Abstract

Disinformation spreads among the public and in scientific discourse through the actions of individuals, organizations, and governments that distort scholarly communications, media narratives, and institutional trust. This taxonomy introduces a structured framework and specialized set of definitions to elucidate the key participants, platforms, and strategies employed in the propagation of disinformation. Enhanced comprehension of the mechanisms and pathways of scientific disinformation equips journalists and policymakers with the tools necessary to more effectively recognize and address these issues. The authors developed this taxonomy of disinformation through a multi-faceted approach, encompassing a literature review, expert review, and case study analysis. The literature review revealed a scarcity of taxonomical models amidst prevalent algorithmic detection studies. Subsequently, an expert review process refined our taxonomy through collaborative analysis of twenty-two cases of identified disinformation, categorized by their methods, motives, and impacts. Finally, we validated and fine-tuned our taxonomy through detailed case studies of twelve diverse disinformation instances, assessing the taxonomy's effectiveness in capturing the essential characteristics of each case and making necessary adjustments to ensure its relevance and accuracy in real-world applications., Comment: 10 pages, 3 tables

Abstract

In the wake of increased behavioral issues in the peri- and post-COVID environment, schools have resorted to zero-tolerance policies that imposed uniform and predetermined punishments for even the slightest infraction. In this article, the authors argue that these policies represent a failure of imagination that installs punitive policies that do more harm than good. Further, they suggest that restorative justice and/or school-wide positive behavior support systems are better means of creating educational environments in which all students flourish.

Abstract

T cell receptors (TCRs) enable T cells to specifically recognize mutations in cancer cells1-3. Here we developed a clinical-grade approach based on CRISPR-Cas9 non-viral precision genome-editing to simultaneously knockout the two endogenous TCR genes TRAC (which encodes TCRα) and TRBC (which encodes TCRβ). We also inserted into the TRAC locus two chains of a neoantigen-specific TCR (neoTCR) isolated from circulating T cells of patients. The neoTCRs were isolated using a personalized library of soluble predicted neoantigen-HLA capture reagents. Sixteen patients with different refractory solid cancers received up to three distinct neoTCR transgenic cell products. Each product expressed a patient-specific neoTCR and was administered in a cell-dose-escalation, first-in-human phase I clinical trial ( NCT03970382 ). One patient had grade 1 cytokine release syndrome and one patient had grade 3 encephalitis. All participants had the expected side effects from the lymphodepleting chemotherapy. Five patients had stable disease and the other eleven had disease progression as the best response on the therapy. neoTCR transgenic T cells were detected in tumour biopsy samples after infusion at frequencies higher than the native TCRs before infusion. This study demonstrates the feasibility of isolating and cloning multiple TCRs that recognize mutational neoantigens. Moreover, simultaneous knockout of the endogenous TCR and knock-in of neoTCRs using single-step, non-viral precision genome-editing are achieved. The manufacture of neoTCR engineered T cells at clinical grade, the safety of infusing up to three gene-edited neoTCR T cell products and the ability of the transgenic T cells to traffic to the tumours of patients are also demonstrated.

Abstract

Undergraduate seniors at a residential liberal-arts college reflected on their experiences attending on-campus cultural events. These focus groups were transcribed, coded, and analyzed using a grounded theory-derived approach to understand how these events shaped their college experience. The resulting communication artifact revealed a variety of student outcomes tied to on-campus cultural event attendance. The findings have implications for the design, implementation, and assessment of academic and co-curricular programming, particularly in terms of articulating the purpose of such programming and enlisting faculty in its delivery.

33. The dissociative subtype of post-traumatic stress disorder : understanding prevalence, course, prognosis, characteristics, severity, and functional impairment

Abstract

A separate dissociative subtype of post-traumatic stress disorder (PTSD-DS) has been described in the fifth edition of the Diagnostic and Statistical Manual of Mental Disorders. It has been suggested that PTSD-DS represents a small proportion of individuals with PTSD and may be a more pathological and severe subtype, however, there has been limited investigation of this subtype, especially in children, following various forms of traumatic experience. A systematic review and meta-analysis were conducted, synthesising the current evidence relating to point prevalence of a PTSD-DS in children and adults (as a proportion of those with PTSD). In addition, empirical analysis was conducted of pre-existing data from two longitudinal studies of children and adolescents; one in which they had experienced a single-event trauma, the second in which they were under the care of a local authority and had experience of abuse or neglect. Meta-analysis of 49 studies (53 samples; N = 8214) estimated the point prevalence of PTSD-DS in children and adults to be 38.1% (95% CI 31.5-45.0%). Prevalence of PTSD-DS was significantly higher for children compared to adults, and when prevalence was determined by diagnostic or clinical cut-off methods compared to latent class and profile analyses (exploratory methods that determine hidden groups based on the means of categorical and continuous variables respectively). Results of the empirical study found that: PTSD-DS was common in children with PTSD, early PTSD-DS showed similar natural recovery to early PTSD, and PTSD-DS was predictive of later PTSD following single-event trauma. Dissociation did not appear to be an important factor in post-traumatic stress symptoms or functional impairment following single- or multi-event trauma. The results of both studies suggest that PTSD-DS is relatively common, especially in children. PTSD-DS may offer little clinical utility to the extant PTSD diagnosis, and dissociation may simply be a typical part of the PTSD presentation in children. Further research is warranted into PTSD-DS following different forms of trauma and in larger samples of participants with this subtype.

50. The influence of solute diffusion and anisotropic elasticity on the deformation of iron : a study using planar discrete dislocation plasticity

Abstract

Plastic deformation of crystalline metals is facilitated by crystallographic defects called dislocations. In certain alloys at certain temperatures and loading rates, the motion of these dislocations can become chaotic and unstable. This is a phenomenon related to dynamic strain ageing called the Portevin-Le Châtelier effect. The effect occurs when impurities, such as carbon and nitrogen, segregate to dislocations and obstruct their motion, leading to reduced ductility, hardening, and in the worst case, mechanical failure. In this thesis, I model plasticity with planar discrete dislocation plasticity. I extend planar discrete dislocation plasticity using anisotropic elasticity and introduce discrete diffusing solutes. This is used to understand the deformation of iron; and the onset and progression of the Portevin- Le Châtelier effect. Results from the model show that the nature of the diffusing solute field significantly affects the manifestation of the Portevin-Le Châtelier effect. As the solute concentration increases, the iron specimen's proof stress, flow stress and solute strengthening increases; and the serrations recorded in a stress-strain curve (symptomatic of the Portevin-Le Châtelier effect) become smaller and occur at a slower frequency. Conversely, as the solute diffusivity increases, the serrations become larger and occur at a faster frequency. Furthermore, I show that anisotropic elasticity alters the mechanisms of plastic deformation of α-iron at elevated temperatures; specifically, that a<100> type edge dislocations, which are energetically unfavourable at moderate temperatures, are nucleated in increasing numbers. This agrees with experimental observations found in the literature but cannot be predicted by isotropic elastic models. The increase in a<100> edge dislocations is symptomatic of the precipitous decrease in the yield strength of iron. My work has important consequences for the application of steels in elevated temperature environments such as generation IV very high temperature nuclear fission reactors.
