Abstract

BackgroundThe genomic landscape of head and neck cancer has been reported through The Cancer Genome Atlas project. We attempt to determine if high-risk human papillomavirus (HPV) or frequently mutated genes are correlated with survival in an oral cancer cohort.MethodsPatient demographic data along with data from final pathology was collected. Tumor DNA was analyzed using a custom Illumina targeted sequencing panel. Five high-risk HPV types were tested by qPCR. Statistical analyses were used to identify associations between patient outcome and mutational status.ResultsHigh-risk HPV types were identified in 7% of cases; HPV status was not associated with survival. Mutations were identified in TP53, TERT promoter, & PIK3CA. Mutations in TP53 were significantly associated with poorer overall survival on multi-variate analysis (p = 0.03).ConclusionsMutations in TP53 were associated with poor patient survival. Expanding our sample size may identify further predictors of outcome to direct customized cancer care.

Abstract

CRISPR is revolutionizing the ability to do somatic gene editing in mice for the purpose of creating new cancer models. Inactivation of the VHL tumor suppressor gene is the signature initiating event in the most common form of kidney cancer, clear cell renal cell carcinoma (ccRCC). Such tumors are usually driven by the excessive HIF2 activity that arises when the VHL gene product, pVHL, is defective. Given the pressing need for a robust immunocompetent mouse model of human ccRCC, we directly injected adenovirus-associated viruses (AAVs) encoding sgRNAs against VHL and other known/suspected ccRCC tumor suppressor genes into the kidneys of C57BL/6 mice under conditions where Cas9 was under the control of one of two different kidney-specific promoters (Cdh16 or Pax8) to induce kidney tumors. An AAV targeting Vhl, Pbrm1, Keap1, and Tsc1 reproducibly caused macroscopic ccRCCs that partially resembled human ccRCC tumors with respect to transcriptome and cell of origin and responded to a ccRCC standard-of-care agent, axitinib. 39. Borylative cyclisation of alkynes using BCl3

Abstract

Boron trichloride, a cheap and commercially available Lewis acid, has been demonstrated to activate alkynes possessing appropriate nucleophiles, facilitating borylative cyclisation. This reaction furnishes polycyclic compounds possessing a new C(sp2)-B bond externally to the newly formed ring (through concomitant C-C and C-B bond formation). The RBCl2 intermediates generated from cyclisation were esterified with pinacol to furnish air/moisture stable boronic esters. This methodology has been applied to the following classes of starting materials: 1,4-disubstituted but-1-ynes (including N- and O- linked analogues), 2-alkynylanisoles, 2-alkynylthioanisoles and 1,2-bis(alkynyl)benzenes. Thus, borylated scaffolds such as dihydronaphthalenes, dihydroquinolines, 2H-chromenes, benzofurans, benzothiophenes, dibenzopentalenes and benzofulvenes have been synthesised. A variety of functionalities (e.g. amines, esters, nitriles) were tolerated by the reaction, with a number of substrates cyclised on either a gram scale, or under ambient conditions, demonstrating the robust nature of this methodology. An oxidation reaction with [Ph3C][BF4] was carried out on some of the borylated dihydronaphthalene compounds to obtain borylated naphthalenes. Suzuki-Miyaura cross-coupling reactions were carried out on certain borylated cycles to furnish new C-C bonds and generate analogues of established pharmaceuticals such as Nafoxidine or Raloxifene, demonstrating the synthetic value of these borylated cycles. Additionally, a one-pot borylative cyclisation/Suzuki-Miyaura cross-coupling reaction was also developed. Throughout this investigation, alternative reactivity has been observed when using BCl3 to activate certain alkynes, including intermolecular 1,2-trans-carboboration and a rare example of N- and O-directed 1,2-trans-haloboration. Additionally, multiple borylative cyclisations have been carried out on an appropriate alkyne to obtain a B-doped polyaromatic hydrocarbon (PAH), which has potential material-based applications.

Abstract

Genetically engineered T cell therapy can induce remarkable tumor responses in hematologic malignancies. However, it is not known if this type of therapy can be applied effectively to epithelial cancers, which account for 80-90% of human malignancies. We have conducted a first-in-human, phase 1 clinical trial of T cells engineered with a T cell receptor targeting HPV-16 E7 for the treatment of metastatic human papilloma virus-associated epithelial cancers (NCT02858310). The primary endpoint was maximum tolerated dose. Cell dose was not limited by toxicity with a maximum dose of 1 × 10.sup.11 engineered T cells administered. Tumor responses following treatment were evaluated using RECIST (Response Evaluation Criteria in Solid Tumors) guidelines. Robust tumor regression was observed with objective clinical responses in 6 of 12 patients, including 4 of 8 patients with anti-PD-1 refractory disease. Responses included extensive regression of bulky tumors and complete regression of most tumors in some patients. Genomic studies, which included intra-patient tumors with dichotomous treatment responses, revealed resistance mechanisms from defects in critical components of the antigen presentation and interferon response pathways. These findings demonstrate that engineered T cells can mediate regression of common carcinomas, and they reveal immune editing as a constraint on the curative potential of cellular therapy and possibly other immunotherapies in advanced epithelial cancer.
