• Modern varieties are derived from wild progenitors through crop domestication. • Limited genetic diversity is a constraint of crop improvement programs to keep pace with the challenges of climate change. • Crop wild relatives and their rich reservoir of gene pool provide the armor against crop stresses. • Genome editing tools are used to explore the germplasm of popular crops for domestication and climate-resilient genes. • Crop domestication is accelerated by Cis-regulating mutation, CRISPR-Cas-, and multiplex editing. Crop domestication is one of the oldest crop improvement methods, significantly impacting socioeconomic conditions globally. Modern varieties are derived from wild progenitors through crop domestication. These varieties are generated by crossing compatible genotypes to obtain the desired phenotype-containing progenies. The uncertainties and stress conditions caused by climate change deeply impact the current agricultural scenario. However, limited genetic diversity is the major constraint of the present crop improvement programs in keeping pace with climate change and several resulting biotic and abiotic stresses. Therefore, using crop wild relative and their rich reservoir of resistant genes to biotic and abiotic stressors must provide the armors against the crop stresses. However, crop wild relatives are unsuitable for direct intensive cropping because of many undesirable agronomic traits and are incompatible for gene transfer with conventional breeding. With the dawn of new-generation sequencing techniques and genome editing tools, scientists are trying to explore the germplasm of popular crops for domestication and climate-resilient genes. Considering the above context, the present review discusses the genetics of crop domestication, different strategies for a targeted mutation to accelerate crop domestication, and the current status of various crops.
