Cadmium (Cd) contamination of soil can strongly impact human health through the food chain due to uptake by crop plants. Inorganic immobilizing agents such as silicates and phosphates have been shown to effectively reduce Cd transfer from the soil to cereal crops. However, the effects of such agents on total Cd and its bioaccessibility in leafy vegetables are not yet known. Pak choi (Brassica rapa L. ssp. chinensis) was here selected as a representative leafy vegetable to be tested in pots to reveal the effects of silicate–phosphate amendments on soil Cd chemical fractions, total plant Cd levels, and plant bioaccessibility. The collected Cd contaminated soil was mixed with control soil at 1:0, 1:1, 1:4, 0:1 with a view to Cd high/moderate/mild/control soil samples. Three heavy metal-immobilizing agents: wollastonite (W), potassium tripolyphosphate (KTPP), and sodium hexametaphosphate (SHMP) were added to the soil in order to get four different treatment groups, i.e., control (CK), application of wollastonite alone (W), wollastonite co-applied with KTPP (WKTPP), application of wollastonite co-applied with SHMP (WSHMP) for remediation of soils with different levels of Cd contamination. All three treatments increased the effective bio-Cd concentration in the soils with varying levels of contamination, except for W under moderate and heavy Cd contamination. The total Cd concentration in pak choi plants grown in mildly Cd-contaminated soil was elevated by 86.2% after WKTPP treatment compared to the control treatment could function as a phytoremediation aid for mildly Cd-contaminated soil. Using an in vitro digestion method (physiologically based extraction test) combined with transmission electron microscopy, silicate and phosphorus agents were found to reduce the bioaccessibility of Cd in pak choi by up to 66.13% with WSHMP treatment. Application of silicate alone reduced soil bio-Cd concentration through the formation of insoluble complexes and silanol groups with Cd, but the addition of phosphate may have facilitated Cd translocation into pak choi by first co-precipitating with Ca in wollastonite while simultaneously altering soil pH. Meanwhile, wollastonite and phosphate treatments may cause Cd to be firmly enclosed in the cell wall in an insoluble form, reducing its translocation to edible parts and decreasing the bioaccessibility of Cd in pak choi. This study contributes to the mitigation of Cd bioaccessibility in pak choi by reducing soil Cd concentration through in situ remediation and will help us to extend the effects of wollastonite and phosphate on Cd bioaccessibility to other common vegetables. Therefore, this study thus reveals effective strategies for the remediation of soil Cd and the reduction of Cd bioaccessibility in crops based on two indicators: total Cd and Cd bioaccessibility. Our findings contribute to the development of methods for safer cultivation of commonly consumed leafy vegetables and for soil remediation.

The degree and amount of sand production in hydrate reservoirs is related to the selection of stable production processes, but there is currently a lack of quantitative sand production prediction research using real logging data and formation samples from hydrate reservoirs. To reveal the dynamic change characteristics of in-situ reservoirs during hydrate decomposition, and explore quantitative prediction methods for guiding production practice, it is conducted a series of numerical simulations and quantitative prediction experiments. The numerical simulations are carried out using different sand-out prediction methods by using hydrate logging data during drilling, while quantitative prediction experiments of water production and sand-out are carried out based on in-situ reservoir samples. Our experiments indicate that hydrate mining is facing a serious risk of sand-out. The particle transport in the reservoir changes from “large-channel seepage” to “umbrella seepage” and then to “uniform fine flow” as the replacement flow rate decreases. A quantitative prediction model for water and sand production is also established. As a result, our study can provide support for the advancement of technology for long-term stable production and sand control of hydrates, laying the groundwork for developing a stable production plan for natural gas hydrates in offshore areas and determining the optimal depressurisation method.

Natural gas hydrate is widely distributed, shallow-buried, clean, and pollution-free and has enormous reserves, it is regarded as the alternative clean energy source in the oil and gas field with the most potential. Pressure coring is the only way to drill for gas hydrate core on the surface under in situ conditions, which is of great value for analyzing its occurrence conditions and reserves comprehensively. Based on this, a new wireline pressure coring system (WPCS) with a ball valve seal was designed and developed in this paper; it was applied in the deep sea for the first time in the South China Sea hydrate survey voyage of the Guangzhou Marine Geological Survey (GMGS). A total of 15 runs of deep-sea gas hydrate drilling and coring applications were carried out, and they tested well. The experimental water depth was 1700–1800 m, and the coring depth below the seafloor was about 100–150 m. The formation consisted of sandy hydrate and argillaceous hydrate. The results showed the following. (1) The success rate of ball valve turn-over could reach almost 100% in the argillaceous hydrate reservoir, although there are some isolated cases of pressure relief. Meanwhile, drilling in the sandy hydrate reservoir, the success rate was only 54.55%. (2) When drilling in the argillaceous hydrate reservoir, the core recovery rate could reach 80%, while in the sandy hydrate reservoir, it was almost 0%. In practice, the sandy formation with gas hydrate is stiff to drill compared to the performance in argillaceous formations. After our analysis, it was believed that the ball valve and core tube could be easily plugged by sand debris during the sampling of sandy hydrate formation. Moreover, the sandy core is easily plugged into the core liner because of the high friction of sand grains in clearance. (3) The pressure-holding effect of the core drilling tool was related to the formation of hydrate, the sealing form of the ball valve, and the environmental pressure. Sandy hydrate formations often caused the ball valve to jam, while the muddy hydrate formation did not. The research results of this paper have reference value for the further optimization of the WPCS structure, the optimization of drilling parameters, and the design parameters of the ball valve structure, which could be better used for the pressure coring of gas hydrate and subsequent research work in the future.

Abstract The dental operative microscope has been widely employed in the field of dentistry, particularly in endodontics and operative dentistry, resulting in significant advancements in the effectiveness of root canal therapy, endodontic surgery, and dental restoration. However, the improper use of this microscope continues to be common in clinical settings, primarily due to operators’ insufficient understanding and proficiency in both the features and established operating procedures of this equipment. In October 2019, Professor Jingping Liang, Vice Chairman of the Society of Cariology and Endodontology, Chinese Stomatological Association, organized a consensus meeting with Chinese experts in endodontics and operative dentistry. The objective of this meeting was to establish a standard operation procedure for the dental operative microscope. Subsequently, a consensus was reached and officially issued. Over the span of about four years, the content of this consensus has been further developed and improved through practical experience.

To further understand the characteristics of clay and sand production (hereafter collectively referred to as sand production) and to provide optimization designs of sand control schemes are critical for gas production from clayey silt natural gas hydrate reservoirs in the South China Sea. Thus, gas-water-sand production behavoirs and coupling reservoir subsidence characteristics before, during, and after hydrate dissociation of the clayey silt hydrate reservoirs with different clay contents (5%, 10%, 15%, 20%, 25%, and 30%) have been studied through a self-developed experimental system. The results show that with the increase of clay content, the total mass of sand production first increases and then decreases, and it reaches maximum when the clayey content is 20%. The sand production is the lowest before hydrate dissociation and increases significantly during hydrate dissociation, which mainly occurs in the high-speed gas and water production stage at the beginning of hydrate dissociation. After hydrate dissociation, the sand production decreases significantly. During the whole depressurization process, the clay and free sand particles generally move to the sand outlet due to the fluid driving force and overlying stress extrusion. However, for conditions of high clay contents, those particles fail to pass through the sand control screen and gradually accumulate and block the screen by forming a mud cake, which greatly reduce the permeability of the screen and limite sand production as well as gas and water production. Our research lays a foundation for sand production prediction and sand control scheme selection during gas recovery from clayey silty hydrate reservoirs that greatly need to consider a balance between sand control and gas productivity.

Abstract The centenary of insulin discovery represents an important opportunity to transform diabetes from a fatal diagnosis into a medically manageable chronic condition. Insulin is a key peptide hormone and mediates the systemic glucose metabolism in different tissues. Insulin resistance (IR) is a disordered biological response for insulin stimulation through the disruption of different molecular pathways in target tissues. Acquired conditions and genetic factors have been implicated in IR. Recent genetic and biochemical studies suggest that the dysregulated metabolic mediators released by adipose tissue including adipokines, cytokines, chemokines, excess lipids and toxic lipid metabolites promote IR in other tissues. IR is associated with several groups of abnormal syndromes that include obesity, diabetes, metabolic dysfunction-associated fatty liver disease (MAFLD), cardiovascular disease, polycystic ovary syndrome (PCOS), and other abnormalities. Although no medication is specifically approved to treat IR, we summarized the lifestyle changes and pharmacological medications that have been used as efficient intervention to improve insulin sensitivity. Ultimately, the systematic discussion of complex mechanism will help to identify potential new targets and treat the closely associated metabolic syndrome of IR.

Objective. Human dental pulp stem cells (hDPSCs) were recognized as a suitable and promising source of stem cells in dental pulp regeneration. However, the mechanism by which hDPSCs differentiation into osteo-/odontogenic lineage remains unclear. Ena/VASP-like protein (EVL) has been found to be involved in diverse biological processes. In this study, we explored the role and underlying mechanism of EVL in osteo-/odontogenic differentiation of hDPSCs. Methods. Expression of EVL was detected in hDPSCs by quantitative reverse transcription polymerase chain reaction (qRT-PCR) and western blot (WB) analyses during osteo-/odontogenic differentiation. The function of EVL in osteo-/odontogenic differentiation and involvement of MAPK signaling pathways were evaluated by alkaline phosphatase (ALP) staining and activity, alizarin red staining (ARS), and qRT-PCR and western blot analyses. Results. The expression of EVL was upregulated during osteo-/odontogenic differentiation of hDPSCs. Overexpression of EVL significantly increased osteo-/odontogenic capacity of hDPSCs, which was reflected in increased alkaline phosphatase (ALP) staining, ALP activity, mineralized nodule formation, and the expressions of genes related to osteo-/odontogenic differentiation, while downregulation of EVL inhibited it. In addition, EVL activated the JNK pathway and phosphorylation of p38 MAPK during differentiation procedure of hDPSCs. The EVL-enhanced differentiation of DPSCs was suppressed by blocking the JNK pathway, rather than the p38 MAPK pathway. Conclusion. EVL promotes the osteo-/odontogenic differentiation of hDPSCs by activating the JNK pathway, providing a future target for osteo-/odontogenic differentiation and dental pulp regeneration.

Abstract Background Cardiovascular disease (CVD), one of the most common comorbidities of coronavirus disease 2019 (COVID-19), has been suspected to be associated with adverse outcomes in COVID-19 patients, but their correlation remains controversial. Method This is a quantitative meta-analysis on the basis of adjusted effect estimates. PubMed, Web of Science, MedRxiv, Scopus, Elsevier ScienceDirect, Cochrane Library and EMBASE were searched comprehensively to obtain a complete data source up to January 7, 2021. Pooled effects (hazard ratio (HR), odds ratio (OR)) and the 95% confidence intervals (CIs) were estimated to evaluate the risk of the adverse outcomes in COVID-19 patients with CVD. Heterogeneity was assessed by Cochran’s Q-statistic, I2test, and meta-regression. In addition, we also provided the prediction interval, which was helpful for assessing whether the variation across studies was clinically significant. The robustness of the results was evaluated by sensitivity analysis. Publication bias was assessed by Begg’s test, Egger’s test, and trim-and-fill method. Result Our results revealed that COVID-19 patients with pre-existing CVD tended more to adverse outcomes on the basis of 203 eligible studies with 24,032,712 cases (pooled ORs = 1.41, 95% CIs: 1.32-1.51, prediction interval: 0.84-2.39; pooled HRs = 1.34, 95% CIs: 1.23-1.46, prediction interval: 0.82-2.21). Further subgroup analyses stratified by age, the proportion of males, study design, disease types, sample size, region and disease outcomes also showed that pre-existing CVD was significantly associated with adverse outcomes among COVID-19 patients. Conclusion Our findings demonstrated that pre-existing CVD was an independent risk factor associated with adverse outcomes among COVID-19 patients.

Recently, deepwater resource exploration has grown rapidly. Because the conditions of marine environment and seabed geology are more complex, deepwater drilling needs to numerous confront challenges, such as more complicated wellbore situations, low drilling efficiency, and high cost. Advanced novel drilling methods serve as significant impetus to facilitate the rapid advancement in deepwater oil-and-gas exploration and development. However, adopting riserless drilling methods may pollute marine environment and yield poor wall protective effects, while drilling methods with risers may suffer from relatively high cost and risk. Based on these dilemmas, in this study, a novel deepwater closed-cycle riserless drilling method with a subsea pump + gas combined lift is proposed. The proposed novel closed-cycle method has also established a multiphase flow drilling model and analyzed the effects of drilling fluid displacement, gas injection displacement, gas injection site and seawater depth on the multiphase flow in the wellbore. The simulation results revealed the following: As the gas migrates upward along the pipeline, its flow velocity first increases slowly and then rapidly owing to the volume expansion of gas. Larger displacement of drilling fluid demands greater working power of the subsea lifting pump, which is characterized by a nonlinear relationship. The gas injection displacement can effectively mitigate the load-bearing capacity of the pump, and increasing gas injection displacement leads to a decreased subsea lifting pump working power requirement; the decreasing effect on pump power load is more significant in the case of low gas injection displacement. Increasing the depth of gas injection sites reduces the subsea pump working with a decreasing slope with respect to the power descent. Finally, the subsea pump lifting power demand increases approximately linearly with an increasing seawater depth. Subsequently, an optimization method of hydraulic parameters for deepwater closed-cycle riserless drilling was proposed, which provides a theoretical foundation for the selection of subsea pumping power as well as the optimization of gas injection sites and displacement.

Hydrate-bearing sediments provide excellent materials for studying the primary sources and diagenetic alterations of organic matter. In this study, the elemental and isotopic signatures of total organic carbon (TOC), total inorganic carbon (TIC), total nitrogen (TN), and total sulfur (TS) are systematically investigated in three hydrate-bearing sediment cores (∼240 m) retrieved from the Shenhu area, South China Sea. All sediment layers from three sites are with low TOC content (average 0.35%) with marine and terrestrial mixed sources (-23.6‰ < δ13Corg < -21.4‰). However, the generally low δ15N (2.49–5.31‰) and C/N ratios (4.35–8.2) and their variation with depth cannot be explained by the terrestrial sources (Pearl River) and marine sources, binary end-member mixing processes. Contribution from lateral allochthonous organic matter from the mountainous river is considered after excluding other possible factors and ingeniously elucidating the organic matter origins. Furthermore, specific layers in W01B and W02B exhibit elevated S/C ratios (up to 2.39), positive bias of δ34S-TS (up to +29.7‰), and negative excursion of δ13C-TIC (up to -8.29‰), which are the characteristics of sustained occurrence of sulfate-driven anaerobic oxidation of methane. The occurrence of coupled carbon–sulfur anomaly may be accompanied by deep hydrocarbon leakage and the formation of hydrate with high saturation.

The hard rock stratum will seriously affect the efficiency of deep-sea drilling and greatly increase the cost of drilling operations due to its complexity. In ocean drilling operations, the design of the bit is critical. Therefore, the core bit should be able to drill in a variety of formations, especially in hard rock stratum, with the function of reducing the need to change the bit due to formation changes and improving the drilling efficiency. In this paper, three different core bits were designed according to the hard rock stratum of the seabed: the roller bit, diamond bit and bionic bit. According to the test results, it was observed that: (1) It is fully proven that the roller core bit has the characteristics of stable drilling in hard strata, small torque and high drilling efficiency. The same method can realize the core in shallow soft strata through the setting of drilling parameters. (2) For the diamond bit, the field test data show that the new formula diamond bit has a good adaptability to the changing hard rock strata in this area, with an average rate of penetration (ROP) of 4.4 m/h and a bit life of 137.75 m. (3) For the bionic bit, field experiment data showed that drilling in the formation lithology was gray green tuff, which had a core recovery up to 100%. The average ROP was 1.35 m/h, and the highest ROP was 1.88 m/h. The design and experiment of three kinds of drill bits have effectively solved the technical problem of efficient drilling in deep-sea hard-rock stratum.

BackgroundThe association between environmental and socioeconomic risk factors and the occurrence of hepatocellular carcinoma (HCC) are still inconclusive. A meta-analysis was conducted to address this issue.MethodsWe systematically searched the databases including PubMed, Web of Science, and Google Scholar and collected the related risk factors of HCC before March 6, 2020. Statistical analysis was performed on the odds ratio (OR) value and 95% CI of the correlation between environmental and socioeconomic factors and HCC. Begg's rank correlation test, Egger's linear regression test, and the funnel plot were employed for identification of the publication bias.ResultsOut of 42 studies, a total of 57,892 participants were included. Environmental and socioeconomic risk factors including ever educated (illiteracy); race (Black, Hispanic, and Asian); medium and low incomes; occupations (farmer and labor); passive smoking; place of residence (rural); blood aflatoxin B1 (AFB1) adduct level; exposure of pesticide, etc., were statistically increased with the occurrence of HCC (P < 0.05) and OR values and 95% CIs were 1.37 (1.00, 1.89), 2.42 (1.10–5.31), 1.90 (0.87–4.17), 5.36 (0.72–40.14), 1.48 (1.11, 1.96), 1.74 (1.00–3.03), 1.49 (1.06–2.08), 1.52 (1.07–2.18), 1.43 (0.27, 7.51), 1.46 (1.09, 1.96), 2.58 (1.67–3.97), and 1.52 (0.95–2.42), respectively. We found 6–9, 9–12, and ≥12 years of education that statistically reduced the risk of the occurrence of HCC (P < 0.05) and OR values and 95% CIs were 0.70 (0.58, 0.86), 0.52 (0.40, 0.68), and 0.37 (0.23, 0.59), respectively. No significant associations (P > 0.05) were observed between race (Hispanic and Asian), passive smoking, marital status, place of birth, place of residence, and HCC. In stratified analysis, exposure of pesticide was statistically significant (P < 0.05), while race of black was on the contrary.ConclusionEnvironmental and socioeconomic risk factors have great impacts on the incidence rate of HCC. Abstract Background Osteoporosis (OP) is one of the commonly seen bone diseases with low bone mineral densities and trauma fractures. Accumulative studies have demonstrated that the occurrence of OP is closely related to osteoclasts differentiation. LncRNA FTX has been demonstrated to inhibit the development of some human cancers. However, its potential functions in human OP remains to be elusive. Methods The expressions of FTX and miR-137 in bone and serum samples of patients with or without OP were measured. Bioinformatics analysis, RIP assays and luciferase reporter assays were performed to examine the upstream and downstream transactional factors of miR-137. Functional assays were conducted to check the roles of the Notching1 signaling pathway OP. Results FTX was suppressed in OP samples and serums, however, miR-137 was greatly elevated. FTX reduced osteoclast-genesis and inhibited osteogenic differentiation by targeting miR-137. This also inhibited the Notch1 signaling pathway. Conclusion Our experiments and results pointed out that lncRNA FTX up-regulated miR-137 in OP through the Notch1 signaling pathway.

Staphylococcus aureus (S. aureus), which is one of the most important species of Staphylococci, poses a great threat to public health. Clustered regularly interspaced short palindromic repeats (CRISPR) and their CRISPR-associated proteins (Cas) are an adaptive immune platform to combat foreign mobile genetic elements (MGEs) such as plasmids and phages. The aim of this study is to describe the distribution and structure of CRISPR-Cas system in S. aureus, and to explore the relationship between CRISPR and horizontal gene transfer (HGT). Here, we analyzed 67 confirmed CRISPR loci and 15 companion Cas proteins in 52 strains of Staphylococci with bioinformatics methods. Comparing with the orphan CRISPR loci in Staphylococci, the strains harboring complete CRISPR-Cas systems contained multiple CRISPR loci, direct repeat sequences (DR) forming stable RNA secondary structures with lower minimum free energy (MFE), and variable spacers with detectable protospacers. In S. aureus, unlike the orphan CRISPRs away from Staphylococcal cassette chromosome mec (SCCmec), the complete CRISPR-Cas systems were in J1 region of SCCmec. In addition, we found a conserved motif 5′-TTCTCGT-3′ that may protect their downstream sequences from DNA interference. In general, orphan CRISPR locus in S. aureus differed greatly from the structural characteristics of the CRISPR-Cas system. Collectively, our results provided new insight into the diversity and characterization of the CRISPR-Cas system in S. aureus.

Identifying and extracting male and female parent of hybrid rice and then accurately judging the spikelet flowering of male parents is the basis of hybrid rice pollination. Currently, male parent flowering information extraction for hybrid rice is basically obtained by manual recognition. In this study, remote sensing images of parental rice fields were obtained with a multispectral camera carried by a UAV (Umanned Aerial Vehicle). Six kinds of visible light vegetation indices and four kinds of multispectral vegetation indices, together with two classification methods, pixel-based supervised classification and sample-based object-oriented classification, were applied to identify the male and female parents of hybrid rice, after which the accuracies of the methods were compared. The results showed that the visible vegetation index had a better effect in pixel-based supervised classification. The kappa coefficient of ExGR (Excess Green minus Excess Red index) classification was 0.9256 and the total accuracy was 0.9552. The extraction accuracy was higher than that of the other vegetation indices and object-oriented classification. In pixel-based supervised classification, the maximum likelihood method achieved the highest identification accuracy and shortest calculation time. Taking the remote sensing images obtained with a UAV as a data source, maximum likelihood supervised classification based on ExGR index can more effectively and quickly identify the field information of male and female parents of hybrid rice so as to provide a reference for determining optimal pollination timing for hybrid rice in large-scale seed production farms.

Molecular dynamics simulations were performed to study the effects of temperatures, pressures, and methane mole fractions on the generation behaviors, dynamic properties, and interfacial structures of methane gas bubbles. Methane gas bubbling can be promoted by high temperatures and high mole fractions of methane, which come from the generation of larger methane clusters in solution. Bubbles were found to be highly dynamic, with more methane molecules exchanging between bubbles and the surrounding solution at high pressures and in systems with high mole fractions of methane. The interfacial structures between bubbles and the surrounding solution were rough at a molecular level, and the roughness of the outermost methane and water molecules was high at high temperatures, low pressures, and in systems with high methane mole fractions. The dissolution of methane molecules depended on the interactions between the outermost methane and water molecules, which would become stronger with decreasing temperatures, increasing pressures, and decreasing methane mole fractions. The results obtained can help in understanding both the generation behaviors of bubbles when gas hydrates decompose and the re-nucleation behaviors of gas hydrates in the presence of bubbles.

Objectives: To evaluate the efficacy of hydrocortisone in patients with septic acute kidney injury (SAKI). Methods: This retrospective cohort study consisted of all consecutive patients with SAKI who were admitted to the Taizhou First People's Hospital from March 2016 to February 2018. The patients who were treated with usual care including antibiotics, fluid resuscitation, and blood glucose control were regarded as the control group, and those received add-on hydrocortisone by the clinicians' discretion was considered in the intervention group. Hydrocortisone was administered as a 50 mg intravenous bolus every six hours for seven days. To adjust the potential baseline differences between the hydrocortisone and control groups, a 1:1 propensity score matching (PSM) was performed to identify a matched control subject for each patient in the hydrocortisone group. Results: In the propensity-matched cohort, the 28-day mortality was significantly lower for patients in the hydrocortisone group (p = .04). Rock-Eval pyrolysis and the biomarker composition of organic matter were systematically studied in hydrate-bearing sediments from the Shenhu area, South China Sea. The n-alkane distribution patterns revealed that the organic matter in the sediments appeared to originate from mixed sources of marine autochthonous input, terrestrial higher plants, and ancient reworked organic matter. The low total organic carbon contents (average < 0.5%) and the low hydrogen index (HI, 2+ hydrate-bound gases. The biomarker of methanogens, squalane, was recognized in the sediments of this study, possibly suggesting the generation of secondary microbial gases which are coupled with the biodegradation of the deep allochthonous hydrocarbons.

Although numerous COVID-19 vaccines are effective against COVID-19 infection and variants of concern (VOC) in the real world, it is imperative to obtain evidence of the corresponding vaccine effectiveness (VE). This study estimates the real-world effectiveness of the BNT162b2 and mRNA-1273 vaccines against COVID-19 infection and determines the influence of different virus variants on VE by using test-negative design (TND) studies. We systematically searched for published articles on the efficacy of BNT162b2 and mRNA-1273 against COVID-19 infection. Two researchers independently selected and extracted data from eligible studies. We calculated the VE associated with different vaccine types, SARS-CoV-2 variants, and vaccination statuses, using an inverse variance random-effects model. We selected 19 eligible studies in the meta-analysis from 1651 records. For the partially vaccinated group, the VE of BNT162b2 and mRNA-1273 was 61% and 78% against COVID-19 infection, respectively. For the completely vaccinated group, the VE of BNT162b2 and mRNA-1273 was 90% and 92% against COVID-19 infection, respectively. During subgroup analyses, the overall VE of BNT162b2 and mRNA-1273 against the Delta variant was 53% and 71%, respectively, for the partially vaccinated group; the respective VE values were 85% and 91% for the fully vaccinated group. Irrespective of the BNT162b2 or mRNA-1273 vaccines, the Delta variant significantly weakened vaccine protection for the partially vaccinated group, while full vaccination was highly effective against COVID-19 infection and various VOC. The mRNA-1273 vaccine is more effective against COVID-19 infection and VOC than the BNT162b2 vaccine, especially for the partially vaccinated group. Overall, the results provide recommendations for national and regional vaccine policies.

Abstract Salinization is one of the major factors that threaten the existence of plants worldwide. Populus euphratica has been deemed to be a promising candidate for stress response research because of its high capacity to tolerate extreme salt stress. We carried out a genome-wide transcriptome analysis to identify the differentially expressed genes (DEGs) response to salt shock and elucidate the early salt tolerance mechanisms in P. euphratica. Both hierarchical clustering and DEG analysis demonstrated a predominant variation from time-course rather than NaCl intensity within 24 hours salt shock. Among the identified 1,678 salt-responsive DEGs, 74.1% (1,244) have not been reported before. We further created an integrated regulatory gene network of the salt response in P. euphratica by combining DEGs, transcription factors (TFs), Helitrons, miRNAs and their targets. The prominent pathways in this network are plant hormone transduction, starch and sucrose metabolism, RNA transport, protein processing in endoplasmic reticulum, etc. In addition, the network indicates calcium-related genes play key roles in P. euphratica response to salt shock. These results illustrated an overview of the systematic molecular response in P. euphratica under different intensities of salt shock and revealed the complex regulatory mechanism.

Three hundred gas samples recovered from SHSC-4 during China’s first gas hydrate production test in the South China Sea were examined for gas component and isotopic composition. According to the gas chromatography analysis, all the gas samples from SHSC-4 are predominated by CH4, with minor N2 + O2, as well as trace amounts of CO2, C2H6, and C3H8. No H2S was detected. The molecular and isotopic data of the gas samples fall into the region of “mixed origin” on the plot of C1/(C2 + C3) − δ13C1, which is close to the microbial origin. The discrimination diagram of δ13C1 − δDCH4 shows that the methane in all of the samples is of microbial origin, and is derived from the CO2 reduction.

The formation and development of maize kernel is a complex dynamic physiological and biochemical process that involves the temporal and spatial expression of many proteins and the regulation of metabolic pathways. In this study, the protein profiles of the endosperm and pericarp at three important developmental stages were analyzed by isobaric tags for relative and absolute quantification (iTRAQ) labeling coupled with LC-MS/MS in popcorn inbred N04. Comparative quantitative proteomic analyses among developmental stages and between tissues were performed, and the protein networks were integrated. A total of 6,876 proteins were identified, of which 1,396 were nonredundant. Specific proteins and different expression patterns were observed across developmental stages and tissues. The functional annotation of the identified proteins revealed the importance of metabolic and cellular processes, and binding and catalytic activities for the development of the tissues. The whole, endosperm-specific and pericarp-specific protein networks integrated 125, 9 and 77 proteins, respectively, which were involved in 54 KEGG pathways and reflected their complex metabolic interactions. Confirmation for the iTRAQ endosperm proteins by two-dimensional gel electrophoresis showed that 44.44% proteins were commonly found. However, the concordance between mRNA level and the protein abundance varied across different proteins, stages, tissues and inbred lines, according to the gene cloning and expression analyses of four relevant proteins with important functions and different expression levels. But the result by western blot showed their same expression tendency for the four proteins as by iTRAQ. These results could provide new insights into the developmental mechanisms of endosperm and pericarp, and grain formation in maize.

Grain weight is one of the most important yield components and a developmentally complex structure comprised of two major compartments (endosperm and pericarp) in maize (Zea mays L.), however, very little is known concerning the coordinated accumulation of the numerous proteins involved. Herein, we used isobaric tags for relative and absolute quantitation (iTRAQ)-based comparative proteomic method to analyze the characteristics of dynamic proteomics for endosperm and pericarp during grain development. Totally, 9539 proteins were identified for both components at four development stages, among which 1401 proteins were non-redundant, 232 proteins were specific in pericarp and 153 proteins were specific in endosperm. A functional annotation of the identified proteins revealed the importance of metabolic and cellular processes, and binding and catalytic activities for the tissue development. Three and 76 proteins involved in 49 Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways were integrated for the specific endosperm and pericarp proteins, respectively, reflecting their complex metabolic interactions. In addition, four proteins with important functions and different expression levels were chosen for gene cloning and expression analysis. Different concordance between mRNA level and the protein abundance was observed across different proteins, stages, and tissues as in previous research. These results could provide useful message for understanding the developmental mechanisms in grain development in maize.

A high-performance liquid chromatography method is presented for the enantioseparation and quantitation of tetramethrin. The separation is achieved on amylose 3,5-dimethylphenyl-carbamate CSP (Chiralpak AD-H column) using a mobile phase consisting of a mixture of n-hexane-ethanol-2-propanol (99:0.9:0.1, v/v/v). Baseline chiral separation for the four isomers of tetramethrin is obtained within 20 min. Each of the resolutions of the two pairs of enantiomers is more than 2.0. The absolute configurations of each isomer are determined. The accuracy, precision, linearity, and limits of detection and quantitation of the method are investigated. For the determination of isomers in soil, the soil sample is extracted with acetone in an ultrasonic bath. The percentage recoveries from soil are in the range of 73.5% to 87.9%.
