For vapor condensation, the control of heterogeneous nucleation and spatial distribution of nuclei are crucial for regulating droplet dynamics and improving condensation efficiency. However, due to the complex characteristics of multicomponent, multiphase, and multiscale, the underlying mechanism of mixed vapor condensation remains unclear, especially at the nucleation stage. In this paper, we focus on the enrichment effects of non-uniform wettability surfaces by molecular dynamics simulation, which could intensify the droplet nucleation and growth processes in a water-air mixed system. The results clarify the inhibitory effect of non-condensable gas on droplet nucleation and prove that only 1% of non-condensable gas could reduce one half of the condensation performance from a molecular perspective. Since apoptosis of foam, cells can induce plaque instability, reducing intracellular lipid content while protecting foam cells from apoptosis is beneficial for the safe and efficient therapy of atherosclerosis. In this study, osteopontin-coupled polydopamine (PDA-OPN) nanoparticles were synthesized and applied to target mild photothermal therapy (PTT) of atherosclerosis. The results from laser confocal microscopy indicate that PDA-OPN nanoparticles can be specially recognized and absorbed by foam cells. Under near-infrared laser irradiation, the mild photothermal generated by PDA-OPN decreases intracellular lipid accumulation but does not induce cell apoptosis. In vivo treatments demonstrate that mild PTT can substantially reduce plaque area and improve plaque stability by upregulating the expression of plaque fibrosis in ApoE−/− mice. Our findings reinforce that the PDA-OPN nanoparticle-mediated mild PTT can inhibit atherosclerotic progression, which provides new insights for developing safe and effective treatment methods for atherosclerosis.

Matrix metalloproteinases (MMPs), a class of zinc-contained endopeptidases, are closely involved in tumor growth, infiltration, metastasis, and angiogenesis. By virtue of the specifically enzymatic hydrolysis, MMPs have been widely used to turn on imaging and/or therapy function of elaborately designed enzyme-responsive nanoprobes, which is expected to realize precise diagnosis and treatment of cancer. This review systematically summarizes the classifications of MMPs, their substrates and recognized sequences, and overexpressed tumor types. The advances of MMPs-responsive nanoprobes for cancer diagnosis and therapy are focused, including trigger mechanism, design principle, and various imaging or therapy modes. Finally, this review analyzes the challenges of MMPs-responsive nanoprobes in clinical application, and provides constructive opinions for future study.

Peptide-mediated biomimetic synthesis of 2D photothermal conversion materials with enhanced photothermal therapy of tumors.

Long noncoding RNAs (lncRNAs) are expressed in different species and different tissues, and perform different functions, but little is known about their involvement in the synthesis or secretion of follicle-stimulating hormone (FSH). In general, we have revealed lncRNA‍‒‍microRNA (miRNA)‍‒‍‍messenger RNA (mRNA) interactions that may play important roles in rat primary pituitary cells. In this study, a new lncRNA was identified for the first time. First, we analyzed the gene expression of lncRNA-m18as1 in different tissues and different stages by reverse transcription-quantitative polymerase chain reaction (RT-qPCR) and observed the localization of lncRNA-m18as1 with fluorescence in situ hybridization, which indicated that this lncRNA was distributed mainly in the cytoplasm. Next, we used RT-qPCR and enzyme-linked immunosorbent assay (ELISA) to analyze the regulation of FSH synthesis and secretion after overexpression or knockdown of lncRNA-m18as1 and found that lncRNA-m18as1 was positively correlated with FSH synthesis and secretion. In addition, mothers against decapentaplegic homolog 2 (Smad2) was highly expressed in our sequencing results. We also screened miR-18a-5p from our sequencing results as a miRNA that may bind to lncRNA-m18as1 and Smad2. We used RNA immunoprecipitation-qPCR (RIP-qPCR) and/or dual luciferase assays to confirm that lncRNA-m18as1 interacted with miR-18a-5p and miR-18a-5p interacted with Smad2. Fluorescence in situ hybridization (FISH) showed that lncRNA-m18as1 and miR-18a-5p were localized mainly in the cytoplasm. Finally, we determined the relationship among lncRNA-m18as1, miR-18a-5p, and the Smad2/3 pathway. Overall, we found that lncRNA-m18as1 acts as a molecular sponge of miR-18a-5p to regulate the synthesis and secretion of FSH through the Smad2/3 pathway.

As nanomedicine-based clinical strategies have continued to develop, the possibility of combining chemotherapy and singlet oxygen-dependent photodynamic therapy (PDT) to treat pancreatic cancer (PaC) has emerged as a viable therapeutic modality. The efficacy of such an approach, however, is likely to be constrained by the mechanisms of drug release and tumor oxygen levels. In the present study, we developed an Fe(III)-complexed porous coordination network (PCN) which we then used to encapsulate PTX (PCN-Fe(III)-PTX) nanoparticles (NPs) in order to treat PaC via a combination of chemotherapy and PDT. The resultant NPs were able to release drug in response to both laser irradiation and pH changes to promote drug accumulation within tumors. Furthermore, through a Fe(III)-based Fenton-like reaction these NPs were able to convert H2O2 in the tumor site to O2, thereby regulating local hypoxic conditions and enhancing the efficacy of PDT approaches. Also these NPs were suitable for use as a T1-MRI weighted contrast agent, making them viable for monitoring therapeutic efficacy upon treatment. Our results in both cell line and animal models of PaC suggest that these NPs represent an ideal agent for mediating effective MRI-guided chemotherapy-PDT, giving them great promise for the clinical treatment of PaC.

Atherosclerosis is a chronic progressive disease, which may result in serious clinical outcomes, such as acute heart events or stroke with high mortality. At present, the clinical problems of atherosclerosis mainly consist of the difficulty in confirming the plaques or identifying the stability of the plaques in the early phase and the shortage of valid treatments. Fortunately, with the development of nanotechnology, various inorganic nanoparticles with imaging enhancement and noninvasive therapy functions have been studied in the imaging and treatment of atherosclerosis, which has brought new hope to patients. This review focuses on the recent progress in the use of inorganic nanoparticles in the diagnosis and therapy of atherosclerosis, including the key processes in the development of atherosclerosis and the mainly involved cells, inorganic nanoparticle-based dual-mode imaging methods classified by the types of targeting cells, and inorganic nanoparticle-based therapeutic approaches, such as photothermal therapy (PTT), photodynamic therapy (PDT), sonodynamic therapy (SDT), drug delivery, gene therapy and imaging-guided therapy for atherosclerosis. Finally, this review discusses the challenges and directions of inorganic nanoparticles in potential clinical translation of anti-atherosclerosis in future. We believe this review will enable readers to systematically understand the progress of the inorganic nanoparticle-based imaging and therapy of atherosclerosis and therefore promote the further development of anti-atherosclerosis.

Exploring novel surface-enhanced Raman scattering (SERS) active materials with high detection sensitivity, excellent biocompatibility, low biotoxicity, and good spectral stability is urgently required for efficacious cancer cell diagnosis. Herein, black TiO2 nanoparticles (B-TiO2 NPs) with crystal-amorphous core-shell structure are successfully developed. Remarkable SERS activity is derived from the synergistic effect of the promising crystal-amorphous core-shell structure. Abundant excitons can be generated by high-efficiency exciton transitions in the crystal core, a feature that provides sufficient charge source. Significantly, the novel crystal-amorphous heterojunction enables the efficient exciton separation at the crystal-amorphous interface, which can effectively facilitate charge transfer from the crystal core to the amorphous shell and results in exciton enrichment at the amorphous shell. Kelvin probe force microscopy (KPFM) confirms the Fermi level of the amorphous layer shifting to a relatively low position compared to that of the crystal core, allowing efficient photoinduced charge transfer (PICT) between the amorphous shell and probe molecules. The first-principles density functional theory (DFT) calculations further indicate that the amorphous shell structure possesses a narrow band gap and a relatively high electronic density of state (DOS), which can effectively promote vibration coupling with target molecules. Moreover, MCF-7 drug-resistant (MCF-7/ADR) breast cancer cells can be quickly and accurately diagnosed based on the high-sensitivity B-TiO2-based SERS bioprobe. To the best of our knowledge, this is the first time the crystal-amorphous core-shell heterojunction enhancement of the TiO2-molecule PICT process, which widens the application of semiconductor-based SERS platforms in precision diagnosis and treatment of cancer, has been investigated.

Simple Summary Triple-negative breast cancer (TNBC) with negative expression of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor 2 (HER2) is considered to be associated with poorer outcomes and a higher risk of recurrence or metastasis owing to a lack of effective targeted therapeutic drugs. The epidermal growth factor receptor (EGFR) functions is a driver of disease progression in most of TNBC that represents a viable target that can be leveraged to guide the intra-tumoral delivery of chemotherapeutic drugs in TNBC patients. Moreover, ultrasound-mediated cavitation (UMC) strategies increase tissue permeability and extravasation through nuclei-dependent cavitation via sonoporation, thus enabling drugs to better enter target tissues. In this research, a combination of active, targeting nano-micelles and UMC was able to inhibit TNBC tumor growth effectively at lower concentrations while reducing treatment-related toxicity. Thus, this is a very promising treatment strategy in the clinical therapy with TNBC and other cancer types. Abstract Triple-negative breast cancer (TNBC) is highly recurring and metastatic breast cancer with overexpressing epidermal growth factor receptor (EGFR). Herein, a series of in vitro and in vivo analyses were used to explore the therapeutic effect of EGFR-targeting nano-micelles (PLGA-PEG/DOX@anti-EGFR) combined with ultrasound-mediated cavitation (UMC). The prepared nano-micelle drug carriers have good biocompatibility and can greatly increase the drug accumulation in tumor regions, thereby reducing off-target toxicity while enhancing anti-tumor efficacy. Moreover, an in vivo analysis of the practical utility of this treatment modality was conducted by using SonoVueTM microbubbles to achieve cavitation under different power intensity levels, with an ultrasonic power intensity of 0.5 W/cm2 maximizing the intra-tumoral blood perfusion. Relative to PLGA-PEG@DOX/anti-EGFR nano-micelles treatment alone, the combination with UMC was better able to suppress tumor growth even at low concentrations. As such, combining actively targeted drug-carrier molecules with UMC represents an effective approach to enhancing therapeutic efficacy while reducing the adverse, systemic effects associated with DOX and other chemotherapeutic drugs, and it can be considered as a promising clinical prospect in the treatment of TNBC.

The development of a simplified theranostic system with high-efficiency for multifunctional imaging-guided photodynamic therapy/photothermal therapy (PDT/PTT) is a great challenge. Therefore, a versatile fabrication strategy was introduced to design new Fe3O4-black TiO2 nanocomposites (Fe-Ti NCs). The Fe-Ti NCs exhibit an intense broad light absorption, high photothermal conversion efficiency, inherited phototherapy, and favorable magnetic resonance imaging (MRI) properties. The in vitro results demonstrate synergistic PTT and PDT capability of Fe-Ti NCs under 808 nm irradiation at low concentration and power density. Fe-Ti NCs also show superior phototherapy performance (PTT/PDT) under 671 nm laser irradiation. The confocal microscopy analysis demonstrates reactive oxygen species (ROS)-mediated synergistic phototherapy. Hematological and histological analysis confirms no evident toxicity of Fe-Ti NCs. The in vivo photoinduced tumor ablation capability of Fe-Ti NCs was assessed and monitored, and a rapid increase in temperature (60 ± 2 °C) after being exposed to 808 nm laser at 0.7 W cm-2 for 5 min was observed. Then, the same change in temperature is observed under 671 nm laser at 0.5 W cm-2. Thus, in vitro and in vivo dual-wavelength laser tumor ablation ability of Fe-Ti NCs verified excellent synergistic phototherapy efficacy against tumors. Moreover, Fe-Ti NCs exhibit superparamagnetic behavior, high magnetization value (48 emu g-1), good r2 relaxivity value (38.2 mM-1 s-1), and excellent T2 imaging capability to monitor therapeutic performance.

Constructing nanocarriers with high drug loading capacity is a challenge, which limits the effective delivery of drugs to solid tumors. Here, we reported a one-pot synthesis of hollow nanoparticles (NPs) encapsulated by doxorubicin (DOX) and modified with polydopamine (PDA) to form PDA@DOX NPs for breast cancer treatment. PDA@DOX NPs demonstrated exceptionally high capacity (53.16%) for loading DOX. In addition, when PDA@DOX NPs were administered systemically, they exhibited responsive aggregation in the tumor sites and demonstrated a good controlled release effect for DOX due to the weak acidic environment of the tumor sites and targeting near-infrared (NIR) light irradiation. The PDA outer layer absorbed the near-infrared (NIR) light and facilitated simultaneous generation of heat energy for destroying the tumor cells to release the drug upon NIR irradiation. Moreover, this NIR-activated combined/synergistic therapy exhibited remarkably complete tumor growth suppression in a breast cancer mouse model. Importantly, NPs exhibited a good ultrasound performance both in vitro and in vivo, which could monitor the treatment process. In conclusion, this NIR-activated PDA@DOX NP system is demonstrated as a good US-guided combination (chemotherapy + PTT) therapy platform with high loading capacity and controlled drug release characteristics, which is promising for the treatment of breast cancer.

Janus nanoparticles (JNPs) have multiple configurations for molecular imaging, targeting, and therapeutic effects on cancers; these properties have made these particles attractive for biomedical applications. Nonetheless, smart strategies for the controlled synthesis in a liquid phase and exploration of the appropriate applications of JNPs remain a challenge. In this study, a unique liquid-phase method was applied to fabricate Mn3O4-TiO2/ZnO/Fe3O4 multifunctional binary transition metal oxide-based JNPs, using the concept of epitaxial growth and lattice mismatch among synthesized materials. Transmission electron microscopy and scanning transmission electron microscopy results revealed that the created materials are embedded in the form of dimers with good dispersion and homogeneous growth in a nonpolar solvent. Pluronic® F-127-coated Mn3O4- TiO2 JNPs were utilized as a contrast agent in T1-weighted magnetic resonance imaging (MRI) and in photodynamic therapy (PDT) for cancers in vitro and in vivo. In vivoT1-weighted MRI of the heart, liver, and kidneys in mice after intravenous injection of the nanoparticles revealed high sensitivity and biocompatibility of as-synthesized Mn3O4-TiO2 JNPs. Results of synchrotron X-ray fluorescence microscopy mapping showed the stability of the nanocomposites and efficiency of penetration into the cytoplasm and perinuclear area. Inorganic TiO2 photosensitizers showed promising tumor ablation performance in PDT in vitro and in vitro at low intensity of UV irradiation (5.6 mW.cm–2) because of their ultrasmall size and photodegradable stability. These results reveal that multifunctional Mn3O4-TiO2 JNPs enhance a T1-weighted MRI contrast and have excellent properties for PDT and therefore, may be a novel agent for cancer theranostics.

Mitochondria-targeted mild-temperature photothermal therapy (MT-PTT) is a promising strategy that can maximize anticancer effects and reduce adverse reactions. Here, a novel photosensitizer with mitochondrial targeting based on IR780 iodide and heat shock protein 90 inhibitor (BIIB021), which can passively accumulate in MCF-7 cells and achieve effective MT-PTT effect is synthesized. The prepared PEG-IR780-BIIB021 nano-micelles possess considerable biocompatibility and biological stability, with an encapsulation efficiency of about 84% for BIIB021. They can selectively enrich in mitochondria, and release BIIB021 after NIR irradiation to reduce cell tolerance to heat, thereby reducing the mitochondrial membrane potential and rapidly affecting key intrinsic apoptotic factors (Cyt-C, Caspase-9, Bcl-2 and Bax) to achieve the effect of MT-PTT. It is believed that mitochondria-targeted MT-PTT generated by the PEG-IR780-BIIB021 nano-micelles is a promising therapeutic strategy in clinical practice.

The antimicrobial peptides (AMPs) are a group of unique naturally occurring anti-microbial compounds with around 50 amino acids. It represents promising therapeutic agents to the infectious disease without concerning about drug resistance. However, commercial development of these peptides for even the simplest application has been hindered by the limitations of sources, instability, toxicity and bioavailability. To improve the properties of the artificial synthesized AMPs, the modification and design are the hotspots of the AMPs research. In fact, more than half of the known AMPs are naturally modified. In this review, two types of modification strategies, biochemical modification and chemical modification were summarized. Although, the chemical modification is versatile and direct, the manufacturing cost is greatly increased compared to the antibiotics. With the recent progress of the protein modification enzyme, the biochemical modification of the antimicrobial peptide followed by heterologous expression has great application prospects.

Aims The present study aimed to investigate the possible effects of metformin on the progression of atherosclerosis in a rabbit model. Main methods Rabbits were randomly divided into three groups (n = 10): the control (Ctrl) group (fed with a chow diet), and two experimental groups, the AS group and the Met group (both received an atherogenic diet). After 2 weeks of acclimatization, the rabbits in the AS and Met groups were given a placebo and metformin, respectively, daily by gavage for 10 weeks. Plasma lipids and inflammatory cytokines were measured. The aorta was isolated for histological and immunohistochemical analysis. In vitro, human umbilical vein endothelial cells (HUVECs) were treated with metformin, and monocyte adhesion and adhesion molecule expression were measured. Key findings Metformin reduced plasma inflammatory cytokine levels but did not alter lipid content. Compared with that in the AS group, the atherosclerosis burden in the Met group was significantly decreased. The lesional macrophage content was reduced, but the lesional collagen content was not affected in the metformin-treated rabbits, compared with the corresponding levels in the non-treated controls. Furthermore, the aortic mRNA expression levels of adhesion molecules and inflammatory cytokines in the Met group were also significantly reduced compared with those in the AS group. Metformin treatment reduced monocyte adhesion to endothelial cells (ECs) and adhesion molecule expression, and inhibited rabbit monocyte differentiation into macrophages and the macrophage inflammatory response. Significance Our results suggest that metformin impeded the progression of atherosclerosis, possibly by suppressing macrophage infiltration and inflammatory responses.

Nanotechnology has introduced new techniques and phototherapy approaches to fabricate and utilize nanoparticles for cancer therapy. These phototherapy approaches, such as photothermal therapy (PTT) and photodynamic therapy (PDT), hold great promise to overcome the limitations of traditional treatment methods. In phototherapy, magnetic iron oxide nanoparticles (IONPs) are of paramount importance due to their wide range of biomedical applications. This review discusses the basic concepts, various therapy approaches (PTT, PDT, magnetic hyperthermia therapy (MHT), chemotherapy and immunotherapy), intrinsic properties, and mechanisms of cell death of IONPs; it also provides a brief overview of recent developments in IONPs, with focus on their therapeutic applications. Much attention is devoted to elaborating the various parameters, intracellular behaviors and limitations of MHT. Bimodal therapies which act alone or in combination with other modalities are also discussed. The review highlights some limitations in the explored research areas and suggests future directions to overcome these limitations.

At present, transmembrane glycoprotein CD133 highly expressed pancreatic cancer stem cells (PCSCs), with the features of chemotherapeutic/radiotherapeutic resistance and exclusive tumorigenic potential, are considered as the primary cause of metastasis and recurrence in pancreatic cancer, and therefore are an effective target in the disease treatment. Furthermore, with the launch of precision medicine, multifunctional nanoprobes have been applied as an efficient strategy for the magnetic resonance imaging (MRI)-guided photothermal therapy (PTT) of pancreatic cancer. In this research, with the aim of achieving precise MRI-guided PTT in CD133 highly expressed PCSCs, novel bTiO2-Gd-CD133mAb nanoprobes were designed and successfully prepared by loading Gd-DOTA and CD133 monoclonal antibodies on black TiO2 nanoparticles. It was very interesting to find that the r1 relaxivity value of the nanoprobes was 34.394 mM-1 s-1, about 7.5 times that of commercial Magnevist (4.5624 mM-1 s-1), which indicates that the nanoprobes have good potential as MRI T1 contrast agents with excellent performance. Herein, CD133 highly expressed PANC-1 cells were selected and verified as PCSCs model. In vitro experiments demonstrated that the nanoprobes exhibited active-targeting ability in PANC-1 cells, and consequently could specially enhance T1-weighted MR imaging and 808 nm near-infrared (NIR)-triggered PTT efficiency in the PCSCs model. Our study not only provides a new strategy for the effective treatment of pancreatic cancer and its' stem cells, but also further broadens the application of black TiO2 in the field of cancer theranostics.

Photothermal therapy (PTT) has emerged as one of the promising cancer therapy approaches. However, nanoparticles (NPs) which are used for PTT might be biopersistent and potentially toxic. The current research explores the promising use of Fe3O4 nanoflowers as nontoxic, efficient photothermal, and strong T2 type magnetic resonance imaging (MRI) contrast agents for imaging-guided photothermal cancer therapy. In this study, a facile solvothermal method is used to fabricate PEG-coated Fe3O4 nanoflowers with controllable dimensions. Their successful fabrication, the effect of the reaction parameters, and their magnetic properties are investigated in depth. The therapeutic performance of the Fe3O4 nanoflowers (Fe-NFs) is evaluated and compared with commercially available black TiO2 nanoparticles (b-TiO2) under an 808 nm laser. The photothermal therapy efficiency of the Fe-NFs is observed to be better than that of the reported Fe3O4 nanoparticles. In vitro and in vivo investigation demonstrates that the therapeutic performance of the Fe-NFs is comparable to that of b-TiO2. Moreover, the Fe-NFs show excellent magnetic properties and magnetic resonance imaging capability to monitor therapeutic performance.

The present study was performed to investigate the seroprevalence and risk factors for Dirofilaria immitis infection in cats from Liaoning province, northeastern China. From October 2014 to September 2016, sera of 651 cats, including 364 domestic cats and 287 feral cats (332 females and 319 males) were assessed. They were tested for the presence of D. immitis antigen using SNAP Heartworm RT test kit. In this population, the average prevalence was 4.5%. Age and rearing conditions (feral or domestic) were found to be associated with the prevalence of D. immitis. The prevalence was significantly higher in feral cats compared with domestic cats (8.4% vs 1.4%, P0.05), but older cats (≥3 years old) showed a statistically higher prevalence compared with younger cats (0.05), all these results suggest that outdoor exposure time may be one of the most important factors for D. immitis prevalence in cats. Results reveal that D. immitis are prevalence in domestic and feral cats in northeastern China, which indicates that appropriate preventive measures should be taken to decrease the incidence of feline heartworm disease in Liaoning province, northeastern China.

Here, we fabricated fluorescent nanobubbles (NBs) by encapsulation of liquid tetradecafluorohexane (C6F14) within biodegradable photoluminescent polymers (BPLPs) through an emulsion-evaporation process and conjugation of PNBL-NPY ligand for specific targeting of Y1 receptors overexpressed in breast tumors. The developed PNBL-NPY modified NBs were uniform in size with good dispersibility and photostability, presenting good ultrasound enhancement. Further, in vitro and in vivo results indicated that the fabricated NBs exhibit high affinity and specificity to Y1 receptor-overexpressing breast cancer cells and tumors with minimal toxicity and damage to organs. Our developed PNBL-NPY-modified NBs are novel targeted UCAs for safe, efficient and specific targeted breast cancer imaging, and may provide a new nanoplatform for early cancer diagnosis and treatment in the future.

TiO2 nanomaterials have been widely used for anticancer drug carriers and UV/980 nm NIR triggered cancer synergistic platforms. However, traditional pure TiO2 nanocarriers encounter some serious drawbacks, such as low drug loading ability, limited tissue penetration of UV light, and heating effect of 980 nm NIR on normal tissue, which obstruct their further application in cancer treatment. To overcome those challenges, novel mesoporous silica (mSiO2) coated black TiO2 core-shell nanocomposites are designed and constructed as doxorubicin carriers for 808 nm NIR triggered thermal imaging guided photothermal therapy combined chemotherapy of breast cancer. Properties of the nanocomposites such as micro-morphology, size, drug loading ability and release, targeting performance, and therapy efficiency in vitro and in vivo were evaluated. The results indicated the core-shell nanocomposites with dramatically increased loading ability were pH-responsive/NIR-accelerated doxorubicin release nanocarriers and showed synergistic breast cancer treatment in vitro and in vivo. This study verifies that the newly prepared mSiO2 coated black TiO2 core-shell nanocarriers can overcome the limitations of traditional TiO2 nanocarriers and thus improve and broaden usage of TiO2 nanoparticles in nanomedicine.
