 years (1.0–4.1). The most common seizure types were focal and atypical absence seizures. In the whole cohort, 87.3% had hemiparesis and 67.1% had electrical status epilepticus during slow sleep (ESES). There were significant differences in age at epilepsy onset, extent of lesion, and EEG interictal discharges between the two groups. At the last follow‐up (median 14.1 years), the rates of seizure‐freedom (81.6% vs. 57.1%, p = 0.032) and ASM discontinuation (44.4% vs. 6.3%, p = 0.006) were higher in the surgical group than in the nonsurgical group. Patients in the surgical group had a higher rate of seizure‐freedom with complete resection/disconnection than with subtotal resection (87.5% vs. 55.6%, p = 0.078), but with no statistically significant difference. In the nonsurgical group, more extensive lesions were associated with worse seizure outcomes. Cognition improved postoperatively in 90% of surgical patients. Significance: In patients with unilateral multilobar and hemispheric PMG, the age of seizure onset, the extent of the lesion and EEG features can help determine whether surgery should be performed early. Additionally, surgery could be more favorable for achieving seizure freedom and cognitive improvement sooner. Plain Language Summary: We aim to summarize clinical characteristics and compare the long‐term outcomes after surgical and nonsurgical (ASM) treatment to provide a basis for treatment decisions for patients with unilateral multilobar and hemispheric polymicrogyria (PMG)‐related epilepsy. We found that patients with unilateral hemispheric and multilobar PMG had significantly higher rates of seizure freedom and ASM discontinuation with surgical treatment than with nonsurgical treatment. In the surgical group, seizure outcomes were better in patients treated with complete resection/disconnection than in those treated with subtotal resection, but the difference was not statistically significant.
