Abstract

Abstract [Objective] Astragalus membranaceus (Fisch.) Bge. var. mongholicus (Bge.) Hsiao is an important primitive plant of astragali radix, and its main planting areas are arid and semi-arid areas in Inner Mongolia, Shanxi, Gansu, and other places. Irregular intermittent rainfalls cause drought and rehydration cycle in plants. Studying the characteristics of metabolites in A . membranaceus var. mongholicus during drought and rehydration is crucial to understand the drought tolerance mechanism in response to natural intermittent rainfalls. [Methods] The seedlings of annual A . membranaceus var. mongholicus were used as materials. After repeated drought and rehydration treatment, soil nutrients and root growth in potted plants were measured. The primary metabolites of A . membranaceus var. mongholicus were analyzed using NMR based technology. Differential metabolite screening and metabolic pathway were analyzed. Total flavonoids, total saponins, and polysaccharides in the roots of A . membranaceus var. mongholicus were determined. [Results] (1) A . membranaceus var. mongholicus showed a trend of decreasing in root diameter and increasing in root length and fibrous root density under drought stress. (2) Under drought and rehydration treatment, a total of 42 metabolites were detected, containing mainly amino acids and their derivatives, organic acids, amines, ammonia compounds, and sugars. Metabolic pathway analysis showed that amino acid metabolism pathways were mainly affected during repeated drought stress. Plant drought resistance was enhanced by increasing the content of aspartate, alanine, glutamate, proline, and arginine, as well as reducing the content of asparagine, tryptophan, and 4-aminobutyrate. Specifically, the increase in metabolites such as alanine, glutamate, and proline was 1-3 times, while metabolites such as tryptophan and 4-aminobutyric acid were decreased by 1-2 times. (3) Under the first round of drought stress, the accumulation of astragaloside was decreased; after the second round of drought stress, the production of three active substances, namely astragaloside, total flavonoids, and polysaccharides, showed a downward trend. [Conclusion] Amino acids and their derivatives are closely related in response to drought stress in A . membranaceus var. mongholicus. Plant regulates its secondary metabolites in response to environmental changes

10. Degradation of amyloid β-peptides catalyzed by nattokinase in vivoand in vitro

Abstract

Amyloid-β 1-42 (Aβ42) plays a pivotal role in Alzheimer disease (AD) pathogenesis. Peripheral clearance of Aβ42 largely affects its level in the brain and affects AD progression. Although nattokinase (NK) degrades Aβ40, the details of NK’s capture of various Aβ species and reduction of plasma Aβ42/Aβ40 are uncharacterized. In this study, the Aβ42/Aβ40-degrading ability of NK was investigated using five Aβs and AD model mice. The C-terminal region of Aβ42/Aβ40 (Gly29 to Val40) was primarily required for NK capture, and the integrated conformation in Aβ42/Aβ40 aggregates was a more efficient target for NK catalysis. Further, suspended Aβ42/Aβ40 oligomers were more easily captured by NK than suspended Aβ42/Aβ40 fibrils, while deposited Aβ42/Aβ40 fibrils recruited more NK than deposited Aβ42/Aβ40 oligomers. Although most NK was likely lost during NK uptake and/or entry into the blood, a small fraction of NK showed good plasma Aβ42/Aβ40-degrading efficacy after entering the blood due to NK’s stability in the plasma of AD mice for at least 9 days. It was concluded that oral administration of NK is a feasible approach for peripheral Aβ42/Aβ40 clearance. This implies that NK might serve as an anti-Aβ42 agent for the treatment of Aβ42/Aβ40-related diseases such as AD.

Abstract

Halloysite (HNT) is a natural inorganic mineral that has many applications in manufacturing. This review (with 192 references) covers (a) the chemical properties of halloysites, (b) the effects of alkali and acid etching on the loading capacity and the release behavior of halloysites, (c) the use of halloysite nanotubes in analytical sciences and drug delivery, and (d) recent trends in the preparation of magnetic HNTs. Synthetic methods such as co-precipitation, thermal decomposition, and solvothermal method are discussed, with emphasis on optimal magnetization. In the analytical field, recent advancements are summarized in terms of applications of HNT-nanocomposites for extraction and detection of heavy metal ions, dyes, organic pollutants, and biomolecules. The review also covers methods for synthesizing molecularly imprinted polymer-modified HNTs and magnetic HNTs. With respect to drug delivery, the toxicity, techniques for drug loading and the various classes of drug-halloysite nanocomposites are discussed. This review gives a general insight on the utilization of HNT in analytical determination and drug delivery systems which may be useful for researchers to generate new ideas.
