We then established a mouse model with a notable gene alteration by CRISPR/Cas9 to confirm the biological role.Results: The long-term recurrence-free survivors more frequently had germline heterozygous variant rs2185379 of the PRDM1 gene exon than patients with early recurrence (6.8-fold, P=0.013) and the general population. In the mouse model, primary intraperitoneal disseminated tumors of allograft ID8 were significantly smaller in the germline heterozygous rs2185379 group than in the wild-type group (57.4% decrease, P=0.008). Immunohistochemistry showed that the area of distribution of infiltrating T lymphocytes with positive CD8 staining was significantly increased in the germline heterozygous rs2185379 group in comparison to the wild-type group.Conclusion: Germline heterozygous rs2185379 in PRDM1 is correlated with an excellent prognosis and can be used to establish a new strategy for treating advanced ovarian cancer.Keywords: ovarian cancer, long-term survivors, rs2185379, PRDM1

Abstract

Background: Olaparib, a poly (ADP-ribose) polymerase (PARP) inhibitor, has previously been shown to extend progression-free survival versus placebo when given to patients with relapsed high-grade serous or endometrioid ovarian cancer who were platinum sensitive and who had a BRCA1 or BRCA2 (BRCA1/2) mutation, as part of the SOLO2/ENGOT-Ov21 trial. The aim of this final analysis is to investigate the effect of olaparib on overall survival. Methods: This double-blind, randomised, placebo-controlled, phase 3 trial was done across 123 medical centres in 16 countries. Eligible patients were aged 18 years or older, had an Eastern Cooperative Oncology Group performance status at baseline of 0–1, had histologically confirmed, relapsed, high-grade serous or high-grade endometrioid ovarian cancer, including primary peritoneal or fallopian tube cancer, and had received two or more previous platinum regimens. Patients were randomly assigned (2:1) to receive olaparib tablets (300 mg in two 150 mg tablets twice daily) or matching placebo tablets using an interactive web or voice-response system. Stratification was by response to previous chemotherapy and length of platinum-free interval. Treatment assignment was masked to patients, treatment providers, and data assessors. The primary endpoint of progression-free survival has been reported previously. Overall survival was a key secondary endpoint and was analysed in all patients as randomly allocated. Safety was assessed in all patients who received at least one treatment dose. This trial is registered with ClinicalTrials.gov, NCT01874353, and is no longer recruiting patients. Findings: Between Sept 3, 2013 and Nov 21, 2014, 295 patients were enrolled. Patients were randomly assigned to receive either olaparib (n=196 [66%]) or placebo (n=99 [34%]). Abstract

Byline: T. Haga, H. Sakaguchi, H. Watari, S. Kumai A High-Speed Twin-Roll Caster (HSTRC) was designed and used in order to increase casting speeds. Several Al-Mg-Si alloys were cast at a speed of 60 m/min. The Si content in these alloys is in the range of 0.57mass%, and the Mg content is 0.3mass%. Strips thinner than 4 mm can be cast continuously from the Al-Mg-Si alloys. The strips have an equiaxed, rather than columnar, grain microstructure. Rolled strips of 0.5 mm thickness with T4 heat treatments are not broken after 180-degree bending. The retained eutectic Si particles after T6 heat treatment are smaller than 5 mm and spherical.

Abstract

A systematic analysis of saturation transfer (ST) electron paramagnetic resonance spectroscopy was developed to separate an observed absorption signal either into an amplitude and a phase spectrum or into an in-phase and an ST spectrum. We established a phase reference procedure, replacing the low-power phase null method by a high-power phase maximum method. This is an extension of the finding that a peak-to-peak height and a single integration of the second harmonic absorption display vary sinusoidally as functions of the modulation phase [Y. Shimoyama and H. Watari, Appl. Spectrosc. 39, 170 (1985)]. Based on the sinusoidal variation in the peak-to-peak height, we found a phase shift at each magnetic field and could evaluate the amplitude and phase of a signal vector. The sinusoidal variation of the single integrated value allowed the absorption signal to be separated into an in-phase and an ST spectrum. The high-power phase maximum method provides a remedy of the phase setting problem in ST spectroscopy by which the in-phase signal can be detected at any microwave field. [ABSTRACT FROM AUTHOR]

Abstract

The aim of this study was to analyse the influence of diabetes mellitus as a prognostic factor for overall survival in endometrial cancer.Charts were reviewed from patients with endometrial carcinoma from 1985 to 2003. Data on clinicopathologic variables, adjuvant treatment, site of recurrence and survival were collected. The chi-square test was used to examine associations between variables. The Kaplan-Meier method was used for survival analysis and Cox's proportional hazards model for multiple regression analysis.Multivariate analysis revealed that diabetes mellitus, FIGO stage and depth of myometrial invasion were significantly associated with overall survival.

50. Mutant p53 gain-of-function induces epithelial–mesenchymal transition through modulation of the miR-130b–ZEB1 axis

Abstract

The tumor suppressor gene p53 has been implicated in the regulation of epithelial–mesenchymal transition (EMT) and tumor metastasis by regulating microRNA (miRNA) expression. Here, we report that mutant p53 exerts oncogenic functions and promotes EMT in endometrial cancer (EC) by directly binding to the promoter of miR-130b (a negative regulator of ZEB1) and inhibiting its transcription. We transduced p53 mutants into p53-null EC cells, profiled the miRNA expression by miRNA microarray and identified miR-130b as a potential target of mutant p53. Ectopic expression of p53 mutants repressed the expression of miR-130b and triggered ZEB1-dependent EMT and cancer cell invasion. Loss of an endogenous p53 mutation increased the expression of miR-130b, which resulted in reduced ZEB1 expression and attenuation of the EMT phenotype. Furthermore, re-expression of miR-130b suppressed mutant p53-induced EMT and ZEB1 expression. Importantly, the expression of miR-130 was significantly reduced in EC tissues, and patients with higher expression levels of miR-130b survived longer. These data provide a novel understanding of the roles of p53 gain-of-function mutations in accelerating tumor progression and metastasis through modulation of the miR-130b–ZEB1 axis.
