16. Mechanism of Cryptotanshinone Inhibiting Proliferation of Human Breast Cancer MCF7 Cells

Abstract

Objective To investigate the inhibitory effect of cryptotanshinone (CPT) on human breast cancer cell MCF7 and its mechanism. Methods The survival rate of MCF7 cells was measured by MTT assay. Cell apoptosis was detected by Annexin V/PI assay and Hoechst 33258 fluorescence staining assay. Cell cycle and reactive oxygen species were detected by flow cytometry. Cell migration and invasion were detected by cell scratch test and Transwell chamber test. The surface molecules CD44 and CD24 were detected by flow cytometry and microsphere culture. The expression of cell-associated proteins was detected by Western blot. Results CPT inhibited the proliferation of MCF7 cells in a dose-dependent manner, and the 24 h IC50 value was 19.24 μmol/L. Compared with the untreated group, the CPT-treated group showed cell cycle arrested in the S phase, and apoptosis was induced. The results of the cell scratch and Transwell chamber tests showed that CPT significantly inhibited the migration and invasion of MCF7 cells. Furthermore, CPT reduced the CD24-/LowCD44+ cell population in MCF7 cell-derived microspheres. Western blot results showed that CPT could up-regulate the expression of Bax protein, down-regulate the expression of BCL-2, PI3K-p85, Akt, N-cadherin, Twist1, Sox2, Oct4, and Nanog protein, effectively inhibit the phosphorylation of ER-α, and decrease the expression of ABCG2. Conclusion CPT can inhibit the proliferation of MCF7 cells by inhibiting the migration and invasion of MCF7 cells, decreasing the number of CD24-/lowCD44+ cells and affecting the expression of tumor stem cell-related proteins.

Abstract

Vinegar is traditionally produced by fermentation with a mixed system composed of naturally inoculated microorganisms. The complex microbial communities in the fermentation system are crucial to the formation of vinegar flavor. Studies on the microbial communities and their interactions are helpful to reveal the relationship between microorganisms and the quality of vinegar and are of great significance for targeted regulation of microorganisms in the fermentation process, productivity enhancement and vinegar quality improvement. Therefore, recent studies on the microbial diversity and the interactions among core functional microorganisms in the traditional fermentation process of vinegar are reviewed in this paper. Moreover, the positive effects of the interaction among core functional microorganisms on the flavor of vinegar are discussed. Finally, the problems existing in the current research are summarized. This review is expected to provide a reference for targeted regulation of vinegar quality.

Abstract

In order to prolong the service life of wireless vibration sensors when collecting a large number of high-frequency vibration data, this paper studied the existing vibration data compression algorithms, put forward and analyzed the existing problems, and on this basis, proposed an effective mechanism of K-means clustering-discrete cosine transform (DCT) dual data compression. According to the characteristics of predictive maintenance data, K-means clustering-DCT dual compression algorithm firstly used K-means algorithm to aggregate and classify vibration data, and then carried out DCT compression according to the frequency domain characteristics of vibration signals. The verification results showed that the algorithm significantly improved the data compression efficiency and reduced the transmission of redundant data by aggregating vibration data. In addition, under the condition of the same amount of data, the algorithm had better application performance after improving the peak signal-to-noise ratio compared with other algorithms.

Abstract

When trained on multimodal image datasets, normal Generative Adversarial Networks (GANs) are usually outperformed by class-conditional GANs and ensemble GANs, but conditional GANs is restricted to labeled datasets and ensemble GANs lack efficiency. We propose a novel GAN variant called virtual conditional GAN (vcGAN) which is not only an ensemble GAN with multiple generative paths while adding almost zero network parameters, but also a conditional GAN that can be trained on unlabeled datasets without explicit clustering steps or objectives other than the adversary loss. Inside the vcGAN's generator, a learnable ``analog-to-digital converter (ADC)" module maps a slice of the inputted multivariate Gaussian noise to discrete/digital noise (virtual label), according to which a selector selects the corresponding generative path to produce the sample. All the generative paths share the same decoder network while in each path the decoder network is fed with a concatenation of a different pre-computed amplified one-hot vector and the inputted Gaussian noise. We conducted a lot of experiments on several balanced/imbalanced image datasets to demonstrate that vcGAN converges faster and achieves improved Frech\'et Inception Distance (FID). In addition, we show the training byproduct that the ADC in vcGAN learned the categorical probability of each mode and that each generative path generates samples of specific mode, which enables class-conditional sampling.
