The development of effective targeted therapies for triple negative breast cancer (TNBC) remains a challenge. This targeted drug delivery system used a near-infrared fluorescence dye cyanine 5.5 (Cy5.5) and an ICAM-1 antibody on thioether-bridged periodic mesoporous organosilica nanoparticles (PMOs). The ICAM-1 antibody and cyanine 5.5-engineered PMOs (PMO-Cy5.5-ICAM) offer excellent in vivo and in vitro biocompatibility. The PMO-Cy5.5-ICAM shows a loading capacity up to 400 mg/g of doxorubicin (DOX). The drug release profile of the DOX-loaded targeted delivery system (DOX@PMO-Cy5.5-ICAM) is pH-sensitive. Confocal microscopy showed that the PMO-Cy5.5-ICAM efficiently targets and enters TNBC cells. In in vivo experiments, the DOX@PMO-Cy5.5-ICAM accumulates more in TNBCs than in the control groups and exhibits better therapeutic effects on TNBC; thus, it is a promising treatment strategy for TNBC.

The development of early, accurate diagnostic strategies for triple-negative breast cancer (TNBC) remains a significant challenge. Intercellular adhesion molecule-1 (ICAM-1) overexpressed in human TNBC cells is a potential molecular target and biomarker for diagnosis. In this study, small-molecule probe (denoted as γ3-Cy5.5) constructed with a short 17-mer linear peptide (γ3) and near-infrared fluorescence (NIRF) dye cyanine 5.5 (Cy5.5) was used to detect the expression of ICAM-1 in vitro and in vivo, and to diagnose TNBC via NIRF imaging. Western blotting and flow cytometric analysis were used for the detection of ICAM-1 expression in MDA-MB-231 and MCF-7 cells. The cytotoxicity of the small-molecule probe γ3-Cy5.5 was detected using the CCK8 assay. The in vitro targeting of the small-molecule probe γ3-Cy5.5 was verified via flow cytometry and a laser scanning confocal microscope. Finally, the targeting of small-molecule probe in vivo and ex vivo was observed by NIRF imaging. Western blotting and flow cytometry demonstrate that ICAM-1 was highly expressed in the MDA-MB-231 TNBC cell line. Laser confocal microscopy and flow cytometry results show that TNBC cells have an increased cellular uptake of γ3-Cy5.5 compared to the control probe γ3S-Cy5.5. With in vivo NIRF, a significantly higher Cy5.5 signal appeared in the tumors of mice administered γ3-Cy5.5 than those treated with γ3S-Cy5.5. The target-to-background ratio observed on the NIRF images was significantly higher in the γ3-Cy5.5 group (10.2, 13.6) compared with the γ3S-Cy5.5 group (4.4, 4.0) at 1 and 2 h, respectively. This is the first report of the use of ICAM-1-specific small-molecule probe for in vivo NIRF optical imaging of TNBC. This method provides a noninvasive and specific strategy for the early diagnosis of TNBC.

Aim: To investigate the diagnostic efficiency of apparent diffusion coefficient value (ADC) in differentiating benign from malignant breast lesions at 3.0 T diffusion-weighted imaging with different pair of b value. Methods: Total 110 patients with 107 lesions (44 benign and 63 malignant) were selected for our study with five different b-values 0, 400, 800, 1200 and 1600 s/mm2. ADC values were calculated using different pairs of b values. The cut-off ADC values and diagnostic efficiency were evaluated by receiver operating characteristic analysis. Comparison of Mean ADC value for breast lesions was determined by using independent sample t test. ROC curves were used for diagnostic efficiency of ADC using different pairs of b values. Results: With increase of b value, mean ADC value decreases. The mean ADC values for benign were 1.73 × 10-3 mm2/s for b 0 and 400, 1.57 × 10-3 mm2/s for b 0 and 800, 1.43 × 10-3 mm2/s for b 0 and 1200 and 1.30 × 10-3 mm2/s for b 0 and 1600 s/mm2. The mean ADC values for the malignant breast lesion were 1.21 × 10-3 mm2/s for b 0 and 400, 1.06 × 10-3 mm2/s for b 0 and 800, 0.94 × 10-3 mm2/s for b 0 and 1200 and 0.86 × 10-3 mm2/s for b 0 and 1600 s/mm2. ADC diagnostic efficiency for benign and malignant lesion for all the pair of b value combination was significant (p > 0.05). The sensitivity, specificity, PPV, NPV and accuracy were 80.95%, 90.9%, 92.72%, 76.92%, 85.04% for b 0 and 400; 84.12%, 90.9%, 92.98%, 80%, 86.91% for b 0 and 800; 84.12%, 90.9%, 92.98%, 80%, 86.91% for b 0 and 1200; 84.12%, 90.9%, 92.98%, 80%, 86.91% for b 0 and 1600 s/mm2 respectively. Conclusion: DWI is effective in differentiating benign and malignant breast lesion at 3.0 Tesla using ADC with higher b value combination.

To investigate the diagnostic efficiency of decline rate of signal intensity and apparent diffusion coefficient with different b values for differentiating benign and malignant breast lesions on diffusion-weighted 3.0 T magnetic resonance imaging.A total of 152 patients with 162 confirmed histopathologically breast lesions (85 malignant and 77 benign) underwent 3.0 T diffusion-weighted magnetic resonance imaging. Four b values (0, 400, 800 and 1 000 s/mm²) were used. The signal intensity and ADC values of breast lesions were measured respectively. The signal intensity decline rate (SIDR) and apparent diffusion coefficient decline rate (ADCDR) were calculated respectively. SIDR = (signal intensity of lesions with low b value-signal intensity of lesions with high b value)/signal intensity of lesions with low b value, ADCDR = (ADC value of lesions with low b value-ADC value of lesions with high b value) /ADC value of lesions with low b value. The independent sample t-test was employed for statistical analyses and the receiver operating characteristic (ROC) curve for evaluating the diagnosis efficiency of SIDR and ADCDR values.Significant differences were observed in SIDR between benign and malignant breast lesions with b values of 0-400, 400-800 and 800-1 000 s/mm². The sensitivities of SIDR for differentiating benign and malignant breast lesions were 61.2%, 68.2% and 67.1%, the specificities 74.0%, 85.7% and 67.5%, the diagnosis accordance rates 67.3%, 76.5% and 67.3%, the positive predictive values 72.2%, 84.1% and 69.5% and the negative predictive values 63.3%, 71.0% and 65.0% respectively. Significant differences were observed in ADCDR between benign and malignant breast lesions with b values of 400-800 s/mm² and 800-1 000 s/mm². The sensitivities of SDR for differentiating benign and malignant breast lesions were 80.0% and 65.9%, the specificities 72.7% and 65.0%, the diagnostic accordance rates 76.5% and 65.4%, the positive predictive values 76.4% and 67.5% and the negative predictive values 76.7% and 63.3% respectively.The decline rate of signal intensity and apparent diffusion coefficient with different b values may be used for differentiating benign and malignant breast lesions. And the diagnostic efficiency with b values of 400-800 s/mm² is optimal.

OBJECTIVE: To explore the effect of dexamethason e by local treatment on cerebral edema and brain damage after brain injury. METHODS: Twenty-two rabbits were classified into 2 groups, Gro up A (the control group, n=11) and Group B (the treated gr oup, n=11). An rabbit brain contusion model was made by bo ne windowplasty by extradural hitting. Group B was treated by local infiltrating and spraying of dexamethasone at equidistance to lesions. Group A was given nor mal saline in the same way as Group B. The changes of moisture in brain tissues and serum myelin basic protein (MBP) were observed. RESULTS: The percentage of water content in damaged hemisphere in Group A and Group B was 81.75%plus minus0.56% and 79.45%plus minus0.52% respe ctively. There was a significant difference between the 2 groups (P0.05). The normal level of MBP was 1.66 mug/Lplus minus0.71 mug/L, while the value of MBP in Group A and Group B were 5.98 mug/Lplus minus2.08 mug /L and 3.15 mug/Lplus minus1.09 mug/L separately. The level of MBP in Group A an d Group B were higher than normal level and there was also a significant differe nce between Group A and Group B (P0.05). CONCLUSIONS: The results of our study showed that the brain moi sture and MBP in serum were increased after brain injury while reduced after tre atment with dexamethasone. It is demonstrated that local treatment of brain inju ry with dexamethasone has an obvious therapeutic effect on cerebral edema and se rum MBP.

To study migration at county scale and measurement of destination attractivity accurately can reveal the migration flows in more details and explain what kinds of county have higher attractivity ability that will help local governors make better policies to enhance county attractivity and attract more migrants for regional development. The county-scale relative intrinsic attractivity (RIA) of Guangdong Province is computed by using the number of migration and the corresponding distance. The results show that there is an obvious distance decay phenomenon on RIA. The closer to destination, the more migrants move to destination. Moreover, the RIAs have higher correlation relation with migrants to destination. The RIA appears as core-periphery belt pattern. The center of Pearl River Delta is the RIA high core, and the outside areas of the delta are the low RIA belt. The RIA spatial difference is determined by economic factors, living standards and population quality.
