Abstract

Steroid and hopanoid biomarkers can be found in ancient rocks and, in principle, can give a glimpse of what life was present at that time. Sterols and hopanoids are produced by two related enzymes, though the evolutionary history of this protein family is complicated by losses and horizontal gene transfers, and appears to be widely misinterpretted. Here, I have added sequences from additional key species, and re-analysis of the phylogeny of SHC and OSC indicates a single origin of both enzymes among eukaryotes. This pattern is best explained by vertical inheritance of both enzymes from a bacterial ancestor, followed by widespread loss of SHC, and two subsequent HGT events to ferns and ascomycetes. Thus, the last common ancestor of eukaryotes would have been bifunctional for both sterol and hopanoid production. Later enzymatic innovations allowed diversification of sterols in eukaryotes. Contrary to previous interpretations, the LCA of eukaryotes potentially would have been able to produce hopanoids as a substitute for sterols in anaerobic conditions. Without invoking any other metabolic demand, the LCA of eukaryotes could have been a facultative aerobe, living in unstable conditions with respect to oxygen level.

Abstract

The igneous crust of the oceans and the continents represents the major part of Earth's lithosphere and has recently been recognized as a substantial, yet underexplored, microbial habitat. While prokaryotes have been the focus of most investigations, microeukaryotes have been surprisingly neglected. However, recent work acknowledges eukaryotes, and in particular fungi, as common inhabitants of the deep biosphere, including the deep igneous provinces. The fossil record of the subseafloor igneous crust, and to some extent the continental bedrock, establishes fungi or fungus-like organisms as inhabitants of deep rock since at least the Paleoproterozoic, which challenges the present notion of early fungal evolution. Additionally, deep fungi have been shown to play an important ecological role engaging in symbiosis-like relationships with prokaryotes, decomposing organic matter, and being responsible for mineral weathering and formation, thus mediating mobilization of biogeochemically important elements. In this review, we aim at covering the abundance and diversity of fungi in the various igneous rock provinces on Earth as well as describing the ecological impact of deep fungi. We further discuss what consequences recent findings might have for the understanding of the fungal distribution in extensive anoxic environments and for early fungal evolution.

Abstract

Tooth extraction is a common procedure in dentistry. The normal healing response to the procedure results in a significant loss of bone and collapse of the surrounding gingiva. In addition to normal healing, a substantial percentage of extraction sites suffer postoperative complications. This article presents histology that supports the concept that the first response to extraction is bone death and resorption of the socket wall. The stages of extraction socket healing also will be discussed. Additionally, the article will present a regenerative method that skips the resorptive phase, the clotting phase, the granulation of tissue phase, and the collagen-producing phase of normal extraction-socket healing, while avoiding extraction-socket complications.

Abstract

Risdiplam (EVRYSDI®) is an oral survival of motor neuron 2 (SMN2) pre-mRNA splicing modifier approved by the EMA and MHRA for the treatment of patients aged ≥2 months, with a clinical diagnosis of Type 1, 2 or 3 spinal muscular atrophy (SMA) or 1–4 SMN2 copies.JEWELFISH (NCT03032172) is an open-label study of risdiplam in patients with SMA who previously received RG7800 (RO6885247), nusinersen (SPINRAZA®), olesoxime or onasemnogene abeparvovec (ZOLGENSMA®).JEWELFISH assesses the safety, tolerability and pharmacokinetic/pharmacodynamic (PD) relationship of risdiplam in a broad range of ages (1–60 years), SMA types (1–3), SMN2 copy numbers (1–4) and motor function (nonsitters, sitters and walkers). We previously presented safety data from 173 patients (data cut-off: 31 July 2020): 13 previously received RG7800, 76 received nusinersen, 70 received olesoxime and 14 received onasemnogene abeparvovec. Risdiplam led to a rapid and sustained >2-fold increase in SMN protein (data cut-off: 1 June 2020), consistent with data from treatment-nai¨ve patients with Types 2/3 SMA (SUNFISH; NCT02908685).No drug-related safety findings leading to withdrawal were reported for any patients in JEWELFISH. The safety profile of risdiplam was consistent with observations in treatment-nai¨ve patients. Here we present 12-month safety, PD and exploratory efficacy data.

Abstract

The problem of deriving an analytic expression which would relate low flying aircraft trajectories to terrain cross-section has not been solved. A computer simulation has been devised which generates synthetic flight paths which are coupled to a terrain input and are dependent on the values of five input parameters. Simulations have been generated which best match a series of actual test flights. The input parameters which generated the best fit to actual flights are examined to determine their statistical properties. Tests of hypotheses are conducted to determine goodness of fit and equality of means and variances among parameter sets related to specific terrains and speeds. A sampling procedure is suggested whereby a random series of flights might be generated over an arbitrary terrain cross-section subject to speed restrictions. (Author)
