Abstract Powdery mildew is a devastating disease that affects wheat yield and quality. Wheat wild relatives represent valuable sources of disease resistance genes. Cloning and characterization of these genes will facilitate their incorporation into wheat breeding programs. Here, we report the cloning of Pm57, a wheat powdery mildew resistance gene from Aegilops searsii. It encodes a tandem kinase protein with putative kinase-pseudokinase domains followed by a von Willebrand factor A domain (WTK-vWA), being ortholog of Lr9 that mediates wheat leaf rust resistance. The resistance function of Pm57 is validated via independent mutants, gene silencing, and transgenic assays. Stable Pm57 transgenic wheat lines and introgression lines exhibit high levels of all-stage resistance to diverse isolates of the Bgt fungus, and no negative impacts on agronomic parameters are observed in our experimental set-up. Our findings highlight the emerging role of kinase fusion proteins in plant disease resistance and provide a valuable gene for wheat breeding.

Abstract As potent approaches for addressing computationally expensive optimization problems, surrogate-assisted evolutionary algorithms (SAEAs) have garnered increasing attention. Prevailing endeavors in evolutionary computation predominantly concentrate on expensive continuous optimization problems, with a notable scarcity of investigations directed toward expensive combinatorial optimization problems (ECOPs). Nevertheless, numerous ECOPs persist in practical applications. The widespread prevalence of such problems starkly contrasts the limited development of relevant research. Motivated by this disparity, this paper conducts a comprehensive survey on SAEAs tailored to address ECOPs. This survey comprises two primary segments. The first segment synthesizes prevalent global, local, hybrid, and learning search strategies, elucidating their respective strengths and weaknesses. Subsequently, the second segment furnishes an overview of surrogate-based evaluation technologies, delving into three pivotal facets: model selection, construction, and management. The paper also discusses several potential future directions for SAEAs with a focus towards expensive combinatorial optimization.

Heat stress (HS) disrupts intestinal microbiota, glycolipid metabolism, and hepatic mitochondrial function in late gestational mice. Baicalin (BAI), a Chinese herbal medicine known for its heat-clearing and anti-inflammatory properties, has shown promise in modulating intestinal microecology and mitigating inflammation in various organs. This study investigates whether baicalin attenuates HS-induced intestinal microbial dysbiosis and liver damage in pregnant mice during late gestation. Twenty-four pregnant mice were randomly assigned to four groups, including thermoneutral (TN) (24 ± 1 ℃), HS (35 ± 1 ℃), HS+BAI200 (oral gavaged with 200 mg/kg BW of BAI), and HS+BAI400 (oral gavaged with 400 mg/kg BW of BAI). 400 mg/kg BAI treatment markedly decreased the rectal temperature and increased fetal weight in HS pregnant mice. Furthermore, 400 mg/kg BAI administration effectively ameliorated HS-induced hepatic damage and lipid disorders, reducing HSP70, AST, and ALT levels while increasing TG concentration. Notably, it activated a network of genes involved in lipid synthesis, including fatty acid synthase (FAS), acetyl-CoA carboxylase (ACC), and oxidation, such as peroxisome proliferator-activated receptor alpha (PPARα), carnitine palmityl transferase 1 beta (CPT1β). Moreover, BAI intervention restored the intestinal morphology and barrier function, evidenced by increased intestinal villus height, the ratio of villus height to crypt depth, and colonic goblet cells numbers. 400 mg/kg of BAI treatment up-regulated the expression of tight junction proteins, such as claudin-1 and Zonula Occludens-1 (ZO-1), in the jejunum and ileum, counteracting HS-induced downregulation. High-throughput sequencing showed that BAI treatment altered cecal microbial composition, increasing the relative abundance of beneficial Bacteroidota and decreasing Deferribacterota, Turicibacter, and Akkermansia. Spearman’s correlation analysis highlighted significant correlations between differential cecal microbiota and physiological indexes. In conclusion, BAI administration alleviated adverse impacts in heat-exposed mice during late gestation, improving maternal physiological parameters, and ameliorating hepatic damage with altered cecal microbial composition. The findings suggest that BAI may regulate the gut-liver axis by modulating intestinal morphology, microecology, and hepatic function.

Di(2-ethylhexyl) phthalate (DEHP) is a widely recognized environmental endocrine disruptor that potentially impacts female reproductive function, although the specific mechanisms leading to such impairment remain unclear. A growing body of research has revealed that the endoplasmic reticulum and mitochondrial function significantly influence oocyte quality. The structure of mitochondria-associated endoplasmic reticulum membranes (MAMs) is crucial for facilitating the exchange of Ca2+, lipids, and metabolites. This study aimed to investigate the alterations in the composition and function of MAMs after DEHP exposure and to elucidate the underlying mechanisms of ovarian toxicity. The female mice were exposed to DEHP at doses of 5 and 500 mg/kg/day for one month. The results revealed that DEHP exposure led to reduced serum anti-Müllerian hormone levels and increased atretic follicles in mice. DEHP induced endoplasmic reticulum stress and disrupted calcium homeostasis in oocytes. Furthermore, DEHP impaired the mitochondrial function of oocytes and reduced their membrane potential, and promoting apoptosis. Similar results were observed in human granulosa cells after exposure to mono-(2-ethylhexyl) phthalate (MEHP, metabolites of DEHP) in vitro. Proteomic analysis and transmission electron microscopy revealed modifications in the functional proteins and structure of the MAMs, and the suppression of oxidative phosphorylation pathways. The findings of this investigation provide a new perspective on the mechanism underlying the reproductive toxicity of DEHP in females.

IntroductionNeuromyelitis Optica spectrum disorder (NMOSD) is an autoimmune disease characterized by anti-aquaporin-4 (AQP4) auto-antibodies. The discovery of antibodies AQP4 and myelin oligodendrocyte glycoprotein (MOG) has expanded our understanding of the pathogenesis of neuromyelitis optica. However, the molecular mechanisms underlying the disease, particularly AQP4-associated optic neuritis (AQP4-ON), remain to be fully elucidated.MethodsIn this study, we utilized Weighted Gene Co-expression Network Analysis (WGCNA) to investigate the transcriptomic profiles of peripheral blood samples from patients with AQP4-ON and MOG-positive optic neuritis (MOG-ON), compared to healthy controls.ResultsWGCNA revealed a brown module (ME brown) strongly associated with AQP4-ON, which correlated positively with post-onset visual acuity decline. A total of 132 critical genes were identified, mainly involved in histone modification and microtubule dynamics. Notably, genes HDAC4, HDAC7, KDM6A, and KDM5C demonstrated high AUC values in ROC analysis, indicating their potential as biomarkers for AQP4-ON.ConclusionOur findings provide novel insights into the molecular signature of AQP4-ON and highlight the potential of systems biology approaches in identifying biomarkers for NMOSD. The identified histone modification genes warrant further investigation for their role in disease pathogenesis and as therapeutic targets.

Abstract Sugar Will Eventually be Exported Transporter (SWEET) proteins are highly conserved in various organisms and play crucial roles in sugar transport processes. However, SWEET proteins in peanuts, an essential leguminous crop worldwide, remain lacking in systematic characterization. Here, we identified 94 SWEET genes encoding the conservative MtN3/saliva domains in three peanut species, including 47 in Arachis hypogea, 23 in Arachis duranensis, and 24 in Arachis ipaensis. We observed significant variations in the exon-intron structure of these genes, while the motifs and domain structures remained highly conserved. Phylogenetic analysis enabled us to categorize the predicted 286 SWEET proteins from eleven species into seven distinct groups. Whole genome duplication/segment duplication and tandem duplication were the primary mechanisms contributing to the expansion of the total number of SWEET genes. In addition, an investigation of cis-elements in the potential promoter regions and expression profiles across 22 samples uncovered the diverse expression patterns of AhSWEET genes in peanuts. AhSWEET24, with the highest expression level in seeds from A. hypogaea Tifrunner, was observed to be localized on both the plasma membrane and endoplasmic reticulum membrane. Moreover, qRT-PCR results suggested that twelve seed-expressed AhSWEET genes were important in the regulation of seed development across four different peanut varieties. Together, our results provide a foundational basis for future investigations into the functions of SWEET genes in peanuts, especially in the process of seed development.

Objective Vanin-1 (VNN1) is a pantetheinase that catalyses the hydrolysis of pantetheine to produce pantothenic acid and cysteamine. Our previous studies have shown that the VNN1 is specifically expressed in chicken liver which negatively regulated by microRNA-122. However, the functions of the VNN1 in lipid metabolism in chicken liver haven’t been elucidated. Methods First, we detected the VNN1 mRNA expression in 4-week chickens which were fasted 24 hours. Next, knocked out VNN1 via CRISPR/Cas9 system in the chicken Leghorn Male Hepatoma cell line. Detected the lipid deposition via oil red staining and analysis the content of triglycerides (TG), low-density lipoprotein-C (LDL-C), and high-density lipoprotein-C (HDL-C) after VNN1 knockout in Leghorn Male Hepatoma cell line. Then we captured various differentially expressed genes (DEGs) between VNN1-modified LMH cells and original LMH cells by RNA-seq. Results Firstly, fasting-induced expression of VNN1. Jiulong area is the most concentrated area of Yanshanian granite found in western Sichuan, including Tiechanghe, Qiaopengzi-Landiao and Xinhuoshangranite. In this paper, LA-ICP-MS zircon U-Pb dating were carried out to determine the formation age of the granite， and its temporal-spatial relationship with the surrounding granites, providing a basis for the comparative study of Liwu copper deposit in the southern margin of Songpan-Ganzi block. The results show thatthe diagenetic age of Huajiaoping granite is (163.1±1.4) Ma, andit underwent magmatic or tectonic thermal events at (146±3) Ma, which is consistent withthe geological events ofTiechanghe, Xinhuoshan and Luomogranite. The magma temperature of Huajiaoping granite is 592～727 ℃ (average temperature 648 ℃), which is significantly lower than that of Qiaopengzi granite (766～804 ℃). Combined with the regional geological background, about 170 ～ 140 Ma, the lithosphere in the southern margin of Songpan Ganzi block is in the extension stage. The granite magma activity dominated in the early stage (163 Ma) by Huajiaoping granite, and in the late stage (146 Ma) by magmatism or tectonic thermal events. This discovery enriched the Yanshannian granitic magmatic thermal events in Jiulong area.

Taka dome and Jianglang dome are located in the southern margin of Songpan-Ganzi block, and their geological characteristics indicate similar composition and metamorphic-deformation characteristics. Liwu, Heiniudong and Zhongsui deposits have been found in Jianglang Dome, while no similar deposits have been found in the same horizon of Taka Dome. To investigate the medium-deep part structure characteristics and mineralization of the Taka Dome, the geophysical electrical parameters and audio magnetotelluric sounding profiles of different strata of different ages were measured to obtain the electrical characteristics of the middle and deep subsurface. The results show that the overall characteristics of the dome are low and medium resistance on both sides, and high resistance in the core is mainly mixed with low resistance. The overall electrical characteristics are consistent with the spatiotemporal law of the dome structure. There are three main low-resistivity anomaly zones in the deep electrical profile, which are inferred to reflect the low-resistivity lithology, fault or structural fracture zone. The three main high-resistivity anomaly belts are inferred to be reflected by the upper member of the Mesoproterozoic Liwu Group (Pt2l3) with complete medium-high resistivity characteristics in the Taka dome. The above understanding provides geophysical evidence for exploring the medium-deep geological spatial structure of the Taka Dome.

Abstract In this paper, a fully distributed control protocol is presented based on adaptive technology to solve the time‐varying formation problem of multi‐unmanned aerial vehicle (UAV) systems, where the coupling weights between neighbouring UAVs can be adjusted through adaptive method. Firstly, the proposed control protocol eliminates the need for global information on the interaction topology of multi‐UAV systems, the calculation of Laplacian matrix eigenvalues is avoided and reduces the computational complexity of the system. Secondly, an algorithm with two steps is proposed to determine the distributed adaptive control gain matrices, where the feasibility condition of time‐varying formation of multi‐UAV systems is given. Then, the closed‐loop stability of multi‐UAV systems is proved through Lyapunov theory. Finally, numerical simulation is conducted to verify the effectiveness of the theoretical results.

Detection of flow fields constitutes a critical role in the advancement and innovation of hypersonic aircraft. Under hypersonic conditions, aircraft aerodynamics manifest a multitude of complex phenomena, including intense turbulence and fluctuations, transitions within the boundary layer, interactions between shock waves and boundary layers, as well as the effects of high-temperature gas. Thus, the surveillance of hypersonic aircraft flow fields is imperative not only for flight safety but also for the progression of hypersonic technologies. Given the practical limitations that restrict sensors installed to only key areas for detection, we propose a GNN-based method for hypersonic aircraft flow field reconstruction from limited sensors. Moreover, we have introduced a novel graph modeling technique for enhancing the reconstruction motivated by that solely the most significant edges within the graph are efficacious for field reconstruction. The methodology encompasses the following steps, modeling graphs from diverse perspectives, and transforming them into minimum spanning trees. These sparse graphs are then integrated with learnable weights optimized by automatic differentiation. Our method has been tested on an open-source turbulence dataset, the two components of graph pruning and weight optimization bring about 34% and 10% improvements on average. Furthermore, our approach has been validated in the reconstruction of hypersonic aircraft flow fields for at least 10% error reduction among all the situations.

Abstract Thermal layout optimization problems are common in integrated circuit design, where a large number of electronic components are placed on the layout, and a low temperature (i.e., high efficiency) is achieved by optimizing the positions of the electronic components. The operating temperature value of the layout is obtained by measuring the temperature field from the expensive simulation. Based on this, the thermal layout optimization problem can be viewed as an expensive combinatorial optimization problem. In order to reduce the evaluation cost, surrogate models have been widely used to replace the expensive simulations in the optimization process. However, facing the discrete decision space in thermal layout problems, generic surrogate models have large prediction errors, leading to a wrong guidance of the optimization direction. In this work, the layout scheme and its temperature field are represented by images whose relation can be well approximated by a deep neural network. Therefore, we propose an online deep surrogate-assisted optimization algorithm for thermal layout optimization. First, the iterative local search is developed to explore the discrete decision space to generate new layout schemes. Then, we design a deep neural network to build an image-to-image mapping model between the layout and the temperature field as the approximated evaluation. The operating temperature of the layout can be measured by the temperature field predicted by the mapping model. Finally, a segmented fusion model management strategy is proposed to online updates the parameters of the network. The experimental results on three kinds of layout datasets demonstrate the effectiveness of our proposed algorithm, especially when the required computational budget is limited.

Abstract Background Umbilical cord blood transplantation (UCBT) is a curable therapy for hematological disease; however, the impact of nutritional status on UCBT outcomes remains controversial. To evaluate the joint effect of clinical characteristics and nutritional status on the prognosis of patients who underwent UCBT, we screened various factors to establish a predictive model of overall survival (OS) after UCBT. Methods We performed an integrated clinical characteristic and nutritional risk factor analysis and established a predictive model that could be used to identify UCBT recipients with poor OS. Internal validation was performed by using the bootstrap method with 500 repetitions. Results Four factors, including disease status, conditioning regimen, calf skinfold thickness and albumin level, were identified and used to develop a risk score for OS, which showed a positive predictive value of 84.0%. A high-risk score (≥ 2.225) was associated with inferior 3-year OS post-UCBT [67.5% (95% CI 51.1–79.4%), P = 0.001]. Then, we built a nomogram based on the four factors that showed good discrimination with a C-index of 0.833 (95% CI 0.743–0.922). The optimism-corrected C-index value of the bootstrapping was 0.804. Given the advantages of softness, lightness, low cost, and interaction safety, inverse kinematic modeling and control of soft actuators has caused a research boom. However, in realizing dexterous manipulation of space large soft manipulators, it is much more difficult to achieve precise control not only because of the greater accumulation of errors in the multiple degrees of freedom and nonlinear properties of soft materials at large scales but also because of the inability of directly solving the inverse kinematics in the cases of singular pure elongation. In this work, a model-free intelligent kinematic control strategy is proposed for these problems that exhibit a mapping relationship between the output end-effector position and the input pressure. For multiple-degree-of-freedom robots, especially pneumatic soft manipulators, traditional inverse kinematic modeling methods are complex and inverse Jacobian matrix solution often encounters geometric singularities. To address this issue, this paper proposes an inverse kinematics–multilayer perceptron (IK-MLP) method for soft manipulators. In this strategy, the trained intelligent controller can be applied to control pneumatic manipulators without establishing a traditional inverse kinematic model. The control algorithm is experimentally tested based on the ground experiment system of the space soft manipulator. Simulations and experiments are carried out to validate the given model-free intelligent controller, proving that the IK-MLP method can effectively solve the singularity of inverse kinematics.

Abstract Tuning the coordination structures of metal sites is intensively studied to improve the performances of single‐atom site catalysts (SASC). However, the pore structure of SASC, which is highly related to the accessibility of active sites, has received little attention. In this work, single‐atom ZnN4 sites embedded in P‐functionalized carbon with hollow‐wall and 3D ordered macroporous structure (denoted as H‐3DOM‐ZnN4/P‐C) are constructed. The creation of hollow walls in ordered macroporous structures can largely increase the external surface area to expose more active sites. The introduction of adjacent P atoms can optimize the electronic structure of ZnN4 sites through long‐rang regulation to enhance the intrinsic activity and selectivity. In the electrochemical CO2 reduction reaction, H‐3DOM‐ZnN4/P‐C exhibits high CO Faradaic efficiency over 90% in a wide potential window (500 mV) and a large turnover frequency up to 7.8 × 104 h−1 at −1.0 V versus reversible hydrogen electrode, much higher than its counterparts without the hierarchically ordered structure or P‐functionalization.

IntroductionPersonal care products (PCPs) contain a number of endocrine-disrupting chemicals (EDCs) that could potentially affect the reproductive function in women of childbearing age. However, studies focused on the effects of PCPs use on reproductive outcomes are very limited. The current study aimed to explore the relationships between PCPs use patterns and reproductive outcomes in women undergoing in vitro fertilization/intracytoplasmic sperm injection (IVF/ICSI) treatment.MethodsA total of 1500 women from the Tongji Reproductive and Environmental (TREE) study between December 2018 and January 2020 were included in this study. Participants provided characteristics of PCPs use within the previous three months. Retrieved oocyte number, mature oocyte number, two distinct pronuclei (2PN) zygote number, fertilization rate, cleavage rate, blastocyst formation rate, implantation, clinical pregnancy, miscarriage, and live birth were followed up as reproductive endpoints. Generalized linear regression model was utilized to assess the associations between various categories of PCPs use and reproductive endpoints of IVF/ICSI.ResultsAfter adjusting for relevant covariates, women who used skin care products ≥14 times per week had a reduction of 22.4% in the maturation rate (95% CI: -39.2%, -1.6%) compared to participants who did not use skin care products. After transferring fresh embryos, women who used cosmetics 1–2 times per week (adjusted OR = 2.2, 95% CI: 1.0, 4.8) or 3–7 times per week (adjusted OR = 2.5, 95% CI: 1.2, 5.2) had a higher possibility of miscarriage than those who did not use cosmetics. There was negative association between the use of gel or soap and the cleavage rate among women aged < 30 years old (P for interaction = 0.01). Among women with BMI ≥ 24 kg/m2, the use of gel or soap was negatively associated with the blastocyst formation rate (P for interaction = 0.04), while cosmetics use was negatively associated with the maturation rate (P for interaction = 0.001).ConclusionOur findings suggest that the use of PCPs in women of reproductive age have a potential adverse impact on IVF/ICSI outcomes, particularly skin care and cosmetic products.

Abstract Recovering the global accurate complex physics field from limited sensors is critical to the measurement and control of the engineering system. General reconstruction methods for recovering the field, especially the deep learning with more parameters and better representational ability, usually require large amounts of labeled data which is unaffordable in practice. To solve the problem, this paper proposes uncertainty guided ensemble self-training (UGE-ST), using plentiful unlabeled data to improve reconstruction performance and reduce the required labeled data. A novel self-training framework with the ensemble teacher and pre-training student designed to improve the accuracy of the pseudo-label and remedy the impact of noise is first proposed. On the other hand, uncertainty guided learning is proposed to encourage the model to focus on the highly confident regions of pseudo-labels and mitigate the effects of wrong pseudo-labeling in self-training, improving the performance of the reconstruction model. Experiments including the airfoil velocity and pressure field reconstruction and the electronic components’ temperature field reconstruction indicate that our UGE-ST can save up to 90% of the data with the same accuracy as supervised learning.

Abstract Background To explore the effect of intervention programs constructed under the guidance of the comprehensive unit-based safety program (CUSP) model on chemotherapy-induced nausea and vomiting (CINV) in patients with ovarian cancer. Method According to the time of admission, 90 ovarian cancer chemotherapy patients in the first affiliated Hospital of Anhui Medical University from June 2019 to September 2020 were divided into an intervention group and a control group with 45 cases each. Both groups of patients received routine intervention, and the intervention group implemented the CUSP program on this basis. The intervention lasted 8 months. Before and after the intervention, the patients in the ward were used the Multinational Association of Supportive Care in Cancer (MASCC) Antiemesis Tool, the Functional Living Index-Emesis (FLIE), and the Hospital Anxiety and Depression Scale (HADS) for the effect evaluation. Abstract Deep Neural Networks have been successfully applied in hyperspectral image classification. However, most of prior works adopt general deep architectures while ignore the intrinsic structure of the hyperspectral image, such as the physical noise generation. This would make these deep models unable to generate discriminative features and provide impressive classification performance. To leverage such intrinsic information, this work develops a novel deep learning framework with the noise inclined module and denoise framework for hyperspectral image classification. First, the spectral signature of hyperspectral image is modeled with the physical noise model to describe the high intra‐class variance of each class and great overlapping between different classes in the image. Then, a noise inclined module is developed to capture the physical noise within each object and a denoise framework is then followed to remove such noise from the object. Finally, the CNN with noise inclined module and the denoise framework is developed to obtain discriminative features and provides good classification performance of hyperspectral image. Experiments are conducted over two commonly used real‐world datasets and the experimental results show the effectiveness of the proposed method. The present study aimed to investigate the impacts of dietary standardized ileal digestible lysine to net energy (SID Lys:NE) ratio on lipid metabolism in pigs fed high-wheat diets. Thirty-six crossbred growing barrows (65.20 ± 0.38 kg) were blocked into two treatment groups, fed high-wheat diets with either a high SID Lys:NE ratio (HR) or a low SID Lys:NE ratio (LR). Each treatment group consisted of three replicates, with six pigs per pen in each replicate. The diminishing dietary SID Lys:NE ratio exhibited no adverse impacts on the carcass trait (p > 0.05) but increased the marbling score of the longissimus dorsi muscle (p < 0.05). Meanwhile, LR diets tended to increase the serum triglyceride concentration (p < 0.1). LR diets upregulated fatty acid transport protein 4 and acetyl-coA carboxylase α expression levels and downregulated the expression level of adipose triglyceride lipase (p < 0.05). LR diets improved energy metabolism via decreasing the expression levels of AMP-activated protein kinase (AMPK) α1, sirtuin 1 (SIRT1), and peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) (p < 0.05). Additionally, LR diets stimulated hepatic bile acid synthesis via upregulating the expression levels of cytochrome P450 family 7 subfamily A member 1 and cytochrome P450 family 27 subfamily A member 1, and downregulating farnesol X receptor (FXR) and small heterodimer partner (SHP) expression levels (p < 0.05). A lowered SID Lys:NE ratio affected the colonic microbial composition, characterized by increased relative abundances of YRC22, Parabacteroides, Sphaerochaeta, and Bacteroides, alongside a decreased in the proportion of Roseburia, f_Lachnospiraceae_g_Clostridium, Enterococcus, Shuttleworthia, Exiguobacterium, Corynebacterium, Subdoligranulum, Sulfurospirillum, and Marinobacter (p < 0.05). The alterations in microbial composition were accompanied by a decrease in colonic butyrate concentration (p < 0.1). The metabolomic analysis revealed that LR diets affected primary bile acid synthesis and AMPK signaling pathway (p < 0.05). And the mantel analysis indicated that Parabacteroides, Sphaerochaeta, f_Lachnospiraceae_g_Clostridium, Shuttleworthia, and Marinobacter contributed to the alterations in body metabolism. A reduced dietary SID Lys:NE ratio improves energy metabolism, stimulates lipogenesis, and inhibits lipolysis in finishing pigs by regulating the AMPKα/SIRT1/PGC-1α pathway and the FXR/SHP pathway. Parabacteroides and Sphaerochaeta benefited bile acids synthesis, whereas f_Lachnospiraceae_g_Clostridium, Shuttleworthia, and Marinobacter may contribute to the activation of the AMPK signaling pathway. Overall, body metabolism and colonic microbiota collectively controlled the lipid metabolism in finishing pigs.

Murrayae exotica is a traditional Chinese medicine widely grown in southeast China. A Box–Behnken design of response surface methodology was employed to further optimise ultrasonic-assisted extraction conditions for M. exotica leaves total flavonoids (MELTF). The results showed that the optimised extraction conditions were an ultrasonic power of 240 W, an ultrasonic temperature of 60 °C, a solvent concentration of 76%, an ultrasonic time of 55 min and a liquid–solid ratio of 22 mL · g−1. Under these conditions, 8.59 ± 0.34 mg · g−1 was achieved as the mean experimental value of extraction yield, which amounts to 2.56 times that of ethanol leaching extraction. As compared to MELTF, MELTF after purification (MELPTF) had a higher purity of 9.96%, which was nearly nine times higher than that of MELTF (1.26%). As compared to MELTF, MELPTF had higher α-glucosidase and α-amylase inhibitory activities as well as DPPH· and ABTS·+ scavenging activities with IC50 values of 0.021, 0.094, 0.245 and 0.113 mg · mL−1, which are 1.33, 2.12, 3.17 and 1.78 times higher than those of MELTF (0.028, 0.199, 0.777 and 0.201 mg · mL−1). The study thus demonstrates the eligibility of MELPTF to be considered as a multifunctional bioactive ingredient having potential applications in anti-hyperglycaemic pharmaceutical formulation and as an antioxidant in functional foods.

Abstract Background Lactulose as an effective prebiotic protects intestinal mucosal injury. Bacillus coagulans is widely used in feed additives because of its ability to promote intestinal health. Our previous study suggests that the combination of lactulose and Bacillus coagulans may be a good candidate as alternative for antibiotic growth promoters. However, the in vivo effects of lactulose and Bacillus coagulans on growth and intestinal health under immune challenge in piglets remains unclear. The objective of this study is to explore the protective effects of synbiotic containing lactulose and Bacillus coagulans on the intestinal mucosal injury and barrier dysfunction under immune challenge in weaned piglets. Methods Twenty four weaned piglets were assigned to 4 groups. Piglets in the CON-saline and LPS-LPS group were fed the basal diet, while others were fed either with chlortetracycline (CTC) or synbiotic mixture of lactulose and Bacillus coagulans for 32 d before injection of saline or lipopolysaccharide (LPS). Piglets were sacrificed 4 h after LPS injection to collect samples to determine intestinal morphology, integrity and barrier functions as well as relative genes and proteins. Results Our data showed that no differences were observed in the growth performance of the four test groups. LPS injection induced higher serum diamine oxidase activities, D-lactic acid levels, and endotoxin status, lower villus height and ratio of villus height to crypt depth, greater mRNA and lower protein expression related tight junction in both jejunum and ileum. In addition, a higher apoptosis index, and protein expression of Bax and caspase-3 were also observed in the LPS challenge group. Interestingly, dietary synbiotic mixture with lactulose and Bacillus coagulans protected against LPS-induced intestinal damage, barrier dysfunction and higher apoptosis as well as CTC. Conclusions Our data suggest that dietary supplementation of synbiotic mixture with lactulose and Bacillus coagulans showed resilience to LPS-induced intestinal morphological damage, barrier dysfunction and aggressive apoptosis in piglets as well as the protective effects of CTC. These results indicate that synbiotic mixture of lactulose and Bacillus coagulans showed beneficial effects on performance and resilience to acute immune stress in weaned piglets.

Abstract The high-resolution (HR) spatio-temporal flow field plays a decisive role in describing the details of the flow field. In the acquisition of the HR flow field, traditional direct numerical simulation (DNS) and other methods face a seriously high computational burden. To address this deficiency, we propose a novel multi-scale temporal path UNet (MST-UNet) model to reconstruct temporal and spatial HR flow fields from low-resolution (LR) flow field data. Different from the previous super-resolution (SR) model, which only takes advantage of LR flow field data at instantaneous (SLR) or in a time-series (MTLR), MST-UNet introduces multi-scale information in both time and space. MST-UNet takes the LR data at the current frame and the predicted HR result at the previous moment as the model input to complete the spatial SR reconstruction. On this basis, a temporal model is introduced as the inbetweening model to obtain HR flow field data in space and time to complete spatio-temporal SR reconstruction. Finally, the proposed model is validated by the spatio-temporal SR task of the flow field around two-dimensional cylinders. Experimental results show that the outcome of the MST-UNet model in spatial SR tasks is much better than those of SLR and MTLR, which can greatly improve prediction accuracy. In addition, for the spatio-temporal SR task, the spatio-temporal HR flow field predicted by the MST-UNet model has higher accuracy either.

IntroductionThe gut microbiota closely relates to host health, whereas the relationship between gut microbiota and testosterone during the development of Meishan male pigs remains unclear. This study investigated the fecal microbiota composition and testosterone level during development in Meishan male pigs.MethodsFresh fecal samples of 20 healthy Meishan male pigs were individually collected at 10 and 22 weeks (wk) of age for testosterone content detection and bacteria pyrosequencing analysis. Anaerobic culture experiment of fecal bacteria in vitro was performed for bacteria pyrosequencing analysis.ResultsThe fecal testosterone content increased significantly from 10 weeks (wk) to 22 wk of age (P < 0.05). Meanwhile, the boars at 22 wk had a lower abundance of phylum Bacteroidetes and Proteobacteria, and genus Alloprevotella, Prevotella_1, Prevotellaceae_NK3B31_group, and Streptococcus in the fecal microbiota composition (P < 0.05). but higher proportions of the phylum Actinobacteria, Firmicutes, Kiritimatiellaeota, and Tenericutes, and genus Clostridium_sensu_stricto_1, Muribaculaceae and Terrisporobacter than that at 10 wk (P < 0.05), and the Firmicutes to Bacteroidetes ratio was higher at 22 wk than 10 wk (P < 0.05). Moreover, the fecal testosterone level significantly correlated with the relative abundance of the phylum Actinobacteria, Firmicutes, and Tencuteseri, and genus Alloprevotella, Clostridium_sensu_stricto_1, Muribaculaceae, Prevotella_1 and Streptococcus. Many studies have showed that phthalates have reproductive and embryonic toxicity, while the potential mechanisms are mostly unknown. Inflammation may play a mediating part in phthalate exposure and adverse reproductive endpoints. A cross-sectional survey was conducted to investigate the associations of phthalate metabolites with inflammatory cytokines in the follicular fluid (FF) of women undergoing in vitro fertilization (IVF). We determined the levels of eight phthalate metabolites and five cytokines in the FF of 76 women, including interleukin (IL)- 6, IL-8, IL-10, monocyte chemoattractant protein-1 (MCP-1) and tumor necrosis factor-α (TNF-α). The associations of individual phthalate exposure with cytokines in FF samples were explored by multiple linear regression. We further evaluated the combined effects of multiple phthalate exposures on FF levels of cytokines by using Bayesian kernel machine regression (BKMR) models. We found that there was a positive relationship between mono-ethyl phthalate (MEP) and IL-6 in the FF (percent change:12.4%; 95% CI: 1.3%, 24.9%). In contrast, elevated mono-benzyl phthalate (MBzP), mono(2-ethylhexyl) phthalate (MEHP) and %MEHP levels were associated with decreased MCP-1. In the BKMR models, phthalate metabolite mixtures were positively associated with TNF-α when the mixtures were lower than 65th percentile compared with their medians. Abstract Convolutional neural networks (CNNs) have successfully demonstrated their powerful predictive performance in a variety of tasks. However, it remains a challenge to estimate the uncertainty of these predictions simply and accurately. Deep Ensemble is widely considered the state-of-the-art method which can estimate the uncertainty accurately, but it is expensive to train and test. MC-Dropout is another popular method that is less costly but lacks the diversity of predictions resulting in less accurate uncertainty estimates. To combine the benefits of both, we introduce a ReLU-Based Uncertainty Estimation (RBUE) method. Instead of using the randomness of the Dropout module during the test phase (MC-Dropout) or using the randomness of the initial weights of CNNs (Deep Ensemble), RBUE uses the randomness of activation function to obtain diverse outputs in the testing phase to estimate uncertainty. Under the method, we propose strategy MC-DropReLU and develop strategy MC-RReLU. The uniform distribution of the activation function’s position in CNNs allows the randomness to be well transferred to the output results and gives a more diverse output, thus improving the accuracy of the uncertainty estimation. Moreover, our method is simple to implement and does not need to modify the existing model. We experimentally validate the RBUE on three widely used datasets, CIFAR10, CIFAR100, and TinyImageNet. The experiments demonstrate that our method has competitive performance but is more favorable in training time.

Soybean (Glycine max (L.) Merr.) is a globally significant crop, widely cultivated for oilseed production and animal feeds. In recent years, the rapid growth of multi-omics data from thousands of soybean accessions has provided unprecedented opportunities for researchers to explore genomes, genetic variations, and gene functions. To facilitate the utilization of these abundant data for soybean breeding and genetic improvement, the SoybeanGDB database (https://venyao.xyz/SoybeanGDB/) was developed as a comprehensive platform. SoybeanGDB integrates high-quality de novo assemblies of 39 soybean genomes and genomic variations among thousands of soybean accessions. Genomic information and variations in user-specified genomic regions can be searched and downloaded from SoybeanGDB, in a user-friendly manner. To facilitate research on genetic resources and elucidate the biological significance of genes, SoybeanGDB also incorporates a variety of bioinformatics analysis modules with graphical interfaces, such as linkage disequilibrium analysis, nucleotide diversity analysis, allele frequency analysis, gene expression analysis, primer design, gene set enrichment analysis, etc. In summary, SoybeanGDB is an essential and valuable resource that provides an open and free platform to accelerate global soybean research.

Abstract Objectives To characterize the clinical features and outcomes of childhood-onset primary Sjögren’s syndrome (pSS). Methods Patients less than 18 years old who were diagnosed with pSS by paediatric rheumatologists were included, and all patients were applied the 2002 American-European Consensus Group (ACEG) criteria, the 2016 American College of Rheumatology/European League Against Rheumatism (ACR/EULAR) criteria for pSS, or the 1999 proposed juvenile pSS criteria. The electronic medical records of patients with pSS from 2013 to 2020 were collected and analysed. Results Thirty-nine patients were included. Of them, 27 (69.2%), 38 (97.4%) and 35 (89.7%) patients fulfilled the AECG criteria, ACR/EULAR criteria and proposed juvenile pSS criteria, respectively. The female:male ratio was 3.9:1. The median ages at first signs or symptoms and at diagnosis were 9.2 (4.7, 14.5) years and 10.9 (6.3, 15.0) years, respectively. The main clinical manifestations were rash or purpura (20, 51.3%), followed by fever (12, 30.8%), glandular enlargement/recurrent parotitis (10, 25.6%), and dry mouth and/or dry eyes (9, 23.1%). Twenty-eight (56.4%) patients had systemic damage, the most common of which was haematological involvement (14, 35.9%), followed by hepatic (13, 33.3%) and renal involvement (8, 20.5%). Thirty-eight (97.4%) patients underwent labial minor salivary gland biopsy, and all exhibited focal lymphocytic sialadenitis. All patients had a global ESSDAI score ≥ 1 at diagnosis, and the median total score at diagnosis was 8 (2, 31). Thirty-six (92.3%) patients were followed up for a median time of 23.6 (7.9, 79.5) months, and three patients developed systemic lupus erythematosus (SLE) at follow-up times of 13.3, 38.8 and 63.8 months. Conclusions The presentation of childhood-onset pSS is atypical, and extraglandular manifestations and systemic involvement are more common than in adult-onset pSS. Labial salivary gland biopsy is vital for patients with probable pSS. Some patients may develop SLE over time.

Abstract Ionomics-metabolomics association analysis is a novel method to elucidating the potential mechanisms underlying the effects of dietary copper on the overall health parameters of suckling piglets model. Few studies have elucidated the relationship between the changes of ionic and metabolic homeostasis responses to dietary copper level. The growth performance data was obtained from 180 suckling piglets which access to different copper levels: 6 (low copper diet, LC), 20 (control diet, CON), and 300 (high copper diet, HC) mg·kg−1 copper (based on diet, supplementation from CuSO4), and offered ad libitum from d 14 until weaning at 40 d of age. Dietary high level copper (300 mg·kg−1) increased the ADG and ADFI during d 14 to 28 of piglets. Six elements (Mg, Na, K, P, Cu, and Mn) concentrations significantly changes in hair among the three treatment diets. The significant increased concentrations of Na and K, and decreased concentration of Mg and Mn in 300 mg·kg−1 than 20 mg·kg−1 copper diet was observed. In current study, with the increase in copper level from 20 to 300 mg·kg−1 in diet, the correlation between hair Na, K and Cu, Mn, Zn vanish. Hair Na and K were positively correlated with serum total antioxidant capacity (T-AOC) and negatively correlated with tumor necrosis factor-α (TNF-α). The hair Cu was negatively correlated with serum malondialdehyde (MDA), total bile acid (TBA). The fecal Cu was positively correlated with serum growth hormone (GH). The results suggested that the average daily gain (ADG) in 6 mg·kg−1 copper diet and the average daily feed intake (ADFI) in 20 mg·kg−1 copper diet were decreased than 300 mg·kg−1 copper diet during d 14 to 28 and the ADG was decreased in 6 and 20 mg·kg−1 copper diets in d 29 to 40 of piglets. Dietary 20 mg·kg−1 copper maintain ion homeostasis due to increase the number of positive correlations between macroelements-microelements in hair and serum. Significantly changed Na, K, Mg, Mn and Cu concentrations in hair can reflect the adverse effects of dietary 300 mg·kg−1 copper of suckling piglets. We believe our results may benefit people to gain a better understanding of the ion interactions and metabolic homeostasis of heavy metal elements that are critical to human and animal health.

Day length is a critical environmental factor for regulating animal growth and development. This study aimed to investigate the effects of different day lengths on the developmental changes of growth parameters, testicular sizes, testosterone secretion in Meishan male pigs, and steroidogenesis proteins and melatonin receptors. Fourteen Meishan male pigs (10 weeks (wks) of age) with the same parity, paired in litter and body weight (BW), were evenly allocated into a short-day-length group (SDL, 10 light/14 dark) and long-day-length group (LDL, 14 light/10 dark). After 12 wks of the experiment, the LDL-treated boars had more lying time and less exploring time. The LDL treatment led to significant increases in body height, chest circumference, testicular length, testicular weight, crude protein digestibility, and fecal testosterone at the 10th and 12th wks of the experiment, and cortisol at the 10th wk, compared to the SDL treatment, with no differences in the final BW, testicular width, and epididymis weight. Furthermore, the LDL treatment significantly increased the protein levels of melatonin receptor 1b (MT2), aromatase (CYP19), and steroidogenic factor 1 (SF1) in the testis, with no differences in the protein levels of melatonin receptor 1a (MT1), steroidogenic acute regulatory (StAR), 3β-hydroxysteroid dehydrogenase (3β-HSD), and cholesterol side-chain cleavage enzyme (P450scc). The present study suggests that day length has an effect on the growth and gonadal development in male pigs maybe via MT2 and influences steroid synthesis and secretion in the testis. Therefore, proper day length should be considered in male pig breeding.

Utilizing data from the Tropical Rainfall Measuring Mission (TRMM) satellite’s precipitation radar (PR) and lightning imaging sensor (LIS), this study explores the spatiotemporal distributions of thunderstorm and lightning structures over the Qinghai–Tibet Plateau (QTP), an aspect that has not been explored previously. The structural aspects are crucial when considering the impact of thunderstorm and lightning activity in the atmospheric processes. Thunderstorms over the QTP show clear spatial variations in both vertical height and horizontal extension. In the southern region, the average heights of 20 dBZ and 30 dBZ echo tops typically exceed 11.2 and 9.3 km, respectively. Meanwhile, in the eastern part, the average coverage areas for reflectivity greater than 20 dBZ and 30 dBZ consistently surpass 1000 and 180 km2, respectively. The spatial distribution of thunderstorm vertical development height relative to the surface aligns more closely with the horizontal extension, indicating stronger convection in the eastern QTP. The thunderstorm flash rate shows an eastward and northward prevalence, while the thunderstorm flash density peaks in the western and northeastern QTP, with a minimum in the southeast. Furthermore, in the eastern QTP, lightning duration, spatial expansion, and radiance are more pronounced, with the average values typically exceeding 0.22 s, 14.5 km, and 0.50 J m−2 sr−1 μm−1, respectively. Monthly variations reveal heightened values during the summer season for thunderstorm vertical extension, areas with reflectivity greater than 30 dBZ, and lightning frequency. Diurnal variations highlight an afternoon increase in thunderstorm vertical and horizontal extension, lightning frequency, duration, and spatial scale. From a statistical perspective, under weak convective conditions, lightning length exhibits a positive correlation with thunderstorm convection intensity, contrasting with the opposite relationship suggested by previous studies. This article further analyzes and discusses the correlations between various thunderstorm and lightning structural parameters, enhancing our understanding of the distinctive features of thunderstorm and lightning activities in the QTP.

Introduction: Amisulpride is primarily eliminated via the kidneys. Given the clear influence of renal clearance on plasma concentration, we aimed to explicitly examine the impact of renal function on amisulpride pharmacokinetics (PK) via population PK modelling and Monte Carlo simulations.Method: Plasma concentrations from 921 patients (776 in development and 145 in validation) were utilized.Results: Amisulpride PK could be described by a one-compartment model with linear elimination where estimated glomerular filtration rate, eGFR, had a significant influence on clearance. All PK parameters (estimate, RSE%) were precisely estimated: apparent volume of distribution (645 L, 18%), apparent clearance (60.5 L/h, 2%), absorption rate constant (0.106 h−1, 12%) and coefficient of renal function on clearance (0.817, 10%). No other significant covariate was found. The predictive performance of the model was externally validated. Covariate analysis showed an inverse relationship between eGFR and exposure, where subjects with eGFR= 30 mL/min/1.73 m2 had more than 2-fold increase in AUC, trough and peak concentration. Simulation results further illustrated that, given a dose of 800 mg, plasma concentrations of all patients with renal impairment would exceed 640 ng/mL.Discussion: Our work demonstrated the importance of renal function in amisulpride dose adjustment and provided a quantitative framework to guide individualized dosing for Chinese patients with schizophrenia.

Magnesium hydride (MGH), a highly promising hydrogen-producing substance/additive for hydrogen production through its hydrolysis reaction, has the potential to enhance broiler production. However, before incorporating MGH as a hydrogen-producing additive in broiler feed, it is crucial to fully understand its impact on microbiota and metabolites. In vitro fermentation models provide a fast, reproducible, and direct assessment tool for microbiota metabolism and composition. This study aims to investigate the effects of MGH and coated-magnesium hydride (CMG) on fermentation characteristics, as well as the microbiota and metabolome in the culture of in vitro fermentation using cecal inocula from broilers. After 48 h of incubation, it was observed that the presence of MGH had a significant impact on various factors. Specifically, the content of N-NH3 decreased, while the total hydrogen gas and total SCFAs increased. Furthermore, the presence of MGH promoted the abundance of SCFA-producing bacteria such as Ruminococcus, Blautia, Coprobacillus, and Dysgonomonas. On the other hand, the presence of CMG led to an increase in the concentration of lactic acid, acetic acid, and valeric acid. Additionally, CMG affected the diversity of microbiota in the culture, resulting in an enrichment of the relative abundance of Firmicutes, as well as genera of Lactobacillus, Coprococcus, and Eubacterium. Conversely, the relative abundance of the phylum Proteobacteria and pathogenic bacteria Shigella decreased. Metabolome analysis revealed that MGH and CMG treatment caused significant changes in 21 co-regulated metabolites, primarily associated with lipid, amino acid, benzenoids, and organooxygen compounds. Importantly, joint correlation analysis revealed that MGH or CMG treatments had a direct impact on the microbiota, which in turn indirectly influenced metabolites in the culture. In summary, the results of this study suggested that both MGH and coated-MGH have similar yet distinct positive effects on the microbiota and metabolites of the broiler cecal in an in vitro fermentation model.

Abstract Considering the limited communication resources and slow convergence speed of multi-unmanned aerial vehicle (UAV) systems, this paper presents a finite-time even-triggered control framework for multi-UAV systems to achieve formation-containment tracking control. First, a virtual leader with time-varying output is introduced so that the trajectory of the whole system can be manipulated in real time. Second, the finite-time control enables that the systematic error converge to a small neighborhood of origin in finite time. Third, in order to save communication resources, an event-triggering function is developed to generate the control event sequences, which avoids continuous update of the controller. Rigorous proof shows the finite-time stability of the proposed control algorithm, and Zeno behavior is strictly excluded for each UAV. Finally, some numerical simulations are given to verify the effectiveness of the proposed controllers.

Abstract Background Allogeneic hematopoietic stem cell transplantation (allo-HSCT) is an important curative therapy for adult acute lymphoblastic leukemia (ALL). For patients who lack a human leukocyte antigen (HLA)-matched sibling donor, unrelated cord blood (UCB) is an alternative graft option. Previous studies have focused mainly on all T- and B-cell ALL (B-ALL) patients, while data related specifically to adult B-ALL patients after UCB transplantation (UCBT) are scarce. Methods We retrospectively compared the outcomes of UCBT and HLA-matched sibling transplantation (MST) in the treatment of adult B-ALL patients in complete remission (CR) at our center. From June 2006 to December 2020, 156 adult B-ALL patients who achieved CR before transplantation were enrolled. The main clinical outcomes of UCBT and MST were analyzed. Results Hematopoietic recovery was significantly faster in MST recipients than in UCBT recipients. Abstract Based on the detection data of the Guangdong‐Hong Kong‐Macau lightning location system from 2013 to 2022, this paper analyzes the characteristics of lightning strikes in an offshore wind turbine cluster (WTC) in Guangdong. This is the first time to quantify the lightning strikes difference between the inner and outer wind turbines (WTs) and discuss the impact mechanisms of WTCs on the characteristics of lightning strikes by the simulation results of electric potential distribution. The observation results show that the stroke density, cloud‐to‐ground (CG) flash density, and average peak current within the affected area increase by 10.1%, 11%, and 5.1%, respectively, but the lightning multiplicity changes little. The inner and outer WTs show different characteristics of lightning strikes. The stroke density, lightning multiplicity, and average peak current in the area of outer WTs increase by 21%, 4.1%, and 6.8%, respectively, while in the area of inner WTs decrease by 20.2%, 19%, and 3.9%, respectively. The CG flash density in the area of outer WTs increases by 16.1% but changes little in the area of inner WTs. The simulation results show there is a significant difference in the electric potential distribution between the inner and outer WTs: the distortion effect of the outer WTs on spatial electric potential is stronger than that of the inner WTs at a greater height, but the difference is not obvious at a lower height. Such a difference in electric potential distribution leads to different characteristics of lightning strikes between the inner and outer WTs.

Abstract We previously developed shinyCircos, an interactive web application for creating Circos diagrams, which has been widely recognized for its graphical user interface and ease of use. Here, we introduce shinyCircos‐V2.0, an upgraded version of shinyCircos that includes a new user interface with enhanced usability and many new features for creating advanced Circos plots. To help users get started with shinyCircos‐V2.0, we provide detailed tutorials and example input data sets. The application is available online at https://venyao.xyz/shinyCircos/ and https://asiawang.shinyapps.io/shinyCircos/, or can be installed locally using the source code deposited in GitHub (https://github.com/YaoLab-Bioinfo/shinyCircos-V2.0).

Rice bran is a high-quality and renewable livestock feed material rich in nutrients and bioactive substances. To investigate the effects of dietary supplementation with fermented heat-treated rice bran on the performance, apparent digestibility of nutrients, cecal microbiota and metabolites in laying hens, a total of 128 18-week-old Hy-Line brown layers were randomly assigned to four treatment groups: 2.5% HRB (basal diet contained 2.5% heat-treated rice bran), 5.0% HRB (5.0% heat-treated rice bran), 2.5% FHRB (2.5% fermented heat-treated rice bran), 5.0% FHRB (5.0% fermented heat-treated rice bran). Results showed that FHRB supplementation significantly increased the average daily feed intake (ADFI) during 25–28 weeks, and improved apparent digestibility of dry matter (DM), crude protein (CP), ether extract (EE) and crude fiber (CF) in laying hens. Moreover, feeding 5.0% of HRB and FHRB resulted higher egg production (EP) and average egg weight (AEW) during the feeding period, and decreased the feed conversion ratio (FCR) during 21 to 28 weeks. The alpha and beta diversity indices indicated that FHRB altered the cecal microbiota. In particular, dietary supplementation with FHRB significantly increased the relative abundances of Lachnospira and Clostridium. Compared with the 2.5% level of supplementation, supplementing 5.0% HRB and 5.0% FHRB increased the relative abundances of Firmicutes, Ruminococcus and Peptococcus, and lowered the relative abundance of Actinobacteria. Furthermore, dietary FHRB supplementation significantly increased the concentration of short-chain fatty acids in cecum and changed the overall metabolome. The results of correlation analysis showed a close interaction between cecal microbiota, metabolites and apparent digestibility of nutrients. Taken together, we revealed that FHRB supplementation can induce characteristic structural and metabolic changes in the cecal microbiome, which could potentially promote nutrient digestion and absorption, and improve the production performance of laying hens.

Umbilical cord blood transplantation (UCBT) and peripheral blood stem cell transplantation (PBSCT) are effective allogeneic treatments for patients with malignant and non-malignant refractory hematological diseases. However, the differences in the immune cell reconstitution and the immune reactions during initial stages post-transplantation are not well established between UCBT and PBSCT. Therefore, in this study, we analyzed the differences in the immune reactions during the early stages (days 7-100 post-transplantation) such as pre-engraftment syndrome (PES), engraftment syndrome (ES), and acute graft-versus-host disease (aGVHD) and the immune cell reconstitution between the UCBT and the PBSCT group of patients. We enrolled a cohort of patients that underwent UCBT or PBSCT and healthy controls (n=25 each) and evaluated their peripheral blood mononuclear cell (PBMC) samples and plasma cytokine (IL-10 and GM-CSF) levels using flow cytometry and ELISA, respectively. Our results showed that the incidences of early immune reactions such as PES, ES, and aGVHD were significantly higher in the UCBT group compared to the PBSCT group. Furthermore, in comparison with the PBSCT group, the UCBT group showed higher proportion and numbers of naïve CD4+ T cells, lower proportion and numbers of Tregs, higher proportion of CD8+ T cells with increased activity, and higher proportion of mature CD56dim CD16+ NK cells during the early stages post-transplantation. Moreover, the plasma levels of GM-CSF were significantly higher in the UCBT group compared to the PBSCT group in the third week after transplantation. Overall, our findings demonstrated significant differences in the post-transplantation immune cell reconstitution between the UCBT and the PBSCT group of patients. These characteristics were associated with significant differences between the UCBT and the PBSCT groups regarding the incidences of immune reactions during the early stages post transplantation.

This study investigates the location accuracy distribution of the lightning detection networks (LDNs) deployed on inclined terrains, an aspect frequently encountered in complex terrains but hitherto disregarded in previous studies. By designing eight substation LDNs deployed on slope-type (ST), mountain-type (MT) and basin-type (BT) terrains, respectively, we employed Monte Carlo simulations to analyze their spatial location accuracy distribution based on time-of-arrival technology. Significant differences among the LDNs on inclined terrains and between them and the LDN on plain-type (PT) terrain were revealed. Compared to the PT LDN, LDNs on inclined terrains exhibited a reduction in high-precision location regions and a shift in the distribution pattern of location accuracy. The ST LDN showed marked deviations in the high-precision vertical location toward the lower slope side with increases in the elevation angle and consistently smaller high-precision vertical location areas compared to MT and BT LDNs. The variations in elevation angles of MT and BT LDNs had a substantial impact on the spatial distribution patterns of both horizontal and vertical location accuracy, with BT LDNs featuring larger vertical high-precision areas than MT LDNs. Our conclusions were further corroborated through an analysis of an actual LDN, which combined characteristics from both ST and MT terrain patterns.

Screening and identifying the active compounds in foods are important for the development and utilization of functional foods. In this study, the anti-enteritis activity of ethanol extract from Camellia oleifera oil (PECS) was quickly evaluated using a Smurf Drosophila model and the metabolomics approach, combined with molecular docking techniques, were performed to rapidly screen and identify compounds with potential anti-enteritis activity in PECS. PECS showed good anti-enteritis activity and inhibited the activity of 5-lipoxygenase (LOX), cyclooxygenase 2 (COX-2) and inducible nitric oxide synthase (iNOS). In particular, wighteone and p-octopamine were newly identified in C. oleifera oil and were proven to have good anti-enteritis activity. The inhibitory activity of kaempferitrin (IC50 = 0.365 mmol L−1) was higher than that of wighteone (IC50 = 0.424 mmol L−1) and p-octopamine (IC50 = 0.402 mmol L−1). Of note, the IC50 value of salazosulfapyridine was 0.810 mmol L−1. Inhibition of LOX activity is likely one of the anti-enteritis mechanisms of PECS. These new findings lay the foundation for further investigations into the underlying mechanisms of anti-enteritis activity in C. oleifera oil.

Latent tuberculosis infection (LTBI) widely exists in patients with unexplained infertility, and whether LTBI would affect the ovarian reserve and pregnancy outcome of infertile women undergoing intrauterine insemination (IUI) is still unknown. A single-center, retrospective, cohort study was designed that included infertile women undergoing IUI at the Department of Reproductive Medicine, Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology in Wuhan, China, from January 2018 to December 2020. The primary outcomes of this study were ovarian reserve and live birth rate. Secondary outcomes included pregnancy outcomes and maternal and neonatal complications. As a result, 3066 IUI cycles were eventually enrolled in this study. Of these women, 9.6% (295/3066) had LTBI evidence. After propensity score matching (PSM), there was no significant difference in the baseline between the LTBI and non-LTBI groups. The data showed that women who had LTBI had trends toward lower biochemical pregnancy rates (12.9% vs. 17.7%, p-value 0.068), lower clinical pregnancy rates (10.8% vs. 15.1%, p-value 0.082) and lower live birth rates (8.1% vs. 12.1%, p-value 0.076), with no significant differences. There were also no significant differences in ovarian reserve and other secondary outcomes between the two groups. In conclusion, there were no significant differences in ovarian reserve, perinatal or neonatal complications between women with and without LTBI. Women with LTBI tended to have worse pregnancy outcomes after receiving IUI, but the difference was not significant.

Abstract As a major food crop and model organism, rice has been mostly studied with the largest number of functionally characterized genes among all crops. We previously built the funRiceGenes database including ~ 2800 functionally characterized rice genes and ~ 5000 members of different gene families. Since being published, the funRiceGenes database has been accessed by more than 54,400 users with over 540,000 pageviews. The funRiceGenes database has been continuously updated with newly cloned rice genes and newly published literature, based on the progress of rice functional genomics studies. Up to Nov 2021, ~ 4100 functionally characterized rice genes and ~ 6000 members of different gene families were collected in funRiceGenes, accounting for 22.3% of the 39,045 annotated protein-coding genes in the rice genome. Here, we summarized the update of the funRiceGenes database with new data and new features in the last 5 years.

Both ceftiofur (CTO) and high copper are widely utilized in animal production in China, and the occurrence of CTX-M-carrying Escherichia coli in food-producing animals is increasing. There are some specific associations between in-feed high-level copper and antibiotic resistance, but research in Gram-negative bacteria such as E. coli remains scarce. This study aimed to evaluate the effect of high-copper diet on the horizontal transfer of blaCTX−M−1 among E. coli. A total of 32 male SPF rats aged 21 days were randomly assigned to the following four groups: control (6 mg/kg in-feed copper, C−), high copper (240 mg/kg in-feed copper, H−), CTO (6 mg/kg in-feed copper with oral CTO administration, C+), and high copper plus CTO (240 mg/kg in-feed copper with oral CTO administration, H+). All rats were orally inoculated with an E. coli strain harboring a conjugative plasmid carrying blaCTX−M−1, and the C+ and H+ groups were given 10 mg/kg of body weight (BW) CTO hydrochloride at 26, 27, and 28 days, while the C− and H− groups were given salad oil at the same dose. Fecal samples collected at different time points were used for the enumeration of E. coli on Mac plates or for molecular analysis using PCR, pulsed-field gel electrophoresis (PFGE), S1-PFGE, and Southern-blot hybridization. The results showed that the number of the blaCTX−M−1 gene in the H− group was higher and that the loss speed of this gene was slower compared with the C− group. After administration of CTO, the counts of cefotaxime-resistant E. coli were significantly higher in the C+ group than that in the corresponding control group (C+ vs. C−; H+ vs. H−). In the in vitro test, the results showed that the transfer rates of the conjugation induced by the H− (12 mmol/L) group were significantly higher than that of low copper (2 mmol/L) group. The indigenous sensitive isolates, which were homologous to the blaCTX−M-positive isolates of rat feces, were found by PFGE. The further analysis of S1-PFGE and Southern-blot hybridization confirmed that the blaCTX−M−1 gene in new transconjugants was derived from the inoculated strain. Taken together, high-copper diet facilitates the horizontal transfer and maintenance of the resistant genes in the intestine of rats, although the effects of antibiotics on bacterial resistance appearance and maintenance are more obvious.

ABSTRACT Chromosome evolution drives species evolution, speciation, and adaptive radiation. Accurate genome assembly is crucial to understanding chromosome evolution of species, such as dikaryotic fungi. Rust fungi (Pucciniales) in dikaryons represent the largest group of plant pathogens, but the evolutionary process of adaptive radiation in Pucciniales remains poorly understood. Here, we report a gapless genome for the wheat leaf rust fungus Puccinia triticina determined using PacBio high-fidelity (HiFi) sequencing. This gapless assembly contains two sets of chromosomes, showing that one contig represents one chromosome. Comparisons of homologous chromosomes between the phased haplotypes revealed that highly frequent small-scale sequence divergence shapes haplotypic variation. Genome analyses of Puccinia triticina along with other rusts revealed that recent transposable element bursts and extensive segmental gene duplications synergistically highlight the evolution of chromosome structures. Comparative analysis of chromosomes indicated that frequent chromosomal rearrangements may act as a major contributor to rapid radiation of Pucciniales. This study presents the first gapless, phased assembly for a dikaryotic rust fungus and provides insights into adaptive evolution and species radiation in Pucciniales. IMPORTANCE Rust fungi (Pucciniales) are the largest group of plant pathogens. Adaptive radiation is a predominant feature in Pucciniales evolution. Chromosome evolution plays an important role in adaptive evolution. Accurate chromosome-scale assembly is required to understand the role of chromosome evolution in Pucciniales. We took advantage of HiFi sequencing to construct a gapless, phased genome for Puccinia triticina. Further analyses revealed that the evolution of chromosome structures in rust lineage is shaped by the combination of transposable element bursts and segmental gene duplications. Chromosome comparisons of Puccinia triticina and other rusts suggested that frequent chromosomal arrangements may make remarkable contributions to high species diversity of rust fungi. Our results present the first gapless genome for Pucciniales and shed light on the feature of chromosome evolution in Pucciniales.

This paper reports a numerical research on MEMS (microelectromechanical system) micronozzles through multiphysics coupling simulation along with design optimization based on simulation results. The micronozzle, which is a core component of the electrothermal microthruster, features a micron-scale geometry, a 2-dimensional (2D) Laval configuration, a rectangular cross section, and a highly thermal conductive silicon wall due to MEMS fabrication. As a result, viscous loss in the flow field and heat transfer to the nozzle wall can strongly influence nozzle performance, namely, thrust force and specific impulse. To accurately understand the flow field inside the micronozzle and how the highly thermal conductive silicon wall interacts with gas flow, a numerical simulation that couples fluid dynamics field and solid heat transfer field is employed in the research. The influence of different structural parameters on micronozzle performance is then investigated to set a basis for design optimization. The optimum design of the linear expander micronozzle is obtained through constrained optimization by linear approximation. To further improve micronozzle performance, the bell-shaped expander is adapted. The optimization result shows that the bell-shaped expander is not suitable for micronozzle featuring 2D Laval configuration, and the reason behind the phenomenon is thoroughly discussed.

Introduction: Gliadins are the major components of gluten proteins with vital roles on properties of end-use wheat product and health-relate quality of wheat. However, the function and regulation mechanisms of γ-gliadin genes remain unclear. Objectives: Dissect the effect of DNA methylation in the promoter of γ-gliadin gene on its expression level and gluten strength of wheat. Methods: The prokaryotic expression and reduction–oxidation reactions were performed to identify the effect of TaGli-γ-2.1 on dough strength. Bisulfite analysis and 5-Aza-2′-deoxycytidine treatment were used to verify the regulation of TaGli-γ-2.1 expression by pTaGli-γ-2.1 methylation. The content of gluten proteins composition was measured by RP-HPLC, and the gluten strength was measured by Gluten Index and Farinograph. Results: TaGli-γ-2.1 was classified into a subgroup of γ-gliadin multigene family and was preferentially expressed in the later period of grain filling. Addition of TaGli-γ-2.1 protein fragment into strong gluten wheat flour significantly decreased the stability time. Hypermethylation of three CG loci of pTaGli-γ-2.1 conferred to lower TaGli-γ-2.1 expression. Treatment with 5-Aza-2′-deoxycytidine in seeds of strong gluten wheat varieties increased the expression levels of TaGli-γ-2.1. Furthermore, the accumulations of gliadin and γ-gliadin were significantly decreased in hypermethylated wheat varieties, corresponding with the increasing of gluten index and dough stability time. Conclusion: Epigenetic modification of pTaGli-γ-2.1 affected gluten strength by modulating the proportion of gluten proteins. Hypermethylation of pTaGli-γ-2.1 is a novel genetic resource for enhancing gluten strength in wheat quality breeding.
