Biodegradable polymers are an important part of the solution toolbox to achieve circularity in the plastic economy and overcome negative impacts of a linear plastic economy. Biodegradable polymers need to excel not only on a mechanical performance level in the application to fulfill their function during the use phase but also on a biodegradation performance level after use. The biodegradation performance is tailored to the application and the receiving environment of the polymer product after use, which can be both engineered systems (e.g., compost, anaerobic digestors, wastewater treatment plants) and natural systems (e.g., soils, freshwater, or marine environments). This chapter addresses key aspects of polymer biodegradability and biodegradation in both natural and engineered systems with the goal to advance a more holistic view on the topic and, thereby, provide guidance for all stakeholders working on developing, testing, and regulating biodegradable polymers. These aspects include definitions of biodegradability and biodegradation, elucidating polymer- and environmental factors that control the biodegradation process, a discussion of the analytical chemistry of polymer biodegradation, polymer biodegradability testing and certification, as well as a brief overview of research needs. In accordance with the diverse backgrounds of the authors of the chapter, this chapter targets all stakeholder groups from academics to industry and regulators. [ABSTRACT FROM AUTHOR]

Objective: Although delirium is well known to acute care clinicians, the features required for its diagnosis and how to understand and operationalize them remain sticking points in the field. To clarify the delirium phenotype, we present a close reading of past and current sets of delirium diagnostic criteria. Methods: We first differentiate the delirium syndrome (i.e., features evaluated at bedside) from additional criteria required for diagnosis. Next, we align related features across diagnostic systems and examine them in context to determine intent. Where criteria are ambiguous, we review common delirium instruments to illustrate how they have been interpreted. Results: An acute disturbance in attention is universally attested across diagnostic systems. A second core feature denotes confusion and has been included across systems as disturbance in awareness, impaired consciousness, and thought disorganization. This feature may be better understood as a disturbance in thought clarity and operationalized in terms of neuropsychological domains thereby clearly linking it to global neurocognitive disturbance. Altered level of activity describes a third core feature, including motor and sleep/wake cycle disturbances. Excluding stupor (wherein mental content cannot be assessed due to reduced arousal) from delirium, as in DSM‐5‐TR, is appropriate for a psychiatric diagnosis, but the brain injury exclusion in ICD‐11 is unjustified. Conclusions: The delirium phenotype involves a disturbance in attention, qualitative thought clarity, and quantitative activity level, including in relation to expected sleep/wake cycles. Future diagnostic systems should include a severity threshold and specify that delirium diagnosis refers to a 24‐h period. Keratin intermediate filaments are an essential and major component of the cytoskeleton in epithelial cells. They form a stable yet dynamic filamentous network extending from the nucleus to the cell periphery, which provides resistance to mechanical stresses. Mutations in keratin genes are related to a variety of epithelial tissue diseases. Despite their importance, the molecular structure of keratin filaments remains largely unknown. In this study, we analyzed the structure of keratin 5/keratin 14 filaments within ghost mouse keratinocytes by cryo-electron microscopy and cryo-electron tomography. By averaging a large number of keratin segments, we have gained insights into the helical architecture of the filaments. Two-dimensional classification revealed profound variations in the diameter of keratin filaments and their subunit organization. Computational reconstitution of filaments of substantial length uncovered a high degree of internal heterogeneity along single filaments, which can contain regions of helical symmetry, regions with less symmetry and regions with significant diameter fluctuations. Cross-section views of filaments revealed that keratins form hollow cylinders consisting of multiple protofilaments, with an electron dense core located in the center of the filament. These findings shed light on the complex and remarkable heterogenic architecture of keratin filaments, suggesting that they are highly flexible, dynamic cytoskeletal structures. Amyotrophic lateral sclerosis (ALS) is a rapidly progressing neurodegenerative disease characterized by the loss of motor control. Current understanding of ALS pathology is largely based on post-mortem investigations at advanced disease stages. A systematic in vivo description of the microstructural changes that characterize early stage ALS, and their subsequent development, is so far lacking. Recent advances in ultra-high field (7 T) MRI data modelling allow us to investigate cortical layers in vivo. Given the layer-specific and topographic signature of ALS pathology, we combined submillimetre structural 7 T MRI data (qT1, QSM), functional localizers of body parts (upper limb, lower limb, face) and layer modelling to systematically describe pathology in the primary motor cortex (M1), in 12 living ALS patients with reference to 12 matched controls. Longitudinal sampling was performed for a subset of patients. We calculated multimodal pathology maps for each layer (superficial layer, layer 5a, layer 5b, layer 6) of M1 to identify hot spots of demyelination, iron and calcium accumulation in different cortical fields. We show preserved mean cortical thickness and layer architecture of M1, despite significantly increased iron in layer 6 and significantly increased calcium in layer 5a and superficial layer, in patients compared to controls. The behaviourally first-affected cortical field shows significantly increased iron in L6 compared to other fields, while calcium accumulation is atopographic and significantly increased in the low myelin borders between cortical fields compared to the fields themselves. A subset of patients with longitudinal data shows that the low myelin borders are particularly disrupted and that calcium hot spots, but to a lesser extent iron hot spots, precede demyelination. Finally, we highlight that a very slow progressing patient (Patient P4) shows a distinct pathology profile compared to the other patients. Our data show that layer-specific markers of in vivo pathology can be identified in ALS patients with a single 7 T MRI measurement after first diagnosis, and that such data provide critical insights into the individual disease state. Our data highlight the non-topographic architecture of ALS disease spread and the role of calcium, rather than iron accumulation, in predicting future demyelination. We also highlight a potentially important role of low myelin borders, that are known to connect to multiple areas within the M1 architecture, in disease spread. Finally, the distinct pathology profile of a very-slow progressing patient (Patient P4) highlights a distinction between disease duration and progression. The unprecedented rate of CO2 increase in our atmosphere and subsequent ocean acidification (OA) threatens coastal ecosystems. To forecast the functioning of coastal seagrass ecosystems in acidified oceans, more knowledge on the long-term adaptive capacities of seagrass species and their epibionts is needed. Therefore we studied morphological characteristics of Posidonia oceanica and the structure of its epibiont communities at a Mediterranean volcanic CO2 vent off Panarea Island (Italy) and performed a laboratory experiment to test the effect of OA on P. oceanica photosynthesis and its potential buffering capacity. At the study site east of Basiluzzo Islet, venting of CO2 gas was controlled by tides, resulting in an average pH difference of 0.1 between the vent and reference site. P. oceanica shoot and leaf density was unaffected by these levels of OA, although shorter leaves at the vent site suggest increased susceptibility to erosion, potentially by herbivores. The community of sessile epibionts differed in composition and was characterized by a higher species richness at the vent site, though net epiphytic calcium carbonate concentration was similar. These findings suggest a higher ecosystem complexity at the vent site, which may have facilitated the higher diversity of copepods in the otherwise unaffected motile epibiont community. In the laboratory experiment, P. oceanica photosynthesis increased with decreasing pHT (7.6, 6.6, 5.5), which induced an elevated pH at the leaf surfaces of up to 0.5 units compared to the ambient seawater pHT of 6.6. This suggests a temporary pH buffering in the diffusive boundary layer of leaves, which could be favorable for epibiont organisms. The results of this multispecies study contribute to understanding community-level responses and underlying processes in long-term acidified conditions. Increased replication and monitoring of physico-chemical parameters on an annual scale are, however, recommended to assure that the biological responses observed during a short period reflect long-term dynamics of these parameters. Topographic maps form a critical feature of cortical organization, yet are poorly described with respect to their microstructure in the living aging brain. We acquired quantitative structural and functional 7T-MRI data from younger and older adults to characterize layer-wise topographic maps of the primary motor cortex (M1). Using parcellation-inspired techniques, we show that quantitative T1 and Quantitative Susceptibility Maps values of the hand, face, and foot areas differ significantly, revealing microstructurally distinct cortical fields in M1. We show that these fields are distinct in older adults and that myelin borders between them do not degenerate. We further show that the output layer 5 of M1 shows a particular vulnerability to age-related increased iron, while layer 5 and the superficial layer show increased diamagnetic substance, likely reflecting calcifications. Taken together, we provide a novel 3D model of M1 microstructure, where body parts form distinct structural units, but layers show specific vulnerability toward increased iron and calcium in older adults. Oxygenic and anoxygenic photosynthesis were studied with microsensors in microbial mats found at 9-10 m depth in anoxic and sulfidic water in Little Salt Spring (Florida, USA). The lake sediments were covered with a 1-2 mm thick red mat dominated by filamentous Cyanobacteria, below which Green Sulfur Bacteria (GSB, Chlorobiaceae) were highly abundant. Within 4 mm inside the mats, the incident radiation was attenuated to undetectable levels. In situ microsensor data showed both oxygenic photosynthesis in the red surface layer and light-induced sulfide dynamics up to 1 cm depth. Anoxygenic photosynthesis occurred during all daylight hours, with complete sulfide depletion around midday. Oxygenic photosynthesis was limited to 4 h per day, due to sulfide inhibition in the early morning and late afternoon. Laboratory measurements on retrieved samples showed that oxygenic photosynthesis was fully but reversibly inhibited by sulfide. In patches Fe(III) alleviated the inhibition of oxygenic photosynthesis by sulfide. GSB were resistant to oxygen and showed a low affinity to sulfide. In this study, indicators are proposed to characterize the intrinsic environmental properties and external value of urban parks. The intrinsic properties involve both acoustic factors and non-acoustic factors, such as visual aspects and size. To assess external value, the restoration level is defined, which measures the nearby presence of a quiet, ‘green’ area at residential areas outside parks. The restoration levels of green areas are based on intrinsic properties and the distances of each dwelling to urban park areas. The overall environmental value of a park, the group restoration level, is defined as a logarithmic summation of the restoration levels over its surrounding residential areas. Restoration levels were determined for sixteen public parks in the city of Rotterdam and compared with survey data from questionnaires. Results show that the investigated parks display a large variation in the group restoration level levels, mainly due to differences in size and average noise levels. Abundant microbial mats, recently discovered in underwater freshwater springs in the hypersaline Dead Sea, are mostly dominated by sulfur-oxidizing bacteria. We investigated the source of sulfide and the activity of these communities. Isotopic analysis of sulfide and sulfate in the spring water showed a fractionation of 39-50‰ indicative of active sulfate reduction. Sulfate reduction rates ( SRR) in the spring sediment (< 2.8 nmol cm−3 day−1) are too low to account for the measured sulfide flux. Thus, sulfide from the springs, locally reduced salinity and O2 from the Dead Sea water are responsible for the abundant microbial biomass around the springs. The springs flow is highly variable and accordingly the local salinities. We speculate that the development of microbial mats dominated by either Sulfurimonas/ Sulfurovum-like or Thiobacillus/ Acidithiobacillus-like sulfide-oxidizing bacteria, results from different mean salinities in the microenvironment of the mats. SRR of up to 10 nmol cm−3 day−1 detected in the Dead Sea sediment are surprisingly higher than in the less saline springs. While this shows the presence of an extremely halophilic sulfate-reducing bacteria community in the Dead Sea sediments, it also suggests that extensive salinity fluctuations limit these communities in the springs due to increased energetic demands for osmoregulation. Cyanobacteria and diatom mats are ubiquitous in hypersaline environments but have never been observed in the Dead Sea, one of the most hypersaline lakes on Earth. Here we report the discovery of phototrophic microbial mats at underwater freshwater seeps in the Dead Sea. These mats are either dominated by diatoms or unicellular cyanobacteria and are spatially separated. Using in situ and ex situ O microsensor measurements we show that these organisms are photosynthetically active in their natural habitat. The diatoms, which are phylogenetically associated to the Navicula genus, grew in culture at salinities up to 40 % Dead Sea water (DSW) (14 % total dissolved salts, TDS). The unicellular cyanobacteria belong to the extremely halotolerant Euhalothece genus and grew at salinities up to 70 % DSW (24.5 % TDS). As suggested by a variable O penetration depth measured in situ, the organisms are exposed to drastic salinity fluctuations ranging from brackish to DSW salinity within minutes to hours. We could demonstrate that both phototrophs are able to withstand such extreme short-term fluctuations. Nevertheless, while the diatoms recover better from rapid fluctuations, the cyanobacteria cope better with long-term exposure to DSW. We conclude that the main reason for the development of these microbial mats is a local dilution of the hypersaline Dead Sea to levels allowing growth. Environmental policy is characterised by complexity, in causes and effects, resulting in various combinations of policy instruments. However, evaluating these policy instrument mixes and assessing their effectiveness is difficult because of a lack of methodological approaches. This paper therefore proposes a methodology which comprises: (a) describing the underlying policy theory; (b) describing the policy instruments; (c) analysing goal attainment; and (d) evaluating effectiveness, focusing on coverage of points of intervention, steering power of policy instruments and coherence of the policy instruments mix. The methodology is illustrated with an evaluation of noise policy in the Netherlands – a typical complex policy domain in which a mix of policy instruments has been in place for decades, and thus provides a good empirical case. This paper is an outcome of a workshop that addressed the question how soundscape research can improve its impact on the local level. It addresses a number of topics by complementing existing approaches and practices with possible future approaches and practices. The paper starts with an analysis of the role of sound annoyance and suboptimal soundscapes on the lives of individuals and concludes that a good soundscape, or more generally a good sensescape, is at the same time pleasant as well as conducive for the adoption of healthy habits. To maintain or improve sensescape quality, urban planning needs improved design tools that allow for a more holistic optimization and an active role of the local stakeholders. Associated with this is a gradual development from government to governance in which optimization of the soundscape at a local (administrative or geographic) level is directly influenced by the users of spaces. The paper concludes that soundscape research can have a greater impact by helping urban planners design for health and pleasant experiences as well as developing tools for improved citizen involvement in local optimization. Noise exposure has harmful effects on human health. Despite policy on the prevention and reduction of noise, the environmental burden is increasing, specifically due to road traffic noise. Noise policy in the Netherlands is organised in a rather complex way, with different legal frameworks for the various sources of noise. Whereas noise limits have frequently been adjusted in the traffic noise policy subsystem, the industrial and aviation noise policy subsystems are characterised by stability in norm setting. This paper aims to explain the differences in dynamics within the noise policy subsystems, by applying the Advocacy Coalition Framework (ACF). We conclude that the dynamics in the traffic noise policy subsystem is mainly due to two adversary coalitions advocating legislative arrangements to accommodate respective spatial claims. The stability in industrial and aircraft noise policy subsystems is explained by ‘balanced’ coalitions and a dominant economy coalition, respectively. We identified the (only) path to policy change in Dutch noise policy to be cross-coalition learning in which ‘policy brokerage’ might be crucial. We conclude with some reflections on the use of ACF in empirical research and the role of professional forums and institutional arrangements in stability and/or change in policy subsystems. Background: Coral reefs degrade globally at an alarming rate, with benthic algae often replacing corals. However, the extent to which benthic algae contribute to coral mortality, and the potential mechanisms involved, remain disputed. Recent laboratory studies suggested that algae kill corals by inducing hypoxia on the coral surface, through stimulated microbial respiration. Methods/Findings: We examined the main premise of this hypothesis by measuring in situ oxygen microenvironments at the contact interface between the massive coral Porites spp. and turf algae, and between Porites spp. and crustose coralline algae (CCA). Oxygen levels at the interface were similar to healthy coral tissue and ranged between 300-400 mM during the day. At night, the interface was hypoxic (∼70 μM) in coral-turf interactions and close to anoxic (∼2 μM) in coral-CCA interactions, but these values were not significantly different from healthy tissue. The diffusive boundary layer (DBL) was about three times thicker at the interface than above healthy tissue, due to a depression in the local topography. A numerical model, developed to analyze the oxygen profiles above the irregular interface, revealed strongly reduced net photosynthesis and dark respiration rates at the coral-algal interface compared to unaffected tissue during the day and at night, respectively. Conclusions/Significance: Our results showed that hypoxia was not a consistent feature in the microenvironment of the coral-algal interface under in situ conditions. Therefore, hypoxia alone is unlikely to be the cause of coral mortality. Due to the modified topography, the interaction zone is distinguished by a thicker diffusive boundary layer, which limits the local metabolic activity and likely promotes accumulation of potentially harmful metabolic products (e.g., allelochemicals and protons). Interest in environmental policy integration (EPI) has recently been strong, both in the literature and in practice. We explore Dutch initiatives to integrate noise management into spatial planning policy in light of the body of literature on EPI. The main approaches of EPI are translated into a conceptual framework consisting of organizational, procedural, and contextual factors. The objective of this literature review is to relate paradigm shifts and policy innovations regarding noise management and spatial planning to empirical windows of opportunity for and barriers to implementation of EPI. It shows how instruments allowing a flexible approach and deviation from standards at the local level fit in with the discourse on decentralized and area-oriented policy. The analysis suggests that procedural and decision-making rules and organizational arrangements can bridge implementation gaps in local-level planning practice. However, EPI in the Netherlands has not solved the noise problem, and the number of affected inhabitants is increasing. Steroid receptors belonging to the superfamily of nuclear receptors do not exist as single monomeric proteins but mediate their effects by the interaction with numerous other proteins, e.g., cofactors for transcription, but also other proteins involved in cellular signaling. This interaction may be ligand dependent, which explains the differential effects of receptor ligands. Whereas some receptors, e.g., the estrogen receptor, have been studied in great detail, much less is known about proteins interacting with the mineralocorticoid receptor (MR). In this study, we aimed to identify interacting proteins using a proteomics approach involving tagged receptor constructs. After affinity isolation of MR complexes, blue native electrophoresis revealed the presence of several populations of MR complexes differing in size and composition. During the identification of interacting proteins, various heat shock proteins but also several previously undescribed potential interactors were found, including 14-3-3-ε. We also demonstrate here that the cytosolic MR in the presence of detergent interacts in a ligand-selective manner with glucose-regulated protein 78 and propionyl-CoA carboxylase-β precursor, which are found in the unliganded or aldosterone-containing complex but not with spironolactone. Microsensors are powerful tools for microenvironment studies, however their use has often been restricted to laboratory applications due to the lack of adequate equipment for in situ deployments. Here we report on new features, construction details, and examples of applications of an improved diver-operated motorized microsensor profiler for underwater field operation to a water depth of 25 m. The new motorized profiler has a final precision of 5 µm, and can accommodate amperometric Clark-type microsensors for oxygen and hydrogen sulfide, potentiometric microsensors (e.g., for pH, Ca2+, and fiber-optic irradiance microsensors. The profiler is interfaced by a logger with a signal display, and has pushbuttons for underwater operation. The system can be pre-programmed to autonomous operation or interactively operated by divers. Internal batteries supply power for up to 24 h of measurements and 36 h of data storage (max. 64 million data points). Two flexible stands were developed for deployment on uneven or fragile surfaces, such as coral reefs. Three experimental pilot studies are presented, where (1) the oxygen distribution in a sand ripple was 3-0-mapped, (2) the microenvironment of sediment accumulated on a stony coral was studied, and (3) oxygen dynamics during an experimental sedimentation were investigated. The architecture of protein assemblies and their remodeling during physiological processes is fundamental to cells. Therefore, providing high-resolution snapshots of macromolecular complexes in their native environment is of major importance for understanding the molecular biology of the cell. Cellular structural biology by means of cryo-electron tomography (cryo-ET) offers unique insights into cellular processes at an unprecedented resolution. Recent technological advances have enabled the detection of single impinging electrons and improved the contrast of electron microscopic imaging, thereby significantly increasing the sensitivity and resolution. Moreover, various sample preparation approaches have paved the way to observe every part of a eukaryotic cell, and even multicellular specimens, under the electron beam. Imaging of macromolecular machineries at high resolution directly within their native environment is thereby becoming reality. In this review, we discuss several sample preparation and labeling techniques that allow the visualization and identification of macromolecular assemblies in situ, and demonstrate how these methods have been used to study eukaryotic cellular landscapes. Simple Summary: Patients with brain tumors often experience changes in memory and other aspects of thinking. Many may also have difficulty with social cognition, affecting the abilities that facilitate social behavior and maintain social relationships; however, the data are limited. Whether treatments such as cranial radiation can impact social cognition is not well-studied. We sought to understand how radiation dose exposure to the amygdala–orbitofrontal network, which subserves social cognition and emotion recognition, impacted performance on an emotion recognition task. We found that radiation dose to the amygdala and associated structures was associated with performance on an emotion recognition task, including longer response times with increasing radiation doses. Radiation techniques that reduce the dose to the amygdala-orbitofrontal network may decrease side effects for patients receiving cranial radiation. Background: Although data are limited, difficulty in social cognition occurs in up to 83% of patients with brain tumors. It is unknown whether cranial radiation therapy (RT) dose to the amygdala–orbitofrontal network can impact social cognition. Methods: We prospectively enrolled 51 patients with low-grade and benign brain tumors planned for cranial RT. We assessed longitudinal changes on an emotion recognition task (ERT) that measures the ability to recognize emotional states by displaying faces expressing six basic emotions and their association with the RT dose to the amygdala–orbitofrontal network. ERT outcomes included the median time to choose a response (ERTOMDRT) or correct response (ERTOMDCRT) and total correct responses (ERTHH). Results: The RT dose to the amygdala–orbitofrontal network was significantly associated with longer median response times on the ERT. Increases in median response times occurred at lower doses than decreases in total correct responses. The medial orbitofrontal cortex was the most important variable on regression trees predicting change in the ERTOMDCRT. Discussion: This is, to our knowledge, the first study to show that off-target RT dose to the amygdala–orbitofrontal network is associated with performance on a social cognition task, a facet of cognition that has previously not been mechanistically studied after cranial RT. The article discusses the mechanisms by which corals are damaged upon exposed to sedimentation. It reports that coral death is caused by adverse conditions within enriched sediment and highlights that its death is accelerated by the hydrogen sulfide produced from microbial degradation of coral tissue. The experimental results point out that after twenty four hours of exposure to enriched sediments the first dead tissues in coral is observed.

Vestimentiferan Tws colonize hydrothermal vents and cold seeps worldwide. They lack a digestive system and gain nutrition from endosymbiotic sulfur-oxidizing bacteria. It is currently assumed that vestimentiferan Tws harbour only a single endosymbiont type. A few studies found indications for additional symbionts, but conclusive evidence for a multiple symbiosis is still missing. We investigated Tws from Marsili Seamount, a hydrothermal vent in the Mediterranean Sea. Molecular and morphological analyses identified the Tws as L amellibrachia anaximandri. 16S ribosomal RNA clone libraries revealed two distinct gammaproteobacterial phylotypes that were closely related to sequences from other L amellibrachia symbionts. Catalysed reporter deposition fluorescence in situ hybridization with specific probes showed that these sequences are from two distinct symbionts. We also found two variants of key genes for sulfur oxidation and carbon fixation, suggesting that both symbiont types are autotrophic sulfur oxidizers. Our results therefore show that vestimentiferans can host multiple co-occurring symbiont types. Statistical analyses of vestimentiferan symbiont diversity revealed that host genus, habitat type, water depth and geographic region together accounted for 27% of genetic diversity, but only water depth had a significant effect on its own. Phylogenetic analyses showed a clear grouping of sequences according to depth, thus confirming the important role water depth played in shaping vestimentiferan symbiont diversity. The cytoskeleton of eukaryotic cells is primarily composed of networks of filamentous proteins, F-actin, microtubules, and intermediate filaments. Interactions among the cytoskeletal components are important in determining cell structure and in regulating cell functions. For example, F-actin and microtubules work together to control cell shape and polarity, while the subcellular organization and transport of vimentin intermediate filament (VIF) networks depend on their interactions with microtubules. However, it is generally thought that F-actin and VIFs form two coexisting but separate networks that are independent due to observed differences in their spatial distribution and functions. In this paper, we present a closer investigation of both the structural and functional interplay between the F-actin and VIF cytoskeletal networks. We characterize the structure of VIFs and F-actin networks within the cell cortex using structured illumination microscopy and cryoelectron tomography. We find that VIFs and F-actin form an interpenetrating network (IPN) with interactions at multiple length scales, and VIFs are integral components of F-actin stress fibers. From measurements of recovery of cell contractility after transient stretching, we find that the IPN structure results in enhanced contractile forces and contributes to cell resilience. Studies of reconstituted networks and dynamic measurements in cells suggest direct and specific associations between VIFs and F-actin. From these results, we conclude that VIFs and F-actin work synergistically, both in their structure and in their function. These results profoundly alter our understanding of the contributions of the components of the cytoskeleton, particularly the interactions between intermediate filaments and F-actin. Marine coastlines colonized by seagrasses are a net source of methane to the atmosphere. However, methane emissions from these environments are still poorly constrained, and the underlying processes and responsible microorganisms remain largely unknown. Here, we investigated methane turnover in seagrass meadows of Posidonia oceanica in the Mediterranean Sea. The underlying sediments exhibited median net fluxes of methane into the water column of ca. 106 μmol CH4 ( m22 ( d21. Our data show that this methane production was sustained by methylated compounds produced by the plant, rather than by fermentation of buried organic carbon. Interestingly, methane production was maintained long after the living plant died off, likely due to the persistence of methylated compounds, such as choline, betaines, and dimethylsulfoniopropionate, in detached plant leaves and rhizomes. We recovered multiple mcrA gene sequences, encoding for methyl-coenzyme M reductase (Mcr), the key methanogenic enzyme, from the seagrass sediments. Most retrieved mcrA gene sequences were affiliated with a clade of divergent Mcr and belonged to the uncultured Candidatus Helarchaeota of the Asgard superphylum, suggesting a possible involvement of these divergent Mcr in methane metabolism. Initiation of combination antiretroviral therapy (cART) reduces inflammation in HIV-infected (HIV+) individuals. Recent studies demonstrated that diffusion MRI based extracellular free water (FW) modeling can be sensitive to neuroinflammation. Here, we investigate the FW in HIV-infection, its temporal evolution, and its association with blood markers, and cognitive scores. Using 96 age-matched participants, we found that FW was significantly elevated in grey and white matter in cART-naïve HIV+ compared to HIV-uninfected (HIV−) individuals at baseline. These increased FW values positively correlated with neurofilament light chain (NfL) and negatively correlated with CD4 counts. FW in grey and white matter, as well as NfL decreased in the HIV+ after 12 weeks of cART treatment. No significant FW differences were noted between the HIV+ and HIV− cohorts at 1 and 2-year follow-up. Results suggest that FW elevation in cART-naïve HIV+ participants is likely due to neuroinflammation. The correlation between FW and NfL, and the improvement in both FW and NfL after 12 weeks of cART treatment further reinforces this conclusion. The longer follow-up at 1 and 2 years suggests that cART helped control neuroinflammation as inferred by FW. Therefore, FW could be used as a biomarker to monitor HIV-associated neuroinflammation. Objectives: To perform a scoping literature review of cognitive, psychiatric, and quality of life outcomes in adults undergoing extracorporeal membrane oxygenation for any indication.Data Sources: We searched PubMed, EMBASE, Cochrane Library, Web of Science, CINAHL, and PsycINFO from inception to June 2019.Study Selection: Observational studies, clinical trials, qualitative studies, and case series with at least 10 adult subjects were included for analysis. Outcomes of interest consisted of general or domain-specific cognition, psychiatric illness, and quality of life measures that included both mental and physical health.Data Extraction: Study selection, data quality assessment, and interpretation of results were performed by two independent investigators in accordance with the PRISMA statement.Data Synthesis: Twenty-two articles were included in this review. Six described cognitive outcomes, 12 described psychiatric outcomes of which two were qualitative studies, and 16 described quality of life outcomes. Cognitive impairment was detected in varying degrees in every study that measured it. Three studies examined neuroimaging results and found neurologic injury to be more frequent in venoarterial versus venovenous extracorporeal membrane oxygenation, but described a variable correlation with cognitive impairment. Rates of depression, anxiety, and post-traumatic stress disorder were similar to other critically ill populations and were related to physical disability after extracorporeal membrane oxygenation. Extracorporeal membrane oxygenation survivors' physical quality of life was worse than population norms but tended to improve with time, while mental quality of life did not differ significantly from the general population. Most studies did not include matched controls and instead compared outcomes to previously published values.Conclusions: Extracorporeal membrane oxygenation survivors experience cognitive impairment, psychiatric morbidity, and worse quality of life compared with the general population and similar to other survivors of critical illness. Physical disability in extracorporeal membrane oxygenation patients plays a significant role in psychiatric morbidity. The pollution of the natural environment, especially the world's oceans, with conventional plastic is of major concern. Biodegradable plastics are an emerging market bringing along potential chances and risks. The fate of these materials in the environment and their possible effects on organisms and ecosystems has rarely been studied systematically and is not well understood. For the marine environment, reliable field test methods and standards for assessing and certifying biodegradation to bridge laboratory respirometric data are lacking. In this work we present newly developed field tests to assess the performance of (biodegradable) plastics under natural marine conditions. These methods were successfully applied and validated in three coastal habitats (eulittoral, benthic and pelagic) and two climate zones (Mediterranean Sea and tropical Southeast Asia). Additionally, a stand-alone mesocosm test system which integrated all three habitats in one technical system at 400-L scale independent from running seawater is presented as a methodological bridge. Films of polyhydroxyalkanoate copolymer (PHA) and low density polyethylene (LD-PE) were used to validate the tests. While LD-PE remained intact, PHA disintegrated to a varying degree depending on the habitat and the climate zone. Together with the existing laboratory standard test methods, the field and mesocosm test systems presented in this work provide a 3-tier testing scheme for the reliable assessment of the biodegradation of (biodegradable) plastic in the marine environment. In the last few years, a large amount of research have been done in the field of youth care services. However, there is still a substantial need for empirical findings, especially on institutions with specific missions such as therapeutic residential groups for young people. This article grew out of cooperation between practitioners and researchers and presents the results of a German study entitled "Process Evaluation of Therapeutic Residential Groups for Young People" (TRGs). The main research question of the evaluation study was whether the treatment in the TRGs is specifically stabilizing, protective, and, above all, effective in dealing with young people with severe problems. The study employs multiple methodologies (qualitative and quantitative procedures) to obtain a satisfactorily comprehensive analysis of both outcome and process quality. The article presents selected results from the quantitative part of the study and the qualitative interview study and discusses them considering recent research in the field of child and adolescent care services. Cancer-related cognitive impairment (CRCI) has not been objectively assessed in chronic lymphocytic leukemia (CLL). It is currently unclear how much of CRCI is attributable to disease, treatment, or both. We used CLL as a novel model to study the differential roles of disease and treatment in CRCI. One hundred and fifty CLL patients (100 treatment-naïve and 50 chemotherapy-treated) including 84 patients with higher-risk of CLL progression completed objective neuropsychological tests. Sociodemographic-adjusted linear regression models examined cognitive outcomes in relation to risk and treatment. Higher-risk patients recalled two fewer words on a memory task (β = −1.8, 95%CI –3.3,−0.3) and took 15 s longer on an executive function task (β = 15.4, 95%CI 3.1, 27.6) than lower-risk patients, independent of treatment. Treated patients reported greater cognitive difficulties than treatment-naive patients (β = −6.1, 95%CI –10.1, −2.2) but did not perform worse on objective measures. We performed first experiments to evaluate the potential of photoluminescence spectroscopy to detect microplastics using a 405 nm laser diode for sample excitation. A set of nine bulk plastic samples and nine samples of natural materials typically found in the marine environment were examined and compared. The plastic and non-plastic materials could be differentiated based on the shape of the acquired photoluminescence spectra. Summary: The release of abiotic methane from marine seeps into the atmosphere is a major source of this potent greenhouse gas. Methanotrophic microorganisms in methane seeps use methane as carbon and energy source, thus significantly mitigating global methane emissions. Here, we investigated microbial methane oxidation at the sediment–water interface of a shallow marine methane seep. Metagenomics and metaproteomics, combined with 13C‐methane stable isotope probing, demonstrated that various members of the gammaproteobacterial family Methylococcaceae were the key players for methane oxidation, catalysing the first reaction step to methanol. We observed a transfer of carbon to methanol‐oxidizing methylotrophs of the betaproteobacterial family Methylophilaceae, suggesting an interaction between methanotrophic and methylotrophic microorganisms that allowed for rapid methane oxidation. From our microcosms, we estimated methane oxidation rates of up to 871 nmol of methane per gram sediment per day. This implies that more than 50% of methane at the seep is removed by microbial oxidation at the sediment–water interface, based on previously reported in situ methane fluxes. The organic carbon produced was further assimilated by different heterotrophic microbes, demonstrating that the methane‐oxidizing community supported a complex trophic network. Seepage of methane (CH4) on land and in the sea may significantly affect Earth’s biogeochemical cycles. However processes of CH4 generation and consumption, both abiotic and microbial, are not always clear. We provide new geochemical and isotope data to evaluate if a recently discovered CH4 seepage from the shallow seafloor close to the Island of Elba (Tuscany) and two small islands nearby are derived from abiogenic or biogenic sources and whether carbonate encrusted vents are the result of microbial or abiotic processes. Emission of gas bubbles (predominantly CH4) from unlithified sands was observed at seven spots in an area of 100 m2 at Pomonte (Island of Elba), with a total rate of 234 ml m-2 d-1. The measured carbon isotope values of CH4 of around -18‰ (VPDB) in combination with the measured δ2H value of -120‰ (VSMOW) and the inverse correlation of δ13C-value with carbon number of hydrocarbon gases are characteristic for sites of CH4 formation through abiogenic processes, specifically abiogenic formation of CH4 via reduction of CO2 by H2. The H2 for methanogenesis likely derives from ophiolitic host rock within the Ligurian accretionary prism. The lack of hydrothermal activity allows CH4 gas to become decoupled from the stagnant aqueous phase. Hence no hyperalkaline fluid is currently released at the vent sites. Within the seep area a decrease in porewater sulphate concentrations by ca. 5 mmol/l relative to seawater and a concomitant increase in sulphide and dissolved inorganic carbon (DIC) indicate substantial activity of sulphate-dependent anaerobic oxidation of methane (AOM). In absence of any other dissimilatory pathway, the δ13C-values between -17 and -5‰ in dissolved inorganic carbon and aragonite cements suggest that the inorganic carbon is largely derived from CH4. I-ACT is a theoretical framework designed to guide environmental health improvement efforts. I-ACT identifies four interacting drivers of change that influence an environmental health aim: (1) Information systems; (2) public Awareness; (3) leadership and Coordination; and, (4) Tools. Actors can use I-ACT to clarify roles and identify strategies to impact their aim. Here, we apply the I-ACT framework to a ubiquitous environmental hazard, noise pollution, comparing three Western countries: the United States, the Netherlands, and the United Kingdom. Our approach statutorily defines each country's designated aims, qualitatively evaluates its information systems, awareness, coordination, and tools, and assesses the role of these drivers in achieving the aims. While the Netherlands and the United Kingdom demonstrated robust activity for some drivers, the United States showed limited activity and achievement. There appeared to be an association between achievement of aims and demonstrated elements of each driver, providing support for the utility of I-ACT. [ABSTRACT FROM AUTHOR]

Our study aimed to investigate the short-term effect of combination antiretroviral therapy (cART) on cognitive performance and functional and structural connectivity and their relationship to plasma levels of antiretroviral (ARV) drugs. Seventeen ARV treatment-naïve HIV-infected individuals (baseline mean CD4 cell count, 479 ± 48 cells/mm) were age matched with 17 HIV-uninfected individuals. All subjects underwent a detailed neurocognitive and functional assessment and magnetic resonance imaging. HIV-infected subjects were scanned before starting cART and 12 weeks after initiation of treatment. Uninfected subjects were assessed once at baseline. Functional connectivity (FC) was assessed within the default mode network while structural connectivity was assessed by voxel-wise analysis using tract-based spatial statistics (TBSS) and probabilistic tractography within the DMN. Tenofovir and emtricitabine blood concentration were measured at week 12 of cART. Prior to cART, HIV-infected individuals had significantly lower cognitive performance than control subjects as measured by the total Z-score from the neuropsychological tests assessing six cognitive domains ( p = 0.020). After 12 weeks of cART treatment, there remained only a weak cognitive difference between HIV-infected and HIV-uninfected subjects ( p = 0.057). Mean FC was lower in HIV-infected individuals compared with those uninfected ( p = 0.008), but FC differences became non-significant after treatment ( p = 0.197). There were no differences in DTI metrics between HIV-infected and HIV-uninfected individuals using the TBSS approach and limited evidence of decreased structural connectivity within the DMN in HIV-infected individuals. Tenofovir and emtricitabine plasma concentrations did not correlate with either cognitive performance or imaging metrics. Conclusions: Twelve weeks of cART improves cognitive performance and functional connectivity in ARV treatment-naïve HIV-infected individuals with relatively preserved immune function. The production of biodegradable plastic is increasing. Given the augmented littering of these products an increasing input into the sea is expected. Previous laboratory experiments have shown that degradation of plastic starts within days to weeks. Little is known about the early composition and activity of biofilms found on biodegradable and conventional plastic debris and its correlation to degradation in the marine environment. In this study we investigated the early formation of biofilms on plastic shopper bags and its consequences for the degradation of plastic. Samples of polyethylene and biodegradable plastic were tested in the Mediterranean Sea for 15 and 33 days. The samples were distributed equally to a shallow benthic (sedimentary seafloor at 6 m water depth) and a pelagic habitat (3 m water depth) to compare the impact of these different environments on fouling and degradation. The amount of biofilm increased on both plastic types and in both habitats. The diatom abundance and diversity differed significantly between the habitats and the plastic types. Diatoms were more abundant on samples from the pelagic zone. We anticipate that specific surface properties of the polymer types induced different biofilm communities on both plastic types. Additionally, different environmental conditions between the benthic and pelagic experimental site such as light intensity and shear forces may have influenced unequal colonisation between these habitats. The oxygen production rate was negative for all samples, indicating that the initial biofilm on marine plastic litter consumes oxygen, regardless of the plastic type or if exposed in the pelagic or the benthic zone. Mechanical tests did not reveal degradation within one month of exposure. However, scanning electron microscopy (SEM) analysis displayed potential signs of degradation on the plastic surface, which differed between both plastic types. This study indicates that the early biofilm formation and composition are affected by the plastic type and habitat. Benthic cyanobacterial mats (BCMs) are impacting coral reefs worldwide. However, the factors and mechanisms driving their proliferation are unclear. We conducted a multi-year survey around the Caribbean island of Curaçao, which revealed highest BCM abundance on sheltered reefs close to urbanised areas. Reefs with high BCM abundance were also characterised by high benthic cover of macroalgae and low cover of corals. Nutrient concentrations in the water-column were consistently low, but markedly increased just above substrata (both sandy and hard) covered with BCMs. This was true for sites with both high and low BCM coverage, suggesting that BCM growth is stimulated by a localised, substrate-linked release of nutrients from the microbial degradation of organic matter. This hypothesis was supported by a higher organic content in sediments on reefs with high BCM coverage, and by an in situ experiment which showed that BCMs grew within days on sediments enriched with organic matter (Spirulina). We propose that nutrient runoff from urbanised areas stimulates phototrophic blooms and enhances organic matter concentrations on the reef. This organic matter is transported by currents and settles on the seabed at sites with low hydrodynamics. Subsequently, nutrients released from the organic matter degradation fuel the growth of BCMs. Improved management of nutrients generated on land should lower organic loading of sediments and other benthos (e.g. turf and macroalgae) to reduce BCM proliferation on coral reefs. An intertidal hypersaline cyanobacterial mat from Abu Dhabi (United Arab Emirates) exhibited a reversible change in its surface colour within several hours upon changes in salinity of the overlying water. The mat surface was orange-reddish at salinities above 15% and turned dark green at lower salinities. We investigated this phenomenon using a polyphasic approach that included denaturing gradient gel electrophoresis, microscopy, high-performance liquid chromatography, hyperspectral imaging, absorption spectroscopy, oxygen microsensor measurements and modelling of salinity dynamics. Filaments of Microcoleus chthonoplastes, identified based on 16S rRNA sequencing and morphology, were found to migrate up and down when salinity was decreased below or increased above 15%, respectively, causing the colour change of the mat uppermost layer. Migration occurred in light and in the dark, and could be induced by different salts, not only NaCl. The influence of salinity-dependent and independent physico-chemical parameters, such as water activity, oxygen solubility, H2S, gravity and light, was excluded, indicating that the observed migration was due to a direct response to salt stress. We propose to term this salinity-driven cyanobacterial migration as ‘halotaxis’, a process that might play a vital role in the survival of cyanobacteria in environments exposed to continuous salinity fluctuations such as intertidal flats. Submarine methane emissions in the Tuscan Archipelago have been studied since the 1960s, both for economic and research purposes. Offshore gas seepage is mainly concentrated southward and westward of Elba island, along N–S faults related to recent extensional activity in the Tuscan shelf and N–S trending positive magnetic anomalies, which have been interpreted as serpentinites associated with ophiolitic rocks due to their very high magnetic susceptibility. This study focuses on the gas chemistry of a new emission site corresponding to a shallow water mud volcano in the Scoglio d'Affrica area. The Scoglio d'Affrica seep has a gas composition typical of mud volcanoes, with methane as the prevalent component (95 vol%) and minor gases which include carbon dioxide, nitrogen and trace amounts of helium. The combined stable C and H isotope composition of CH 4 (δ13C and δ2H) and the enrichment in heavy carbon isotopes of CO 2 , highlight a prevalent secondary microbial origin for these fluids (δ13C~−35.8‰ vs VPDB; δ2H~−166‰ vs VSMOW; δ13C CO2 up to + 21.7‰ vs VPDB). Thus, in spite of the occurrence of positive magnetic anomalies, a possible abiotic origin of methane is excluded. Moreover, the gas from the mud volcano is extremely depleted in 3He and presents typical 3He/4He ratios of a geological setting in which radiogenic crustal helium is strongly predominant. A photo-mosaic of the mud volcano is also reported. A possible connection with other submarine methane emissions in the Tuscan Archipelago is limited to emissions located few kilometers from the Scoglio d'Affrica area. Recent emissions in the area suggest that gases similar in composition from distinct reservoirs, find their way to the surface from Eocene deposits in different time intervals and through different faults and fractures, placed along the Elba-Pianosa ridge. • A photo-mosaic of the shallowest mud volcano in the Tyrrheanian Sea is reported. • Analysis of isotopic and molecular composition of submarine methane emissions. • Carbon dioxide is enriched in heavy carbon isotopes. • Radiogenic crustal helium reveals discontinuities in shallow crustal levels. • Secondary microbial origin is predominant; abiotic origin is discarded.
