Background: Face image animation generates a synthetic human face video that harmoniously integrates the identity derived from the source image and facial motion obtained from the driving video. This technology could be beneficial in multiple medical fields, such as diagnosis and privacy protection. Previous studies on face animation often relied on a single source image to generate an output video. With a significant pose difference between the source image and the driving frame, the quality of the generated video is likely to be suboptimal because the source image may not provide sufficient features for the warped feature map. Methods: In this study, we propose a novel face-animation scheme based on multiple sources and perspective alignment to address these issues. We first introduce a multiple-source sampling and selection module to screen the optimal source image set from the provided driving video. We then propose an inter-frame interpolation and alignment module to further eliminate the misalignment between the selected source image and the driving frame. Conclusions: The proposed method exhibits superior performance in terms of objective metrics and visual quality in large-angle animation scenes compared to other state-of-the-art face animation methods. It indicates the effectiveness of the proposed method in addressing the distortion issues in large-angle animation.

Abstract In this paper, we explored the influence of lipase on cuticle wax and anthocyanin metabolism in blueberries. The results demonstrated a progressive alteration in wax crystal structure, transitions from rod‐like to short rod‐like and flaky, and even disappearance of the cuticle wax. Gas chromatography–mass spectrometry analysis revealed a reduction in esters, aldehydes, long‐chain alkanes, and total wax content following lipase treatment of blueberry fruits. Additionally, ultra high performance liquid chromatography–mass spectrometry analysis showed significant decreases in peonidin, petunidin, cyanidin, and delphinidin, accompanied by an increase in malvidin content due to lipase treatment. Subsequent investigations uncovered that lipase treatment inhibited the activities of key anthocyanin synthetic enzymes, namely phenylalanine ammonia‐lyase (PAL), chalcone isomerase (CHI), and dihydroflavonol reductase (DFR). Moreover, lipase treatment downregulated the expression of synthesis‐related genes, including PAL, 4‐coumarate: CoA ligase, chalcone synthase (CHS), CHI, and DFR. In summary, our findings indicate that lipase disrupts the integrity of cuticle wax structure, inhibits anthocyanin synthesis, and reduces anthocyanin content. Accordingly, our studies suggest that inhibiting lipase could offer a novel approach to preserving the freshness and anthocyanin concentration of blueberries.

Abstract Rheumatoid arthritis (RA) is a chronic autoimmune disease with an unknown etiology. RA cannot be fully cured and requires lengthy treatment, imposing a significant burden on both individuals and society. Due to the lack of specific drugs available for treating RA, exploring a key new therapeutic target for RA is currently an important task. Activated fibroblast-like synoviocytes (FLSs) play a crucial role in the progression of RA, which release interleukin (IL)-1β, IL-6 and tumor necrosis factor (TNF)-α resulting in abnormal inflammatory reaction in the synovium. A previous study has highlighted the correlation of m6A reader insulin-like growth factor 2 mRNA-binding protein 2 (IGF2BP2) with inflammation-related diseases in human. However, the role of IGF2BP2 in the inflammatory reaction of FLSs during RA progression has not been assessed. In this study, IGF2BP2 expression was decreased in the synovial tissues of RA patients and collagen-induced arthritis (CIA) rats. Intra-articular injection of an adeno-associated virus (AAV) vector overexpressing IGF2BP2 relieved paw swelling, synovial hyperplasia and cartilage destruction in CIA rats. IGF2BP2 overexpression also inhibited lipopolysaccharide (LPS)-mediated RA fibroblast-like synoviocytes (RA-FLSs) migration and invasion accompanied by a decreased level of inflammatory factors in vitro. Conversely, IGF2BP2 suppression promoted RA-FLSs migration and invasion with an elevated level of inflammatory factors in vitro. The sequencing result showed that glutathione S-transferase Mu 5 (GSTM5), a key antioxidant gene, was the target mRNA of IGF2BP2. Further experiments demonstrated that IGF2BP2 strengthened the stability of GSTM5 mRNA, leading to weakened inflammatory reaction and reduced expression of matrix metalloproteinase 9 and 13 (MMP9, MMP13). Therefore, IGF2BP2-GSTM5 axis may represent a potential therapeutic target for RA treatment.

Abstract Background Recent researches have found a strong correlation between the triglyceride-glucose (TyG) index or the atherogenic index of plasma (AIP) and cardiovascular disease (CVD) risk. However, there is a lack of research on non-invasive and rapid prediction of cardiovascular risk. We aimed to develop and validate a machine-learning model for predicting cardiovascular risk based on variables encompassing clinical questionnaires and oculomics. Methods We collected data from the Korean National Health and Nutrition Examination Survey (KNHANES). The training dataset (80% from the year 2008 to 2011 KNHANES) was used for machine learning model development, with internal validation using the remaining 20%. An external validation dataset from the year 2012 assessed the model’s predictive capacity for TyG-index or AIP in new cases. We included 32122 participants in the final dataset. Machine learning models used 25 algorithms were trained on oculomics measurements and clinical questionnaires to predict the range of TyG-index and AIP. The area under the receiver operating characteristic curve (AUC), accuracy, precision, recall, and F1 score were used to evaluate the performance of our machine learning models. Results Based on large-scale cohort studies, we determined TyG-index cut-off points at 8.0, 8.75 (upper one-third values), 8.93 (upper one-fourth values), and AIP cut-offs at 0.318, 0.34. Values surpassing these thresholds indicated elevated cardiovascular risk. The best-performing algorithm revealed TyG-index cut-offs at 8.0, 8.75, and 8.93 with internal validation AUCs of 0.812, 0.873, and 0.911, respectively. External validation AUCs were 0.809, 0.863, and 0.901. For AIP at 0.34, internal and external validation achieved similar AUCs of 0.849 and 0.842. Slightly lower performance was seen for the 0.318 cut-off, with AUCs of 0.844 and 0.836. Significant gender-based variations were noted for TyG-index at 8 (male AUC=0.832, female AUC=0.790) and 8.75 (male AUC=0.874, female AUC=0.862) and AIP at 0.318 (male AUC=0.853, female AUC=0.825) and 0.34 (male AUC=0.858, female AUC=0.831). Gender similarity in AUC (male AUC=0.907 versus female AUC=0.906) was observed only when the TyG-index cut-off point equals 8.93. Conclusion We have established a simple and effective non-invasive machine learning model that has good clinical value for predicting cardiovascular risk in the general population.

Abstract During postharvest storage, the metabolism of cell wall components greatly affects the quality of fruits and vegetables. Utilizing 1‐methylcyclopropene (1‐MCP) as an ethylene receptor inhibitor and modified atmosphere (MA) separately or in combination (1‐MCP+MA) were investigated for their impact on Zizania latifolia quality during postharvest storage. The treatments effectively controlled firmness increase, color degradation, and reductions in soluble protein, reducing sugar, total phenol, and ascorbic acid content, with the combined 1‐MCP+MA showing the most significant effect. These treatments also moderately changed soluble pectin, protopectin, cellulose, and hemicellulose content, preserving the activities of essential enzymes and the expression of enzyme genes. The combined treatment particularly demonstrated superior preservation capabilities, holding promise for reducing postharvest losses and maintain the storage quality of edible produce, offering insights for efficient postharvest storage and freshness maintenance of fruits and vegetables.

Abstract The lotus seed embryo, known as “Plumula nelumbinis,” holds significance in Chinese culture as a traditional medicinal and edible herb rich in beneficial bioactive compounds like flavonoids, alkaloids, and polyphenols. Using ultrahigh‐performance liquid chromatography‐quadrupole‐exactive Orbitrap mass spectrometry, we meticulously analyzed the total flavonoids in Plumula nelumbinis (PNF) sourced from Hangzhou's West Lake. Our investigation preliminarily identified 29 flavonoid compounds, including 21 flavonoid O‐glycosides, five flavonoid C‐glycosides, and three aglycones, through comparisons with standards and literature references. Subsequent cellular anti‐inflammatory experiments using lipopolysaccharide (LPS)‐induced RAW264.7 cells demonstrated that treatment with 300 μg/mL of lotus seed flavonoids significantly reduced inflammatory factors' production, such as nitric oxide, prostaglandin E2, tumor necrosis factor, interleukin‐6, and interleukin‐1β. Additionally, PNF effectively lowered intracellular reactive oxygen species levels, mitigated LPS‐induced cell apoptosis, and nuclear translocation of nuclear factor‐κB (NF‐κB) p65 protein, collectively suppressing the NF‐κB p65 inflammatory pathway and demonstrating potent anti‐inflammatory properties. Crucially, the observed gene expression patterns of inflammatory factors aligned with their respective protein secretion levels. This study provides a comprehensive exploration of lotus seed flavonoids' anti‐inflammatory potential, highlighting their significance in potential therapeutic applications and laying the groundwork for future advancements in related functional foods.

Abstract Familial adenomatous polyposis (FAP) patients face an almost certain 100% risk of developing colorectal cancer, necessitating prophylactic colectomy to prevent disease progression. A crucial goal is to hinder this progression. In a recent clinical trial involving 14 FAP patients, half received 60 g of black raspberry (BRB) powder orally and BRB suppositories at bedtime, whereas the other half received only BRB suppositories at bedtime over 9 months. This intervention led to a notable reduction in rectal polyps for 11 patients, although 3 showed no response. In this study, we delved into the metabolic changes induced by BRBs in the same patient cohort. Employing mass spectrometry–based non‐targeted metabolomics, we analyzed pre‐ and post‐BRB urinary and plasma samples from the 11 responders. The results showed significant alterations in 23 urinary and 6 plasma metabolites, influencing various pathways, including polyamine, glutathione metabolism, the tricarboxylic acid cycle, inositol metabolism, and benzoate production. BRBs notably elevated levels of several metabolites associated with these pathways, suggesting a potential mechanism through which BRBs facilitate rectal polyp regression in FAP patients by modulating multiple metabolic pathways. Notably, metabolites derived from BRB polyphenols were significantly increased post‐BRB intervention, emphasizing the potential therapeutic value of BRBs in FAP management.

QP (Quenching and Partitioning) steel, the third generation of advanced high-strength steel, has garnered significant attention due to its exceptional mechanical performance. This paper investigates the interplay of grain rotation, slip transfer, and back stress strengthening through quasi in-situ deformation scanning electron microscope (SEM), electron backscatter diffraction (EBSD), and load-unload-reload (LUR) cycle tests. The findings reveal the substantial differences between QP980 and QP1180. QP980, characterized by heterostructures, exhibits non-uniform deformation characteristics. In regions with heterostructures, grain rotation inhibits slip transfer, leading to the accumulation of geometrically necessary dislocations (GNDs) and the formation of dislocation substructures (walls and cells), resulting in significant back stress strengthening. Conversely, due to the lack of heterogeneous structural regions, QP1180 displays uniform deformation characteristics, grain rotation and slip transfer promote each other, showing a weaker and different back stress strengthening compared to QP980. The interplay of grain rotation, slip transmission, and dislocation substructure formation influences the back stress behavior, impacting the mechanical properties and deformation behavior of the materials. This research provides a novel perspective on plastic deformation and strengthening theory, and serves as a valuable reference for material design and performance optimization.

Abstract Objective Rheumatoid arthritis (RA) is a chronic, progressive autoimmune disease with a complex pathogenesis that has not yet been fully elucidated, and T-cell pyroptosis is an important pathogenetic factor in RA. This study aimed to investigate the role of endoplasmic reticulum aminopeptidase 2 (ERAP2) in the pyroptosis of CD4+ T cells in RA and the specific molecular mechanism. Methods Peripheral venous blood was collected from human subjects, and CD4+ T cells were isolated and activated to measure the level of pyroptosis and ERAP2 expression. Pyroptosis levels were assessed using immunofluorescence, flow cytometry, qRT-PCR, and Western blotting. Changes in pyroptosis levels were observed upon knockdown or overexpression of ERAP2. To detect activated Caspase-1 in tissues, chimeric mice were engrafted with human synovial tissue and reconstituted with human CD4+ T cells. CD4 + T cells were treated with GLI1 antagonists and SMO receptor agonists to detect changes in pyroptosis levels. Results CD4+ T cell levels undergoing pyroptosis were found to be elevated in the blood and synovium of RA patients. The gene and protein expression of ERAP2 were significantly higher in CD4+ T cells from RA patients. Deletion of ERAP2 suppressed pyroptosis of these cells, attenuated the activation of Caspase-1 in tissue T cells, and reduced tissue inflammatory responses. Reciprocally, overexpression of ERAP2 triggered inflammasome assembly, activated Caspase-1, and induced pyroptosis in CD4+ T cells. Mechanistically, ERAP2 inhibits the Hedgehog signaling pathway and upregulates the expression of nucleotide-binding oligomerization segment-like receptor family 3(NLRP3), cleaved Caspase-1, and Gasdermin D to promote pyroptosis in CD4+ T cells. Conclusions Taken together, our results identify a novel mechanism by which ERAP2 regulates RA development and document the effect of the ERAP2/Hedgehog signaling axis on pyroptosis of CD4+ T cells from RA patients.

Abstract To investigate the impact of abscisic acid (ABA) on the postharvest flavor and quality of blueberries, this study employed different concentrations of ABA (400 μM ABA, 600 μM ABA, and 800 μM ABA) to treat the blueberries. The treated blueberries were then stored at 4°C (~80% relative humidity) for 35 day. Subsequently, genes associated with the metabolism of volatile aromatic substances and fatty acids in postharvest blueberries were assessed using real‐time PCR. Our results revealed that the treatment of ABA could maintain a high level of nutrient and aromatic substances in blueberries. Particularly, 600 μM ABA caused most significant changes in fatty acid metabolism‐related enzyme activities, such as lipoxygenase, hydroperoxide lyase, alcohol dehydrogenase, as well as related gene expressions, like VcLOX2S, VcHPL, VcADH1‐1, VcADH1‐2, VcADH1‐3, and VcADH1‐4. In addition, Furthermore, the blueberries treated with 600 μM ABA exhibited a superior quality when compared to the control group. This superior quality included higher levels of total acid, ascorbic acid, total phenol, anthocyanin, soluble sugar contents, and so forth. In conclusion, postharvest ABA treatment decelerated the decrease in volatile aroma substances and effectively maintained the flavor of blueberries.

Objective: Acupuncture as an alternative therapy for post-stroke cognitive impairment (PSCI) has emerged as a research focus. The inclusion of additional external treatments in many previous studies prevents a clear, direct assessment of acupuncture's impact on PSCI. In order to prevent patients from developing hypersensitivity to other treatments and misinterpreting acupuncture's true therapeutic value, this study establish stricter intervention criteria and exclude therapies beyond acupuncture. The review aimed to offering a clearer evaluation of acupuncture's efficacy and safety in PSCI treatment. Methods: This research involved a comprehensive search for randomized controlled trials (RCTs) across eight databases, adhering to the Cochrane Systematic Reviewer's Handbook 5.1.0 for risk-of-bias and quality assessments. A meta-analysis was conducted using RevMan 5.3 software. Results: The inclusion of 18 publications, totaling 1361 patients, was achieved. The meta-analysis demonstrated a significantly higher overall efficacy of acupuncture for PSCI compared to controls (OR = 4.06, 95 % CI 2.86–5.76, Z = 7.82). Notable statistical differences were observed in the Montreal Cognitive Assessment scores (MD = 2.32, 95 % CI 1.68–2.97, Z = 7.10) and the Mini-Mental State Examination scores (MD = 2.02, 95 % CI 1.06–2.98, Z = 4.13) between the groups. Improvements in the Barthel Index scores were noted for the experimental group (MD = 5.70, 95 % CI 4.68–6.72, Z = 10.92). Conclusion: Integrating acupuncture with Western medications offers significant benefits for treating PSCI over Western medications alone. However, the long-term efficacy of acupuncture in PSCI treatment and its potential in reducing recurrence rates remain undetermined. Further high-standard RCTs are essential to explore acupuncture's effectiveness in PSCI treatment more thoroughly.

Abstract Cognitive disorders, including dementia, Parkinson's and Alzheimer's disease, pose significant public health issues. Preventing/delaying their onset is essential as effective medical treatments are unavailable. Nuts contain bioactive compounds, including fatty acids, macronutrients such as proteins and dietary fibers, micronutrients, and polyphenols; these act synergistically preventing age‐related diseases. Epidemiological investigations have established that high nut consumption reduces cognitive impairment risk, suggesting it is useful for cognitively impaired individuals; however, clinical data on the effects of nut consumption on cognitive function are lacking. This review summarized nut consumption effects on oxidative stress, inflammatory response, and gut microbiota. Nuts reduce oxidative stress; their active substances, including glansreginin A, proanthocyanidins, quercetin, and nut oil, have significant anti‐inflammatory effects. Furthermore, they comprise complex mixtures, which synergistically induce beneficial changes in the intestinal microbial community.

In order to increase the utilization rate of shiitake mushroom by-products, the shiitake mushroom stem was crushed after superfine grinding with a jet mill, with coarse powder and 40 mesh powder as the control. The effect of jet milling on the physicochemical properties of shiitake mushroom stem powder and the dissolution amount of functional components represented by ergosterol and polysaccharides were studied. The cumulative dissolution rate of ergosterol and polysaccharides was fitted by the Weibull model. The results showed that after superfine grinding by jet milling, the average particle size (D50) of powder decreased to 3.21 μm, bulk density, tap density and L* value increased from 0.15 g/mL to 0.25 g/mL, 0.23 g/mL to 0.42 g/mL, 65.31 to 73.49, respectively. In order to develop soft fruit candy products with hypoglycemic function, the preparation method was optimized by single-factor and response surface studies. The texture profile analysis and sensory score was regarded as the evaluation indices, while bayberry juice was used as the main raw material, carrageenan and gelatin as the gelling agent, and xylitol as the sweetener. Additionally, the hypoglycemic effect of bayberry soft candy extract was evaluated by in vitro study, and the components of bayberry juice was detected by using UPLC-MS/MS assay. Finally, the hypoglycemic components and related pathways of action were predicted by network pharmacology. The results showed that in the 100 mL bayberry soft candy gel solution system, the optimal formulation of bayberry soft candy obtained by response surface methodology was as follows: 89.37% bayberry juice for swelling and constant volume, with 9.90% gelatin, 1.41% carrageenan, and 30.86% xylitol addition. The sensory score of bayberry soft candy produced under this process was 87.30, which was close to the theoretical value. The in vitro hypoglycemic study showed that the inhibition of α-glucosidase and α-amylase by 4 mg/mL of bayberry soft candy extract reached 98.58% and 86.89%, respectively. The network pharmacological analysis postulated that 3,5-diacetyltambrin (YM16), azaleatin (YM17) and raspberry ketone glucoside (YM1) were the key hypoglycemic components in bayberry juice, in addition, the human cancer pathway and PI3K-Akt pathway were important pathways of their action. The soft candy prepared under the optimal process condition showed good elasticity, fantastic taste and certain hypoglycemic effects. These results provide a certain theoretical basis for the development of fruit-flavored functional soft candy.

Abstract Skeletal disorders are commonly diagnosed by X-ray imaging, but the radiation limits its use. Optical imaging through the near-infrared-II window (NIR-II, 1000–1700 nm) can penetrate deep tissues without radiation risk, but the targeting of contrast agent is non-specific. Here, we report that lanthanide-doped nanocrystals can passively target the bone marrow, which can be effective for over two months. We therefore develop the high-resolution NIR-II imaging method for bone disease diagnosis, including the 3D bone imaging instrumentation to show the intravital bone morphology. We demonstrate the monitoring of 1 mm bone defects with spatial resolution comparable to the X-ray imaging result. Moreover, NIR-II imaging can reveal the early onset inflammation as the synovitis in the early stage of rheumatoid arthritis, comparable to micro computed tomography (μCT) in diagnosis of osteoarthritis, including the symptoms of osteophyte and hyperostosis in the knee joint.

Under the background of carbon neutrality, it is important to construct a large number of high-permeability power grid engineering (HPGE) systems, since these can aid in addressing the security and stability challenges brought about by the high proportion of renewable energy. Construction and engineering frequently involve multiple risk considerations. In this study, we constructed a three-stage comprehensive risk management model of HPGE, which can help to overcome the issues of redundant risk indicators, imprecise risk assessment techniques, and irrational risk warning models in existing studies. First, we use the fuzzy Delphi model to identify the key risk indicators of HPGE. Then, the Bayesian best–worst method (Bayesian BWM) is adopted, as well as the measurement alternatives and ranking according to the compromise solution (MARCOS) approach, to evaluate the comprehensive risks of projects; these methods are proven to have more reliable weighting results and a larger sample separation through comparative analysis. Finally, we established an early warning risk model on the basis of the non-compensation principle, which can help prevent the issue of actual risk warning outcomes from being obscured by some indicators. The results show that the construction of the new power system and clean energy consumption policy are the key risk factors affecting HPGE. It was found that four projects are in an extremely high-risk warning state, five are in a relatively high-risk warning state, and one is in a medium-risk warning state. Therefore, it is necessary to strengthen the risk prevention of HPGE and to develop a reasonable closed-loop risk control mechanism.

Dielectric metasurfaces with tunable functions are increasingly appealing to both the academia and industrial communities. Among the many tuning mechanisms reported so far, metasurface immersed in liquid crystal (LC) stands out as one of the most viable approaches for industrial applications such as Lidar and 3D sensing. However, as with traditional LC devices, alignment processes are necessary for LC-based tunable metasurfaces, leading to additional costs in terms of time and materials. In this paper, we present a novel LC-based tunable metasurface, wherein the alignment of LC is provided by the metasurface structures. LC serves as the dielectric environment for the metasurface with an electrically tunable refractive index. Simultaneously, purposely engineered metasurface induces alignment effects within the LC. The alignment effect of the metasurface on the LC was quantitatively characterized using crossed polarizers. A marked transmission contrast was observed, indicating the high-quality alignment provided solely by the metasurface. Electrical tunability was also measured, recording a maximum modulation depth in transmission amplitude of 94% at near-infrared telecommunication wavelengths. This work highlights the remarkable advantages of LC-based tunable metasurface devices, making them highly valuable for next-generation metasurface-integrated LC on silicon devices.

Abstract In this study, the hepatoprotective effect of alkali extract (NE) of water bamboo shoot (WBS) against acute alcoholic liver injury (ALI) in mice was evaluated, and its underlying mechanisms were explored. Animal experiment demonstrated that NE exhibited hepatoprotective effect on alcoholic intake‐induced ALI via significantly enhancing the hepatic activities of alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH), superoxide dismutase (SOD), glutathione peroxidase (GSH‐Px), and catalase (CAT), whereas significantly attenuated hepatic levels of malondialdehyde (MDA), interleukin‐1β (IL‐1β), interleukin‐6 (IL‐6), and tumor necrosis factor‐α (TNF‐α). The potential molecular mechanism and bioactive compounds of NE against ALI were explored by bioinformatics analysis and isosinensetin was found to be the major bioactive compounds in NE against ALI. The western blot analysis showed that NE inhibited phosphorylation of p65 and IκBα to suppress the NF‐κB pathway. Overall our result indicated that WBS possesses potential hepatoprotective effects against ALI.

Abstract Objective Rheumatoid arthritis (RA) is a chronic, progressive autoimmune disease. Over-activation of fibroblast-like synoviocytes is responsible for the hyperplasia of synovium and destruction of cartilage and bone and pyroptosis of FLS plays a key role in those pathological processes during RA. This study investigated the detailed mechanisms that SMAD2 regulates the pyroptosis of FLS and secretion of inflammatory factors in rheumatoid arthritis. Methods We collected synovial tissues of RA patients and FLS-RA and cultured FLS for detection of expression of SMAD2. ASC, NLRP3, cleaved-caspase-1, and GSDMD-N were detected by Western blot after overexpression of SMAD2. Besides, flow cytometry, electron microscope, ELISA, HE staining, and Safranin O staining were performed to further demonstrate that SMAD2 can affect the pyroptosis of FLS-RA. Results The expression of SMAD2 was down-regulated in synovial tissues of RA patients and FLS-RA. Overexpression of SMAD2 can inhibit the expression of ASC, NLRP3, cleaved-caspase-1, and GSDMD-N. Flow cytometry and electron microscope further demonstrated that SMAD2 attenuated pyroptosis of FLS-RA. In addition, overexpression of SMAD2 also inhibited inflammatory factors such as IL-1β, IL-18, IL-6, and IL-8 secretion and release of LDH. Besides, overexpression of SMAD2 can reverse the decrease of p-SMAD2 and TGF-TGF-β induced by nigericin. In vivo experiments on CIA rats further demonstrated that overexpression of SMAD2 by local intra-articular injection of LV-SMAD2 can effectively alleviate joint redness, swelling, and destruction of cartilage and bones. Conclusion SMAD2 inhibited FLS-RA pyroptosis by down-regulating of NLRP3 inflammasomes (NLRP3, ASC, and caspase-1 complex) and eased the secretion of inflammatory factors via the TGF-β signaling pathway, thereby improving the symptom of RA. We hope that this study may provide a new research idea for RA and a potential target for the treatment of RA.

The seismic behavior of the end-plate connections between a steel beam and the weak axis of the H-shaped steel column using a U-shaped connector was investigated using numerical analysis. Finite element (FE) models were established using ABAQUS 6.14 software, and the applicability of the modeling approach was verified by comparing the numerical results with the relevant experimental results. This parametric study of the joint was carried out to analyze the effects of the thickness of the U-shaped connectors, the thickness of the end-plates, the axial compression ratio of the columns, and the linear stiffness ratio of the beam to the column. The results show that the U-shaped connector set in the weak axis of the H-shaped column can form a box-shaped panel zone with the column flange and web. The volume of the panel zone and its resistance to shear deformation are increased through this connection. Finally, the recommended reasonable ranges for the thickness of the U-shaped connector, the thickness of the end-plates, the axial compression ratio of the columns, and the linear stiffness ratio of the beam to the column are proposed in this paper.

Abstract Background A new two-piece abutment design consisting of an upper prosthetic component and tissue-level base has been introduced; however, the biomechanical behavior of such a design has not been documented. This study aimed to investigate the effect of a two-piece abutment design on the stress in the implant components and surrounding bone, as well as its influence on microgap formation. Methods To simulate the implant models in the mandibular left first molar area, we established nine experimental groups that included three bone qualities (type II, III, and IV) and three implant–abutment designs (internal bone level, tissue level, and a two-piece design). After the screw was preloaded, the maximum occlusal (600 N) and masticatory (225 N) forces were established. Finite element analysis was performed to analyze the maximum and minimum principal stresses on the peri-implant bone; the von Mises stresses in the implants, abutments, bases, and screws, and the microgaps at the implant–abutment, implant–base, and base–abutment interfaces. Results For all three loading methods, the two-piece abutment design and bone-level connection exhibited similarities in the maximum and minimum principal stresses in the peri-implant bone. The von Mises stresses in both screws and bases were greater for the two-piece design than for the other connection types. The smallest microgap was detected in the tissue-level connection; the largest was observed at the implant–base interface in the two-piece design. Conclusions The present study found no evidence that the abutment design exerts a significant effect on peri-implant bone stress. However, the mechanical effects associated with the base and screws should be noted when using a two-piece abutment design. The two-piece abutment design also had no advantage in eliminating the microgap.

Cuticular wax plays a major role in the growth and storage of plant fruits. The cuticular wax coating, which covers the outermost layer of a fruit’s epidermal cells, is insoluble in water. Cuticular wax is mainly composed of very long-chain fatty acids (VLCFAs); their derivatives, including esters, primary alcohols, secondary alcohols, aldehydes, and ketones; and triterpenoids. This complex mixture of lipids is probably biosynthesized in the epidermal cells of most plants and exuded onto the surface. Cuticular wax not only makes the fruit less susceptible to microbial infection but also reduces mechanical damage to the fruit, thereby maintaining the fruit’s commodity value. To date, research has mostly focused on the changes, function, and regulation of fruit wax before harvest, while ignoring the changes and functions of wax in fruit storage. This paper reviews on the composition, structure, and metabolic regulation of cuticular wax in fruits. It also focuses on postharvest factors affecting wax composition, such as storage temperature, relative humidity (RH), gas atmosphere, and as exogenous hormones; and the effects of wax on fruit postharvest quality, including water dispersion, fruit softening, physiological disorders, and disease resistance. These summaries may be of assistance in better understanding the changes in cuticular wax in postharvest fruit and the resulting effects on fruit quality.

Mulberry leaf has potential in lipid-lowering. Here, we compared the chemical composition and lipid-lowering effects of different mulberry leaf-derived teas, and investigated their underlying gut microbiome-related mechanisms. The main components of mulberry leaf-derived tea were flavonoids, phenolic acids and alkaloids, with green tea (0.60% polyphenols, 3.35% polysaccharides, 0.20% DNJ) containing higher levels than black tea (0.40% polyphenols, 2.95% polysaccharides, 0.15% DNJ). Accordingly, the lipid-lowering efficacy of green tea was superior to that of black tea in high-fat (HFD)-induced hyperlipidemic mice. Black tea increased 2 genera and 130 metabolites, decreased 6 genera and 90 metabolites, while green tea increased 5 genera and 127 metabolites, decreased 15 genera and 64 metabolites. Correlation analysis showed that the alterations of gut microbiota and metabolic profiles caused by mulberry leaf tea were tightly associated with its lipid-lowering effect. Altogether, this study provided a promising functional food targeting gut microbiota for the management of lipid metabolism.

In this paper, we combine NTGK with flow and heat transfer model to perform numerical calculations on battery and cold plate to investigate thermal characteristics of li-ion batteries cooled by liquid and performance of cold plate during entire discharge process. A battery heat generation model is created using Kriging method after impacts of ambient temperature, SOC, and discharge rate are examined. Additionally, a new sort of cold plate is suggested based on constructal-theory, which performs better than conventional cold plate. Constructal cold plate reduces time needed for battery average temperature and maximum temperature differential to 25 °C and 5 °C by 15.25 % and 10.58 % for TimeT and TimeΔT, respectively, and pressure drop Δp by 22.89 % when compared to straight channel cold plate. Cooling effect of constructal cold plate is marginally inferior to that of serpentine cold plate, but flow resistance is greatly decreased, and pressure drop is only about one-tenth that of serpentine cold plate. Performance of constructal cold plate was examined using orthogonal optimization to determine effects of width of inlet and outlet and length of major channels. TimeT, TimeΔT, and Δp are decreased by 11 %, 14.69 %, and 15.22 % after optimization, and comprehensive performance is nearly doubled with improved performance.

Abstract Nitric oxide (NO) is a highly reactive gaseous small molecule that regulates plant growth. Sodium nitroprusside (SNP) was used as an NO donor to study the effects of NO treatment on browning and antioxidant activity of button mushrooms (Agaricus bisporus). The findings revealed that exogenous NO treatment alleviated the browning, decreased hardness, and increased weight loss of mushrooms stored at 4°C for 15 days, and reduced the accumulation of reactive oxygen species and membrane lipid peroxidation. Moreover, exogenous NO treatment dramatically increased the anabolism of endogenous NO, maintained the content of antioxidant substances including flavonoids and total phenols, and kept catalase and superoxide dismutase activities at a high level. Further studies revealed that exogenous NO treatment inhibited the activities of enzymes that are involved in browning, which including polyphenol oxidase (PPO) and tyrosinase (TYR), and increased phenylalanine ammonia‐lyase (PAL) activity, with SNP treatment at 0.05 mM being the most effective. Furthermore, SNP treatment at 0.05 mM also inhibited PPO and TYR gene expressions while increasing PAL gene expression. Therefore, our findings suggest that exogenous NO has the potential for development in delaying postharvest browning and maintaining the quality of mushrooms.

The objective of the present research is to investigate the influence of the microstructural evolution mechanism on mechanical properties during deformation. For this purpose, hot rolled Fe-0.22C-6.12Mn-3.08Al steel is subjected to intercritical annealing at 740 °C for 10 min. The annealed specimen contains a mixture of lath-like α-ferrite and reverted γ, whose volume fraction is approximately 38.7%. During deformation, once the critical stress caused by the severe dislocation pile-ups is reached, the strain/stress-induced phase transformation occurs. First, grain boundaries of γ/α serve as the preferential nucleation sites for fresh α′-martensite. Subsequently, fresh α′-martensite nucleates inside reverted γ. In the meantime, the interfacial α′-martensite grows continuously into the reverted γ interior. During deformation (prestrain at 0.0%, 5.1% and 7.8%), the misorientation (MO) in α-ferrite+α′-martensite continuously rises from 0.63° to 0.67°; nevertheless, the MO in reverted γ increases from 0.65° to 0.73° and then rapidly decreases to 0.69°. Hence, the phase transformation of reverted γ is accompanied by obvious stress softening; thus, effective work hardening occurs in the subsequent deformation region due to α-ferrite and fresh α′-martensite. This phenomenon greatly enhances the strength and delays necking, e.g., the annealed specimen possesses outstanding mechanical properties (992 MPa, 47.6% and 47.2 GPa × %). Once necking occurs, the residual reverted γ is rapidly consumed in the necking region. Hence, the hardness in the necking region (5.31 ± 0.20 GPa) is much higher than that in the uniform deformation region (4.53 ± 0.17 GPa) due to the abundant hard fresh α′-martensite.

Background: Systemic chemotherapy (SC) remains the only first-line treatment for unresectable intrahepatic cholangiocarcinoma (iCCA). Hepatic arterial infusion chemotherapy (HAIC) has been recently proven to be effective in managing hepatocellular carcinoma (HCC). Hence, our study aims to investigate the safety and efficacy of HAIC in treating unresectable iCCA patients.Methods: We reviewed 146 patients with unresectable iCCA who had received HAIC or SC between March 2016 and March 2022 in a retrospective manner. Outcomes of patients and safety were compared between the HAIC and SC groups.Results: There were 75 and 71 patients in the HAIC and SC groups, respectively. The median OS in the HAIC and SC groups was 18.0 and 17.8 months (p = 0.84), respectively. The median PFS in the HAIC and SC groups was 10.8 and 11.4 months (p = 0.59), respectively. However, the HAIC group had significantly longer intrahepatic progression-free survival (IPFS) than the SC group (p = 0.035). The median IPFS in the HAIC and SC groups was 13.7 and 11.4 months, respectively. According to the OS (p = 0.047) and PFS (p = 0.009), single-tumor patients in the HAIC group appeared to benefit more. In addition, the overall incidence of adverse events (AEs) was lower in the HAIC group than that in the SC group.Conclusion: Our study revealed that HAIC was a safe and effective therapeutic regimen for unresectable iCCA with better intrahepatic tumor control when compared to SC. Meanwhile, patients with single tumor were more likely to benefit from HAIC than SC.

Summary: Cyclin-dependent kinase 4 (CDK4) and retinoblastoma protein (RB) are both important cell-cycle regulators that function in different scenarios. Here, we report that FERM domain-containing 8 (FRMD8) inhibits CDK4 activation and stabilizes RB, thereby causing cell-cycle arrest and inhibiting colorectal cancer (CRC) cell growth. FRMD8 interacts separately with CDK7 and CDK4, and it disrupts the interaction of CDK7 with CDK4, subsequently inhibiting CDK4 activation. FRMD8 competes with MDM2 to bind RB and attenuates MDM2-mediated RB degradation. Frmd8 deficiency in mice accelerates azoxymethane/dextran-sodium-sulfate-induced colorectal adenoma formation. The FRMD8 promoter is hypermethylated, and low expression of FRMD8 predicts poor prognosis in CRC patients. Further, we identify an LKCHE-containing FRMD8 peptide that blocks MDM2 binding to RB and stabilizes RB. Combined application of the CDK4 inhibitor and FRMD8 peptide leads to marked suppression of CRC cell growth. Therefore, using an LKCHE-containing peptide to interfere with the MDM2-RB interaction may have therapeutic value in CDK4/6 inhibitor-resistant patients.

To reduce the waste of resources, kiwifruit residue was fermented by Lactobacillus plantarum to prepare the beverage. The effects of solid-liquid ratio, inoculation volume, initial TSS and fermentation time on sensory score and viable count of the product were investigated by single-factor experiments. On these grounds, the fermentation process was optimized by response surface methodology with the sensory score as the response value. Furthermore, the changes in its nutritional quality, antioxidant activity and flavor during fermentation were analyzed. Results showed that, the optimal fermentation process of kiwifruit residue beverage was as follows: Initial TSS 12.4%, inoculation volume 3.3%, fermentation time 36 h, and the sensory score of the product was 87.17 scores. The nutritional quality assay showed that, as time fermentation, the viable count and the contents of soluble protein and total acid increased, total phenol content initially decreased and then increased, flavonoids content had no significant difference, and the contents of total sugar and ascorbic acid decreased. Under the optimum process, ABTS+· scavenging rate, DPPH· scavenging rate as well as iron reduction increased by 9.59%, 6.89%, and 4.08%, respectively. After 36 h fermentation, the product reached the best comprehensive nutritional quality. In addition, the assay of headspace-gas chromatography-ion mobility spectroscopy (HS-GC-IMS) showed that the first 24 h of fermentation was the transformation stage of flavor compounds, and 36 h was an important node in the flavor formation, while the compounds with pungent odor were over-accumulated after 48 h fermentation. Among them, the relative contents of ketones and esters increased, while the relative contents of aldehydes and alcohols decreased. Further study found that, during fermentation, the fruit aroma and caramel aroma decreased first and then increased, the fermentation aroma increased, and the characteristic aroma as well as green aroma decreased. The olfactory sensory score (31.00) was the highest at 36 h, and the main aroma changed from green aroma to fermented aroma. To sum up, the product under the optimal process conditions (fermentation 36 h) had excellent sensory and was rich in nutrition and flavor, while the fermentation process had a great impact on the overall quality of the product and was required to be strictly controlled.

Measurement-device-independent QKD schemes suffer from a trade-off between ease of implementation (avoiding the need for global phase locking) and high rates (quadratic improvement in rate). Here, the authors propose a protocol which offers both simple implementation and strong performances.

ObjectiveTo explore the value of the FT4/TSH ratio in the etiological diagnosis of newly diagnosed patients with thyrotoxicosis.MethodsThe retrospective study was conducted on 287 patients with thyrotoxicosis (122 patients with subacute thyroiditis and 165 patients with Graves’ disease) and 415 healthy people on their first visit to our hospital. All patients underwent thyroid function tests including the measurement of T3, T4, FT3, FT4, TSH, T3/TSH, and T4/TSH. The receiver operating characteristic (ROC) curve was employed to evaluate the value of FT4/TSH in the differential diagnosis of Graves’ disease and subacute thyroiditis, and compared with other related indicators.ResultsThe area under the curve of FT4/TSH for diagnosing Graves’ disease and thyroiditis was 0.846, which was significantly larger than the area under the curve of T3/T4 ratio (P< 0.05) and FT3/FT4 ratio (P< 0.05). When the cut-off value of the FT4/TSH ratio was 5731.286 pmol/mIU, the sensitivity was 71.52%, the specificity was 90.16%, the positive predictive value was 90.77% and the negative predictive value was 70.06%. The diagnostic accuracy was 79.44%.ConclusionFT4/TSH ratio can be used as a new reference index for the differential diagnosis of thyrotoxicosis.

Intelligent packaging was prepared and evaluated to determine the quality of shiitake mushrooms using colorimetric sensing. Bromothymol blue/methyl red composite dyes were incorporated into polyvinyl alcohol (PVA) to form a porous indicator via freeze-drying. The PVA indicator was then modified with polylactic acid (PLA). The porous structure of the indicator was investigated using scanning electron microscopy (SEM) imaging, and it was found that the hydrophobicity of the PVA/PLA indicator was significantly improved following modification. A sensitive colorimetric response toward the presence of CO2 in packaging was achieved using the naked eye. During investigations into the potential application of this system, the ΔE value of the indicator was found to correlate significantly with the quality indices (i.e., the browning index, firmness value, malondialdehyde content, and sensory score) of shiitake mushrooms during storage. Based on these correlations, qualitative identification of the freshness of shiitake mushrooms was studied using principal component analysis, while a prediction model of this freshness was obtained by partial least squares regression. This indicator, therefore, shows potential for application in intelligent packaging for the detection of CO2 due to the clear visualization of color changes by the naked eye.

Introduction: Polarization-sensitive optical coherence tomography (PS-OCT) is a functional extension of standard OCT. PS-OCT systems can be generally categorized into two categories based on the number of input polarization states on the sample: multi-input polarization state (multi-IPS) and single IPS. In addition, each category includes two configurations: fiber-based system and bulk optics-based system. However, there are complex and time-consuming steps to calibrate the polarization states of light among the reference, the sample, and detection arms for fiber-based system. And it is not compact and robust enough for bulk optics-based system.Methods: In the modified SD PS-OCT system with structural symmetry in both arms of the reference and sample, there are no bulk polarization optical elements in both arms of the reference and the sample. A circularly polarized light was used to incident on sample, and Stokes vector of backscattered light was employed to characterize the birefringence of biological tissues based on the quaternion approach, which directly establishes the relationship between Stokes vectors of backscattered light and Jones matrix of the sample.Results and discussion: The new algorithm provides the analytic solution of retardance and fast-axis orientation. To evaluate the performance of the developed system, an eighth-wave plate is used. Then, the polarization properties of the myocardial tissue in vivo are quantitatively reconstructed based on the quaternion approach. The results demonstrated that the proposed method has an advantage over Jones formalism based on a single input state and two polarization input states. In the future, the modified SD PS-OCT could be improved as a common path SD PS-OCT for clinical applications.

Dendrobium officinale polysaccharides (DOPs) are important active polysaccharides found in Dendrobium officinale, which is commonly used as a conventional food or herbal medicine and is well known in China. DOPs can influence the composition of the gut microbiota and the degradation capacity of these symbiotic bacteria, which in turn may determine the efficacy of dietary interventions. However, the necessary analysis of the relationship between DOPs and the gut microbiota is lacking. In this review, we summarize the extraction, structure, health benefits, and related mechanisms of DOPs, construct the DOPs-host axis, and propose that DOPs are potential prebiotics, mainly composed of 1,4-β-D-mannose, 1,4-β-D-glucose, and O-acetate groups, which induce an increase in the abundance of gut microbiota such as Lactobacillus, Bifidobacterium, Akkermansia, Bacteroides, and Prevotella. In addition, we found that when exposed to DOPs with different structural properties, the gut microbiota may exhibit different diversity and composition and provide health benefits, such as metabolism regulations, inflammation modulation, immunity moderation, and cancer intervention. This may contribute to facilitating the development of functional foods and health products to improve human health.

To ensure the security of information exchange in software-defined optical networks, quantum key distribution (QKD) based on quantum mechanics is introduced. However, the slow and valuable process of generating quantum key resources contradicts the high-speed data transmission requirements of optical networks. To address this issue, this paper proposes the CPSR-HQKDN scheme, which takes into account factors such as security requests, key demand, key residual, and key update rates for trusted and untrusted links. This approach improves resource utilization and service efficiency by optimizing the processing order of key requests. Moreover, the routing strategy dynamically adjusts based on the network resource environment, thereby increasing the success rate of key requests. Through simulation experiments comparing the performance of the CPSR-HQKDN routing scheme with existing schemes, it is observed that in high-concurrent scenarios, the CPSR-HQKDN routing scheme can improve the success rate of key requests by at least 5%.

Dendrobium officinale leaves have the potential to be processed into natural antioxidants, functional foods, and food additives. To maximally maintain their quality, fresh D. officinale leaves were dehydrated using different drying methods, i.e., hot air drying (HD), microwave drying (MD), infrared drying (IRD), and freeze drying (FD), and then the physicochemical properties, microstructure, and biological activities of the dried samples were compared. The results showed that, with the FD method, the samples had a porous microstructure, maintained the highest phenolic content, and demonstrated the highest antioxidant and hypoglycemic activities. Among the three thermal drying methods, with the IRD method, the samples retained higher phenolic contents, showed stronger DPPH free-radical scavenging, ferric ion reducing, ferrous ion chelating, and α-glucosidase inhibitory abilities, and more strongly promoted glucose metabolism in insulin-resistant HL-7702 cells than the samples with the MD and HD methods. These results suggested that FD was the most suitable method. However, IRD might be a promising alternative, owing to the high cost and long time needed for FD for the large-scale drying of D. officinale leaves.

The objective of this study was to investigate the possible association between the single nucleotide polymorphism (SNP), rs35569394, of the vascular endothelial growth factor gene (VEGF) and the risk of esophageal cancer (EC) in the Han Chinese population. A total of 290 EC subjects and 322 ethnically matched unrelated healthy controls free from the esophageal disease were studied. Genomic DNA was isolated from peripheral blood by salting out. Genotyping of VEGF rs35569394 polymorphism was carried out via polymerase chain reaction followed by agarose gel electrophoresis. The results showed that the distribution of genotypes was significantly different across the gender groups (p=0.032) and clinical stages (p=0.034). VEGF rs35569394 was associated with EC risk (p= 0.012, OR=1.34). A gender analysis break-down showed that rs35569394-D allele frequency was significantly higher in females than in the controls (p=0.0004, OR=1.81). Moreover, significant associations were also found in females under the dominant model (II versus ID+DD: χ2=8.18, p=0.003, OR=2.12) and the recessive model (II+ID versus DD: χ2=8.25, p=0.004, OR=2.39). Additionally, we found that the genotype, rs35569394-DD, was associated with a complete response + partial response to chemotherapy when compared with rs35569394-II (χ2=4.67, p=0.030, OR=0.47). In conclusion, our case-control study showed that the VEGF rs35569394 was significantly associated with the clinical stages and the increased risk of EC in Han Chinese females. In addition, the genotype rs35569394-DD showed a better response to chemotherapy.

Systems of spins engineered with tunable density and reduced dimensionality enable a number of advancements in quantum sensing and simulation. Defects in diamond, such as nitrogen-vacancy (NV) centers and substitutional nitrogen (P1 centers), are particularly promising solid-state platforms to explore. However, the ability to controllably create coherent, two-dimensional spin systems and characterize their properties, such as density, depth confinement, and coherence, is an outstanding materials challenge. We present a refined approach to engineer dense (≳1 ppm ⋅ nm), 2D nitrogen, and NV layers in diamond using delta-doping during plasma-enhanced chemical vapor deposition epitaxial growth. We employ both traditional materials techniques, e.g., secondary ion mass spectrometry, alongside NV spin decoherence-based measurements to characterize the density and dimensionality of the P1 and NV layers. We find P1 densities of 5–10 ppm ⋅ nm, NV densities between 1 and 3.5 ppm ⋅ nm tuned via electron irradiation dosage, and depth confinement of the spin layer down to 1.6 nm. We also observe high (up to 0.74) ratios of NV to P1 centers and reproducibly long NV coherence times, dominated by dipolar interactions with the engineered P1 and NV spin baths.

ObjectivesLotus root (Nelumbo nucifera Gaertn) is a common medicinal-food dual-use vegetable. In this study, the effects of lotus root extract on acute alcoholism were investigated.MethodsThe Walle-Hoch method was used to determine the ADH activity of lotus root extracts in vitro. Lotus root methanol extract were identified by UPLC-QTOF-MS/MS based metabolomics analysis. Then 109 active ingredients with achievable oral doses and drug-like properties were explored using the TCMSP platform. SwissTargetPrediction Database to predict lotus root treatment targets for acute alcoholismSTRING database (https://www.string-db.org/) was used to construct protein-protein interaction network graphs. Gene ontology (GO) functional, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of genes common to lotus root and alcoholism by Metascap database. Molecular docking simulations were performed using AutoDock 1.5.6 software. Animal experiments verified the relieving effect of lotus root extract on acute alcoholism after intervention.ResultsResults indicated the methanol extract of lotus root showed the highest activation rate of ethanol dehydrogenase in vitro (18.87%). The 433 compounds of lotus root methanol extract were identified by UPLC-QTOF-MS/MS based metabolomics analysis. Bioinformatics analysis indicate that there were 224 intersectioning targets between lotus root extract and acute alcoholism. KEGG enrichment analysised shows that lotus root extract may play a role in treating acute alcoholism by intervening with the neuroactive ligand-receptor interaction pathway. The protein-protein interaction network (PPI) analysis found that HSP90AA1, MAPK1 and STAT3 played a key role in lotus root extract-modulated PPI networks. Molecular docking showed that (7R, 8S)-dihydrodihydrodipine cypressol had the best binding ability with MAPK1. Experiments in mice indicate that lotus root extract improved the activity of liver alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH), catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), increase glutathione (GSH) and reduce malondialdehyde (MDA) levels, decrease glutamate transaminase (AST), alanine transaminase (ALT) and alkaline phosphatase (AKP) in the serum of mice with acute alcoholism, and accelerate the metabolic rate of alcohol after drinking. This study reveals the mechanism of lotus root to alleviate acute alcoholism, which provides a basis for further research on functional foods using lotus root and offers new possibilities for the treatment of acute alcoholism.ConclusionsThe results of the current study showed that the methanolic extract of lotus root had the highest activation rate of ethanol dehydrogenase. Network pharmacology results suggest that lotus root extract may play a role in the treatment of alcoholism by regulating signaling pathways, such as neuroactive ligand-receptor interactions, as well as biological processes, such as regulation of secretion, regulation of ion transport, response to lipopolysaccharides, and response to alcohol. Animal experiments confirmed the therapeutic effect of lotus root on acute alcoholism mechanistically through activation of alcohol catabolic enzyme, reduction of oxidative stress and protection of liver function.

The melting of ice creates favorable conditions for the development of the Arctic by elevating the Arctic’s geostrategic value and stoking national competition. As a result, many states, both within and beyond the Arctic region, have recently released Arctic strategies or updated existing ones, clarifying their roles, interests, priorities, and policies for Arctic international cooperation. In this study, a database is generated with a collection of Arctic strategies from 16 states, and then grounded theory is applied to code the texts in these strategy documents. Using this code, word-frequency analysis, cross-tabulation, correlation analysis, and cooperative-network analysis are implemented to explore competition and collaboration in the Arctic among these 16 states. The results indicate that Arctic states are significantly concerned with governance, economic and social development, and security and stability, while environmental protection and scientific research are given more attention by non-Arctic states. In addition, the highest value of the Pearson correlation coefficient is identified as 0.85 for collaboration and Arctic public security affairs, which indicates that collaboration on Arctic security will dominate the development of Arctic affairs in the near future.

Cuticular wax contributes to maintaining postharvest storage quality against fruit water loss and softening. Triterpenoids, such as oleanolic acid (OA) and ursolic acid (UA), are the main components in blueberry cuticular wax, but their role in water migration during the storage of blueberries remains to be determined. Here, we examined the relationship between the content of OA and UA and the storage quality of blueberry fruit (25 °C). The results revealed that the UA content during eight-day postharvest storage ranged from 58 to 77 μg cm−2, which was negatively related to weight loss. Additionally, we investigated the effect of exogenous OA and UA on water migration in the blueberry fruit during storage at room temperature; the weight loss was significantly lower (by 22%) with UA treatment than in the control fruit. Our findings indicate that OA and UA effectively affect water migration in blueberry fruit during postharvest storage, which could contribute to improving postharvest preservation techniques.

Abstract Shiitake mushrooms (Lentinus edodes) form a great source of protein; however, the effect of enzymatic hydrolysis to improve its flavor has not yet been investigated. To explore the effects of different protease treatments on protein degradation and flavor compounds of shiitake mushrooms, papain, neutral protease, and flavourzyme were selected for the hydrolysis of shiitake mushrooms. Proteolysis index, Trichloroacetic acid‐nitrogen soluble index, and protein content were estimated to evaluate protein degradation. Flavor amino acids and volatile components of the hydrolysate were also analyzed. Results showed flavourzyme has an advantage over the others in promoting protein degradation and improving flavor. Compared to single‐enzyme treatment, a combination treatment with three enzymes, at 1:1:1 ratio, was found to perform greater protein hydrolysis. After protease‐mediated hydrolysis, the variety and content of flavor substances (free amino acids and eight‐carbon compounds) in L. edodes increased. We concluded that protease treatment is an effective method for improving the quality and flavor of L. edodes.

Fruit acidity is an essential factor affecting blueberry organoleptic quality. The organic acid content in blueberry fruit mainly contributes to fruit acidity. This study aims to evaluate the effect of exogenous salicylic acid (SA), the principal metabolite of aspirin, on the organoleptic quality and organic acid metabolism in rabbiteye blueberry (Vaccinium virgatum Ait, ‘Powderblue’) during cold storage (4 °C). Results showed that SA-treated fruit reduced fruit decay and weight loss delayed fruit softening, and decline of total soluble solids (TSS). TA and total organic acid amounts stayed the same during the late storage period in SA-treated fruit. Four kinds of organic acid components, malic acid, quinic acid, citric acid, and succinic acid, were at higher levels in fruit treated by SA as compared to control. SA enhanced the activities of PEPC, NAD-MDH, and CS to promote the synthesis of malic acid and citric acid. Meanwhile, the activities of NADP-ME, ACL, and ACO, which participated in the degradation of malic acid and citric acid, were inhibited by SA. qPCR results also showed that the expression of VcPEPC, VcNAD-MDH, and VcCS genes were upregulated. In contrast, SA downregulated the expression of VcNADP-ME, VcACL, and VcACO genes. In conclusion, SA could regulate the key genes and enzymes that participated in organic acids metabolism to maintain the freshness of blueberry during cold storage, therefore minimizing the economic loss.

Most of the current research only explored the loading of an active substance in active packaging. In this study, cinnamaldehyde essential oil (CEO) and chlorogenic acid (CA) were co-encapsulated in chitosan (CS) nanoparticles based Pickering emulsion. The morphology and wettability of CS-CA particles were determined. In addition, physicochemical characterizations and stability of the Pickering emulsion were also investigated. Results showed that the wettability of nanoparticles was improved with increasing the ratios of CS to CA, which is helpful to stabilize the emulsion. CEO Pickering emulsion was stabilized by CS-CA nanoparticles and CEO emulsion showed the best stability by using CS-CA nanoparticles with the ratios of CS to CA 1:0.75 with the minimum creaming index value of 26.5 ± 4.6% after 5 days of storage. These overall results presented in this work demonstrate, for the first time, the potential of Pickering emulsion for the co-encapsulation of water-soluble and water-insoluble ingredients.

Recent evidence suggests that endoplasmic reticulum (ER) stress plays a vital role in inflammatory bowel disease (IBD). Therefore, the aim of this study was to investigate the mechanism by which ER stress promotes inflammatory response in IBD. The expression of Gro-α, IL-8 and ER stress indicator Grp78 in colon tissues from patients with Crohn’s disease (CD) and colonic carcinoma was analyzed by immunohistochemistry staining. Colitis mouse model was established by the induction of trinitrobenzene sulphonic acid (TNBS), and the mice were treated with ER stress inhibitor tauroursodeoxycholic acid (TUDCA). Then the body weight, colon length and colon inflammation were evaluated, and Grp78 and Gro-α in colon tissues were detected by immunohistochemistry. Epithelial cells of colon cancer HCT116 cells were treated with tunicamycin to induce ER stress. Grp78 was detected by Western blot, and chemokines were measured by PCR and ELISA. The expression levels of Grp78, Gro-α and IL-8 were significantly upregulated in intestinal tissues of CD patients. Mice with TNBS induced colitis had increased expression of Grp78 and Gro-α in colonic epithelia. TUDCA reduced the severity of TNBS-induced colitis. In HCT116 cells, tunicamycin increased the expression of Grp78, Gro-α and IL-8 in a concentration-dependent manner. Furthermore, p38 MAPK inhibitor significantly inhibited the upregulation of Gro-α and IL-8 induced by tunicamycin. In conclusion, ER stress promotes inflammatory response in IBD, and the effects may be mediated by the activation of p38 MAPK signaling pathway.

Melatonin, a crucial bioactive molecule, involved in several physiological processes in plants. This study investigated the effects of melatonin (MT) treatment on lignification, including firmness, lignin, lignified-enzyme activities, the expression patterns of genes encoding corresponding enzymes and transcription factors in water bamboo shoot during storage for 8 days. MT treatment decreased the firmness and content of lignin. It inhibited the degradation of total phenols and ascorbic acid and delayed the lignin biosynthesis, via reducing the activities of phenylalanine ammonia-lyse cinnamyl alcohol dehydrogenase and peroxidase, as well as lignin biosynthesis-related genes expression levels. Transcription factors of ZlNAC1, ZlNAC2, ZlNAC3 and ZlNAC4 from NAC family and ZlMYB1 and ZlMYB2 from MYB family were increased in water bamboo shoot after harvest and MT-treated markedly reduced their expression. Therefore, our findings supply a fundamental understanding of MT treatment suppression of lignification and establish a foundation for further research on transcriptional regulation.

Abstract Suspension array technology (SAT) has aroused substantial interest due to its strong multiplexed detection capability, quick binding kinetics, high throughput, and high detection sensitivity. Although rapid developments in material science have led to tremendous progress with regards to SAT, several challenges still exist that limit the performance of SAT, including limited barcode numbers, unpredictable barcode signal and suboptimal detection sensitivity. Therefore, major efforts are still needed to further improve performance to fulfill the higher requirements of precision diagnosis. In this review, we focus on summarizing recent advances regarding encoded microspheres and labels for functional nanomaterial‐based suspension array technology, then analyze the current challenges and propose corresponding solutions. We anticipate that this review will be helpful for the future design of functional nanomaterial‐based SAT platforms with superior performance.
