Although liverworts are widely distributed around the world with a large number of extant species, reliable fossil records are relatively rare. Here, we report a new species, Ricciopsis baojishanensis Han and Yan, n. sp. (Ricciaceae) and an unnamed species, Hepaticites sp. from the Late Triassic Nanying’er Formation in Baojishan Basin, Baiyin City, Gansu Province, Northwest China. The generic designation is based on detailed comparison of the gross morphology with related fossil and extant species. The new species is characterized by its rosette-forming thallus, dichotomous branching, ribbon-like segments and entire margins. The current fossils represent the first record of liverwort from the Late Triassic in Baojishan Basin, Gansu Province. Based on the different fossil records of the Ricciaceae, we suggest that these taxa were widely distributed during Late Triassic to Oligocene worldwide, mainly in warm temperate and tropical environments, similar with their current distribution. The discovery of the present fossils indicates that the climate of Baojishan Basin in Late Triassic is warmer and more humid than that of today.

A warming climate may increase flood hazard through boosting the global hydrological cycle. However, human impact through modifications to the river and its catchment is not well quantified. Here, we show a 12,000-year-long record of Yellow River flood events by synthesizing sedimentary and documentary data of levee overtops and breaches. Our result reveals that flood events in the Yellow River basin became almost an order of magnitude more frequent during the last millennium than the middle Holocene and 81 ± 6% of the increased flood frequency can be ascribed to anthropogenic disturbances. Our findings not only shed light on the long-term dynamics of flood hazards in this world’s most sediment-laden river but also inform policy of sustainable management of large rivers under anthropogenic stress elsewhere.

A comparative study was conducted for the first time on the form and valence of arsenic in the dry and fresh Cordyceps breeding products to clarify the specific morphology and valence of arsenic in Cordyceps breeding products and its safety. Arsenic betai-ne(AsB), arsenite(AsⅢ), dimethyl arsenic(DMA), arsenocholine(AsC), monomethyl arsenic(MMA), and arsenate(AsⅤ) in the dry and fresh samples were investigated using a bionic extraction method combined with HPLC-ICP-MS. The HPLC separation was performed on a DioncxIonPac~(TM) AS7 anion exchange column with a mobile phase of 100 mmol·L~(-1) ammonium carbonate-water for gradient elution at room temperature and the flow rate of 0.8 mL·min~(-1). HPLC was coupled with ICP-MS for the determination. The arsenic content was characterized in combination with chemometrics. The health safety risk of inorganic arsenic in the samples was assessed using the margin of exposure(MOE). The results of methodological validation showed that the six arsenic compounds showed good linearity(R~2gt;0.999) from 10 to 500 ng·mL~(-1), with precision RSDs of 1.8%-3.0%, recoveries(n=6) of 84.15%-98.28%, reproducibility RSDs of 6.4%-7.7%, and sample stability RSDs of 8.3%-14% within 10 h. Trace AsⅢ and AsⅤ were detected in 30 batches of dry and fresh Cordyceps breeding products, while arsenic compounds in other forms and valence were not detected. In the dry products, AsⅢ was 0.019-0.040 mg·kg~(-1) and AsV was 0.024-0.061 mg·kg~(-1), while in the fresh products, AsⅢ was 0.002 3-0.006 1 mg·kg~(-1) and AsⅤ was 0.008-0.016 mg·kg~(-1). The risk assessment results showed that the MOE of inorganic arsenic was much higher than 1 in both dry and fresh products, and the potential health safety risk of inorganic arsenic was low. The HPLC-ICP-MS method established in this study was efficient, rapid, accurate, and stable for the determination of six arsenic compounds in Cordyceps breeding products. The results of this study provide a basis for the safety and quality control of Cordyceps breeding products.

Influenza virus (IV) infections usually cause acute lung injury characterized by exaggerated proinflammatory responses. The paucity of therapeutic strategies that target host immune response to attenuate lung injury poses a substantial challenge in management of IV infections. In this study, we chemically synthesized a novel fatty acid (2Z,4E)-deca-2,4-dienoic acid (DDEA) identified from Chinese Cordyceps by using UHPLC-Q-TOF-MS techniques. The DDEA did not inhibit H1N1 virus replication but attenuated proinflammatory responses by reducing mRNA and protein levels of TNF-α, IFN-α, IFN-β, IL-6, CXCL-8/IL-8, CCL-2/MCP-1, CXCL-10/IP-10, CCL-3/MIP-1α, and CCL-4/MIP-1β in A549 cells and U937-derived macrophages. The anti-inflammatory effect occurred through downregulations of TLR-3-, RIG-I-, and type I IFN-activated innate immune signaling pathways. Altogether, our results indicate that DDEA may potentially be used as an anti-inflammatory therapy for the treatment of IV infections.

The Yellow River floodplain represents a fertile landmass that contributes significantly to human welfare and thus has been colloquially known as the birthplace of Chinese civilization. The sediment-laden nature of the Yellow River gave rise to a super-elevated channel belt, which is prone to failure particularly in the summer months when excessive precipitation occurs, resulting in cataclysmic floods traditionally regarded as “China’s Sorrow.” Therefore, a deeper understanding of levee breach frequency in this area is especially important for the assessment of socio-economic risk of levee breaches associated with future climate changes. To better understand the nature, evolution, and driving mechanisms of levee breaches on the lower Yellow River, it is necessary to place the instrumental data within a longer time framework. Here, we retrieve past information about levee breaches on the lower Yellow River since AD 11 from various documentary sources such as official histories of China. We evaluated each line of descriptions and narratives about the location, timing, and nature of each event in these documents, ending up with a detailed timeline of levee breaches on the lower Yellow River during the last 2000 years on an annual time scale. Our results reveal remarkable variations in the frequency of levee breaches superimposed on a long-term increasing trend. In addition to climate changes, the iterative embankment-siltation-breaching process caused a feedback: more breaches result in much more channel siltation, which in turn leads to even more breaches. The enhanced farming in the Loess Plateau played a pivotal role in the formation and operation of this positive feedback. Our findings may not only help improve the assessment of socio-economic risk of levee breaches associated with future climate changes, but also provide consulting information for hydraulic engineering and infrastructural designs in the lower Yellow river area.

As the largest inland basin in the northeastern Tibetan Plateau, Qaidam Basin has traditionally been thought of as the key region to study the Cenozoic climatic changes in the plateau; however, the information from a palaeobotanic respect is still lacking because of the paucity of the fossil plants. Fossil fruits and foliage ofAilanthus(Simaroubaceae) are reported from the Oligocene Shangganchaigou Formation in the northwestern Qaidam Basin, Qinghai Province, China. All the characters suggest that the fossil materials should be assigned toA.confucii, which is one widely distributed fossil species in the Cenozoic, regarded as the analogue of extantA. altissima. Current materials represent the first fossil record of Simaroubaceae in the northern Tibetan Plateau. Based on the climate requirements of the livingA. altissima, the ranges of mean annual temperature (MAT) and mean annual precipitation (MAP) reflected by current fossils from Qaidamare 6.9-17.0 degrees C, and 376-1,383 mm, respectively, compared with the current climate at the fossil site with MAT of -1.03 degrees C and MAP of 60.5 mm. The results indicate that the climate conditions of the fossil site during the Early Oligocene were much warmer and more humid than that of the current. We hereby suggest the warm and moist air could approach the northwestern Qaidam and to accommodate the ecological environment in Early Oligocene.

Characterization of aqueous extract in traditional Chinese medicine (TCM) is challenging due to the poor retention of the analytes on conventional C18 columns. This study presents a systematic characterization method based on a rapid chromatographic separation (8 min) on a polar-modified C18 (Waters Cortecs T3) column of aqueous extract of Cordyceps sinensis. UHPLC-HRMS method was used to profile components in both untargeted and targeted manners by full MS/PIL/dd-MS2 acquisition approach. The components were identified or tentatively identified by reference standards comparison, fragmentation rules elucidation and available databases search. A total of 91 components, including 10 nucleobases, 20 nucleosides, 39 dipeptides, 18 amino acids and derivatives and 4 other components, were characterized from the aqueous extract of C. sinensis. And this was the first time to systematically report the presence of nucleosides and dipeptides in C. sinensis, especially for modified nucleosides. The chemical basis inquiry of this work would be beneficial to mechanism exploration and quality control of C. sinensis and related products. Meanwhile, this work also provided an effective solution for characterization of aqueous extract in TCM.

In this study, a titanium ion (Ti4+) functionalized polydopamine coated ferroferric oxide (Fe3O4@PDA@Ti4+) core-shell magnetic particle was prepared for the selective extraction of nucleotides. Firstly, different metal ions including Ti4+, Zr4+, Fe3+, Al3+, Cu2+, Zn2+, Ni2+ and Mg2+ were respectively immobilized onto Fe3O4@PDA particles and their extraction efficiency for five nucleotides [cytidine-5′-monophosphate (CMP), uridine-5′-monophosphate (UMP), guanosine-5′-monophosphate (GMP), thymidine-5′-monophosphate (TMP) and adenosine-5′-monophosphate (AMP)] were compared. Among these prepared materials, Fe3O4@PDA@Ti4+, which exhibited the highest extraction efficiency for nucleotides, was further characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, transmission electron microscopy and energy dispersive X-ray spectroscopy. After being optimized of the extraction parameters including adsorbent amounts, extraction time, extraction temperature, type and concentration of the eluent, the prepared Fe3O4@PDA@Ti4+ magnetic particles were successfully applied for the selective extraction and determination of CMP, UMP, GMP, TMP and AMP in Cordyceps and Lentinus edodes. Good linearity (varying from 0.063 to 19.000 μg/mL, R2 > 0.999) and low limit of detection (LODs) (ranging between 0.0047 and 0.0141 μg/mL) for target analytes were achieved. These results demonstrated that the synthesized material in this study had potential for selective extraction of phosphorylated small molecular compounds in complicated matrix.

A new method for reconstructing paleoenvironments using the pavement cell characteristics of Middle Jurassic ginkgoalean fossils from the Yaojie and Baojishan basins, Gansu Province, China, is presented. Specifically, we use present-day relationships between the length-to-width (L/W) ratio of leaf pavement cells of Ginkgo biloba and mean annual temperature (MAT), mean annual precipitation (MAP), and relative humidity (RH). Primary analyses indicate that the L/W ratio of Ginkgo biloba pavement cells is negatively correlated with MAT, MAP, and RH, and linear regression equations can be established. Based on these equations, paleoclimatic parameters were inferred using the ginkgoalean fossil data. The estimated paleotemperatures in the Yaojie and Baojishan basins during the Middle Jurassic are approximately 6 °C higher than those of the present day, which is consistent with the results of the currently used stomata ratio method and the global CO2 prediction model. We deduce that paleoprecipitation in the basins was approximately 460 mm higher and paleohumidity approximately 20% higher than current conditions and thus the two basins exhibited humid subtropical conditions during the Middle Jurassic. Fossil flora and spore-pollen assemblage data from the study areas corroborate the new observations.

To accelerate the breeding process of cultivated Ophiocordyceps sinensis and increase its yield, it is important to identify molecular fingerprint of dominant O. sinensis. In the present study, we collected 3 batches of industrially cultivated O. sinensis product with higher yield than the others and compared their internal transcribed spacer (ITS) sequences with the wild and the reported. The ITS sequence was obtained by bidirectional sequencing and analyzed with molecular systematics as a DNA barcode for rapid and accurate identification of wild and cultivated O. sinensis collected. The ITS sequences of O. sinensis with detailed collection loci on NCBI were downloaded to construct a phylogenetic tree together with the sequences obtained from the present study by using neighbor-joining method based on their evolution relationship. The information on collection loci was analyzed with ArcGIS 10.2 to demonstrate the geographic distribution of these samples and thus to determine the origin of the dominant samples. The results showed that all wild and cultivated samples were identified as O. sinensis and all sequences were divided into seven phylogenetic groups in the tree. Those groups were precisely distributed on the map and the process of their system evolution was clearly presented. The three cultivated samples were clustered into two dominant groups, showing the correlation between the industrially cultivated samples and the dominant wild samples, which can provide references for its optimized breeding in the future.

In this study, an effective and portable method for enzyme activity detection and inhibitory activity evaluation was developed based on the alkaline phosphatase (ALP)-mediated reaction in a personal glucose meter (PGM). In this method, ALP catalyzes the hydrolysis of substrate amifostine (WR-2721) to produce ethanethiol (WR-1065), which can trigger the reduction of ferricyanide (K3[Fe(CN)6]), an electron transfer mediator in glucose test strips, to ferrocyanide ([K4Fe(CN)6]) and generate a PGM-detectable signal. Thus, WR-1065 can be directly quantified by a PGM as simply as detecting glucose in blood. After being systematically optimized, the method was applied to evaluate the inhibitory activity of ten small-molecule compounds and six Cordyceps sinensis (CS) extracts on ALP. The results showed that adenosine-5-monophosphate and theophylline had high inhibitory activity, but two CS extracts have promotion potency on ALP with the values of -20.7 ± 1.3% and -46.6 ± 2.1%, respectively. Moreover, the binding sites and modes of small-molecule compounds to ALP were investigated by molecular docking, while a new substrate competitor with theoretically good inhibitory activity against ALP was designed by scaffold hopping. Finally, the accuracy of the PGM method for enzyme activity detection was assessed by detecting ALP from milk samples, and the recovery ranged from 87.7% to 116.9%. These results indicate that it is feasible to evaluate enzyme activity and the inhibitory activity of small-molecule compounds and CS extracts on ALP using a PGM based on ALP-mediated reaction. Deep profiling of glycosphingolipids and sphingomyelins in wild Cordyceps was carried out by using offline chromatographic enrichment followed by ultrahigh performance liquid chromatography-ultrahigh definition-quadrupole time-of-flight mass spectrometry (UHPLC-UHD-Q-TOF-MS). A total of 119 glycosphingolipids (72 new ones) and 87 sphingomyelins (43 new ones) were identified from wild Cordyceps on the basis of the accurate mass and MS/MS fragmentations, isotope patterns, sphingolipid (SPL) database matching, confirmation by SPL standards, and the reversed-phase liquid chromatographic retention rule. This study is the most comprehensive report on the identification of glycosphingolipids and sphingomyelins from fungus. A subsequent lipopolysaccharide-induced mouse splenic lymphocyte proliferation assay showed that the Cordyceps glycosphingolipid fraction exhibits higher immunosuppressive activity compared to that of Cordyceps sphingomyelins. Our findings provided insight into the chemical diversity of sphingolipids in Cordyceps and chemical evidence for the therapeutic application of wild Cordyceps.

This study aimed to investigate the anti-inflammatory activity of Musca domestica cecropin-A2 (Mdc-A2) toward Staphylococcus aureus (S. aureus) to learn more about their immunological functions. RAW264.7 cells were transfected with recombinant lentiviruses introduce pLEX-Mdc-A2into the RAW264.7 cell line (RAW-Mdc-A2). The RAW264.7 cell line with empty pLEX (RAW-pLEX) was produced in the same manner as a negative control. Real-time quantitative reverse transcription PCR (RT-PCR) was performed to analyze the mRNA expression of TNF-a, IL-1β, NFκB-1 and NFκB-2 in S. aureus-stimulated RAW-Mdc-A2 cells and RAW-pLEX cells in untreated cells and cells treated for 3 h, 6 h, 12 h and 24 h. RT-PCR was performed to analyze the mRNA expression of TNF-a, NFκB-1 and NFκB-2 stimulated by Lipoteichoic acid (LTA). Production of TNF-a was detected by enzyme-linked immunosorbent assay (ELISA). Colony counts were used to calculate the number of CFU per mL of cell culture supernatants. The results showed that compared to RAW-pLEX cells, stable transfection of Mdc-A2 in RAW264.7 cells stimulated by S. aureus significantly down-regulated the mRNA expression of TNF-a transcript variant 1 (TNF-a-tv-1) at 6 h and 12 h and the mRNA expression of TNF-a transcript variant 2 (TNF-a-tv-2) at 3 h, 6 h and 12 h. Compared to RAW-pLEX cells, stable transfection of Mdc-A2 in RAW264.7 cells stimulated by S. aureus significantly down-regulated the mRNA expression of IL-1β-T at 3 h, 6 h and 12 h as well as the mRNA expression of IL-1β at 3 h and 6 h. The expression and production of TNF-a and bacterial burden of cell culture supernatants were significantly down-regulated in RAW-Mdc-A2 cells stimulated by S. aureus, and the expression and production of TNF-a were significantly down-regulated in RAW-Mdc-A2 cells stimulated by LTA. Compared to RAW-pLEX cells, stable transfection of Mdc-A2 in RAW264.7 cells stimulated by S. aureus significantly down-regulated the mRNA expression of NFκB-1 at 3 h, 6 h and 12 h as well as the mRNA expression of NFκB-2 at 6 h. Additionally, stable transfection of Mdc-A2 in RAW264.7 cells stimulated by LTA significantly down-regulated the mRNA expression of NFκB-1. In conclusion, Mdc-A2 possesses potent anti-inflammatory activity and potent antimicrobial activity. Additionally, Mdc-A2 may interact with LTA and execute strong anti-inflammatory activity by blocking the activation of NF-κB signaling pathways.

Background: As one of the bioactive components in Cordyceps sinensis (CS), proteins were rarely used as index components to study the correlation between the protein components and producing areas of natural CS. Objective: Protein components of 26 natural CS samples produced in Qinghai, Tibet, and Sichuan provinces were analyzed and compared to investigate the relationship among 26 different producing areas. Materials and Methods: Proteins from 26 different producing areas were extracted by Tris-HCl buffer with Triton X-100, and separated using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional electrophoresis (2-DE). Results: The SDS-PAGE results indicated that the number of protein bands and optical density curves of proteins in 26 CS samples was a bit different. However, the 2-DE results showed that the numbers and abundance of protein spots in protein profiles of 26 samples were obviously different and showed certain association with producing areas. Conclusions: Based on the expression values of matched protein spots, 26 batches of CS samples can be divided into two main categories (Tibet and Qinghai) by hierarchical cluster analysis. Cordyceps is one of the most valuable traditional Chinese medicines. There are various counterfeits in markets because of high price and limited output. In this study,116 Cordyceps,146 hosts and 29 related products were collected and detected by using normal DNA barcoding technology and specific PCR method. The results indicated that Cordyceps and its adulterants could be distinguished from each other through DNA barcoding technology based on ITS and COⅠsequences. Two pairs specific primers ITSSF1/ITSSR1 and ITSSF2/ITSSR2 were developed to amplify 297 bp and 136 bp ITS regions of Cordyceps sinensis,respectively. It could be used to identify C. sinensis specifically and rapidly. Furthermore,specific primers ITSSF1/ITSSR1 and ITSSF2/ITSSR2 combined with ITS and COⅠsequences could differentiate powder Cordyceps from fermentation mycelia and could identify related products. Therefore,the method developed from this study could be applied to identify the powder of Cordyceps from fermentation mycelia and related products efficiently.

In this study,the protein in different Cordyceps samples,which include fresh sample( S1),22 ℃ drying sample( S2),37 ℃ drying sample( S3) and 60 ℃ drying sample( S4),were analyzed by sodium dodecylsupinate-polyacrylamide gel electrophoresis( SDS-PAGE) and two-dimensional electrophoresis( 2-DE). The total protein contents in Cordyceps samples were from 1. 655-4. 493 mg·g~(-1) and the protein contents in fresh sample was the highest. The results of SDS-PAGE showed that the mainly ranges of protein molecular weight of Cordyces samples were 10-100 kDa and the numbers of protein bands were 28 to 41,the fresh sample had the maximum number of protein bands. The 2-DE profiles were analyzed by PDQuest software. The resulted indicated that 488-876 protein spots were detected in different Cordyceps samples and the isoelectric point( pI) was distributed between 4. 5 and 6. 5,the protein molecular weight was distributed in 10-20 kDa and 25-100 kDa,the fresh sample had the maximum number of protein spots. Therefore,the drying process could decrease contents and species of protein in Cordyceps,and the different drying conditions had different effects on protein. These results provide a reference for improving the drying process of Cordyceps.

In this study, determination of adenosine in Cordyceps was accomplished by an online pressurized liquid extraction HPLC, and a poroshell column was employed for rapid HPLC separation. The Cordyceps sample powder was filled into a hollow guard column and extracted by thermal and pressurized water (75°C, 150 bar) for 3 min. Chromatographic separation was conducted on a poroshell 120 SB-Aq (4.6 × 50 mm, 2.7 μm) column and gradient elution was performed by water and methanol at a flow rate of 1.5 mL/min. The detection wavelength was set at 260 nm. The total measurement time including sample extraction and separation was 0.9993) within tested range (4.03-80.5 μg/mL). The intra- and inter-day precisions of adenosine were 0.24% and 0.94%, respectively, and the recoveries of adenosine was 96.1% (relative standard deviation, 4.49%). The validated method was then applied to determinate adenosine in six batches of Cordyceps, and the contents of adenosine were 0.28-0.48 mg/g. The developed method is beneficial for the quality control of Cordyceps.

Background Water is generally considered to be a safe and green solvent suitable for use in natural product extraction. In this study, an eco-friendly subcritical water method was used to extract pectin from waste jackfruit peel (JFP-S), which was compared with pectin obtained by the traditional citric acid method (JFP-C). Results The extraction process was optimized using response surface methodology (RSM), and the optimum process parameters were as follows: extraction temperature 138 °C, extraction time 9.15 min, liquid / solid (L/S) ratio 17.03 mL g-1 . Under these conditions, the pectin yield was 149.6 g kg-1 (dry basis). Pectin obtained from the two extraction methods displayed a high degree of esterification and the monosaccharide composition was consistent. The galacturonic acid content of JFP-S and JFP-C was 52.27% and 56.99%, respectively. JFP-S had more hairy regions and side chains than JFP-C. The molecular weight of JFP-S was 113.3 kDa, which was significantly lower than that of JFP-C (174.3 kDa). Fourier-transform infrared spectroscopy (FTIR) indicated that two samples had similar pectin typical absorption peaks. According to differential scanning calorimetry (DSC), both JFP-S and JFP-C had relatively good thermal stability. JFP-S demonstrated lower apparent viscosity and elasticity than JFP-C. Meanwhile, the G' and G'' moduli of JFP-S were lower, which found expression in the gel textural characterization of the samples. Conclusion This work showed that the subcritical water method is an efficient, time-saving, and eco-friendly technology for the extraction of pectin from jackfruit peel compared with the traditional citric acid method. The physicochemical properties of pectin could be changed during subcritical water extraction. © 2019 Society of Chemical Industry.

Objectives To evaluate the diagnostic utility of DWI in the assessment of node metastases and investigate whether the ADC value could be used to discriminate between metastatic and non-metastatic lymph nodes in breast cancer patients. Materials and methods 13 studies with a total of 676 metastatic and 811 non-metastatic lymph nodes were included. Results (1) The pooled sensitivity, specificity, PPV and NPV of DWI were 0.83, 0.82, 0.83 and 0.85, respectively. The PLR and NLR were 4.95 and 0.23, respectively. The AUC and Q* index were 0.91 and 0.85, respectively. (2) The ADC value of metastatic lymph nodes was lower than non-metastatic lymph nodes ( WMD =−0.213, 95% CI −0.349 to −0.076, Z=3.05, P meta -analysis of the group of b(0800): The pooled sensitivity, specificity, PPV and NPV of DWI were 0.86, 0.86, 0.82 and 0.90, respectively. The PLR and NLR were 6.76 and 0.18, respectively. The AUC and Q* index were 0.93 and 0.87. The ADC value of metastatic lymph nodes was lower than non-metastatic lymph nodes( WMD =−0.267, 95% CI −0.348 to −0.185, Z=6.40, P Conclusions DWI and ADC value appear to be a reliable method to differentiate metastatic and non-metastatic lymph nodes. The combination of b=0 and 800s/mm 2 resulted in higher diagnostic accuracy and more pronounced ADC value difference. If only a couple of b values are used, those of b=0 and 800s/mm 2 are recommended.

Ganoderma lucidum (Chizhi in Chinese) is one of the most valuable and widely used medicinal fungi in traditional Chinese medicines (TCMs). Most of previous studies were focused on the triterpenoids and polysaccharides of G. lucidum, whereas less attention had been paid on the protein, which is another bioactive compound in it. In the present study, protein maps of fourteen G. lucidum samples were comprehensively analyzed by sodium dodecyl sulfate - polyacrylamide gel electrophoresis (SDS-PAGE) and two-dimensional electrophoresis (2-DE). The results indicated that there were significant differences in protein profiles of G. lucidum samples from different origins. Furthermore, previous reported bioactive proteins from the fruiting bodies of G. lucidum, were mainly distributed in 4 taxa (A, B, C and D) based on their molecular weights on the 2-DE maps. The proteins should be considered as marker for the quality control of G. lucidum, because the proteomic variation may affect on their pharmacological activities.

In this study, determination of adenosine in Cordyceps was accomplished by an online pressurized liquid extraction HPLC, and a poroshell column was employed for rapid HPLC separation. The Cordyceps sample powder was filled into a hollow guard column and extracted by thermal and pressurized water (75°C, 150 bar) for 3 min. Chromatographic separation was conducted on a poroshell 120 SB-Aq (4.6 × 50 mm, 2.7 μm) column and gradient elution was performed by water and methanol at a flow rate of 1.5 mL/min. The detection wavelength was set at 260 nm. The total measurement time including sample extraction and separation was6 min. The calibration curve of adenosine was Y = 4.502 × 106X + 1.731 × 104, which showed good linearity (r20.9993) within tested range (4.03-80.5 μg/mL). The intra- and inter-day precisions of adenosine were 0.24% and 0.94%, respectively, and the recoveries of adenosine was 96.1% (relative standard deviation, 4.49%). The validated method was then applied to determinate adenosine in six batches of Cordyceps, and the contents of adenosine were 0.28-0.48 mg/g. The developed method is beneficial for the quality control of Cordyceps.

The present study was designed to investigate the effect of cultivated Cordyceps sinensis (CCS) on leukemia-derived K562 cells, and further explore the underlying mechanisms. After routine culture of K562 cells, MTT assay was used to detect the effect of CCS on survivel of human leukemia cell lines K562;DAPI staining was used to observe the morphological changes of the nucleus and AO/EB staining was used to observe cell apoptosis. JC-1 staining was employed to detect the changes in mitochondrial membrane potential. Flow cytometry (FCM) was used to detect cell cycle distribution, and Western blot analysis was used to detect the expression levels of Bax, Bcl-2, caspase 3, caspase 8, cyclin D1, CDK2, and CDK4 in K562 cells. The results showed that CCS (0.345-5.524 g·L⁻¹) substantially suppressed proliferation of K562 cells and induced G₁/S phase arrest in a dose-dependent manner. DAPI and AO/EB staining indicated that cell apoptosis was significantly induced by CCS treatment, accompanied by decreased mitochondrial membrane potential demonstrated by JC-1 staining. Western blot results showed that CCS significantly increased the expression of Bax and, meanwhile, decreased the expression levels of Bcl-2, cyclin D1, CDK2, CDK4, caspase 3 and caspase 8. Collectively, our data demonstrated that CCS dose-dependently suppressed cell proliferation and induced cell apoptosis in K562 cells, and the mechanism might be associated with inducing cell cycle arrest, regulating Bcl-2/Bax ratio and activating the mitochondrial apoptosis pathway.

This study is to establish the HPLC specific chromatogram and determine four main nucleosides of wild and cultivated Cordyceps sinensis. Uridine, inosine, guanosine and adenosine were selected as reference substance. HPLC analysis was performed on a Waters XSelect HSS T3 C₁₈ (4.6 mm×250 mm, 5 μm), with a mobile phase consisting of water(A)-acetonitrile (B) at a flow rate of 0.6 mL•min⁻¹ (0-5 min,0% B;5-15 min,0%-10% B, 15-30 min,10%-20% B, 30-33 min, 20%-50% B, 33-35 min, 50%-0% B, 35-40 min, 0% B). The detection wavelength was 260 nm and the column temperature was controlled at 30 ℃, and the injection volume was 5 μL. HPLC specific chromatogram of wild and cultivated C. sinensis was established and four main nucleosides were simultaneously determined by the above method. Specific chromatograms and contents of four main nucleosides showed no significant differences between cultivated and wild C. sinensis. These results can provide scientific evidences for further development and utilization of cultivated C. sinensis.
