We examined the apoptotic response of two glioblastoma cells, p53 wild type U87 and p53 mutated T98G, to doxorubicin, bortezomib, and vorinostat, which respectively target DNA, 26S proteasome and histone deacetylase, to clarify p53ʹs function in apoptosis. We demonstrated that doxorubicin induced apoptosis in U87 cells but not in T98G cells. The level of p53 was definitively correlated to the extent of DNA damage and apoptosis initiation. Dominant-negative p53 reduced p21 expression, but did not affect doxorubicin-induced apoptosis, so the transcriptional activity of p53 seemed not to participate in doxorubicin-induced apoptosis. However, p53 concentrated into the nucleus during heavy apoptosis. Bortezomib could induce apoptosis in U87 with high sensitivity and T98G cells with low sensitivity. In contrast, vorinostat promoted apoptosis in both U87 and T98G cells and reduced the basal level of p53 in U87 cells, indicating that p53 played no role in the vorinostat-induced apoptosis. To clearly define the role of p53 in bortezomib- and doxorubicin-induced apoptosis, we combined doxorubicin with bortezomib to treat U87 cells to assess this combination’s effect on apoptosis and p53 status. Interestingly, the combination of doxorubicin with bortezomib engendered compound stress, resulting in a synergistic outcome for apoptosis in U87 cells. However, the amounts of p53 in the total count and in the nucleus were much lower with the combination than with doxorubicin alone, suggesting that p53 played no role in either the compound stress, doxorubicin-only or bortezomib-induced apoptosis.

As a necessary tool for understanding, prediction, and design (especially on a microscopic scale), Quantum chemical (QC) methods have a profound impact on the field of energetic materials (EMs). This study focuses upon the QC methods applicable to energetic molecules and their related applications. They generally include the Hartree-Fock method, semi-empirical QC methods, density functional theory (DFT), and high-accuracy ab initio methods. This study includes a detailed discussion about the application scope and accuracy of the descriptions of the geometric structure, electronic structure, thermodynamic property, and reactivity of energetic molecules. Additionally, this study stresses machine learning combined with DFT calculations that becomes increasingly popular as an important way to establish models for accurate property predictions. This work is expected to be instructive and constructive for the use of QC methods in EM study.

In this study, to develop new advanced corrosion inhibitors with high corrosion inhibition efficiency at low concentration, a simple but effective strategy that is the adjustment of bridge heteroatoms in a mixed corrosion inhibitor named 2-(n-hexylamino)-4-(3-N,N-dimethylaminopropyl)amino-6-(2-aminobenzimidazol)-1,3,5-s-triazine (BDCT) was used to improve the anti-corrosive properties. Two series of new compounds were designed by adjusting three bridge N atoms in BDCT by different number of O and S atoms at three different positions. The molecular structures and different kinds of anti-corrosive properties of 14 designed compounds were investigated by the DFT and MD methods. The results showed that when compared to BDCT, the molecular area, electrophilicity, electronegativity, strength of feed-back bonding, softness, chemical reactivity and binding energy on the Fe(110) surface were improved obviously by the introduced bridge O and S atoms, leading to higher corrosion inhibition efficiency. This effect could be further enhanced with the increased number of them, and S atoms were found to better than O atoms obviously. The introduction positions of O and S atoms also made effects on anti-corrosive properties, introducing them at the R1 position was more helpful for improving anti-corrosive properties than R2 than R3. As a result, the compound with three bridge S atoms (B7) has the best anti-corrosive properties. The adsorption sites, type of bonding with metals and corrosion inhibition mechanism of BDCT were also changed completely by O and S atoms.

Background and aims Three to 10 minutes of smoking cessation advice by physicians is effective to increase quit rates, but is not routinely practised. We examined the effectiveness of physicians' very brief (approximately 30 sec) smoking cessation intervention on quit rates among Chinese outpatient smokers. Design A pragmatic, open-label, individually randomized controlled trial. Setting Seventy-two medical outpatient departments of hospitals and/or community health centers in Guangdong, China. Participants Chinese adults who were daily cigarette smokers (n = 13 671, 99% males) were invited by their physician to participate during outpatient consultation. Smokers who were receiving smoking cessation treatment or were judged to need specialist treatment for cessation were excluded. Interventions The intervention group (n = 7015) received a 30-sec intervention including physician's very brief advice, a leaflet with graphic warnings and a card with contact information of available cessation services. The control group (n = 6656) received a very brief intervention on consuming vegetables and fruit. A total of 3466 participants in the intervention group were further randomized to receive a brief booster advice from trained study personnel via telephone 1 month following their doctor visit. Measurements The primary outcome was self-reported 7-day point prevalence abstinence (PPA) in the intervention and control groups at the 12-month follow-up. Secondary outcomes included self-reported 30-day abstinence and biochemically validated abstinence at 12-month follow-up. Findings By intention-to-treat, the intervention (versus control) group had greater self-reported 7-day abstinence [9.1 versus 7.8%, odds ratio (OR) = 1.14, 95% confidence interval (CI) = 1.03-1.26, P = 0.008] and 30-day abstinence (8.0 versus 6.9%, OR = 1.14, 95% CI = 1.03-1.27, P = 0.01) at 12-month follow-up. The effect size increased when only participants who received the intervention from compliant physicians were included (7-day PPA, OR = 1.42, 95% CI = 1.11-1.74). The group difference in biochemically validated abstinence was small (0.8 versus 0.8%, OR = 1.00, 95% CI = 0.71-1.42, P = 0.99). Conclusion A 30-sec smoking cessation intervention increased self-reported abstinence among mainly male smokers in China at 12-month follow-up (risk difference = 1.3%), and should be feasible to provide in most settings and delivered by all health-care professionals.

Due to the large number of operations, surgeons sometimes need to work overtime or even stay up late to perform pancreaticoduodenectomy. Fatigue and sleep deprivation can result in an increased error rate at work. There have been numerous studies about the effect of overtime surgery on the prognosis of patients. However, the effect of overtime work for pancreaticoduodenectomy on the prognosis of patients is unclear. This study explores the impact of overtime work for pancreaticoduodenectomy on the prognosis of patients.To explore the impact of overtime work for pancreaticoduodenectomy on the short-term prognosis of patients.This was a single-center, retrospective cohort study. The patients who underwent pancreaticoduodenectomy between January 2017 and December 2019 were included. Patients were stratified by operative start time into the control group (surgery that started between 8:00 and 16:49) and the overtime group (surgery that started between 17:00 and 22:00) and compared intraoperative and postoperative parameters. The following parameters were compared between the overtime group and the control group: Operative time, blood loss, number of lymph nodes removed, duration of treatment in the Intensive Care Unit (ICU), and incidence of complications.From January 2017 to December 2019, a total of 239 patients underwent pancreaticoduodenectomy in the Department of Hepatobiliary Surgery of our institution. Four patients were excluded from this study due to lack of clinical data. A total of 235 patients were included, with 177 in the control group and 58 in the overtime group. There was no difference between the two groups in operative time, blood loss, number of lymph nodes removed, ICU length of stay, hospital length of stay, mortality during hospitalization. Compared with the control group, the overtime group had a higher incidence of pancreatic fistula (32.8%Overtime work for pancreaticoduodenectomy increases the incidence of pancreatic fistula. The effect of overtime surgery on the long-term prognosis of patients' needs to be further studied.

Based on the parent tetrazole 2N-oxide, six series of novel carbon-linked ditetrazole 2N-oxides with different energetic substituent groups (-NH2, -N3, -NO2, NF2, -NHNO2) and energetic bridge groups (-CH2-, -CH2−CH2-, -NH-, -N=N-, -NH−NH-) were designed. The overall performance and the effects of different energetic substituent groups and energetic bridge groups on the performance were investigated by density functional theory and electrostatic potential methods. The results showed that most of designed compounds have oxygen balance around zero, high heats of formation, high density, high energy, and acceptable sensitivity, indicating that tetrazole N-oxide is a useful parent energetic compound employed for obtaining high energy compounds, even only combined with some very common energetic substituent groups and bridge groups. In prostate cancer, p53 maximizes apoptosis in response to severe DNA damage, not DNA replication stress. Here, we examined the apoptotic response of two glioblastoma cells, p53-wild type U87 and a p53-mutated T98G cell, for the same stresses. We ascertained that p53 intensified apoptosis in response to severe DNA damage, not DNA replication stress in glioblastoma. We further asked if p53-mediated apoptosis can be induced by cellular stress other than severe DNA damage. We analyzed two compounds, bortezomib and vorinostat, respective inhibitors of 26S proteasome and histone deacetylase, to evaluate their capacity to activate p53-mediated apoptosis. The cellular stress incited by bortezomib, not vorinostat, activated p53-mediated apoptosis. Next, we asked if the cellular stress generated by combining the two compounds had a synergistic effect on apoptosis. Our results demonstrated that doxorubicin with bortezomib or CFS-1686, or bortezomib with vorinostat have a significant synergistic effect on apoptosis only in p53-wild type cell. Under high stress, p53 translocates from cytosol into the nucleus to cause apoptosis possibly. Together, p53 maximizes apoptosis for cellular stress caused by severe DNA damage, disruption of protein turnover, and for the stress induced by drug combination including doxorubicin with bortezomib or CFS-1686, and bortezomib with vorinostat.

Results Of patients with Bismuth and Corlette (B & C) type Ⅲ (n=19) and Ⅳ (n=25) HC lesions, 33 (55.9%) were treated with hilar resection combined with major liver resection (MLR), while the other 11 patients with type Ⅲ and Ⅳ, and those with type Ⅰ (n=8) and Ⅱ (n=7) HC lesions were treated with hilar resection. The overall surgical mortality was 5.1% and surgical morbidity was 35.6%. There was no statistical difference in the mortality between MLR group and hilar resection group (6.1% vs. 3.8%; X2=0.703, P=0.145). The median follow-up period was 18 months (range, 1-94 months). The 1-, 3-, 5-year survival rate was 59.3%, 36.5%, and 17.7%, respectively. The overall survival after resections was 18 months. In HC patients with B & C type Ⅲ and Ⅳ lesions, the median survival was 23 months for hilar resection with MLR and 8 months for hilar resection alone; the 1-, 3-, 5-year cumulative survival rate was 63.9%, 23.3%, and 15.5%, respectively for hilar resection with MLR, and 11.1%, 0, and 0, respectively for hilar resection alone, with significant differene observed (HR, 9.902; 95% CI, 2.636-19.571, P=0.001). This study focuses on the test performance discrepancies for filter cartridges resulting from different kinds of test dust. A comprehensive experimental set-up including pulse-jet cleaning system for cleanable pleated filter cartridges was developed. Then, three identical cartridge samples were tested in this experimental set-up using different synthetic test dust, i.e., 325 mesh talcum powder, ASHRAE dust and ISO A2 dust respectively. Results showed that the type of test dust had significant impacts on dust holding capacity and cleaning effectiveness of pulse-jet cleaned cartridges. Cartridge sample loaded with ASHRAE dust has the smallest dust holding capacity and the worst cleaning effectiveness. However, considering its short experimental period, size distribution and actual simulation effects of atmospheric aerosol, ASHRAE dust is still recommended.

From numerous approaches studying the prediction of stock price, this paper proposed a new approach which was the combination of RBF neural network and Markov chain to forecast the stock closing price of the Shanghai composite index. Markov chain was aimed at making the error between the actual price and predicted price obtained by RBF neural network correct. Besides, for higher prediction accuracy, genetic algorithm was used to optimize the state division of Markov chain. The experimental result confirmed its effectiveness and superiority in comparison with the other two methods in some time interval.

The study endpoint was the proportion of patientswithsuccessfulprophylaxisat4 weeksaftertreatment.Theprimaryanalysiswasbasedontheevaluablepopulation(PP population) and the pre-specified non-inferiority margin was set at 215%. ClinicalTrials.gov: NCT01254344.Results: Of 599 patients randomized, 499 (251 ertapenem and 248 ceftriaxone) were eligible for inclusion inthe PP population. The proportions of patients with successful prophylaxis in the ertapenem and ceftriaxonegroups were 90.4% (227/251) and 90.3% (224/248), respectively. The difference in the proportion of successfuloutcomes was 0.1% (95% CI 25.2%, 5.5%). Unexplained antibioticusewasthe mostfrequentreasonfor prophy-laxis failure in both groups [ertapenem 4.8% (12/251), ceftriaxone 4.4% (11/248); difference 0.3%; 95% CI 23.6,4.3]. Pathogen species isolated from SSI sources were consistent with previously conducted studies and theproduct package insert. The incidence of adverse events (AEs) was similar between the groups, with the mostcommon AE being pyrexia [ertapenem 7.6% (22/290), ceftriaxone 5.7% (17/297)].Conclusions: Ertapenem is as effective as ceftriaxone/metronidazole for SSI prophylaxis in patients undergoingelective colorectal surgery, and is well tolerated.Keywords: b-lactams, ceftriaxone, metronidazole

In this paper, the power spectral density method based on ANSYS is carried out to study the intake tower finite element model under an earthquake. A power spectral density function is obtained and disposed to be smooth. Then it is used to study the intake tower when the seismic wave occurs in the direction of flow and perpendicular to the direction of flow .This study provides theoretical basis for reinforce and transform of the intake tower structure.

In this paper, we depart from the basic vibration equation deduced from Vibro sand tower structure recurrence formula. Using ANSYS finite element software to simulate the red sand of the top ten tower structural vibration modal. Of which the first two modal solutions in accordance with the conventional prismatic cantilever closer to theoretical calculations, the results of the finite element solution proved authentic, we provide a theoretical basis for the use of finite element methods for solving the structural problems of sand washing tower.

Instrument analysis method is used in this experiment, which detecting the zirconium content of zirconium powder by using plasma-atomic emission spectrometer. In this experiment, the instrumental detective conditions were determined, and analyzed several main factors affecting the experimental results, concluded the effects of temperature, hydrofluoric acid and deionized water on dissolving zirconium powder, summarized the method to detect zirconium powder content directly by ICP.

Based on the finite element method, three models are built: classical solid model, potential flow model and a combination of fluid-structure coupling model. Based on the above model the damping mechanism of tuned liquid damper has been investigated. Study focused on classical solid model and the boundary constrained potential fluid model respectively. After the completion of the above calculation, the analysis of bidirectional fluid-structure interaction model being carried out with the implicit dynamics algorithm. The results shows the contrast of the dynamic response of structure in the hydrated and anhydrous state. The calculations show that: the additional water mass can effectively reduce the natural vibration frequency of the structure, and it also can effectively reduce the displacement and velocity response of structure under vibration loads.

Concentration distribution of Nd2O3 in 3KA rare earth cell was studied by a self-restraint sample instrumentation of high temperature molten salts. The results were shown that concentration of Nd2O3 in this system was less than 4%, and the concentration of Nd2O3 decreased from top to bottom on the portrait, but it would be increased a little bit in the bottom point, and the concentration of Nd2O3 would be gradually increasing from cathode to anode.

This paper introduces an intrusion detection system which uses IPV6 principle to work out a new intrusion mode so that the intrusion behaviors could be identified. This system adopts the intrusion features to analyze the consistency and deviations which is different from the common methodology which utilizes the historic data for this purposes. A Snort system is proposed to test the Network Intrusion Detection System (NIDS) that is based on IPV6.

The tidal motion of the ocean water through the ambient magnetic field, generates secondary electric and magnetic field. The magnetic fields generated by the diurnal (O1) ocean flow can be clearly detected. We simulate the magnetic signals for tidal constituents –diurnal (O1) tides. The idea of exploiting tidal signals for EM studies of the Earth is not new, but so far it was used only for interpretation of inland and transoceanic magnetic field data due to O1. Emphasis in this work is made on a discussion of sea bottom electric field of the same origin.

This paper deals with some issues during the synthesis of DF signals such as change of phase and sine amplitude, smooth output and wave filter which features dynamics and variability. Utilizing AD9833 chip, this paper design the circuit that is used for synthesizing DF signals, including external circuit, wave filter circuit and amplify circuit. Given the characteristics of this chip, this paper provides the procedure design methodology that includes AD9833 initiative, write control and synthesis frequency. The testing experiments indicate the accurateness and reliability of this design.

For the shortcomings that computation speed of the DDS decreases with iterations increasing in CORDIC algorithm., the traditional algorithm of multiple iterations is displaced by a point of decompose predict the direction of rotation and multi-level iterative parallel computing method in this paper. The function simulation results show that the improved algorithm enhance the computation speed and maintain data high precision. This design has high computing speed, high precision and simple hardware implementation etc.

Solar radiation between 100 and 400 nm has a number of effects on human health. The UV Index and exposure time are particularly important in the evaluation of the risk of sunburn, skin cancer and other injury. This paper used the UVM-30 photodetector to measure the UVI and the max exposure time in sun real time with a simple linear function relationship circuit. The results suggest you should bath in sun not more than 36 minutes at noon, or not more than 106 minutes at 17:00 in the afternoon with 5 UVI.

This paper discusses how to address some issues when contemplating the global optimal transportation path (GOTP) such as dynamics, the ability of real-time analysis as well as complexity of prediction. Using shortest path methodology, this paper abstracts the real-life problem to a graphic context. Based on the solution of ant colony optimization (ACO) algorithm, the simulation indicates that this manner is efficient and effective in dealing with these problems. The indicators utilized ACO are achieved through simulation results analysis, providing the range of exact elements.

A Macpherson suspension model of a car was built based on ADAMS/ CAR. The kinematics discipline of the suspension was shown based on the general theory. Through the comparison of the simulation results of the two kinds of suspension, the L type Macpherson suspension in the wheel alignment parameter, steer angle and anti-lift/dive performance aspects is improved.

The development of Electric Vehicle is not only a replacement of power source, but a new process of reasonable design and development on the mechanical structure and power system. Considering the strained condition of the support structure when the Electric Vehicle is under the four typical working conditions: accelerating, braking, steering and vertical impact, the dynamic design method and finite element theory are adapted to analysis its strength and mode, while the orthogonal test method is used to analyze the influencing factors of its modes. Combining the size optimization, the weight of the power system’s support structure has been reduced. This method, which has been adapted in the independent development of an Electric Vehicle，will provide a useful reference for the dynamical and lightweight design of similar mechanical structure.
