10. Lactobacillus acidophilus alleviates lung inflammation in LPS-induced acute pneumonia in mice: efficacy and mechanism

Abstract

Objective To investigate the effect of Lactobacillus acidophilus (L. acidophilus) on the inflammation of mice with lipopolysaccharide (LPS)-induced acute pneumonia and the possible mechanism. Methods Twenty-one mice were divided into a control group (NC, n=6), a model group (LPS, n=8) and a L. acidophilus-treated group (Probiotics, n=7). The mice in the control group were fed normally, while those in the LPS and Probiotics groups were treated with 10 mg/kg LPS through tracheal drip to establish a mouse model of acute pneumonia, and those in the Probiotics group were given L. acidophilus. All of the mice were executed in 7 d after modeling. The pathological changes of lung tissue were assessed by HE staining and wet/dry ratio (W/D) of lung tissue. ELISA was used to detect the levels of inflammatory factors in bronchoalveolar lavage fluid and the activity of myeloperoxidase (MPO) in lung tissue. 16S rDNA sequencing was employed to observe the diversity of intestinal flora, and immunohistochemical (IHC) staining was applied to determine the expression of ZO-1 and TRPV4. Results The Probiotics group had significantly reduced W/D ratio and IL-1β, TNF-α and IL-6 levels, increased IL-10 level (P < 0.05), diminished MPO activity (P < 0.05), down-regulated TRPV4 expression in colon and lung tissues, and upregulated ZO-1 expression in colon tissue, meanwhile, the abundance of Helicobacter spp. and Desulfovibrio spp. was significantly altered when compared with the LPS group. Conclusion L. acidophilus alleviates lung inflammation in LPS-induced pneumonia in mice, which may be associated with intestinal abundance of Desulfovibrio spp. in the, activity of TRPV4 channel and intestinal mucosal permeability.

Abstract

In eukaryotes, DNA wraps around histones to form nucleosomes, which are compacted into chromatin. DNA-templated processes, including transcription, require chromatin disassembly and reassembly mediated by histone chaperones. Additionally, distinct histone variants can replace core histones to regulate chromatin structure and function. Although replacement of H2A with the evolutionarily conserved H2A.Z via the SWR1 histone chaperone complex has been extensively studied, in plants little is known about how a reduction of H2A.Z levels can be achieved. Here, we show that NRP proteins cause a decrease of H2A.Z-containing nucleosomes in Arabidopsis under standard growing conditions. nrp1-1 nrp2-2 double mutants show an over-accumulation of H2A.Z genome-wide, especially at heterochromatic regions normally H2A.Z-depleted in wild-type plants. Our work suggests that NRP proteins regulate gene expression by counteracting SWR1, thereby preventing excessive accumulation of H2A.Z.

Abstract

Aiming at the requirements of rapid cooling of the burned site in early first-aid treatment of burns, our research team designed a device for cooling the wound surface of burns. To meet the purpose of reducing of wound temperature, swelling and seepage rapidly, the device which uses the principle of urea and water mixed reaction refrigeration consisted of the adsorption of shell, water storage bag, bag and cover body. The device, portable and easy to operate, can withstand high temperature UV rays. It is up to the mustard for outdoor environments lacking water sources such as field battles. At the same time, it reduces the possibility of secondary wounds caused by chemical residues in traditional cooling therapy. (本研究设计了一款烧伤用创伤面降温装置, 运用尿素与水混合反应制冷的原理, 通过吸附壳体、蓄水袋、收容袋和盖体的配合使用以达到迅速降低创面温度, 减少创面肿胀渗液等目的。该装置轻便易于携带, 能耐受高温和紫外线, 适合野战现场等缺乏水源的户外环境, 且简单易操作, 能有效避免创面二次损伤。)
