AIM: To explore the changes and significance in JIA-U and to detect the levels of CDld(hi)CD5+CD19+ regulatory B cells(Breg)in the peripheral blood of children with juvenile idiopathic arthritis-associated uveitis(JIA-U).METHODS: From April 2018 to May 2020, 95 children with JIA-U were selected as JIA-U group; 70 children with juvenile idiopathic arthritis(JIA)were selected as JIA group, and all of them were diagnosed and treated in our hospital; another 75 healthy children in the same period were selected as the control group. The ratio of CDld(hi)CD5+CD19+Breg in peripheral blood was detected by flow cytometry; the level of IL-10 in peripheral blood was detected by ELISA; the correlation between CDld(hi)CD5+CD19+Breg ratio and the expression of IL-10, the severity of JIA-U patients was analyzed by Pearson correlation coefficient method; the influencing factors of JIA-U were analyzed by Logistic regression. To elucidate the mechanism of Euodiae Fructus stir-fried with water decoction of Coptidis Rhizoma in the treatment of chronic colitis, this study employed ultra performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC-Q-TOF-MS), network pharmacology, and experimental verification to predict the involved targets and signaling pathways. The chronic colitis mouse model was constructed to verify the core targets. A total of 48 compounds in the herbal medicine were identified by UPLC-Q-TOF-MS. SwissTargetPrediction was used to screen the potential active components and drug targets. GeneCards, OMIM, PharmGKB, and TDD were used to search for the disease targets. A total of 31 active ingredients, 453 targets of the herbal medicine, and 3 960 targets of chronic colitis were obtained. The common targets shared by the herbal medicine and chronic colitis were introduced into STRING to construct the protein-protein interaction(PPI) network, and CytoNCA plug-in was used to screen the key targets. A total of 90 key targets were obtained, and the key active components included isorhamnetin, quercetin, limonin, and oxyberberine. GO annotation and KEGG pathway enrichment for the key targets were carried out via DAVID. The targets were mainly involved in the positive regulation of protein phosphorylation, positive regulation of nitric oxide biosynthetic process, and negative regulation of apoptotic process. The medicine may treat chronic colitis through PI3 K-Akt, VEGF, HIF-1, and TNF signaling pathways. A mouse model of chronic colitis was established and then treated with Euodiae Fructus stir-fried with the water decoction of Coptidis Rhizoma. The experimental results demonstrated that the medicine can alleviate the pathological damage of colon, significantly reduce the levels of IL-1β, IL-6, and TNF-α, inhibit the activation of PI3 K/Akt pathway, and down-regulate the expression of VEGFA in the treatment of chronic colitis.

Background Gastric cancer (GC) is a prevalent malignancy, leading to a high incidence of cancer-associated death. Cisplatin (DDP)-based chemotherapy is the principal therapy for clinical GC treatment, but DDP resistance is a severe clinical challenge and the mechanism remains poorly understood. Circular RNAs (circRNAs) have been identified to play crucial roles in modulating the chemoresistance of gastric cancer cells. Aim To explore the effect of circVAPA on chemotherapy resistance during GC progression. Methods The effect of circVAPA on GC progression and chemotherapy resistance was analyzed by MTT assay, colony formation assay, Transwell assay, wound healing assay, and flow cytometry analysis in GC cells and DDP resistant GC cell lines, and tumorigenicity analysis in nude mice in vivo. The mechanism was investigated by luciferase reporter assay, quantitative real-time PCR, and Western blot analysis. Results CircVAPA expression was up-regulated in clinical GC tissues compared with normal samples. CircVAPA depletion inhibited proliferation, migration, and invasion and increased apoptosis of GC cells. The expression of circVAPA, STAT3, and STAT3 downstream genes was elevated in DDP resistant SGC7901/DDP cell lines. CircVAPA knockdown attenuated the DDP resistance of GC cells. Mechanically, circVAPA was able to sponge miR-125b-5p, and miR-125b-5p could target STAT3 in the GC cells. MiR-125b-5p inhibitor reversed circVAPA depletion-enhanced inhibitory effect of DDP on GC cells, and STAT3 knockdown blocked circVAPA overexpression-induced proliferation of DDP-treated SGC7901/DDP cells. The depletion of STAT3 and miR-125b-5p inhibitor reversed circVAPA depletion-induced GC cell apoptosis. Functionally, circVAPA contributed to the tumor growth of SGC7901/DDP cells in vivo. Conclusion CircVAPA promotes chemotherapy resistance and malignant progression in GC by miR-125b-5p/STAT3 signaling. Our findings present novel insights into the mechanism by which circVAPA regulates chemotherapy resistance of GC cells. CircVAPA and miR-125b-5p may be considered as the potential targets for GC therapy.

Background Reprogrammed glucose metabolism of enhanced Warburg effect (or aerobic glycolysis) is considered as a hallmark of cancer. Long non-coding RNAs (lncRNAs) have been certified to play a crucial role in tumor progression. The current study aims to inquire into the potential regulatory mechanism of long intergenic non-protein coding RNA 242 (LINC00242) on aerobic glycolysis in gastric cancer. Method LINC00242, miR-1-3p and G6PD expression levels in gastric cancer tissues and cells were determined by qRT-PCR. Cell apoptosis or viability were examined by Flow cytometry or MTT assay. Western blot was utilized to investigate G6PD protein expression levels. Immunohistochemical (IHC) and hematoxylin and eosin (H&E) staining were used for histopathological detection. The targeted relationship between LINC00242 or G6PD and miR-1-3p was verified by luciferase reporter gene assay. Nude mouse xenograft was utilized to detect tumor formation in vivo. Result LINC00242 and G6PD was high-expressed in gastric cancer tissues and cells, and LINC00242 is positively correlated with G6PD. Silencing of LINC00242 or G6PD within gastric cancer cells prominently inhibited cell proliferation and aerobic glycolysis in vitro and relieved the tumorigenesis of gastric cancer in vivo. miR-1-3p was predicted to directly target both LINC00242 and G6PD. Overexpression of miR-1-3p suppressed gastric cancer cells proliferation and aerobic glycolysis. LINC00242 competitively combined miR-1-3p, therefore relieving miR-1-3p-mediated suppression on G6PD. Conclusion LINC00242 plays a stimulative role in gastric cancer aerobic glycolysis via regulation of miR-1-3p/ G6PD axis, therefore affecting gastric cancer cell proliferation.

Engineering complete periodontal tissue regeneration remains a considerable clinical challenge because the native periodontium is composed of hard mineralized and soft unmineralized tissues. Stem cell therapies that use endogenous extracellular matrix (ECM) have the potential for structural and functional periodontal regeneration. Here, mineralized ECM/dental pulp stem cell microspheroids (MMCMs) mimicking native mesenchymal microspheroid precursors were developed, and showed an enhanced Young’s modulus ranging from 0.4 GPa to 1.9 GPa. Furthermore, biomineralization promoted cell viability, inhibited cell apoptosis, and enhanced ECM secretion and osteogenic differentiation potential in dental pulp stem cells. After implantation in the full periodontal defect, MMCMs regenerated complete periodontium with native-like hierarchically organized periodontal ligament collagen fibers inserted into the newly formed alveolar bone and cementum; they also recruited CD90+CD73+ host mesenchymal stem cells. Mechanistically, MMCMs promoted functional periodontal regeneration through activation of the transforming growth factor beta 1 (TGF-β1)/Smad3 signaling pathway. Inhibition of TGF-β1 signaling pathway significantly impaired osteogenic potential and matrix secretion of MMCMs, while exogenous TGF-β1 treatment enhanced the osteogenesis and periodontal regeneration outcomes of MMCMs. These results indicate the promising application of MMCMs in complex tissue regeneration.

On the basis of on-site investigation and data collection, this paper summarizes the specific measures, which were taken by Hangzhou West Lake during its development of intelligent tourism. Many aspects are included, which are the intelligence of tourist communication, intelligence of ticketing, intelligence of public information distribution, intelligence of payment, intelligence of self-service and so on. Besides, this paper also analyzes the shortcomings and then it will put forward the following suggestions: strengthening the propaganda of intelligent tourism service facilities, applying intelligent tourism system to security insurance, increasing the humanized intelligent electronic guidance, designing tourism products based on the cultural heritage of West Lake, and exploring the functions of intelligent tourism in improving tourists' quality experience.

As an important type of additive, the redispersible powder is widely applied in cement-based and gyp-based materials, so as to improve the cementing strength, impermeability, workability, hydrophobicity and so on. However, the chemical composition of the redispersible powders determines its applicability, as well as the degree of improvement to the matrix materials. In this paper, the chemical composition of an unknown redispersible powder was analyzed by several instruments. Firstly, Fourier transform infrared spectroscopy (FTIR) analysis indicated that the polymer was polymerized with monomers of vinyl esters. Secondly, pyrolysis-gas chromatography-mass spectrometry (Py-GC/MS) analysis showed the pyrolysis products were mainly acetic acid, methyl acetate and methyl versatates. Considering the pyrolysis mechanism of synthetic polymers and the results from FTIR spectra, the polymer in the redispersible powder was deduced to be the copolymer of vinyl acetate and vinyl ester of versatic acid (VeoVa), which was a monomer with a unique highly branched carbon-rich structure. Thermogravimetry (TG) analysis revealed that the powder contained 74.0% of polymer, 14.5% of calcium carbonate and 11.4% of other inorganic materials.

A new type of strongly sub-feasible SQP (Strong Quadratic Programming) method is used to establish a nonlinear model to realize the automatic search of the rational cable force of the cable-stayed bridge. The objective function is established based on the stress and displacement of the main girder and the bridge tower. The cable force of the stay cable is the design variable, the stress and displacement of the main girder and the bridge tower are the constraints, and the numerical model of the earthquake force is analyzed under different cable loading conditions. The results show that the new type of strongly sub-feasible SQP method is suitable for the complicated cable force optimized parameters of large-scale cable-stayed bridges, the calculated results are in good agreement with the designed cable force, and the bending moment of the main beam in the bridge state is uniform. The main tower is close to the axial compression force, and structural deformation is much smaller than the specification limits. Under the excitation of earthquake, the optimized cable force can effectively improve the uneven distribution of bending moment of the main beam, limit the longitudinal mid-span deformation and the lateral deformation of the main tower, so that the whole structure is safer in the earthquake.

The coverage is important for the wireless networking cost. Wide coverage means less base station and less cost. In this paper, we first describe the structure of power wireless network. Then we compared the LTE 230 MHz and 1.8 GHz. These two cellular networks cannot fulfill deep coverage requirement, so we use LoRa to solve this problem. Finally, we do some measurements with LoRa in some real environments. The measured results shows the LoRa can fulfill the deep coverage requirement.

Streptomycin (STR) was the bactericidal antibiotic used extensively in human and veterinary medicine with a high aqueous solubility. In this study, the adsorption of STR on soil was studied as a function of pH, cationic types and ionic strength by using adsorption modeling and bath experiments. Adsorption modeling of the adsorption of STR was better fitted with the Freundlich as compared to the Langmuir. The absorbed STR on soil kept relatively constant at pH 3.5-8 and decreased at pH>8. And the order of cationic influence was CaCl2> NaCl≈KCl> the blank sample for STR sorption capacity, suggesting that the sorption of STR increased as the presence of low concentrations of the cationic. The amount of STR adsorbed was inversely proportional to the logarithmic concentration of the background cations.

Catalytic combustion technology can effectively reduce the temperature of the combustion reaction and significantly inhibit the formation of NOx. Perovskites catalyst materials have high stability, varied chemical composition, and low cost etc. A combination of catalytic combustion technology and perovskite catalyst materials has great significance for energy industry and environment protection. However, improving lower temperature catalytic activity and high temperature thermostability performance of perovskite catalyst is still major problem. Some researchers have improved the activity and stability of perovskite catalysts for methane combustion by different preparation methods. The structure and preparation methods of perovskite-type oxide catalysts are presented.

This article discusses the NMT communities on the impact of urban environment and social issues, proposing the main problems existing in NMT communities are follows: The NMT paths are lack of systematic; the wide of NMT is not enough; mixed with the motor transportation; NMT environmental quality is not high. Then put forward some design methods: Set up a network of NMT combining with community environment; build up the NMT landscape suitable to human scale; community facilities design for NMT.

Although surfactants have been considered in surfactant-aided soil washing systems, there is little information on the adsorption of the impact of surfactant on the adsorption of antibiotic, and this may have significant implications for the soil. In this study, Triton X-100 and SDBS were selected to study its effect on the sorption of Streptomycin sulfate from soil under equilibrium sorption. The adsorption of Streptomycin sulfate on soils in surfactant free and surfactant solutions of different critical micelle concentrations (CMCs) has been studied .The applied surfactant concentrations (X) ranged from below the (nominal) CMC to 5 times the CMC. For relatively water-soluble Streptomycin sulfate, the distribution coefficients with anionic surfactant (Kd*) deceeded those without surfactant (Kd), while non-ionic (Kd*) all exceeded those without surfactant (Kd). The Kd*/Kd ratios were used to evaluate the efficiency of surfactants and it was found that anionic surfactant is a better choice for remediation of contaminated soils whereas non-ionic surfactants leads to poor remediation efficiency.

An ionic liquid, 1-butyl-3-methylimidazolium tetrafluoroborate( [BMIM] BF4) / salt aqueous two-phase systems (ATPS) was presented as a simple, rapid and effective sample pretreatment technique coupled with double-beam UV-Vis spectrophotometer for analysis of gatifloxacin in water. For the purpose to choose optimal conditions, various factors were considered systematically, and the results indicated that the pH value, the salt-out ability of salt and the amount of [BMIM] BF4 had great influence on phase separation and extraction. Under the optimal condition, gatifloxacin in different concentration were enriched and determined, the recovery was 87.3–91.5%.

Poly[ethyl trifluorobuty-2-noate] (PETFB) was prepared from ethyltrifluorobuty-2-noate by anionic polymerization. The polymer was examined by UV-Vis, IR and NMR spectrometries. It possessed π-conjugated backbone in the main chain. No significant variation in the electrical conductivity of BF3-doped PETFB was observed after more than three months' storage, indicating improved conductive stability in air as compared with polyacetylene.

The main purpose of applying fuzzy clustering method to dividing customer group is to divide series or establish serial platform, satisfying customer demand and promoting the robustness of product platform. Aiming at the deficiencies of current customer group dividing method based on fuzzy clustering, in this paper, an improved customer group dividing method based on fuzzy transitive closure dynamic clustering method is presented and a method to find the same result elements is adopted to simplify the calculation process of the transitive closure of a fuzzy similar matrix. And then, based on customer group dividing result, a product platform model based on customer group dividing is established in order to improve products based on platform. Finally, an instance of A electronic company manufacturing speakers is given in order to verify the effectiveness of the method proposed.

A method was proposed for the determination of trace lead with flame atomic absorption spectrometry after preconcentration of lead by rapid coprecipitation technique with PAR-Fe (III) at pH 6.0. The analytical parameters including pH, amount of iron (III), amount of reagent, the standing time of the precipitate, etc., were examined. The detection limits (DL) were found to be 18.7 microg x L(-1) for Pb (II). In analysis of lake water and the milk tea powder samples, RSD's and the standard addition recovery of this method were in the ranges of 1.03%-2.24% and 94.2%-98.3% respectively. The effect of matrix can be overcome by the method and the results are satisfyiog. The method shows good application prospect in the determination of trace lead owing to its rapidness and reproducibility.

MicroRNAs (miRNAs) play important roles in the development of various cancers. MiRNA-497 functions as a tumor-suppressor that is downregulated in several malignancies; however, its role in non-small cell lung cancer (NSCLC) has not been examined in detail. Here, we showed that miR-497 is downregulated in NSCLC tumors and cell lines and its ectopic expression significantly inhibits cell proliferation and colony formation. Integrated analysis identified HDGF as a downstream target of miR-497, and the downregulation of HDGF by miR-497 overexpression confirmed their association. Rescue experiments showed that the inhibitory effect of miR-497 on cell proliferation and colony formation is predominantly mediated by the modulation of HDGF levels. Furthermore, tumor samples from NSCLC patients showed an inverse relationship between miR-497 and HDGF levels, and ectopic expression of miR-497 significantly inhibited tumor growth and angiogenesis in a SCID mouse xenograft model. Our results suggest that miR-497 may serve as a biomarker in NSCLC, and the modulation of its activity may represent a novel therapeutic strategy for the treatment of NSCLC patients.

Vibrio parahaemolyticus ATCC 27519 was chosen as indicator of aquacultural pathogenic bacteria to determine the antibacterial activity and mechanism of copper-bearing montmorillonite (Cu-MMT) in vitro. The results indicated that montmorillonite (MMT) had no antibacterial activity. The minimum inhibitory concentration (MIC) and bactericidal concentration (MBC) of Cu-MMT on Vibrio parahaemolyticus were 75 and 300 mg/L, respectively. The activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT) and lactate dehydrogenase (LDH) of bacteria were examined and the results showed treatment with Cu-MMT could lead to significant release of intracellular enzymes from the tested bacteria suggesting that the permeability of the cell membrane increased and bacteria suffered injury. Three typical inhibitors (malonic acid, iodine acetic acid and phosphate sodium) were used to further study the inhibitory pathways of respiratory metabolism. Cu-MMT effectively inhibited respiratory metabolism of Vibrio parahaemolyticus, with the respiratory inhibition percent (I(R)) of 31.8%. The respiratory superposing inhibition percent after addition of phosphate sodium, iodine acetic acid and malonic acid was 48.6%, 27.8% and 17.5%, respectively. These results indicated that the effect of malonic acid on superposing inhibition percent of Cu-MMT for bacteria is the lowest; thus, the synergic action between Cu-MMT and malonic acid is the weakest, indicating that they inhibited the same pathway of respiratory metabolism, i.e. the TCA pathway, which is the most important pathway of carbohydrate metabolism. The atomic force microscope image of Vibrio parahaemolyticus exposed to Cu-MMT showed that Cu-MMT could rupture the bacterial cell membrane.

To investigate the effects of granulocyte colony-stimulating factor (G-CSF) on peripheral endothelial progenitor cells (EPC) and atherosclerosis (AS) in cholesterol-fed rabbits.Male New Zealand white rabbits were randomly divided into control group, G group (Recombinant Human Granulocyte Colony Stimulating Factor Injection rhG-CSF 50 microg/d), AS group (high cholesterol diet) and G + AS group (rhG-CSF 50 microg/d plus high cholesterol diet, n = 8 per group). Peripheral blood was collected at baseline and at 1, 4, 8 and 12 weeks, total mononuclear cells (MNCs) were isolated from peripheral blood by Ficoll density gradient centrifugation, and then the cells were plated on fibronectin-coated culture dishes. After being cultured for 7 days, EPCs were characterized as adherent cells double positive for DiLDL uptake and lectin binding by direct fluorescent staining under a laser scanning confocal microscope. After being cultured for 3 days, the number of EPC (PE-CD34/FITC-CD133 double-stained positive cells) was quantified by flow cytometric analysis. Serum NO was measured and aortic plaque area analyzed at 12 weeks.EPC number was low in control and AS groups and EPC number was significantly increased ( approximately 13-fold, P0.001) compared to baseline at 1 week in G and G + AS groups and remained at this level throughout the study period in G group while decreased gradually in G + AS group and returned to baseline level at 12 weeks. Aortic atherosclerotic plaque was visible in both AS and G + AS groups, however, the aortic atherosclerotic plaque area was smaller in G + AS group than that of in As group (59.8 mm(2) +/- 26.9 mm(2) vs. 251.5 mm(2) +/- 83.4 mm(2), P0.01). Serum NO was similar between AS and G + AS groups and significantly higher than that in control and G groups.CSF could attenuate atherosclerosis in cholesterol-fed rabbits by increasing circulating EPC.

To evaluate the time course of granulocyte-colony-stimulating-factor (G-CSF), estrogen and various doses of atorvastatin on endothelial progenitor cells (EPCs) mobilization.A total of 48 male New Zealand White rabbits were treated with placebo, estrogen (0.25 mg.k(-1).d(-1)), Atorvastatin (2.5, 5, or 10 mg) and G-CSF (50 microg/rabbit/d), respectively. Peripheral EPCs number was surveyed weekly for 4 weeks by FACS analysis (double-positive for PE-CD34/FITC-CD133) and under fluorescent microscope (double-positive for FITC-UEA-1/Dil-acLDL). Serum nitric oxide (NO) and lipids were also measured at the third week.Peripheral EPCs was significantly increased in G-CSF treated animals and remained constant for 4 weeks compared to placebo treated animals. Atorvastatin increased peripheral EPCs dose-dependently from 2.5 to 5 mg and peaked at the third week while peripheral EPCs number was not affected by 10 mg.k(-1).d(-1) atorvastatin during the first 3 weeks and was significantly higher only in the fourth week compared to placebo group. Estrogen also significantly increased peripheral EPCs at the third and fourth week compared to placebo group. At the third week, serum NO was similar in G-CSF group, significantly higher in atorvastatin 5 mg.k(-1).d(-1) and estrogen groups while significantly lower in atorvastatin 10 mg.k(-1).d(-1) group compared to placebo group. Serum lipids were similar among various groups.Atorvastatin, estrogen and G-CSF could mobilize EPCs. The mobilization efficacy is as follows: G-CSFatorvastatin 5 mg.k(-1).d(-1)estrogenatorvastatin 2.5 mg.k(-1).d(-1)atorvastatin 10 mg.k(-1).d(-1). NO might partly contribute to the mobilizing effect of estrogen and atorvastatin.
