Full-thickness oral mucosal defects are accompanied by significant blood loss and frequent infections. Instead of conventional therapies that separate hemostasis and anti-inflammation in steps, emerging hydrogels can integrate multiple functions for the successive process after defect including hemostasis/inflammatory phase, proliferative phase, and remodeling phase. However, these functions can be easily compromised by rapid swelling and degradation of hydrogels in wet oral environment. Herein, a low-swelling adhesive hydrogel with rapid hemostasis and potent anti-inflammatory capability was developed using a dual cross-linking strategy as well as a safe and facile fabrication method. It was double cross-linked hydrogel consisting of gelatin methacrylate (GelMA), nanoclay, and tannic acid (TA) (referred to as GNT). GNT hydrogel exhibited low-swelling (one-eighth of that of GelMA), excellent stretchability (211.86%), and good adhesive properties (5 times the adhesive strength of GelMA). Physicochemical characterization illuminated the close interactions among the three components. A systematic investigation of the therapeutic effects of GNT hydrogels was performed. In vitro and in vivo experimental results demonstrated the potent hemostatic property and excellent antibacterial and anti-inflammatory effects of GNT hydrogels. The RNA sequencing analysis results for rat full-thickness oral mucosal samples showed that GNT reduced inflammation levels by down-regulating the expression of multiple inflammation-related pathways, including TNF and IL-17 pathways. It also enhanced the expression levels of tissue regeneration-related genes and thus accelerated defective mucosal repair. More importantly, the therapeutic effects of GNT were superior to those of a commercial oral tissue repair membrane when applied for full-thickness oral mucosal defect repair in rabbits. In summary, the prepared low-swelling adhesive GNT hydrogel with rapid hemostasis and potent anti-inflammatory is a promising therapy for full-thickness mucosal defect in the moist and dynamic oral environment.

Nowadays, bacterial infection has become a severe threat to human health worldwide. The arising of antibiotic-resistant bacteria resulting from antibiotic abusing further adds the burden of bacterial infection treatment. It is the first priority to find more antibiotic-free solutions for combating bacteria, especially multi-drug-resistant bacteria. Compared to other antibiotic-free solutions, reactive oxidative species (ROS)-mediated strategies have outstanding advantages. Highly active ROS can directly cause oxidative damage and are less possible to bring about bacterial resistance. Photodynamic therapy (PDT), sonodynamic therapy (SDT), and enzyme-like catalytic therapy are three main methods to massively manufacture ROS. With the rapid development of nanotechnology, nanoparticles have been widely used in PDT and SDT. Nanoparticles with enzyme-like activities are also extensively studied. In this review, we introduced the application of nanomaterials in three ROS-mediated methods for combating bacteria and biofilms, which might provide important insights for antibiotic-free antibacterial strategies.

Objective: DNA copy number alterations and difference expression are frequently observed in ovarian cancer. The purpose of this way was to pinpoint gene expression change that was associated with alterations in DNA copy number and could therefore enlighten some potential oncogenes and stability genes with functional roles in cancers, and investigated the bioinformatics significance for those correlated genes. Method: We obtained the DNA copy number and mRNA expression data from the Cancer Genomic Atlas and identified the most statistically significant copy number alteration regions using the GISTIC. Then identified the significance genes between the tumor samples within the copy number alteration regions and analyzed the correlation using a binary matrix. The selected genes were subjected to bioinformatics analysis using GSEA tool. Results: GISTIC analysis results showed there were 45 significance copy number amplification regions in the ovarian cancer, SAM and Fisher’s exact test found there have 40 genes can affect the expression level, which located in the amplification regions. That means we obtained 40 genes which have a correlation between copy number amplification and drastic up- and down-expression, which p-value ere overlapped with the several published studies which were focused on the gene study of tumorigenesis. Conclusion: The use of statistics and bioinformatics to analyze the microarray data can found an interaction network involved. The combination of the copy number data and expression has provided a short list of candidate genes that are consistent with tumor driving roles. These would offer new ideas for early diagnosis and treat target of ovarian cancer.

The application of three-dimensional printed polymer scaffolds in repairing bone defects is a promising strategy. Among them, polycaprolactone (PCL) scaffolds are widely studied due to their good processability and controlled degradation rate. However, as an alternative graft for repairing bone defects, PCL materials have poor hydrophilicity, which is not conducive to cell adhesion and growth. In addition, the poor mechanical properties of PCL materials cannot meet the strength required to repair bone defects. In this paper, nano-zirconium dioxide (ZrO2) powder is embedded in PCL material through a melt-mixing process, and a regular grid scaffold is constructed by 3D printing. The embedding of nanometer zirconium dioxide powder improves the hydrophilicity and water absorption of the composite scaffold, which is conducive to cell adhesion, proliferation and growth and is beneficial to the exchange of nutrients. Therefore, the PCL/ZrO2 composite scaffold showed better biological activity in vitro. At the same time, the PCL/ZrO2 composite material system significantly improves the mechanical properties of the scaffold. Among them, compared with the pure PCL scaffold, the Young’s modulus is increased by about 0.4 times, and the compressive strength is increased by about 0.5 times. In addition, the osteogenic differentiation results also showed that the PCL/ZrO2 composite scaffold group showed better ALP activity and more effective bone mineralization than the pure PCL group. We believe that the 3D printed PCL/ZrO2 composite scaffold has certain application prospects in repairing bone defects.

PURPOSE To assess the influence of ferrule thickness on the fracture resistance and failure mode of endodontically treated bovine incisors and to predict the long-term prognosis, as well as choose the most suitable clinical treatment, for teeth with different ferrule thicknesses. MATERIALS AND METHODS A total of 50 endodontically treated bovine incisors were restored with quartz fiber posts and metal crowns and separated into five groups (n = 10 each): no ferrule (group A); 0.5-mm-thick ferrule (group B); 1.0-mm-thick ferrule (group C); 1.5-mm-thick ferrule (group D); and 2.0-mm-thick ferrule (group E). All specimens were subjected to a fatigue loading test (2.33 Hz, 50 N, 300,000 cycles). Survived specimens were loaded until fracture on a universal testing machine at an angle of 135 degrees and a crosshead speed of 0.5 mm/minute. Failure modes and fracture resistance were recorded. Data were analyzed using one-way ANOVA and least significant difference tests. RESULTS A significant increase (P < .05) was detected in fracture resistance with increase in ferrule thickness. Group D (1.5 mm) and group E (2.0 mm) showed significantly higher fracture resistance than the other three groups. All failures belonged to restorable fracture patterns. CONCLUSION Ferrule thickness contributed significantly to the fracture resistance of endodontically treated bovine incisors restored with quartz fiber posts and metal crowns. Teeth with ferrule thickness of ≥ 1.5 mm can achieve higher fracture resistance and have a better long-term prognosis.

The rapid spread of drug-resistant pathogens threatens human health. To address the current antibacterial dilemma, the development of antibiotic-free strategies using nanotechnology is imperative. In this study, silver nanoparticles (Ag-PC NPs) with pH-sensitive charge reversal and self-aggregation capacities are successfully synthesized. In the acidic microenvironment of bacterial biofilms, protonation of the surface peptide enhances the affinity of Ag-PC NPs for bacteria, which can make Ag-PC NPs prone to target and penetrate into biofilms, and the self-aggregated capacity helps Ag-PC NPs remain in biofilms for a long time to disrupt bacterial biofilm formation. In addition, biocompatible Ag-PC NPs are utilized in three types of bacteria-infected animal models. They exhibit an excellent performance in killing bacteria, inhibiting plaque biofilms, and ameliorating inflammatory responses. In conclusion, this study offers new insights into antibiotic-free antibacterial strategies, and exhibits promising application prospects.

An effective algicidal bacterium, Enterobacter hormaechei F2, was isolated and identified using 16S rDNA sequencing. The increased production of algal catalases and peroxidases as well as the production of reactive oxygen species demonstrated the effective and distinctive algicidal activities of E. hormaechei F2. The comparative effects of different components of bacterial solutions suggested that indirect algicidal disruption was the major mechanism of E. hormaechei F2 performance. Dialysis of the extracellular material revealed that the molecular weight of the algicides was less than 3.5 kDa. These were inferred to be proteins, amino acids, and other small molecules. Gene enrichment analysis shows the pathways such as amino acid metabolism and Type n secretion systems involved in the algicidal process, which can predict that prodigiosin and signaling systems may play a role in the algicidal activity.

With the widespread use of volatile anesthetic agents in the prolonged sedation for COVID-19 pneumonia and ARDS, there is an urgent need to investigate the effects and treatments of lengthy low-concentration inhaled anesthetics exposure on cognitive function in adults. Previous studies showed that general anesthetics dose- and exposure length-dependently induced neuroinflammatory response and cognitive decline in neonatal and aging animals. The anti-diabetes drug metformin has anti-neuroinflammation effects by modulating microglial polarization and inhibiting astrocyte activation. In this study, we demonstrated that the inhalation of 1.3% isoflurane (a sub-minimal alveolar concentration, sub-MAC) for 6 h impaired recognition of novel objects from Day 1 to Day3 in adult mice. Prolonged sub-MAC isoflurane exposure also triggered typically reactive microglia and A1-like astrocytes in the hippocampus of adult mice on Day 3 after anesthesia. In addition, prolonged isoflurane inhalation switched microglia into a proinflammatory M1 phenotype characterized by elevated CD68 and iNOS as well as decreased arginase-1 and IL-10. Metformin pretreatment before anesthesia enhanced cognitive performance in the novel object test. The positive cellular modifications promoted by metformin pretreatment included the inhibition of reactive microglia and A1-like astrocytes and the polarization of microglia into M2 phenotype in the hippocampus of adult mice. In conclusion, prolonged sub-MAC isoflurane exposure triggered significant hippocampal neuroinflammation and cognitive decline in adult mice which can be alleviated by metformin pretreatment via inhibiting reactive microglia and A1-like astrocytes and promoting microglia polarization toward anti-inflammatory phenotype in the hippocampus.

To study the biosorption behaviors of bacteria on heavy metal chelators, the biosorption kinetics, biosorption thermodynamics and pH influence tests of the Ochrobactrum MT180101 on ionic and chelate copper were investigated. Furthermore, the biosorption mechanisms of the Ochrobactrum MT180101 on ionic copper and chelate copper were explained by means of an excitation emission matrix as well as infrared and X-ray photoelectron spectroscopy. The results indicated the following. 1) The biosorption on chelate copper was needed to destroy the complexation group first through metabolic and secretory activities. 2) The biosorption mechanism of the Ochrobactrum MT180101 on copper involved surface biosorption, extracellular chelation and bienzyme-mediated biotransformation. The results suggested that Ochrobactrum had a superior biosorption efficiency to ionic and chelate copper.

As major precursors of disinfection by-products (DBPs), soluble microbial products (SMPs) generated by sewage discharge can adversely affect drinking water quality. It is essential to understand the adsorption behaviours of SMPs onto sediments and the effect of DBPs formation. In this study, the adsorption ability of sediments was evaluated by adsorption isotherms with respect to temperature and salinity. Adsorption behaviours were investigated using X-ray photoelectron spectroscopy, electron microscopy analysis, and excitation emission matrix fluorescence analysis. Chlorination was also employed to explore the influence of sediment adsorption on drinking water quality. The results indicated that the maximum adsorption potential of sediments to SMPs was 1.60 mg/g, which involved exothermic processes. SMPs adsorption declined with increasing temperature and salinity, and fulvic acid and protein in SMPs were more readily adsorbed on sediments than was humic acid. Correlation analysis results indicated that adsorption behaviours of sediments to SMPs could significantly reduce the generation potential of DBPs (r = 0.882-0.938, p 0.01). In addition, the decrease of C-DBPs was considerably greater than that of N-DBPs. These research findings are of importance to assessments of the fate and transport of SMPs in water-sediment systems, as well as the effect of following DBPs formation in the drinking water supply.

The growing demand for charming smiles has led to the popularization of tooth bleaching procedures. Current tooth bleaching products with high-concentration hydrogen peroxide (HP, 30-40%) are effective but detrimental due to the increased risk of enamel destruction, tooth sensitivity, and gingival irritation. Herein, we reported a less-destructive and efficient tooth whitening strategy with a low-concentration HP, which was realized by the remarkably enhanced Fenton-like catalytic activity of oxygen-deficient TiO2 (TiO2-x). TiO2-x nanoparticles were synthesized with a modified solid-state chemical reduction approach with NaBH4. The Fenton-like activity of TiO2-x was optimized by manipulating oxygen vacancy (OV) concentration and further promoted by the near-infrared (NIR)-induced photothermal effect of TiO2-x. The TiO2-x sample named BT45 was chosen due to the highest methylene blue (MB) adsorption ability and Fenton-like activity among acquired samples. The photothermal property of BT45 under 808 nm NIR irradiation was verified and its enhancement on Fenton-like activity was also studied. The BT45/HP + NIR group performed significantly better in tooth whitening than the HP + NIR group on various discolored teeth (stained by Orange II, tea, or rhodamine B). Excitingly, the same tooth whitening performance as the Opalescence Boost, a tooth bleaching product containing 40% HP, was obtained by a self-produced bleaching gel based on this novel system containing 12% HP. Besides, negligible enamel destruction, safe temperature range, and good cytocompatibility of TiO2-x nanoparticles also demonstrated the safety of this tooth bleaching strategy. This work indicated that the photothermal-enhanced Fenton-like performance of the TiO2-x-based system is highly promising in tooth bleaching application and can also be extended to other biomedical applications.

Blockchain cross-chain technology is an important technical means to realize inter-chain interconnection and value transfer. This paper systematically summarizes the current blockchain mainstream cross-chain technology. Firstly, this paper summarizes the basic knowledge of cross-chain technology, and then introduces the mainstream cross-chain techniques. Finally, this paper analyzes the challenges of cross-chain technology and forecasts its future prospect.

A newly isolated algicidal strain, Enterobacter hormaechei F2, showed high algicidal activity against the typical HAB species Microcystis aeruginosa. Potential algicides were detected through liquid chromatograph–mass spectrometer analysis, revealing that prodigiosin is an algicide and PQS is a quorum sensing molecule. RNA-seq was used to understand the algicidal mechanisms and the related pathways. We concluded that the metabolism of prodigiosin and PQS are active at the transcriptional level. The findings indicate that E. hormaechei F2 can be used as a potential biological agent to control harmful algal blooms to prevent the deterioration of the ecological and economic value of water bodies.

Arabidopsis SMAX1/SMXL (SUPPRESSOR OF MAX2 1/SMAX1-LIKE) proteins function as transcriptional repressors in karrikin and strigolactone (SL) signaling pathways and regulate plant architecture. MAX2 is a common factor in the two signaling pathways and a component of the SCF complex that modulates the proteasome-mediated degradation of SMAX1/SMXLs. SMXL6, 7, and 8 proteins promote shoot branching and inhibit petiole elongation. Our study found that the accumulation of SMAX1 suppresses rosette shoot branching and increases cauline branches on the primary inflorescence stem, plant height, petiole length, and leaf length/width ratio. The SMAX1 accumulation enhances the expression of BRC1, HB53, HB40, and HB21 that modulate shoot branching. SMAX1 also regulates the expression of the genes involved in auxin transport, cytokinin signaling pathway, and SL biosynthesis. The expression analyses of these genes suggest that excessive SMAX1 should accelerate the transport of auxin and the biosynthesis of SL in plants. High SL concentration suppresses the bud development in smax1D mutant that accumulates SMAX1 protein in plant. However, the effects of cytokinin and auxin on shoot branching remain elusive in the mutant with excessive SMAX1. SMAX1 regulates leaf shape and petiole length via modulating TCP1 expression. Our findings reveal a novel function of SMAX1 and new mechanism of shoot branching.

The surface modification of titanium (Ti) can enhance the osseointegration and antibacterial properties of implants. In this study, we modified porous Ti discs with calcium phosphate (CaP) and different concentrations of Lactoferrin (LF) by biomimetic mineralization and examined their antibacterial effects and osteogenic bioactivity. Firstly, scanning electron microscopy (SEM), the fluorescent tracing method, X-ray photoelectron spectroscopy (XPS), Fourier transform infrared spectroscopy (FTIR), energy dispersive X-ray spectroscopy (EDX), X-ray diffraction (XRD), and the releasing kinetics of LF were utilized to characterize the modified Ti surface. Then, the antibacterial properties against S. sanguis and S. aureus were investigated. Finally, in vitro cytological examination was performed, including evaluations of cell adhesion, cell differentiation, extracellular matrix mineralization, and cytotoxicity. The results showed that the porous Ti discs were successfully modified with CaP and LF, and that the LF-M group (200 μg/mL LF in simulated body fluid) could mildly release LF under control. Further, the LF-M group could effectively inhibit the adhesion and proliferation of S. sanguis and S. aureus and enhance the osteogenic differentiation in vitro with a good biocompatibility. Consequently, LF-M-modified Ti may have potential applications in the field of dental implants to promote osseointegration and prevent the occurrence of peri-implantitis.

This paper analyzes the differences between online and offline courses in teaching organization, course content and practice, as well as the differences between hybrid teaching and hybrid courses in concept and emphasis. This paper puts forward the basic principles and requirements of hybrid courses teaching design, and discusses the key points and application scenarios of hybrid courses teaching design from three aspects: teaching objectives, teaching elements and teaching activities. And the key issues that need to be figured out are explained.

It is necessary to study the mechanism of resistance to heavy metals in microbiological processes. In this study, Ochrobactrum MT180101 was used as the microbial source of an membrane bioreactor to investigate its degradation efficiency for electroplating wastewater and the copper-resistant mechanism. Meanwhile, excitation emission matrix-parallel factor, scanning electron microscope, atomic force microscope, fourier transform infrared spectroscopy, X-ray photoelectron spectroscopy and proteome analyses were applied to explain the comprehensive mechanism of the Ochrobactrum MT180101 resisting heavy metal toxicity. The results indicated that the Ochrobactrum MT180101 resisted heavy metal toxicity with the following pathways: i) binding metal cations on cell wall surfaces, ii) generating microbial products such as protein to chelate and stabilize the metal cations, iii) bio-transporting heavy metals from the intramembrane to the outer membrane by means of intracellular transport, and iv) reducing heavy metals through enzyme-mediated biotransformation. The results ensure that Ochrobactrum MT180101 was a copper-resistant bacterium that can be used in the pretreatment or deep treatment of electroplating wastewater.

OBJECTIVES To assess the tear meniscus height (TMH), noninvasive tear breakup time (NITBUT), and bulbar redness in pediatric patients after orthokeratology (OK). METHODS Pediatric patients who underwent overnight OK between December 2014 and November 2017 were retrospectively investigated. According to the time of wearing, subjects were divided into three groups: 1-week, 1-month, and over 1-year. Age-matched subjects were recruited as a control group. Keratograph 5M was used to assess TMH, NITBUT-first, NITBUT-average, and bulbar redness score. RESULTS A total of 106 subjects aged from 7 to 15 years were included. Compared with control group, NITBUT-first was significantly lower in 1-week group (P=0.036) and NITBUT-average was significantly lower in 1-month group (P=0.019). Compared with over 1-year group, NITBUT-first was significantly lower in 1-week and 1-month groups (P=0.014, P=0.035, respectively), and NITBUT-average was significantly lower in 1-month group (P=0.022). Bulbar redness score was significantly lower in 1-month group than 1-week group (P=0.004). No significant difference in TMH was found between all groups. MicroRNAs (miRNAs), a promising therapeutic nucleic acid drugs being secreted extracellularly in exosomes, have been reported to elicit a regulatory role in a wide array of biological process, including osteogenesis and angiogenesis. However, poor cellular uptake and endosomal entrapment are still central issues that limited their therapeutic outcomes. Herein, nanoscale zeolitic imidazolate framework (ZIF) was attempted as a vector of miRNA delivery to achieve efficient cellular uptake and the release of payloads at a target intracellular site. Proangiogenic miR-21 and pro-osteogenic miR-5106 were selected as the model miRs and incorporated into miR@ZIF-8 nanocomposites via a one-pot method. The results verified that ZIF-8 vector could not only exhibit a high-payload loading efficiency but also promote the cellular uptake, as well as improve the endosomal escape capability of miRNAs. The therapeutic effects of miR@ZIF-8 were further systematically investigated. RNA sequencing analysis demonstrated that MAPK signaling pathway and PID-HIF1-TF pathway were upregulated by miR-21@ZIF-8 transfection in HUVECs, finally leading to the promotion of angiogenesis, demonstrated by in vitro and in vivo evaluations. Moreover, the miR-5106@ZIF-8 significantly improved the osteogenic differentiation of BMSCs in vitro and efficiently enhanced bone formation in vivo. In summary, we envisage that the ease of fabrication, versatility, low cytotoxicity, promoted cellular uptake, and promising endosomal escape performance make the miR@ZIF-8 nanocomposite be a potential vector for the efficient delivery of therapeutic nucleic acid drugs.
