The BESIII experiment located in Beijing, China, is investigating physics in the energy region of the charm-quark via electron positron annihilation reactions. A small detector to be placed in the very forward/backward region around theta = 0 degree at BESIII is foreseen to measure photons from the initial state. This is especially interesting, because it opens the door for various physics measurements over a wide range of energies, even below the experiment’s designated energy threshold, which is fixed by the accelerator. This thesis is investigating the capabilities of a crystal zero degree detector (cZDD) consisting of PbWO4 crystals placed in that region of BESIII. Detailed Geant4-based simulations have been performed and the energy resolution of the detector has been determined to be sigma/mu = 0.06 + 0.025/sqrt( E[GeV] ). The determination of the center-of-mass energy sqrt(s)_isr after the emission of the photon is of great importance for the study of such events. Preliminary simulations estimated the resolution of the reconstructed sqrt(s)_isr using the cZDD information to be significantly better than 10 % for appropriate photon impacts on the detector. Such events can only be investigated, when data from the cZDD and other detectors of BESIII can be correlated. A fast and powerful Data Acquisition (DAQ) capable of performing event correlation in real time is needed. DAQ modules capable of performing real time event correlation are being developed for the PANDA experiment at the future FAIR facility in Darmstadt, Germany. Investigating these modules in a realistic high-rate environment such as provided at BESIII, offers a great opportunity to gain experience in real time event correlation before the start of PANDA. Developments for the cZDD’s DAQ using prototype PANDA DAQ modules have been done and successfully tested in experiments with radioactive sources and a beamtest with 210 MeV electrons at the Mainz Microtron.

In den letzten Jahren wurde die strukturelle Verankerung von Bildung für nachhaltige Ent- wicklung (BNE) in den Bildungsbereichen vorangetrieben. Auch in der beruflichen Bildung lassen sich Bildungsprozesse nachhaltigkeitsorientiert ausrichten (Holst/Singer-Brodowski, 2020). Mit der neuen Standardberufsbildposition „Umweltschutz und Nachhaltigkeit”, die seit August 2021 verpflichtender Bestandteil aller neugeordneten und neu geschaffenen Ausbil- dungsberufe ist, hat die strukturelle Verankerung von Nachhaltigkeit in der Berufsbildung einen wichtigen Impuls erfahren. Dennoch wird in der Zwischenbilanz des „Nationalen Aktionsplans BNE“ konstatiert, dass eine „transformative berufliche Bildung, die über eine Anpassung an berufliche und gesell- schaftliche Veränderungsprozesse hinausgeht und aus sich heraus aktiv eine gerechte Ent- wicklung für Mensch und Umwelt anstrebt” (Nationale Plattform BNE c/o BMBF, 2020, S. 33), erst am Anfang steht. Um Berufliche Bildung für nachhaltige Entwicklung (BBNE) zielgerich- tet zu implementieren, sind geeignete Indikatoren erforderlich, die den Umsetzungsstand kontinuierlich erfassen, kommunizieren und so Veränderungen anstoßen können. Dabei müs- sen die Besonderheiten des dualen Ausbildungssystems berücksichtigt werden. Neben dem Lernen an den Lernorten Betrieb, überbetriebliche Bildungsstätte und Berufsschule variieren duale Ausbildungsberufe hinsichtlich der curricularen Grundlagen (Ausbildungsrahmen- plan, Rahmenlehrplan, Prüfungsordnungen) in ihrer „Nähe“ zur Leitidee nachhaltiger Ent- wicklung. Hinzu kommt die Heterogenität des Berufsbildungspersonals (haupt- und neben- amtliches Ausbildungspersonal, Berufsschullehrkräfte). Neben den strukturellen Charakteris- tika der Berufsbildung sind vielfältige Akteure an der Gestaltung und Weiterentwicklung des dualen Ausbildungssystems beteiligt, wie z. B. Sozialpartner, Kammern und Verbände. Um die Akzeptanz der Indikatoren zu erhöhen, sollten diese Akteure im Entwicklungsprozess der Indikatoren einbezogen werden. Das vom BMBF geförderte Verbundprojekt „Indikatoren berufliche Bildung für nachhaltige Entwicklung“ (kurz: iBBnE) entwickelte in diesem Kontext Indikatoren, die sich speziell auf das Berufsbildungssystem beziehen. Die Indikatoren verfolgen hierbei zwei maßgebliche Ziele: Sie erfassen zum einen den Ist-Zustand der Umsetzung von nachhaltiger Entwicklung in der Berufsbildung (Zapf, 1973). Zum anderen erfüllen sie durch einen theoriebasierten und normativ begründeten Soll-Zustand eine Hebelfunktion für weitere nötige Umsetzungsmaß- nahmen im Berufsbildungssystem zur Verankerung von Nachhaltigkeit.

Basal-like breast cancer (BLBC) is a highly aggressive breast cancer subtype frequently associated with poor prognosis. Due to the scarcity of targeted treatment options, conventional cytotoxic chemotherapies frequently remain the standard of care. Unfortunately, their efficacy is limited as BLBC malignancies rapidly develop resistant phenotypes. Using transcriptomic and proteomic approaches in human and murine BLBC cells, we aimed to elucidate the molecular mechanisms underlying the acquisition of aggressive and chemotherapy-resistant phenotypes in these mammary tumors. Specifically, we identified and characterized a novel short isoform of Roundabout Guidance Receptor 3 (ROBO3s), upregulated in BLBC in response to chemotherapy and encoding for a protein variant lacking the transmembrane domain. We established an important role for the ROBO3s isoform, mediating cancer stem cell properties by stimulating the Hippo-YAP signaling pathway, and thus driving resistance of BLBC cells to cytotoxic drugs. By uncovering the conservation of ROBO3s expression across multiple cancer types, as well as its association with reduced BLBC-patient survival, we emphasize its potential as a prognostic marker and identify a novel attractive target for anti-cancer drug development.

Chromatin is organized into compartments enriched with functionally-related proteins driving non-linear biochemical activities. Some compartments, e.g. transcription foci, behave as liquid condensates. While the principles governing the enrichment of proteins within condensates are being elucidated, mechanisms that coordinate condensate dynamics with other nuclear processes like DNA replication have not been identified. We show that at the G1/S cell cycle transition, large transcription condensates form at histone locus bodies (HLBs) in a cyclin-dependent kinase 1 and 2 (CDK1/2)-dependent manner. As cells progress through S phase, ataxia-telangiectasia and Rad3-related (ATR) accumulates within HLBs and dissolves the associated transcription condensates. Integration of CDK1/2 and ATR signaling creates a phosphorylation code within the intrinsically-disordered region of mediator subunit 1 (MED1) coordinating condensate dynamics with DNA replication. Disruption of this code results in imbalanced histone biosynthesis, and consequently, global DNA damage. We propose the spatiotemporal dynamics of transcription condensates are actively controlled via phosphorylation and essential for viability of proliferating cells.
