ABSTRACT Malva sylvestris is a species used worldwide as an alternative to anti-inflammatory therapies; however, its mechanism of action remains unknown. In this paper, the anti-inflammatory effects of M. sylvestris alcoholic extracts were evaluated by measuring the pro-inflammatory mediators PGE
2 and PGD2 in desferrioxamine-stimulated phorbol 12-myristate 13-acetate-differentiated U937 cells. An HPLC-DAD fingerprint of the M. sylvestris extract was performed and caffeic acid, ferulic acid and scopoletin were identified and quantified. An HPLC-MS/MS method was developed and validated to separate and measure the prostaglandins. The lower limits of detection (~0.5 ng/mL for PGE2 and PGD2 ) and quantification (1.0 ng/mL for PGE2 and PGD2 ) indicated that the method is highly sensitive. The calibration curves showed excellent coefficients of correlation ( r > 0.99) over the range of 1.0-500.0 ng/mL, and at different levels, the accuracy ranged from 96.4 to 106.4% with an RSD < 10.0% for the precision study. This method was successfully applied using U937-d cells. A significant dose-dependent reduction of PGE2 and PGD2 levels occurred using 10 µg/mL (10.74 ± 2.86 and 9.60 ± 6.89%) and 50 µg/mL of extract (48.37 ± 3.24 and 53.06 ± 6.15%), suggesting that the anti-inflammatory mechanisms evoked by M. sylvestris may be related to modulation of these mediators. Background: Alzheimer´s disease, a progressive and degenerative disorder of the brain, is the most common cause of dementia among the elderly. To face its multifactorial nature, the use of single compounds that can simultaneously modulate different targets involved in the neurodegenerative cascade has emerged as an interesting therapeutic approach. Objective: This work investigated the ability of uleine, the major indole alkaloid purified from stem barks of the Brazilian medicinal plant Himatanthus lancifolius, to interact with crucial Alzheimer´s disease disruptive targets associated with two of its major neurodegenerative pathways: acetylcholinesterase and butyrylcholinesterase (cholinergic pathway) and β-secretase and β-amyloid peptide (amyloidogenic pathway). Methods: Uleine’s capacity to inhibit human acetylcholinesterase and butyrylcholinesterase enzymes was determined measuring the difference between reaction rates with and without uleine monitored at 412 nm using 5,5’- dithiobis-(2- nitrobenzoic acid) as colorimetric agent. FRET based assay was used to evaluate β-secretase inhibition using DABCYL- Ser-Glu-Val-Asn-Leu-Asp-Ala-Glu-Phe-EDANS as substrate and β-amyloid peptide spontaneous aggregation assay was performed using the thioflavin T spectroscopy assay. Cell viability and toxicity experiments with PC12 and SH-SY5Y cell lines were performed using the MTT colorimetric assay. Results: Uleine demonstrated strong inhibitory activities for both cholinesterases (IC50 279.0±4.5 and 24.0±1.5 μM, respectively) and β-secretase (IC50 180±22 nM). Above all, uleine significantly inhibited the self-aggregation of amyloid- β peptide and was not toxic for PC12 or SH-SY5Y neuronal cells. Conclusion: These data show for the first time that the natural compound uleine has a novel, multieffective ability to decelerate or even inhibit the development of Alzheimer´s disease.

Defesa: Curitiba, 23/08/2016 Inclui referências : f. 146-158 Área de concentração: Insumos, medicamentos e correlatos Resumo: Tendo em vista a necessidade contínua de novas moléculas com potencial farmacológico e o crescente interesse pelas atividades biológicas de alcaloides quinolínicos, principalmente a leishmanicida, o objetivo deste trabalho foi produzir derivados da fenilquinolina e da pentilquinolina, por meio da ação de microrganismos, células de fungos e bactérias, visando modificações estruturais nessas substâncias. Do processso de biotransformação com a 2-npentilquinolina não houve obtenção de derivados. Das reações de biotransformação da 2-fenilquinolina com os fungos Aspergillus fumigatus e Aspergillus flavus, dois derivados foram obtidos e, respectivamente, denominados DFQ1, com hidroxilações nas posições C5 e C6, uma substância não descrita até o momento, e DFQ2, com uma hidroxilação na posição C4'. Primeiramente, esses derivados foram testados com relação à atividade antileishmania contra as formas promastigotas das linhagens Leishmania amazonensis, L. braziliensis e L. infantum após 24, 48 e 72 h de tratamento. Os resultados demonstraram que a presença de um grupamento hidroxila na posição C4' alterou positivamente a atividade antileishmania para o derivado DFQ2 (CI50 > 100 M), com uma atividade citotóxica pelo menos cinco vezes superior à ação observada para a substância de origem fenilquinolina (CI50 > 500 M). Entretanto, a atividade citotóxica deste derivado foi consideravelmente menor que a observada para o derivado DFQ1 (CI50 < 10 M), demonstrando que a inserção de duas hidroxilas, nas posições C5 e C6, foram capazes de potencializar essa atividade duas mil vezes mais que a molécula original. Através da microscopia eletrônica de transmissão, foi possível visualizar as principais alterações provocadas nas estruturas intracelulares das formas promastigotas de L. amazonensis e L. braziliensis pelos tratamentos com as substâncias DFQ1 e DFQ2. Com relação à atividade anti-inflamatória, embora com intensidade variável, todos os alcaloides testados apresentaram atividade inibitória sobre a quimiotaxia de polimorfonucleares humanos induzida pela caseína, sendo seus efeitos a 5 M, a maior concentração testada, consideravelmente elevados, sendo inclusive superior àquele demonstrado pela dexametasona, usada como fármaco padrão nas condições experimentais propostas, sugerindo uma ação adicional desses alcaloides na supressão do processo inflamatório. Coletivamente, os resultados aqui descritos demonstraram que as modificações na estrutura da 2- fenilquinolina são potencializadoras da ação leishmanicida dessa substância e, também, que os alcaloides dela derivados são capazes de interferir em pelo menos uma das etapas do processo inflamatório. Dessa forma, sugere-se que a ação deste grupo de substâncias no tratamento da leishmaniose afeta diretamente o parasito e pode, também, influenciar o processo inflamatório gerado, o que é um dos agravantes principais da doença. Palavras-chave: biotransformação, 2-fenilquinolina, 2-n-pentilquinolina, citotoxicidade, leishmaniose Abstract: Having in mind the continuous need of new molecules with pharmacological potential and the growing interest in biological activities of quinolinic alkaloids, mainly leishmanicide, the objective of this work was to produce derivatives of phenylquinoline and pentylquinoline potentially more efficient through the action of micro-organisms, fungal cells and bacteria, aiming structural modifications of these substances. The procedures of biotransformation with 2-n-pentylquinoline there was no obtaining derivatives. Through the biotransformation reactions of 2-phenylquinoline with Aspergillus fumigatus and Aspergillus flavus, two derivatives were obtained and respectively called DFQ1 with hydroxylation at C5 and C6 positions, which led to a substance not described to date, and DFQ2 with a hydroxylation at the C4' position. Firstly, these derivatives were tested for anti-leishmania activity against the promastigote forms of L. amazonensis, L. infantum and L. braziliensis lineages after 24, 48 and 72 hours of treatment. The results demonstrated that the insertion of a hydroxyl group at the C4' position positively changed the anti-leishmania activity for the derivative DFQ2 (IC50 > 100 M), with a cytotoxic activity at least five times higher than the activity observed for the original phenylquinoline compound (IC50 > 500 M). However, the cytotoxic activity of this derivative was considerably lower than that observed for the derived DFQ1 (IC50 < 10 M), demonstrating that the insertion of two hydroxyls at C5 and C6 positions was able to potentiate the activity two thousand times more than the original molecule. By transmission electron microscopy, it was possible to view the main changes caused in the intracellular structures of promastigotes of L. amazonensis and L. braziliensis by treatment with DFQ1 and DFQ2 substances. With regard to the antiinflammatory activity, although to varying degrees, all alkaloids and quinolines tested showed inhibitory activity on the chemotaxis of human polymorphonuclear induced by casein, being their effect on 5 M, the highest concentration tested, surprisingly high, and being even higher than those shown by dexamethasone, used as standard drug in the proposed experimental conditions, suggesting an additional action of these alkaloids in the suppression of the inflammatory process. Collectively, the results described herein demonstrate that changes in the 2-phenylquinoline structure are intensifiers of this substance leishmanicide action, and also that its alkaloid derivatives are capable of interfering with at least one of the steps of the inflammatory process. Thus, it is suggested that the action of this group of compounds in the treatment of leishmaniasis directly affects the parasite and can also influence the inflammatory process generated, which is a major worsening of the disease. St.-Hil., Lythraceae, conhecida popularmente como dedal e com a indicação de seu uso para tratamento de câncer, primeiramente, sobre o comportamento das linhagens celulares leucêmicas humanas, U-937, Daudi e Jurkat, em sistemas de cultura in vitro. Os ensaios realizados com essas linhagens demonstraram claramente que o extrato e as frações preparados dessa planta exercem efeitos citostático e citotóxico significativos para as linhagens U-937, Daudi e Jurkat, os quais são independentes da presença do ácido elágico, marcador fitoquímico da espécie. Na avaliação do efeito tóxico pelo método de quantificação da lactato desidrogenase (LDH), o extrato e as frações hidrofóbica (F1) e ActOEt (F2) apresentaram toxicidade significativa a 100 ?g/mL, enquanto que as frações n- BuOH (F3) e aquosa (F4) em 1000 ?g/mL. O extrato de L. pacari foi capaz de inibir a formação de colônias de progenitores leucêmicos de células U-937 em meio semissólido. Citocentrifugados das mesmas linhagens tratadas com 1000 ?g/mL de extrato mostraram características típicas de apoptose, com a presença de vacúolos citoplasmáticos, condensação da cromatina, fragmentação do núcleo e formação de corpos apoptóticos. A exposição de células U-937, assim como outras representativas de câncer de colo do útero (HeLa), de cólon (HRT-18) e de neuroblastoma (SY5Y), além de células mononucleares (MNC) ao extrato de L. pacari levou à alterações características de apoptose no ciclo celular, bem como co-expressão da Anexina V e do 7-AAD. Interessante ressaltar que, em contraste com os resultados descritos para as linhagens tumorais, o extrato de L. pacari pouco interferiu na viabilidade ou influenciou na proliferação de células MNC de medula óssea de camundongos, como também apresentou maior indução de apoptose em células MNC isoladas do sangue periférico de voluntários sadios quando estimuladas com fitohemaglutinina, o que sugere que o efeito indutor de apoptose parece repousar sobre células que estão em atividade metabólica intensa e/ou que estejam em ciclo celular. Embora mais estudos sejam necessários, o conjunto de resultados apresentados revelou alguns aspectos interessantes e ainda não relatados com relação às atividades biológicas dessa espécie brasileira, sugerindo investigações futuras para assegurar seu uso com fins terapêuticos. Abstract: The aim of this work was to evaluate the influence of the methanolic extract and its fractions prepared from the stem barks of Lafoensia pacari A. St.-Hil., Lythraceae, popularly known as "dedal" and used for treating cancer, on the behavior of the human U-937, Daudi, and Jurkat leukemic cell lines, in in vitro culture systems. The assays conducted with these leukemic lineages have clearly shown their cytostatic and cytotoxic effects, which were independent of the presence of ellagic acid, the phytochemical marker of the species. Significant toxicity, determined by the quantification of lactate dehydrogenase (LDH), has been shown for the extract as well as for its hydrophobic (F1) and EtOAc (F2) fractions only at a 100 ?g/mL along with the n-BuOH (F3) and the aquous (F4) fractions at 1000 ?g/mL. The extract was also able to inhibit the formation of colonies by U-937 cells in clonogenic assays. Cytospun preparations of cells treated with 1000 ?g/mL of extract showed typical characteristics of apoptosis, with the presence of cytoplasmic vacuoli, chromatin condensation, nuclear fragmentation, and formation of apoptotic bodies. Exposition of U-937 cells as well as other cell lines representatives of cervical (HeLa), colon (HRT-18), and neuroblastoma (SY5Y) cancers, along with human mononuclear cells to L. pacari extract led to changes characteristics of apoptosis induction, including morphological aspects, alteration of the cell cycle status, and co-expression levels of Annexin V and 7-AAD. Interesting to note that, in contrast with the results described for the tumor cell lines, the extract has little interfered on the viability or influenced the proliferation of mice bone marrow mononuclear cells. Higher induction of apoptosis in peripheral blood mononuclear cells isolated from healthy volunteers was observed when these cells were stimulated to proliferate with phytohemagglutinin, suggesting that the effect of inducing apoptosis seems to rest on cells that are under intense metabolic activity or cycling. Although further studies are needed, the findings herein described revealed some interesting aspects not yet reported with respect to the biological activities of this species, suggesting future investigations to ensure its use for therapeutic purposes.

Para guiar os procedimentos de análise e facilitar a elucidação dos casos periciados faz-se necessário o uso de técnicas de análise mais gerais, conhecidas como técnicas de triagem. Com o objetivo de estudar diferentes métodos de triagem toxicológica, o presente trabalho teve o objetivo de selecionar, avaliar e aplicar técnicas de triagem dentro de uma rotina de análise toxicológica sistemática com escopo forense. Foram selecionadas duas técnicas, a cromatografia líquida de alta eficiência acoplada a detecção por arranjo de diodos e imunoensaio com quimioluminescencia por Biochip, para as quais foram submetidas amostras de sangue total e urina coletadas de cadáveres necropsiados. Ambas as técnicas apresentaram boa sensibilidade para as substâncias elencadas como de interesse, assim como tiveram influência do tipo de matriz biológica analisada. Apesar de bons níveis de concordância, as técnicas estudadas apresentaram resultados divergentes importantes e que demandariam intervenção. Cada técnica apresentou vantagens e desvantagens particulares que devem ser avaliadas caso a caso, de acordo com a demanda e a necessidade do laboratório. Abstract: The best known activity of the Forensic Toxicology is the detection and quantification of chemicals in postmortem biological matrices. To guide the procedures and facilitate the elucidation of juridical cases it is necessary to use more general techniques of analysis, known as screening techniques. With the aim of studying different toxicological techniques of screening, the present study offered to select, evaluate and apply screening techniques within a routine of systematic toxicological analysis in a forensic ambient. It was selected two techniques, high performance liquid chromatography coupled with diode array detection and chemiluminescence immunoassay by using Biochip. They were subjected to real samples of whole blood and urine collected from autopsied corpses. Both techniques showed good sensitivity for substances listed as of interest, and were influenced by the type of biological matrix analyzed. Despite good agreement, the techniques studied showed important divergent results that require intervention. Each technique has particular advantages and disadvantages that should be evaluated case by case, according to the demand and need of the laboratory.

O resveratrol e uma substancia fenolica conhecido por varias atividades farmacologicas, das quais se destacam as antioxidantes e anticancerigenas. Em contraste, possui baixa biodisponibilidade in vivo. Visando maior atividade biologica, mudancas na estrutura quimica do transresveratrol envolvendo biotransformacao com dezenove especies de fungos foram realizadas. Desses, somente o Bipolaris sorokiniana e o Penicillium sp. (64) levaram a resultados positivos em sua biotransformacao. Neste contexto, a biotransformacao do trans-resveratrol pelo B. sorokiniana resultou na sua dimerizacao, levando ao transdehidrodimero do resveratrol, enquanto a reacao com Penicillium sp. (64) levou a formacao de trans-resveratrol-4'-O-metil-5-O-acetil-3-O-?-glucosideo, substancia pela primeira vez descrita. Ao se avaliar o efeito dessas substancias sobre o comportamento de celulas das linhagens McCoy, B16F10 e Hela com o ensaio do MTT, observou-se que o trans-resveratrol-4'-O-metil-5-O-acetil-3-O-?-glucosideo inibiu a proliferacao com perfil semelhante, porem significativamente maior, que o seu precursor, em contraste com o trans-dehidrodimero do resveratrol, o qual promoveu significativo aumento do numero de celulas. Interessante ressaltar que ambos os efeitos, tanto de inibicao quanto de inducao da proliferacao, foram dependentes da dose. Estudos mais aprofundados sao necessarios para definicao do real potencial biologico desses produtos. Abstract: Considering that an organic compound hardly ever resists the catalystic action of the several enzymatic systems present in living organisms, changes in chemical structure of organic molecules can be accomplished through systems involving microorganisms. The structural changes in the ending products may result in considerable enhancement of their pharmacological potential. Contrasting with its low bioavailability in vivo, resveratrol is a phenolic compound widely known by its different pharmacological activities, especially as an antioxidant and anticancer agent. With the aim of achieving higher biological activity, changes in the chemical structure of the trans-resveratrol involving biocatalysis with nineteen species of fungi have been performed. Of these, only Bipolaris sorokiniana and Penicillium sp. (64) led to positive results in which the biotransformation of trans-resveratrol by B. sorokiniana resulted in its dimerization, leading to the formation of trans-resveratrol dehydrodimer, while the reaction with Penicillium sp. (64) resulted in the formation of trans-resveratrol-4'-Omethyl- 5-O-acetyl-3-O-?-glucoside, a compound never reported. To evaluate whether these compounds were biologically active, their influence on the behavior of McCoy, B16F10, and Hela cell lines were studied using the MTT assay. The results showed that the trans-resveratrol-4'-O-methyl-5-O-acetyl-3-O-?-glucoside derivative inhibited the proliferation of all cell lines tested with similar profile, but significantly higher, than its precursor. In contrast, the trans-dehydrodymer led to a significant increase in the number of cells. Worthy of note is the fact that, although having opposite effects, both were dose-dependent. Further studies are required for defining the real biological potential of these products.

Quando em excesso, a melanina produz eritema, sardas e hiperpigmentação. Entre as enzimas envolvidas na melanogênese, a tirosinase é a mais importante. O gênero Passiflora possui como substâncias majoritárias os flavonoides C-glicosilados, compostos polifenólicos. Os polifenóis representam o maior grupo de inibidores da tirosinase. Assim, o objetivo deste trabalho foi caracterizar os efeitos dos extratos preparados de folhas de P. alata, P. edulis, P. actinia e P. incarnata sobre a atividade de tirosinase purificada de Agaricus bisporus e, também, daquela obtida de células B16F10. Usando o ensaio do MTT como teste de citotoxicidade, estabeleceu-se que a maior concentração não tóxica dos extratos foi de 100 ?g/mL, a qual foi utilizada para o estudo da atividade de tirosinase in vitro. A mutação é herdada de um dos pais, mas confere baixa penetrância (aproximadamente 3 a 4%) aos seus portadores, o que sugere que outros fatores são necessários para o desenvolvimento do tumor. Um dos eventos genéticos mais comumente encontrados em TCA, associado à mutação R337H, é a perda de heterozigose com retenção do alelo mutado. Este é um dos principais mecanismos de inativação de genes supressores de tumor, sendo demonstrado em vários tipos de cânceres. A proteína p53 mutante é altamente instável em condições de pH e temperatura elevados e desencadeia um desequilíbrio genômico, resultando em perda da sua função e no acúmulo de alterações no DNA. A perda de imprinting genômico na região 11p15 é o principal fator que leva a perda de controle da expressão de IGF2 e de outros genes situados nesta região. O presente estudo foi realizado com o objetivo de identificar outras alterações de risco para o desenvolvimento do TCA em crianças portadoras da mutação R337H. Para isso, foram analisados 13 tumores de córtex adrenal (4 adenomas e 9 carcinomas). A idade em que estes tumores foram diagnosticados variou de 1 mês a 9 anos e a proporção entre meninas e meninos foi de 3,3:1,0. As amostras foram analisadas por Hibridização Genômica Comparativa em Microarranjos (aCGH ou SNP-CGH). A alta resolução desta técnica permite a identificação de alterações no número de cópias de DNA em regiões genômicas muito menores do que a permitida pela CGH convencional. Encontramos várias alterações, sendo os ganhos de DNA mais freqüentes que as perdas. A perda do alelo materno de IGF2 em 92% (12/13) das amostras parece ser o principal mecanismo de perda de imprinting que ocorre em 11p15. Um SNP foi encontrado em 6p21 em 92% das amostras, sugerindo uma perda de função do gene HLA-DRB5 que poderia ser responsável pela diminuição na apresentação de antígeno pelo TCA. As alterações encontradas no cromossomo 9 foram bastante frequentes, abrangendo quase toda sua extensão e foram marcadas predominantemente por ganhos genômicos. Estes ganhos em 9q33 confirmam achados anteriores, mas não permitem afirmar conclusivamente sobre a participação de outro gene ou sobre alguma informação nova nos mecanismos relacionados à formação de tumor no córtex adrenal. Abstract: It has been documented in Parana state (southern Brazil) that the mutation R337H in the TP53 gene is the main cause of childhood adrenocortical tumor (ACT). The mutation is inherited from a parent, but it causes low risk (approximately 3-4%) to their carriers, which suggests that other factors are required for tumor development. One of the genetic events most commonly found in ACT associated with the R337H mutation is the loss of heterozygosis with retention of the mutant allele in the tumor. This is one of the main mechanisms of inactivation of tumor suppressor genes, and it has been demonstrated in several types of cancers. The mutant p53 protein is highly unstable under conditions of high pH and temperature and triggers a genomic imbalance, resulting in loss of function, and hence the accumulation of DNA alterations. Loss of genetic imprinting in 11p15 is the main factor that leads to alteration in the expression of IGF2 and other genes located in this region. This study was conducted to identify other genomic changes in children with the germline mutation R337H that may contribute to ACT development. We have analyzed 13 childhood ACT (4 adenomas and 9 carcinomas). The age at which these tumors were diagnosed ranged from 1 month to 9 years and the ratio between girls and boys was 3.3:1.0. The samples were analyzed by comparative genomic hybridization on microarrays (aCGH or SNP-CGH). The high resolution of this technique allows the identification of changes in the DNA in genomic regions much smaller than allowed by conventional CGH. There are a number changes, with gains of DNA more frequently found than losses. We have found loss of the maternal allele of IGF2 in 92% (12/13) of the samples that seems to be the main mechanism of loss of imprinting that occurs in 11p15. One SNP was found in 6p21 in 92% of the samples, suggesting a possible loss of function of the gene HLA-DRB5 that could account for decreased presentation of antigen by the ACT. The abnormalities found in chromosome 9 were quite frequent and extensive, covering almost all parts of this chromosome and were predominantly gain of chromosome. These findings in 9q33 confirm earlier findings, but do not allow any new conclusion about the participation of other genes or about any new information in the mechanism related to tumorigenesis in adrenal cortex.

Defesa: Curitiba, 31/03/2009 Bibliografia : fls. 66-68 Resumo: O vírus de Dengue necessita de diversas proteínas da célula hospedeira humana para que possa completar o ciclo de infecção viral. Nas diversas etapas do ciclo, ocorrem interações com diferentes tipos de proteínas da célula. O presente trabalho, utilizando-se de ferramentas de biologia molecular e de bioinformática, teve como objetivo identificar e analisar, pela construção de um interatoma, as interações entre as proteínas do vírus da Dengue e as da célula hospedeira humana, a fim de identificar aquelas essenciais ao ciclo viral e ao desenvolvimento do quadro fisiopatológico da doença. Para tanto, realizou-se o cruzamento entre as proteínas estruturais Capsídeo, pré-Membrana e Envelope do vírus da Dengue, clonadas no vetor pBT, com as bibliotecas de cDNA construídas de células de cérebro e fígado humanos. Os dados obtidos por este cruzamento, assim como as respectivas seqüências, foram analisados e utilizados para alimentar o programa Cytoscape, no qual uma rede foi gerada. Análise desta rede permitiu a identificação de proteínas com funções relacionadas à infecção viral da célula, como a Clusterina, envolvida na ativação do sistema complemento e na resposta apoptótica, e das moléculas COX-1, -2 e -3, envolvidas na resposta inflamatória. Subseqüentemente, os dados obtidos no duplo-híbrido foram cruzados com dados de expressão gênica obtidos de células infectadas com o vírus da Dengue no programa Matisse e uma segunda rede foi gerada, possibilitando a identificação de módulos funcionais, dentre os quais o da apoptose, de interesse do presente trabalho, visto não ser interessante para o vírus promover apoptose da célula que o abriga, ao menos nos momentos iniciais da infecção. A seqüência das proteínas estruturais do vírus foi analisada, o que possibilitou a identificação de um domínio anti-apoptótico Bcl-2 na seqüência da proteína de Membrana. Este domínio também foi encontrado na seqüência protéica de outros vírus que apresentam função anti-apoptótica conhecida. A abordagem diferenciada adotada neste trabalho possibilitou não só observar e elaborar hipóteses sobre o modo pelo qual o vírus atua na célula hospedeira após ser infectada, como também abordar, de forma global, aspectos envolvidos nos processos celulares da interação entre o vírus da Dengue e as células humanas, os quais deverão ser biologicamente validados, a fim de comprovar ou refutar sua essencialidade. Abstract: The Dengue virus uses several of the human host cell proteins to complete the cycle of viral infection. Therefore, during the progressive stages of this cycle, many interactions with different types of cellular proteins take place. The aim of this work, which involved bioinformatics, was to identify and analyse some essential interactions for the virus replication cycle and for the development of the host disease established between the proteins from the Dengue virus and from the human host cells, resulting in a protein interactome. To achieve this, a screening of the in vitro binding between the Dengue virus Capsid, Premembrane and Envelope structural proteins, which were cloned in the pBT vector, and human brain and liver cDNA libraries was performed. The binding data obtained were analysed and the respective protein sequences involved determined. Both results were then used to feed the Cytoscape software in which a network of interacting proteins was generated. Analyses of this network identified proteins with functions related to cellular viral infection, such as Clusterin, involved in the activation of the complement system and cellular apoptosis, and Cox-1, -2 and 3, all involved in the inflammatory response. Subsequently, using the Matisse software, data obtained with the double-hybrid system were matched up against gene expression results obtained from cells infected with the Dengue virus. The network generated allowed the identification of some cellular functional modules, such as the apoptosis one, which is of particular interest in this work as interference in this cellular process is expected given the fact that, at least initially, it is not advantageous for the virus to kill the host cell. Analyses of the structural proteins sequences revealed the presence of a Bcl-2 anti-apoptotic domain in the Membrane viral protein, which has been reported for some other viruses with already known anti-apoptotic activity. The innovative approach used in this study not only allowed the formulation of questions and hypothesis about the way the virus behaves inside cells after infection, but also to speculate, in a more global view, about some of the cellular aspects involved in the relationship between the proteins of the Dengue virus and host cell interaction, which must be biologically validated in the future in order to confirm or refute their essentiality.

- Abstract
The purpose of the present work is to report the effects shown by the uleine-rich fraction extracted from the barks of Himatanthus lancifolius (Muell. Arg.) Woodson (Apocynaceae) on human peripheral blood mononuclear cells in which we were able to inhibit the proliferation of phytohemagglutinin-stimulated lymphocytes by blocking their transformation into blast-dividing cells. Furthermore, it inhibited the proliferation of Daudi and Reh cells, two leukemic cell lines of lymphoid origin. The present study widens the biological applications of H. lancifolius' alkaloids to include its possible use as a modulator of the immune system., (Georg Thieme Verlag KG Stuttgart. New York.)

- Abstract
The aim of this work was to investigate the anti-inflammatory activities of the uleine-rich fraction extracted from the barks of Himatanthus lancifolius (Muell. Arg.) Woodson (Apocynaceae). To achieve this, we focused on its in vitro effects on some steps of the inflammatory response using peripheral human leukocytes. The results presented herein show that the uleine-rich fraction significantly inhibits the migration of casein-induced granulocytes and their adhesion to fibronectin and vitronectin, along with mononuclear cells, by down-regulating the expression of alpha 4beta1 and alpha5beta1 integrins. The data suggest that H. LANCIFOLIUS has the potential of interferring with leukocyte trafficking through its uleine-rich fraction, emphasizing its usefulness in inflammatory conditions.
