Objective To construct hns gene overexpression strains of Burkholderia thailandensis BPM, and explore the effects of hns overexpression on bacterial growth, motility, biofilm formation ability and susceptibility to antimicrobial drugs. Methods The hns genes 03253 and 05307 were respectively cloned into the expression vector pTAC-Cm to yield recombinant vectors pTAC-03253 and pTAC-05307. Then the 2 types of plasmids were electroporated into the BPM competent strain to obtain recombinant strains, and their phenotypic changes in growth, motility, biofilm, and drug sensitivity were detected. Results The strains of hns overexpression were successful constructed, and they presented significantly inhibited growth and motility, markedly enhanced ability to form biofilms, decreased sensitivity to levofloxacin and minocycline when compared with the wild-type BPM strain. Conclusion H-NS protein plays an important role in the regulation of virulence and drug resistance in Burkholderia thailandensis BPM, and can become a new target for the treatment of infections caused by Burkholderia thailandensis.

Abstract Purpose This study aimed to evaluate the clinical efficacy of applying real-time dynamic navigation (RDN) in the extraction of deep horizontal mandibular impacted third molars, hypothesizing that RDN reduces surgical time and minimizes the risk of injury to adjacent anatomical structures. Methods A prospective study was conducted on 160 patients aged between 18 and 37 years with deep horizontal impaction of the mandibular third molar. The participants were randomly assigned to either the experimental group (receiving RDN-assisted extractions) or the control group (undergoing traditional extraction methods). Objective To investigate the effects of long non-coding RNA FEZ family zinc finger 1 antisense RNA 1(lncRNA FEZF1-AS1) on enhancer of zeste homolog 2(EZH2) in regulation of proliferation, migration, invasion and epithelial-mesenchymal transition (EMT) of pulmonary interstitial cells and its mechanism. Methods The A549 cells human lung adenocarcinoma cell line were divided into control group and model group [model cells were induced into lung interstitial cells after being treated with transforming growth factor β1(TGF-β1)20 ng/mL for 48 h]. The protein expression of E-cadherin, N-cadherin and vimentin in each group was detected by Western blot. The expression of lncRNA FEZF1-AS1 and EZH2 in the two groups was detected by RT-qPCR. Cells in the transfection group were divided into si NC group,lncRNA FEZF1-AS1+OE vector group and si lncRNA FEZF1-AS1+OE EZH2 group. Cell proliferation was examined by CCK-8 method, cell migration was detected by cell scratch, and cell invasion was detected by Transwell assays. The protein expression of E-cadherin, N-cadherin, vimentin and EZH2 in each group was detected by Western blot. The direct binding effect of FEZF1-AS1 and EZH2 was determined by RNA immuno-precipitation (RIP). Results Compared with the control group, the protein expression level of E-cadherin in the model group was significantly decreased (P＜0.05),and the protein expression of N-cadherin and vimentin was significantly increased(P＜0.05). Compared with the control group, the expression level of lncRNA FEZF1-AS1 and EZH2 genes was significantly increased in the model group (P＜0.05). Compared with si NC group, the proliferation, migration and invasion ability of si lncRNA FEZF1-AS1+OE vector group were decreased, the expression of E-cadherin protein was increased while the expression of N-cadherin, vimentin and EZH2 was decreased(P＜0.05). Compared with si lncRNA FEZF1-AS1+OE vector group, the proliferation, invasion and migration of si lncRNA FEZF1-AS1+OE EZH2 group were increased (P＜0.05). E-cadherin expression was decreased, while N-cadherin, vimentin and EZH2 expressions were increased (P＜0.05). RIP experiment further confirmed that lncRNA FEZF1-AS1 had direct binding effect with EZH2. Conclusions LncRNA FEZF1-AS1 can promote the proliferation, invasion, metastasis and EMT process of pulmonary fibrosis cells by regulating EZH2.

Objective To investigate the distribution of human papillomavirus (HPV) subtypes in women with different age and analyze the relationship between HPV and cervical lesions. Methods A cross-sectional study was conducted on 4 084 patients who underwent colposcopy for pathological examination and HPV genotyping assay in our hospital from January 2015 to June 2020. The patients are classified into 5 groups in respect to the age and 6 groups (the cervicitis, CIN Ⅰ, CIN Ⅱ, CIN Ⅲ, cervical carcinoma in situ and cervical cancer groups) in respect to pathological results respectively. Chi-square test was used to analyze the difference among groups mentioned and to analyze the overall situation of HPV infection, the positive rate of detection of HPV, subtype distribution, infection with single and multiple HPV genotypes among different age groups. Spearman's correlation analysis was used to analyze distribution characteristics of HPV subtypes infection and cervical lesion among different age groups. Results The positive rate of HPV was 93.24% (3808/4 084) different in 5 age groups(Chi-square=11.400 P=0.022). The single subtype infection accounted for 64.67%, and the difference of single subtype infection and multiple subtypes infection was statistically significant among different age groups(Chi-square=70.736, P < 0.001). HPV16(31.88%)、HPV52(25.20%)、HPV58 (14.57%)、HPV18 (11.12%)and HPV51(7.81%)were the top 5 among 23 subtypes of HPV detected. The distribution difference of positive rate of HPV was statistically significant among different lesion groups (P < 0.05). In the cervical cancer group, most patients were infected with HPV16, HPV 18, HPV 52, HPV 58, and HPV 33. Spearman's correlation analysis showed there was a significant correlation between age and cervical lesions (r=0.051, P=0.001). Positive rate of cervical cancer were 0.29% in age 18~29, 1.08% in 30~39, 3.52% in age 40~49, 4.09% in age 50~59 and 10.37% in age 60~82. Conclusion HPV16, HPV 52, HPV 58 and HPV 18 are the most prevalent subtypes among 4 084 patients. HPV16 and HPV18 are relevant with cervical cancer. The risk of cervical cancer is significantly higher for women older than 40. The positive rate of cervical cancer increases with the age.

Parkinson’s disease (PD) is an age-related neurodegenerative disorder which is assessed based on the motor symptoms. A number of microRNAs (miRNAs) are dysregulated and involved in the pathogenesis or development of PD. However, no confirmed markers are used for the early detection of PD. The present study aimed to elucidate the potential two miRNAs (miR-132-3p and miR-146-5p) as novel markers for early PD diagnosis. In the present study, the expression levels of miR-132-3p and miR-146-5p in serum samples from 82 patients with PD and 44 healthy volunteers were measured by reverse transcription-quantitative polymerase chain reaction. Furthermore, the correlation analysis was performed between aberrant miRNAs and Braak staging, Part V of the Unified Parkinson’s Disease Rating Scale (UPDRS-V; the modified Hoehn and Yahr staging of PD) and Part III of the UPDRS-III. Subsequently, the receiver–operating characteristic (ROC) curve results of miR-132-3p and miR-146-5p from healthy volunteers for PD prediction and from severe PD patients were assessed. From the results it was observed that miR-132-3p and miR-146a-5p expressions were significantly decreased in the serum samples of patients with PD compared to those in the healthy volunteers. Moreover, the expressions of miR-132-3p and miR-146a-5p showed a dramatic decrease in severe PD patients as compared to the normal PD patients. Meanwhile, miR-132-3p and miR-146-5p expressions were negatively correlated with Braak staging (r = −0.45, P < 0.0001; r = −0.51, P < 0.0001), UPDRS-III (r = −0.55, P < 0.0001; r = −0.51, P < 0.0001) and UPDRS-V scores (r = − 0.46, P < 0.0001; r = −0.45, P < 0.0001) in PD patients. The area under the curve (AUC) results of miR-132-3p and miR-146a-5p in discriminating PD patients from the healthy controls were 0.7325 (95% CI = 0.6400–0.8251) and 0.7295 (95% CI = 0.3658–0.8232). Moreover, the AUC results of miR-132-3p and miR-146-5p concerning discriminating severe PD patients from normal PD patients were 0.8175 (95% CI = 0.7229–0.9121) and 0.7921 (95% CI = 0.6937–0.8905). In other words, both miR-132-3p and miR-146a-5p may function as promising biomarkers for early diagnosis of PD.

Abstract Background Previous studies have reported that long-term use of valproic acid can cause changes in bone metabolism in children. We conducted this meta-analysis to determine the effects of valproic acid on bone metabolism and bone mineral density (BMD) in children with epilepsy. Methods Studies were searched from the databases of PubMed, Embase, Ovid, Cochrance Library, Springer Link and Web of Science. The effects of valproic acid on bone metabolism indicators and BMD were assessed through calculating the standardized mean difference (SMD) with 95% confidence interval (CI). Results Fourteen studies with 987 individuals were included in this analysis. The long-term use of valproic acid did not affect the levels of serum calcium (p = 0.99), phosphorus (p = 0.28), ALP (p = 0.76), PTH (p = 0.36) and osteocalcin (p = 0.72), but it led to a decrease in 25-OH-VitD (p = 0.01) and BMD (p = 0.002 for the vertebra; p = 0.004 for the femur) in treating children with epilepsy. Conclusion Long-term use of valproic acid in treating children with epilepsy can lead to a reduction in 25-OH-VitD and BMD. Measurements of 25-OH-VitD and BMD should be performed regularly in children taking the drug to detect early osteopenia caused by the drug.

Considering the influence of deformation and displacement of supporting device on modification results,the gear modification is carried out by using KISSsoft analysis software,the gears are modified in generally drum shape and spiral with drum shape respectively,according to Weber slice theory,the stiffness matrix of displacement of tooth contact is established,by using the way of permutation and combination of incremental,the Hertz pressure stress,transmission error,peak load distribution coefficient of tooth meshing performance data of gear under different amount of modification are obtained,considering the deformation and displacement of the supporting device of the gear system accords with the actual working environment,comparing the two kinds of modification of gear meshing performance,contact stress,transfer the error of peak,longitudinal load distribution coefficient than the average crown modification to the tooth surface with drum shaped spiral gears,gear modification in improving the full use of the carrying capacity essential.

A convenient one-pot approach to alkylcyanobiaryls is described. The method is based on biaryl cross-coupling between the sodium salt of the terephthalonitrile dianion and a neutral aromatic nitrile in liquid ammonia, and successive alkylation of the long-lived anionic intermediate with alkyl bromides. The reaction is compatible with benzonitriles that contain methyl, methoxy and phenyl groups, fluorine atoms, and a 1-cyanonaphthalene residue. The variety of ω-substituted alkyl bromides, including an extra bromine atom, a double bond, cyano and ester groups, as well as a 1,3-dioxane fragment are suitable as alkylation reagents.

Taking the double arc gear in the national standard GB/T12759-1991 as the analysis object,by using the theory of fuzzy mathematics and reliability optimization design method,considering arc gear transmission allowable stress randomness and fuzziness,taking the fuzzy reliability of arc gear bending modulus strength and contact strength and the modulus,tooth number,helix angle,longitudinal coincidence degree of gear as constraint condition,a minimum volume of arc gear optimization as target,the fuzzy reliability optimum design model of double arc gear is established,and a practical example is given,the results indicate that,the circular gear design with fuzzy reliability optimization design method has better rationality.

Abstract In this paper, we have demonstrated that europium oxide (Eu2O3) is a new type of active catalyst for single-walled carbon nanotubes (SWNTs) growth under suitable conditions. Both random SWNT networks and horizontally aligned SWNT arrays are efficiently grown on silicon wafers. The density of the SWNT arrays can be altered by the CVD conditions. This result further provides the experimental evidence that the efficient catalyst for SWNT growth is more size dependent than the catalysts themselves. Furthermore, the SWNTs from europium sesquioxides have compatibly higher quality than that from Fe/Mo catalyst. More importantly, over 80% of the nanotubes from Eu2O3 are semiconducting SWNTs (s-SWNTs), indicating the preferential growth of s-SWNTs from Eu2O3. This new finding could open a way for selective growth of s-SWNTs, which can be used as high-current nanoFETs and sensors. Moreover, the successful growth of SWNTs by Eu2O3 catalyst provides new experimental information for understanding the preferential growth of s-SWNTs from Eu2O3, which may be helpful for their controllable synthesis.

Abstract In this letter, we report that dysprosium is an effective catalyst for single-walled carbon nanotubes (SWNTs) growth via a chemical vapor deposition (CVD) process for the first time. Horizontally superlong well-oriented SWNT arrays on SiO2/Si wafer can be fabricated by EtOH-CVD under suitable conditions. The structure and properties are characterized by scanning electron microscopy, transition electron microscopy, Raman spectroscopy and atomic force microscopy. The results show that the SWNTs from dysprosium have better structural uniformity and better conductivity with fewer defects. This rare earth metal provides not only an alternative catalyst for SWNTs growth, but also a possible method to generate high percentage of superlong semiconducting SWNT arrays for various applications of nanoelectronic device.
