18. In-Gel Detection of DNA.

Abstract

The introduction of pulsed-field agarose gel electrophoresis (PFGE) has expanded the list of particles separable by use of gel electrophoresis to include: (1) linear DNAs as long as 3-6 Mbp, (2) DNA- protein complexes and circular DNAs that become arrested during invariant field agarose gel electrophoresis; and (3) micron-sized spheres that also become arrested during invariant field agarose gel electrophoresis (reviewed in refs. 1-3). During the replication, recombination and packaging of DNA by the various double-stranded DNA bacteriophages, circular DNAs, protein-DNA complexes, and end-to- end joined mature DNA multimers (concatemers) as long as 0.5-1.0 Mbp are formed (reviewed in refs. 4,5). Thus, in addition to being useful for genome mapping (1,2,6,7 and see alsoChapter 18), PFGE also is useful for studying the DNA metabolism of both bacteriophages and, presumably, other viruses. 22. Influence of growth media on vancomycin resistance of Enterococcus isolates and correlation with resistance gene determinants

Abstract

The effect of Mueller–Hinton (MH), MH+blood or brain heart infusion medium (agar or broth) on 13 Enterococcus isolates was determined, when testing their antibiotic susceptibility. Disk diffusion and Vitek methods were used to determine vancomycin resistance, while broth dilution and E-test methods were used to measure the minimum inhibitory concentration. The data were correlated with the presence of vancomycin resistance genes. A definite correlation pattern could not be established between the presence of van genes and vancomycin resistance in any plating medium, when tested by the disk diffusion assay. The broth dilution, irrespective of the plating medium, and Vitek methods were more reliable than the E-test method in testing isolates with vanA or vanB genes. However, for vanC2/C3 genotypes, the E-test method, irrespective of the plating medium, tested better than the broth dilution assay. [Copyright &y& Elsevier]

Abstract

Fundamental income tax concepts should be taught in the elementary accounting course. The important reasons for this contention are: (1) Students are interested in the subject of income tax, and they have a need for tax knowledge. (2) Some accounting topics are taught better through the comparison of income tax and conventional accounting treatment. (3) A benefit may accrue to the teaching of the income tax accounting courses since accounting students will bring a better grounding in tax fundamentals to the course. Greater depth of treatment may be possible than is presently the case in courses in income tax accounting. Morphology, antigenicity, and nucleic acid content of the Bacteroidessp. used in the culture of Entamoeba histolytica. J. Bacteriol. 90:1045–1053. 1965.—Certain changes in morphology, antigenicity, and nucleic acid content that occur in a culture of Bacteroidessp. in the presence of penicillin G in CLG medium are described. This “variant” is one of seven recovered in several laboratories, all of which are descendants of the original Bacteroidesisolated by Shaffer and Frye. Penicillin-inhibited cells of this culture are currently being used in the routine propagation of Entamoeba histolyticain CLG medium. Evidence is presented for the loss of ability to react with antibody in these penicillin-inhibited bacteria in CLG medium, when studied by fluorescent-antibody techniques. The implications of the antigenic changes observed as they pertain to similar antigenic studies of the amoebas are discussed. A pronounced reduction in the ribonucleic acid (RNA) content of such penicillin-inhibited cells was also observed. The potential importance of the changes that occur in the RNA of these cells with respect to considerations of the growth requirements of the amoebas is also discussed.
