Abstract Endothelial-mesenchymal transition (EndMT) is defined as an important process of cellular differentiation by which endothelial cells (ECs) are prone to lose their characteristics and transform into mesenchymal cells. During EndMT, reduced expression of endothelial adhesion molecules disrupts intercellular adhesion, triggering cytoskeletal reorganization and mesenchymal transition. Numerous studies have proved that EndMT is a multifaceted biological event driven primarily by cytokines such as TGF-β, TNF-α, and IL-1β, alongside signaling pathways like WNT, Smad, MEK-ERK, and Notch. Nevertheless, the exact roles of EndMT in complicated diseases have not been comprehensively reviewed. In this review, we summarize the predominant molecular regulatory mechanisms and signaling pathways that contribute to the development of EndMT, as well as highlight the contributions of a series of imperative non-coding RNAs in curbing the initiation of EndMT. Furthermore, we discuss the significant impact of EndMT on worsening vasculature-related diseases, including cancer, cardiovascular diseases, atherosclerosis, pulmonary vascular diseases, diabetes-associated fibrotic conditions, and cerebral cavernous malformation, providing the implications that targeting EndMT holds promise as a therapeutic strategy to mitigate disease progression.

Abstract The application of biochar to soil is widely recognized as a promising strategy for enhancing the accumulation and stability of soil organic carbon (SOC), which is crucial in mitigating climate change. However, the influence of interactions between plants and biochar on soil microbial communities and their involvement in SOC mineralization and stability remains unclear. This understanding is essential for optimizing carbon (C) sequestration in systems involving plants, soil, and biochar. In this study, employing a 13C natural abundance approach, we investigated the effect of biochar on the maize rhizosphere priming effect (RPE) in paddy soil. We also examined alterations in microbial communities and functional genes related to C degradation and fixation. Over the 99 days of maize growth, biochar application increased RPE and total SOC while decreasing dissolved organic C. It also elevated soil pH, resulting in shifts in fungal and bacterial community structure, favoring oligotrophic species. Fungal and bacterial assemblies were dominated by deterministic and stochastic processes, respectively. While the abundance of fungal guilds varied irregularly, bacterial guilds were uniformly enriched under biochar-plant interactions. Functional traits such as ecoenzymatic activities, bacterial guilds, and functional genes predominantly affected RPE under biochar application. Bacterial functional genes associated with C degradation and fixation were concurrently enhanced with biochar application. Our results indicate that interactions between plants and biochar can enhance native SOC mineralization and accumulation in a short timeframe by modulating functional traits of soil microorganisms, particularly the bacterial community involved in C degradation and fixation. Abstract Piercing-sucking pests are the most notorious group of pests for global agriculture. RNAi-mediated crop protection by foliar application is a promising approach in field trials. However, the effect of this approach on piercing-sucking pests is far from satisfactory due to the limited uptake and transport of double strand RNA (dsRNA) in plants. Therefore, there is an urgent need for more feasible and biocompatible dsRNA delivery approaches to better control piercing-sucking pests. Here, we report that foliar application of layered double hydroxide (LDH)-loaded dsRNA can effectively disrupt Panonychus citri at multiple developmental stages. MgAl-LDH-dsRNA targeting Chitinase (Chit) gene significantly promoted the RNAi efficiency and then increased the mortality of P. citri nymphs by enhancing dsRNA stability in gut, promoting the adhesion of dsRNA onto leaf surface, facilitating dsRNA internalization into leaf cells, and delivering dsRNA from the stem to the leaf via the vascular system of pomelo plants. Finally, this delivery pathway based on other metal elements such as iron (MgFe-LDH) was also found to significantly improve the protection against P. citri and the nymphs or larvae of Diaphorina citri and Aphis gossypii, two other important piercing-sucking hemipeteran pests, indicating the universality of nanoparticles LDH in promoting the RNAi efficiency and mortality of piercing-sucking pests. Collectively, this study provides insights into the synergistic mechanism for nano-dsRNA systemic translocation in plants, and proposes a potential eco-friendly control strategy for piercing-sucking pests. The fatigue crack initiation process accounts for the main portion of the high-cycle fatigue (HCF) life. Although coated superalloys are well-known to be prone to surface microcracks, the influence of the coating-substrate microstructure on the damage mechanism during HCF of the coated single crystal (SX) superalloys remains unclear. In the current study, HCF failure and interrupted tests were conducted at 900 °C under a low applied maximum stress on a PtAl-coated third-generation SX superalloy using sheet specimens. The HCF crack initiation was investigated by analyzing the evolution of surface microcracks and coating-substrate microstructure. The results show that the surface microcracks nucleation and growth are controlled by the initial microstructure of the coating-substrate region and the oxidation process during the HCF crack initiation stage. The rapid nucleation and growth of the surface microcracks are promoted by grain boundaries (GBs) in the coating and the interdiffusion zone (IDZ). Comparatively, the surface microcracks are blunted by oxidation in the SX substrate due to the loss of GBs, leading to a slower microcrack growth. However, the surface microcracks can gradually grow further into the substrate by repeating the processes as follows: oxidation at the crack tip, formation of γ′-depleted region due to oxidation, recrystallization at the γ′-depleted region and cracking along the GBs in the recrystallized γ′-depleted region. Subsequently, the surface microcracks, which reach the critical size, propagate along the slip bands during the progressive propagation stage. This study will contribute to improve the HCF durability of the PtAl-coated SX superalloy.

Abstract Background Though deep learning has consistently demonstrated advantages in the automatic interpretation of breast ultrasound images, its black-box nature hinders potential interactions with radiologists, posing obstacles for clinical deployment. Methods We proposed a domain knowledge-based interpretable deep learning system for improving breast cancer risk prediction via paired multimodal ultrasound images. The deep learning system was developed on 4320 multimodal breast ultrasound images of 1440 biopsy-confirmed lesions from 1348 prospectively enrolled patients across two hospitals between August 2019 and December 2022. The lesions were allocated to 70% training cohort, 10% validation cohort, and 20% test cohort based on case recruitment date. Results Here, we show that the interpretable deep learning system can predict breast cancer risk as accurately as experienced radiologists, with an area under the receiver operating characteristic curve of 0.902 (95% confidence interval = 0.882 – 0.921), sensitivity of 75.2%, and specificity of 91.8% on the test cohort. With the aid of the deep learning system, particularly its inherent explainable features, junior radiologists tend to achieve better clinical outcomes, while senior radiologists experience increased confidence levels. Multimodal ultrasound images augmented with domain knowledge-based reasoning cues enable an effective human-machine collaboration at a high level of prediction performance. Conclusions Such a clinically applicable deep learning system may be incorporated into future breast cancer screening and support assisted or second-read workflows.

Abstract Colorectal cancer (CRC) arises via the progressive accumulation of dysregulation in key genes including oncogenes and tumor-suppressor genes. Prostaglandin-endoperoxide synthase 2 (PTGS2, also called COX2) acts as an oncogenic driver in CRC. Here, we explored the upstream transcription factors (TFs) responsible for elevating PTGS2 expression in CRC cells. The results showed that PTGS2 silencing repressed cell growth, migration and invasion in HCT116 and SW480 CRC cells. The two fragments (499–981 bp) and (1053–1434 bp) were confirmed as the core TF binding profiles of the PTGS2 promoter. PTGS2 expression positively correlated with RUNX1 level in colon adenocarcinoma (COAD) samples using the TCGA-COAD dataset. Furthermore, RUNX1 acted as a positive regulator of PTGS2 expression by promoting transcriptional activation of the PTGS2 promoter via the 1086–1096 bp binding motif. In conclusion, our study demonstrates that PTGS2 upregulation induced by the TF RUNX1 promotes CRC cell growth, migration and invasion, providing an increased rationale for the use of PTGS2 inhibitors in CRC prevention and treatment.

Abstract Resorcinol‐formaldehyde (RF) resin represents a promising visible‐light responding photocatalyst for oxygen reduction reaction (ORR) toward H2O2 production. However, its photocatalytic ORR activity toward H2O2 generation is still unsatisfied for practical application. Herein, 3‐hydroxythiophenol‐formaldehyde (3‐HTPF) resin microspheres synthesized through polycondensation reaction between 3‐HTP and formaldehyde at room temperature and subsequent hydrothermal treatment exhibit enhanced photocatalytic ORR activity is reported. The experimental results show that the partial substitution of hydroxy group (─OH) by sulfhydryl one (─SH) through using 3‐HTP to replace resorcinol could slow the rates of nucleation and growth of the resin particles and lead to strongly π‐stacked architecture in 3‐HTPF. The introduction of ─SH group can also improve adsorption ability of 3‐HTPF to O2 molecules and enhance ORR catalytic activity of the photocatalysts. Stronger built‐in electric field, better adsorption ability to O2 molecules, and increased surface catalytic activity collectively boost photocatalytic activity of 3‐HTPF microspheres. As a result, H2O2 production rate of 2010 µm h−1 is achieved over 3‐HTPF microspheres at 273 K, which is 3.4 times larger than that obtained using RF submicrospheres (591 µm h−1). The rational substituent group modulation provides a new strategy for designing polymeric photocatalysts at the molecular level toward high‐efficiency artificial photosynthesis.

Abstract Viologen, as a type of strong electron acceptor, is prone to undergo electron transfer (ET) and change color under external stimuli. However, due to the easy aggregation of viologen molecules, they usually suffer from poor fluorescence emission in the condensed phase. Herein, a new viologen derivative of VioCl2·2Cl (12+·2Cl) was designed and synthesized, in which the fluorescence was enhanced by introducing Me‐β‐CD to weaken the interactions between viologen molecules. Then a viologen‐based host–guest supramolecule of 12+@Me‐β‐CD was obtained by electrostatic interactions. Photo‐/chemo‐responded guest 12+ supplies 12+@Me‐β‐CD, a green and dark purple caused by intramolecular and intermolecular ET. Furthermore, 12+@Me‐β‐CD displays an additional thermal responsive purple color. The triple chromic behaviors all exhibit excellent reversibility and cycling stability. As expected, 12+@Me‐β‐CD exhibits strong photoluminescence (PL) in solid–liquid dual states, presenting an improved quantum yield (Φ) from 12+ (Φs = 0.37%, Φl = 16.74%) to 12+@Me‐β‐CD (Φs = 10.45%, Φl = 25.86%), and the fluorescence intensity can be dynamically modulated by light, heat, and acid/base vapors. The multi‐responsive chromism and tunable fluorescence of 12+@Me‐β‐CD allow for potential applications in information security and smart windows.

To meet the demands for non-destructive testing of uniaxial mechanical properties of welded joints of thick-wall hydrogenation reactors, this study provides an experimental investigation on whether the spherical indentation tests (SITs) can accurately characterize the uniaxial mechanical property variations along the thickness of welded joints. The phenomenologically summarized empirical method (i.e., the Kwon method) and the semi-analytical method (i.e., the simplified-IIEM) were selected as representatives, and their reliability was judged from the viewpoints of stress-strain prediction, the inversion accuracy and repeatability of strength, and the ability to characterize the variation of uniaxial mechanical properties along the thickness of welded joints. Characteristics of the inverse predictions were analyzed, and the source of errors in each method were extensively investigated. This study provides a theoretical and technical guidance for the engineering application and promotion of SITs.

The incessant arms race between viruses and hosts has led to numerous evolutionary innovations that shape life’s evolution. During this process, the interactions between viral receptors and viruses have garnered significant interest since viral receptors are cell surface proteins exploited by viruses to initiate infection. Our study sheds light on the arms race between the MDA5 receptor and 5’ppp-RNA virus in a lower vertebrate fish, Miichthys miiuy. Firstly, the frequent and independent loss events of RIG-I in vertebrates prompted us to search for alternative immune substitutes, with homology-dependent genetic compensation response (HDGCR) being the main pathway. Our further analysis suggested that MDA5 of M. miiuy and Gallus gallus, the homolog of RIG-I, can replace RIG-I in recognizing 5’ppp-RNA virus, which may lead to redundancy of RIG-I and loss from the species genome during evolution. Secondly, as an adversarial strategy, 5’ppp-RNA SCRV can utilize the m6A methylation mechanism to degrade MDA5 and weaken its antiviral immune ability, thus promoting its own replication and immune evasion. In summary, our study provides a snapshot into the interaction and coevolution between vertebrate and virus, offering valuable perspectives on the ecological and evolutionary factors that contribute to the diversity of the immune system.

Miiuy croaker, Miichthys miiuy is an ecologically important teleost species which is widely distributed in southeast coast of China. In this study, we present a chromosomal-scale genome assembly of the miiuy croaker which is an important Sciaenidae fish and economical species. We adopted Oxford Nanopore and Hi-C sequencing techniques to achieve an assembly with high accuracy and completeness. The investigation of genome characteristic and functional features may provide insights into the study of phylogenetic diversity of miiuy croaker. This study can also be beneficial to improve molecular assisted breeding techniques. Moreover, it can be a great resource to better conduct further study of other sciaenids.

Non-targeted screening (NTS) technology has been showing significant potential in identifying contaminants of emerging concern (CECs) in water and has attracted great attention in academia in recent years. It is a method that analyzes samples without pre-selecting substances, enabling the detection and identification of unknown compounds, which is crucial for environmental health and public protection. This study uses the Bibliometrix package in R 4.4.1 and CiteSpace 6.3.R1 software to statistically analyze 589 relevant publications from the Web of Science Core Collection from 2007 to 2024. Our work concentrates on NTS of water bodies; thus, articles that only analyze water sediments without analyzing the water were not considered for inclusion. By conducting a quantitative analysis and visualizing the publication trends, countries, authors, journals, and keywords, the present study identifies research hotspots, compositions, and paradigms within this field, trying to analyze the horizontal and vertical development trends and structural evolution of the research area. The research found that the application of NTS in water pollution studies has progressed through three phases: theoretical exploration, rapid development, and steady progress. From the national level, China leads with the highest number of publications (131), followed by Germany (105), Spain (50), and the United States (39). The top three authors by publication volume are J. Hollender, Nikolaos S. Thomaidis, and Emma L. Schymanski, while the top three by citation count are J. Hollender, Emma L. Schymanski, and M. Krauss. However, international collaboration between countries and researchers still remains an area for improvement. Science of the Total Environment is the journal with the highest number of publications (81), and Environmental Science & Technology holds the highest number of citations. Research on NTS methodologies, suspect screening, and health risk assessments are hot topics in the academic community. Future research is expected to be multidisciplinary, with emerging hotspots likely to focus on including the identification of novel pollutants through NTS, toxicity assessments of biotransformed compounds, and the health impacts and mechanisms of related compounds.

Abstract Background Periodontitis is a chronic oral inflammatory disease that seriously affects people's quality of life. The purpose of our study was to investigate the correlation between the systemic immune inflammation index (SII) and periodontitis by utilizing a large national survey. This will establish a reference for the early identification and management of periodontitis. Methods This study comprised the adult US population who participated in a national periodontitis surveillance project during the six years from 2009 to 2014. Through the utilization of univariate and multivariate weighted logistic regression, we investigated the correlation between the systemic immune inflammation index and periodontitis. Additionally, we employed sensitivity analyses to evaluate the robustness of our findings. Results The study involved 10,366 participants with an average age of 51.00 years, of whom 49.45% were male (N = 5126) and 50.55% were female (N = 5240). The prevalence of periodontitis is estimated to be about 38.43% in the US adults aged 30 or older population. Our logistic regression models indicated a positive association between a SII higher than 978 × 109/L and periodontitis. The elder group (aged 50 or older) with SII higher than 978 × 109/L demonstrated a significant correlation with periodontitis in the fully adjusted model (Odds Ratio [OR] = 1.409, 95% Confidence Interval [CI] 1.037, 1.915, P = 0.022). However, there is no statistical difference among adults aged 30 to 50. The robustness of our findings was confirmed through sensitivity analyses. Conclusions Our study highlights that SII is associated with periodontitis in a nationally representative sample of US adults. And the SII is significantly associated with a high risk of periodontitis in individuals aged 50 or older.

In female animals, metabolic homoeostasis and reproductive fitness are critical to population expansion. The trade-off between lipid storage and reproduction inevitably occurs. However, most studies have focused on the complex network of relationships between reproductive and metabolic physiology at the transcriptional level. In this study, we identified a microRNA, miR-2b-2-5p, in a highly invasive quarantine pest, Bactrocera dorsalis. Knockdown of miR-2b-2-5p by antagomiR microinjection impaired ovarian development, reduced fecundity, and decreased triglyceride (TAG) storage in the fat body, whereas overexpression of miR-2b-2-5p by injection of its mimic caused reproductive defects similar to knockdown but increased TAG. Bioinformatics analysis and dual luciferase assay indicated that cyclic AMP response element (CRE)-binding protein (CREB) was the target gene of miR-2b-2-5p. RNAi-mediated knockdown of CREB led to excessive lipid storage and reproductive defects. Further starvation treatment revealed that miR-2b-2-5p functions by fine-tuning CREB expression in response to dietary stimuli. These results suggest that miR-2b-2-5p acts as a monitor to regulate CREB mRNA levels in the fat body, maintaining lipid homoeostasis and keeping the reproductive system on track. Thus, our study not only provides new insights into the interaction between metabolism and reproduction at the posttranscriptional level in B. dorsalis, but also providing a potential eco-friendly control strategy (RNAi-based biopesticides targeting essential miRNAs) for this notorious agricultural pest.

Abstract The assembly of W and Y chromosomes poses significant challenges in vertebrate genome sequencing and assembly. Here, we successfully assembled the W chromosome of Verasper variegatus with a length of 20.48 Mb by combining population and PacBio HiFi sequencing data. It was identified as a young sex chromosome and showed signs of expansion in repetitive sequences. The major component of the expansion was Ty3/Gypsy. The ancestral Osteichthyes karyotype consists of 24 protochromosomes. The sex chromosomes in four Pleuronectiformes species derived from a pair of homologous protochromosomes resulting from a whole-genome duplication event in teleost fish, yet with different sex-determination systems. V. variegatus and Cynoglossus semilaevis adhere to the ZZ/ZW system, while Hippoglossus stenolepis and H. hippoglossus follow the XX/XY system. Interestingly, V. variegatus and H. hippoglossus derived from one protochromosome, while C. semilaevis and H. stenolepis derived from another protochromosome. Our study provides valuable insights into the evolution of sex chromosomes in flatfish and sheds light on the important role of whole-genome duplication in shaping the evolution of sex chromosomes.

Hepatic stellate cells (HSCs) represent a significant component of hepatocellular carcinoma (HCC) microenvironments which play a critical role in tumor progression and drug resistance. Tumor-on-a-chip technology has provided a powerful in vitro platform to investigate the crosstalk between activated HSCs and HCC cells by mimicking physiological architecture with precise spatiotemporal control. Here we developed a tri-cell culture microfluidic chip to evaluate the impact of HSCs on HCC progression. On-chip analysis revealed activated HSCs contributed to endothelial invasion, HCC drug resistance and natural killer (NK) cell exhaustion. Cytokine array and RNA sequencing analysis were combined to indicate the iron-binding protein LIPOCALIN-2 (LCN-2) as a key factor in remodeling tumor microenvironments in the HCC-on-a-chip. LCN-2 targeted therapy demonstrated robust anti-tumor effects both in vitro 3D biomimetic chip and in vivo mouse model, including angiogenesis inhibition, sorafenib sensitivity promotion and NK-cell cytotoxicity enhancement. Taken together, the microfluidic platform exhibited obvious advantages in mimicking functional characteristics of tumor microenvironments and developing targeted therapies.

For near-β high-strength titanium alloys, rheological softening is the key step during thermal deformation to determine the microstructure and mechanical properties. Nevertheless, the intrinsic softening mechanism during the rheological stage is still an open question due to the special complexity of the process. In this study, the deformation process of near-β titanium alloy TC18 (Ti–5Al–5Mo–5V–1Cr–1Fe, wt.%) in a dual-phase region at a high strain rate was investigated. The micromechanical behavior of TC18 alloy during thermal deformation at 785 °C and 0.1/s was revealed by using in-situ high-energy X-ray diffraction (HEXRD). The stress partitioning and microstructure evolution mechanism for this alloy were discussed in detail. It has been confirmed that the main softening mechanisms of TC18 alloy during thermal deformation are dynamic recovery (DR) and continuous dynamic recrystallization (cDRX). The stress partitioning between constituent phases featured by higher stress in the α phase than in the β phase induces an inhomogeneous strain field near α phase lamellae to promote the dislocation pile-up in subgrains. The cDRX process of β grains is composed of dislocation slipping, DR and subgrains formation, and transformation of recrystallized grains with high-angle grain boundaries (HAGB). Schmid factor and micro stress have a remarkable effect on dynamic recrystallization (DRX) behavior. The soft β//LD oriented grains are more likely to undergo cDRX than the hard β//LD oriented grains. This work may have implications for understanding the deformation mechanism and provide a fundamental basis for selecting appropriate processing technology for near-β titanium alloys.

Abstract The mRNA therapy is widely used in the treatment of diseases due to its efficient characteristics, and the COVID-19 vaccine is the application of mRNA therapy. However, due to the instability of mRNA, mRNA vaccines often need lots of modifications to ensure its stability. Recent research shows that circRNA with stable RNA structure can encode protein, which provides a new direction for mRNA therapy. Here, we discovered a novel circRNA (circMIB2) derived from E3 ubiquitin-protein ligase MIB2 (MIB2) gene in lower vertebrate fish, which can translate into a 134 amino acid protein (MIB2-134aa) through m6A modification, and is involved in innate immunity. MIB2-134aa is completely consistent with the amino acid sequence of the two domains of host gene MIB2 protein; host gene MIB2 can target TRAF6 through the two domains and inhibit the innate immune response by promoting the ubiquitination degradation of the K11-link of TRAF6, MIB2-134aa also targets TRAF6 through these same domains. Interestingly, MIB2-134aa greatly reduced the degradation of TRAF6 by its host gene MIB2. More importantly, we found that circRNA therapy of circMIB2 can significantly inhibit the colonization of Vibrio anguillarum in zebrafish, and it provides a new direction for the treatment of pathogenic diseases of fish.

Cuticular wax is the first line of structural defense for plants against external stresses. This study investigated the effects of melatonin (MT) on chemical composition and accumulation profile of wax, as well as fruit quality of rabbiteye blueberry during storage. The results indicated a significant reduction in the overall wax content during storage. Nevertheless, MT effectively delayed the decline, with a higher amount of 9.8% and 15.17% in the treated ‘Baldwin’ and ‘Garden Blue’ compared to their respective controls at 21st day of storage. The wax composition significantly varied depending on storage time, MT treatment, and cultivars. Additionally, MT markedly improved the fruit quality of rabbiteye blueberries. Correlation analysis revealed water loss and decay rates were negatively correlated with triterpenoids and fatty acids. Taken together, this study highlights the positive effects of post-harvest MT application on shelf life and fruit quality of blueberry by modifying the wax profile during storage.

Abstract Durable superhydrophobic anti‐erosion/anticorrosion coatings are highly demanded across various applications. However, achieving coatings with exceptional superhydrophobicity, mechanical strength, and corrosion resistance remains a grand challenge. Herein, a robust microstructure coating, inspired by the cylindrical structures situated on the surface of conch shell, for mitigating erosion and corrosion damages in gas transportation pipelines is reported. Specifically, citric acid monohydrate as a pore‐forming agent is leveraged to create a porous structure between layers, effectively buffering the impact on the surface. As a result, the coating demonstrates remarkable wear resistance and water repellency. Importantly, even after abrasion by sandpaper and an erosion loop test, the resulting superhydrophobic surfaces retain the water repellency. The design strategy offers a promising route to manufacturing multifunctional materials with desired features and structural complexities, thereby enabling effective self‐cleaning and antifouling abilities in harsh operating environments for an array of applications, including self‐cleaning windows, antifouling coatings for medical devices, and anti‐erosion/anticorrosion protection, among other areas.

Edwardsiella tarda is a major causative pathogen of bacterial ascites in Japanese flounder, leading to massive economic losses, and the discovery of molecular markers linked to disease resistance is an effective strategy in resistance breeding programs. The Rho GTPase family comprises small proteins with a molecular weight of 20~30 kDa. Rho GTPase family members are involved in diverse cellular processes, such as cytoskeleton, cell adhesion, vesicle transport, and proliferation. In addition, they play pivotal roles in infection by different pathogens. Rho-related GTP-binding protein Rho6 (Rnd1), a member of the Rho-GTPase family, participates in various biological functions, including neural junction formation, axonal extension, tumorigenesis, neuronal function, and apoptosis. Some members of the Rho family, such as Rac1 and Rac2, regulate immune response in grass carp, large yellow croaker, zebrafish, and half-smooth tongue sole. However, the function of Rnd1 in fish is poorly understood. Japanese flounder (Paralichthys olivaceus) is greatly affected by E. tarda infections during the breeding process. In previous studies, whole-genome sequencing and assembly of Japanese flounder were performed, and subsequently, various disease resistance genes were screened to support the improvement of Japanese flounder germplasm resources. To study the role of Pornd1 in resistance against E. tarda infection in Japanese flounder, Pornd1 was cloned and identified using PCR. The full-length Pornd1 cDNA was 699 bp, containing an open reading frame encoding a 232-amino acid protein. The predicted molecular weight of PoRnd1 was 26 kDa. Sequence and homology analyses showed that the Rnd1 protein harbors a Rho-GTP superfamily structural domain, which is highly conserved in various species. PoRnd1 shares the highest homology with Rnd1 from Hippoglossus hippoglossus (98.28%). On phylogenetic tree, PoRnd1 was clustered with Rnd1 from other fish species. The single-nucleotide polymorphism (SNP) locus associated with E. tarda resistance is located at 4 575 720 bp on chromosome 14 of Japanese flounder. The frequency of the T allele in disease-resistant families (freqT=0.92) was significantly higher than that in susceptible families (freqT=0.20). The SNP was located at the 2nd intron of Pornd1. Real-time quantitative PCR was employed to characterize the expression profiles of Pornd1 in the tissues of healthy and E. tarda-infected fish. Pornd1 expression was the highest in the heart, followed by the liver, kidney, head kidney, and spleen, but its expression was low in the skin, blood, gills, and muscle. In E. tarda-infected fish, the expression of Pornd1 mRNA decreased after 6 h, then gradually increased, and subsequently reached the highest level after 48 h in the liver, kidney, and spleen. Pornd1 expression in the kidney and spleen in the 48 h group was significantly higher than that in the 6 and 12 h groups. Furthermore, Pornd1 expression in the liver of resistant families was significantly higher than that in susceptible families. Based on its His tag, the PoRnd1 recombinant protein was purified using an Ni column and subjected to SDS-PAGE. The target band of PoRnd1 at 32 kDa was observed in the gel after Coomassie Blue staining. The PoRnd1 recombinant protein (0.5 mg/mL) was used to study antibacterial activity through the Oxford cup assay. PoRnd1 significantly inhibited the growth of Staphylococcus aureus, Escherichia coli, E. tarda, and Vibrio harveyi. In summary, Pornd1 may be closely linked to disease resistance in Japanese flounder and can thus serve as an effective gene marker for disease resistance breeding. Our findings provide a theoretical basis for further elucidating the molecular mechanisms of immunity in Japanese flounder.

Background: Organophosphate flame retardants (OPFRs) are compounds with a wide range of industrial and commercial applications and are mainly used as flame retardants and plasticizers. The global consumption of OPFRs has risen rapidly in recent decades, and they have been widely detected in environmental media. Unfortunately, OPFRs have been associated with many adverse health outcomes. The issue of the health risks of OPFRs is attracting increasing attention. Therefore, there is a need to review the current state of research and trends in this field to help researchers and policymakers quickly understand the field, identify new research directions, and allocate appropriate resources for further development of the OPFR health risk research field. Methods: This study statistically analyzed 1162 relevant publications included in the Web of Science Core Collection from 2003–2023. The internal and external features of the literature, such as publication trends, countries, authors, journals, and keywords, were quantitatively analyzed and visually presented to identify the research hotspots, compositions, and paradigms of the field and to horizontally and vertically analyze the development trends and structural evolution of the field. Results: The development of the field can be divided into three stages, and the field entered a period of rapid development in 2016. China (649 papers) is the most prolific country, followed by the United States (188 papers). The authors STAPLETON HM and WANG Y have the highest combined impact. International collaboration between countries and researchers still needs to be strengthened. Science of The Total Environment is the most frequently published journal (162 papers), and Environmental Science and Technology is the most frequently cited journal (5285 citations). Endocrine disruption, developmental toxicity, and neurotoxicity are the health effects of greatest interest. Conclusions: Future research is expected to be multidisciplinary, and research hotspots may involve a comprehensive assessment of OPFR exposure in the population, exploration of the mechanisms of endocrine-disrupting effects and in vivo metabolic processes, and examination of the health effects of OPFR metabolites.

Cadmium (Cd), a prevalent environmental contaminant, exerts widespread toxic effects on human health through various biochemical and molecular mechanisms. This review encapsulates the primary pathways through which Cd inflicts damage, including oxidative stress induction, disruption of Ca2+ signaling, interference with cellular signaling pathways, and epigenetic modifications. By detailing the absorption, distribution, metabolism, and excretion (ADME) of Cd, alongside its interactions with cellular components such as mitochondria and DNA, this paper highlights the extensive damage caused by Cd2+ at the cellular and tissue levels. The role of Cd in inducing oxidative stress—a pivotal mechanism behind its toxicity—is discussed with emphasis on how it disrupts the balance between oxidants and antioxidants, leading to cellular damage and apoptosis. Additionally, the review covers Cd’s impact on signaling pathways like Mitogen-Activated Protein Kinase (MAPK), Nuclear Factor kappa-light-chain-enhancer of activated B cells (NF-κB), and Tumor Protein 53 (p53) pathways, illustrating how its interference with these pathways contributes to pathological conditions and carcinogenesis. The epigenetic effects of Cd, including DNA methylation and histone modifications, are also explored to explain its long-term impact on gene expression and disease manifestation. This comprehensive analysis not only elucidates the mechanisms of Cd toxicity but also underscores the critical need for enhanced strategies to mitigate its public health implications.

Conventional moiré magnifier suffers from high manufacturing cost and poor scalability due to the stringent requirement on alignment between the microfocusing element array and micropattern array (MPA). By superposition of the microreflective element array and MPA together to form hierarchical microstructure, not only the alignment can be done during mold fabrication process, which is easily scalable and compatible to semiconductor, but also the synthetically magnified image is visible as viewed from both front and back side. The investigation of the moiré magnifier with hierarchical microstructure unveils new imaging phenomenon in moiré imaging device, and thereby offers a facile and low-cost platform to address challenges to open up a potential avenue in micro-optic involved three-dimensional display or anti-counterfeiting applications.

Controlled immune response, enhanced osteogenesis, and antibacterial effect are essential for successful implanted biomaterials in the long-term. However, the previous research only considers one of the above-mentioned materials’ properties, and the potential synergetic effects are still unknown. Inspired by the mussel adhesion-mediated ions-coordinated strategy, a multifunctional coating integrated with bioactive Sr2+ and antibacterial Cu2+ was fabricated the multifunctional PEEK implants (PEEK-PDA-Sr/Cu). Compared to the uncoating PEEK, the PEEK implants with a multifunctional coating can regulate the M1 subtypes of macrophages to M2 subtypes due to the presence of Sr2+, which further promote the osteogenic differentiation of BMSCs. Even under the bacterial infection environment, the PEEK implants with a multifunctional coating can still enhance the osseointegration of bone implants due to the antibacterial properties of Cu2+. Taken together, PEEK implants with a multifunctional coating can provide an osteogenic immune microenvironment with enhanced osteogenesis and antibacterial property, which is essential for successful implantation in the long-term.

Tumor cell extravasation across endothelial barrier has been recognized as a pivotal event in orchestrating metastasis formation. This event is initiated by the interactions of extravasating tumor cells with endothelial cells (ECs). Therefore, targeting the crosstalk between tumor cells and ECs might be a promising therapeutic strategy to prevent metastasis. In this study, we demonstrated that Rh1, one of the main ingredients of ginseng, hindered the invasion of breast cancer (BC) cells as well as diminished the permeability of ECs both in vitro and in vivo, which was responsible for the attenuated tumor cell extravasation across endothelium. Noteworthily, we showed that ECs were capable of inducing the epithelial-mesenchymal transition (EMT) and invadopodia of BC cells that are essential for tumor cell migration and invasion through limiting the nuclear translocation of hematopoietically expressed homeobox (HHEX). The decreased nuclear HHEX paved the way for initiating the CCL20/CCR6 signaling axis, which in turn contributed to damaged endothelial junctions, uncovering a new crosstalk mode between tumor cells and ECs. Intriguingly, Rh1 inhibited the kinase activity of casein kinase II subunit alpha (CK2α) and further promoted the nuclear translocation of HHEX in the BC cells, which resulted in the disrupted crosstalk between chemokine (C-C motif) ligand 20 (CCL20) in the BC cells and chemokine (C-C motif) receptor 6 (CCR6) in the ECs. The prohibited CCL20-CCR6 axis by Rh1 enhanced vascular integrity and diminished tumor cell motility. Taken together, our data suggest that Rh1 serves as an effective natural CK2α inhibitor that can be further optimized to be a therapeutic agent for reducing tumor cell extravasation.

The protein-coding ability of circRNAs has recently been a hot topic, but the role of protein-coding circRNAs in antiviral innate immunity of teleost fish has rarely been reported. Here, we identified a novel circRNA, termed circMORC3, derived from Microrchidia 3 (MORC3) gene in Miichthys miiuy. circMORC3 can inhibit the expression of antiviral cytokines. In addition, circMORC3 encodes a novel peptide with a length of 84 amino acids termed MORC3-84aa. MORC3-84aa not only significantly inhibited TRIF-mediated activation of IRF3 and NF-κB signaling pathways, but also effectively suppressed the expression of antiviral cytokines triggered by RNA virus Siniperca chuatsi rhabdovirus (SCRV). We found that MORC3-84aa directly interacted with TRIF and negatively regulated TRIF protein expression. In addition, host gene MORC3 attenuates SCRV-induced IFN and ISG expression. Mechanistically, MORC3-84aa promotes autophagic degradation of TRIF by enhancing K6-linked ubiquitination and inhibits TRIF-mediated activation of the type I interferon signaling pathway. And the host gene MORC3 not only repressed IRF3 protein expression but also inhibited IRF3 phosphorylation levels. Our study shows that circMORC3 and host gene MORC3 played a synergistic role in viral immune escape.

Summary: The losses caused by Vibrio infections in the aquaculture industry are challenging to quantify. In the face of antibiotic resistance, a natural and environmentally friendly alternative is urgently needed. In this study, we identify E3 ubiquitin-protein ligase RNF103 (rnf103) as a crucial target involved in immune evasion by Vibrio anguillarum. Our research demonstrates that Rnf103 promotes immune escape by inhibiting Traf6. Interestingly, we discover a circular RNA (circRNA), circRnf103, formed by reverse splicing of the Rnf103 gene. Predictive analysis and experimentation reveal that circRnf103 encodes Rnf103-177aa, a protein that competes with Rnf103 and binds to Traf6, preventing its degradation. Notably, circRnf103 therapy induces Rnf103-177aa protein production in zebrafish. In zebrafish models, circRnf103 exhibits significant effectiveness in treating V. anguillarum infections, reducing organ burden. These findings highlight the potential of circRNA therapy as a natural and innovative approach to combat infectious diseases sustainably, particularly in aquaculture and environmental management.

In aquaculture, selective breeding for survival till harvest have become an alternative strategy for improving disease resistance and production. However, knowledge of genetic parameters of harvest survival, e.g., heritability and genetic correlations between survival and growth rate traits, is still scarce. The aims of this study were to estimate genetic parameters for harvest survival and growth rate traits under commercial farming conditions in olive flounder (Paralichthys olivaceus). Harvest survival was defined as a binary trait; growth traits were measured as average daily gain (ADG), specific growth rate (SGR), daily growth coefficient (DGC) and body weight (BW). Data from a population of 241 full-sib families (involving 39,904 individuals, four generations) were used. Heritabilities of survival were low but significant, which were 0.15 ± 0.04 and 0.22 ± 0.01 on observed and underlying scale, respectively. Heritability estimates for ADG, SGR and DGC were medium to high, which were 0.33 ± 0.06, 0.83 ± 0.07, 0.58 ± 0.07, respectively. While the heritability of BW is of low magnitude (0.17 ± 0.08). The genetic correlations between harvest survival and three growth rate traits (i.e., ADG, SGR and DGC) were very strong (ranging from 0.66 to 0.79), which is an exciting result. However, the genetic correlation between harvest survival and BW was much lower (0.17 ± 0.08). These results suggest that selection for harvest survival would consequentially result in concomitant increase of growth rate, and vice versa. Our findings revealed novel insights into the genetic improvement of growth rate and harvest survival through genetic selection in olive flounder.

The microstructure and corrosion property of super 13Cr martensitic stainless steel (S13CrMSS) processed by quenching, one-step tempering and two-step tempering were investigated by XRD, SEM, EBSD and TEM-EDS analysis. It has been found that volume fraction of the reversed austenite in the sample treated by two-step tempering treatment was 13.5 vol. % up to the maximum. The reversed austenite almost always nucleated and distributed in martensite laths boundary during tempering and the diffusion of Ni, Mn and C promoted its growth. Enrichment of such elements as Ni, Mn, C and the ultrafine size of the reversed austenite were considered to be the main factor that accounted for the stability of the reversed austenite. At the same time, martensite and reversed austenite satisfy the (1 1¯ 1)γ // (011)α' and [011]γ // [1¯1¯ 1]α' Kurdjumov-Sachs (K–S) orientation relationship. Potentiodynamic polarization curves and electrochemical impedance spectroscopy indicated that two-step tempered specimen exhibited a higher pitting potential, lower corrosion current density, thicker passivation film and superior pitting resistance than the quenching specimen (without reversed austenite) as well as one-step tempering specimen (3.6 vol.%). It has been confirmed that the reversed austenite has a beneficial effect on the corrosion property of S13CrMSS.

The blackspotted croaker (Protonibea diacanthus) is an endangered coastal marine fish. It is also a valuable species that is cultured on the southeast coast of China. While some genetic studies have been conducted to protect this species, genomic resources are lacking. Here, we report a chromosome-scale assembly of P. diacanthus genome by high-depth genome sequencing, assembly, and annotation. The genome scale was 635.69 Mb with contig and scaffold N50 length of 3.33 Mb and 25.60 Mb, respectively. Hi-C scaffolding of the genome resulted in 24 chromosomes of 94.15% total genome. We predicted 23,971 protein-coding genes. In addition, we constructed a phylogenetic tree using 2755 single-copy gene families and identified 462 unique gene families in P. diacanthus genome compared to three other sciaenids. What's more, from the analysis of gene families, we found that several gene families related to innate immunity were significantly expanded in the blackspotted croaker genome compared to other teleost genomes. The high-quality genome can improve our understanding of the molecular mechanisms behind economically valuable traits and provide insights into characteristics of the immune system.

Objective: Yinchen Sini decoction (YCSND), a traditional Chinese medicine (TCM) formula, plays a crucial role in the treatment of liver disease. However, the bioactive constituents and pharmacological mechanisms of action remain unclear. The present study aimed to reveal the molecular mechanism of YCSND in the treatment of acute liver injury (ALI) using integrated network analysis and metabolomics.Methods: Ultra-high-performance liquid chromatography coupled with Q-Exactive focus mass spectrum (UHPLC-QE-MS) was utilized to identify metabolites in YCSND, and high-performance liquid chromatography (HPLC) was applied to evaluate the quality of four botanical drugs in YCSND. Cell damage and ALI models in mice were established using CCl4. 1H-NMR metabolomics approach, along with histopathological observation using hematoxylin and eosin (H&E), biochemical measurements, and reverse transcription quantitative real-time PCR (RT-qPCR), was applied to evaluate the effect of YCSND on CCl4- induced ALI. Network analysis was conducted to predict the potential targets of YCSND in ALI.Result: Our results showed that 89 metabolites in YCSND were identified using UHPLC-QE-MS. YCSND protected against ALI by reducing the levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), and malondialdehyde (MDA) contents and increasing those of superoxide dismutase (SOD), and glutathione (GSH) both in vivo and in vitro. The 1H-NMRmetabolic pattern revealed that YCSND reversed CCl4-induced metabolic abnormalities in the liver. Additionally, the Kyoto Encyclopedia of Genes and Genome (KEGG) pathway enrichment analysis identified five pathways related to liver injury, including the PI3K-AKT, MAPK, HIF-1, apoptosis, and TNF signaling pathways. Moreover, RT-qPCR showed YCSND regulated the inflammatory response (Tlr4, Il6, Tnfα, Nfκb1, Ptgs2, and Mmp9) and apoptosis (Bcl2, Caspase3, Bax, and Mapk3), and inhibited PI3K-AKT signaling pathway (Pi3k and Akt1). Combined network analysis and metabolomics showed a link between the key targets (Tlr4, Ptgs2, and Mmp9) and vital metabolites (choline, xanthine, lactate, and 3-hydroxybutyric acid) of YCSND in ALI.Conclusion: Overall, the results contribute to the understanding of the therapeutic effects of YCSND on ALI, and indicate that the integrated network analysis and metabolomics could be a powerful strategy to reveal the pharmacological effects of TCM.

Abstract The protein Talin1 encoded by the TLN1 gene is a focal adhesion-related protein that binds to various cytoskeletal proteins and plays an important role in cell adhesion and movement. Recent studies have shown that it is overexpressed in prostate cancer, liver cancer, and oral squamous cell carcinoma, and is closely related to tumor progression and metastasis. This study integrated bioinformatics and functional analysis to reveal the prognosis and potential functions of TLN1 in AML. The results showed that the expression level of TLN1 was abnormally increased in AML and localized in the cell membrane and cytoplasm, and TLN1 is a significant prognostic indicator of overall survival (OS). Enrichment analysis of related genes showed that TLN1 is related to neutrophil mediated immunity, neutrophil activation and may regulate important signal pathways in hematological tumors including tyrosine kinase receptor, FLT3 and PIK3/AKT. The PPI network shows that TLN1 and MYH9 may be involved in the process of AML tumors together with PIP5K1C, ROCK1, S100A4, MY01A and WAC. Immune infiltration analysis explains that TLN1 is associated with multiple immune cells and may be an important immune marker in AML. Furthermore, molecular biology experiments confirmed that TLN1 is related to the proliferation, differentiation and cycle of AML cells. Silencing TLN1 can inhibit the proliferation of AML cells and promote differentiation through the Talin1/P-AKT/CREB signaling pathway.

This study investigated the association between the enactment of the new policy and changes in the number of births, and health-related birth outcomes. Methods We used difference-in-difference model to analyse the birth record data in Pudong New Area, Shanghai.The design is descriptive before-and-after comparative study. Results The data covered three policy periods: the one-child policy period (January 2008 to November 2014); the partial two-child policy period (December 2014 to June 2016); the universal two-child policy period (July 2016 to December 2017). There was an estimate of 7656 additional births during the 18 months of the implementation of the universal two-child policy. Summary: Autophagy and apoptosis are two recognized mechanisms of resistance to bacterial invasion. However, bacteria have likewise evolved the ability to evade immunity. In this study, we identify ACKR4a, a member of an atypical chemokine receptor family, as a suppressor of the NF-κB pathway, which cooperates with Beclin-1 to induce autophagy to inhibit NF-κB signaling and block apoptosis, facilitating Vibrio harveyi infection. Mechanistically, V. harveyi-induced Ap-1 activates ACKR4a transcription and expression. ACKR4a forms a complex with Beclin-1 and MyD88, respectively, inducing autophagy and transporting MyD88 into the lysosome for degradation to suppress inflammatory cytokine production. Meanwhile, ACKR4a-induced autophagy blocks apoptosis by inhibiting caspase8. This study proves for the first time that V. harveyi uses both autophagy and apoptosis to evade innate immunity, suggesting that V. harveyi has evolved the ability to against fish immunity.

IntroductionFlumatinib is a novel, oral breakpoint cluster region-abelson (BCR-ABL) tyrosine kinase inhibitor that has demonstrated manageable safety and promising efficacy in patients with newly diagnosed chronic phase (CP) chronic myeloid leukemia (CML). MethodsThis study evaluated the pharmacokinetic (PK) profiles of flumatinib mesylate tablets at a dose of 400 mg and 600 mg in patients with CML-CP. The study was registered at chictr.org Identifier (ChiCTR2100044700). In this open-label, pharmacokinetic study, eligible patients were administered a single-dose of flumatinib 400 mg or 600 mg on day 1, followed by 2-day washout and 8 consecutive days of once-daily administration. Serial plasma samples were assayed for flumatinib and its metabolites (N-demethylate metabolite M1 and amide-bond hydrolytic metabolite M3).ResultsTwenty-nine patients were assigned to flumatinib 400 mg (n=14) or 600 mg (n=15). Serum concentrations of flumatinib reached maximum measured plasma concentration (Cmax) at a median time of 2 hours after each single dose, and then eliminated slowly with a mean apparent terminal disposition half-life (t1/2) from 16.0 to 16.9 hours. Following single- and multiple-dose administration, flumatinib exposure (Cmax, area under the concentration-time curve from 0 to t hours (AUC0-t), area under the concentration-time curve from 0 hours to infinity (AUC0-∞)) increased in an approximately dose-proportional manner. There was approximately 4.1- and 3.4- fold drug accumulation at steady-state after multiple-dose administration at 400 mg and 600 mg, respectively. The drug-related AEs associated with both treatments were primarily low-grade and tolerable events.ConclusionAnalysis of PK parameters indicated that flumatinib exposure increased in an approximately dose-proportional manner. Further research needs to be conducted in a large sample-size study.

Background: Natural killer (NK) cells play an indispensable role in anti-tumor immunity. TGF-β1 is the main accomplice of tumor immune escape, inhibiting tumor immunity mediated by NK cells. It is reported that Salvia miltiorrhiza can promote the immune killing effect of NK cells. In this study, Tanshinol, a water-soluble active component of Salvia miltiorrhiza, was used to investigate its effect on the inhibition of NK cell functions mediated by TGF-β1 in breast cancer. Methods: We constructed a mouse model of breast cancer by tail vein injection, H&E staining and ELISA were used to verify the role of TGF-β1 and the effects of Tanshinol on breast cancer and NK cells. In vitro, we used CCK8 and cytotoxicity assays to preliminarily evaluate the effect of Tanshinol on the anti-tumor effect of NK cells intervention by TGF-β1. We explored the killing activity of NK cells and related signal pathways by immunofluorescence imaging technology, RT-PCR, ELISA and flow cytometry. Also, Western blot, RT-PCR and immunofluorescence experiments were applied to investigate the expression level of the natural killer group 2 member D (NKG2D)-NKG2D ligands (NKG2DL) signal axis, and combined with immunoprecipitation, to detect the formation of NKG2D-DNAX-activating protein of 10 kD (DAP10) complex. Results: TGF-β1 played a role in promoting lung metastasis of breast cancer and inhibiting the secretion of cytotoxic mediators from NK cells, but Tanshinol could reverse it. High-dose Tanshinol also significantly optimized the survival rate of tumor-bearing mice. TGF-β1 could destroy the NKG2D-NKG2DL axis, down-regulate the expression and nuclear accumulation of p-smad2/3. Moreover, TGF-β1 inhibited the activation of PI3K-ERK1/2-PLCγ2 signaling pathway that is related to the degranulation of NK cells, and diminished the expression of degranulation marker CD107a and the release of anti-tumor cytotoxic killing medium of NK cells. However, Tanshinol was able to interfere with the negative regulation of TGF-β1 on the functions of NK cells, mainly through promoting the expression of NKG2D and its molecular chaperone DAP10, thereby propelling the formation of NKG2D-DAP10 complex. Conclusions: Collectively, Tanshinol enables NK cells to activate and release multiple killing mediators to carry out immune attacks on tumor cells.

Objectives: To explore the optical coherence tomography (OCT) biomarkers to predict the transition to chronic central serous chorioretinopathy (cCSC) after retinal laser photocoagulation. Methods: Patients enrolled in this study were from a 12-week clinical trial comparing the efficacy and safety of subthreshold micropulse laser (SML) with threshold conventional laser (TCL) for CSC and had extended follow-up for more than 1 year. They were divided into two groups, transited to cCSC group (cCSC group) and did not transited to cCSC group (non-cCSC group) according to fundus examination at the extended follow-up. Collect the best-corrected visual acuity (BCVA) and OCT characteristics of patients at baseline and 12 weeks after laser treatment. Results: Twenty-seven patients were enrolled (42.6 ± 7.7 years old), and duration of follow-up was 178.9 ± 88.8 (57.0–312.0) weeks. Nine patients (33.3%) were assigned to cCSC group, and the other 18 patients (66.7%) were assigned to the non-cCSC group. Twelve weeks after the laser treatment, subretinal fluid (SRF) of 15 patients (83.3%) in non-cCSC group and 5 patients (55.6%) in cCSC group absorbed completely; the height of SRF had statistical difference between two groups ( p = 0.035); rough RPE was less common in cCSC group ( p = 0.030); hyper reflective mass (HRM) was more common in cCSC group ( p = 0.024); more number of hyper reflective foci (HRF) in outer segment of photoreceptor layer were detected in cCSC group ( p = 0.035). From baseline to 12 weeks after laser treatment, the number of HRF in outer segment photoreceptor layer did not change significantly in cCSC group ( p = 0.665) but decreased significantly in non-cCSC group ( p = 0.000). A total of five patients suffered binocular CSC, three of them in the non-cCSC group occurred later than the other two in the cCSC group (129.9–278.3 weeks vs 96.1–114.9 weeks after baseline). Conclusion: SRF, rough RPE, HRM, and number of HRF in outer segment photoreceptor layer 12 weeks after laser treatment, and change in the number of HRF in outer segment photoreceptor layer from baseline to 12 weeks after laser treatment may predict the transition to cCSC.

Long non-coding RNAs (lncRNAs) have been revealed to harbor open reading frames (ORFs) that can be translated into small peptides. The peptides may participate in the pathogenesis of colorectal cancer (CRC). Herein, we investigated the role of a lncRNA BVES-AS1-encoded peptide in colorectal tumorigenesis. Through bioinformatic analysis, lncRNA BVES-AS1 was predicted to have encoding potential and to be associated with poor prognosis of patients with CRC. In CRC cells, BVES-AS1 was validated to encode a 50-aa-length micro-peptide, named BVES-AS1-201-50aa, through a western blotting method. BVES-AS1-201-50aa enhanced cell viability and promoted the migratory and invasive capacities of HCT116 and SW480 CRC cells in vitro, validated via CCK-8 assay and transwell assay, respectively. Immunofluorescence assay showed that BVES-AS1-201-50aa increased the expression of proliferating cell nuclear antigen (PCNA) and matrix metalloproteinase 9 (MMP9) in CRC cells. We further verified that BVES-AS1-201-50aa targeted and activated the Src/mTOR signaling pathway in CRC cells by co-immunoprecipitation (Co-IP) experiment, qualitative proteomic analysis, and western blotting. Our findings demonstrated that BVES-AS1 could encode a micro-peptide, which promoted CRC cell viability, migration, and invasion in vitro. Our current work broadens the diversity and breadth of lncRNAs in human carcinogenesis.

Recurrent pregnancy loss (RPL) is both mental and physical health problem affecting about 1–5% of women of childbearing age. The etiology of RPL is complex, involving chromosomal abnormalities, autoimmune diseases, metabolic disorders, and endometrial dysfunction. The causes of abortion are still unknown in more than 50% of these cases. With the development of science and technology, an increasing number of scholars focus on this field and find that genetic factors may play an essential role in unexplained RPL, such as embolism-related genes, immune factor-related genes, and chromosomal numeric, and structural variation. This review summarizes the genetic factors associated with RPL, including genetic mutations and genetic polymorphisms, chromosomal variants, and chromosomal polymorphisms. Many related genetic factors have been found to be demographically and geographically relevant, some of which can be used for risk prediction or screening for the etiology of RPL. However, it is difficult to predict and prevent RPL due to uncertain pathogenesis and highly variable clinical presentation. Therefore, the genetic factors of RPL still need plentiful research to obtain a more accurate understanding of its pathogenesis and to provide more detection means for the screening and prevention of RPL.

Summary: N6-methyladenosine (m6A), the most abundant internal mRNA modification in eukaryotes, plays a vital role in regulating innate immunity. However, its underlying mechanism remains largely unknown, especially in lower vertebrates. The results of the present study show that fat-mass- and obesity-associated protein (FTO), also known as a m6A demethylase, improved the innate immunity and prevented Siniperca chuatsi rhabdo virus and Vibrio anguillarum infection in miiuy croaker. Significantly, FTO-promoted immunity was dependent on its m6A demethylase activity. In terms of mechanism, NOD1 has abundant methylation modification in its CDS and 3′UTR regions, and FTO can reduce its methylation level, thus avoiding its degradation by YTHDF2. In summary, our results indicate that the FTO-mediated m6A modification in NOD1 mRNA promotes innate immunity by activating the NOD-like receptor pathway, which provides a molecular mechanism for the regulation of immune response via RNA methylation in teleost.

Immunotherapy has been recognized as an effective and important therapeutic modality for multiple types of cancer. Nevertheless, it has been increasing recognized that clinical benefits of immunotherapy are less than expected as evidenced by the fact that only a small population of cancer patients respond favorably to immunotherapy. The structurally and functionally abnormal tumor vasculature is a hallmark of most solid tumors and contributes to an immunosuppressive microenvironment, which poses a major challenge to immunotherapy. In turn, multiple immune cell subsets have profound consequences on promoting neovascularization. Vascular normalization, a promising anti-angiogenic strategy, can enhance vascular perfusion and promote the infiltration of immune effector cells into tumors via correcting aberrant tumor blood vessels, resulting in the potentiation of immunotherapy. More interestingly, immunotherapies are prone to boost the efficacy of various anti-angiogenic therapies and/or promote the morphological and functional alterations in tumor vasculature. Therefore, immune reprograming and vascular normalization appear to be reciprocally regulated. In this review, we mainly summarize how tumor vasculature propels an immunosuppressive phenotype and how innate and adaptive immune cells modulate angiogenesis during tumor progression. We further highlight recent advances of anti-angiogenic immunotherapies in preclinical and clinical settings to solidify the concept that targeting both tumor blood vessels and immune suppressive cells provides an efficacious approach for the treatment of cancer.

There is a lot of evidence that oral hypoglycemic drugs work by affecting gut microbes, but the key strains responsible for this effect are not well known. Huang-Qi-Ling-Hua-San (HQLHS), composed of Astragalus Membranaceus, Ganoderma lucidum, Inonotus obliquus, and Momordica charantia L., is a specially designed Chinese medicine formula to treat type 2 diabetes (T2D). In this study, a mouse model of T2D induced by high-fat diet and streptozotocin was used to explore the mechanism of HQLHS in improving hyperglycemia and hyperlipidemia through multiple rounds of animal experiments, such as HQLHS feeding, fecal microbiota transplantation (FMT), and live bacteria feeding, so as to explore the potential target intestinal flora in its hypoglycemic effect. Results show that such specific taxa as Bifidobacterium, Turicibacter, Alistipes, Romboutsia, and Christensenella were identified to be preferably enriched by HQLHS and then assumed to be the target microbes. Herein, FMT was used to test if the upregulated beneficial bacteria by HQLHS play a therapeutic role. The strain Christensenella minuta DSM 22607 and the strain Christensenella timonensis DSM 102800 were selected to test the beneficial effect of Christensenella taxa on T2D. Diabetic animals supplemented with these strains showed the improvement in blood glucose and lipid metabolism, the promotion of GLP-1 secretion, the increase in antioxidant capacity, the inhibition of hepatic gluconeogenesis, the suppression of intestinal glucose absorption, the enhancement of intestinal barrier, reduced LPS-induced inflammation, and the reduction of branched amino acids (BCAAs) content in the liver. Overall, these data demonstrate that Christensenella plays a beneficial role in T2D and is a target for the action of HQLHS therapy.

The mitogen-activated protein kinase (MAPK) gene family performs crucial roles in cell division, migration, development, apoptosis, inflammatory response, and abiotic and biotic stress responses. However, very little information is available about the MAPKs in turbot (Scophthalmus maximus). In this study, 15 turbot MAPKs (SmMAPKs) were identified throughout the whole genome, and their basic chemical and physical properties and subcellular localization were illustrated. All SmMAPKs contained the serine/threonine protein kinases, catalytic domain (S_TKc, SMART00220). Phylogenetic analysis revealed that SmMAPKs were classified into three subfamilies, namely, c-Jun NH2-terminal kinase (JNK), extracellular signal-regulated kinase (ERK), and p38. Conserved motif and gene structure analysis revealed high levels of conservation within and between phylogenetic subfamilies. Expression patterns of MAPKs in distinct tissues and under diverse abiotic and biotic stresses were examined using the published available RNA-seq data sets. As a result, SmMAPKs showed obviously tissue-specific expression. Furthermore, 7 and 10 candidate stress-responsive MAPK genes were detected under abiotic and biotic stresses, respectively, among which five common MAPK genes, namely, SmMAPK4 (ERK4), SmMAPK6 (ERK3), SmMAPK11 (p38β), SmMAPK12b (p38γ), and SmMAPK15 (ERK7/8) showed extremely significant responses to both abiotic and biotic stresses, demonstrating their potential functions in comprehensive antistress. These results demonstrate that MAPKs might play vital roles in response to various abiotic and biotic stresses in turbot, which would contribute to making scientific preventive measures to environmental changes in the process of farming and promoting the development of selective breeding for comprehensive stress resistance in turbot.

Abstract Background Cadmium (Cd) is a common heavy metal that mainly causes renal damage. There is a lack of research on the large-scale and systematic evaluation of the association between urinary Cd (U-Cd) and various effect biomarkers among Chinese residents. Methods Based on the establishment process of dietary Cd limit standards by European Food Safety Authority (EFSA), the dose–response relationships between U-Cd and four biomarkers, β2-microglobulin (β2-MG), N-acetyl-β-glucosidase (NAG), microalbumin (mALB), and retinol binding Protein (RBP) were explored, respectively. Toxicokinetic model was used to derive the dietary Cd exposure limit for Chinese residents after critical U-Cd concentration was calculated. Results As the sensitive biomarkers of renal injury, β2-MG and NAG were selected to estimate the 95% confidence interval lower limit of the U-Cd benchmark dose (BMDL5) to be 3.07 and 2.98 μg/g Cr, respectively. Dietary Cd exposure limit was calculated to be 0.28 μg/kg bw/day (16.8 μg/day, based on the body weight of 60 kg), which was lower than the average Chinese Cd exposure (30.6 μg/day) by the China National Nutrient and Health Survey. Conclusion This study established an overall association between U-Cd and renal injury biomarkers, and explored the Chinese dietary Cd exposure limits, which helps improve Chinese Cd exposure risk assessment and provides a reference basis for formulating reasonable exposure standards.

Sciaenops ocellatus, also known as red drum, is a valuable commercial fish that is widely cultured in China. In recent years, while the genetic research and molecular breeding on S. ocellatus has been widely performed, whole-genome resources are lacking. In our study, we assembled the S. ocellatus genome by high-quality genome sequencing, followed by complete annotation. The size of final genome was 686.62 Mb and N50 length of contig and scaffold is 99.71 Kb and 25.62 Mb, respectively. Using the Hi-C technology, we obtained 24 pseudochromosomes containing 87.82% of the total scaffolds. We annotated 20,053 protein-coding genes in S. ocellatus genome, of which 99.93% were homologous to proteins in different database. Totally, we detected 133.54 Mb repeat sequences, which accounts for 19.45% of the assembly. According to the phylogenetic analysis, S. ocellatus is most closed to L. croea with a divergence time about 30 million years ago. We assembled a high-quality genome that will be a valuable resource to further study the biology of S. ocellatus and will inform how to breed the red drum to enhance its economic traits.

The main drawback of correlation filter scheme is that it can only predict translation of the object, ignoring the other affine transformations such as rotation, aspect ratio change, and scale change. This paper tries to address this problem by a two-step affine transformation prediction method. The first step is to predict coarse target translation by the correlation filter model with adaptive model update rate generation. Compared with fixed update rate in existing correlation filter based trackers, the presented method generates the update rate by Long Short-Term Memory model with inputs of historical target templates. The second step maps well-designed features to affine transformation parameters. The designed features contain the information of affine transformation which makes the learning of non-linear mapping function possible. The training samples for on-line filter model update are transformed to the same pose, and they help the learnt filter to represent the object better than non-aligned samples. The proposed network is trained from end to end and achieves competitive performance when comparing with state-of-the-arts trackers on four benchmarks, OTB100, UAV123, VOT2018, and VOT2020.
