7. Angelica sinensis polysaccharide antagonizes D-galactose induced brain aging in rats

Abstract

Objective To explore the antagonistic effect of Angelica sinensis polysaccharide (ASP) on brain aging induced by D-galactose (D-gal) in rats. Methods SD rats were randomly divided into control group, ASP group, aging model group, and ASP aging model group. Their spatial learning and memory abilities were detected by water maze test. The aging of brain tissue cells were detectded by aging-related β-galactosidase (SA-β-Gal) staining. The contents of SOD, MDA and GSH in hippocampal homogenate were measured by chromatography, and those of IL-1β and IL-6 in hippocampal homogenate were detected by ELISA. Southern blotting and TRAP-PCR were used to detect telomere length and telomerase activity in the hippocampal cells. Neural stem cells (NSCs) were isolated, cultured and identified from the brain tissue of SD fetal rats. The third-generation NSCs were divided into control group, ASP group, aging model group, and ASP aging model group. The content of MDA in the cells was determined by chromatography. The intracellular ROS level was determined by flow cytometry. qRT-PCR was used to detect the mRNA expression of p19, p21 and p53 of senescence related genes. Results Compared with the aging model group, the injection of ASP significantly improved the spatial learning and memory abilities of aging rats. The number of SA-β-Gal positive cells in the brain tissue was decreased (P < 0.05). The contents of SOD and GSH in the hippocampus were increased, but that of MDA was decreased (P < 0.05). The secretion levels of IL-1β and IL-6 were decreased (P < 0.05). The telomere length and telomerase activity were increased (P < 0.05). NSCs were successfully isolated and identified. Compared with the aging group, the MDA content and ROS level were significantly reduced in the ASP aging group (P < 0.05), and the mRNA levels of senescence-related genes p19, p21, and p53 were decreased (P < 0.05). Conclusion ASP may regulate cell telomere length and telomerase activity, down-regulate the expression of aging-related genes, reduce the expression of inflammatory factors, improve the cell's antioxidant capacity, and then antagonize the degeneration effect of D-gal on NSCs and hippocampal cells.

Abstract

In the present work, we evaluated different widely accepted methyl derivatization procedures which are almost obligatory in gas chromatography analysis of fatty acids (FAs), both acid- and base-catalyzed ones. We determined their derivatization efficiency and rapidity and found that combined base–acid methylation, the NaOH–BF3–MeOH method, out-performed all other methods. The analyses of FAME standards and samples were performed on a conventional 30 m × 0.25 mm Wax column, with a single run of 51 min. To accelerate the analysis while maintaining resolution, a more polar 0.1 mm ID column was used to analyze 52 FAMEs. This way, we were able to obtain results within one-third of the analysis time with almost equal separation, if not better. To further evaluate the quantification methods that are performed with or without internal standards, we constructed new calibration curves by removing a known quantity from the original calibration curves, and used them to recalculate the concentration of the known removed quantity. Comparison of the calculated and actual concentration values revealed that, when performed carefully, the external standard method could achieve results that were comparable to results obtained with the internal standard method, and thus is recommended for daily analysis.

Multiple sclerosis (MS) is an inflammatory disease of the central nervous system (CNS) and is characterized by the destruction of myelin and axons leading to progressive disability. Peptide epitopes from CNS proteins, such as myelin oligodendrocyte glycoprotein (MOG), possess promising immunoregulatory potential for treating MS; however, their instability and poor bioavailability is a major impediment for their use clinically. To overcome this problem, we used molecular grafting to incorporate peptide sequences from the MOG35-55 epitope onto a cyclotide, which is a macrocyclic peptide scaffold that has been shown to be intrinsically stable. Using this approach, we designed novel cyclic peptides that retained the structure and stability of the parent scaffold. One of the grafted peptides, MOG3, displayed potent ability to prevent disease development in a mouse model of MS. These results demonstrate the potential of bioengineered cyclic peptides for the treatment of MS.
