An ergonomic upright body posture is maintained by the alignment of the spine, pelvis, and lower extremities, and the muscle strength of body trunk and lower extremities. The posture varies with age because of the degenerative changes in the involved structures and the weakening of the muscles. The compensatory mechanisms underlying these changes have recently been evaluated, and the loss of lumbar lordosis results in spinal kyphosis, pelvic retroversion, hip extension, knee flexion, and ankle dorsiflexion. These mechanisms are referred to as the hip–spine and knee–spine syndromes. The spine, hip, and knee are anatomically connected, and the pain and discomfort of the lower back, hip, and knee frequently arise due to degenerative changes of these structures. Thus, these mechanisms are considered as the knee–hip–spine syndrome. Spinal fusion, total hip arthroplasty, and total knee arthroplasty are the surgical procedures for severe degeneration, and their clinical outcomes for the affected sites are promising. However, despite surgeries, other structures may degenerate and result in complications, such as proximal junctional kyphosis and hip dislocation, following spinal fusion. Therefore, it is necessary to evaluate each patient under specific conditions and to treat each section while considering associations between the target structure and entire body. The purpose of this article is to introduce postural maintenance, variations with age, and improvements with surgical interventions of spine, hip, and knee as the knee–hip–spine syndrome. The common 'three-pistil' (TP) wheat mutation line expresses TPs in a floret normally containing TPs forming three grains set close back-to-back. The developmental origin of the TP trait in common wheat had been diagnosed non-destructively using the blue aleurone trait. The aleurone colour of F2 seeds grown in F1 plants of cross TP/UC66049 was evaluated. Due to xenia, the hue of blue grain colour depended on dose of the Ba1 gene for blue aleurone in the triploid endosperm. The TP trait produced four types of segregation in three-seed clusters: (i) white grain only, (ii) two white grains and one blue, (iii) one white grain and two blue, and (iv) three blue grains only. The observed frequency of blue-white seed within clusters followed the binominal distribution 3Cr (0.75)r∙(0.25)3-r, where r is the number of colour variants in three-seed clusters (r = 0-3). Intrafloret segregation of seed colour and F2 segregation derived from aleurone colour of F3 seeds indicated an independent origin of the TP trait. [ABSTRACT FROM AUTHOR]

Abstract: Gherkin (Cucumis anguria L.), also known as West Indian gherkin, burr gherkin, and maxixe, is mainly cultivated and consumed in Brazil and the United States. Marker-assisted selection (MAS) would be a highly desirable tool for breeding of gherkin as gherkin cultivation generally requires considerable time, labor, and space. However, DNA markers available for gherkin have rarely been reported, although many microsatellite linkage maps have been constructed for cucumber and whole genomic sequences have been reported. In this study, we attempted to transfer 86 microsatellite markers derived from cucumber to gherkin and assess amplification and polymorphism between two gherkin accessions to enable the use of the microsatellite markers in gherkin. In PI 320052 and PI 364475, 58 (67.4%) and 55 (64.0%) microsatellite markers were successfully amplified, respectively. Of the amplified markers, the polymorphism number between the two accessions was 40. Thus, an adequate number of cucumber microsatellite markers can be transferred to gherkin. Current polymorphic microsatellite markers could then be made available for the study of landmarks in linkages, genomic structures, evolutionary ecology, and MAS in gherkin and Cucurbitaceae. Abstract: To analyze the mechanisms of mRNA export from the nucleus to the cytoplasm, we have isolated eleven mutants, ptr [poly(A)+ RNA transport] 1 to 11, which accumulate poly(A)+ RNA in the nucleus at a nonpermissive temperature in Schizosaccharomyces pombe. Of those, the ptr5–1 mutant shows dots- or a ring-like accumulation of poly(A)+ RNA at the nuclear periphery after shifting to the nonpermissive temperature. We cloned the ptr5 + gene and found that it encodes a component of the nuclear pore complex (NPC), nucleoporin 85 (Nup85). The ptr5–1 mutant shows no defects in protein transport, suggesting the specific involvement of Ptr5p/Nup85p in nuclear mRNA export in S. pombe. We identified Seh1p, a nucleoporin interacting with Nup85p, an mRNA-binding protein Mlo3p, and Sac3p, a component of the TREX-2 complex involved in coupling of nuclear mRNA export with transcription, as multi-copy suppressors for the ptr5–1 mutation. In addition, we found that the ptr5–1 mutation is synthetically lethal with a mutation of the mRNA export factor Rae1p, and that the double mutant exaggerates defective nuclear mRNA export, suggesting that Ptr5p/Nup85p is involved in nuclear mRNA export through Rae1p. Interestingly, the ptr5–1 mutation also showed synthetic effects with several prp pre-mRNA splicing mutations, suggesting a functional linkage between the NPCs and the splicing apparatus in the yeast nucleus. Purpose: Balancing the gap is essential in total knee arthroplasty (TKA). The purpose of this study was to quantify the influence of patellar position on femoro-tibial joint load in TKA. We hypothesized that resetting of the patella increased medial joint load and decreased lateral joint load. Methods: Our original tensor system was used during posterior-stabilized (PS) and cruciate-retaining (CR) total knee arthroplasty (TKA) using medial para-patellar approach (MPP) or sub-vastus approach (SV). Results: In PS-TKA, by resetting the patella, the ratios between medial and lateral compartments were not changed in both extension and flexion position using MPP and were significantly changed in flexion position using SV. In CR-TKA, by resetting the patella, the load of the lateral component decreased and the ratios between medial and lateral compartments were changed significantly in both extension and flexion position using SV. Conclusion: It is important to be aware that, when performing CR-KA, the load of the lateral compartment will decrease by resetting the patella using SV. The article presents a case study of a 14-year-old boy who felt severe pain and noticed deformity in both knees after landing from a jump over a fence. Physical examination showed flexion deformity of the knees and inability to lift the knees, and he was diagnosed with type 1A osteogenesis imperfecta. The article discusses simultaneous bilateral Salter-Harris type II injuries followed by unilateral Salter-Harris type III injury of the proximal tibia in a patient with osteogenesis imperfecta.

An original tensor system was developed to directly measure the load between femoral trial component and tibial cut surface in vivo in both patella everted and reset positions during total knee arthroplasty (TKA). We used this system during posterior-stabilized (PS) and cruciate-retaining (CR) TKA. In PS-TKA, there was no significant difference between the loads in extension in patella everted position and reset position. In flexion, however, there was significant increase of load in patella reset position compared to in everted position. In CR-TKA, there was no significant difference between the loads in patella everted position and in patella reset position in either extension or flexion. It was found that the effect of patella position on joint gap load was different between PS-TKA and CR-TKA. It is important to be aware that, when performing PS-KA, the load in flexion gap will increase, in other words, flexion gap distance will decrease by resetting the patella. [ABSTRACT FROM AUTHOR]

Delayed gadolinium-enhanced MRI of cartilage (dGEMRIC) was used for the measurement of relative proteoglycan depletion of articular cartilage in the patellofemoral (PF) joint following a proprietary protocol, which was compared with the X-ray images, proton density weighted MR images (PDWI) and arthroscopic findings. The study examined 30 knees. The ages ranged from 16 to 74 (average 40.3) years. The Gd-DTPA2–containing contrast medium was used in a single dose. The subjects were made to exercise the knee joint for 10 min; and MR images were taken 2 h after intravenous injection of contrast medium. T1-calculated images were produced and the region of interest (ROI) was set as follows. (1) ROI1: entire articular cartilage in a slice through the center of the patella. (2) ROI2: low signal region in T1-calculated images, which were set in a blind fashion by two observers. (3) ROI3: articular cartilage on one side that includes ROI2 where low signal region were detected (medial or lateral). ROI3 was set to examine the contrast of ROI2 with surrounding articular cartilage. The average T1 values of ROI1 was 393.5±33.6 ms for radiographic grade 0 and 361.3±11.1 ms for grade I, which showed a significant difference ( P=0.036). The T1 value of ROI2 was 351.6±28.2 ms for grade I, 361.9±38.3 ms for grade II, 362.1±67.7 ms for grade III, and 297.8±54.1 ms for grade IV according to arthroscopic Outerbridge classification. All cases, that demonstrated decrease of T1 values on dGEMRIC (ROI2), showed abnormal arthroscopic or direct viewing findings. The ratio (ROI3/ROI2) in cases of only slight damage classified as Outerbridge grade I (6 cases) was an average of 1.04±0.02 and was 1.0 or greater in all cases, thereby indicating well-defined contrast with the surrounding cartilage. The diagnosis of damage in articular cartilage was possible in all 16 cases with radiographic K–L grade I on dGEMRIC, while the intensity changes were not found in 10 of 16 cases on PDWI. The dGEMRIC with a single-dose would be useful on a diagnosis of the area demonstrating early relative proteoglycan depletion in the articular cartilage of the PF joint prior to any discernible changes in the subchondral bone on X-ray images and exceeds to plain MR images for examining deterioration of articular cartilage. Abstract: A 52 year-old woman suffered bilateral femoral shaft fractures when she was 5 years old and they were treated surgically. She had been able to walk without pain and giving way throughout her young and adult life, however, when she came to us with a 1-year history of the right knee pain, she could walk only 10 m continuously. The right knee revealed valgus deformity with complete lateral patellar dislocation. The femoro–tibial angle was 154° and arthritic change was seen in the femorotibial joint. Surfaces of both the patella and the femur were degraded. Twenty-two degrees of rotational deformity was also found internally in the involved side. Varus osteotomy with external rotation of the distal fragment and lateral retinacular release combined with reefing of vastus medialis muscle were performed. Twenty-four months after surgery, knee score and functional score were, respectively, 85/100 and 71/100, using knee society clinical rating system and there was no pain associating patella incongruity. Background: The behavior of host and graft cells during the healing process after autologous tendon graft has not been elucidated. Hypothesis: Host cells will integrate into the bone-tendon interface and contribute to cellular repopulation of the graft. Study Design: Controlled laboratory study. Methods: Twelve-week-old, genetically identical, female green fluorescent protein transgenic rats (n = 20) and wild-type rats (n = 20) were used. The rats were divided into 2 experimental groups. In group A, the Achilles tendons of wild-type rats were harvested and transplanted into the transcondylar femoral bone tunnels of green fluorescent protein rats. In group B, the Achilles tendons of green fluorescent protein rats were transplanted into a transcondylar femoral bone tunnel of wild-type rats. Immediately after transplantation (time zero) and at 1, 2, and 4 weeks after the transplantation, distal femoral epiphyses were harvested and cut into 14-μm serial sagittal frozen sections. The sections were examined with a confocal laser-scanning micro-scope to quantify green fluorescent protein-positive cell survival. Results: At time zero, only host cells in group A and only graft cells in group B demonstrated green fluorescent protein signals. At 1 week in group A, many green fluorescent protein-positive cells were found in the graft. In group B, a few green fluorescent protein-positive cells were found in the graft. At 2 and 4 weeks in group A, many green fluorescent protein-positive cells were detected in the graft, but green fluorescent protein-positive cells had disappeared completely in group B. Conclusion: Host cells, rather than graft cells, contribute to repair of the bone-tendon interface and the remodeling of grafts after simulated autologous tendon graft. [ABSTRACT FROM AUTHOR]

Italian and Tunisian durum wheats from different eras of breeding were assessed for the presence of a gene for brittle rachis. Nine of 15 Italian durum landraces had brittle rachides. Strampelli’s achievement was the release of the well-known variety, Senatore Cappelli, which was derived from a Tunisian landrace, Jenah Rhettifah, which has a brittle rachis. Rachides of two Tunisian landraces were also brittle. Since the 1950’s, 16 accessions were released as selections from crosses or mutagenesis involving Senatore Cappelli. Seven of these accessions have brittle rachides. F2 segregation in intercrosses of Senatore Cappelli with other durum accessions with brittle rachides indicated a common allele for brittle rachis. Segregation of the F2 from Senatore Cappelli/ANW 10A and the F2 of Senatore Cappelli/ANW 10B showed that the gene for brittle rachis of Senatore Cappelli was allelic to the brittle allele of the Br-B1 locus on chromosome 3B. Senatore Cappelli was presumably the only source of brittle rachis used in Italian breeding programmes. The genes for brittle rachis have been retained in the gene pool of durum wheat, suggesting that the brittle rachis character is not associated with an appreciable yield loss in modern farming systems in Mediterranean environments. Growth factors have been shown to play a critical role in enhancing tissue healing. In this study, we evaluate the effect of growth factors on cell proliferation and matrix synthesis. Fibroblasts were derived from medial collateral ligaments (MCLs) of goats and grown in culture. Basic fibroblast growth factor (bFGF), platelet-derived growth factor (PDGF-BB), epidermal growth factor (EGF), and transforming growth factor (TGF-β[sub 1]) were used for the treatment of cells. It was found that bFGF, PDGF-BB and EGF significantly increased proliferation of the goat MCL fibroblasts. In addition, among these three growth factors, bFGF, at the concentration of 1.0 ng/ml, had the maximal effect on cell proliferation. Furthermore, both TGF-β[sub 1] and EGF significantly increased collagen and noncollagenous protein syntheses of goat MCL fibroblasts. The results of this study may be used as baseline data for the application of growth factors to enhance MCL healing in a goat model. [ABSTRACT FROM AUTHOR]

Highlights from the article: The hip-spine syndrome was first described by C. M. Offierski, wherein the degenerative change of the hip and the lumbar spine are interrelated [3]. Thus, the relationship among the spine, the pelvis, and the hip joint for the treatment of the hip-spine syndrome needs further investigations. Similarly, the knee-spine syndrome demonstrates that the symptoms from the lumbar spine may be caused by osteoarthritis (OA) of the knee and that the limitation of knee extension in turn affects the loss of lumbar lordosis [4].

The boutonniere deformity is a well-known deformity in the fingers, however, its appearance on a lessor toe is extremely rare. In the present case, the deformity resulted from a rupture of the central slip of the extensor tendon and the shift of the lateral bands to the planter side after reduction of a traumatic planter dislocation of the PIP joint of the second toe. We describe a case of popliteofibular ligament (PFL) injury, successfully treated with a new anatomic reconstruction technique looping the biceps femoris tendon (BT). The anterior half of the BT was split longitudinally from the fibular insertion, cut at the proximal end and left attached at the insertion. The proximal end of the BT was looped back in a slit made in the popliteal tendon (PT) at the original anatomical insertion site of the PFL, and passed through the tunnel from the posterior and the baseball suture was tightened on the anterior cortex of fibular head. Background: Leaves of Poaceae have a unique morphological feature: they consist of a proximal sheath and a distal blade separated by a ligular region. The sheath provides structural support and protects young developing leaves, whereas the main function of the blade is photosynthesis. The auricles allow the blade to tilt back for optimal photosynthesis and determine the angle of a leaf, whereas the ligule protects the stem from the entry of water, microorganisms, and pests. Liguleless variants have an upright leaf blade that wraps around the culm. Research on liguleless mutants of maize and other cereals has led to identification of genes that are involved in leaf patterning and differentiation. Results: We characterized an induced liguleless mutant (LM) of Aegilops tauschii Coss., a donor of genome D of bread wheat Triticum aestivum L.. The liguleless phenotype of LM is under dominant monogenic control (Lgt). To determine precise position of Lgt on the Ae. tauschii genetic map, highly saturated genetic maps were constructed containing 887 single-nucleotide polymorphism (SNP) markers derived via diversity arrays technology (DArT)seq. The Lgt gene was mapped to chromosome 5DS. Taking into account coordinates of the SNP markers, flanking Lgt, on the pseudomolecule 5D, a chromosomal region that contains this gene was determined, and a list of candidate genes was identified. Morphological features of the LM phenotype suggest that Lgt participates in the control of leaf development, mainly, in leaf proximal–distal patterning, and its dominant mutation causes abnormal ligular region but does not affect reproductive development. Conclusions: Here we report characterization of a liguleless Ae. tauschii mutant, whose phenotype is under control of a dominant mutation of Lgt. The dominant mode of inheritance of the liguleless trait in a Triticeae species is reported for the first time. The position of the Lgt locus on chromosome 5DS allowed us to identify a list of candidate genes. This list does not contain Ae. tauschii orthologs of any well-characterized cereal genes whose mutations cause liguleless phenotypes. Key message: We propose the MADS-box transcription factor SVP-A1 as a promising candidate gene for the elongated glume locus P2, which maps to chromosome 6A instead of the previously proposed chromosome 7B. In rice and wheat, glume and floral organ length are positively correlated with grain size, making them an important target to increase grain size and potentially yield. The wheat subspecies Triticum ispahanicum is known to develop elongated glumes and floral organs as well as long grains. These multiple phenotypic effects are controlled by the P2 locus, which was previously mapped to wheat chromosome 7B. Using three mapping populations, we show that the long glume locus P2 does not map to chromosome 7B, but instead maps to a 1.68 Mbp interval on chromosome 6A. Within this interval, we identified SVP-A1, a MADS box transcription factor which is the direct ortholog of the maize gene underlying the 'pod corn' Tunicate locus and is a paralog to the T. polonicum elongated glume P1 gene. In T. ispahanicum, we identified a unique allele which has a 482-bp deletion in the SVP-A1 promoter and is associated with ectopic and higher expression of SVP-A1 in the elongated glumes and floral organs. We used near-isogenic lines (NILs) to show that P2 has a consistent positive effect on the length of glume, lemma, palea, spike and grain. Based on the mapping data, natural variation, biological function of SVP genes in cereals and expression analyses, we propose the MADS-box transcription factor SVP-A1 as a promising candidate for P2. Abstract: Wild Cucumis spp., which have resistance to various diseases, have the potential to be used for melon (C. melo L.) breeding. Nevertheless, few interspecific hybrids between melon and wild Cucumis spp. have been obtained because of strong reproductive barriers such as pollen–pistil incongruity and hybrid seed abortion. For this study, we examined the temperature conditions enhancing pollen tube growth and fruit set in interspecific crosses between melon and wild Cucumis spp. Before temperature-conditioned pollination, four wild Cucumis spp. were pollinated with the melon line ‘MR-1’ and two lines of C. anguria, PI 147065 and PI 320052, and a line of C. metuliferus, PI 526242, were selected for later experiments. These three lines were crossed reciprocally with ‘MR-1’ under controlled temperatures of 24–34°C at 2°C intervals. When ‘MR-1’ pistils were pollinated with three lines of wild Cucumis spp., pollen tube growth was arrested after germination at all temperatures. However, in the pistil of wild Cucumis spp., pollen tube growth of ‘MR-1’ was significantly greater at 32°C and at 34°C than that under 30°C. Furthermore, pollen tube length in pistils of PI 320052 and PI 526242 at 32°C was comparable to that of ‘MR-1’ self-pollination at 28°C. Fruit set frequency was also improved at 32°C. These results demonstrated that the pollen tube inhibition of Cucumis spp. interspecific crosses might be partially or fully recoverable in higher-temperature conditions. Moreover, 32°C is apparently the threshold temperature for promoting pollen tube growth in these cross combinations. [Copyright &y& Elsevier]

The healing of an injured musculoskeletal system requires an influx of mesenchymal cells that can differentiate into osteoblasts, fibroblasts, chondroblasts, and skeletal myoblasts. However, whether these mesenchymal cells arise from the circulation (bone marrow) or the injured tissues themselves has been controversial. To reveal the spatiotemporal characteristics of the reparative mesenchymal cells, we investigated the healing process after patellar tendon injury using two types of green fluorescent protein (GFP) chimeric rats; one expressing GFP in the circulating cells, and the other expressing it in the patellar tendon. We analyzed the behavior of GFP-positive cells after experimental tendon injury in both chimeric rats to clarify the origin of reparative cells. At 24 h after the injury, the wound contained circulation-derived cells but not tendon-derived cells. Tendon-derived cells first appeared in the wounded area at 3 days after the injury, and had significantly increased in number with time and had maintained a high level of proliferative activity until 7 days after the injury, whereas the circulation-derived cells had decreased in number and had been replaced by the tendon-derived cells. These findings suggest that circulation-derived and tendon-derived cells contribute to the healing of tendons in different periods as part of a biphasic process. J. Cell. Abstract: The photocatalytic reaction in the partial oxidation of α-methylstyrene (α-MS) to acetophenone (AP) was investigated for TiO2 supported on Y-type, moredenite, and ZSM-5 zeolites. XRD and FT-Raman results demonstrated that the anatase is the stable phase of TiO2 supported on zeolites and no rutile phase was observed in the supported catalysts calcined at 723–973K. TiO2 supported on zeolites was found to catalyze the partial oxidation of α-MS to AP under light irradiation (>290nm). The photocatalytic activity of TiO2 supported on zeolites was significantly improved by the addition of the water to the organic solvent. It was found that the optimized amount of water added for TiO2 supported on zeolite catalysts was smaller than that for TiO2 catalyst. [Copyright &y& Elsevier]

Background: Plant height is an important wheat trait that is regulated by multiple genes, among which Rht is of the utmost value. In wheat, Rht-B1p (=Rht17) is a mutant allele of the Rht gene that encodes for a DELLA-protein and results in the development of gibberellin-insensitive plants with a dwarfing phenotype. The pleiotropic effects of dwarfing genes on yield are highly dependent on both the genetic background and the environmental conditions. In Russia, the Central Non-Black Earth Region and Krasnodar Krai are two economically important regions that require differing management for sustainable wheat production for food, feed and industry. The purpose of our study was to compare the pleiotropic effects of Rht-B1p on the main valuable agronomic traits in the F3:4 families of the spring bread wheat Chris Mutant/Novosibirskaya 67 in the genetic background of Vrn-B1a/vrn-B1 (spring/winter phenotype) and Ppd-D1a/Ppd-D1b (insensitivity/sensitivity to photoperiod) alleles in a field experiment in Moscow and Krasnodar Krai. Results: Plant height was reduced on average by 21 cm (28%) and 25 cm (30%), respectively; Ppd-D1a slightly strengthened the dwarfing effect in Moscow and mitigated it in Krasnodar Krai. Grain weight of the main spike was reduced by Rht-B1p in Moscow and to lesser extent in Krasnodar; Ppd-D1a and Vrn-B1a tended to partially compensate for this loss in Krasnodar Krai. Thousand grain weight was reduced on average by 5.3 g (16%) and 2.9 g (10%) in Moscow and Krasnodar Krai, respectively, but was partially compensated for by Ppd-D1a in Krasnodar Krai. Harvest index was increased due to Rht-B1p by 6 and 10% in Moscow and Krasnodar Krai, respectively. Rht-B1p resulted in a delay of heading by 1–2 days in Moscow. Ppd-D1a accelerated heading by 1 day and 6 days in Moscow and in Krasnodar Krai, respectively. Conclusions: Rht-B1p could be introduced into wheat breeding along with dwarfing genes such as Rht-B1b and Rht-D1b. Special attention should be paid to its combination with Ppd-D1a and Vrn-B1a as regulators of developmental rates, compensators of adverse effects of Rht-B1p on productivity and enhancers of positive effect of Rht-B1p on harvest index. On the basis of rapid population aging, in 2007, the Japanese Orthopaedic Association (JOA) proposed a new disease concept "locomotive syndrome" as a degenerative condition of reduced mobility due to the impairment of the musculoskeletal system. Worsened locomotive components, which consist of bones, joints, and intervertebral discs, and muscles and nerves, can lead to symptoms such as pain, limited range of motion, malalignment, impaired balance, and difficulty in walking, ultimately resulting in the requirement of nursing care. "Locomotive syndrome" has gained increased interest in Japan but still not worldwide. Hence, in this brief review, we summarize an updated definition, assessment, and management of "locomotive syndrome". The JOA recommends "locomotion training" exercise intervention to be effective in maintaining motor function that comprises two simple exercises—squatting and single-leg standing. However, the extent to which exercises affect "locomotive syndrome" is unknown. Here, we further report hypothesis-generating patient cases who presented the improved sagittal spinopelvic alignment in standing radiographs and postural stability in piezoelectric force-plate measurements through our 6-month "locomotion training" outpatient rehabilitation program. It is noteworthy that "locomotion training" facilitated these improvements despite the presence of specific disorders including thoracic kyphosis and symptomatic lumbar spinal canal stenosis. This raises the need for further investigations to clarify effects of "locomotion training" exercises on the spinal alignment, global balance, and quality of life in patients with "locomotive syndrome". Background: Inflorescences of wheat species, spikes, are characteristically unbranched and bear one sessile spikelet at a spike rachis node. Development of supernumerary spikelets (SSs) at rachis nodes or on the extended rachillas is abnormal. Various wheat morphotypes with altered spike morphology, associated with the development of SSs, present an important genetic resource for studies on genetic regulation of wheat inflorescence development. Results: Here we characterized diploid and tetraploid wheat lines of various non-standard spike morphotypes, which allowed for identification of a new mutant allele of the WHEAT FRIZZY PANICLE (WFZP) gene that determines spike branching in diploid wheat Ttiticum monococcum L. Moreover, we found that the development of SSs and spike branching in wheat T. durum Desf. was a result of a wfzp-A/TtBH-A1 mutation that originated from spontaneous hybridization with T. turgidum convar. compositum (L.f.) Filat. Detailed characterization of the false-true ramification phenotype controlled by the recessive sham ramification 2 (shr2) gene in tetraploid wheat T. turgidum L. allowed us to suggest putative functions of the SHR2 gene that may be involved in the regulation of spikelet meristem fate and in specification of floret meristems. The results of a gene interaction test suggested that genes WFZP and SHR2 function independently in different processes during spikelet development, whereas another spike ramification gene(s) interact(s) with SHR2 and share(s) common functions. Conclusions: SS mutants represent an important genetic tool for research on the development of the wheat spikelet and for identification of genes that control meristem activities. Further studies on different non-standard SS morphotypes and wheat lines with altered spike morphology will allow researchers to identify new genes that control meristem identity and determinacy, to elucidate the interaction between the genes, and to understand how these genes, acting in concert, regulate the development of the wheat spike. Key message: Different blue-aleurone wheats display major differences in chromosome composition, ranging from disomic chromosome additions, substitutions, single chromosome arm introgressions and chromosome translocation of Thinopyrum ponticum. Abstract: Anthocyanins are of great importance for human health due to their antioxidant, anti-inflammatory, anti-microbial and anti-cancerogenic potential. In common wheat ( Triticum aestivum L.) their content is low. However, elite lines with blue aleurone exhibit significantly increased levels of anthocyanins. These lines carry introgressed chromatin from wild relatives of wheat such as Thinopyrum ponticum and Triticum monococcum. The aim of our study was to characterize genomic constitutions of wheat lines with blue aleurone using genomic and fluorescence in situ hybridization. We used total genomic DNA of Th. ponticum and two repetitive DNA sequences (GAA repeat and the Afa family) as probes to identify individual chromosomes. This enabled precise localization of introgressed Th. ponticum chromatin. Our results revealed large variation in chromosome constitutions of the blue-aleurone wheats. Of 26 analyzed lines, 17 carried an introgression from Th. ponticum; the remaining nine lines presumably carry T. monococcum chromatin undetectable by the methods employed. Of the Th. ponticum introgressions, six different types were present, ranging from a ditelosomic addition (cv. Blue Norco) to a disomic substitution (cv. Blue Baart), substitution of complete (homologous) chromosome arms (line UC66049) and various translocations of distal parts of a chromosome arm(s). Although Japanese morning glory (Ipomoea nil (L.) Roth.) has been used intensively for genetic studies, DNA markers have not been developed in Ipomoea nil sufficient to cover all chromosomes. Therefore, we conducted microsatellite (simple sequence repeats, SSR) marker development in I. nil for future genetic studies. From 92,662 expressed sequence tag (EST) sequences, 514 unique microsatellite-containing ESTs were identified. Primer pairs were designed automatically in 326 SSRs. Of 150 SSRs examined, 75 showed polymorphisms among strains. A phenogram based on the SSR genotypes revealed the genetic relation among seven Japanese morning glories from five different regions of the world and an ivyleaf morning glory (I. hederacea Jacq.). A 24-year-old male presented with an open fracture of the left patella (Gustilo–Anderson grade III-A) and fracture of the left femoral lateral condyle, and subsequent patella alta suggesting rupture of the left patellar tendon. This initial injury was treated 6 months earlier in another hospital. He was referred because of limited knee motion and radiographs showed severe patella infera. By means of a combination of a contralateral BTB graft and soft tissue repair named anterior capsular shift, the knee contracture with patella infera was resolved successfully. This study evaluates the potential of flow cytometry for chromosome sorting in durum wheat (Triticum turgidum Desf. var. durum, 2n = 4x = 28). Histograms of fluorescence intensity (flow karyotypes) obtained after the analysis of DAPI-stained chromosomes consisted of three peaks. Of these, one represented chromosome 3B, a small peak corresponded to chromosomes 1A and 6A, and a large peak represented the remaining 11 chromosomes. Chromosomes sorted onto microscope slides were identified after fluorescence in situ hybridization (FISH) with probes for GAA microsatellite, pSc1 19.2, and Afa repeats. Genomic distribution of these sequences was determined for the first time in durum wheat and a molecular karyotype has been developed for this crop. Flow karyotyping in double-ditelosomic lines of durum wheat revealed that the lines facilitated sorting of any arm of the wheat A- and B-genome chromosomes. Compared to hexaploid wheat, flow karyotype of durum wheat is less complex. This property results in better discrimination of telosomes and high purities in sorted fractions, ranging from 90 to 98%. We have demonstrated that large insert libraries can be created from DNA purified using flow cytometry. This study considerably expands the potential of flow cytogenetics for use in wheat genomics and opens the possibility of sequencing the genome of this important crop one chromosome arm at a time. • A disturbance in photosynthetic electron flow affects the chlorophyll-depleted wheat mutants. • Unexpectedly long-term acclimation to light fluctuations can be allowed in mutants. • Successful development of a fluctuating-light phenotype depends on the mutated locus. • Compensation of the mutation involves use of alternative electron sinks and thylakoid structural re-organization. The search for more productive crops is exploring accessions with reduced chlorophyll accumulation in leaves, which would promote plant growth mainly because of a higher light transmittance throughout the canopy. Under continuous light, chlorophyll-depleted chlorina mutants of wheat can reach yields similar to WT; however, their performance under fluctuating light could possibly be lowered by a disturbed photosynthetic electron transport. Six chlorina mutants of Triticum aestivum (ANBW4A, ANBW4B, ANK32A) or T. durum (ANDW7A, ANDW7B, ANDW8A) were compared to WT genotypes under continuous and fluctuating light regimes, the latter obtained through a randomized pattern of light intensity changes. After two weeks of plant acclimation under either regimes, light energy management was thoroughly analysed for four weeks. All chlorina mutants showed a defective ability to regulate the electron poise during a fast rise in irradiance. Different extent of deregulation depended on mutation and genomic background, durum wheat being more severely impaired than bread wheat. However, a great acclimative capacity to a fluctuating light regime unexpectedly still characterized the chlorina wheat plants, with the only exception of the mutants carrying the cn-B1b mutated locus (ANDW7B and ANBW4B). Under fluctuating light, all other chlorina mutants developed indeed an ability to improve their control of electron transport against the recurrent reducing bursts caused by lightflecks. Compensatory responses included the regulation of energy distribution between photosystems, the use of alternative electron sinks, and a structural reorganization of the thylakoid system. However, in no case a reduced chlorophyll content led to more productive plants. Among 23 measured photosynthetic parameters, especially the non-regulatory energy dissipation in photosystem II - Y(NO) –, when probed during a light rise, can be proposed in automated phenotyping experiments as a straightforward index to be used for chlorina wheat screening with respect to potential productivity.
