Parabens are alkyl esters of p‐hydroxybenzoic acid that are commonly used as preservatives in personal care products such as cosmetics. Recent studies have revealed the presence of parabens in surface and tap water because of their use as disinfection products; however, little is known about their occurrence in biological samples and their bioaccumulation potential, particularly in raptor birds known as sentinels for pollutant detection. We examined the occurrence and tissue distribution of parabens, their metabolites, and halogenated byproducts in the liver, kidney, brain, and muscle of birds of prey from Texas and North Carolina (USA). Methylparaben (MeP), propylparaben (PrP), and butylparaben (BuP) were detected in more than 50% of all tissues examined, with the kidney exhibiting the highest concentration of MeP (0.65–6.84 ng/g wet wt). Para‐hydroxybenzoic acid (PHBA), a primary metabolite, had the highest detection frequency (>50%) and a high accumulation range in the liver, of 4.64 to 12.55 ng/g. The chlorinated compounds chloromethylparaben and chloroethylparaben were found in over half of the tissues, of which dichloromethylparaben (2.20–3.99 ng/g) and dichloroethylparaben (1.01–5.95 ng/g) in the kidney exhibited the highest concentrations. The dibrominated derivatives dibromideethylparaben (Br2EtP) was detected in more than 50% of samples, particularly in muscle and brain. Concentrations in the range of 0.14 to 17.38 ng/g of Br2EtP were detected in the kidney. Dibromidepropylparaben (Br2PrP) was not frequently detected, but concentrations ranged from 0.09 to 21.70 ng/g in muscle. The accumulations of total amounts (sum) of parent parabens (∑P), metabolites (∑M), and halogenated byproducts (∑H) in different species were not significantly different, but their distribution in tissues differed among the species. Positive correlations were observed among MeP, PrP, BuP, and PHBA in the liver, suggesting similar origins and metabolic pathways. Environ Toxicol Chem 2024;43:2365–2376. © 2024 The Author(s). Global amphibian declines have the highest incidence in tropical regions, but most of the ecotoxicological data on amphibians is collected on temperate northern hemisphere anuran species. We tested the hypothesis that tropical anuran larvae (Epipedobates anthonyi) would be more sensitive to pesticides than a North American native species (Lithobates sphenocephalus). For 12 pesticides, 96-hr range-finding acute toxicity tests were conducted to determine if mortality occurred at environmentally relevant levels. Based on those studies, two substances were selected for additional time-to- event analyses in both species as well as median lethal concentration (LC50) calculations. Time-to- event results indicated that the two species appear to be roughly equivalent in their sensitivity to the two tested pesticides. Significant differences between species were not consistent across concentrations for either the insecticide terbufos or the herbicide pendimethalin. The utility of LC50 data was mixed with one LC50 providing an arbitrarily large standard error around the LC50 precluding informative comparisons across species. However, standard LC50 methods allowed data collection that continues to contribute to our understanding of the protectiveness of fish as surrogates for anuran larvae. While our data set is limited, it appears that testing temperate species would be protective for tropical species in ecological risk assessments. Our data also support the continued use of fish as surrogates for amphibian larvae as none of the species were more sensitive to the tested pesticides than rainbow trout (Oncorhynchus mykiss), the standard sensitive fish species used for acute toxicity testing. Despite widespread recognition of significant data deficiencies, reptiles remain a relatively understudied taxon in ecotoxicology. To conduct ecological risk assessments on reptiles frequently requires using surrogate taxa such as birds, but recent research suggests that reptiles have significantly different exposure profiles and toxicant sensitivity. We exposed western fence lizards, Sceloporus occidentalis, to the same quantities of three model chemicals via oral (gavage) and dermal (ventral skin application) exposure for either 24 or 48 hours. Three phthalate esters (di-methyl phthalate [DMP], di-iso-butyl phthalate [DIBP], and di-n-octyl phthalate [DNOP]) were chosen as model chemicals because they represent a gradient of lipophilicity but are otherwise structurally similar. Overall, the more lipophilic phthalates (DIBP and DNOP) were found to have higher concentrations in tissues than the less lipophilic DMP. Significant differences in tissue concentrations between DIBP and DNOP were tissue-dependent, suggesting that delivery to a site of action following exposure is not only a simple function of lipophilicity. In dermal treatments, DMP usually had fewer detections (except in ventral skin samples), suggesting that lipophilicity (log Kow>2) is a requirement for uptake across the skin. In general, tissue residues were greater in oral treatments than dermal treatments (significant in adipose and liver tissue), but differences were driven strongly by differences in DMP which did not appear to be absorbed well across skin. When differences in tissue residue concentrations between oral and dermal exposure did occur, the difference was not drastic. Taken together these results suggest that dermal exposure should be considered in risk assessments for reptilian receptors. Dermal exposure may be an especially important route for reptiles as their ectothermic physiology translates to lower energetic demands and dietary exposure compared to birds and mammals.
